    • The observation of neutrinoless double-beta decay (0νββ) would show that lepton number is violated, reveal that neu-trinos are Majorana particles, and provide information on neutrino mass. A discovery-capable experiment covering the inverted ordering region, with effective Majorana neutrino masses of 15 - 50 meV, will require a tonne-scale experiment with excellent energy resolution and extremely low backgrounds, at the level of ∼0.1 count /(FWHM·t·yr) in the region of the signal. The current generation 76Ge experiments GERDA and the Majorana Demonstrator, utilizing high purity Germanium detectors with an intrinsic energy resolution of 0.12%, have achieved the lowest backgrounds by over an order of magnitude in the 0νββ signal region of all 0νββ experiments. Building on this success, the LEGEND collaboration has been formed to pursue a tonne-scale 76Ge experiment. The collaboration aims to develop a phased 0νββ experimental program with discovery potential at a half-life approaching or at 1028 years, using existing resources as appropriate to expedite physics results.

  • Genome-wide association meta-analysis in 652,134 participants identifies 9 novel susceptibility loci for aortic stenosis

    • Background: Aortic stenosis (AS) is the most common form of incident valvular heart disease. While valve replacement is effective, the absence of an approved medical therapy provides no alternatives to patients with contraindications or mild disease. An improved understanding of the genetics of AS could identify targets for pharmacological intervention.Methods: An inverse variance-weighted, fixed effects meta-analysis of the association of 11,591,806 variants with AS was undertaken using data from 10 European cohorts totalling 652,134 participants (13,758 cases of AS). We queried publicly available datasets to characterize the functional consequences of genome-wide significant variants, conducted a phenome-wide association study to assess their association with other outcomes, and constructed polygenic risk scores to examine their association with AS. We also performed gene- and gene-set enrichment analyses, estimated genetic correlation with cardiovascular traits, and assessed whether five lipid or immunological biomarkers were causally associated with AS using Mendelian randomization.Results: Eighteen independent variants at 16 loci attained genome-wide significance in the meta-analysis, including variants at all seven previously reported loci. Many of the significant variants were intronic or intergenic, and the phenome-wide association study revealed extensive pleiotropy with apolipoprotein B, C-reactive protein, and other cardiovascular and immunological traits. A weighted polygenic risk score composed of the 18 variants was strongly associated with AS (adjusted OR per SD, 1.38; 95% CI, 1.33 to 1.44; p=4.6×10–57), and improved the discriminatory ability for AS when added to a model that contained clinical risk factors (difference in the area under the curve p=2.0×10–11). Gene-based approaches indicated higher IL6R expression in the blood among AS cases compared to controls (p=3.1×10–6), and the association of LDLR with AS (p=2.3×10–10). Gene set analyses revealed that genes bound by the transcription factor TCF7 or micro-RNAs miR-21, miR-219, miR-491, and miR-19 were differentially expressed in the liver depending on AS status (p≤5.7×10–4), suggesting disease development may be mediated by tissue-specific transcriptional and post-transcriptional regulation. Mendelian randomization supported a causal association of five lipid and immunological biomarkers with AS, including low-density lipoprotein cholesterol (OR per mmol/L, 1.61; 95% CI, 1.48 to 1.75; p=1.3×10–30).Conclusions: Evidence from large-scale genetic analyses indicate that lipid metabolism, inflammation, and calcification are key contributors to AS.

  • Librator: a platform for the optimized analysis, design, and expression of mutable influenza viral antigens

    • Artificial mutagenesis and protein engineering have laid the foundation for antigenic characterization and universal vaccine design for influenza viruses. However, many methods used in this process require manual sequence editing and protein expression, limiting their efficiency and utility in high-throughput applications. More streamlined in silico tools allowing researchers to properly analyze and visualize influenza viral protein sequences with accurate nomenclature are necessary to improve antigen design and productivity. To address this need, we developed Librator, a system for analyzing and designing custom protein sequences of influenza virus hemagglutinin (HA) and neuraminidase (NA) glycoproteins. Within Librator’s graphical interface, users can easily interrogate viral sequences and phylogenies, visualize antigen structures and conservation, mutate target residues and design custom antigens. Librator also provides optimized fragment design for Gibson Assembly of HA and NA expression constructs based on peptide conservation of all historical HA and NA sequences, ensuring fragments are reusable and compatible across related subtypes, thereby promoting reagent savings. Finally, the program facilitates single-cell immune profiling, epitope mapping of monoclonal antibodies and mosaic protein design. Using Librator-based antigen construction, we demonstrate that antigenicity can be readily transferred between HA molecules of H3, but not H1, lineage viruses. Altogether, Librator is a valuable tool for analyzing influenza virus HA and NA proteins and provides an efficient resource for optimizing recombinant influenza antigen synthesis.

