    • Background Interleukin (IL)-15 levels are increased in serum, synovium and bone marrow of patients with rheumatoid arthritis (RA). IL-15 influences both the innate and the adaptive immune response; its major role is activation and proliferation of T cells. There are also emerging data that IL-15 affects osteoclastogenesis. The authors investigated the association of genetic variants in IL15 with the rate of joint destruction in RA. Method 1418 patients with 4885 x-ray sets of both hands and feet of four independent data sets were studied. First, explorative analyses were performed on 600 patients with early RA enrolled in the Leiden Early Arthritis Clinic. Twenty-five single-nucleotide polymorphisms (SNPs) tagging IL-15 were tested. Second, SNPs with significant associations in the explorative phase were genotyped in data sets from Groningen, Sheffield and Lund. In each data set, the relative increase of the progression rate per year in the presence of a genotype was assessed. Subsequently, data were summarised in an inverse weighting meta-analysis. Results Five SNPs were significantly associated with rate of joint destruction in phase 1 and typed in the other data sets. Patients homozygous for rs7667746, rs7665842, rs2322182, rs6821171 and rs4371699 had respectively 0.94-, 1.04-, 1.09-, 1.09- and 1.09- fold rate of joint destruction compared to other patients (p = 4.0x10(-6), p = 3.8x10(-4), p = 5.0x10(-3), p = 5.0x10(-3) and p = 9.4x10(-3)). Discussion Independent replication was not obtained, possibly due to insufficient power. Meta-analyses of all data sets combined resulted in significant results for four SNPs (rs7667746, p < 0.001; rs7665842, p < 0.001; rs4371699, p = 0.01; rs6821171, p = 0.01). These SNPs were also significant after correction for multiple testing. Conclusion Genetic variants in IL-15 are associated with progression of joint destruction in RA.

  • Mapping the 12CO J = 1-0 and J = 2-1 emission in AGB and early post-AGB circumstellar envelopes. I. The COSAS program, first sample

    • We present COSAS (CO Survey of late AGB Stars), a project to map and analyze the 12CO J = 1−0 and J = 2−1 line emission in a representative sample of circumstellar envelopes around AGB and post-AGB stars. The survey was undertaken with the aim of investigating small- and large-scale morphological and kinematical properties of the molecular environment surrounding stars in the late AGB and early post-AGB phases. For this, COSAS combines the high sensitivity and spatial resolving power of the IRAM Plateau de Bure interferometer with the better capability of the IRAM 30 m telescope to map extended emission. The global sample encompasses 45 stars selected to span a range in chemical type, variability type, evolutionary state, and initial mass. COSAS provides means to quantify variations in the mass-loss rates, assess morphological and kinematical features, and to investigate the appearance of fast aspherical winds in the early post-AGB phase. This paper, which is the first of a series of COSAS papers, presents the results from the analyses of a first sample of 16 selected sources. The envelopes around late AGB stars are found to be mostly spherical, often mingled with features such as concentric arcs (R Cas and TX Cam), a broken spiral density pattern (TX Cam), molecular patches testifying to aspherical mass-loss (WX Psc, IK Tau, V Cyg, and S Cep), and also with well-defined axisymmetric morphologies and kinematical patterns (X Her and RX Boo). The sources span a wide range of angular sizes, from relatively compact (CRL 2362, OH 104.9+2.4 and CRL 2477) to very large (χ Cyg and TX Cam) envelopes, sometimes partially obscured by self-absorption features, which particularly for IK Tau and χ Cyg testifies to the emergence of aspherical winds in the innermost circumstellar regions. Strong axial structures with more or less complex morphologies are detected in four early post-AGB stars (IRAS 20028+3910, IRAS 23321+6545, IRAS 19475+3119 and IRAS 21282+5050) of the sub-sample.

