    • The primary scientific goal of the GRIPS mission is to revolutionize our understanding of the early universe using gamma-ray bursts. We propose a new generation gamma-ray observatory capable of unprecedented spectroscopy over a wide range of gamma-ray energies (200 keV-50 MeV) and of polarimetry (200-1000 keV). The gamma-ray sensitivity to nuclear absorption features enables the measurement of column densities as high as 10(28)cm (-aEuro parts per thousand 2). Secondary goals achievable by this mission include direct measurements of all types of supernova interiors through gamma-rays from radioactive decays, nuclear astrophysics with massive stars and novae, and studies of particle acceleration near compact stars, interstellar shocks, and clusters of galaxies.

  • Properties of the 7He ground state from 8He neutron knockout

    • The unbound nucleus 7He, produced in neutron-knockout reactions with a 240 MeV/u 8He beam in a liquid-hydrogen target, has been studied in an experiment at the ALADIN-LAND setup at GSI. From an R-matrix analysis the resonance parameters for 7He as well as the spectroscopic factor for the 6He(0+) + n configuration in its ground-state have been obtained. The spectroscopic factor is 0.61 confirming that 7He is not a pure single-particle state. An analysis of 5He data from neutron-knockout reactions of 6He in a carbon target reveals the presence of an s-wave component at low energies in the α+n relative energy spectrum. A possible low-lying exited state in 7He observed in neutron knockout data from 8He in a carbon target and tentatively interpreted as a Iπ=1/2− state, could not be observed in the present experiment. Possible explanations of the shape difference between the 7He resonance obtained in the two knockout reactions are discussed in terms of target-dependence or different reaction mechanisms at relativistic energies.

  • The cantharelloid clade: dealing with incongruent gene trees and phylogenetic reconstruction methods

    • We reassessed the circumscription of the cantharelloid clade and identified monophyletic groups by using nLSU, nSSU, mtSSU and RPB2 sequence data. Results agreed with earlier studies that placed the genera Cantharellus, Craterellus, Hydnum, Clavulina, Membranomyces, Multiclavula, Sistotrema, Botryobasidium and the family Ceratobasidiaceae in that clade. Phylogenetic analyses support monophyly of all genera except Sistotrema, which was highly polyphyletic. Strongly supported monophyletic groups were: (i) Cantharellus-Craterellus, Hydnum, and the Sistotrema confluens group; (ii) Clavulina-Membranomyces and the S. brinkmannii-oblongisporum group, with Multiclavula being possibly sister of that clade; (iii) the Sistotrema eximum-octosporum group; (iv) Sistotrema adnatum and S. coronilla. Positions of Sistotrema raduloides and S. athelioides were unresolved, as were basal relationships. Botryobasidium was well supported as the sister taxon of all the above taxa, while Ceratobasidiaceae was the most basal lineage. The relationship between Tulasnella and members of the cantharelloid clade will require further scrutiny, although there is cumulative evidence that they are probably sister groups. The rates of molecular evolution of both the large and small nuclear ribosomal RNA genes (nuc-rDNA) are much higher in Cantharellus, Craterellus and Tulasnella than in the other cantharelloid taxa, and analyses of nuc-rDNA sequences strongly placed Tulasnella close to Cantharellus-Craterellus. In contrast analyses with RPB2 and mtSSU sequences placed Tulasnella at the base of the cantharelloid clade. Our attempt to reconstruct a "supertree" from tree topologies resulting from separate analyses that avoided phylogenetic reconstruction problems associated with missing data and/or unalignable sequences proved unsuccessful.

    • We compared combination fibrinolytic plus glycoprotein IIb/IIIa inhibitor therapy with stand-alone fibrinolysis with respect to speed and stability of reperfusion in patients who had acute ST-segment elevation myocardial infarction; data were obtained from 654 patients in 4 trials (Integrilin to Manage Platelet Aggregation to Combat Thrombosis in Acute Myocardial Infarction, Platelet Aggregation Receptor Antagonist Dose Investigation and Reperfusion Gain in Myocardial Infarction, Integrilin and Tenecteplase in Acute Myocardial Infarction, and the Fifth Global Use of Strategies to Open Occluded Coronary Arteries) that compared thrombolytics plus lamifiban, eptifibatide, or abciximab with standard thrombolysis. We found significantly faster and more stable ST-segment recovery with combination therapy starting at 60 minutes (56.7% vs 48.0% with >/=50% ST-segment resolution, p = 0.03) and sustained over 180 minutes after drug administration; this transient benefit may suggest a time frame when more optimal percutaneous coronary intervention can be performed.

