    • The innate immune system and in particular the pattern-recognition receptors Toll-like receptors have recently been linked to atherosclerosis. Consequently, inhibition of various signaling molecules downstream of the Toll-like receptors has been tested as a strategy to prevent progression of atherosclerosis. Receptor-interacting protein 2 (Rip2) is a serine/threonine kinase that is involved in multiple nuclear factor-κB (NFκB) activation pathways, including Toll-like receptors, and is therefore an interesting potential target for pharmaceutical intervention.

  • Migration and autism-spectrum disorder : population-based study

    • Background: Migration has been implicated as a risk factor for autism, but evidence is limited and inconsistent. Aims: To investigate the relationship between parental migration status and risk of autism spectrum disorder, taking into consideration the importance of region of origin, timing of migration and possible discrepancies in associations between autism subtypes. Method: Record-linkage study within the total child population of Stockholm County between 2001 and 2007. Individuals with high- and low-functioning autism were defined as having autism spectrum disorder with and without comorbid intellectual disability, and ascertained via health and habilitation service registers. Results: In total, 4952 individuals with autism spectrum disorder were identified, comprising 2855 children with high-functioning autism and 2097 children with low-functioning autism. Children of migrant parents were at increased risk of low-functioning autism (odds ratio (OR) = 1.5, 95% CI 1.3-1.7); this risk was highest when parents migrated from regions with a low human development index, and peaked when migration occurred around pregnancy (OR=2.3, 95% CI 1.7-3.0). A decreased risk of high-functioning autism was observed in children of migrant parents, regardless of area of origin or timing of migration. Parental age, income or obstetric complications did not fully explain any of these associations. Conclusions: Environmental factors associated with migration may contribute to the development of autism presenting with comorbid intellectual disability, especially when acting in utero. High- and low-functioning autism may have partly different aetiologies, and should be studied separately.

    • ntroduction Simulation has become an established method for education and training of Emergency Medical Services personnel in different skills such as advanced airway techniques, also in evaluation and initial care of stroke patients and in pre - hospital trauma. Simula tion can be a useful method to improve learning. To our knowledge, there are only a few studies that relate simulation to the effect on clinical skills. The aim of the present study was to investigate the effect of simulation on the clinical competence in a pre - hospital trauma care course for ambulance nurses. Methods The study was performed using a before - after design. Seventeen ambulance nurse students who participated in a trauma care course were evaluated in two simulated trauma cases. All subjects had passed the initial theoretical part of the course. The pre - test was performed in the beginning of the following part of the course involving simulation and the post - test at the end of the course. The analysis was performed by assessment of performance, as seen on video - tapes from the pre - tests and the post - tests. A validated instrument was used to determine the level of student’ s clinical competence. Paired t - test was used to confirm differences between the pre - test and post - test results . Results There was a significant increase in the over - all results for the post - test, with a difference of 1.12 points (t=4.642, df=16, p= 0.001) Situation Awareness, Patient Assessment and Decision Making showed the most pronounced improvements. Conclusion The results imply t hat simulation in addition to traditional theoretical education improves the clinical competence of the students, in comparison to traditional education and training without any significant amount of simulation.

    • En mycket viktig faktor vid inventering av vägnätets tillstånd med hjälp av mätbilar är att med tillräckligt bra noggrannhet kunna positionsbestämma mätdata.I mars 2012 lades korrektionstjänsten EPOS, som hittills använts för att ge en förbättrad positionering, ner. I och med detta har VTI på uppdrag av Trafikverket utvärderat om den kostnadsfria korrektionstjänsten EGNOS, som drivs av ESA, är möjlig att använda istället. Svaret är ja. Inga systematiska skillnader i positionsangivelser har kunnat observeras då den gamla och nya korrektionstjänsten använts.