  • A genetic variant in granzyme B is associated with progression of joint destruction in rheumatoid arthritis

    • Objective Genetic factors account for an estimated 4558% of the variance in joint destruction in rheumatoid arthritis (RA). The serine proteinase granzyme B induces target cell apoptosis, and several in vitro studies suggest that granzyme B is involved in apoptosis of chondrocytes. Serum levels of granzyme B are increased in RA and are also associated with radiographic erosions. The aim of this study was to investigate GZMB as a candidate gene accounting for the severity of joint destruction in RA. Methods A total of 1,418 patients with 4,885 radiograph sets of the hands and feet from 4 independent cohorts were studied. First, explorative analyses were performed in 600 RA patients in the Leiden Early Arthritis Clinic cohort. Fifteen single-nucleotide polymorphisms (SNPs) tagging GZMB were tested. Significantly associated SNPs were genotyped in data sets representing patients from the Groningen, Sheffield, and Lund cohorts. In each data set, the relative increase in the annual rate of progression in the presence of a genotype was assessed. Data were summarized in a meta-analysis. The association of GZMB with the RNA expression level of the GZMB genomic region was tested by mapping expression quantitative trait loci (QTLs) on 1,469 whole blood samples. Results SNP rs8192916 was significantly associated with the rate of joint destruction in the first cohort and in the meta-analysis of all data sets. Patients homozygous for the minor allele of rs8192916 had a higher rate of joint destruction per year compared with other patients (P = 7.8 x 104). Expression QTL of GZMB identified higher expression in the presence of the minor allele of rs8192916 (P = 2.27 x 105). Conclusion SNP rs8192916 located in GZMB is associated with the progression of joint destruction in RA as well as with RNA expression in whole blood.

    • ObjectiveThe progression of joint destruction in rheumatoid arthritis (RA) is determined by genetic factors. Changes in IL4 and IL4R genes have been associated with RA severity, but this finding has not been replicated. This study was undertaken to investigate the association between IL4- and IL4R-tagging single-nucleotide polymorphisms (SNPs) and the progression rate of joint damage in RA in a multicohort candidate gene study. MethodsIL4- and IL4R-tagging SNPs (n = 8 and 39, respectively) were genotyped in 600 RA patients for whom 2,846 sets of radiographs of the hands and feet were obtained during 7 years of followup. Subsequently, SNPs significantly associated with the progression of joint damage were genotyped and studied in relation to 3,415 radiographs of 1,953 RA patients; these included data sets from Groningen (The Netherlands), Lund (Sweden), Sheffield (UK), the North American Rheumatoid Arthritis Consortium (US), Wichita (US), and the National Data Bank (US). The relative increase in progression rate per year in the presence of a genotype was determined in each cohort. An inverse variance weighting meta-analysis was performed on the 6 data sets that together formed the replication phase. ResultsIn the discovery phase, none of the IL4 SNPs and 7 of the IL4R SNPs were significantly associated with the joint damage progression rate. In the replication phase, 2 SNPs in the IL4R gene were significantly associated with the joint damage progression rate (rs1805011 [P = 0.02] and rs1119132 [P = 0.001]). ConclusionGenetic variants in IL4R were identified, and their association with the progression rate of joint damage in RA was independently replicated.

    • The HIV-1 spike is a trimer of the transmembrane gp41 and the peripheral gp120 subunit pair. It is activated for virus-cell membrane fusion by binding sequentially to CD4 and to a chemokine receptor. Here we have studied the structural transition of the trimeric spike during the activation process. We solubilized and isolated unliganded and CD4-bound spikes from virus-like particles and used cryoelectron microscopy to reconstruct their 3D structures. In order to increase the yield and stability of the spike, we used an endodomain deleted and gp120-gp41 disulfide-linked variant. The unliganded spike displayed a hollow cage-like structure where the gp120-gp41 protomeric units formed a roof and bottom, and separated lobes and legs on the sides. The tripod structure was verified by fitting the recent atomic core structure of gp120 with intact N- and C-terminal ends into the spike density map. This defined the lobe as gp120 core, showed that the legs contained the polypeptide termini, and suggested the deleted variable loops V1/V2 and V3 to occupy the roof and gp41 the bottom. CD4 binding shifted the roof density peripherally and condensed the bottom density centrally. Fitting with a V3 containing gp120 core suggested that the V1/V2 loops in the roof were displaced laterally and the V3 lifted up, while the core and leg were kept in place. The loop displacements probably prepared the spike for coreceptor interaction and roof opening so that a new fusion-active gp41 structure, assembled at the center of the cage bottom, could reach the target membrane.

