    • We report the first measurement of the near oxygen K-edge auger spectrum of the glycine molecule. Our work employed an x-ray free electron laser as the photon source operated with input photon energies tunable between 527 and 547 eV. Complete electron spectra were recorded at each photon energy in the tuning range, revealing resonant and non-resonant auger structures. Finally ab initio theoretical predictions are compared with the measured above the edge auger spectrum and an assignment of auger decay channels is performed.

  • Gridmapping the Northern Plains of Mars: A New Overview of Recent Water‐ and Ice‐Related Landforms in Acidalia Planitia.

    • We used a grid‐mapping technique to analyze the distribution of 13 water‐ and ice‐related landforms in Acidalia Planitia as part of a joint effort to study the three main basins in the northern lowlands of Mars, that is, Acidalia, Utopia, and Arcadia Planitiae. The landforms were mapped at full Context Camera resolution along a 300‐km‐wide strip from 20°N to 84°N. We identified four landform assemblages: (1) Geologically recent polar cap (massive ice), which superposes the latitude‐dependent mantle (LDM) (LA1); (2) ice‐related landforms, such as LDM, textured terrain, small‐scale polygons, scalloped terrain, large‐scale viscous flow features, and gullies, which have an overlapping distribution (LA2); (3) surface features possibly related to water and subsurface sediment mobilization (LA3; kilometer‐scale polygons, large pitted mounds, small pitted mounds, thumbprint terrain); and (4) irregularly shaped pits with raised rims on equator‐facing slopes. Pits are likely the result of an energetic release of volatiles (H2O, CO2, and CH4), rather than impact‐, volcanism‐, or wind‐related processes. LDM occurs ubiquitously from 44°N to 78°N in Acidalia Planitia. Various observations suggest an origin of air fall deposition of LDM, which contains less ice in the uppermost tens of meters in Acidalia Planitia than in Arcadia and Utopia Planitiae. However, LDM may be thicker and more extended in the past in Acidalia Planitia. The transition between LDM‐free terrain and LDM is situated further north than in Utopia and Arcadia Planitiae, suggesting different past and/or present climatic conditions among the main basins in the northern lowlands.

    • A project of mapping ice‐related landforms was undertaken to understand the role of subsurface ice in the northern plains. This work is the first continuous regional mapping from CTX (ConTeXt Camera, 6 m/pixel; Malin et al., 2007) imagery in Arcadia Planitia along a strip 300 km across stretching from 30°N to 80°N centered on the 170°W line of longitude. The distribution and morphotypes of these landforms were used to understand the permafrost cryolithology. The mantled and textured signatures occur almost ubiquitously between 35°N and 78°N and have a positive spatial correlation with inferred ice stability based on thermal modeling, neutron spectroscopy, and radar data. The degradational features into the LDM (latitude‐dependent mantle) include pits, scallops, and 100‐m polygons and provide supporting evidence for subsurface ice and volatile loss between 35 and 70°N in Arcadia with the mantle between 70 and 78°N appearing much more intact. Pitted terrain appears to be much more pervasive in Arcadia than in Acidalia and Utopia suggesting that the Arcadia study area had more widespread near‐surface subsurface ice and thus was more susceptible to pitting or that the ice was less well buried by sediments. Correlations with ice stability models suggest that lack of pits north of 65–70°N could indicate a relatively young age (~1 Ma); however, this could also be explained through regional variations in degradation rates. The deposition of the LDM is consistent with an air fall hypothesis; however, there appears to be substantial evidence for fluvial processes in southern Arcadia with older, underlying processes being equally dominant with the LDM and degradation thereof in shaping the landscape.

