    • The difficulty associated with an accurate determination of transition rates for forbidden lines in lowly ionized heavy elements is illustrated in the case of Nd II. We have investigated the radiative decay of the low-lying metastable levels in Nd+ including the two levels 4f(4)(I-5) 5d K-6(11/2) and 4f(4)(I-5) 5d I-6(13/2). In these two particular cases, using di. erent theoretical approaches, we. nd that the decay is dominated by the M1 channels but that the E2 contributions are of the same order of magnitude. These levels have also been studied experimentally by lifetime measurements with the heavy ion storage ring CRYRING of Stockholm University. The difficulties encountered when performing such experiments are underlined and discussed.

    • The two metastable levels 5d(2)D(3/2) and 5d(2) D-5/2 in Ba II both show extremely long lifetimes of the order of several tens of seconds each. This has been found both by experiments and by theoretical predictions. The small transition probabilities associated with these two levels make them interesting and challenging for theoreticians as well as for experimentalists. Several calculations and measurements of these two lifetimes have been made previously but discrepancies between the results are present. This article presents values of ??? = 89.4 +/- 15.6 s for the D-2(3/2) level and ??? = 32.0 +/- 4.6 s for the D-2(5/2) level measured in a beam-laser experiment performed at the ion storage ring CRYRING. These values are supported by our new calculations resulting in ???= 82.0 s for the D-2(3/2) level and ???= 31.6 s for the D-2(5/2) level.

    • A laser probing investigation has yielded the lifetimes of the 3s23p4(1D)3d 2G7/2,9/2 metastable doublet states of Ar+. The results, obtained with the CRYRING ion storage ring of Stockholm, are 3.0±0.4 and 2.1±0.1  s, respectively. Comparisons with theoretical values calculated with two independent theoretical approaches, i.e., the pseudorelativistic Hartree-Fock method and the multiconfiguration Breit-Pauli approach, have allowed us to establish the unexpected and extraordinary strong contribution of an electric octupole (E3) transition to the ground state, in addition to the M1 decay channels to the 3d 2,4F states and the E2 contributions to the 4s 2P, 2D states.

    • We have investigated the radiative decay of the metastable level 5d(4)D(7/2) in Xe+. Theoretically we find the decay to be heavily dominated by an M2 transition and not by M1/E2 transitions. Lifetime measurements of 5d(4)D(7/2) in a storage ring are difficult since magnetic mixing of the metastable with a short-lived level quenches its population. Decay rates were determined at different magnetic field strengths (B) in order to allow a nonlinear extrapolation to B=0. The experimental lifetime of 2.4 +/- 0.8 s was in agreement with the calculated value, but much smaller than previously estimated.

    • We present a technique for measuring the radiative lifetimes of metastable states of negative ions that involves the use of a heavy-ion storage ring. The method has been applied to investigate the radiative decay of the np(3) P-2(1/2) levels of Te-(n=5) and Se-(n=4) and the 3p(3) D-2 state of Si- for which the J=3/2 and 5/2 levels were unresolved. All of these states are metastable and decay primarily by emission of E2 and M1 radiation. Multi Configuration Dirac-Hartree-Fock calculations of rates for the transitions in Te- and Se- yielded lifetimes of 0.45 s and 4.7 s, respectively. The measured values agree well with these predicted values. In the case of the D-2 state of Si-, however, our measurement was only able to set a lower limit on the lifetime. The upper limit of the lifetime that can be measured with our apparatus is set by how long the ions can be stored in the ring, a limit determined by the rate of collisional detachment. Our lower limit of 1 min for the lifetime of the D-2 state is consistent with both the calculated lifetimes of 162 s for the D-2(3/2) level and 27.3 h for the D-2(5/2) level reported by O'Malley and Beck and 14.5 h and 12.5 h, respectively, from our Breit-Pauli calculations.

    • Assessment of impairment and function is essential in order to monitor joint status and evaluate therapeutic interventions in patients with haemophilia. The improvements in the treatment of haemophilia have required the development of more sensitive tools to detect the more minor dysfunctions that may now be apparent. This paper outlines some of the recent developments in this field. The Haemophilia Joint Health Score (HJHS) provides a systematic and robust measure of joint impairment. The MRI Scoring System has been designed to provide a comprehensive scoring system combining both progressive and additive scales. The Functional Independence Score for Haemophilia (FISH) has been developed to assess performance of functional activities and can be used in conjunction with the Haemophilia Activities List (HAL) which provides a self report measure of function. It is recommended that both measures are evaluated as these tools measure different constructs. Further refinement and testing of the psychometric properties of all of these tools is in progress. More widespread use of these tools will enable the sharing of data across the world so promoting best practice and ultimately enhancing patient care.