    • Denna VTI-studie konstaterar att det går att mäta ojämnheter på vinterväg på samma sätt som på barmarksväg. Både ojämnheter och buller vid låga frekvenser ökar vid visst vinterväglag och är kopplade till hastighet. De mätningar som utförts inom projektets ramar visar att det går utmärkt att utföra mätningar på en snöklädd yta. Med vinterväglag menas här en snöklädd väg cirka ett dygn efter ett kraftigt snöfall. Baserat på de vägar som mättes visar studien att ojämnheter i våglängdsintervallet 0,05 till 1 meter påverkas allra mest av vinterväglaget. Ojämnheterna ökar cirka 5 gånger i medeltal för hela testslingan från barmarks- till vinterförhållanden. Det går även att se att ytans struktur, som beskrivs av de kortaste våglängderna som undersöktes, <10 mm, blir slätare på den snöklädda ytan vilket var en av hypoteserna och som indikerar att mätningen är pålitlig. Kopplingen mellan buller och ojämnheter är relaterade till hastighet. Den största ljudskillnaden mellan vinter- och sommarväg var vid låga frekvenser. Det uppfattade ljudet i bilen var då mellan 3 och 6 decibel högre för körningen på vinterväg. I de mer högfrekventa områdena var skillnaden i ljudnivå tvärtom, lägre för vintervägen än för sommarvägen och en trolig orsak till detta kan vara att ljudet absorberas av snön.

Databasen ULF innehåller statistisk information om hur levnadsförhållanden i Sverige ser ut bland olika grupper. Områden som behandlas i är bland annat boende, ekonomi, hälsa, fritid, sociala relationer, sysselsättning och trygghet. Uppgifterna som publiceras i samlas in genom telefonintervjuer med ett urval av Sveriges befolkning över 16 år.

  • Constructing communities : The establishment and demographic development of sawmill communities in the Sundsvall district, 1850-1890

    • This dissertation studies the establishment and demographic development of the sawmill communities that emerged in the Sundsvall district during the latter half of the 19th century.  The intention is to highlight the importance of the sawmill communities and their resident populations by discussing community construction from a demographic perspective as well as socially and symbolically. Based on church registers, this is a longitudinal study that includes information from 31 individual sawmill communities. This study has shown that the establishment and demographic development of the sawmill communities was not an instant process that necessarily followed the construction of the sawmill industries. The prerequisites of the geographical locations and year of establishment influenced population development, but the speed and size of the settlements were individual to each mill site. More prosperous times for the industry during the 1870s resulted in that migration increased consequently leading to quickly populated communities and larger registered core populations in residence. Migration to the sawmill communities from within the parishes was infrequent and the geographical backgrounds revealed that an extremely small proportion of the populations had been born within the district, implying a migratory hesitation among locally born. The sawmill populations were male-dominated due to the large groups of temporary workers inhabiting the communities, although, adult males barely made up one-third of the registered populations. The largest demographic group was children aged 0-14 years. The strong presence of children and high proportions of married individuals suggests that the sawmill communities were family oriented communities, more so than non-sawmill areas. Long-time settled families had usually formed kinship networks with other residents. This dissertation concludes that while time was important for the development of the sawmill communities, so were the registered populations residing in these communities. Residency would have been key in claiming belonging to the sawmill communities and to be considered as a real sawmill worker. Residency, family and kin therefore contributed to the construction of community structures, geographically, socially and symbolically.

  • Annexin A1 expression in blood mononuclear cells : a potential marker of glucocorticoid activity in patients with coronary artery disease

    • An imbalance between pro- and anti-inflammatory actions is believed to drive progression of atherosclerosis. Annexin A1 (AnxA1) is a key player in resolution of inflammation and a mediator of anti-inflammatory effects of glucocorticoids. Here, we investigated whether expression of AnxA1 in peripheral blood mononuclear cells (PBMCs) was altered in patients with coronary artery disease (CAD) and also related findings to glucocorticoid sensitivity ex vivo.We included 57 patients 6-12 months after acute coronary syndrome (ACS), 10 patients with ACS, and healthy controls. AnxA1 mRNA was measured in PBMCs and AnxA1 protein was assessed in monocytes and lymphocyte subsets by flow cytometry. In post-ACS patients and controls, glucocorticoid sensitivity was determined by measuring inhibitory effects of dexamethasone on LPS46 induced cytokine secretion.AnxA1 mRNA levels in PBMCs were higher in patients compared with controls, although most pronounced in ACS patients. AnxA1 protein was most abundant in the monocyte fraction. ACS patients exhibited the highest levels of cell surface-associated AnxA1 protein while levels in post-ACS patients and controls were similar. Ex vivo assays showed that PBMCs from post-ACS patients were more prone to release IL-6. Glucocorticoid sensitivity correlated with cell surface-associated AnxA1 protein in peripheral monocytes. Dexamethasone also induced upregulation of AnxA1 mRNA.AnxA1 expression in PBMCs is closely associated with glucocorticoid actions and cell surface associated AnxA1 appears to be a marker of glucocorticoid sensitivity. Although still speculative, a “normal” expression of cell surface-associated AnxA1 in post-ACS patients may suggest that glucocorticoid actions in vivo are insufficient to provide adequate anti-inflammatory effects in these patients.

