  • Genomic and drug target evaluation of 90 cardiovascular proteins in 30,931 individuals.

    • Circulating proteins are vital in human health and disease and are frequently used as biomarkers for clinical decision-making or as targets for pharmacological intervention. Here, we map and replicate protein quantitative trait loci (pQTL) for 90 cardiovascular proteins in over 30,000 individuals, resulting in 451 pQTLs for 85 proteins. For each protein, we further perform pathway mapping to obtain trans-pQTL gene and regulatory designations. We substantiate these regulatory findings with orthogonal evidence for trans-pQTLs using mouse knockdown experiments (ABCA1 and TRIB1) and clinical trial results (chemokine receptors CCR2 and CCR5), with consistent regulation. Finally, we evaluate known drug targets, and suggest new target candidates or repositioning opportunities using Mendelian randomization. This identifies 11 proteins with causal evidence of involvement in human disease that have not previously been targeted, including EGF, IL-16, PAPPA, SPON1, F3, ADM, CASP-8, CHI3L1, CXCL16, GDF15 and MMP-12. Taken together, these findings demonstrate the utility of large-scale mapping of the genetics of the proteome and provide a resource for future precision studies of circulating proteins in human health.

  • Time without PSA recurrence after radical prostatectomy as a predictor of future biochemical recurrence, metastatic disease and prostate cancer death : a prospective Scandinavian cohort study

    • Objective: Although surveillance after radical prostatectomy routinely includes repeated prostate specific antigen (PSA)-testing for many years, biochemical recurrence often occurs without further clinical progression. We therefore hypothesised that follow-up can be shortened for many patients without increasing the risk of prostate cancer death. We investigated the long-term probabilities of PSA recurrence, metastases and prostate cancer death in patients without biochemical recurrence five and 10 years after radical prostatectomy.Design: Prospective cohort study. Stratification by Gleason score (<= 3+4=7or >= 4+3=7), pathological tumour stage (pT2 or >= pT3) and negative or positive surgical margins.Setting: Between 1989 and 1998, 14 urological centres in Scandinavia randomised patients to the Scandinavian Prostate Cancer Group study number 4 (SPCG-4) trial.ParticipationAll 306 patients from the SPCG-4 trial who underwent radical prostatectomy within 1year from inclusion were eligible. Four patients were excluded due to surgery-related death (n=1) or salvage radiotherapy or hormonal treatment within 6weeks from surgery (n=3).Primary outcome measures: Cumulative incidences and absolute differences in metastatic disease and prostate cancer death.Results: We analysed 302 patients with complete follow-up during a median of 24 years. Median preoperative PSA was 9.8ng/mL and median age was 65 years. For patients without biochemical recurrence 5 years after radical prostatectomy the 20-year probability of biochemical recurrence was 25% among men with Gleason score <= 3+4=7and 57% among men with Gleason score >= 4+3=7; the probabilities for metastases were 0.8% and 17%; and for prostate cancer death 0.8% and 12%, respectively. The long-term probabilities were higher for pT >= 3versus pT2 and for positive versus negative surgical margins. Limitations include small size of the cohort.Conclusion: Many patients with favourable histopathology without biochemical recurrence 5years after radical prostatectomy could stop follow-up earlier than 10 years after radical prostatectomy.

    • BACKGROUND: Previous population-based studies investigating the relationship between physical activity and the gut microbiota have relied on self-reported activity, prone to reporting bias. Here, we investigated the associations of accelerometer-based sedentary (SED), moderate-intensity (MPA), and vigorous-intensity (VPA) physical activity with the gut microbiota using cross-sectional data from the Swedish CArdioPulmonary bioImage Study.METHODS: In 8416 participants aged 50-65, time in SED, MPA, and VPA were estimated with hip-worn accelerometer. Gut microbiota was profiled using shotgun metagenomics of faecal samples. We applied multivariable regression models, adjusting for sociodemographic, lifestyle, and technical covariates, and accounted for multiple testing.FINDINGS: Overall, associations between time in SED and microbiota species abundance were in opposite direction to those for MPA or VPA. For example, MPA was associated with lower, while SED with higher abundance of Escherichia coli. MPA and VPA were associated with higher abundance of the butyrate-producers Faecalibacterium prausnitzii and Roseburia spp. We observed discrepancies between specific VPA and MPA associations, such as a positive association between MPA and Prevotella copri, while no association was detected for VPA. Additionally, SED, MPA and VPA were associated with the functional potential of the microbiome. For instance, MPA was associated with higher capacity for acetate synthesis and SED with lower carbohydrate degradation capacity.INTERPRETATION: Our findings suggest that sedentary and physical activity are associated with a similar set of gut microbiota species but in opposite directions. Furthermore, the intensity of physical activity may have specific effects on certain gut microbiota species.FUNDING: European Research Council, Swedish Heart-Lung Foundation, Swedish Research Council, Knut and Alice Wallenberg Foundation.

