  • Plasma glial fibrillary acidic protein and tau: predictors of neurological outcome after cardiac arrest.

    • The purpose was to evaluate glial fibrillary acidic protein (GFAP) and total-tau in plasma as predictors of poor neurological outcome after out-of-hospital (OHCA) and in-hospital cardiac arrest (IHCA), including comparisons with neurofilament light (NFL) and neuron-specific enolase (NSE).Retrospective multicentre observational study of patients admitted to an intensive care unit (ICU) in three hospitals in Sweden 2014-2018. Blood samples were collected at ICU admission, 12h, and 48h post-cardiac arrest. Poor neurological outcome was defined as Cerebral Performance Category 3-5 at 2-6months after cardiac arrest. Plasma samples were retrospectively analysed for GFAP, tau, and NFL. Serum NSE was analysed in clinical care. Prognostic performances were tested with the area under the receiver operating characteristics curve (AUC).Of the 428 included patients, 328 were OHCA, and 100 were IHCA. At ICU admission, 12h and 48h post-cardiac arrest, GFAP predicted neurological outcome after OHCA with AUC (95% CI) 0.76 (0.70-0.82), 0.86 (0.81-0.90) and 0.91 (0.87-0.96), and after IHCA with AUC (95% CI) 0.77 (0.66-0.87), 0.83 (0.74-0.92) and 0.83 (0.71-0.95). At the same time points, tau predicted outcome after OHCA with AUC (95% CI) 0.72 (0.66-0.79), 0.75 (0.69-0.81), and 0.93 (0.89-0.96) and after IHCA with AUC (95% CI) 0.61 (0.49-0.74), 0.68 (0.56-0.79), and 0.77 (0.65-0.90). Adding the change in biomarker levels between time points did not improve predictive accuracy compared to the last time point. In a subset of patients, GFAP at 12h and 48 h, as well as tau at 48h, offered similar predictive value as NSE at 48h (the earliest time point NSE is recommended in guidelines) after both OHCA and IHCA. The predictive performance of NFL was similar or superior to GFAP and tau at all time points after OHCA and IHCA.GFAP and tau are promising biomarkers for neuroprognostication, with the highest predictive performance at 48h after OHCA, but not superior to NFL. The predictive ability of GFAP may be sufficiently high for clinical use at 12h after cardiac arrest.

  • A generalizable data-driven model of atrophy heterogeneity and progression in memory clinic settings

    • Memory clinic patients are a heterogeneous population representing various aetiologies of pathological aging. It is unknown if divergent spatiotemporal progression patterns of brain atrophy, as previously described in Alzheimer's disease (AD) patients, are prevalent and clinically meaningful in this group of older adults. To uncover distinct atrophy subtypes, we applied the Subtype and Stage Inference (SuStaIn) algorithm to baseline structural MRI data from 813 participants enrolled in the DELCODE cohort (mean ± SD age = 70.67 ± 6.07 years, 52% females). Participants were cognitively unimpaired (CU; n = 285) or fulfilled diagnostic criteria for subjective cognitive decline (SCD; n = 342), mild cognitive impairment (MCI; n = 118), or dementia of the Alzheimer's type (n = 68). Atrophy subtypes were compared in baseline demographics, fluid AD biomarker levels, the Preclinical Alzheimer Cognitive Composite (PACC-5), as well as episodic memory and executive functioning. PACC-5 trajectories over up to 240 weeks were examined. To test if baseline atrophy subtype and stage predicted clinical trajectories before manifest cognitive impairment, we analysed PACC-5 trajectories and MCI conversion rates of CU and SCD participants. Limbic-predominant and hippocampal-sparing atrophy subtypes were identified. Limbic-predominant atrophy first affected the medial temporal lobes, followed by further temporal and, finally, the remaining cortical regions. At baseline, this subtype was related to older age, more pathological AD biomarker levels, APOE ε4 carriership, and an amnestic cognitive impairment. Hippocampal-sparing atrophy initially occurred outside the temporal lobe with the medial temporal lobe spared up to advanced atrophy stages. This atrophy pattern also affected individuals with positive AD biomarkers and was associated with more generalised cognitive impairment. Limbic-predominant atrophy, in all and in only unimpaired participants, was linked to more negative longitudinal PACC-5 slopes than observed in participants without or with hippocampal-sparing atrophy and increased the risk of MCI conversion. SuStaIn modelling was repeated in a sample from the Swedish BioFINDER-2 cohort. Highly similar atrophy progression patterns and associated cognitive profiles were identified. Cross-cohort model generalizability, both on the subject and group level, were excellent, indicating reliable performance in previously unseen data. The proposed model is a promising tool for capturing heterogeneity among older adults at early at-risk states for AD in applied settings. The implementation of atrophy subtype- and stage-specific end-points may increase the statistical power of pharmacological trials targeting early AD.

