    • The performance of COATEST® APC® Resistance on different coagulation instruments has been further evaluated through analysis of plasma from 100 blood donors on KC 10, ST 4, ACL 300R, Electra 900 and Thrombolyzer. The electromechanical instruments KC-10 and ST 4 showed a lower response to APC than the turbidimetric or photometric instruments with median APC ratios of 2.7 and 2.8 for the two former versus 3.1, 3.4 and 3.5, respectively, for the other three, which is in agreement with earlier initial findings. Similarly, the cut-off value varied between 2.1 and 2.6 for these instruments. The correlation of APC ratios between instruments was strong with r values ranging between 0.71 and 0.93 and, furthermore, none of the six plasmas with the lowest APC ratios on the Thrombolyzer ranked higher than 8 on any of the other instruments. Analysis of control plasmas with six consecutive kit batches on ACL and ST4 resulted in APC ratio ranges of 3.3-3.7 and 2.7-3.0 for a normal control and 1.8-2.0 and 1.9-2.0 for an abnormal control on ACL and ST4, respectively, illustrating a high reproducibility between batches. Repeated freezing and thawing of samples is disrecommended since this often resulted in increased APC ratios. In contrast, in spite of up to 40% decrease in FVIII activity upon storage of 10 different plasma samples for 5 h, the effect on the APC ratio was only minor as was also the effect of addition of 1.0 IU/ml of FVIII. In neither case was any sample misclassified. Altogether, the results support the applicability of this kit for measuring the response of plasma to APC.

  • Cell-surface charge and cell-surface hydrophobicity of collagen-binding Aeromonas and Vibrio strains

    • Abstract Partitioning in aqueous polymer two-phase systems of polyethylene glycol and dextran was used to detect and compare cell-surface charge and cell-surface hydrophobicity of Aeromonas hydrophila, A. caviae, A. sobria, Vibrio cholerae, and V. anguillarum strains. These strains have cell-surface components that bound either native or thermally denatured type I collagen (i.e., a mixture of the alpha1+alpha2 chains) and gelatin immobilized on latex beads. Our goals were: (1) to compare the possible relationship between the cell-surface charge/hydrophobicity and binding to collagen and (2) to evaluate the influence of the culture media on the expression of surface properties. There was no apparent relationship between cell-surface charge, cell-surface hydrophobicity, and binding to collagen. The expression of surface properties was dependent on the culture media. There was no relationship between binding to immobilized collagen and binding to soluble 125I-labeled collagen. Particle-agglutination reactivity differed when using various collagen-coated microbead preparations. There were general differences in the particle-agglutination reactivity when collagen-coated latex beads were prepared using different coating procedures. The negative charge and hydrophobicity of the various collagen-coated microbead preparations were also studied by partitioning in the two-phase system of polyethylene glycol and dextran. Under these conditions, the alpha1+alpha2 collagen-chain mixture covalently immobilized on carboxy-modified latex beads was less hydrophobic and negatively charged than gelatin and native collagen immobilized on the same kind of latex beads. For latex beads passively coated with collagen preparations, the alpha1+alpha2 collagen-chain mixture was more hydrophobic than gelatin and native collagen. We suggest that for screening collagen-binding among Vibrio and Aeromonas strains, a reliable and sensitive particle-agglutination assay should consider the collagen preparation and the coating procedure for the immobilization of collagen onto the latex beads. In this regard, carboxy-modified latex beads coated with an alpha1+alpha2 collagen-chain mixture gave the best results.

    • We report the first results of the AMANDA detector. During the antarctic summer 1993-94 four strings were deployed between 0.8 an 1 km depth, each equipped with 20 photomultiplier tubes (PMTs). A laser source was used to investigate the optical properties of the ice in situ. We find that the ice is intrinsically extremely transparent. The measured absorption length is 59 ± 3 m, i.e. comparable with the quality of the ultra-pure water used in the IMB and Kamiokande proton-decay and neutrino experiments [1,2] and more than two times longer than the best value reported for laboratory ice [3]. Due to a residual density of air bubbles at these depths, the motion of photons in the medium is randomized. For spherical, smooth bubbles we find that, at 1 km depth, the average distance between collisions is about 25 cm. The measured inverse scattering length on bubbles decreases linearly with increasing depth in the volume of ice investigated. © 1995 Elsevier Science B.V. All rights reserved.