  • MIND THE GAP: THE INTERFACE BETWEEN AUTONOMOUS VEHICLES AND THE LOGISTICS SYSTEM

    • Purpose Using autonomous vehicles in goods transport has potential benefits. However, the transition to a future autonomous (and electric) freight system needs to take account of the interface between the vehicle and other parts of the logistics system in order to exploit potential benefits and overcome challenges related to the absence of a driver. The purpose of the research is to identify the prerequisites for achieving automated loading, unloading, related information handling and in-plant transport at these interfaces. Research Approach Cases studies consider senders and receivers of various products as well as organisations that have a role including real estate companies that develop warehouses and terminals. The case studies consider vehicle and handling equipment technology alongside the processes that may need to change if the efficiency gains of an autonomous vehicle are to be realised. Research questions are addressed at the logistics system level. The cases studies combine questions about logistics process and technology alongside those concerned with underlying business models. Findings and Originality Results presented are at the half way stage of a two-year project and will be based on the initial series of interviews for four case studies. Findings so far include: (1) lack of development in this topic compared with interest in autonomous and automated transport (2) importance of organisational ssues in the logistics chain e.g. contractual relationships (3) importance of taking a broad view of the companies that need to be involved (4) product type and characteristics are important but other factors may have more influence on the scope to develop initiatives in this area. Research Impact The research provides insights into the complexity of the interfaces in autonomous and automated transport and logistics systems. The empirical findings enrich the discussion of this field. Combining insights from logistics, technology, materials handling and business has demonstrated the benefits of an interdisciplinary approach. Practical Impact Autonomous transport systems may bring many commercial benefits. However, the scale of these benefits will be significantly diminished if questions about the interface points in the logistics system are not addressed. The outputs of the research will provide important insights for managers addressing these challenges.

In as-sintered materials, Y was present as yttrium aluminium garnet. The materials were exposed in air at 1500 °C for up to 250 h to study the effect of Y on oxidation behaviour. The oxides formed were investigated using scanning electron microscopy (SEM)-based techniques and X-ray diffraction. While the Y-free Mo(Si,Al)2 formed a scale consisting of Al2O3 and a small amount of mullite, the Y-containing samples formed oxides containing both yttrium silicate and larger fractions of mullite, in addition to Al2O3. Oxidation rate, scale spallation, as well as the evaporation of Mo, all increased with Y addition.