  • “You fuel the car with gas, you fuel the body with food” – Educational functions of food for health in home and consumer studies

    • This study aims to contribute to the understanding of food for health as educational content in Home and Consumer Studies (HCS), specifically targeting three educational functions: (i) qualification, (ii) socialization, and (iii) subjectification. Data came from a comprehensive case study, where one school class and two HCS teachers were followed during one school year. Fourteen classroom observations and eight teacher interviews were included. It is shown how qualification functions comprise nutrition knowledge and cooking skills, how socialization functions entail, e.g., dichotomous values of food as “good” or “bad,” and how conscious consumers who take personal responsibility for themselves and the environment become promoted subjects. Taken together, the results illustrate how educational content can have consequences beyond those intended. Based on the results, an argument is made about the relevance for HCS teachers to critically reflect on their teaching and its potential educational consequences.

  • The Ghost Boulevard

    • In 1947, Soviet architect Alexey Shchusev developed a large-scale urban renewal project for the post-war city of Chisinau, the then-capital of the SSR of Moldova. Part of the master plan was the construction of Boulevard D. Cantemir, which would cut through the city’s historic fabric. Only two sections of the boulevard were built before the project was abandoned. During the period of radical institutional political and economic shift towards a market economy in the early 1990s, initiatives to build the boulevard re-emerged through red lines, zoning documents, and planning regulations. The lack of political consensus caused planning paralyses over the city, creating a legal void where different actors competed to appropriate spaces. The power of the red lines has prompted various kinds of materializations of the boulevard. The real battle takes place in the sphere of the imaginary, and memory management is one of the main planning tools. Exploring the trajectory of the “Ghost Boulevard,” I reveal conflicting political and economic agendas and the many forces that constitute complex processes of planning today.

    • This report documents the conversations that occurred during two seminars, “Caring for Plans: Narratives of the Parallel Society Package”, held at the Copenhagen Architecture Festival CAFx, October 17, 2021,1 and “Solidarity in Times of Repressive Politics: A Seminar on the Effects of the Concepts ‘Particularly/Vulnerable Areas’”, held at Folkets Husby, October 15, 2022, in the Stockholm suburb of Husby.Narratives about the “failure” of large-scale housing from the postwar decades are now guiding major physical, social, and economic changes in neighborhoods all over Europe. Denmark and Sweden have long been known for their welfare-state systems and benevolent housing policies. However, in recent years, both countries have enacted new national “anti-segregation” measures that call for major physical and social changes to neighborhoods built in the 1960s and 1970s. In these processes, the opinions of local communities and residents of the neighborhoods have seldom been heard. By working with “witness seminars,” a method adopted from oral history, it is our aim to foreground residents’ perspectives and how they have enacted solidarity and collective resistance to these measures.

    • Narratives about the ‘failure’ of large-scale post-World War II housing are now guiding major physical, social, and economic changes in neighborhoods all over Europe. This is true even in Denmark and Sweden, which have long been known for their welfare states and benevolent housing policies. Today, however, both countries have enacted new national anti-segregation measures that call for major physical and social changes to neighborhoods built in the postwar era, even as the opinions of local communities and residents of such neighborhoods have been only sparsely heard – if at all. By working with the method ‘witness seminars’, we – as the research collective Aktion Arkiv – foreground residents’ perspectives and their collective resistance: the effects and affects of top-down changes. While sharing their lived experiences and actions, residents say that architects and planners can ‘simply say no’ and thereby refuse to participate in these actions.

  • Evaluation of NTRK immunohistochemistry as a screening method for NTRK gene fusion detection in non-small cell lung cancer