  • Sub-glass transition annealing enhances polymer solar cell performance

    • Thermal annealing of non-crystalline polymer: fullerene blends typically results in a drastic decrease in solar cell performance. In particular aggressive annealing above the glass transition temperature results in a detrimental coarsening of the blend nanostructure. We demonstrate that mild annealing below the glass transition temperature is a viable avenue to control the nanostructure of a non-crystalline thiophene-quinoxaline copolymer: fullerene blend. Direct imaging methods indicate that coarsening of the blend nanostructure can be avoided. However, a combination of absorption and luminescence spectroscopy reveals that local changes in the polymer conformation as well as limited fullerene aggregation are permitted to occur. As a result, we are able to optimise the solar cell performance evenly across different positions of the coated area, which is a necessary criterion for large-scale, high throughput production.

  • Bora and Aurora-A continue to activate Plk1 in mitosis

    • Polo-like kinase-1 (Plk1) is required for proper cell division. Activation of Plk1 requires phosphorylation on a conserved threonine in the T-loop of the kinase domain (T210). Plk1 is first phosphorylated on T210 in G2 phase by the kinase Aurora-A, in concert with its cofactor Bora. However, Bora was shown to be degraded prior to entry into mitosis, and it is currently unclear how Plk1 activity is sustained in mitosis. Here we show that the Bora/Aurora-A complex remains the major activator of Plk1 in mitosis. We show that a small amount of Aurora-A activity is sufficient to phosphorylate and activate Plk1 in mitosis. In addition, a fraction of Bora is retained in mitosis, which is essential for continued Aurora-A dependent T210 phosphorylation of Plk1. We find that once Plk1 is activated, minimal amounts of the Bora/Aurora-A complex are sufficient to sustain Plk1 activity. Thus, the activation of Plk1 by Aurora-A may function as a bistable switch; highly sensitive to inhibition of Aurora-A in its initial activation, but refractory to fluctuations in Aurora-A activity once Plk1 is fully activated. This provides a cell with robust Plk1 activity once it has committed to mitosis.

    • Thermal power plants produce SO2 during combustion of fuel containing sulfur. One way to decrease the SO2 emission from power plants is to introduce a sensor as part of the control system of the desulphurization unit. In this study, SiC-FET sensors were studied as one alternative sensor to replace the expensive FTIR (Fourier Transform Infrared) instrument or the inconvenient wet chemical methods. The gas response for the SiC-FET sensors comes from the interaction between the test gas and the catalytic gate metal, which changes the electrical characteristics of the devices. The performance of the sensors depends on the ability of the test gas to be adsorbed, decomposed, and desorbed at the sensor surface. The feature of SO2, that it is difficult to desorb from the catalyst surface, makes it known as catalyst poison. It is difficult to quantify the SO2 with static operation, even at the optimum operation temperature of the sensor due to low response levels and saturation already at low concentration of SO2. The challenge of SO2 desorption can be reduced by introducing dynamic operation in a designed temperature cycle operation (TCO). The intermittent exposure to high temperature can help to desorb SO2. Simultaneously, additional features extracted from the sensor data can be used to reduce the influence of sensor drift. The TCO operation, together with pattern recognition, may also reduce the baseline and response variation due to changing concentration of background gases (4-10% O-2 and 0-70% RH), and thus it may improve the overall sensor performance. In addition to the laboratory experiment, testing in the desulphurization pilot unit was performed. Desulphurization pilot unit has less controlled environment compared to the laboratory conditions. Therefore, the risk of influence from the changing concentration of background gas is higher. In this study, linear discriminant analysis (LDA) and partial least square (PLS) were employed as pattern recognition methods. It was demonstrated that using LDA quantification of SO2 into several groups of concentrations up to 2000 ppm was possible. Additionally, PLS analysis indicated a good agreement between the predicted value from the model and the SO2 concentration from the reference instrument of the pilot plant.