    • OBJECTIVES: The aim included investigation of the associations between sedentary (SED), low-intensity physical activity (LIPA), moderate-to-vigorous intensity PA (MVPA) and the prevalence of subclinical atherosclerosis in both coronaries and carotids and the estimated difference in prevalence by theoretical reallocation of time in different PA behaviours.DESIGN: Cross-sectional.SETTING: Multisite study at university hospitals.PARTICIPANTS: A total of 22 670 participants without cardiovascular disease (51% women, 57.4 years, SD 4.3) from the population-based Swedish CArdioPulmonary bioImage study were included. SED, LIPA and MVPA were assessed by hip-worn accelerometer.PRIMARY AND SECONDARY OUTCOMES: Any and significant subclinical coronary atherosclerosis (CA), Coronary Artery Calcium Score (CACS) and carotid atherosclerosis (CarA) were derived from imaging data from coronary CT angiography and carotid ultrasound.RESULTS: High daily SED (>70% ≈10.5 hours/day) associated with a higher OR 1.44 (95% CI 1.09 to 1.91), for significant CA, and with lower OR 0.77 (95% CI 0.63 to 0.95), for significant CarA. High LIPA (>55% ≈8 hours/day) associated with lower OR for significant CA 0.70 (95% CI 0.51 to 0.96), and CACS, 0.71 (95% CI 0.51 to 0.97), but with higher OR for CarA 1.41 (95% CI 1.12 to 1.76). MVPA above reference level, >2% ≈20 min/day, associated with lower OR for significant CA (OR range 0.61-0.67), CACS (OR range 0.71-0.75) and CarA (OR range 0.72-0.79). Theoretical replacement of 30 min of SED into an equal amount of MVPA associated with lower OR for significant CA, especially in participants with high SED 0.84 (95% CI 0.76 to 0.96) or low MVPA 0.51 (0.36 to 0.73).CONCLUSIONS: MVPA was associated with a lower risk for significant atherosclerosis in both coronaries and carotids, while the association varied in strength and direction for SED and LIPA, respectively. If causal, clinical implications include avoiding high levels of daily SED and low levels of MVPA to reduce the risk of developing significant subclinical atherosclerosis.

    • The present study aims to describe accelerometer-assessed physical activity (PA) patterns and fulfillment of PA recommendations in a large sample of middle-aged men and women, and to study differences between subgroups of socio-demographic, socio-economic, and lifestyle-related variables. A total of 27 890 (92.5% of total participants, 52% women, aged 50–64 years) middle-aged men and women with at least four days of valid hip-worn accelerometer data (Actigraph GT3X+, wGT3X+ and wGT3X-BT) from the Swedish CArdioPulmonary bioImage Study, SCAPIS, were included. In total, 54.5% of daily wear time was spent sedentary, 39.1% in low, 5.4% in moderate, and only 0.1% in vigorous PA. Male sex, higher education, low financial strain, born in Sweden, and sedentary/light working situation were related to higher sedentary time, but also higher levels of vigorous PA. High BMI and having multiple chronic diseases associated strongly with higher sedentary time and less time in all three PA intensities. All-year physically active commuters had an overall more active PA pattern. The proportion fulfilling current PA recommendations varied substantially (1.4% to 92.2%) depending on data handling procedures and definition used. Twenty-eight percent was defined as having an “at-risk” behavior, which included both high sedentary time and low vigorous PA. In this large population-based sample, a majority of time was spent sedentary and only a fraction in vigorous PA, with clinically important variations between subgroups. This study provides important reference material and emphasizes the importance of a comprehensive assessment of all aspects of the individual PA pattern in future research and clinical practice.

    • Autoimmune Addison's disease (AAD) is characterized by the autoimmune destruction of the adrenal cortex. Low prevalence and complex inheritance have long hindered successful genetic studies. We here report the first genome-wide association study on AAD, which identifies nine independent risk loci (P < 5 × 10-8). In addition to loci implicated in lymphocyte function and development shared with other autoimmune diseases such as HLA, BACH2, PTPN22 and CTLA4, we associate two protein-coding alterations in Autoimmune Regulator (AIRE) with AAD. The strongest, p.R471C (rs74203920, OR = 3.4 (2.7-4.3), P = 9.0 × 10-25) introduces an additional cysteine residue in the zinc-finger motif of the second PHD domain of the AIRE protein. This unbiased elucidation of the genetic contribution to development of AAD points to the importance of central immunological tolerance, and explains 35-41% of heritability (h2).