    • Trees represent the largest terrestrial carbon sink and a renewable source of ligno-cellulose. There is significant scope for yield and quality improvement in these largely undomesticated species, and efforts to engineer elite varieties will benefit from improved understanding of the transcriptional network underlying cambial growth and wood formation. We generated high-spatial-resolution RNA sequencing data spanning the secondary phloem, vascular cambium, and wood-forming tissues of Populus tremula. The transcriptome comprised 28,294 expressed, annotated genes, 78 novel protein-coding genes, and 567 putative long intergenic noncoding RNAs. Most paralogs originating from the Salicaceae whole-genome duplication had diverged expression, with the exception of those highly expressed during secondary cell wall deposition. Coexpression network analyses revealed that regulation of the transcriptome underlying cambial growth and wood formation comprises numerous modules forming a continuum of active processes across the tissues. A comparative analysis revealed that a majority of these modules are conserved in Picea abies. The high spatial resolution of our data enabled identification of novel roles for characterized genes involved in xylan and cellulose biosynthesis, regulators of xylem vessel and fiber differentiation and lignification. An associated web resource (AspWood, http://aspwood.popgenie.org) provides interactive tools for exploring the expression profiles and coexpression network.

We generated RNA Sequencing transcriptome data for four mature, wild-growing aspens (Populus tremula) from high-spatial-resolution tangential cryosection series spanning the secondary phloem, vascular cambium, expanding and secondary cell wall forming xylem cells, cell death zone and the previous years annual ring. The transcriptome comprised 28,294 expressed, previously annotated protein-coding genes, 78 novel protein-coding genes and 567 long intergenic non-coding RNAs. Most paralogs originating from the Salicaceae whole genome duplication had diverged expression, with the notable exception of those with high expression during secondary cell wall deposition. We performed co-expression network analysis to identify central transcriptional modules and associated several of these with known biological processes. This revealed previously uncharacterized complexity underlying the regulation of cambial growth and wood formation, with modules forming a continuum of activated processes across the tissues. The high spatial resolution suggested novel roles for known genes involved in xylan and cellulose biosynthesis, regulators of xylem vessel and fiber differentiation and components of lignification. The associated web resource (AspWood, http://aspwood.popgenie.org) integrates the data within a set of interactive tools for exploring the co-expression network of cambial growth and wood formation.

  • Salmonid stocking in five North Atlantic jurisdictions: Identifying drivers and barriers to policy change

    • New knowledge challenges long-established practices of fish stocking and transfer because of increasing scientific consensus that the release of cultivated fish can pose risks to biodiversity; however, stocking can also improve fisheries, creating difficult decision trade-offs regarding its use. Accordingly, controversy persists about fish stocking and transfer. No studies, however, have embraced a multinational perspective to understand the important governance dimensions of the success and failure of salmonid stocking and transfer policies. The present study has analysed the historical development and contemporary governance of the stocking and transfer of native and non-native salmonids of the genera Salmo, Salvelinus, and Oncorhynchus in five legislative units around the North Atlantic Ocean: the Atlantic Provinces of Canada, France, Germany, Norway, and Sweden. The study is based on the analyses of published and unpublished literature, and a survey of experts. Current salmonid stocking policies and practices varied significantly among jurisdictions; the degree of policy change varied, from radical and rapid changes de jure and de facto in Atlantic Canada and Norway to incremental mostly de jure changes in France and Germany. Rapid policy change in Atlantic Canada, Norway, and partly in Sweden can be explained by the socio-political importance of salmonid fisheries, stocking regulations based on policy objectives to conserve wild Atlantic salmon (Salmo salar), well-documented examples of the harmful consequences of transfers of non-native species, and well-developed vertical governance linkages. The policy changes resemble that of the 'punctuated equilibrium policy framework'. By contrast, France and Germany place less socio-political emphasis on salmonids, have stocking regulations less directed at wild salmonids, more local-level decision making, more species-rich fish communities, and little evidence of adverse ecological impacts of the transfer and stocking of salmonids. This has led to small, incremental changes in stocking policy de facto that are reflective of the 'advocacy coalition policy framework'.