    • The v=1, 2 J=1–0 and v=1 J=2–1 are intense SiO masers, often mapped in AGB stars. Their distribution displays ring-like structures in the regions close to the star. The distribution of the v=1, 2 J=1–0 masers are similar, but the spots are rarely coincident. The v=1 J=2–1 maser ar-rises, however, from a well separated region. It has been argued that this difference can only be explained by models including overlap of two IR lines of SiO and H2O. The v=3 J=1–0 line is not expected to be affected by any line overlap, and its spot structure should be that predicted by the standard models. Our first results on the relative positions of the three lines are surprising but yet consistent with current models including line overlap.

    • We present the results of SiO maser observations at 43 GHz toward two AGB stars using the VLBA. Our preliminary results on the relative positions of the different J=1–0 SiO masers (v=1,2 and 3) indicate that the current ideas on SiO maser pumping could be wrong at some fundamental level. A deep revision of the SiO pumping models could be necessary.

Those of the v = 1, v = 2 J = 1-0 masers are similar, but the spots are rarely coincident, while the v = 1 J = 2-1 maser arises from a well-separated region farther out. These relative locations can be explained by models tools that include the overlap of two IR lines of SiO and H2O. The v = 3 J = 1-0 line is not directly affected by any line overlap and its spot structure and position, relative to the other lines, is a good test to the standard pumping models. Aims. The aim of this project are to gain insight into the properties and the general theoretical considerations of the different SiO masers that can help to understand them. Methods. We present single-dish and simultaneous VLBI observations of the v = 1, v = 2, and v = 3 J = 1-0 maser transitions of (SiO)-Si-28 in several AGB stars. The results are compared to the predictions of radiative models of SiO masers that both include and not include the effect of IR line overlap. Results. The spatial distribution of the SiO maser emission in the v = 3 J = 1-0 transition from AGB stars is systematically composed of a series of spots that occupy a ring-like structure (as often found in SiO masers). The overall ring structure is extremely similar to that found in the other 43 GHz transitions and is very different from the structure of the v = 1 J = 2-1 maser. The positions of the individual spots of the different 43 GHz lines are, however, very rarely coincident, which in general is separated by about 0.3 AU (between 1 and 5 mas). These results are very difficult to reconcile with standard pumping models, which predict that the masers of rotational transitions within a given vibrational state require very similar excitation conditions (since the levels are placed practically at the same energy from the ground), while the transitions of different vibrational states (which are separated by an energy of 1800 K) should appear in different positions. However, models including line overlap tend to predict v = 1, v = 2, v = 3 J = 1-0 population inversion to occur under very similar conditions, while the requirements for v = 1 J = 2-1 appear clearly different, and are compatible with the observational results.

    • The NMDA-receptor antagonist ketamine has proven efficient in reducing symptoms of suicidality, although the mechanisms explaining this effect have not been detailed in psychiatric patients. Recent evidence points towards a low-grade inflammation in brains of suicide victims. Inflammation leads to production of quinolinic acid (QUIN) and kynurenic acid (KYNA), an agonist and antagonist of the glutamatergic N-methyl-D-aspartate (NMDA) receptor, respectively. We here measured QUIN and KYNA in the cerebrospinal fluid (CSF) of 64 medication-free suicide attempters and 36 controls, using gas chromatography mass spectrometry and high-performance liquid chromatography. We assessed the patients clinically using the Suicide Intent Scale and the Montgomery Asberg Depression Rating Scale (MADRS). We found that QUIN, but not KYNA, was significantly elevated in the CSF of suicide attempters (Pandlt;0.001). As predicted, the increase in QUIN was associated with higher levels of CSF interleukin-6. Moreover, QUIN levels correlated with the total scores on Suicide Intent Scale. There was a significant decrease of QUIN in patients who came for follow-up lumbar punctures within 6 months after the suicide attempt. In summary, we here present clinical evidence of increased QUIN in the CSF of suicide attempters. An increased QUIN/KYNA quotient speaks in favor of an overall NMDA-receptor stimulation. The correlation between QUIN and the Suicide Intent Scale indicates that changes in glutamatergic neurotransmission could be specifically linked to suicidality. Our findings have important implications for the detection and specific treatment of suicidal patients, and might explain the observed remedial effects of ketamine.     • Background: Motivational interviewing (MI) is a collaborative, client-centred counselling style aimed at eliciting and strengthening clients' intrinsic motivation to change. There is strong research evidence supporting the efficacy of MI, notably in its application among alcohol and drug abuse populations. MI interventions in smoking cessation may yield modest but significant increases in quitting. The present study sought to assess the acquisition and retention of MI skills in counsellors at the Swedish National Tobacco Quitline. Methods: Three audio-recorded sessions from each of three counsellors were assessed using the Motivational Interviewing Treatment Integrity (MITI) Code Version 3.0 over 11 assessment periods at fixed intervals in a two-and-a-half year period during which counsellors received ongoing supervision. Results: The mean skill for all counsellors improved throughout the study period in most MITI variables. However, great variations in MI skill between counsellors were observed, as well as fluctuations in performance in counsellors over time. Conclusion: The present exploratory study covers a longer time period than most evaluations of MI training, and has several advantages with regard to study design. It may provide a basis for (larger sample) replication to test MI skill (as measured by the MITI) in relation to behaviour change in clients, to evaluate MI training, and to assess the acquisition and retention of MI skill over time. Difficulties in acquiring and retaining MI skill may raise the issue of a selection policy for MI training. Moreover, fluctuations in MI skill over time emphasise the greater importance of continuous feedback and supervision over initial MI training, and the need for the use of validated treatment integrity assessment instruments in ordinary clinical implementations of MI.