  • Glucagonlike Peptide-1 Receptor Imaging in Individuals with Type 2 Diabetes

    • The glucagonlike peptide-1 receptor (GLP1R) is a gut hormone receptor, intricately linked to regulation of blood glucose homeostasis via several mechanisms. It is an established and emergent drug target in metabolic disease. The PET radioligand 68Ga-DO3A-VS-exendin4 (68Ga-exendin4) has the potential to enable longitudinal studies of GLP1R in the human pancreas.Methods: 68Ga-exendin4 PET/CT examinations were performed on overweight-to-obese individuals with type 2 diabetes (n = 13) as part of a larger target engagement study (NCT03350191). A scanning protocol was developed to optimize reproducibility (target amount of 0.5 MBq/kg [corresponding to peptide amount of <0.2 µg/kg], blood sampling, and tracer stability assessment). The pancreas and abdominal organs were segmented, and binding was correlated with clinical parameters.Results: Uptake of 68Ga-exendin4 in the pancreas, but not in other abdominal tissues, was high but variable between individuals. There was no evidence of self-blocking of GLP1R by the tracer in this protocol, despite the high potency of exendin4. The results showed that a full dynamic scan can be simplified to a short static scan, potentially increasing throughput and reducing patient discomfort. The 68Ga-exendin4 concentration in the pancreas (i.e., GLP1R density) correlated inversely with the age of the individual and tended to correlate positively with body mass index. However, the total GLP1R content in the pancreas did not.Conclusion: In summary, we present an optimized and simplified 68Ga-exendin4 scanning protocol to enable reproducible imaging of GLP1R in the pancreas. 68Ga-exendin4 PET may enable quantification of longitudinal changes in pancreatic GLP1R during the development of type 2 diabetes, as well as target engagement studies of novel glucagonlike peptide-1 agonists.

    • Despite the importance of the glucagon receptor (GCGR) in disease and in pharmaceutical drug development, there is a lack of specific and sensitive biomarkers of its activation in humans. The PET radioligand Ga-68-DO3A-VS-Tuna-2 (Ga-68-Tuna-2) was developed to yield a noninvasive imaging marker for GCGR target distribution and drug target engagement in humans. Methods: The biodistribution and dosimetry of Ga-68-Tuna-2 was assessed by PET/CT in 13 individuals with type 2 diabetes as part of a clinical study assessing the occupancy of the dual GCGR/glucagon like peptide-1 receptor agonist SAR425899. Binding of Ga-68-Tuna-2 in liver and reference tissues was evaluated and correlated to biometrics (e.g., weight or body mass index) or other biomarkers (e.g., plasma glucagon levels). Results: Ga-68-Tuna-2 binding was seen primarily in the liver, which is in line with the strong expression of GCGR on hepatocytes. The kidneys demonstrated high excretion-related retention, whereas all other tissue demonstrated rapid washout. The SUV55 (min) (SUV during the last 10-min time frame, 50-60 min after administration) uptake endpoint was sensitive to endogenous levels of glucagon. Ga-68-Tuna-2 exhibited a safe dosimetry profile and no adverse events after intravenous administration. Conclusion: Ga-68-Tuna-2 can be used for safe and accurate assessment of the GCGR in human. It may serve as an important tool in understanding the in vivo pharmacology of novel drugs engaging the GCGR.

  • Receptor occupancy of dual glucagon-like peptide 1/glucagon receptor agonist SAR425899 in individuals with type 2 diabetes

    • Unimolecular dual agonists for the glucagon-like peptide 1 receptor (GLP1R) and glucagon receptor (GCGR) are emerging as a potential new class of important therapeutics in type 2 diabetes (T2D). Reliable and quantitative assessments of in vivo occupancy on each receptor would improve the understanding of the efficacy of this class of drugs. In this study we investigated the target occupancy of the dual agonist SAR425899 at the GLP1R in pancreas and GCGR in liver by Positron Emission Tomography/Computed Tomography (PET/CT). Patients with T2D were examined by [Ga-68]Ga-DO3A-Tuna-2 and [Ga-68]Ga-DO3A-Exendin4 by PET, to assess the GCGR in liver and GLP1R in pancreas, respectively. Follow up PET examinations were performed after 17 (GCGR) and 20 (GLP-1R) days of treatment with SAR425899, to assess the occupancy at each receptor. Six out of 13 included patients prematurely discontinued the study due to adverse events. SAR425899 at a dose of 0.2 mg daily demonstrated an average GCGR occupancy of 11.2 +/- 14.4% (SD) in N=5 patients and a GLP1R occupancy of 49.9 +/- 13.3%. Fasting Plasma Glucose levels (-3.30 +/- 1.14 mmol/L) and body weight (-3.87 +/- 0.87%) were lowered under treatment with SAR425899. In conclusion, SAR425899 demonstrated strong interactions at the GLP1R, but no clear occupancy at the GCGR. The study demonstrates that quantitative target engagement of dual agonists can be assessed by PET.