    • Purpose: The small molecule inhibitors larotrectinib and entrectinib have recently been approved as cancer agnostic drugs in patients with tumours harbouring a rearrangement of the neurotrophic tropomyosin receptor kinase (NTRK). These oncogenic fusions are estimated to occur in 0.1-3 % of non-small cell lung cancers (NSCLC). Although molecular techniques are most reliable for fusion detection, immunohistochemical analysis is considered valuable for screening. Therefore, we evaluated the newly introduced diagnostic immunohistochemical assay (clone EPR17341) on a representative NSCLC cohort.Methods: Cancer tissue from 688 clinically and molecularly extensively annotated NSCLC patients were comprised on tissue microarrays and stained with the pan-TRK antibody clone EPR17341. Positive cases were further analysed with the TruSight Tumor 170 RNA assay (Illumina). Selected cases were also tested with a NanoString NTRK fusion assay. For 199 cases, NTRK RNA expression data were available from previous RNA sequencing analysis.Results: Altogether, staining patterns for 617 NSCLC cases were evaluable. Of these, four cases (0.6 %) demonstrated a strong diffuse cytoplasmic and membranous staining, and seven cases a moderate staining (1.1 %). NanoString or TST170-analysis could not confirm an NTRK fusion in any of the IHC positive cases, or any of the cases with high mRNA levels. In the four cases with strong staining intensity in the tissue microarray, whole section staining revealed marked heterogeneity of NTRK protein expression.Conclusion: The presence of NTRK fusion genes in non-small cell lung cancer is exceedingly rare. The use of the immunohistochemical NTRK assay will result in a small number of false positive cases. This should be considered when the assay is applied as a screening tool in clinical diagnostics.

    • Purpose The purpose of this study is to identify, characterize and assess supplier flexibility measurement practices in the order-to-delivery process. Design/methodology/approach The study involved a survey; participants were 224 purchasing managers at Swedish manufacturing companies that had more than 20 employees. Findings Scrutiny of the details of measurement practices revealed that most respondents actually do not specifically measure supplier flexibility. Instead they measure other measures like delivery reliability, conduct qualitative follow-ups, or cannot specify how supplier flexibility is measured. It was acknowledged that they measure different supplier flexibility aspects, and the applied measures were characterized, e.g. in terms of which flexibility dimension they represent. Research limitations/implications Conceptual clarifications and adaptations to measuring supplier flexibility in the order-to-delivery process are provided. The identified measures can be a contribution in further developing literature on flexibility performance measurement. Practical implications Purchasing, logistics and supply chain managers in search of supplier flexibility performance measurement can find ways to measure and an extended flexibility vocabulary. This has the potential to improve flexibility in the supply chain. Originality/value Even though flexibility is claimed as being an important competitive advantage, few empirical studies and operationalized measures exist, particularly in the order-to-delivery process.

    • Flexibilitet är ett viktigt konkurrensmedel. Att ha flexibla leverantörer är ett alternativ för ett köparföretag till att själv hålla kostsamma buffertar för att säkerställa egen flexibilitet mot kunder. Frågan är hur och med vilka strategier företag kan uppnå kundorder-flexibilitet i order-till-leveransprocessen? Få empiriska studier förekommer på området. Syftet med denna studie är att utveckla ett ramverk bestående av strategier för hur kundorderflexibilitet i order-till-leveransprocessen kan uppnås, samt att empiriskt undersöka hur leverantörer försöker uppnå kundorderflexibilitet. En konceptuell analys av befintlig litteratur genomfördes och sammanställdes i form av ett ramverk med strategier kopplade till två flexibilitetsdimensioner. Flexibilitet i orderkvantitet och flexibilitet i leveranstid har studerats och täcker därmed order- respektive leverans-delprocesserna. Denna struktur ger fyra kombinationer som täcker 21 proaktiva såväl som 18 reaktiva strategier.Efter att ha sammanställt ramverket av strategier genomfördes en empirisk intervjustudie där nio viktiga leverantörer till fyra stora svenska tillverkare ingick. Leverantörerna är alla viktiga materialleverantörer, som hanterar kundorder med leveranstid, d. v. s.  tillämpar montering mot order (ATO) eller tillverkning mot order (MTO). Deras marknadschefer, key account managers, försäljningschefer eller exportchefer svarade på en intervju som utgick från flexibilitetsstrategierna i ramverket. Svaren avsåg hur de agerar för att uppnå flexibilitet mot den givna köparen, vilket skattades på en skala 1 (inte alls) till 5 (i mycket stor omfattning). De kunde även lägga till ytterligare använda strategier. Det visade sig att den strategi som hade högst medelvärde var “vi förbereder oss genom kontinuerliga kontakter med kundens inköpare”. Medelvärdet för använda strategier var högre i order-delprocessen än i leverans-delprocessen. Med de empiriskt genererade strategierna expanderade ramverket till 23 proaktiva och 19 reaktiva strategier. Det utvecklade ramverket är ett bidrag till flexibilitetslitteraturen. Leverantörer kan använda ramverket som ett hjälpmedel för att på ett systematiskt sätt utveckla och uppnå en högre kundorderflexibilitet i order-till-leveransprocessen. Köparföretagen kan använda ramverket som en checklista eller ett verktyg vid leverantörsutveckling.