  • Mass composition of the escaping plasma at Mars

    • Data from the Ion Mass Analyzer (IMA) sensor of the ASPERA-3 instrument suite on Mars Express have been analyzed to determine the mass composition of the escaping ion species at Mars. We have examined 77 different ion-beam events and we present the results in terms of flux ratios between the following ion species: CO2+/O+ and O-2(+)/O+. The following ratios averaged over all events and energies were identified: CO2+/O+ = 0.2 and O-2(+)/O+ = 0.9. The values measured are significantly higher, by a factor of 10 for O-2(+)/O+, than a contemporary modeled ratio for the maximum fluxes which the martian ionosphere can supply. The most abundant ion species was found to be O+, followed by O-2(+) and CO2+. We estimate the loss of CO2+ to be 4.0 x 10(24) s(-1) (0.29 kg s(-1)) by using the previous measurements of Phobos-2 in our calculations. The dependence of the ion ratios in relation to their energy ranges we studied, 0.3-3.0 keV, indicated that no clear correlation was found.

    • In the spectrum of an emission-line region ejected from the massive star Eta Carinae, called the strontium filament (SrF), forbidden lines from many elements, in particular [Sr II] and [Ti II], are observed. These lines are strong in this specific region and valuable for plasma diagnostics. Forbidden lines are not easily produced in laboratory light sources and the atomic parameters for these lines can thus not be measured in a straightforward way. We use a combination of laboratory and astrophysical measurements to determine transition probabilities for the [Ti II] lines. Lifetimes for metastable levels in Ti II are measured using a laser probing technique on a stored ion beam at CRYRING, MSL, Stockholm. Branching fractions from some of these levels are derived from Hubble Space Telescope/STIS spectra of the SrF. The astrophysical branching fractions are combined with the experimental lifetimes to determine absolute transition probabilities. We report lifetimes for the Ti II levels b(4)P(3/2), b(2)P(1/2), c(2)D(3/2) and c(2)D(5/2), in the range 0.29-17 s, and transition probabilities for eight parity-forbidden lines from the levels c(2)D(3/2) and c(2)D(5/2), along with uncertainty estimates.

The method has been in use for about eight years and it has been applied to various ions. Levels with radiative lifetimes from a few milliseconds up to 28 s have been measured. The method has been continuously developed and this article is focused on recent experimental developments of the method in order to widen its applicability and to increase its efficiency and accuracy.

  • Experimental oscillator strengths for forbidden lines in complex spectra

    • In dilute astrophysical plasmas, such as planetary nebulae and H II regions, strong lines appear that only have weak, if any, correspondence to the spectrum of laboratory light sources. Some of these are parity forbidden lines, i.e. emission from long-lived metastable states that cannot decay via normal electric dipole (E1) routes but only via the slower M1 and E2 transitions. The long lifetime of the upper levels and their sensitivity to collisions make the lines good diagnostics of the emitting plasma, but then the transition probability, or A-value, of these lines must be known. We report on a technique to experimentally determine A-values for forbidden lines using the method of combining the lifetime of the upper level with the branching fractions for the different decay channels. The lifetime is measured using the laser probing technique (LPT) on a stored ion beam. Since these lines are rarely produced in laboratory plasmas, we use astrophysical spectra to determine the branching fractions (BF). The lifetime and the BF then give the A-value, which is one quantity needed for modeling the spectrum of the plasma emitting the forbidden lines. The present measurements are performed within the FERRUM project, an international collaboration producing and evaluating transition probabilities for iron group element lines of astrophysical importance [Johansson, P. I. S. et al., Physica Scripta T100, 71 (2002).]

    • A fast ion beam stored in an ion storage ring will suffer from collisional interaction with the residual gas even in extreme ultrahigh vacuum. Normally these effects are negligible or easy to correct for. For measurements of extremely long radiative lifetimes of metastable levels we have found that correction for collisional effects is very important and usually limits the accuracy. We now suggest that by monitoring the process of collisional excitation of the metastable level of interest the lifetime value could be determined directly with improved accuracy. The method is applied to available data from our recent measurement of the 5d 2D5∕2 level in Ba+, yielding a value of 32.0±2.9 s, thus squeezing the error bar by 35%.