  • Markers of imminent myocardial infarction

    • Myocardial infarction is a leading cause of death globally but is notoriously difficult to predict. We aimed to identify biomarkers of an imminent first myocardial infarction and design relevant prediction models. Here, we constructed a new case–cohort consortium of 2,018 persons without prior cardiovascular disease from six European cohorts, among whom 420 developed a first myocardial infarction within 6 months after the baseline blood draw. We analyzed 817 proteins and 1,025 metabolites in biobanked blood and 16 clinical variables. Forty-eight proteins, 43 metabolites, age, sex and systolic blood pressure were associated with the risk of an imminent first myocardial infarction. Brain natriuretic peptide was most consistently associated with the risk of imminent myocardial infarction. Using clinically readily available variables, we devised a prediction model for an imminent first myocardial infarction for clinical use in the general population, with good discriminatory performance and potential for motivating primary prevention efforts.

  • Uncovering the electrochemical interface of low-index copper surfaces in deep groundwater environments

    • Using a combination of a sophisticated modeling protocol and well-established electrochemical techniques, we unravel the chemical composition of the low-index surfaces of copper in groundwater environments at different ion concentrations, pHs, and redox potentials. By carefully linking density functional theory (DFT) and cyclic voltammetry (CV), we are able to extract fundamental information on interfaces of broad significance. Herein, we focus on the case of groundwater found in deep geological environments of importance to the planned constructions of disposal repositories for spent nuclear fuel around the world. Within the error margins of DFT, we can assign adsorption structures and compositions to the current peaks of the CVs. It is found that among the groundwater ions of main interest (i.e. sulfide, bisulfide, sulfate, chloride and bicarbonate), sulfides (HS-, S2-) bind strongest to the surface, and are likely to dominate at the interfaces under the deep geological conditions relevant for repositories of spent nuclear fuel.

  • Neutron Reflectivity in Corrosion Research on Metals

    • Neutron reflectivity (NR) is potentially a powerful tool for characterizing chemical and morphological changes in thin films and at buried interfaces in corrosion science. While the scope of NR is limited by its inherent demands for low surface roughness and high sample planarity, these drawbacks are compensated for by the unique ability to detect light elements and distinguish between isotopes. Furthermore, the generally weak absorption of neutrons by matter allows the use of bulky sample environments and in situ experiments. In particular, the layer thickness range of 3-100 nm accessible by NR is appropriate for studying air-formed films and passive films, which are crucial for the ability of metallic materials to resist corrosion, as well as for investigating the interaction of metal surfaces with hydrogen and its compounds, e.g., water. Also, NR is suitable for studying early stages of oxide growth on metals at high temperature, including the transition from Cabrera-Mott-type films to Wagner-type growth. Here, we outline key characteristics of NR as applied to the study of corrosion of metals, exemplified by earlier work, and discuss perspectives for future work in the field. The aim of our work is to stimulate the application of the unique capabilities of NR to corrosion science.

    • The influence of alkali- and chlorine-containing compounds on the corrosion of superheater alloys has been studied extensively. The current paper instead investigates the corrosive effects of KCl and HCl under conditions relevant to waterwall conditions. A low-alloy (Fe-2.25Cr-1Mo) steel was exposed to KCl(s), 500 vppm HCl(g) and (KCl + HCl) in the presence of 5%O2 and 20% H2O at 400 °C. The results indicate that alloy chlorination by KCl occurs by an electrochemical process, involving cathodic formation of chemisorbed KOH on the scale surface and anodic formation of solid FeCl2 at the bottom of the scale. The process is accompanied by extensive cracking and delamination of the iron oxide scale, resulting in a complex, convoluted scale morphology. Adding 500 vppm HCl to the experimental environment (KCl + HCl) initially greatly accelerated the formation of FeCl2 at the scale/alloy interface. The accelerated alloy chlorination is attributed to HCl reacting with KOH at the scale surface, causing the cathodic process to be depolarized. A rapid slowing down of the rate of chlorination and corrosion in KCl + HCl environment was observed which was attributed to the electronically insulating nature of the FeCl2 layer which forms at the bottom of the scale, disconnecting the anodic and cathodic regions.