    • Service assurance for cloud applications is a challenging task and is an active area of research for academia and industry. One promising approach is to utilize machine learning for service quality prediction and fault detection so that suitable mitigation actions can be executed. In our previous work, we have shown how to predict service-level metrics in real-time just from operational data gathered at the server side. This gives the service provider early indications on whether the platform can support the current load demand. This paper provides the logical next step where we extend our work by proposing an automated detection and diagnostic capability for the performance faults manifesting themselves in cloud and datacenter environments. This is a crucial task to maintain the smooth operation of running services and minimizing downtime. We demonstrate the effectiveness of our approach which exploits the interpretative capabilities of Self- Organizing Maps (SOMs) to automatically detect and localize different performance faults for cloud services.

  • Coulomb explosion imaging of CH3I and CH2CII photodissociation dynamics

    • The photodissociation dynamics of CH3I and CH2CII at 272 nm were investigated by time-resolved Coulomb explosion imaging, with an intense non-resonant 815nmprobe pulse. Fragment ion momenta over a widem/z range were recorded simultaneously by coupling a velocity map imaging spectrometer with a pixel imaging mass spectrometry camera. For both molecules, delay-dependent pump-probe features were assigned to ultraviolet-induced carbon-iodine bond cleavage followed by Coulomb explosion. Multi-mass imaging also allowed the sequential cleavage of both carbon-halogen bonds in CH2ClI to be investigated. Furthermore, delay-dependent relative fragment momenta of a pair of ions were directly determined using recoil-frame covariance analysis. These results are complementary to conventional velocity map imaging experiments and demonstrate the application of time-resolved Coulomb explosion imaging to photoinduced real-time molecular motion.

    • Developmental exposure to excess glucocorticoids (GCs) has harmful neurodevelopmental effects, which include persistent alterations in the differentiation potential of embryonic neural stem cells (NSCs). The mechanisms, however, are largely unknown. Here, we investigated the effects of dexamethasone (Dex, a synthetic GC analog) by MeDIP-like genome-wide analysis of differentially methylated DNA regions (DMRs) in NSCs isolated from embryonic rat cortices. We found that Dex-induced genome-wide DNA hypomethylation in the NSCs in vitro. Similarly, in utero exposure to Dex resulted in global DNA hypomethylation in the cerebral cortex of 3-day-old mouse pups. Dex-exposed NSCs displayed stable changes in the expression of the DNA methyltransferase Dnmt3a, and Dkk1, an essential factor for neuronal differentiation. These alterations were dependent on Tet3 upregulation. In conclusion, we propose that GCs elicit strong and persistent effects on DNA methylation in NSCs with Tet3 playing an essential role in the regulation of Dnmt3a and Dkk1. Noteworthy is the occurrence of similar changes in Dnmt3a and Dkk1 gene expression after exposure to excess GC in vivo.

    • Due to its element and site specificity, inner-shell photoelectron spectroscopy is a widely used technique to probe the chemical structure of matter. Here, we show that time-resolved inner-shell photoelectron spectroscopy can be employed to observe ultrafast chemical reactions and the electronic response to the nuclear motion with high sensitivity. The ultraviolet dissociation of iodomethane (CH3I) is investigated by ionization above the iodine 4d edge, using time-resolved inner-shell photoelectron and photoion spectroscopy. The dynamics observed in the photoelectron spectra appear earlier and are faster than those seen in the iodine fragments. The experimental results are interpreted using crystal-field and spin-orbit configuration interaction calculations, and demonstrate that time-resolved inner-shell photoelectron spectroscopy is a powerful tool to directly track ultrafast structural and electronic transformations in gas-phase molecules.

    • The dynamics following laser-induced molecular photodissociation of gas-phase CH2BrI at 271.6 nm were investigated by time-resolved Coulomb-explosion imaging using intense near-IR femtosecond laser pulses. The observed delay-dependent photofragment momenta reveal that CH2BrI undergoes C-I cleavage, depositing 65.6% of the available energy into internal product states, and that absorption of a second UV photon breaks the C-Br bond of C(H)2Br. Simulations confirm that this mechanism is consistent with previous data recorded at 248 nm, demonstrating the sensitivity of Coulomb-explosion imaging as a real-time probe of chemical dynamics.