  • Biobased production of succinic acid by Escherichia coli fermentation

    • The prospects of peak oil, climate change and the dependency of fossil carbon have urged research and development of production methods for the manufacture of fuels and chemicals from renewable resources (biomass). The present thesis illustrates different aspects of biobased succinic acid production by a metabolically engineered E. coli strain. The main areas of the thesis are sugar utilisation and feedstock flexibility, and fermentation inhibition, both due to toxic compound derived from the raw material and the fermentation products themselves.The first part of this thesis aimed to investigate the fermentation characteristics of AFP184 in a medium consisting of corn steep liquor, inorganic salts and different sugar sources without supplementation with high-cost nutrients such as yeast extract and peptone. The effects of different sugars, sucrose, glucose, fructose, xylose, equal mixtures of glucose-fructose and glucose-xylose, on succinic acid production kinetics and yields in an industrially relevant medium were investigated. AFP184 was able to utilise all sugars and sugar combinations except sucrose for biomass generation and succinate production. Using glucose resulted in the highest yield, 0.83 (g succinic acid per g sugar consumed anaerobically). Using a high initial sugar concentration resulted in volumetric productivities of almost 3 g L-1 h-1, which is above estimated values for economically feasible production. However, succinic acid production ceased at final concentrations greater than 40 g L-1. To further increase succinic acid concentrations, fermentations using NH4OH, NaOH, KOH, K2CO3, and Na2CO3 as neutralising agents were performed and compared. It was shown that substantial improvements could be made by using alkali bases to neutralise the fermentations. The highest concentrations and productivities were achieved when Na2CO3 was used, 77 g L-1 and 3 g L-1 h-1 respectively. A gradual decrease in succinate productivity was observed during the fermentations, which was shown to be due to succinate accumulation in the broth and not as a result of the addition of neutralising agent or the subsequent increase in osmolarity.To maintain high succinate productivity by keeping a low extracellular succinic acid concentration fermentations were interrupted and cells recovered and resuspended in fresh media. By removing the succinate it was possible to maintain high succinic acid productivity for a prolonged time. Cells subjected to high concentrations of succinate were also able to regain high productivity once transferred into a succinate-free medium.In the last part of the thesis succinic acid production from softwood dilute acid hydrolysates was demonstrated. This study involved establishing the degree of detoxification necessary for growth and fermentation using industrial hydrolysates. Detoxification by treatment with lime and/or activated carbon was investigated and the results show that it was possible to produce succinate from softwood hydrolysates in yields comparable to those for synthetic sugars.The work done in this thesis increases the understanding of succinic acid production with AFP184, illustrate its limitations, and suggests improvements in the current technology with the long term aim of increasing the economical feasibility of biochemical succinic acid production.

    • Resolving the deep relationships of ancient animal lineages has proven difficult using standard Sanger-sequencing approaches with a handful of markers. We thus reassess the relatively well-studied phylogeny of the phylum Nemertea (ribbon worms)—for which the targeted gene approaches had resolved many clades but had left key phylogenetic gaps—by using a phylogenomic approach using Illumina-based de novo assembled transcriptomes and automatic orthology prediction methods. The analysis of a concatenated data set of 2,779 genes (411,138 amino acids) with about 78% gene occupancy and a reduced version with 95% gene occupancy, under evolutionary models accounting or not for site-specific amino acid replacement patterns results in a well-supported phylogeny that recovers all major accepted nemertean clades with the monophyly of Heteronemertea, Hoplonemertea, Monostilifera, being well supported. Significantly, all the ambiguous patterns inferred from Sanger-based approaches were resolved, namely the monophyly of Palaeonemertea and Pilidiophora. By testing for possible conflict in the analyzed supermatrix, we observed that concatenation was the best solution, and the results of the analyses should settle prior debates on nemertean phylogeny. The study highlights the importance, feasibility, and completeness of Illumina-based phylogenomic data matrices.

    • This year theMontreal Protocol celebrates its 20th Anniversary. In September 1987, 24 countries signed the ‘Montreal Protocol on Substances that Deplete the Ozone Layer’. Today 191 countries have signed and have met strict commitments on phasing out of ozone depleting substances with the result that a 95% reduction of these substances has been achieved. The Montreal Protocol has also contributed to slowing the rate of global climate change, since most of the ozone depleting substances are also effective greenhouse gases. Even though much has been achieved, the future of the stratospheric ozone layer relies on full compliance of the Montreal Protocol by all countries for the remaining substances, including methyl bromide, as well as strict monitoring of potential risks from the production of substitute chemicals. Also the ozone depleting substances existing in banks and equipment need special attention to prevent their release to the stratosphere. Since many of the ozone depleting substances already in the atmosphere are long-lived, recovery cannot be immediate and present projections estimate a return to pre-1980 levels by 2050 to 2075. It has also been predicted that the interactions of the effects of the ozone layer and that of other climate change factors will become increasingly important.