  • Exploring failure modes of alumina scales on FeCrAl and FeNiCrAl alloys in a nitriding environment

    • Two high-temperature FeCrAl and FeNiCrAl alloys were exposed in a strongly nitriding environment at 900 °C and the morphology of nitridation was studied. Quasi-in-situ experiments revealed that nitridation started at specific surface sites directly related to the alloy microstructure where the alumina scale was permeable to nitrogen. FeCrAl alloy grains with (112) orientation formed outward-growing alumina scales and were susceptible to nitridation. Outward-growing scales and substrate nitridation was also observed at chromium carbide precipitates in the FeNiCrAl alloy. Both alloys suffered nitridation at reactive element-rich (Y and Zr) inclusions larger than a certain critical size. The latter type of attack is caused by cracks and pores in the scale. The findings open new avenues of research for developing the next generation of high temperature alloys with superior properties.

    • Three FeCrAl alloys and two chromia-formers (a stainless steel, and a Ni-base alloy) have been exposed in four environments (dry air, air + 20% H2O, 20% H-2 + 20% H2O + Ar and 95% N-2 + 5% H-2) for 168 h at 800 degrees C. The corroded samples were investigated by SEM/EDS, XRD and gravimetry, and the formation of CrO2(OH)(2)(g) was measured as a function of time using a denuder technique. The Fe-base alloy formed a Cr-rich protective oxide scale in dry air and wet air but suffered break-away oxidation in 20% H-2 + 20% H2O + Ar. In contrast, the Ni-base alloy suffered extensive NiO formation and internal oxidation in dry air and wet air but formed a protective chromia scale in 20% H-2 + 20% H2O. All three FeCrAl alloys formed protective alumina scales in dry air, wet air and 20% H-2 + 20% H2O + Ar. The FeCrAl alloy Kanthal APMT was severely nitrided in the 95% N-2 + 5% H-2 environment due to defects in the oxide scale associated with RE-rich inclusions which allowed nitrogen to enter the alloy. In contrast, the two Cr-lean FeCrAl alloys Kanthal EF101 and Kanthal EF100 did not suffer nitridation at all.

  • Efficacy of an external chromia layer in reducing nitridation of high temperature alloys

    • Six high temperature alloys have been exposed in N2/H2 environments at 900 °C. In order to study the efficacy of a chromia barrier layer against nitrogen ingress, experiments were performed in two environments having the same N2/H2 ratio but slightly different water content, chromia formation being spontaneous in one case only. The samples were evaluated by SEM/STEM/EDX, XRD, gravimetry and GD-OES. The presence of an external chromia scale reduced nitridation of the alloy by 50–95%. Furthermore, in the presence of a continuous alumina layer no nitridation of the alloy was detected.

    • A new approach to reduce the chromium and aluminium concentrations in FeCrAl alloys without significantly impairing corrosion resistance is to alloy with 1-2 wt.% silicon. This paper investigates the "silicon effect" on oxidation by comparing the oxidation behavior and scale microstructure of two FeCrAl alloys, one alloyed with silicon and the other not, in dry and wet air at 600 degrees C and 800 degrees C. Both alloys formed thin protective oxide scales and the Cr-evaporation rates were small. In wet air at 800 degrees C the Si-alloyed FeCrAl formed an oxide scale containing mullite and tridymite together with alpha- and gamma-alumina. It is suggested that the reported improvement of the corrosion resistance of Al- and Cr-lean FeCrAl's by silicon alloying is caused by the appearance of Si-rich phases in the scale.

    • The localized corrosion of the two magnesium-aluminum alloys AM50 and AZ91 produced by a semisolid casting method was investigated under atmospheric corrosion conditions, in the presence of NaCl. Gravimetry confirmed the beneficial effect from ambient levels of carbon dioxide (CO2) in suppressing corrosion through the formation of magnesium hydroxy carbonates. Both 2D and 3D characterization revealed that ambient CO2 has a strong influence on the morphology and rate of localized corrosion. In CO2-free atmosphere, the alloys suffered localized corrosion resulting in clustered and interconnected cavities that extended deep into the material. In contrast, corrosion in the presence of ambient levels of CO2 (400 ppm) resulted in evenly distributed shallow corrosion sites. The results indicate that CO2 neutralizes the cathodic areas resulting in diminished cathodic activity and inhibiting NaCl(aq) droplet coalescence.