    • Summary. We assessed the reliability and construct validity of the Compatible MRI scale for evaluation of elbows, and compared the diagnostic performance of MRI and radiographs for assessment of these joints. Twenty-nine MR examinations of elbows from 27 boys with haemophilia A and B [age range, 5–17 years (mean, 11.5)] were independently read by four blinded radiologists on two occasions. Three centres participated in the study: (Toronto, n = 24 examinations; Atlanta, n = 3; Cuiaba, n = 2). The number of previous joint bleeds and severity of haemophilia were reference standard measures. The inter-reader reliability of MRI scores was substantial (ICC = 0.73) for the additive (A)-scale and excellent (ICC = 0.83) for the progressive (P)-scale. The intrareader reliability was excellent for both P-scores (ICC = 0.91) and A-scores (ICC = 0.93). The total P- and A-scores correlated poorly (r = 0.36) or moderately (r = 0.54), but positively, with clinical-laboratory measurements. The total MRI scores demonstrated high accuracy for discrimination of presence or absence of arthropathy [P-scale, area-under-the-curve (AUC) = 0.94 ± 0.05; A-scale, AUC = 0.89 ± 0.06], as did the soft tissue scores of both scales (P-scale, AUC = 0.90 ± 0.06; A-scale, AUC = 0.86 ± 0.06). Areas-under-the-curve used to discriminate severe disease demonstrated high accuracy for both P-MRI scores (AUC = 0.83 ± 0.09) and A-MRI scores (AUC = 0.87 ± 0.09), but non-diagnostic ability to discriminate mild disease. Similar results were noted for radiographic scales. In conclusion, both MRI scales demonstrated substantial to excellent reliability and accuracy for discrimination of presence/absence of arthropathy, and severe/non-severe disease, but poor to moderate convergent validity for total scores and non-diagnostic discriminant validity for mild/non-mild disease. Compared with radiographic scores, MRI scales did not perform better for discrimination of severity of arthropathy.

    • We tested the reliability and construct validity of the Compatible magnetic resonance imaging (MRI) scale for the evaluation of haemophilic knees and ankles and compared the diagnostic performance of MRI and plain film radiographs. Sagittal and coronal gradient-echo 1.5-T MR images of knees (n = 22) and ankles (n = 23) were obtained from boys (age range 4-16 years; mean 11 years) in two centres (Toronto, n = 26; Europe, n = 19). The MR images were independently read by four blinded radiologists on two occasions. Number of previous joint bleedings and laboratory level of severity of haemophilia were the reference standards for imaging assessment. Both components of the MRI scale demonstrated high inter- and intrareader intraclass correlation coefficients (progressive (P) scale, 0.91 and 0.94; additive (A) scale, 0.81 and 0.92 respectively). The correlation between the osteochondral domain of the MRI scale and patient's age was moderate. Otherwise, correlations between A- and P-scales and clinical laboratory measurements were weak. The areas under the curve (AUCs) used for discrimination of disease severity were similar for the A- and P-scales (AUCs used for mild disease, A-scale, 0.72 +/- 0.07; P-scale, 0.69 +/- 0.08; P = 0.23; AUCs for severe disease, A-scale, 0.93 +/- 0.05; P-scale, 0.87 +/- 0.08; P = 0.05). No differences were noted between the AUCs of the MRI and radiographic scales used for discrimination of late osteoarticular changes; MRI scales performed better for discrimination of early changes. In conclusion, both MRI scales demonstrated excellent reliability, poor convergent validity, and moderate and excellent validity for discrimination of mild and severe diseases respectively. Compared with radiographic scores, the MRI scales performed better for discrimination of early osteoarticular changes.

    • The temporal variation of the electron energy distribution function (EEDF) was measured with a Langmuir probe in a high power impulse magnetron sputtering (HiPIMS) discharge at 3 and 20 mTorr pressures. In the HiPIMS discharge a high power pulse is applied to a planar magnetron giving a high electron density and highly ionized sputtered vapor. The measured EEDF is Maxwellian-like during the pulse; it is broader for lower discharge pressure and it becomes narrower as the pulse progresses. This indicates that the plasma cools as the pulse progresses, probably due to high metal content of the discharge.