    • The first four strings of phototubes for the AMANDA high-energy neutrino observatory are now frozen in place at a depth of 800 to 1000 m in ice at the South Pole. During the 1995-96 season an additional six strings will be deployed at greater depths. Provided absorption, scattering, and refraction of visible light are sufficiently small, the trajectory of a muon into which a neutrino converts can be determined by using the array of phototubes to measure the arrival times of \v{C}erenkov light emitted by the muon. To help in deciding on the depth for implantation of the six new strings, we discuss models of age vs depth for South Pole ice, we estimate mean free paths for scattering from bubbles and dust as a function of depth, and we assess distortion of light paths due to refraction at crystal boundaries and interfaces between air-hydrate inclusions and normal ice. We conclude that the depth interval 1600 to 1800 m will be suitably transparent for the next six AMANDA strings and, moreover, that the interval 1600 to 2100 m will be suitably transparent for a future 1-km 3   observatory except possibly in a region a few tens of meters thick at a depth corresponding to a peak in the dust concentration at 60 kyr BP.

The ice is intrinsically extremely transparent. The measured absorption length is 59 ± 3 meters, comparable with the quality of the ultrapure water used in the Irvine-Michigan-Brookhaven and Kamiokande proton-decay and neutrino experiments and more than twice as long as the best value reported for laboratory ice. Because of a residual density of air bubbles at these depths, the trajectories of photons in the medium are randomized. If the bubbles are assumed to be smooth and spherical, the average distance between collisions at a depth of 1 kilometer is about 25 centimeters. The measured inverse scattering length on bubbles decreases linearly with increasing depth in the volume of ice investigated.

    • The possible lateral-torsional instability of an I-beam with braced compression flange is studied. It is shown that taking the sagging prior to lateral-torsional buckling into account gives a finite critical moment. A solution for a uniform I-beam under uniform bending is presented. It is further shown that this solution can be approximated by a simple geometrical relation for the critical strain. This approximation is a lower bound also where the bending is causing inelastic deformations. For normal structures tension flange instability will not be a problem but it may be necessary to consider it for very high strength shallow beams or if plastic rotations occur in the sagging region.

    • The use of molecular genetics for introducing fluorescent molecules enables the use of donor-donor energy migration to determine intramolecular distances in a variety of proteins. This approach can be applied to examine the overall molecular dimensions of proteins and to investigate structural changes upon interactions with specific target molecules. In this report, the donor-donor energy migration method is demonstrated by experiments with the latent form of plasminogen activator inhibitor type 1. Based on the known x-ray structure of plasminogen activator inhibitor type 1, three positions forming the corners of a triangle were chosen. Double Cys substitution mutants (V106C-H185C, H185C-M266C, and M266C-V106C) and corresponding single substitution mutants (V106C, H185C, and M266C) were created and labeled with a sulfhydryl specific derivative of BODIPY (=the D molecule). The side lengths of this triangle were obtained from analyses of the experimental data. The analyses account for the local anisotropic order and rotational motions of the D molecules, as well as for the influence of a partial DD-labeling. The distances, as determined from x-ray diffraction, between the C(alpha)-atoms of the positions V106C-H185C, H185C-M266C, and M266C-V106C were 60.9, 30.8, and 55.1 A, respectively. These are in good agreement with the distances of 54 +/- 4, 38 +/- 3, and 55 +/- 3 A, as determined between the BODIPY groups attached via linkers to the same residues. Although the positions of the D-molecules and the C(alpha)-atoms physically cannot coincide, there is a reasonable agreement between the methods.