    • The surface film on pure magnesium and two aluminium-containing magnesium alloys was characterised after 96 h at 95% RH and 22 °C. The concentration of CO2 was carefully controlled to be either 0 or 400 ppm. The exposed samples were investigated using X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and electron microscopy. The results showed that when the alloys were exposed to the CO2-containing environment, aluminium cations (Al3+) was incorporated into a layered surface film comprising a partially “hydrated” MgO layer followed by Mg(OH)2, and magnesium hydroxy carbonates. The results indicated that aluminium-containing magnesium alloys exhibited considerably less localised corrosion in humid air than pure magnesium. Localised corrosion in the materials under investigation was attributed to film thinning by a dissolution/precipitation mechanism.

  • Automated GMP-Compliant Production of [Ga-68]Ga-DO3A-Tuna-2 for PET Microdosing Studies of the Glucagon Receptor in Humans

    • Introduction: [Ga-68]Ga-DO3A-VS-Cys(40)-Tuna-2 (previously published as [Ga-68]Ga-DO3A-VS-Cys(40)-S01-GCG) has shown high-affinity specific binding to the glucagon receptor (GCGR) in vitro and in vivo in rats and non-human primates in our previous studies, confirming the suitability of the tracer for drug development applications in humans. The manufacturing process of [Ga-68]Ga-DO3A-VS-Cys(40)-Tuna-2 was automated for clinical use to meet the radiation safety and good manufacturing practice (GMP) requirements.Methods:The automated synthesis platform (Modular-Lab PharmTrace, Eckert & Ziegler, Eurotope, Germany), disposable cassettes for(68)Ga-labeling, and pharmaceutical-grade(68)Ge/Ga-68 generator (GalliaPharm(R)) used in the study were purchased from Eckert & Ziegler. The parameters such as time, temperature, precursor concentration, radical scavenger, buffer concentration, and pH, as well as product purification step, were investigated and optimized. Process optimization was conducted with regard to product quality and quantity, as well as process reproducibility. The active pharmaceutical ingredient starting material DO3A-VS-Cys(40)-Tuna-2 (GMP-grade) was provided by Sanofi Aventis.Results:The reproducible and GMP-compliant automated production of [Ga-68]Ga-DO3A-VS-Cys(40)-Tuna-2 with on-line documentation was developed. The non-decay-corrected radiochemical yield was 45.2 +/- 2.5% (n= 3, process validation) at the end of the synthesis with a labeling synthesis duration of 38 min and a quality controlincluding release procedure of 20 min. The radiochemical purity of the product was 98.9 +/- 0.6% (n= 17) with the total amount of the peptide in the preparation of 48 +/- 2 mu g (n= 3, process validation). Radionuclidic purity, sterility, endotoxin content, residual solvent content, and sterile filter integrity tests met the acceptance criteria. The product was stable at ambient temperature for at least 2 h.Conclusion:The fully automated GMP-compliant manufacturing process was developed and thoroughly validated. The resulting [Ga-68]Ga-DO3A-VS-Cys(40)-Tuna-2 was used in a clinical study for accurate quantification of GCGR occupancy by a dual anti-diabetic drug in vivo in humans.

    • Most high-temperature processes require monitoring and controlling temperature, preferably with high precision and good lateral resolution. Here we evaluate the use of the technique commonly known as phosphor thermometry, which exploits the temperature dependent photoluminescence from an inorganic phosphor, for the determination of the temperature of a composite material consisting of the metallic alloy FeCrAl dispersed with phosphor particles of yttrium aluminum garnet (Y3Al5O12, YAG) doped with a small amount of luminescent Ce3+ ions (YAG:Ce3+). The results show that with some optimization and by changing the dopant ion, YAG based phosphor particles offer a unique opportunity to measure the surface temperature of metal alloys with high precision and high lateral resolution, all the way up to the maximum working temperature of alumina-forming high temperature alloys at ca. 1300 °C.