  • Higher expression of annexin A1 in 1 CD56+ than in CD56-T cells : Potential implications for coronary artery disease

    • Background: Increased proportions of circulating proinflammatory CD56+ T cells have been reported in patients with coronary artery disease (CAD). Yet, little is known about regulation of these cells. In the present study, we investigated the expression and potential role of the glucocorticoid-mediated protein annexin A1 (AnxA1) in CD56+ and CD56-T cell subsets, with focus on CAD.Methods and Results: We included totally 52 healthy individuals, 28 patients with acute coronary syndrome (ACS) and 57 patients with a history of ACS. AnxA1 mRNA expression was assessed in peripheral blood mononuclear cells. AnxA1 protein expression (total and cell surface-associated) was measured by whole blood flow cytometry in circulating CD56+ and CD56- T cell subsets. Furthermore, inhibitory effects of dexamethasone and/or recombinant AnxA1 on cytokine secretion by CD56+ and CD56- T cells were explored in vitro. We found that CD56+ T cells (the majority CD8+), expressed higher levels of AnxA1 mRNA and protein than did CD56- T cells. When comparing CAD patients with healthy controls, significantly higher levels of cell surface-associated AnxA1 expression were seen in patients, most pronounced in ACS patients. In vitro, dexamethasone reduced cytokine secretion by CD56+ T cells, whereas AnxA1 alone had no effect, and AnxA1 combined with dexamethasone abolished the dexamethasone-induced suppressive effects.Conclusion: AnxA1 was expressed more abundantly in proinflammatory CD56+ T cells. Patients with ACS exhibited increased levels of cell surface-associated AnxA1, thus indicating increased activation of the AnxA1 pathway. Our data further suggested that AnxA1 might counteract glucocorticoid mediated anti-inflammatory effects in T cells.

We examined whether the distribution and phenotype of CD8(+)CD56(+) T cells differed according to the clinical manifestation of CAD. less thanbrgreater than less thanbrgreater thanMethods: Patients with acute coronary syndrome (ACS, n = 30), stable angina (SA, n = 34) and controls (n = 36) were included. Blood was collected before and up to 12 months after referral for coronary investigation. CD8(+)CD56(+) T cells were assessed by flow cytometry for expression of surface markers, apoptosis, and intracellular expression of cytokines. less thanbrgreater than less thanbrgreater thanResults: The proportions of CD8(+)CD56(+) T cells were significantly higher in both ACS and SA patients compared with controls, and remained so after 3 and 12 months. This was independent of age, sex, systemic inflammation and cytomegalovirus seropositivity. CD8(+)CD56(+) T cells differed from CD8(+)CD56(-) T cells in terms of lower CD28 expression and fewer apoptotic cells. Both CD8(+) T cell subsets were positive for interferon (IFN)-gamma and tumor necrosis factor, although IFN-gamma was significantly more confined to the CD8(+)CD56(+) T cells. less thanbrgreater than less thanbrgreater thanConclusion: The persistent accumulation of CD8(+)CD56(+) T cells in ACS and SA patients share several features with immunological aging. It also contributes to a larger IFN-gamma(+) pool in blood, and may thereby hypothetically drive the atherosclerotic process in a less favorable direction.

    • Abstract Objective. There is a need for a technique allowing studies of human mucosal specimens collected during different clinical conditions. This study elucidates if square wave pulse analysis discriminates between epithelial and transmural electrical resistance and if there is an association with transepithelial permeability of molecular probes. Methods. Mucosae from esophagus (surgical resections: n = 14; endoscopic biopsies: n = 15) and jejunum (n = 12) and Caco-2 cell monolayers were investigated in Ussing chambers. Transmural and epithelial electrical resistance were recorded by the use of standardized current pulses. Permeability was assessed using two fluorescein-labeled probes (weight 376 and 4000 Da). Results. Baseline epithelial electrical resistance was higher in esophageal mucosa (∼280 Ω*cm(2)), than in jejunal (∼10 Ω*cm(2)) and Caco-2 cells (∼140 Ω*cm(2)). The subepithelial contribution to the transmural resistance was higher in jejunal preparations (+88%) and Caco-2 cells (+75%), than in esophageal (+30%). During hypoxia the subepithelial resistance was unchanged, whereas the epithelial resistance decreased significantly in jejunal mucosa and Caco-2 cells. These findings coincided with increased transepithelial probe permeability and signs of disturbed morphology. Esophageal epithelia were resistant to hypoxia. However, exposure to deoxycholic acid and trypsin abolished the esophageal epithelial resistance and increased probe permeability. Endoscopic esophageal biopsies from patients with erosive reflux disease exhibited significantly lower epithelial resistance and higher current than healthy subjects. Conclusion. Square wave pulse analysis in Ussing chambers is suitable for assessment of epithelial electrical resistance that can reflect transepithelial permeability of molecular probes with known size. Moreover, the technique discriminated between healthy and reflux-diseased esophageal mucosal biopsies.