  • Predicting Air Compressor Failures Using Long Short Term Memory Networks

    • We introduce an LSTM-based method for predicting compressor failures using aggregated sensory data, and evaluate it using historical information from over 1000 heavy duty vehicles during 2015 and 2016. The goal is to proactively identify trucks that will require maintenance in the near future, so that component replacement can be scheduled before the failure happens, translating into improved uptime. The problem is formulated as a classification task of whether a compressor failure will happen within the specified prediction horizon. A recurrent neural network using Long Short-Term Memory (LSTM) architecture is employed as the prediction model, and compared against Random Forest (RF), the solution used in industrial deployment at the moment. Experimental results show that while Random Forest slightly outperforms LSTM in terms of AUC score, the predictions of LSTM stay significantly more stable over time, showing a consistent trend from healthy to faulty class. Additionally, LSTM is also better at detecting the switch from faulty class to the healthy one after a repair. We demonstrate that this stability is important for making repair decisions, especially in questionable cases, and therefore LSTM model is likely to lead to better results in practice. © Springer Nature Switzerland AG 2019

    • Collaborative CAD systems enabling collaboration in computer-aided design processes among distributed designers are gaining more and more attention. Yet, such systems, especially in support of collaborative assembly modeling, are hardly achievable. Targeting this gap, this paper addresses an Internet-enabled real-time collaborative assembly modeling system, named e-Assembly. This emerging system allows a group of geographically dispersed designers to jointly build an assembly model in real time over the Internet. In particular, this paper proposes a new assembly representation, called Collaborative Assembly Representation, for Internet-based collaborative assembly modeling. Also, collaborative assembly constraint satisfaction is addressed based on three coordination rules embedded in e-Assembly. Furthermore, the system architecture and realization of e-Assembly are provided. Finally, a prototypic implementation of e-Assembly is presented for demonstration and discussion.

    • We report the application of multilayered polyelectrolyte-coated gold nanorods (GNRs) as multifunctional optical contrast agents for cancer cell imaging. The surface modification of GNRs improves their chemical stability and facilitates them to be taken up by cancer cells through electrostatic interaction. The unique longitudinal surface plasmon resonance property of GNRs makes them suitable as both "scattering contrast agents" and "Raman contrast agents". In our experiments, the staining of GNRs in cells was further confirmed by dark field microscopy and Raman microscopy. Our experiment results indicated that GNRs have great potential as multifunctional "optical contrast agents" for future in vivo animal imaging.

  • Obsessive-compulsive disorder : novel insights on executive functions, gut microbiome, and genetics

    • Background: Obsessive-compulsive disorder (OCD) is characterized by intrusive thoughts and images, and repetitive, time-consuming compulsions. It causes disability and impaired quality of life. The neurobiological model of OCD revolves around dysfunctional brain circuits, referred to as the cortico-striato-thalamocortical (CSTC) loop. Aim: This thesis sought to elucidate neurobiological factors associated with OCD. Methods: In study I, we compared executive functions of patients with OCD (n=29), body dysmorphic disorder (BDD) (n=27), and healthy controls (n=28), using computerized and standardized neuropsychological tasks (CANTAB) for response inhibition (stop-signal task, SST), cognitive flexibility (intra-extra dimensional shift task, IED), and working memory (spatial working memory task, SWM). Correlation between task performance and symptom severity was assessed using linear regression models and effect sizes were measured with Pearson’s r. In study II, we systematically reviewed gut microbiota studies of participants with psychiatric disorders, published up to February 2020, following the PRISMA guidelines and the pre-registered study-protocol at PROSPERO. In study III, we compared gut microbiome of patients with OCD (n=32) with healthy controls (n=32) using whole genome sequencing of stool samples. Moreover, we followed a subset of participants longitudinally to compare changes in gut microbiome within patients with OCD before and after exposure- and response prevention (ERP) therapy (n=15). In study IV, we used whole exome sequencing to explore presence of gene-disruptive rare variants (GDRVs) in patients with severe and treatment-resistant OCD (n=5) who have received deep brain stimulation (DBS). Results: In study I we found no significant differences in test performance between individuals with OCD, BDD, and healthy controls. In study II, 69 studies were included in the systematic review. The majority of studies did not report any significant differences in gut microbiota ?-diversity indices (44%), but different composition measured by ?-diversity indices (67%) between patients with psychiatric disorders and healthy controls. However, the results were inconsistent regarding which genera are differentially abundant, except for some studies that reporter lower abundance of butyrate-producing microbes Faecalibacterium prausnitzii and Roseburia species. In study III we found no significant differences in ?-diversity, ?-diversity, or taxonomic dissimilarity at the species-level between patients with OCD and healthy controls, or within patients with OCD before and after ERP. Furthermore, functional analysis, based on gut-brain modules that are relevant for metabolizing neuroactive compounds, did not reveal any significant differences between patients with OCD and healthy controls, or within patients with OCD before and after ERP. In study IV, we found three GDRVs, one of which was a missense variant in the ion transporter domain of KCNB1 (hg19 20-47991077-C-T). The patient with that missense variant was a responder to DBS treatment. Conclusions: Study I. Performance on neuropsychological tests suggest that patients with OCD have comparable executive functions with a related group of patients and healthy controls. Study II and III. Despite the suggested importance of the microbiome-gut-brain axis in psychiatric disorders, evidence for altered gut microbiome in psychiatric disorders including OCD is inconclusive. Improved and consistent study design and methodology are crucial for future studies in this research field. Study IV. Patients with severe and treatment-resistant OCD is a cohort fit for whole exome sequencing studies. Despite the small sample size, we found three GDRVs. More samples are required to evaluate if the frequency of GDRVs is higher in this cohort and if GDRVs could predict treatment outcome from DBS. Patients with OCD represent a heterogeneous group, and individual biological factors cannot explain the pathophysiological underpinning of this disorder. Nevertheless, next-generation sequencing opens new opportunities to study complex psychiatric conditions. Genomic analysis of severe and treatment-resistant patients with OCD could increase our understanding of this complex disorder.

