  • The arthropod community of Scots pine (Pinus sylvestris L.) canopies in Norway

    • We summarise the findings of arthropods collected by fogging the canopy of 24 pine trees in two sites in Eastern and Western Norway. From the samples, taken in 1998 and in 1999, almost 30,000 specimens were determined to 512 species, with Diptera being most species rich (210 species), followed by Coleoptera (76 species) and Araneae (49 species). Of the 96 new species records, nine were new to science (5 Diptera and 4 Oribatida), two were new to the European, three to the Scandinavian and 82 to the Norwegian faunas. The paper demonstrates the need for detailed faunistical inventories of European forests.

    • DNA sequence and annotation of the entire human chromosome 7, encompassing nearly 158 million nucleotides of DNA and 1917 gene structures, are presented. To generate a higher order description, additional structural features such as imprinted genes, fragile sites, and segmental duplications were integrated at the level of the DNA sequence with medical genetic data, including 440 chromosome rearrangement breakpoints associated with disease. This approach enabled the discovery of candidate genes for developmental diseases including autism.

    • BACKGROUND: Most patients who have been surgically treated for secondary hyperparathyroidism (HPT) harbor at least one pathological parathyroid gland with a tumor of monoclonal origin. OBJECTIVE: To elucidate the underlying genetic mechanisms behind secondary HPT, by studying a panel of such tumors for numerical alterations. METHODS: Sixteen parathyroid glands from eight patients (median age 58 years, range 31-74 years), were screened for numerical chromosomal imbalances, using comparative genomic hybridization (CGH). Mutation analysis of the multiple endocrine neoplasia type 1 gene (MEN1) was also performed by sequencing of the coding region. RESULTS: The results show that gross chromosomal alterations occur rarely in secondary HPT. In one of the three glands analyzed from one patient, a complete loss of chromosome 11 was detected. This gland also had an inactivating nonsense mutation, E469X, of the MEN1 gene. The mutation was present neither in the other two glands, nor in the constitutional tissue of the same patient, thus confirming its somatic origin. CONCLUSIONS: The relative lack of numerical chromosomal alterations would suggest that more discrete genetic alterations are responsible for the monoclonal growth in the majority of cases of secondary HPT. Furthermore, somatic inactivation of the MEN1 tumor suppressor gene contributes to the tumorigenesis in a small proportion of the cases.

    • Excited states of Ag-99 were populated via the Cr-50 + Ni-58 (261 MeV) reaction using the NORDBALL detector array equipped with charged-particle and neutron. detector systems for reaction channel separation. On the basis of the measured gammagamma-coincidence relations and angular distribution ratios a significantly extended level scheme has been constructed up to E-x similar to 7.8 MeV and I = 35/2. The experimental results were described within the framework of the shell model. Candidates for states fully aligned in the pig(9/2)(-3)nu(d(5/2),g(7/2))(2) valence configuration space were found at 4109 and 6265 keV.

    • We report here the identification of a gene associated with the hyperparathyroidism-jaw tumor (HPT-JT) syndrome. A single locus associated with HPT-JT (HRPT2) was previously mapped to chromosomal region 1q25-q32. We refined this region to a critical interval of 12 cM by genotyping in 26 affected kindreds. Using a positional candidate approach, we identified thirteen different heterozygous, germline, inactivating mutations in a single gene in fourteen families with HPT-JT. The proposed role of HRPT2 as a tumor suppressor was supported by mutation screening in 48 parathyroid adenomas with cystic features, which identified three somatic inactivating mutations, all located in exon 1. None of these mutations were detected in normal controls, and all were predicted to cause deficient or impaired protein function. HRPT2 is a ubiquitously expressed, evolutionarily conserved gene encoding a predicted protein of 531 amino acids, for which we propose the name parafibromin. Our findings suggest that HRPT2 is a tumor-suppressor gene, the inactivation of which is directly involved in predisposition to HPT-JT and in development of some sporadic parathyroid tumors.

    • The electrochemistry of the ligninolytic redox enzymes, which include lignin peroxidase, manganese peroxidase and laccase and possibly also cellobiose dehydrogenase, is reviewed and discussed in conjunction with their basic biochemical characteristics. It is shown that long-range electron transfer between these enzymes and electrodes can be established and their ability to degrade lignin through a direct electron transfer mechanism is discussed.

    • High-spin states of the nucleus Lu-163 have been populated through the fusion-evaporation reaction La-139(Si-29, 5n) with a beam energy of 157 MeV. In addition to the two lowest excited triaxial strongly deformed (TSD) bands, recently interpreted as one- and two-phonon wobbling excitations, a third excited TSD band has been firmly established decaying to the yrast TSD band. The assignment of this band as a three-quasiparticle band shows together with the normal deformed (ND) level scheme the presence not only of shape coexistence between ND and TSD structures, but also an interplay of wobbling and quasiparticle excitations in the triaxial strongly deformed potential well of Lu-163.

    • High-spin states in Nd-138 have been investigated with the reaction Zr-94(Ca-48,4n), using the GASP array. A new rotational band has been identified, which has close similarities with the well known highly deformed bands of the lighter Nd isotopes, and is therefore interpreted as built on the second minimum of the nuclear potential energy surface. From comparison with calculations a configuration is suggested which involves two neutrons in the i(13/2) deformation driving orbital.