According to aggregated data describingprevalent users compliance to guidelines is fairly high. Objectives:The objective of this study was to investigatechoice and volume (DDD) of substance among incidentusers 75 years and older (75+) of hypnotics (N05C) in Swedenand compare between different county councils andwith incident users below 75 years of age. Methods:Data from the Swedish Prescribed Drug registercovering all dispensed prescriptions in Sweden was analyzedfor incident users of hypnotics (15–44, 45–64, 65–74 and 75+ years of age). Incident users were defined aspeople who filled a prescription (index date) of a hypnotic(N05C) during the period December 2009 - November2010 and had not filled any prescription for a hypnotic duringa wash-out period of 24 months before the index date. Results:Among incident patients 75+ (n= 38,620) zopiclone(52.9%), zolpidem (25.4%), and propiomazine(10.4%) were the most frequently used hypnotics. Therewere however differences between different county councilsin Sweden, with zopiclone ranging from 30 % to 68 %. Theincidence for recommended and appropriate hypnoticsincreased with increasing age, whereas the incidence ofinappropriate hypnotics did not. The prescribed volume ofinappropriate hypnotics was generally larger than the prescribedvolume of recommended hypnotics. Conclusions:The compliance to current guidelines onchoice of hypnotics for the elderly is fairly good, althoughthere is considerable variation between county councils.There are tendencies towards larger mean volumes of inappropriatehypnotics, although further studies are needed toconfirm these findings. Another aspect of quality in treatmentwith sleeping agents is to which extent incident userscontinue their treatment and for how long. This is an issuefor further research.

  • Accessibility in public space as perceived by people with Alzheimer´s disease

    • Most people with dementia remain living at home as long as possible after being diagnosed, and hence their lives also include activities in the public space. The aim of this study was to illuminate experiences of accessibility in public space in people with Alzheimer’s disease. A qualitative grounded theory approach with repeated in-depth interviews was used. The core category, accessibility as a constantly changing experience, was characterized by changes in the relationship between informants and public space. Changes in the relationship took place in activities and use of place and related to familiarity and comfort, individual motives and interests, and planning and protecting. Other changes occurred in places and problematic situations related to everyday technologies, crowded places with high tempo and noise, and change of landmarks. These changes reduced feelings of accessibility and increased difficulties in carrying out activities in public space. These findings may be helpful when providing support, and supporting community living.

    • 25 år av tvärvetenskaplig, internationell forskning visar alltjämt att bemannade skolbibliotek som samspelar nära med undervisningen kan ge högst meningsfulla bidrag till elevers lärande. Det menar sex forskare vid institutionen för Biblioteks- och informationsvetenskap vid Högskolan i Borås. I en lång replik (7/10) i debatten om skolbibliotek, där Gabriel H Sahlgren generöst presenterar färdiga och kategoriska lösningar på svåra och omstridda metodologiska problem inom samhällsvetenskaplig och tvärvetenskaplig forskning, bemöts vårt tidigare inlägg (4/10). Vi noterar att Sahlgren, som professionell opinionsbildare med uppdrag att finna argument för en fri skolmarknad, framhärdar i sin övertygelse om att en enda indisk studie är en bättre indikator på hur skolbibliotek i Sverige kan fungera, än den helhetsbild som övrig forskning presenterar. Skälet för detta är att den indiska studien använder sig av en experimentell metoddesign. Studien ifråga finner inget stöd för att anta att orsakssamband finns mellan förekomsten av skolbibliotek och elevers studieresultat.