    • Results of angle-resolved electron spectroscopy of near-threshold photoionization of Ne atoms by combined femtosecond extreme ultraviolet and near infrared fields are presented. The dressed-electron spectra show an energetic distribution into so-called sidebands, being separated by the photon energy of the dressing laser. Surprisingly, for the low kinetic energy (few eV) sidebands, the photoelectron energy varies as a function of the emission angle. Such behavior has not yet been observed in sideband creation and has not been predicted in commonly used theoretical descriptions such as strong field approximation and soft photon approach. Describing the photoionization with a time-dependent Schrödinger equation allows a qualitative description of the observed effect, as well as the prediction of fine structure in the sideband distribution.

    • Predicting resource requirements for cloud services is critical for dimensioning, anomaly detection and service assurance. We demonstrate a system for real-time estimation of the needed amount of infrastructure resources, such as CPU and memory, for a given service. Statistical learning methods on server statistics and load parameters of the service are used for learning a resource prediction model. The model can be used as a guideline for service deployment and for real-time identification of resource bottlenecks.

  • Linking the Effect of Reservoir Injectivity and CO 2 Transport Logistics in the Nordic Region

    • We compare the cost for CO 2 -transport by ship with cost for pipeline transport in the Nordic region as a function of transport volume and distance. We also calculate the pipeline volumetric break-even point yielding the minimum CO 2 volume required from a specific site for pipeline to become the less costly transport option and finally, we investigate the effect injectivity may have on the choice of reservoir and transport mode. Most stationary CO 2 -emissions in the Nordic region originate from emission intensive industries such as steel, cement and chemical plants and refineries. Typically, their emissions are modest (less than 1 Mt per year) compared to large coal fired power plants, while distances to potential storage sites are considerable, often 300 km or more. Hence, build-up of clusters of emission sources and CO 2 -volumes is likely to take time and be costly. At the same time, many of the emission sources, both fossil based and biogenic, are located along the coast line. The results imply that due to modest CO 2 -volumes and relatively long transport distances CO 2 transport by ship is the least costly transportation option for most of the sources individually as well as for most of the potential cluster combinations during ramp-up of the CCS transport and storage infrastructure. It is furthermore shown that cost of ship transport increases modestly with increasing transport distance which, in combination with poor injectivity in reservoirs in the Baltic Sea, indicate that it may be less costly to transport the CO 2 captured from Finnish and Swedish sources located along the Baltic Sea a further 800-1300 km to the west by ship for storage in aquifers with higher injectivity in the Skagerrak region or in the North Sea.

    • Immunoglobulins are the effector molecules of the adaptive humoral immune system. In a genome-wide association study of 19,219 individuals, we found 38 new variants and replicated 5 known variants associating with IgA, IgG or IgM levels or with composite immunoglobulin traits, accounted for by 32 loci. Variants at these loci also affect the risk of autoimmune diseases and blood malignancies and influence blood cell development. Notable associations include a rare variant at RUNX3 decreasing IgA levels by shifting isoform proportions (rs188468174[C>T]: P = 8.3 × 10(-55), β = -0.90 s.d.), a rare in-frame deletion in FCGR2B abolishing IgG binding to the encoded receptor (p.Asn106del: P = 4.2 × 10(-8), β = 1.03 s.d.), four IGH locus variants influencing class switching, and ten new associations with the HLA region. Our results provide new insight into the regulation of humoral immunity.