Compared to the traditional octave bandwidth feeds, wide bandwidth feeds, such as the Eleven feed, will create the possibility of significantly larger instantaneous frequency coverage observations with existing radio telescopes, such as those used in the VLBI networks, and for future radio telescopes, such as the Square Kilometre Array.     • BACKGROUND: The left ventricular (LV) long axis (Lax) function is very sensitive in documenting myocardial abnormalities in aortic stenosis (AS). We hypothesized that Lax recovery after aortic valve replacement (AVR) is related to the extent of cavity dyssynchrony measured by total isovolumic time (t-IVT).METHODS: A consecutive 107 patients (aged 70±7years, 70 male) with severe AS and Lax impairment were studied. T-IVT was measured before and after AVR. Reduced Lax function and its post-operative recovery were defined as mitral annular plane systolic excursion (MAPSE) ≦10mm and an increase of MAPSE >10%, respectively.RESULTS: LV function improved (EF: 43±8 to 48±10%; MAPSE: 7.9±1.0 to 11.0±2.4mm) and t-IVT shortened (9.7±3.7 to 7.0±2.8s/min, p<0.01 for all) after AVR. Sixty-five (61%) patients had Lax recovery after a median of 32-month follow-up. Univariate predictors were LV size, LA dimensions, the presence of restrictive LV filling and prolonged t-IVT. Only LV end-systolic dimension, restrictive filling and t-IVT (OR 0.61, 95% CI 0.47-0.79, p<0.01) were independent predictors. A pre-operative t-IVT≦9.3s/min was 81% sensitive and 63% specific in predicting Lax recovery (AUC 0.81, p<0.001). The prevalence of CAD or concomitant CABG were similar in 2 patient groups with different t-IVT.CONCLUSIONS: Lax recovery was evident in the majority of AS patients after AVR. The lower prevalence of Lax recovery seen in patients with prolonged t-IVT suggests that dyssynchrony may play an important role in the process of adverse LV remodeling.

  • Hospital organizational factors influence work-family conflict in registered nurses : multilevel modeling of a nation-wide cross-sectional survey in Sweden

    • Background: The present shortage of registered nurses (RNs) in many European countries is expected to continue and worsen, which poses a substantial threat to the maintenance of healthcare in this region. Work-family conflict is a known risk factor for turnover and sickness absence. Objective: This paper empirically examines whether the nurse practice environment is associated with experienced work-family conflict. Design: A multilevel model was fit with the individual RN at the 1st, and the hospital department at the 2nd level using cross-sectional RN survey data from the Swedish part of RN4CAST, an EU 7th framework project. The data analyzed here is based on a national sample of 8356 female and 592 male RNs from 369 hospital departments. Results: We found that 6% of the variability in work-family conflict experienced by RNs was at the department level. Organizational level factors significantly accounted for most of the variability at this level with two of the work practice environment factors examined, staffing adequacy and nurse involvement in hospital affairs, significantly related to work-family conflict. Due to the design of the study, factors on ward and work group levels could not be analyzed, but are likely to account for additional variance which in the present analysis appears to be on the individual level, with private life factors likely explaining another major part. Conclusion: These results suggest that higher level organizational factors in health care have a significant impact on the risk of work-family conflict among RNs through their impact on the nurse practice environment. Lower level organizational factors should be investigated in future studies using hierarchical multilevel sampling.