    • Purpose: The purpose of this study is to develop a framework of strategies to achieving customer order flexibility in and related to the order-to-delivery (OTD) process. The purpose is also to investigate how companies prioritize various strategies to achieve customer order flexibility.Design/methodology/approach: Based on a literature review, pre-tests and conceptual reasoning, a conceptual framework of strategies related to the order-to-delivery process was developed. The strategies were linked to the order quantity and delivery lead-time flexibility dimensions. This structure resulted in six groups covering enabling as well as remedial strategies. An empirical interview study of ten customer–supplier relationships was conducted.Findings: The interviews identified additional strategies, thereby expanding the framework. The enabling strategies with the highest median values were “have continuous contact with the customer's purchaser” and “use safety stock of raw materials/semi-finished products”. The remedial strategy with the highest median was “re-plan/re-prioritize the order backlog”. In the delivery sub-process, it was more common to apply remedial strategies for delivery lead-time than for order quantities.Research limitations/implications: The developed framework is a contribution to the literature on operational flexibility in and related to the OTD process. It complements existing knowledge by taking a supplier perspective.Practical implications: Suppliers can use the framework as a tool to understand and systematically achieve better customer order flexibility in and related to the OTD process. Customers can use the framework as a checklist for supplier evaluation and supplier development.Originality/value: Few identified studies include empirical data on customer order flexibility.

    • Flexibility is one enabler of efficient use of resources, and is therefore an antecedent to sustainability. The purpose of this article is to identify supplier flexibility variables in, and related to, the order-to-delivery (OTD) process and categorize them into a framework, followed by empirically exploring the framework. A perception-based survey was sent to Swedish purchasing managers. 289 responses were received. After descriptive gap analysis, exploratory factor analysis was applied to structure the responses into factors. This formed the basis for hierarchical linear regression analysis, explaining supplier flexibility. A conceptual framework, specifying supplier flexibility into volume, delivery and information exchange dimensions and relating these dimensions to the OTD process, was developed. Significant negative gaps between actual and demanded volume flexibility and delivery flexibility were identified, while significant positive gaps were found for information exchange flexibility. The factor analysis revealed three factors. The regression analysis verified that OTD-related information exchange flexibility and OTD-related volume and delivery flexibility explain the variation in OTD-specific flexibility, and are important antecedents for supplier flexibility in the OTD process. Research limitations/implications A contribution to research is the framework – with defined, related and empirically validated flexibility types. The study proposes a perception-based way to capture supplier flexibility in the OTD process, which is of practical relevance when evaluating suppliers. Identifying, conceptualizing and capturing types of supplier flexibility in the OTD process is new related to academic literature. The wide empirical study, mapping supplier flexibility gaps, is unique in its focus.

    • Objective The aim was to evaluate the effects of cocreated internet-based stepped care (iCAN-DO) on anxiety, depression, posttraumatic stress, and health-related quality of life (HRQoL) in individuals with cancer and self-reported anxiety and/or depression symptoms, compared with standard care.Methods Clinically recruited individuals with breast, colorectal, or prostate cancer underwent online screening with the Hospital Anxiety and Depression Scale (HADS). Those with anxiety and/or depression symptoms (>7 on any of the HADS subscales) were randomized to iCAN-DO or standard care. iCAN-DO comprised psychoeducation and self-care strategies (step 1) and internet-based cognitive behavioral therapy (iCBT, step 2). Data were collected before randomization and at 1, 4, 7, and 10 months and analyzed with intention-to-treat regression analysis and randomization tests.Results Online screening identified 245 (27%) of 909 individuals who reported anxiety and/or depression symptoms. They were randomized to iCAN-DO (n  = 124) or standard care (n = 121). Of them 49% completed the 10-month assessment, and in the iCAN-DO group 85% accessed step 1 and 13% underwent iCBT. iCAN-DO decreased the levels of symptoms of depression (−0.54, 95% confidence interval: −1.08 to −0.01, P < .05) and the proportion of individuals with symptoms of depression (P < .01) at 10 months, compared with standard care, according to HADS. There were no significant effects on anxiety, posttraumatic stress, or HRQoL.Conclusion Internet-based stepped care improves symptoms of depression in individuals with cancer. Further studies are needed to gain knowledge on how to optimize and implement internet-based support in oncology care.