    • METHODS: Multiple Swedish nationwide registers were used to identify 8045 individuals, aged 20-29, with an incident diagnosis of ADHD 2006-2011. Labour market integration was conceptualized according to the core-peripheral model as a continuum from a strong (core) to a weak (peripheral) connection to the labour market. Sequence analyses categorized clusters of labour market integration, from 1 year before to 5 years after their ADHD diagnosis for individuals diagnosed with ADHD and a matched control group without ADHD. Multinomial logistic regression computed odds ratios (ORs) with 95% confidence intervals (CIs) between sociodemographic factors and comorbid disorders and the identified clusters.RESULTS: About one-fourth of the young adults diagnosed with ADHD belonged to clusters characterized by a transition to a mainly peripheral labour market position, which was approximately four-times higher compared to controls without ADHD. Foremost, those living in small cities/villages (OR 1.9; CI 1.5-2.2), those having comorbid autism-spectrum disorder (OR 13.7; CI 6.8-27.5) or schizophrenia/psychoses (OR 7.8; CI 3.8-15.9) were associated with a transition towards a peripheral labour market position throughout the study period. Those with a high educational level (OR 0.1; CI 0.1-0.1), and men (OR 0.7; CI 0.6-0.8) were less likely to have a peripheral labour market position.CONCLUSIONS: Young adults diagnosed with ADHD are four-times more likely to be in the peripheral labour market position compared to those without ADHD. To increase labour market participation, special attention is warranted to those with low educational level, those living outside big cities and those with comorbid mental disorders.

    • OBJECTIVE: It has been reported that diabetes may increase the risk of cadmium-induced kidney damage. The presence of metallothionein antibody (MT-Ab) increased the susceptibility for tubular damage among cadmium workers. This study focused on the relationships between levels of MT-Ab, urinary cadmium, and kidney function in a Chinese type 2 diabetic population. RESEARCH DESIGN AND METHODS: A cross-sectional study was performed on 229 type 2 diabetic patients (92 men and 137 women) who were recruited from two community centers in one district of Shanghai City in China. Information was obtained from interviews, health records, and blood and urine samples. RESULTS: Levels of the tubular biomarker beta2-microglobulin increased significantly when the levels of MT-Ab and urinary cadmium were elevated in male and female subjects; in contrast, the levels of urinary albumin, a glomerular biomarker, did not display such a pattern. After adjusting for potential confounding covariates, logistic regression showed that the odds ratios (ORs) of tubular dysfunction increased upon 1) increasing the MT-Ab concentration from a low to high level (OR 5.56 [95% CI 2.25-13.73]) and 2) increasing the level of urinary cadmium from <1 to >or=1 microg/g creatinine (3.34 [1.17-9.53]); the OR of patients currently smoking was 3.51 (1.14-10.80) relative to that of those who had never smoked. CONCLUSIONS: This study proves that the presence of MT-Ab can potentiate tubular dysfunction among diabetic subjects and that patients with high MT-Ab levels are more prone to development of tubular damage.

    • Amorphous iron-calcium phosphate (Fe-ACP) plays a vital role in the mechanical properties of teeth of some rodents, which are very hard, but its formation process and synthetic route remain unknown. Here, the synthesis and characterization of an iron-bearing amorphous calcium phosphate in the presence of ammonium iron citrate (AIC) are reported. The iron is distributed homogeneously on the nanometer scale in the resulting particles. The prepared Fe-ACP particles can be highly stable in aqueous media, including water, simulated body fluid, and acetate buffer solution (pH 4). In vitro study demonstrates that these particles have good biocompatibility and osteogenic properties. Subsequently, Spark Plasma Sintering (SPS) is utilized to consolidate the initial Fe-ACP powders. The results show that the hardness of the ceramics increases with the increase of iron content, but an excess of iron leads to a rapid decline in hardness. Calcium iron phosphate ceramics with a hardness of 4 GPa can be achieved, which is higher than that of human enamel. Furthermore, the ceramics composed of iron-calcium phosphates show enhanced acid resistance. This study provides a novel route to prepare Fe-ACP, and presents the potential role of Fe-ACP in biomineralization and as starting material to fabricate acid-resistant high-performance bioceramics.