    • Several psychiatric disorders, including major depressive disorder (MDD), are associated with increased blood markers of oxidative stress. The relevance of this to the oxidation-sensitive hippocampus (HC) is unknown. We investigated the relationship between peripheral oxidative stress markers and HC volume in unmedicated individuals with MDD (n=16) and healthy controls (n=19). To conserve power, our primary analysis was carried out in the combined group of subjects, and secondary analyses examined each group separately. Oxidative stress markers (oxidized glutathione (GSSG)) and antioxidants (reduced glutathione (GSH), glutathione peroxidase (Gpx), and Vitamin C) were assessed, and a "total net antioxidant score" was calculated. 4-T MRI estimated total HC volume and HC subfield (CA1, CA1-CA2 transition zone, subiculum and CA3/dentate gyrus [CA3&DG]) volumes. Across groups, the antioxidant score was significantly and positively correlated with total HC volume and CA3&DG subfield volume (normalized to total intracranial volume), adjusting for age and sex. Similar relationships were observed in each individual group but missed statistical significance, likely due to type II errors, with the exception of a significant correlation between the antioxidant score and CA3&DG volume in the MDD group. These preliminary data are consistent with oxidative stress being associated with smaller total HC and CA3&DG subfield volumes.

    • The associations between immune-related conditions and multiple myeloma (MM) and monoclonal gammopathy of undetermined significance (MGUS) have previously been investigated with inconsistent results. In a large population-based study, we identified 19 112 patients with MM, 5403 patients with MGUS, 96 617 matched control subjects, and 262 931 first-degree relatives. We calculated odds ratios (ORs) and 95% confidence intervals (CIs) for the association of MM and MGUS with immune-related conditions by use of logistic regression. A personal history of all infections combined was associated with a significantly increased risk of MM (OR = 1.2; 95% CI, 1.1-1.3), and a personal history of all conditions in the categories infections (OR = 1.6; 95% CI, 1.5-1.7), inflammatory conditions (OR = 1.4; 95% CI, 1.2-1.5), and autoimmune diseases (OR = 2.1; 95% CI, 1.9-2.4) was associated with a significantly increased risk of MGUS. Several specific immune-related conditions elevated the risk of MM and/or MGUS. A family history of autoimmune disease was associated with a significantly increased risk of MGUS (OR = 1.1; 95% CI, 1.00-1.2), but not MM. Our findings suggest that immune-related conditions and/or their treatment are of importance in the etiology of MGUS and possibly MM. The association of both personal and family history of autoimmune disease with MGUS indicates the potential for shared susceptibility for these conditions. (Blood. 2011; 118(24): 6284-6291)

    • The Wnt planar cell polarity (Wnt/PCP) pathway signals through small Rho-like GTPases to regulate the cytoskeleton. The core PCP proteins have been mapped to the Wnt/PCP pathway genetically, but the molecular mechanism of their action remains unknown. Here, we investigate the function of the mammalian PCP protein Vang-like protein 2 (Vangl2). RNAi knockdown of Vangl2 impaired cell-cell adhesion and cytoskeletal integrity in the epithelial cell lines HEK293T and MDCK. Similar effects were observed when Vangl2 was overexpressed in HEK293T, MDCK or C17.2 cells. The effects of Vangl2 overexpression could be blocked by knockdown of the small GTPase Rac1 or by dominant-negative Rac1. In itself, knockdown of Rac1 impaired cytoskeletal integrity and reduced cell-cell adhesion. We found that Vangl2 bound and re-distributed Rac1 within the cells but did not alter Rac1 activity. Moreover, both transgenic mouse embryos overexpressing Vangl2 in neural stem cells and loop-tail Vangl2 loss-of-function embryos displayed impaired adherens junctions, a cytoskeletal unit essential for neural tube rigidity and neural tube closure. In vivo, Rac1 was re-distributed within the cells in a similar way to that observed by us in vitro. We propose that Vangl2 affects cell adhesion and the cytoskeleton by recruiting Rac1 and targeting its activity in the cell to adherens junctions.