  • Nanoparticles of calcium hydroxide for wood deacidification: Decreasing the emissions of organic acid vapors in church organ environments

    • Acetic and formic acid vapors emitted from woodwork in historical organs are very important corrosive agents for lead pipes. These acids are slowly released from the wood both during playing and when the pipes are silent. To inhibit this emission process. the wood surface can be modified, by creating a protective layer with alkaline features. However, a coating of wood is not recommended since this could modify the appearance and create a layer not perfectly compatible with the substrate. For this reason, we propose to use some innovative nanotechnology that has been successfully applied for the deacidification of wood samples coming from the Vasa shipwreck. Application of calcium (or magnesium) hydroxide nanoparticles, with sizes ranging from 30-150 nm, allowed a homogeneous distribution of particles through the surface layer of wood simply by soaking (or spraying) it in a alcohohc (or mixed with less polar solvents) dispersion of nanoparticies. Nanoparticles do not modify the wood appearance and distribute randomly within the first layers of wood. The small size of particles accounts for the high reactivity with CO2 from the air, to give the alkaline reserve of carbonates that provide high efficacy in the neutralization of gaseous acids. The emission of volatile organic compounds (VOC) from the treated wood was determined by using an emission test cell, Field and Laboratory Emission Cell (FLEC). The results show that the emissions of acetic acid vapor from nanoparticles treated wood was very low (

  • An academic ECG core lab perspective of the FDA initiative for digital ECG capture and data management in large-scale clinical trials

    • Maximal utility of accessible data is attractive to all partners in clinical research, whether it directly improves patient care or more accurately allows identification of the safety and efficacy of a new drug or procedure. The Food and Drug Administration (FDA) has presented a guideline draft addressing digitization of electrocardiogram (ECG) data in clinical trials to improve the standards for collection, analysis, and storage of safety information on new medical therapies. This FDA initiative has led to discussions and collaboration among the FDA, the pharmaceutical industry, the electrocardiographj, manufacturers, and the academic as well as the nonacademic EGG core labs. In this article, we present a broad-based viewpoint from two groups of academic EGG core labs, the Alliance of Academic EGG Core Labs and the Virtual Electronic EGG Corelab International Consortium. We have chosen to widen the perspective from using digitized EGG data in safety trials only, as addressed by the FDA guideline draft, to a discussion on the possibilities and the potential problems when using digitized EGG data also in large clinical trials focusing on efficacy measurements. We conclude that the benefit of digital data mining is probably well worth an initial incremental effort and expense.

  • Fabrication of degradable polymer scaffolds to direct the integration and differentiation of retinal progenitors

    • Retinal progenitor cells (RPCs) are self-renewing cells capable of differentiating into the different retinal cell types including photoreceptors, and they have shown promise as a source of replacement cells in experimental models of retinal degeneration. We hypothesized that a biodegradable polymer scaffold could deliver these cells to the subretinal space in a more organized manner than bolus injections, while also providing the graft with laminar organization and structural guidance channels. We fabricated highly porous scaffolds from blends of poly(L-lactic acid) and poly(lactic-co-glycolic acid) using a variety of techniques to produce pores oriented normal to the plane of the scaffold. RPCs were seeded on the polymer scaffolds and cultured for 14 days. Seeded scaffolds were then either fixed for characterization or used in an explant or in vivo rat model. The, scaffolds were fully covered by RPCs in 3 days. Attachment of RPCs to the polymer scaffold was associated with down-regulation of immature markers and up-regulation of markers of differentiation. This suggests that the scaffold may promote differentiation of RPCs. The seeded cells elaborated cellular processes and aligned in the scaffold in conjunction with degenerating retinal explants. The cells also exhibited morphologies consistent with photoreceptors including a high degree of polarization of the cells. This data suggests that the scaffold may be a means to assist in the promotion of photoreceptor phenotypes. Implantation of the seeded scaffold into the rat eye is associated with increased RPC survival. Taken together, these data suggest that these polymer scaffolds provide a useful means for delivering RPCs to the subretinal space and may assist in the formation of retinal cell phenotypes, although it is clear that more cues are needed to direct the differentiation of RPCs into functional photoreceptors.