    • Context. In many plasmas, long-lived metastable atomic levels are depopulated by collisions (quenched) before they decay radiatively. In low-density regions, however, the low collision rate may allow depopulation by electric dipole (E1) forbidden radiative transitions, so-called forbidden lines (mainly M1 and E2 transitions). If the atomic transition data are known, these lines are indicators of physical plasma conditions and used for abundance determination. Aims. Transition rates can be derived by combining relative intensities between the decay channels, so-called branching fractions (BFs), and the radiative lifetime of the common upper level. We use this approach for forbidden [Sc II] lines, along with new calculations. Methods. Neither BFs for forbidden lines, nor lifetimes of metastable levels, are easily measured in a laboratory. Therefore, astrophysical BFs measured in Space Telescope Imaging Spectrograph (STIS) spectra of the strontium filament of Eta Carinae are combined with lifetime measurements using a laser probing technique on a stored ion-beam (CRYRING facility, MSL, Stockholm). These quantities are used to derive the absolute transition rates (A-values). New theoretical transition rates and lifetimes are calulated using the CIV3 code. Results. We report experimental lifetimes of the Sc II levels 3d(2) a(3)P(0,1,2) with lifetimes 1.28, 1.42, and 1.24 s, respectively, and transition rates for lines from these levels down to 3d4s a(3)D in the region 8270-8390 angstrom. These are the most important forbidden [Sc II] transitions. New calculations for lines and metastable lifetimes are also presented, and are in good agreement with the experimental data.

  • Novel patterns of genome rearrangement and their association with survival in breast cancer

    • Representational Oligonucleotide Microarray Analysis (ROMA) detects genomic amplifications and deletions with boundaries defined at a resolution of ∼50 kb. We have used this technique to examine 243 breast tumors from two separate studies for which detailed clinical data were available. The very high resolution of this technology has enabled us to identify three characteristic patterns of genomic copy number variation in diploid tumors and to measure correlations with patient survival. One of these patterns is characterized by multiple closely spaced amplicons, or “firestorms,” limited to single chromosome arms. These multiple amplifications are highly correlated with aggressive disease and poor survival even when the rest of the genome is relatively quiet. Analysis of a selected subset of clinical material suggests that a simple genomic calculation, based on the number and proximity of genomic alterations, correlates with life-table estimates of the probability of overall survival in patients with primary breast cancer. Based on this sample, we generate the working hypothesis that copy number profiling might provide information useful in making clinical decisions, especially regarding the use or not of systemic therapies (hormonal therapy, chemotherapy), in the management of operable primary breast cancer with ostensibly good prognosis, for example, small, node-negative, hormone-receptor-positive diploid cases.

    • Radiographs are important tools to evaluate structural changes in many Joint diseases. In the case of haemophilic arthropathy (HA), the Pettersson score is widely used. The rising of digital radiography enables evaluation of these changes in a more quantitative and detailed manner, potentially improving diagnosis and follow-up. The aim of this study was to evaluate whether digital image analysis ill the case of HA is feasible, using a presently available method for radiographic changes in knee osteoarthritis (OA), knee image digital analysis (KIDA). Sixty-two knee radiographs were scored according to Pettersson and with KIDA, each by two independent observers. Inter-observer variation and correlations between the two scoring methods were determined. The inter-observer variation was smaller for KIDA than for Pettersson and for KIDA not significantly different from evaluation of OA joints. Good correlations were found for the two methods where comparison of parameters was appropriate. Importantly, for each of the parameters within one point in the ordinal Pettersson score, a large window still existed in the continuous KIDA grading. Digital analysis of radiographs to quantify joint damage in HA is feasible. The use of continuous variables, as used in a digital method Such as KIDA has the advantage that it enables objective and much more sensitive detection of small changes than by use of an ordinal analogue method Such as the Pettersson score. Based oil the present results, it would be worthwhile to adapt the KIDA method for the specific characteristics of HA and to extend the method to elbow and ankle radiographs.

    • Objective. Hemophilic arthropathy, with characteristics of inflammatory (rheumatoid arthritis) and degenerative (osteoarthritis) joint damage, occurs at an early age, is associated with minor comorbidity, and is restricted to 3 pairs of large joints. The aim of this study was to determine whether commonly used serum and/or urinary biomarkers of cartilage and bone turnover for which assay kits are commercially available are associated with the severity of joint damage in patients with various degrees of hemophilic arthropathy and, thus, whether this disease could be useful in the identification and evaluation of such biomarkers. Methods. Blood and urine samples were collected from 36 patients with various degrees of hemophilic arthropathy. Commercially available assays for the most frequently investigated serum and urine biomarkers were performed: urinary C-terminal telopeptide of type I collagen (CTX-I), urinary CTX-II, serum CTX-I, serum CTX-II, serum cartilage oligomeric matrix protein (COMP), serum cartilage cleavage products C1,2C and C2C, and serum chondroitin sulfate 846 (CS-846). Radiographs of the ankles, knees, and elbows in all patients were evaluated for the degree of joint damage according to the Pettersson score, which is based on cartilage and periarticular bone changes and is specific for hemophilic arthropathy. Results. Urinary CTX-II, serum C1,2C, and serum CS-846 levels correlated with the overall Pettersson score and with the joint space narrowing component. Regression analysis showed that combined indexes of different markers increased the degree of correlation for the combination of urinary CTX-II, serum COMP, and serum CS-846. Bone-specific markers (urinary/serum CTX-I and serum C1,2C) did not correlate with specific bone-related items of the Pettersson score (osteoporosis and erosions). Conclusion. These results support the idea that a combination of biomarkers relates significantly better to the severity of joint damage than do individual biomarkers. The combination of urinary CTX-II, serum COMP, and serum CS-846 correlated best with the degree of arthropathy. Because of its specific characteristics and restricted involvement, hemophilic arthropathy may prove useful in the screening of newly developed biomarkers of joint damage.