    • The Env protein of murine leukemia virus matures by two cleavage events. First, cellular furin separates the receptor binding surface (SU) subunit from the fusion-active transmembrane (TM) subunit and then, in the newly assembled particle, the viral protease removes a 16-residue peptide, the R-peptide from the endodomain of the TM. Both cleavage events are required to prime the Env for receptor-triggered activation. Cryoelectron microscopy (cryo-EM) analyses have shown that the mature Env forms an open cage-like structure composed of three SU–TM complexes, where the TM subunits formed separated Env legs. Here we have studied the structure of the R-peptide precursor Env by cryo-EM. TM cleavage in Moloney murine leukemia virus was inhibited by amprenavir, and the Envs were solubilized in Triton X-100 and isolated by sedimentation in a sucrose gradient. We found that the legs of the R-peptide Env were held together by trimeric interactions at the very bottom of the Env. This suggested that the R-peptide ties the TM legs together and that this prevents the activation of the TM for fusion. The model was supported by further cryo-EM studies using an R-peptide Env mutant that was fusion-competent despite an uncleaved R-peptide. The Env legs of this mutant were found to be separated, like in the mature Env. This shows that it is the TM leg separation, normally caused by R-peptide cleavage, that primes the Env for receptor triggering.

  • Characteristics of storm time electric fields in the inner magnetosphere derived from Cluster data

    • Storm-time electric fields in the inner magnetosphere measured by Cluster are reported in this study. First, we show two events around the time when Dst index is at a minimum. The electric field possibly related to subauroral ion drifts and/or undershielding is measured inside the inner edge of the electron plasma sheet in the eveningside. For the second event observed in the nightside, the electric field is partly related to dipolarization and is considered as inductive. An electric field without coincident magnetic signatures is also observed. Spatial coherence of the electric field is not large when we check multispacecraft data. It is inferred that the electric field in the magnetotail penetrates inside the region 1 current, while it is not clear about the electric field within the region 2 current from our data. Then superposed epoch analyses using 71 storms are performed. Electric fields at R = 3.5-6R(E) and less than 25 degrees of magnetic latitudes are enhanced around the minimum Dst at all magnetic local times. Electric fields during the recovery phase decay on a time scale shorter than that of Dst index, which could be interpreted in terms of the relation between electric field and ring current during that storm phase. AC electric fields are generally larger than DC electric fields, indicating that the former component might play some role in accelerating ring current particles. These results will be useful to update our empirical electric field model.

    • In this event study, small-scale fluctuations in plasmaspheric plumes with time scales of similar to 10 s to minutes in the spacecraft frame are examined. In one event, plasmaspheric plumes are observed by Cluster, while IMAGE measured density enhancement at a similar location. Fluctuations in density exist in plumes as detected by Cluster and are accompanied by fluctuations in magnetic fields and electric fields. Magnetic fluctuations are transverse and along the direction of the plumes. The E/B ratio is smaller than the Alfv,n velocity. Another similar event is briefly presented. We then consider physical properties of the fluctuations. Alfv,n mode modulated by the feedback instability is one possibility, although non-local generation is likely. It is hard to show that the fluctuations represent a fast mode. Interchange motion is possible due to the consistency between measurements and expectations. The energy source could be a pressure or density gradient in plasmaspheric plumes. When more events are accumulated so that statistical analysis becomes feasible, this type of study will be useful to understand the time evolution of plumes.

