    • Blood low-density lipoprotein (LDL) cholesterol, high-density lipoprotein (HDL) cholesterol and triglyceride levels are risk factors for cardiovascular disease. To dissect the polygenic basis of these traits, we conducted genome-wide association screens in 19,840 individuals and replication in up to 20,623 individuals. We identified 30 distinct loci associated with lipoprotein concentrations (each with P < 5 x 10(-8)), including 11 loci that reached genome-wide significance for the first time. The 11 newly defined loci include common variants associated with LDL cholesterol near ABCG8, MAFB, HNF1A and TIMD4; with HDL cholesterol near ANGPTL4, FADS1-FADS2-FADS3, HNF4A, LCAT, PLTP and TTC39B; and with triglycerides near AMAC1L2, FADS1-FADS2-FADS3 and PLTP. The proportion of individuals exceeding clinical cut points for high LDL cholesterol, low HDL cholesterol and high triglycerides varied according to an allelic dosage score (P < 10(-15) for each trend). These results suggest that the cumulative effect of multiple common variants contributes to polygenic dyslipidemia.

    • High hypoxia-inducible factor-2 alpha (HIF-2 alpha) protein levels predict poor outcome in neuroblastoma, and hypoxia dedifferentiates cultured neuroblastoma cells toward a neural crest-like phenotype. Here, we identify HIF-2 alpha as a marker of normoxic neural crest-like neuroblastoma tumor-initiating/stem cells (TICs) isolated from patient bone marrows. Knockdown of HIF-2 alpha reduced VEGF expression and induced partial sympathetic neuronal differentiation when these TICs were grown in vitro under stem cell-promoting conditions. Xenograft tumors of HIF-2 alpha-silenced cells were widely necrotic, poorly vascularized, and resembled the bulk of tumor cells in clinical neuroblastomas by expressing additional sympathetic neuronal markers, whereas control tumors were immature, well-vascularized, and stroma-rich. Thus, HIF-2 alpha maintains an undifferentiated state of neuroblastoma TICs. Because low differentiation is associated with poor outcome and angiogenesis is crucial for tumor growth, HIF-2 alpha is an attractive target for neuroblastoma therapy.

    • The ATLAS TRT barrel is a tracking drift chamber using 52,544 individual tubular drift tubes. It is one part of the ATLAS Inner Detector, which consists of three sub-systems: the pixel detector spanning the radius range 4 to 20 cm, the semiconductor tracker (SCT) from 30 to 52 cm, and the transition radiation tracker ( TRT) from 56 to 108 cm. The TRT barrel covers the central pseudo-rapidity region |eta| < 1, while the TRT endcaps cover the forward and backward eta regions. These TRT systems provide a combination of continuous tracking with many measurements in individual drift tubes ( or straws) and of electron identification based on transition radiation from fibers or foils interleaved between the straws themselves. This paper describes the recently-completed construction of the TRT Barrel detector, including the quality control procedures used in the fabrication of the detector.

This paper describes the on and off detector electronics for the TRT as well as the TRT portion of the data acquisition (DAQ) system.

    • The ATLAS TRT end-cap is a tracking drift chamber using 245,760 individual tubular drift tubes. It is a part of the TRT tracker which consist of the barrel and two end-caps. The TRT end-caps cover the forward and backward pseudo-rapidity region 1.0 < vertical bar eta vertical bar < 2.0, while the TRT barrel central eta region vertical bar eta vertical bar < 1.0. The TRT system provides a combination of continuous tracking with many measurements in individual drift tubes ( or straws) and of electron identification based on transition radiation from fibers or foils interleaved between the straws themselves. Along with other two sub-systems, namely the Pixel detector and Semi Conductor Tracker (SCT), the TRT constitutes the ATLAS Inner Detector. This paper describes the recently completed and installed TRT end-cap detectors, their design, assembly, integration and the acceptance tests applied during the construction.

    • Ray tracing has been employed to investigate the absorption of light by smooth and random rough metal surfaces. For normally incident light the absorptance of the surface increases with surface roughness. However, for light incident at a tangent to the surface the absorptance-surface roughness relationship is more complex. For example, in certain cases the absorptance can rise, fall, and rise again as the surface roughness increases. In this paper this complex absorptance-roughness relationship is defined and explained. The wavelengths of the light chosen for this study correspond to the primary and secondary output wavelengths of Nd:YAG lasers.

    • The measurement of absorptance is important for the analysis and modelling of laser-material interactions. Unfortunately, most of the absorptance data presently available considers only polished pure metals rather than the commercially available (unpolished, oxidised) alloys which are actually being processed in manufacturing. This paper presents the results of absorptance measurements carried out at room temperature on as-received engineering grade non-ferrous metals (Al, Cu and Zn alloys). The measurements were made using an integrating sphere with an Nd:YLF laser at two wavelengths (1053nm and 527nm � which means that the results are also valid for Nd:YAG radiation at 1064nm and 532nm). The absorptance results obtained differ considerably from existing data for polished, pure metals and should help improve the accuracy of laser-material interaction models. Some clear trends were identified. For all 22 cases studied the absorptance was higher than for ideal pure, polished metals. For all Al- and Cu-samples the absorptance was higher for the green than it was for the infrared wavelength, while for all Zn-coatings this trend was reversed. No clear correlation between absorptance and surface roughness was found at low roughness values (Sa 0.15 � 0.60) but one, rougher set of samples (Sa 2.34), indicated a roughness-absorptance correlation at higher roughness levels.