All challenges had both direct and indirect negative implications for dimensioning safety buffers and were synthesised into a framework. Research limitations/implications This study complements the literature on dimensioning safety buffers with qualitative insights into challenges in dimensioning safety buffers and implications in practice. Practical implications Practitioners can use the framework to understand and overcome challenges in dimensioning safety buffers and their negative implications. Originality/value This study responds to the scarcity of qualitative and empirical studies on dimensioning safety buffers and the absence of any overview of the challenges therein.

  • Situated Freedom : Exploring the Aesthetic Practice of Rem Koolhaas/OMA

    • The thesis advocates that architecture can, as an operative medium, situate freedom. Following the work of Michel Foucault, the thesis develops an argument for an emancipatory endeavor, termed “situated freedom in architecture.” Acknowledging the contingency that can be located within power relations, situated freedom works with the positive and constitutive character of power as it shapes how we can be free (and unfree) in particular situations. To exemplify this approach within architecture practice, the thesis looks at the aesthetic practice of Rem Koolhaas/OMA. The research question asked is: How can architecture, as an aesthetic practice, through its own zone of competence, and by means of a singular, situated project, establish an emancipatory endeavor that is oriented towards enrichment, fulfillment, and joy. In responding to this question, the research analyzes three projects—a private pool (Villa dall’Ava), a media building (China Central Television Headquarters), and a public library (Seattle Public Library)—in three different situations. Through “fabulation” (Deleuze & Guattari) and the use of a “messy method” (Law) the thesis draws on multiple disciplines, such as art, film theory, theatre, architecture, and philosophy. The thesis’s methodology mobilizes a reading of architecture that traces the aesthetic strategies employed in the analyzed projects. Exploring the practice of Koolhaas/OMA offers insights into projective approaches in architecture and situates their work in a new context. The thesis aims to develop an understanding of architecture as an emancipatory endeavor of human wellbeing, enrichment, and joy within the complexity of the present reality.

  • Mäns våld mot kvinnor : Diskurser och kunskap i det sociala arbetets praktik

    • I avhandlingen analyseras vilken kunskap och diskurser som vägleder socialtjänstens arbete med mäns våld mot kvinnor. Avhandlingens övergripande frågeställningar fokuserar på: diskurser om mäns våld i Socialstyrelsens utbildningsmaterial, socialarbetares kunskapsbehov och hur dessa tillgodoses, samt socialarbetares uppfattningar om mäns våld mot kvinnor. Avhandlingen har ett feministiskt och intersektionellt perspektiv och bygger på två delstudier. I den första delstudien används kvalitativa metoder, och empirin består av fem utbildningsmaterial om mäns våld mot kvinnor som publicerats av Socialstyrelsen. Empirin analyserats med hjälp av Carol Bacchis policy approach What’s the Problem Represented to be?, och Norman Faircloughs kritiska diskursanalys. Empirin i den andra delstudien utgörs av enkätdata som inhämtats bland 153 socialarbetare verksamma i socialtjänsten i tre svenska kommuner med olika demografi och geografisk placering. Avhandlingen visar tre centrala resultat. För det första framställs mäns våld i könsneutrala termer och våldet beskrivs som ett relationsvåld och/eller familjevåld. För det andra problematiserar utbildningsmaterialens texter inte mäns överordning, de fäster inte heller någon särskild vikt vid strukturella orsakssammanhang. För det tredje centreras kunskapen och diskurserna kring individuella behov och orsaker. Främst fokuseras behoven hos de grupper av kvinnor som Regeringen menar är särskilt sårbara för våld. Det senare leder till att kvinnor kollektiviseras och att mäns ansvar för våldet, i synnerhet svenska män, och sambandet mellan våld, jämlikhet och andra sociala strukturer förbises. Professionell erfarenhet i ärenden där våld förekommer och utbildning inom området är faktorer som socialarbetarna anser bidragit till att öka deras kunskap. Trots detta uppger respondenter med lång arbetslivserfarenhet, hög utbildning, och fortbildning att de saknar kunskap om mäns våld mot kvinnor. Även fast att socialarbetarna upplever kunskapsbrist, visar resultatet att socialarbetarna endast tagit del stöddokument i begränsad omfattning. Den här avhandlingen bidrar med kunskap om vikten av språkanvändning och hur vi förstår mäns våld mot kvinnor. Resultatet kan användas för att ifrågasätta och utmana rådande normer om mäns våld i samhället.