    • Technologies and digital infrastructures can provide new opportunities for charities to rethink organizational control or potentials for justice. Furthermore, technologies can themselves generate new infrastructure to challenge existing structures. In order for newly adopted technologies to be useful for service delivery and not to simply reinforce existing power imbalances to amplify exclusion, they need to be embedded in the service, to follow “just sustainabilities” where attention is drawn to systemic rather than individual concerns, and be accessible for all. National Ugly Mugs (NUM) is a charity whose aim is to end violence against sex workers. They do this by providing access to justice and protection for sex workers in the UK through a digitally facilitated peer-alerting system, training police and other service providers, and through consensual sharing of intelligence with police forces. This chapter aims to look beyond digital technologies that are employed in sex work support services as “silver bullets” to solving complex socio-cultural, socio-ethical, and socio-technical problems.

    • CdTe/ZnSe core/shell quantum dots were directly synthesized in an aqueous condition by heating a mixed solution of ZnCl2, NaHSe and CdTe QDs in the presence of mercaptosuccinic acid as a stabilizer. By controlling the size and composition, the CdTe/ZnSe QDs with emission wavelength ranging from 540 to 630 nm, high quantum yield (44%) and narrow full width at half maximum (FWHM) could be obtained. Characterization results with HRTEM, XRD and EDX have shown that the synthesized CdTe/ZnSe ON have good monodispersity and a nice crystal structure, and exhibited better stability and less cytotoxicity as compared with CdTe QDs. Furthermore, luminescent QD-IgG bioprobes were produced to detect the breast cancer marker Her2 on the surface of fixed MCF-7 cancer cells for their optical imaging.

    • Prospects for a lossless negative dielectric constant material for optical devices are studied. Simulations show that with sufficient gain, a mixture of two semiconductor quantum dots (QDs) can produce an effective dielectric constant that is lossless and negative. This permits, in concept, arbitrarily small scaling of the optical mode volume, a major goal in the field of nanophotonics. The proposed implementation of a lossless negative dielectric constant material based on colloidal QDs opens a tractable path.

    • Claudius are the major transmembrane protein components of tight junctions in human endothelia and epithelia. Tissue-specific expression of claudin members suggests that this protein family is not only essential for sustaining the role of tight junctions in cell permeability control but also vital in organizing cell contact signaling by protein protein interactions. How this protein family is collectively processed and regulated is key to understanding the role of junctional proteins in preserving cell identity and tissue integrity. The focus of this review is to first provide a brief overview of the functional context, on the basis of the extensive body of claudin biology research that has been thoroughly reviewed, for endogenous human claudin members and then ascertain existing and future proteomics techniques that may be applicable to systematically characterizing the chemical forms and interacting protein partners of this protein family in human. The ability to elucidate claudin-based signaling networks may provide new insight into cell development and differentiation programs that are crucial to tissue stability and manipulation.

  • The detailed chemical composition of the terrestrial planet host Kepler-10

    • Chemical abundance studies of the Sun and solar twins have demonstrated that the solar composition of refractory elements is depleted when compared to volatile elements, which could be due to the formation of terrestrial planets. In order to further examine this scenario, we conducted a line-by-line differential chemical abundance analysis of the terrestrial planet host Kepler-10 and 14 of its stellar twins. Stellar parameters and elemental abundances of Kepler-10 and its stellar twins were obtained with very high precision using a strictly differential analysis of high quality Canada-France-Hawaii Telescope, Hobby-Eberly Telescope and Magellan spectra. When compared to the majority of thick disc twins, Kepler-10 shows a depletion in the refractory elements relative to the volatile elements, which could be due to the formation of terrestrial planets in the Kepler-10 system. The average abundance pattern corresponds to similar to 13 Earth masses, while the two known planets in Kepler-10 system have a combined similar to 20 Earth masses. For two of the eight thick disc twins, however, no depletion patterns are found. Although our results demonstrate that several factors [e.g. planet signature, stellar age, stellar birth location and Galactic chemical evolution (GCE)] could lead to or affect abundance trends with condensation temperature, we find that the trends give further support for the planetary signature hypothesis.

    • Boswellic acids are the compounds isolated from the gum resin of Boswellia serrata and have been used for the treatment of inflammatory diseases for many years in the countries of the east. Recently, a few studies showed that the acids may have anti-cancer effect on leukemia and brain tumours. We investigated the apoptotic and anti-proliferative effects of two types of boswellic acids, keto-beta-boswellic acid and acetyl-keto-beta-boswellic acid, on liver cancer Hep G2 cells. After treating the cells with the boswellic acids, cell proliferation, DNA synthesis, and apoptosis were analysed. The activities of caspase-3, -8 and -9 were assayed. To explore the apoptotic pathway, specific caspase inhibitors were employed. It was found that boswellic acids decreased cell viability and [3H]thymidine incorporation, checked the cells in the G1 phase, and increased percentage of sub-G1. Boswellic acids strongly induced apoptosis accompanied by activation of caspase-3, -8 and -9. The apoptosis was blocked completely by caspase-8 or caspase-3 inhibitor, but inhibited partly by caspase-9 inhibitor. However, these caspase inhibitors did not show any effect on the alternations of cell viability caused by boswellic acids. In conclusion, boswellic acids have anti-proliferation and anti-cancer effects on Hep G2 cells. The apoptotic effect is mediated by a pathway dependent on caspase-8 activation. The acids may be a promising drug for the chemoprevention of liver cancer.