  • Recommendations on CO2 transport solutions

    • The aim of this report is 1) to recommend transport solutions for CO2 sources in the Nordic region, here defined as the least costly transport mode for the selected CCS cases in NORDICCS and 2) to analyze the potential for establishment of CO2 clusters by means of a transportation network around the selected CCS cases in order to reduce the transportation cost. Comparing cost for pipeline transport with cost for ship transport, it is concluded that both for the majority of the selected cases as well as for most of the emission sources in the region, ship transport will be the least costly transport mode for each source individually. It is also concluded that ship transport is the most appropriate transport mode for most of the potential clusters in the region during a ramp-up phase. This is closely related to underutilization of pipelines and risk taking in connection with underutilized pipelines. For distances shorter than 100 km and volumes smaller than 1 Mtpa, e.g. corresponding to a typical collection system containing multiple coastal sources, it has been calculated that onshore pipeline in most cases will be the least costly transport solution. More generally, it can be stated that the break-even distance where ship transport becomes least costly than pipeline transport increases as the volume increases. Yet, it should be emphasized that discharge from a ship offshore and positioning of smaller ships during injection will need to be demonstrated. An obvious but still important conclusion is that constrained storage capability may have a profound impact on design and cost of a CO2 transport system. In fact, a poor storage capability in the reservoirs in the Baltic Sea may render ship transport to Gassum and Utsira a less costly transport and storage option than the reservoirs in the Baltic Sea. Finally, it is concluded that in the Nordic region, the Kattegat-Skagerrak area probably offers the best opportunities for a Nordic CCS system, possibly driven initially by CO2 EOR which potentially may require a start-up already in 2020.

    • Selection induced by human harvest can lead to different patterns of phenotypic change than selection induced by natural predation and could be a major driving force of evolution of wild populations. The vulnerability of individuals to angling depends on the individual decision to ingest the bait, possibly mediated by their neuroendocrine response towards the associated stimulus. To investigate the mechanisms behind individual vulnerability to angling, we conducted angling experiments in replicated ponds and quantified individual behavioral traits and neuroendocrine stress responsiveness in two salmonid species, rainbow trout (Oncorhynchus mykiss) and brown trout (Salmo trutta). We discovered a phenotypic syndrome in rainbow trout, but not in brown trout, where lower serotonergic and dopaminergic brain activity and cortisol levels (i.e., lower stress responsiveness) in response to a standardized experimental stressor were associated with higher activity, forming a proactive phenotype that showed increased vulnerability to angling. Our results show that angling targets the most stress-resilient and active phenotypes of rainbow trout, supporting the suggestion that fishing-induced phenotypic selection may lead to an increased representation of stress-responsive and low-activity phenotypes in harvested populations.

    • The mechanism of multicellular drug resistance, defined as the reduced efficacy of chemotherapeutic drugs in solid tumors is incompletely understood. Here we report that colon carcinoma cells cultured as 3D microtissues (spheroids) display dramatic increases in the expression of a subset of type I interferon-(IFN)-stimulated genes (ISGs). A similar gene signature was associated previously with resistance to radiation and chemotherapy, prompting us to examine the underlying biological mechanisms. Analysis of spheroids formed by different tumor cell lines and studies using knock-down of gene expression showed that cell crowding leads to the induction of IFN regulatory factor-9 (IRF9) which together with STAT2 and independently of IFNs, is necessary for ISG upregulation. Increased expression of IRF9 alone was sufficient to induce the ISG subset in monolayer cells and to confer increased resistance to clinically used cytotoxic drugs. Our data reveal a novel mechanism of regulation of a subset of ISGs, leading to drug resistance in solid tumors. What's new? Drug resistance remains a major challenge in the management of cancer patients. Using a 3D model of tumor cells the authors identify cell crowding and the interferon response as important mediators of drug resistance. They demonstrate that interferon regulatory factor 9 (IRF9) and a panel of interferon-stimulated genes are induced by cell crowding in this model. These results link unexpected new molecular mechanisms with the therapy resistance of solid tumors.