    • Regeringens satsningar mot mäns våld mot kvinnor har varit omfattande under de senaste 15 åren jämfört med hur det sett ut historiskt. Det finns nu en mängd olika kunskapskällor som socialarbetare kan ta del av om våld i nära relationer för att öka sina kunskaper, bland annat har Socialstyrelsen publicerat utbildningsmaterial och det tillkommit en kunskapsbank om mäns våld mot kvinnor, hedersrelaterat våld och förtryck samt våld i samkönade relationer vid Nationellt centrum för kvinnofrid (NCK). Syftet med den här studien är att undersöka socialarbetares kännedom om interna/lokala/regionala policys och metodstöd om våld i nära relationer. Syftet är också att undersöka vilka internetsidor socialarbetare använder för att söka information om sådant våld. Resultatet är baserat på en enkät som besvarats av 153 socialarbetare i tre svenska kommuner. Resultatet visar att 65 procent av respondenterna känner till interna/lokala/regionala policys, och 44 procent känner till interna/lokala/regionala metodstöd. När det kommer till vilka internetsidor socialarbetarna använder för att söka information om våld i nära relationer så visar studien att Socialstyrelsens, kommunens, och kvinnojourers internetsidor är de som flest besökt. NCK tycks inte vara en lika viktig kunskapskälla. Ett viktigt område för kommande studier är att undersöka vilken sorts information om våld i nära relationer socialarbetarna söker.

    • BackgroundPrevious studies have reported high prognostic accuracy of circulating neurofilament light (NfL) at 24-72 h after out-of-hospital cardiac arrest (OHCA), but performance at earlier time points and after in-hospital cardiac arrest (IHCA) is less investigated. We aimed to assess plasma NfL during the first 48 h after OHCA and IHCA to predict long-term outcomes.MethodsObservational multicentre cohort study in adults admitted to intensive care after cardiac arrest. NfL was retrospectively analysed in plasma collected on admission to intensive care, 12 and 48 h after cardiac arrest. The outcome was assessed at two to six months using the Cerebral Performance Category (CPC) scale, where CPC 1-2 was considered a good outcome and CPC 3-5 a poor outcome. Predictive performance was measured with the area under the receiver operating characteristic curve (AUROC).ResultsOf 428 patients, 328 (77%) suffered OHCA and 100 (23%) IHCA. Poor outcome was found in 68% of OHCA and 55% of IHCA patients. The overall prognostic performance of NfL was excellent at 12 and 48 h after OHCA, with AUROCs of 0.93 and 0.97, respectively. The predictive ability was lower after IHCA than OHCA at 12 and 48 h, with AUROCs of 0.81 and 0.86 (p <= 0.03). AUROCs on admission were 0.77 and 0.67 after OHCA and IHCA, respectively. At 12 and 48 h after OHCA, high NfL levels predicted poor outcome at 95% specificity with 70 and 89% sensitivity, while low NfL levels predicted good outcome at 95% sensitivity with 71 and 74% specificity and negative predictive values of 86 and 88%.ConclusionsThe prognostic accuracy of NfL for predicting good and poor outcomes is excellent as early as 12 h after OHCA. NfL is less reliable for the prediction of outcome after IHCA.

    • The authors created a comprehensive developmental cell atlas for spatiotemporal gene expression of the human spinal cord, revealed species-specific regulation during development and used the atlas to infer novel markers for pediatric ependymomas. The spatiotemporal regulation of cell fate specification in the human developing spinal cord remains largely unknown. In this study, by performing integrated analysis of single-cell and spatial multi-omics data, we used 16 prenatal human samples to create a comprehensive developmental cell atlas of the spinal cord during post-conceptional weeks 5-12. This revealed how the cell fate commitment of neural progenitor cells and their spatial positioning are spatiotemporally regulated by specific gene sets. We identified unique events in human spinal cord development relative to rodents, including earlier quiescence of active neural stem cells, differential regulation of cell differentiation and distinct spatiotemporal genetic regulation of cell fate choices. In addition, by integrating our atlas with pediatric ependymomas data, we identified specific molecular signatures and lineage-specific genes of cancer stem cells during progression. Thus, we delineate spatiotemporal genetic regulation of human spinal cord development and leverage these data to gain disease insight.