    • The expanding use of electrical power generated from wind turbines and solar photovoltaic plants is enabled by the decreasing cost of electrical energy from sun and wind. With the advent of electrical energy from the intermittent solar and wind energy resources comes the requirement that electricity must be stored for use over time. The huge demand for materials for such storage systems will require a considerable energy input in extraction, processing and materials formulation, and new and sustainable electrochemical systems need to be developed. Storing electrical energy in bio based batteries is one of the options for handling the rapid expansion of renewable and variable electrical energy generated in wind turbines and in solar photovoltaic systems, from small to large. With projected needs for storage at 300 GWh for the coming decade, there are many niches for new technologies and possibilities. A supply line of materials for energy storage materials could be ultimately based on photosynthesis, in the form of materials derived from plants. Redox activity is possible in lignin, humic acid, and polyphenolic macromolecules, sometimes by electrochemical activation of redox groups.

  • Imbalanced kynurenine pathway in schizophrenia and depression : immunological and genetic aspects

    • Kynurenic acid (KYNA), an end metabolite of the kynurenine pathway along tryptophan degradation, has gained increasing interest in the pathophysiology of psychiatric disorders. Elevated levels of KYNA have been found in the cerebrospinal fluid (CSF) and in the postmortem brain of patients with schizophrenia and bipolar disorder with psychotic symptoms. In contrast, reduced levels of KYNA are associated with depressive symptoms. Rapidly emerging evidence points to the involvement of brain immune activation in psychiatric disorders and to the kynurenine pathway as a causal link between brain immune activation and psychiatric disorders. The aim of present thesis was to investigate the role of the kynurenine pathway in the pathophysiology of schizophrenia and depression in experimental settings. The results show that an infection with neurotropic influenza A virus or a transient elevation of brain KYNA levels in neonatal mice enhanced the sensitivity of d-amphetamine-induced increase in locomotor activity in adulthood. Neonatal elevation of KYNA also impaired prepulse inhibition and working memory in adulthood. These long-lasting behavioral alterations suggest that the kynurenine pathway as a causal link between early-life infection and the development of neuropsychiatric disturbances in adulthood. Reduced KYNA levels were detected in the prefrontal cortex, but not in the frontal cortex, hippocampus, striatum or cerebellum, of Flinders Sensitive Line (FSL) rats, an animal model of depression, compared with their controls, the Flinders Resistant Line (FRL) rats. Inhibition of Kynurenine 3-monooxygenase (KMO) shunts the kynurenine pathway towards enhanced synthesis of KYNA. Mice with a targeted deletion of KMO exhibited impairments in contextual memory and social interaction, potentiated horizontal activity following damphetamine-induced increase in locomotor activity as well as increased anxiety-like behaviors. In addition, genome-wide differential gene expression analyses identified alterations regarding schizophrenia- and psychosis-related genes in these mice. Kynurenine aminotransferase (KAT) II is identified as the main enzyme responsible for most of brain KYNA production. Nevertheless, KAT II KO mice receiving kynurenine injection or repeated injections of Lipopolysaccharide (LPS) exhibited increased concentrations of brain KYNA. Taken together, these results give strong experimental support for the connection between immune activation and KYNA in schizophrenia and suggest that low brain KYNA could be of importance for the depressive-like behaviors observed in FSL rats.

    • The prediction of vehicle trajectory is an important step in safe driving. The relative positions or speeds of vehicles affect the interactions between them, which in turn produce predictions of trajectories. Through the trajectory prediction of each vehicle to form a complete safe driving environment. This paper will be divided into two parts for vehicle track, namely manual setting and machine learning. The advantage of manual setting is that it is convenient to observe the characteristics and then make other vehicles follow the corresponding behaviour. However, the disadvantage is that the characteristics that can be observed are incomplete, which makes the experimental error relatively large. The advantage of machine learning is that through previous experience to make all vehicles change in the corresponding situation. Through previous experience to make all vehicles change in the corresponding situation. An effective extraction and addition of the original data is helpful to reduce the final error.   • A simple, cross-linked collagen tissue substitute for corneal implantation