    • Objectives: To explore differences in body structure and function in systemic lupus erythematosus (SLE) patients and controls, with particular reference to joint hypermobility, and to evaluate the usefulness of the Brighton criteria for diagnosing joint hypermobility syndrome (JHS) in SLE. Method: Female SLE patients were, according to age group, consecutively invited to participate in the study. Controls were healthy females matched for age. All individuals were examined by a physician according to the Brighton criteria, and by an occupational therapist and a physiotherapist to obtain the Beighton scores, overall joint mobility, and manifestations in body structure and function.     • This work discusses the cost optimal capture rate of absorption based carbon capture processes by a combination of process simulations and cost-estimation. The influence of the quality of the CO2 source (quantity, continuity and CO2 concentration) and the availability of low cost heat on the absolute and specific capture cost are highlighted. The results stress that partial capture of CO2 could lower the specific capture cost (€/ton CO2) and that the relation between capital expenditure and lowered energy demand should be reconsidered for cases with access to low-cost heat.

    • In this work, the investment required to apply CO2capture to large-scale industrial sources is assessed and discussed in a case study of Sweden - a highly industrialized region with relative proximity to large and well-documented storage sites in the Norwegian North Sea. The Swedish process industry is characterized by a large share of biogenic emissions, and therefore has a considerable Bio-Energy with Carbon Capture and Storage (BECCS) potential. The capital cost for CO2capture is estimated for a standard MEA-based CO2absorption process. The CO2absorption process is applied to several industries – pulp and paper, oil and gas, steel, cement and chemical production – and dimensioned using process modeling. The equipment cost is subsequently estimated using a detailed individual factor estimation method. The capture costs are compared to estimates of the cost for transport and storage.

  • Rapid growth accelerates telomere attrition in a transgenic fish

    • Background: Individuals rarely grow as fast as their physiologies permit despite the fitness advantages of being large. One reason may be that rapid growth is costly, resulting for example in somatic damage. The chromosomal ends, the telomeres, are particularly vulnerable to such damage, and telomere attrition thus influences the rate of ageing. Here, we used a transgenic salmon model with an artificially increased growth rate to test the hypothesis that rapid growth is traded off against the ability to maintain somatic health, assessed as telomere attrition. Results: We found substantial telomere attrition in transgenic fish, while maternal half-sibs growing at a lower, wild-type rate seemed better able to maintain the length of their telomeres during the same time period. Conclusions: Our results are consistent with a trade-off between rapid growth and somatic (telomere) maintenance in growth-manipulated fish. Since telomere erosion reflects cellular ageing, our findings also support theories of ageing postulating that unrepaired somatic damage is associated with senescence.

  • Grid-based mapping: A method for rapidly determining the spatial distributions of small features over very large areas

    • The increased volume, spatial resolution, and areal coverage of high-resolution images of Mars over the past 15 years have led to an increased quantity and variety of small-scale landform identifications. Though many such landforms are too small to represent individually on regional-scale maps, determining their presence or absence across large areas helps form the observational basis for developing hypotheses on the geological nature and environmental history of a study area. The combination of improved spatial resolution and near-continuous coverage significantly increases the time required to analyse the data. This becomes problematic when attempting regional or global-scale studies of metre and decametre-scale landforms. Here, we describe an approach for mapping small features (from decimetre to kilometre scale) across large areas, formulated for a project to study the northern plains of Mars, and provide context on how this method was developed and how it can be implemented. Rather than. "mapping" with points and polygons, grid-based mapping uses a "tick box" approach to efficiently record the locations of specific landforms (we use an example suite of glacial landforms; including viscous flow features, the latitude dependant mantle and polygonised ground). A grid of squares (e.g. 20 km by 20 km) is created over the mapping area. Then the basemap data are systematically examined, grid-square by grid-square at full resolution, in order to identify the landforms while recording the presence or absence of selected landforms in each grid-square to determine spatial distributions. The result is a series of grids recording the distribution of all the mapped landforms across the study area. In some ways, these are equivalent to raster images, as they show a continuous distribution-field of the various landforms across a defined (rectangular, in most cases) area. When overlain on context maps, these form a coarse, digital landform map. We find that grid-based mapping provides an efficient solution to the problems of mapping small landforms over large areas, by providing a consistent and standardised approach to spatial data collection. The simplicity of the grid-based mapping approach makes it extremely scalable and workable for group efforts, requiring minimal user experience and producing consistent and repeatable results. The discrete nature of the datasets, simplicity of approach, and divisibility of tasks, open up the possibility for citizen science in which crowdsourcing large grid based mapping areas could be applied.