    • Sedan tidigt åttiotal, när Sven-Olov Wallenstein slog ner i Stockholms intellektuella liv, har han vuxit till en kraftfull och mångförgrenad institution i sig, under sitt kärnfulla emblem: Walle. Med avstamp i filosofin och idéhistorien har han gripit in i den estetiska debatten som ingen annan under dessa gångna fyra decennier. Genom egna böcker och genom översättningar, genom tidskrifter, recensioner, katalogtexter, och inte minst genom sin undervisning runtom i Sverige och Norden har han på djupet påverkat diskussionen inom konst, arkitektur, men även inom litteratur, dans, musik och film. Som uttolkare och översättare av den klassiska tyska filosofin – Kant, Hegel, Husserl, Heidegger och Adorno – liksom av dess moderna franska arvtagare – Derrida, Foucault och Deleuze – har han på ett avgörande sätt bidragit till att återknyta det band mellan svensk och europeisk idékultur som den föregående generationens ofta historielösa omfamning av angloamerikansk analytisk filosofi hade klippt av.För att fira Walle har vi satt ihop en bok. Det har inte blivit en pliktbok, ingen sedvanlig samling lärda uppsatser som följer vänbokens konventioner. Istället för en festskrift, fick det bli en skriftfest, en bild- och uttrycks-fest, där skrivande och skapandet självt firas. Walle har arbetat hårdare än de flesta, men till sist var det alltid festen som var målet, releasen, partyt, skrattet och galenskapen. Vad vore en filosof som inte skrattar? Inte en Walle, inte den som förmår förvandla tunga resonemang till klara linjer, komplexa relationer till glad geometri, och mångfaldiga ord till färgstarka, träffande och koncentrerade bilder.

    • In 1960, the Swedish furniture designer Bruno Mathsson built a small one-level summerhouse for himself and his wife Karin at Frösakull in southern Sweden. The house was an experiment and was also dubbed the “House of Tomorrow.” Even though not internationally renown today, the house was praised for its innovative architecture at the time and, as Martin Friedman writes in Design Quarterly in 1965: “Constructed over sand dunes and tucked into a forest of dwarf pines, it is undoubtedly one of the most remarkable buildings in modern Sweden”.  Frösakull reveals a tension between elegant, sleek American mid-century modernism and brutalist anti-aestheticism. The large, illusory window sections and the undulating, transparent ceiling co-exist with recycled steel rafters, wood slats nailed on slightly askew, and the most basic steel draining board. The building oozes pragmatism rather than aestheticism, and the builder underlines this with his affirmation that Frösakull was not built according to the drawing board but that a great deal was left to the handymen to solve. The design and the solutions are usually the simplest and cheapest possible, and they do not consistently adhere to predetermined notions of measurements, proportions or aesthetics.Alongside the emergence of Swedish functionalism, another vital movement evolved: the fitness culture. Already at the Stockholm exhibition in 1930 a new anti-consumerist body culture centring on hygienism, outdoor sports, and nudism was presented in parallel with the new media and consumer culture. Bruno Mathsson was a veritable “health architect” who converted the plans of the health programme into a lifestyle.

    • We have developed a new transmission-based, open-ended coaxial probe for assessing vertebrae strength during spinal fusion surgery. The approach exploits the fact that the probes are within the far field of each other implying that the phase varies linearly with respect to propagation distance. Determining the absolute phase is critical for recovering the associated tissue dielectric properties from which bone strength will be determined. Unfortunately, unwanted multi-path signals corrupt the signals at the lower end of the operating frequency range from which our conventional unwrapping strategy depends. Our new approach requires only three measurements within the prime frequency range and can be determined robustly with a minimum of computations. This will be vital to developing a commercial device since the signal levels will be extremely low power requiring longer than usual data acquisition times, which will be mitigated by measuring the data at only a few frequencies. Fast and efficient operation will be critical for clinical success.

    • We are developing a new transmission-based probe for assessing vertebrae strength via the differences between the dielectric properties of normal and osteoporotic bone. The probes are simple open-ended coaxial cables that will fit easily into the pedicle canals drilled by the surgeons before implanting screws to secure the supporting instrumentation. If the screws pull because of bone weakness, it can inflict a wide range of complications. In the progression towards actual clinical trials, we have already validated the approach on different liquids with varying dielectric properties. In this study, we assess actual animal bone samples as a precursor to human investigations. We exploit the fact that the trabecular bone is considerably different on opposite sides of the growth plate of long bones such as the femur - somewhat mimicking differences between normal and osteoporotic bone. For these experiments, we show that the properties of the distal and proximal sections are considerably different and present opportunities to exploit them in a diagnostic setting

  • Inside the Ideal Home : The Changing Values of Apartment Living and the Promotion of Consumption in Sweden, 1950-1970