    • PURPOSE. To develop a simple corneal substitute from crosslinked collagen. METHODS. Porcine type I collagen (10%, pH 5), was mixed with 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and N-hydroxysuccinimide (NHS). The final homogenous solution was molded to corneal dimensions, cured, and then implanted into rabbits and minipigs by lamellar keratoplasty. The implants were followed for up to 6 months after surgery. Clinical examinations of the cornea included detailed slit lamp biomicroscopy, in vivo confocal microscopy, topography and esthesiometry for nerve function. Histopathologic examinations were also performed on rabbit corneas harvested after 6 months. RESULTS. Cross-linked collagen (refractive index, 1.35) had optical clarity superior to human corneas. Implanted into rabbit and porcine corneas, only 1 of 24 of the surgical corneas showed a slight haze at 6 months after surgery. All other implants showed no adverse reactions and remained optically clear. Topography showed a smooth surface and a profile similar to that of the contralateral nonsurgical eye. The implanted matrices promoted regeneration of corneal cells, tear film, and nerves. Touch sensitivity was restored, indicating some restoration of function. The corneas with implants showed no significant loss of thickness and demonstrated stable host- graft integration. CONCLUSIONS. Collagen can be adequately stabilized, using water soluble carbodiimides as protein cross-linking reagents, in the fabrication of corneal matrix substitutes for implantation. The simple cross-linking methodology would allow for easy fabrication of matrices for transplantation in centers where there is a shortage of corneas, or where there is need for temporary patches to repair perforations in emergency situations.     • This study reports China’s macroeconomic trends in downward pressures and discusses the effects of the ‘micro stimulus’ on the growth performance of Chinese economy. It appears that economic growth rebounds significantly in the short term every time ‘micro stimulus’ is applied, but the economy slows down again once the stimulus dwindles. China’s economic growth thus exhibits a pattern of significant ‘stimulus-dependence’. When facing economic downturn, China has only resorted to stimulus policy to sustain growth. Not surprisingly, our findings indicate that ‘micro stimulus’ cannot realize the strategic intent of growth stabilization and structural adjustment, and may even lead to more structural chaos. One problem that can be attributed to the near-sighted strategy is the worsening productivity performance since the financial crisis of 2008. Therefore both improvement in social security systems and social programs designed for maintaining long run growth are needed in order to improve productivity performance on the one hand and to facilitate structural adjustments on the other. © 2015 The Chinese Economic Association – UK.

    • Scheduling is a critical means for providing on-demand manufacturing services in cloud manufacturing. Multi-agent technologies provide an effective approach for addressing scheduling issues in cloud manufacturing, which, however, have rarely been used for solving the issue. This paper addresses scheduling issues in cloud manufacturing using multi-agent technologies. A multi-agent architecture for scheduling in cloud manufacturing is proposed firstly. Then, a corresponding multi-agent model is presented, which incorporates many-to-many negotiations based on an extended contract net protocol and takes into account dynamic task arrivals. Simulation results indicate the feasibility of the model and approach proposed.

    • Purpose: The purpose of this study is to develop an optimization method for charging plans with the implementation of time-of-day (TOD) electricity tariff, to reduce electricity bill. Design/methodology/approach: Two optimization models for charging plans respectively with fixed and stochastic trip travel times are developed, to minimize the electricity costs of daily operation of an electric bus. The charging time is taken as the optimization variable. The TOD electricity tariff is considered, and the energy consumption model is developed based on real operation data. An optimal charging plan provides charging times at bus idle times in operation hours during the whole day (charging time is 0 if the bus is not get charged at idle time) which ensure the regular operation of every trip served by this bus. Findings: The electricity costs of the bus route can be reduced by applying the optimal charging plans. Originality/value: This paper produces a viable option for transit agencies to reduce their operation costs.

  • Optimism and survivorship after esophageal cancer surgery

    • Dispositional optimism is a personality trait that represents generally favorable expectations about the future. The main aim of this thesis was to examine whether higher dispositional optimism was related to better subjectively reported outcomes and better objectively measured outcomes. We used a self-reported scale, Life Orientation Test-Revised (LOT-R), to measure dispositional optimism in this thesis, with a higher LOT-R sum score representing higher dispositional optimism. Data for the four studies included in this thesis were from a prospective, Swedish nationwide, and population-based cohort entitled “Oesophageal Surgery on Cancer patients - Adaptation and Recovery (OSCAR)”. Study I and II included 192 patients who underwent surgery for esophageal cancer during January 1, 2013 and February 28, 2018 in Sweden. Patients self-reported their dispositional optimism level at 1 year after surgery. At 1, 1.5, and 2 years after esophagectomy, patients repeatedly self-reported their psychological status and health related quality of life (HRQL). Latent growth curve model and linear mixed effects model were used in these two studies. We found that higher dispositional optimism predicted a lower risk of reporting clinically significant psychological distress (anxiety and/or depression) in study I. The odds ratio of reporting clinically significant psychological distress was 0.56 (95% confidence interval 0.40 to 0.79) for one unit increase in the LOT-R sum score. In Study II, we categorized patients into four subgroups with very high, moderately high, moderately low, and very low dispositional optimism based on the quartile of the LOT-R sum score. We found that patients with very high and moderately high dispositional optimism reported better HRQL in several aspects compared to patients with lower dispositional optimism. Study III included 180 patients who underwent surgery for esophageal cancer during January 1, 2013 and February 28, 2018 in Sweden. Patients self-reported their psychological status and HRQL repeatedly at 1, 1.5, and 2 years after esophagectomy. We used fixed effects regression model with adjustment for all time-invariant covariates and observed time-varying confounders, and found that psychological distress was associated with worse HRQL in several aspects. Study IV included 335 patients who underwent surgery for esophageal cancer between January 1, 2013 and December 31, 2019 in Sweden. Patients were followed up until the date of death or until December 31, 2020, whichever occurred first. Cox proportional hazards regression was used. We found that among patients with early and intermediate pathological tumor stages (Tis-II) and with follow-up period before the COVID-19 pandemic, the adjusted hazard ratio for all-cause mortality was 0.89 (95% confidence interval 0.81 to 0.98) for one unit increase in the LOT-R sum score. However, this association was not statistically significant for patients with tumor pathologically staged III-IV (hazard ratio 0.99, 95% confidence interval 0.92 to 1.07) and/or during the COVID-19 pandemic (hazard ratio 1.08, 95% confidence interval 0.94 to 1.25). In conclusion, before the COVID-19 pandemic, higher dispositional optimism was associated with less self-reported clinically significant psychological distress and better HRQL; moreover, among patients with early and intermediate pathological tumor stages, higher dispositional optimism also predicted better overall survival. Measuring dispositional optimism may help identify vulnerable patients with potentially worse prognosis after surgery for esophageal cancer, thus contributing to the development of more tailored and timely interventions to improve postoperative survivorship.