    • In spite of the growing interest in cluster initiatives (CIs) as a means of regional development, there are still few studies of CIs that offer an inside perspective. This article takes such an inside perspective, focusing on the internal legitimacy aspects of the formation and evolving processes of CIs. We propose the inclusion of interpartner legitimacy effects in order to better understand the formation and development of CIs. A case study method is applied on a Swedish CI in the tourism industry. Faced with the situation that their region was lagging behind other Swedish regions in the development of tourism, the actors were spurred to promote a CI. Its central purposes were to consolidate the regional tourism industry, strengthen the regional brand and to establish the entire region as a single coherent destination. This paper contributes to the research body of regional development and CIs by showing how different types of interpartner legitimacies hinder and facilitate the CI process.

    • Network interaction by non-business actors for business purposes is receiving increased interest within research (cf. Hadjikhani and Ghauri, 2001) but much remain to further research. A typical example of such network interaction that still is little researched is collaboration in tourism destination networks. The purposes of this paper are to contribute to research on the formation and evolution of relationships within the tourism industry, and to test the interpartner legitimacy effect on network formation and development. The chosen method is a case study of a Swedish network formation initiative. Faced with the situation that their region was lagging behind other Swedish regions in the development of tourism, actors within the Dalarna region were spurred to form a Regional Strategic Network (RSN). Its central purposes were to consolidate the regional tourism industry, to strengthen the name of the region as a brand and to establish the entire region as a single destination.     • Global classification of the human proteins with regards to spatial expression patterns across organs and tissues is important for studies of human biology and disease. Here, we used a quantitative transcriptomics analysis (RNA-Seq) to classify the tissue-specific expression of genes across a representative set of all major human organs and tissues and combined this analysis with antibody- based profiling of the same tissues. To present the data, we launch a new version of the Human Protein Atlas that integrates RNA and protein expression data corresponding to 80% of the human protein-coding genes with access to the primary data for both the RNA and the protein analysis on an individual gene level. We present a classification of all human protein-coding genes with regards to tissue-specificity and spatial expression pattern. The integrative human expression map can be used as a starting point to explore the molecular constituents of the human body.

    • A gene-centric Human Proteome Project has been proposed to characterize the human protein-coding genes in a chromosome-centered manner to understand human biology and disease. Here, we report on the protein evidence for all genes predicted from the genome sequence based on manual annotation from literature (UniProt), antibody-based profiling in cells, tissues and organs and analysis of the transcript profiles using next generation sequencing in human cell lines of different origins. We estimate that there is good evidence for protein existence for 69% (n = 13985) of the human protein-coding genes, while 23% have only evidence on the RNA level and 7% still lack experimental evidence. Analysis of the expression patterns shows few tissue-specific proteins and approximately half of the genes expressed in all the analyzed cells. The status for each gene with regards to protein evidence is visualized in a chromosome-centric manner as part of a new version of the Human Protein Atlas (www.proteinatlas.org).

    • In 2012 and 2013, the Swedish Secretariat for Gender Research publishes a five-part series on Swedish gender studies, in which Gender Studies Education and Pedagogy is the first part. The aim is to highlight and spread knowledge about gender studies to wider circles, both within and outside universities and other higher education institutions. The publication of this series is being led by editors Anna Lundberg (Linköping University) and Ann Werner (Södertörn University), and includes a reference group of representatives from gender studies disciplines in Sweden as well as the authors who have contributed to the publications.The themes for the four following publications are: What the future holds for students of gender studies – a national alumni survey; Gender studies, politics and social responsibility; Academic challenges in writing and thought; Theoretical/methodological contributions from gender studies. It is not yet decided whether the remaining parts will be translated to English.The publications have been selected based on consultations between the series editors, the Swedish Secretariat for Gender Research, the reference group and the authors.     • The purpose of the study was to investigate if adding a second dearomatization step can minimize the concentration of aromatics in low aromatic city diesel. For diesel fuel to be called low aromatics city diesel (MK1 in Sweden), the concentration of aromatics can be 5 % maximum presently. Low aromatic city diesel was reacted with hydrogen gas over a catalyst bed of Pt/Al 2O 3 in a downflow reactor system. This reaction was studied at different pressures and temperatures. The samples were analyzed with a gas chromatograph and a high performance liquid chromatography. It can be seen that a high concentration of low fraction hydrocarbons (gasoline) has been formed at high temperatures. The analysis with the high performance liquid chromatography points at a concentration of aromatics lower than 2 %.