    • In LaserMaterial Processing, an understanding of the fundamental absorption mechanisms plays a vital role in determining the optimum processing parameters and conditions. The absorptance, which characterizes how much a material absorbs, can alter during a processing application, as a result of melting, boiling, structural changes of the surface, oxidation, plasma formation, etc. To gain knowledge of these complex processes, a combination of experimental as well as of theoretical work is required. In this paper, an overview of the most common experimental methods and techniques to measure laser absorption in opaque solids, such as metals, will be presented.

    • The absorptance of real metallic engineering surfaces used in Laser Material Processing depends strongly upon the particular surface conditions, including the degree of roughness and oxidation. In this paper the influence of surface topography on Nd:YAG/Nd:YLF laser absorptance has been studied, both experimentally and theoretically, on a set of randomly selected as-received metallic engineering surfaces. The surfaces were sputter-coated with a thin layer of gold to eliminate differences in surface chemistry and oxidation. The absorptance was measured using integrating sphere reflectometry and compared to calculations from 3D ray-tracing simulations of AFM-scanned images of the surfaces. The results have increased the understanding of how surface roughness influences the laser-workpiece coupling efficiency.

The measurements were made using an integrating sphere with an Nd:YLF laser at two wavelengths (1053 and 527 nm, which means that the results are also valid for Nd:YAG radiation at 1064 and 532 nm). The absorptance results obtained differ considerably from existing data for polished, pure metals and should help improve the accuracy of laser-material interaction models. Some clear trends were identified; for all materials studied, the absorptance was considerably higher than the previously published values for the relevant pure metals with polished surfaces. For all 15 samples the absorptance was higher for the green than for the infrared wavelength. No clear trend correlating the absorptance with the roughness was found for mild steel in the roughness range Sa 0.4-5.6 μm. A correlation between absorptance and roughness was noted for stainless steel for Sa values above 1.5 μm.

    • Achieving asthma control remains an elusive goat for the majority of patients worldwide. Ensuring a correct diagnosis of asthma is the first step in assessing poor symptom control; this requires returning to the basics of history taking and physical examination, in conjunction with lung function measurement when appropriate. A number of factors may contribute to suboptimal asthma control. Concomitant rhinitis, a common co-pathology and contributor to poor control, can often be identified by asking a simple question. Smoking too has been identified as a cause of poor asthma control. Practical barriers such as poor inhaler technique must be addressed. An appreciation of patients' views and concerns about maintenance asthma therapy can help guide discussion to address perceptual barriers to taking maintenance therapy (doubts about personal necessity and concerns about potential adverse effects). Further study into, and a greater consideration of, factors and patient characteristics that could predict individual responses to asthma therapies are needed. Finally, more clinical trials that enrol patient populations reflecting the real world diversity of patients seen in clinical. practice, including wide age ranges, presence of comorbidities, current smoking, and differing ethnic origins, will contribute to better individual patient management.     • Differential quantification of proteins and peptides by LC-MS is a promising method to acquire knowledge about biological processes, and for finding drug targets and biomarkers. However, differential protein analysis using LC-MS has been held back by the lack of suitable software tools. Large amounts of experimental data are easily generated in protein and peptide profiling experiments, but data analysis is time-consuming and labor-intensive. Here, we present a fully automated method for scanning LC-MS/MS data for biologically significant peptides and proteins, including support for interactive confirmation and further profiling. By studying peptide mixtures of known composition, we demonstrate that peptides present in different amounts in different groups of samples can be automatically screened for using statistical tests. A linear response can be obtained over almost 3 orders of magnitude, facilitating further profiling of peptides and proteins of interest. Furthermore, we apply the method to study the changes of endogenous peptide levels in mouse brain striatum after administration of reserpine, a classical model drug for inducing Parkinson disease symptoms.

    • Tissue constructs for cartilage with native mechanical properties have not been described to date. To address this need the bacterial cellulose (BC) secreted by Gluconacetobacter xylinus (= Acetobacter xylinum) was explored as a novel scaffold material due to its unusual material properties and degradability. Native and chemically modified BC materials were evaluated using bovine chondrocytes. The results indicate that unmodified BC supports chondrocyte proliferation at levels of approximately 50% of the collagen type II substrate while providing significant advantages in terms of mechanical properties. Compared to tissue culture plastic and calcium alginate, unmodified BC showed significantly higher levels of chondrocyte growth. Chemical sulfation and phosphorylation of the BC, performed to mimic the glucosaminoglycans of native cartilage, did not enhance chondrocyte growth while the porosity of the material did affect chondrocyte viability. The BC did not induce significant activation of proinflammatory cytokine production during in vitro macrophage screening. Hence, unmodified BC was further explored using human chondrocytes. TEM analysis and RNA expression of the collagen II from human chondrocytes indicated that unmodified BC supports proliferation of chondrocytes. In addition, ingrowth of chondrocytes into the scaffold was verified by TEM. The results suggest the potential for this biomaterial as a scaffold for tissue engineering of cartilage