  • Constitutively Active SMAD2/3 Are Broad-Scope Potentiators of Transcription-Factor-Mediated Cellular Reprogramming

    • Reprogramming of cellular identity using exogenous expression of transcription factors (TFs) is a powerful and exciting tool for tissue engineering, disease modeling, and regenerative medicine. However, generation of desired cell types using this approach is often plagued by inefficiency, slow conversion, and an inability to produce mature functional cells. Here, we show that expression of constitutively active SMAD2/3 significantly improves the efficiency of induced pluripotent stem cell (iPSC) generation by the Yamanaka factors. Mechanistically, SMAD3 interacts with reprogramming factors and co-activators and co-occupies OCT4 target loci during reprogramming. Unexpectedly, active SMAD2/3 also markedly enhances three other TF-mediated direct reprogramming conversions, from B cells to macrophages, myoblasts to adipocytes, and human fibroblasts to neurons, highlighting broad and general roles for SMAD2/3 as cell-reprogramming potentiators. Our results suggest that co-expression of active SMAD2/3 could enhance multiple types of TF-based cell identity conversion and therefore be a powerful tool for cellular engineering. Ruetz et al. show that constitutively active SMAD2/3 has a surprising ability to boost the efficiency of cell reprogramming both to iPSCs and across lineages and may therefore be a general factor that can enhance transcription-factor-mediated reprogramming in a variety of contexts.

  • Nanoplasma Formation by High Intensity Hard X-rays

    • Using electron spectroscopy, we have investigated nanoplasma formation from noble gas clusters exposed to high-intensity hard-x-ray pulses at similar to 5 keV. Our experiment was carried out at the SPring-8 Angstrom Compact free electron LAser (SACLA) facility in Japan. Dedicated theoretical simulations were performed with the molecular dynamics tool XMDYN. We found that in this unprecedented wavelength regime nanoplasma formation is a highly indirect process. In the argon clusters investigated, nanoplasma is mainly formed through secondary electron cascading initiated by slow Auger electrons. Energy is distributed within the sample entirely through Auger processes and secondary electron cascading following photoabsorption, as in the hard x-ray regime there is no direct energy transfer from the field to the plasma. This plasma formation mechanism is specific to the hard-x-ray regime and may, thus, also be important for XFEL-based molecular imaging studies. In xenon clusters, photo-and Auger electrons contribute more significantly to the nanoplasma formation. Good agreement between experiment and simulations validates our modelling approach. This has wide-ranging implications for our ability to quantitatively predict the behavior of complex molecular systems irradiated by high-intensity hard x-rays.

    • Psoriatic skin differs distinctly from normal skin by its thickened epidermis. Most gene expression comparisons utilize full-thickness biopsies, with substantial amount of dermis. We assayed the transcriptomes of normal, lesional and non-lesional psoriatic epidermis, sampled as split-thickness skin grafts, with 5′-end RNA sequencing. We found that psoriatic epidermis contains more mRNA per total RNA than controls and took this into account in the bioinformatic analysis. The approach highlighted innate immunity-related pathways in psoriasis, including NOD-like receptor (NLR) signaling and inflammasome activation. We demonstrated that the NLR signaling genes NOD2, PYCARD, CARD6 and IFI16 are upregulated in psoriatic epidermis and strengthened these findings by protein expression. Interestingly, PYCARD, the key component of the inflammasome, showed an altered expression pattern in the lesional epidermis. The profiling of non-lesional skin highlighted PSORS4 and mitochondrially encoded transcripts, suggesting that their gene expression is altered already before the development of lesions. Our data suggest that all components needed for the active inflammasome are present in the keratinocytes of psoriatic skin. The characterization of inflammasome pathways provides further opportunities for therapy. Complementing previous transcriptome studies, our approach gives deeper insight into the gene regulation in psoriatic epidermis.