    • A method is described to generate and validate antibodies based on mapping the linear epitopes of a polyclonal antibody followed by sequential epitope-specific capture using synthetic peptides. Polyclonal antibodies directed towards four proteins RBM3, SATB2, ANLN, and CNDP1, potentially involved in human cancers, were selected and antibodies to several non-overlapping epitopes were generated and subsequently validated by Western blot, immunohistochemistry, and immunofluorescence. For all four proteins, a dramatic difference in functionality could be observed for these monospecific antibodies directed to the different epitopes. In each case, at least one antibody was obtained with full functionality across all applications, while other epitope-specific fractions showed no or little functionality. These results present a path forward to use the mapped binding sites of polyclonal antibodies to generate epitope-specific antibodies, providing an attractive approach for large-scale efforts to characterize the human proteome by antibodies.

    • BACKGROUND: Inflammation may play an important role in type 2 diabetes. It has been proposed that dietary strategies can modulate inflammatory activity.METHODS: We investigated the effects of diet on inflammation in type 2 diabetes by comparing a traditional low-fat diet (LFD) with a low-carbohydrate diet (LCD). Patients with type 2 diabetes were randomized to follow either LFD aiming for 55-60 energy per cent (E%) from carbohydrates (n = 30) or LCD aiming for 20 E% from carbohydrates (n = 29). Plasma was collected at baseline and after 6 months. C-reactive protein (CRP), interleukin-1 receptor antagonist (IL-1Ra), IL-6, tumour necrosis factor receptor (TNFR) 1 and TNFR2 were determined.RESULTS: Both LFD and LCD led to similar reductions in body weight, while beneficial effects on glycaemic control were observed in the LCD group only. After 6 months, the levels of IL-1Ra and IL-6 were significantly lower in the LCD group than in the LFD group, 978 (664-1385) versus 1216 (974-1822) pg/mL and 2.15 (1.65-4.27) versus 3.39 (2.25-4.79) pg/mL, both P < 0.05.CONCLUSIONS: To conclude, advice to follow LCD or LFD had similar effects on weight reduction while effects on inflammation differed. Only LCD was found significantly to improve the subclinical inflammatory state in type 2 diabetes.

    • Objective: There is a growing interest for matrix metalloproteinases (MMPs) and their tissue inhibitors (TIMPs) in plasma as novel biomarkers in coronary artery disease (CAD). We aimed to identify the sources of MMP-8, MMP-9, TIMP-1 and TIMP-2 among peripheral blood cells and further explore whether gene expression or protein release was altered in patients with stable angina pectoris (SA). Methods: In total, plasma MMP-9 was measured in 44 SA patients and 47 healthy controls. From 10 patients and 10 controls, peripheral blood mononuclear cells (PBMC) and neutrophils were isolated and stimulated ex vivo. MMPs, TIMPs and myeloperoxidase were measured in plasma and supernatants by ELISA. The corresponding gene expression was measured by real-time PCR. Results: Neutrophils were the dominant source of MMP-8 and MMP-9. Upon moderate stimulation with IL-8, the neutrophil release of MMP-9 was higher in the SA patients compared with controls (p andlt; 0.05). In PBMC, the TIMP-1 and MMP-9 mRNA expression was higher in SA patients compared with controls, p andlt; 0.01 and 0.05, respectively. There were no differences in plasma levels between patients and controls except for TIMP-2, which was lower in patients, p andlt; 0.01. Conclusion: Measurements of MMPs and TIMPs in plasma may be of limited use. Despite similar plasma levels in SA patients and controls, the leukocyte-derived MMP-9 and TIMP-1 are significantly altered in patients. The findings indicate that the leukocytes are more prone to release and produce MMP-9 in symptomatic and angiographically verified CAD-a phenomenon that may have clinical implications in the course of disease.

    • Background: Matrix metalloproteinase (MMP)-9 may play a central role in the development and progression of atherosclerosis. Emerging evidence also indicates an association between MMP-9 and depressive symptomatology. Here, we investigated whether expression of MMP-9 and its inhibitors in blood mononuclear cells and plasma were related to depressive symptoms in patients with a recent myocardial infarction (MI). Methods and Results: Blood sampling was performed between 6 and 18 months after MI in 57 patients. Forty-one clinically healthy subjects were included as controls. Gene expression of MMP-9 and its main tissue inhibitors TIMP-1 and -2 were analyzed in freshly isolated or cultured blood mononuclear cells. Corresponding protein levels were assessed in cell supernatants and plasma. In post-MI patients, mRNA levels of MMP-9 and TIMP-1 and -2 were significantly higher than in controls while protein levels in cell supernatants and plasma did not differ between groups. The Center for Epidemiological Studies - Depression (CES-D) scale was used to assess depressive symptomatology. Repeated assessments during the first 18 months after MI showed significantly higher CES-D scores in patients compared with controls. However, there were no relationships between depressive mood and any of the measurements of MMP-9 or TIMPs. Conclusion: Our findings indicate that overexpression of MMP-9 and TIMPs in blood mononuclear cells and elevated depressive symptoms represent two unrelated phenomena after MI.