    • This study is part of a larger project aimed at establishing criteria to ensure the quality and safety-in-use of recycled and re-used plastics for food packaging. A major concern is that plastic materials can interact with chemicals to a greater or lesser extent. As a consequence, refillable containers may be contaminated by migration of harmful substances into the plastic material due to misuse by consumers before return. On the other hand, there is currently neither any specific national or EU regulation nor a standard test procedure available which could be applied by industry or enforcement laboratories to meet this situation appropriately. In this study, a relative simple inertness test which focuses on the interaction of PET bottle wall strips with a selection of model contaminants was investigated. The model contaminants were selected to include four different sets of chemical functional classes (set A, alcohol-type compounds; set B, ester/ketone type compounds; set C, hydrocarbon-type compounds; set D, chlorinated hydrocarbons). A simple gas chromatographic method using flame ionization detection was developed to allow quantification of the whole range of model contaminants. Linearity and reproducibility of the calibration curves for all contaminants were excellent, which indicates the validity and efficacy of the proposed methodology. The sorption experiments with PET bottle wall strips showed that significant amounts of chemicals can be absorbed into the plastic material if misused, thus establishing a remigration potential in the bottle material after refilling. © 1997 by John Wiley & Sons, Ltd.

    • PURPOSE: To establish the light and electron microscopic morphology of long-term full-thickness embryonic rabbit retinal transplants, with special attention paid to graft- host integration.METHODS: Eighteen rabbits received a complete embryonic neuroretina 19 days after conception. The transplants were positioned under the host retina, flat against the host retinal pigment epithelium with proper polarity, using a vitrectomy technique. After surviving 3 to 10 months, the transplants were examined by light and electron microscopy.RESULTS: The outer retina of the host had degenerated in all specimens. In 16 of the 18 eyes, well-laminated transplants with correct polarity, measuring up to 3.2 mm in length, were found. The transplants displayed long outer segments facing the host retinal pigment epithelium, and they were laminated to the level of the inner plexiform layer in which fusion with the host was often evident. Fusion was more prominent in the oldest transplants. Electron microscopy revealed bundles of neurites at different levels of maturation in close contact with Müller cell fimbriae at regular intervals along the graft-host border.CONCLUSIONS: Full-thickness embryonic rabbit retinal transplants positioned with correct polarity develop into large laminated retinas and survive without immunosuppression for at least 10 months. Host and graft adapt and almost reconstruct the normal retinal appearance. Ultrastructurally, well-developed photoreceptors and many normal synapse types are seen, and neuron sprouting is evident at the graft-host border.

    • Fifty-eight patients, 36 with essential tremor (ET) and 22 with Parkinson's disease (PD), received deep brain stimulation (DBS) in the thalamic ventral intermediate (Vim) nucleus. The mean follow-up was 17 months for ET and 21 months for PD patients. Stimulation parameters were adjusted as needed, at various intervals after surgery. Results were assessed using routine clinical evaluation and established outcome scales. All patients needed incremental increase in stimulation parameters at various intervals during the first 6-12 months after surgery. The mean voltage 1 week postoperatively was 1. 45 V in PD patients, and 1.37 V in ET patients. Twelve months later, the figures were 2.14 V in PD and 2.25 V in ET patients. At 1 year, the Essential Tremor Rating Scale (ETRS) improved from 54 to 28 (p < 0.0001). The motor part of the Unified Parkinson's Disease Rating Scale (UPDRS) improved from 37 to 26 (p < 0.01). Tremor items of the UPDRS improved more markedly (p < 0.0001). One week postoperatively 90% of PD, and 89% of ET patients were tremor free. One year later, 70% of PD and 60% of ET patients remained mostly tremor free. Upon switching off stimulation, there was a clear tendency for tremor rebound (p = 0.07) in the PD group, requiring continuous 24-hour stimulation in some patients. Permanent non-adjustable ataxia was induced by stimulation in 2 PD patients.