    • Gcn5 is a coactivator protein that contributes to gene activation by acetylating specific lysine residues within the N termini of histone proteins. Gcn5 has been intensively studied in the budding yeast, Saccharomyces cerevisiae, but the features of genes that determine whether they require Gcn5 during activation have not been conclusively clarified. To allow comparison with S. cerevisiae, we have studied the genome-wide role of Gcn5 in the distantly related fission yeast, Schizosaccharomyces pombe. We show that Gcn5 is specifically required for adaptation to KCl- and CaCl2-mediated stress in S. pombe. We have characterized the genome-wide gene expression responses to KCl stress and show that Gcn5 is involved in the regulation of a subset of stress response genes. Gcn5 is most clearly associated with KCl-induced genes, but there is no correlation between Gcn5 dependence and the extent of their induction. Instead, Gen5-dependent KCl-induced genes are specifically enriched in four different DNA motifs. The Gcn5-dependent KCl-induced genes are also associated with biological process gene ontology terms such as carbohydrate metabolism, glycolysis, and nicotinamide metabolism that together constitute a subset of the ontology parameters associated with KCl-induced genes.

    • We present seven new vertebrate homologs of the prolactin-releasing hormone receptor (PRLHR) and show that these are found as two separate subtypes, PRLHR1 and PRLHR2. Analysis of a number of vertebrate sequences using phylogeny, pharmacology, and paralogon analysis indicates that the PRLHRs are likely to share a common ancestry with the neuropeptide Y (NPY) receptors. Moreover, a micromolar level of NPY was able to bind and inhibit completely the PRLH-evoked response in PRLHR1-expressing cells. We suggest that an ancestral PRLH peptide started coevolving with a redundant NPY binding receptor, which then became PRLHR, approximately 500 million years ago. The PRLHR1 subtype was shown to have a relatively high evolutionary rate compared to receptors with fixed peptide preference, which could indicate a drastic change in binding preference, thus supporting this hypothesis. This report suggests how gene duplication events can lead to novel peptide ligand/receptor interactions and hence spur the evolution of new physiological functions.

    • Summary. The international MRI expert subgroup of the International Prophylaxis Study Group (IPSG) has developed a consensus for magnetic resonance imaging (MRI) scales for assessment of haemophilic arthropathy. A MRI scoring scheme including a 10 step progressive scale and a 20 step additive scale with identical definitions of mutual steps is presented. Using the progressive scale, effusion/haemarthrosis can correspond to progressive scores of 1, 2, or 3, and synovial hypertrophy and/or haemosiderin deposition to 4, 5, or 6. The progressive score can be 7 or 8 if there are subchondral cysts and/or surface erosions, and it is 9 or 10 if there is loss of cartilage. Using the additive scale, synovial hypertrophy contributes 1–3 points to the additive score and haemosiderin deposition contributes 1 point. For osteochondral changes, 16 statements are evaluated as to whether they are true or false, and each true statement contributes 1 point to the additive score. The use of these two compatible scales for progressive and additive MRI assessments can facilitate international comparison of data and enhance the accumulation of experience on MRI scoring of haemophilic arthropathy.

    • We derive expressions for the coupling coefficients for electromagnetic four-wave mixing in the nonlinear quantum vacuum. An experimental setup for detection of elastic photon-photon scattering is suggested, where three incoming laser pulses collide and generate a fourth wave with a new frequency and direction of propagation. An expression for the number of scattered photons is derived and, using beam parameters for the Astra Gemini system at the Rutherford Appleton Laboratory, it is found that the signal can reach detectable levels. Problems with shot-to-shot reproducibility are reviewed, and the magnitude of the noise arising from competing scattering processes is estimated. It is found that detection of elastic photon-photon scattering may be achieved.