    • Treatment with alkali, particularly overliming, has been widely used as a method for the detoxification of lignocellulose hydrolysates prior to ethanolic fermentation. However, the mechanisms behind the detoxification effect and the influence of the choice of cation have not been well understood. In this study, a dilute acid hydrolysate of spruce and an inhibitor cocktail consisting of six known inhibitors were used to investigate different alkali detoxification methods. The various treatments included the addition of calcium hydroxide, sodium hydroxide, potassium hydroxide, and ammonia to pH 10.0 and subsequent adjustment of the pH to 5.5 with either sulfuric or hydrochloric acid as well as treatment with the corresponding amounts of calcium, sodium, and potassium as sulfate or chloride salts at pH 5.5. An RP-HPLC method was developed for the separation of 18 different inhibitors in the hydrolysate, including furaldehydes and phenolics. Detection and quantification were carded out by means of UV, DAD, and ESI-MS in negative mode. Treatment of the spruce hydrolysate with alkali resulted in up to similar to40% decrease in the concentration of furaldehydes. The effects on the aromatic compounds were complex. Furthermore, SFE was performed on the precipitate formed during alkali treatment to evaluate the inhibitor content of the precipitate, and the following RP-HPLC analysis implied that potential inhibitors were removed mainly through conversion rather than through filtration of precipitate. Parallel experiments in which sulfuric acid or hydrochloric acid was used for acidification to pH 5.5 after alkali treatment indicated that the choice of anion did not affect the removal of inhibitors. Detoxification with calcium hydroxide and ammonia resulted in better fermentability using Saccharomyces cerevisiae than detoxification with sodium hydroxide. The results from the experiments with the inhibitor cocktail indicated that the positive effects of alkali treatment are difficult to explain by removal of the inhibitors only and that possible stimulatory effects on the fermenting organism warrant further attention.

    • This work describes a novel approach to detoxify lignocellulosic hydrolysates and facilitate the analysis of inhibitory compounds, namely supercritical fluid extraction (SFE). The efficiency of the fermentation of lignocellulosic dilute-acid hydrolysates depends upon the composition of the hydrolysate and the organism used. Furthermore, it has been shown that inhibitors in the hydrolysate reduce the fermentation yield. This knowledge has given rise to the need to identify and remove the inhibiting compounds. Sample clean-up or work-up steps, to provide a clean and concentrated sample for the analytical system, facilitate the characterization of inhibitors, or indeed any compound in the hydrolysates. Removal of inhibitors was performed with countercurrent flow supercritical fluid extraction of liquid hydrolysates. Three different groups of inhibitors (furan derivatives, phenolic compounds, and aliphatic acids) and sugars were subsequently analyzed in the hydrolysate, extracted hydrolysate, and extract. The effect of the SFE treatment was examined with respect to fermentability with Saccharomyces cerevisiae. Not only did the extraction provide a clean and concentrated sample (extract) for analysis, but also a hydrolysate with increased fermentability as well as lower concentrations of inhibitors such as phenolics and furan derivatives.   • Stable triaxiality at the highest spins in 138Nd and 139Nd

    • The nuclei 138Nd and 139Nd have been studied at very high spins via the 48Ca+94Zr reaction. Several new rotational bands were observed, four in 138Nd and two in 139Nd. The J(2) moments of inertia calculated from the observed γ-ray energies are very small and almost constant, indicating that these bands are triaxial. Cranked Nilsson-Strutinsky calculations reproduce the general behavior of the bands, supporting this interpretation and suggesting an approximately constant γ value of ∼ + 35° over a large spin range up to the highest observed spins. These bands and a few similar bands in other nuclei of the N≈80 region are a unique example of almost undisturbed triaxial bands.

    • The laser-probing method for lifetime measurements of metastable levels, performed by applying the Fast Ion Beam Laser (FIBLAS) method to ions stored in a storage ring, has been developed by the Stockholm group. Recently, we have applied this method to lifetime measurements of close lying metastable levels. In this paper we discuss experimental studies of ions with complex structure and present the first experimentally obtained lifetimes of selected metastable levels in complex systems as Fe+, Eu+ and La+.

    • High-spin states in Lu-165 were populated in the La-139(Si-30, 4n) reaction at a beam energy of 152 MeV and gamma-ray coincidences were measured with the EUROBALL spectrometer array. Nine new rotational bands were discovered, known band structures were considerably extended and many inter-band transitions were found. Structures with normal deformation coexist with bands associated with the strongly deformed triaxial energy minima found in calculations. Three of these triaxial bands form a family of wobbling excitations with phonon quanta n(w) = 0, 1 and 2. The wobbling mode is a unique signature of nuclear triaxiality. Configuration assignments are discussed for the observed band structures. An exchange of configuration between two of the new bands due to mixing is observed, resulting in different signature partnerships at low and high spins.

    • High-spin states in Lu-165 have been investigated by in-beam gamma-ray coincidence spectroscopy using the EUROBALL spectrometer array. Two new excited rotational bands have been discovered with features similar to a previously known triaxial superdeformed band in that nucleus. Comparison of the decay pattern of these bands, in particular the unusually large E2 transition strength from the first excited to the yrast superdeformed band, to theoretical calculations shows that they belong to a family of wobbling excitations with phonon numbers n(w) = 0, 1 and 2. These results, together with evidence for nuclear wobbling in the neighbouring isotopes Lu-163 and Lu-167, firmly establish this mode of excitation in the A = 165 mass region. The observation of wobbling is a unique signature of stable nuclear triaxiality. (C) 2002 Elsevier Science B.V. All rights reserved.
