  • Antimicrobial resistance (AMR) and molecular characterization of Neisseria gonorrhoeae in Ghana, 2012-2015

    • Neisseria gonorrhoeae antimicrobial resistance (AMR) surveillance is essential for tracking the emergence and spread of AMR strains in local, national and international populations. This is crucial for developing or refining treatment guidelines. N. gonorrhoeae multiantigen sequence typing (NG-MAST) is beneficial for describing the molecular epidemiology of gonococci at national and international levels. Elucidation of AMR determinants to β-lactam drugs, is a means of monitoring the development of resistance. In Ghana, little is known about the current gonococcal AMR prevalence and no characterization of gonococcal isolates has been previously performed. In this study, gonococcal isolates (n = 44) collected from five health facilities in Ghana from 2012 to 2015, were examined using AMR testing, NG-MAST and sequencing of penA. High rates of resistance were identified to tetracycline (100%), benzylpenicillin (90.9%), and ciprofloxacin (81.8%). One isolate had a high cefixime MIC (0.75 μg/ml). Twenty-eight NG-MAST sequence types (STs) were identified, seventeen of which were novel. The isolate with the high cefixime MIC contained a mosaic penA-34 allele and belonged to NG-MAST ST1407, an internationally spreading multidrug-resistant clone that has accounted for most cefixime resistance in many countries. In conclusion, AMR testing, NG-MAST, and sequencing of the AMR determinant penA, revealed high rates of resistance to tetracycline, benzylpenicillin, and ciprofloxacin; as well as a highly diverse population of N. gonorrhoeae in Ghana. It is imperative to continue with enhanced AMR surveillance and to understand the molecular epidemiology of gonococcal strains circulating in Ghana and other African countries.

  • Observation of aerodynamic instability in the flow of a particle stream in a dilute gas
  • 2019

    • Forming macroscopic solid bodies in circumstellar discs requires local dust concentration levels significantly higher than the mean. Interactions of the dust particles with the gas must serve to augment local particle densities, and facilitate growth past barriers in the metre size range. Amongst a number of mechanisms that can amplify the local density of solids, aerodynamic streaming instability (SI) is one of the most promising. This work tests the physical assumptions of models that lead to SI in protoplanetary discs (PPDs). We conduct laboratory experiments in which we track the three-dimensional motion of spherical solid particles fluidised in a low-pressure, laminar, incompressible, gas stream. The particle sizes span the Stokes-Epstein drag regime transition and the overall dust-to-gas mass density ratio, is close to unity. A recently published study establishes the similarity of the laboratory flow to a simplified PPD model flow. We study velocity statistics and perform time-series analysis of the advected flow to obtain experimental results suggesting an instability due to particle-gas interaction: (i) there exist variations in particle concentration in the direction of the mean relative motion between the gas and the particles, that is the direction of the mean drag forces; (ii) the particles have a tendency to catch up to one another when they are in proximity; (iii) particle clumping occurs on very small scales, which implies local enhancements above the background by factors of several tens; (iv) the presence of these density enhancements occurs for a mean approaching or greater than 1; (v) we find evidence for collective particle drag reduction when the local particle number density becomes high and when the background gas pressure is high so that the drag is in the continuum regime. The experiments presented here are precedent-setting for observing SI under controlled conditions and may lead to a deeper understanding of how it operates in nature.

  • No Clear, Direct Evidence for Multiple Protoplanets Orbiting LkCa 15 : LkCa 15 bcd are Likely Inner Disk Signals
  • 2019
  • In: Astrophysical Journal Letters. - : American Astronomical Society. - 2041-8205 .- 2041-8213. ; 877:1
  • Journal article (peer-reviewed)abstract
    • Two studies utilizing sparse aperture-masking (SAM) interferometry and H-alpha differential imaging have reported multiple Jovian companions around the young solar-mass star, LkCa 15 (LkCa 15 bcd): the first claimed direct detection of infant, newly formed planets (protoplanets). We present new near-infrared direct imaging/spectroscopy from the Subaru Coronagraphic Extreme Adaptive Optics (SCExAO) system coupled with Coronagraphic High Angular Resolution Imaging Spectrograph (CHARIS) integral field spectrograph and multi-epoch thermal infrared imaging from Keck/NIRC2 of LkCa 15 at high Strehl ratios. These data provide the first direct imaging look at the same wavelengths and in the same locations where previous studies identified the LkCa 15 protoplanets, and thus offer the first decisive test of their existence. The data do not reveal these planets. Instead, we resolve extended emission tracing a dust disk with a brightness and location comparable to that claimed for LkCa 15 bcd. Forward-models attributing this signal to orbiting planets are inconsistent with the combined SCExAO/CHARIS and Keck/NIRC2 data. An inner disk provides a more compelling explanation for the SAM detections and perhaps also the claimed H-alpha detection of LkCa 15 b. We conclude that there is currently no clear, direct evidence for multiple protoplanets orbiting LkCa 15, although the system likely contains at least one unseen Jovian companion. To identify Jovian companions around LkCa 15 from future observations, the inner disk should be detected and its effect modeled, removed, and shown to be distinguishable from planets. Protoplanet candidates identified from similar systems should likewise be clearly distinguished from disk emission through modeling.

  • Designing for Digital Playing Out
  • 2019
  • In: CHI 2019. - New York, NY, USA : ASSOC COMPUTING MACHINERY.
  • Conference paper (peer-reviewed)abstract
    • We report on a design-led study in the UK that aimed to understand barriers to children (aged 5 to 14 years) 'playing out' in their neighbourhood and explore the potential of the Internet of Things (IoT) for supporting children's free play that extends outdoors. The study forms a design ethnography, combining observational fieldwork with design prototyping and co-creative activities across four linked workshops, where we used BBC micro:bit devices to co-create new IoT designs with the participating children. Our collective account contributes new insights about the physical and interactive features of micro:bits that shaped play, gameplay, and social interaction in the workshops, illuminating an emerging design space for supporting 'digital playing out' that is grounded in empirical instances. We highlight opportunities for designing for digital playing out in ways that promote social negotiation, supports varying participation, allows for integrating cultural influences, and accounts for the weaving together of placemaking and play.