    • The complexity of tissue and the alterations that distinguish normal from cancer remain a challenge for translating results from tumor biological studies into clinical medicine. This has generated an unmet need to exploit the findings from studies based on cell lines and model organisms to develop, validate and clinically apply novel diagnostic, prognostic and treatment predictive markers. As one step to meet this challenge, the Human Protein Atlas project has been set up to produce antibodies towards human protein targets corresponding to all human protein coding genes and to map protein expression in normal human tissues, cancer and cells. Here, we present a dictionary based on microscopy images created as an amendment to the Human Protein Atlas. The aim of the dictionary is to facilitate the interpretation and use of the image-based data available in the Human Protein Atlas, but also to serve as a tool for training and understanding tissue histology, pathology and cell biology. The dictionary contains three main parts, normal tissues, cancer tissues and cells, and is based on high-resolution images at different magnifications of full tissue sections stained with H & E. The cell atlas is centered on immunofluorescence and confocal microscopy images, using different color channels to highlight the organelle structure of a cell. Here, we explain how this dictionary can be used as a tool to aid clinicians and scientists in understanding the use of tissue histology and cancer pathology in diagnostics and biomarker studies.

    • Atherosclerosis is a chronic inflammatory disease promoted by hyperlipidemia. Several studies support FOXP3-positive regulatory T cells (Tregs) as inhibitors of atherosclerosis; however, the mechanism underlying this protection remains elusive. To define the role of FOXP3-expressing Tregs in atherosclerosis, we used the DEREG mouse, which expresses the diphtheria toxin (DT) receptor under control of the Treg-specific Foxp3 promoter, allowing for specific ablation of FOXP3(+) Tregs. Lethally irradiated, atherosclerosis-prone, low-density lipoprotein receptor-deficient (Ldlr(-/-)) mice received DEREG bone marrow and were injected with DT to eliminate FOXP3(+) Tregs. Depletion of Tregs caused a 2.1-fold increase in atherosclerosis without a concomitant increase in vascular inflammation. These mice also exhibited a 1.7-fold increase in plasma cholesterol and an atherogenic lipoprotein profile with increased levels of VLDL. Clearance of VLDL and chylomicron remnants was hampered, leading to accumulation of cholesterol-rich particles in the circulation. Functional and protein analyses complemented by gene expression array identified reduced protein expression of sortilin-1 in liver and increased plasma enzyme activity of lipoprotein lipase, hepatic lipase, and phospholipid transfer protein as mediators of the altered lipid phenotype. These results demonstrate that FOXP3(+) Tregs inhibit atherosclerosis by modulating lipoprotein metabolism.

  • Probiotics and innate immune response in infants

    • We studied the effects of probiotic treatment on the innate immune system during infancy. The study included a subgroup of infants recruited to the pilot study testing the feasibility of probiotics intervention in infants with genetic risk of type 1 diabetes (T1D). A mixture of Lactobacillus rhamnosus GG (5 x 109 cfu), Lactobacillus rhamnosus LC705 (5 x 109 cfu), Bifidobacterium breve Bbi99 (2 x 108 cfu) and Propionibacterium freudenreichii ssp. Shermani JS (2 x 109 cfu) was given to the infants beginning one to three weeks after birth until the age of 6 months. Blood samples were drawn at the age of 6, 12 and 24 months for the analyses of beta-cell autoantibodies and the phenotype and stimulation response of monocytes with flow-cytometry, including surface markers on circulating CD14+ monocytes and expression of co-stimulatory markers on CD14+ monocytes as response to stimulation with lipoteichoic acid (LTA) and lipopolysaccharide (LPS). Also gene expression of toll-like receptor (TLR) signalling molecules was studied in the peripheral blood mononuclear cell (PBMC) population.In the children who received probiotics the number of circulating CD14+ monocytes expressing CD58 was reduced at the age of 6 months, and a tendency for a decreased induction of CCR5, CD80 and CD58 expressing monocytes as response to LTA was seen when compared to the children who received placebo. At the age of 12 months, the number of monocytes expressing CCR5 was decreased in the probiotic group, and a decreased spontaneous expression of TNFRSF1A and an increased spontaneous expression of TLR9 was observed in the PBMC from the children treated with probiotics. In the whole study group, the numbers of circulating monocytes expressing CD80 increased with age as well as the induction of CCR5, CD80 and CD58 on monocytes as response to stimulation. By the age of 24 months one child in both groups developed multiple autoantibodies.We demonstrated that probiotics modulated the activation stage and stimulation response of monocytes, and that prolonged effects of the treatment were seen at the age of 12 months. The findings suggest that early microbial exposure may program the function of the innate immune system for later life.