    • International standards are playing a key role in leading and shaping the smart manufacturing landscape. The integration and consistency among different standards is therefore essential to effectively support industrial automation evolution and to ensure their applicability. This paper focuses on the ISO18828 and ISO22400 standards, related to the production planning process and manufacturing, consequential phases in product lifecycle. In this paper the connections between the information related to production planning process (ISO18828) and the KPI main basic elements in manufacturing operation management (ISO22400) are analysed. The analysis aims at supporting the standards’ users, underlining the aspects that should be taken into account in order to consolidate and improve the considered lifecycle phases.

    • It is well known that fish can learn to avoid angling gear after experiencing a catch-and-release event, that is, after a private hooking experience. However, the possible importance of social information cues and their influence on an individual's vulnerability to angling remains largely unexplored, that is, social experience of a conspecific capture. The effects of private and social experience of hooking on the stress response of fish and subsequent catch rates were examined. Hatchery-reared rainbow trout, Oncorhynchus mykiss (Walbaum), were implanted with heart rate loggers and experimentally subjected to private or social experience of hooking. Private and social experience of angling induced an increased heart rate in fish compared with naive control fish. While private experience of hooking explained most of the reduced vulnerability to capture, no clear evidence was found that social experience of hooking affected angling vulnerability in fish that had never been hooked before. While both private and social experiences of angling constitute significant physiological stressors for rainbow trout, only the private experience reduces an individual's vulnerability to angling and in turn affecting population-level catchability.

    • Genome-wide association studies (GWAS) have transformed our understanding of susceptibility to multiple myeloma (MM), but much of the heritability remains unexplained. We report a new GWAS, a meta-analysis with previous GWAS and a replication series, totalling 9974 MM cases and 247,556 controls of European ancestry. Collectively, these data provide evidence for six new MM risk loci, bringing the total number to 23. Integration of information from gene expression, epigenetic profiling and in situ Hi-C data for the 23 risk loci implicate disruption of developmental transcriptional regulators as a basis of MM susceptibility, compatible with altered B-cell differentiation as a key mechanism. Dysregulation of autophagy/apoptosis and cell cycle signalling feature as recurrently perturbed pathways. Our findings provide further insight into the biological basis of MM.

    • Phenotypic scoring of wild animals under standardized laboratory conditions is important as it allows field ecologists and evolutionary biologists to understand the development and maintenance of interindividual differences in plastic traits (e.g. behaviour and physiology). However, captivity is associated with a shift from a natural familiar environment to an unfamiliar and artificial environment, which may affect estimates of plastic phenotypic traits. In this study, we tested how previous experience with laboratory environments and time spent in captivity affects behavioural (i.e. activity) and metabolic (i.e. standard and maximum metabolic rates) scoring of our model species, wild brown trout Salmo trutta. We found that individuals with previous experience of laboratory captivity (10.5 months earlier) showed higher activity in an open field test than individuals with no prior experience of laboratory captivity. Previous experience with captivity had no significant effect on metabolic rates. However, metabolic rates seemed to increase with increasing time spent in captivity prior to the collection of measurements. Although there are benefits of keeping wild animals in captivity prior to scoring, our results suggest that while allowing for sufficient acclimatization researchers should aim at minimizing time in captivity of wild animals to increase accuracy and ecological relevance of the scoring of plastic phenotypic traits.

    • The design of small and high-affinity lectin inhibitors remains a major challenge because the natural ligand binding sites of lectin are often shallow and have polar character. Herein we report that derivatizing galactose with un-natural structural elements that form multiple non-natural lectin-ligand interactions (orthogonal multipolar fluorine-amide, phenyl-arginine, sulfur-π, and halogen bond) can provide inhibitors with extraordinary affinity (low nanomolar) for the model lectin, galectin-3, which is more than five orders of magnitude higher than the parent galactose; moreover, is selective over other galectins.