    • Using Cluster data from the Electron Drift (EDI) and the Electric Field and Wave (EFW) instruments, we revise our empirically-based, inner-magnetospheric electric field (UNH-IMEF) model at 22.662 mV/m; K-p<1, 1K(p)<2, 2K(p)<3, 3K(p)<4, 4K(p)<5, and K(p)4(+). Patterns consist of one set of data and processing for smaller activities, and another for higher activities. As activity increases, the skewed potential contour related to the partial ring current appears on the nightside. With the revised analysis, we find that the skewed potential contours get clearer and potential contours get denser on the nightside and morningside. Since the fluctuating components are not negligible, standard deviations from the modeled values are included in the model. In this study, we perform validation of the derived model more extensively. We find experimentally that the skewed contours are located close to the last closed equipotential, consistent with previous theories. This gives physical context to our model and serves as one validation effort. As another validation effort, the derived results are compared with other models/measurements. From these comparisons, we conclude that our model has some clear advantages over the others.

    • We have mapped the (CO)-C-12(3-2) line emission around the Mira AB system at 0 ''.5 resolution using the Atacama Large Millimeter/submillimeter Array (ALMA). The CO map shows amazing complexity. The circumstellar gas has been shaped by different dynamical actors during the evolution of the system, and several morphological components can be identified. The companion is marginally resolved in continuum emission and is currently at 0 ''.487 +/- 0 ''.006 separation. In the main line component, centered on the stellar velocity, spiral arcs around Mira A are found. The spiral appears to be relatively flat and oriented in the orbital plane. An accretion wake behind the companion is clearly visible, and the projected arc separation is about 5 ''. In the blue wing of the line emission, offset from the main line, several large (similar to 5-10 '') opposing arcs are found. We tentatively suggest that this structure is created by the wind of Mira B blowing a bubble in the expanding envelope of Mira A.

    • Understanding the effect of particle shape on dry rheology is considered to be an important aspect in order to replace natural sand used in concrete with man-made manufactured sand. There is a significant difference in particle shape and surface texture between crushed rock material (aggregates) and natural sand. The crushed aggregates are normally less rounded and have less equally shaped dimensions compared to natural sand. Thus, the flow ability of the concrete will be different. The main difference is explained by a difference in the wet rheology of the cement paste which in turn is affected by the smallest particles in the size range from microns to a few millimetres. In previous research, it has been demonstrated how different crushing techniques affects the shape of the produced particles. In many cases it is believed that Vertical Shaft Impact (VSI) -technology is the way forward to replace natural sand. The aim of this paper is to investigate the relationship between dry rheology measured with the so called flow cone and particle shape parameters measured with an optical image analyser technique.

    • ObjectiveThe Swedish Early Psoriatic Arthritis Register describes the course of early psoriatic arthritis (PsA) in a real life clinical setting in Sweden. The aim of this study was to obtain information on predictors of clinical outcomes over a 5-year period with special focus on effects of gender, joint patterns, diagnostic delay and initial disease activity.MethodsIn six centres, patients with signs suggestive of PsA were included in the Swedish Early Psoriatic Arthritis Register within 2 years of symptom onset. CASPAR (classification for psoriatic arthritis) criteria were fulfilled by 197 patients who had passed the 5-year follow-up. Disease activity was measured by the Disease Activity Score including 28 joints (DAS28) and the Disease Activity Index for Psoriatic Arthritis (DAPSA). Remission and minimal disease activity (MDA) were used as outcome measures.ResultsMean age at inclusion was 46 years, younger in male than female patients (43 vs 48 years). Mean DAS28 was 3.7 and 3.0 at inclusion and 2.8 and 2.1 at follow-up for women and men, respectively-significantly higher in women at both visits. Likewise, DAPSA scores were significantly higher in women. The degree of improvement (change in DAS28 and DAPSA) was similar. Men achieved MDA or remission (50% vs 33%, 25% vs 13%, respectively) more often, and women had significantly more polyarthritis at inclusion (49% vs 27%) and after 5 years (25% vs 15%). Axial or mono/oligoarticular disease was predominant in men. Independent predictors of MDA at the 5-year follow-up were: shorter symptom duration; greater general wellbeing (global visual analogue scale); and low Health Assessment Questionnaire at inclusion.ConclusionsIn early PsA, short delay between onset of symptoms and diagnosis, preserved function, and male gender are the most important predictors of favourable clinical outcome at the 5-year follow-up. Early recognition of PsA and active treatment may be important, particularly in women with polyarticular disease.