    • Every now and then new railway stations are brought into operation on existing lines. This is a good way of increasing the availability of railway services and attracting more passengers. However, from a capacity point of view, this procedure can be quite tricky, since new stations and additional stops thoroughly alter the traffic properties of the line. The addition of a station like this in Solna, north of Stockholm is under discussion. Here, most of the regional trains, but probably not the long-distance trains, would stop for passenger exchange. A new line, connected to the main line just north of Solna, would also contribute to the traffic flow through the new regional station. The essential question in this project was to determine the number of platform tracks needed to cope with the traffic flow. However, it has proven difficult to find a representative timetable structure to use in the dimensioning work, both the total number of trains and the distribution between stopping and passing trains turned out to be uncertain. A combinatorial method was therefore applied. Using this approach, a large number of timetables, i.e. possible traffic situations, were generated and tested (automatically) for the number of platform tracks needed. Constructing and using this simple model forced the engineers to understand and describe the fundamentals of this operational/scheduling/ dimensioning problem. The procedure hence gave useful insights about the system properties and a direct knowledge of the sensitivity of different factors that are essential for the number of tracks needed at a railway station like this.

  • Aggregation of Color pigments by Surface fixation treatment

    • The quality of a printed image is strongly influenced by the physical and the chemical interactions between the ink and the paper. Print quality can be evaluated either by objective measurements using instruments or by visual assessment studies involving panel of observers judging the final print. In this article, the print quality on commercial papers as well as on non-commercial papers with different amounts of salt for surface fixation has been studied. Perceived detail reproduction depends not only on sharp edge definition but also on the level of color saturation (Chroma). Color saturation and edge definition originate from two different ink and paper interaction processes. Color saturation is heavily dependent on ink penetration while edge definition correlates to ink spreading. In order to gain understanding of the performance of surface treatment by salt, large efforts have been put on splitting up of the increase in color saturation (Chroma) and improved edge definition. The printouts have been made with a desktop printer using pigmented inks. Cross section images have been taken with a light microscope to analyze the ink penetration depth. SEM analysis has been made to analyze the aggregation of the pigments on the surface. The print quality measurements have been both objective measurements such as print density and subjective image evaluation using a test panel of observers in a perceptual study. The perceptual study focused on detail reproduction, and efforts were made to separate the influence of the print density from the edge definition on the detail reproduction. The result from this study shows that an increased level of salt as surface fixation improves the detail reproduction due to aggregation of the pigments on the surface and that the ink penetration depth can be reduced by adding salt as surface fixation resulting in a higher print density.

Målet med verksamheten var tredelat och syftade till 1) att utveckla samarbete mellan forskning och konstnärlig verksamhet genom en modell byggd på metoden aktionsforskning, 2) att forskarna skulle återkoppla sina iakttagelser under repetitionsarbetet med betoning på att det skulle ske innan premiären, 3) att dramatikerna också önskade sig en kritisk analys av pjäsen som text. Forskarna har utforskat iscensättningsprocessen,reflekterat och analyserat återkopplingsprocesser mellan forskare och dramatiker, samt genomfört en genusteoretisk textläsning av pjäsen. Föreliggande artikel behandlar principen om barnets bästa i den norska och svenska utlänningsförvaltningen. Den bygger på en studie av asylbeslut rörande 197 barn och unga som sökte asyl i Norge eller Sverige 2008. Frågan som står i fokus är vilken innebörd idén om barnets bästa har, på papper och i praktik, inom ett område som präglas av en spänning mellan synen på flyktingbarn, barn som sårbara, å ena sidan och av intresset att begränsa invandringen å den andra.