  • A ring-like accretion structure in M87 connecting its black hole and jet

    • The nearby radio galaxy M87 is a prime target for studying black hole accretion and jet formation1,2. Event Horizon Telescope observations of M87 in 2017, at a wavelength of 1.3 mm, revealed a ring-like structure, which was interpreted as gravitationally lensed emission around a central black hole3. Here we report images of M87 obtained in 2018, at a wavelength of 3.5 mm, showing that the compact radio core is spatially resolved. High-resolution imaging shows a ring-like structure of [Formula: see text] Schwarzschild radii in diameter, approximately 50% larger than that seen at 1.3 mm. The outer edge at 3.5 mm is also larger than that at 1.3 mm. This larger and thicker ring indicates a substantial contribution from the accretion flow with absorption effects, in addition to the gravitationally lensed ring-like emission. The images show that the edge-brightened jet connects to the accretion flow of the black hole. Close to the black hole, the emission profile of the jet-launching region is wider than the expected profile of a black-hole-driven jet, suggesting the possible presence of a wind associated with the accretion flow.

    • The electronic structure of GaAs (100) grown by low-temperature molecular beam epitaxy was investigated by means of photoemission spectroscopy. Slight differences are found in the valence band spectra of GaAs layers grown at different As-2/Ga flux ratios. Analysis of As 3d core level spectra does not indicate qualitative differences in respect to high temperature grown GaAs (100) layers. The Ga 3d core level spectra include a new component in comparison to the spectra of high temperature grown GaAs.     • The development of photosensitive slurries for additive manufacturing has attracted great interest due to their correlation with the final properties of the fabricated parts. This paper focus on the printing quality control in digital light processing (DLP) 3D printing of advanced ceramics. Systematic experiments were performed to assess the effects of the exposure time and printing angle on the three-point bending strength of the fabricated samples. The exposure time affected the bending strength of the printed zirconia ceramic dramatically. When the weak exposure time is 1 s and the strong exposure time is 13 s, the average bending strength can reach 580 MPa while Weibull modulus can reach 8.84.     • Objective: To ascertain whether a daily nasal spray with physiological saline could prevent symptoms of common cold in a population of otherwise healthy adults.Material and Methods: This was study involving 10 weeks of daily use of a nasal saline spray and 10 weeks of only recording symptoms. Young adults eligible for military service at an army barrack in Boden, Sweden were invited to participate in the study and 108 healthy conscripts aged approximate to 20 years agreed to do so. Data were recorded by the participants in a diary at home. In the diary the participants noted symptoms such as rhinitis, blocked nose, cough, fever and sore throat (pharyngeal pain). They also recorded inability to perform their duties due to the symptoms, and any medication or antibiotics necessitated by upper respiratory tract infection.Results: A total of 69 subjects completed the 20-week diary period. For 60 of them, compliance during the spray period exceeded 60% and their data were used in the statistical calculations. During the spray period the number of days with nasal secretion and/or blocked nose (mean 6.4 days) was significantly (p= 0.027) lower than that during the observation period (mean 11 days). Furthermore, the participants had a mean of 0.7 episodes of upper respiratory tract infection during the spray period, compared with 1.0 episodes during the observation period (p= 0.05).Conclusion: A daily nasal spray with saline can prevent nasal symptoms of common cold in a population of otherwise healthy adults.

    • This paper investigates the efficiency of cooperative transmission when it is applied in wireless multihop networks. We consider regular linear networks and derive the achievable rate-delay tradeoff when selective relaying through a single relay node is used in each hop. We show that relaying achieves significant gain particularly in the high throughput - high delay regime.