    • This dissertation explores the image of the ideal home in Sweden, an idea closely bound to the spatial dynamics of building norms and the outfitting of the domestic household, from the 1950s through the 1960s. By examining official, commercial, and consumer-cooperative ideals of housing and home, I attempt to understand and analyze correlations between various visions of the ideal apartment home and the social, political, economic, and cultural contexts in which they were conceived during the heyday of the Swedish welfare state. This subject has been extensively researched for the first years of the phenomenon, the 1930s and 1940s, but is relatively ignored for the years following World War II, when the new housing policy was formulated and the Record Years began. The study ends in 1970, when the construction of apartments and the standard of living peaked, followed by a focus on building single-family houses and the questioning of building norms. I argue that the values and visions of an ideal home were expressed in the rhetoric and representations of state, consumer-cooperative, and commercial publications. While scholars have studied individual aspects of this context, I maintain that the interrelationship between them produced a widely circulated vision of domesticity between 1950 and 1970. I highlight how commercial actors—primarily Ikea and the interior design magazine Allt i Hemmet (Everything in the Home)—interacted with state institutions in creating and promoting a discourse of the ideal home.

    • Membrane proteins on enveloped viruses play an important role in many biological functions involving virus attachment to target cell receptors, fusion of viral particles to host cells, host-virus interactions, and disease pathogenesis. Furthermore, viral membrane proteins on virus particles and presented on host cell surfaces have proven to be excellent targets for antivirals and vaccines. Here, we describe a protocol to investigate surface proteins on intact severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) particles using the dual-reporter flow cytometric system. The assay exploits multiplex technology to obtain a triple detection of viral particles by three independent affinity reactions. Magnetic beads conjugated to recombinant human angiotensin-converting enzyme-2 (ACE2) were used to capture viral particles from the supernatant of cells infected with SARS-CoV-2. Then, two detection reagents labeled with R-phycoerythrin (PE) or Brilliant Violet 421 (BV421) were applied simultaneously. As a proof-of-concept, antibody fragments targeting different epitopes of the SARS CoV-2 surface protein Spike (S1) were used. The detection of viral particles by three independent affinity reactions provides strong specificity and confirms the capture of intact virus particles. Dose-dependency curves of SARS-CoV-2 infected cell supernatant were generated with replicate coefficient variances (mean/SD) ˂14%. Good assay performance in both channels confirmed that two virus surface target protein epitopes are detectable in parallel. The protocol described here could be applied for (i) high-multiplex, high-throughput profiling of surface proteins expressed on enveloped viruses; ii) detection of active intact viral particles; and (iii) assessment of specificity and affinity of antibodies and antiviral drugs for surface epitopes of viral antigens.The application can be potentially extended to any type of extracellular vesicles and bioparticles, exposing surface antigens in body fluids or other liquid matrices.

    • Experiment är en relativt ovanlig forskningsmetod i logistikforskningsprojekt, trots att metoden har många fördelar. Genom ett experiment, som genomförs av de deltagande företagen men med stöd av forskarna, är det tydligare för företagen hur resultaten kan vara direkt användbara och därmed att forskningen manifesteras och nyttiggörs på ett konkret sätt. Det här exemplet baseras på en grupp forskare som tillsammans med fyra företag i ett forskningsprojekt kring säkerhetsbuffertdimensionering använt just experiment som metod. Syftet med artikeln är att visa på erfarenheter och lärdomar från företag och forskare med att använda experiment som forskningsmetod. Experimentet har gått ut på att sätta upp en adaptiv dimensioneringsmetod av olika säkerhetsbuffertar, och prova sig fram till en förbättrad leveransförmåga. Data har samlats i form av intervjuer och workshopdiskussioner, samt reflektioner från företagen direkt. Företagens respektive forskarnas erfarenheter och lärdomar presenteras. Företagen upplever t.ex. att de får bidra med input till projektet på ett annat vis än genom att enbart bli intervjuade eller lösa givna uppgifter, och att de samtidigt får ut direkt företagsnytta med sin insatta tid. Samtidigt ställer det här stora krav på att företagen är engagerade i forskningen och har möjlighet att genomföra ett experiment som kanske kräver en del ”handpåläggning” och kan påverka den ordinarie verksamheten. Forskarna upplever att det kräver mer i form av gedigen planering och förarbete, att tydligt specificera experimentet och dess ingående delar så att det är praktiskt tillämpbart. Avstämningsmöten med företagen har visat sig varit viktigt för att vägleda, reda ut oklarheter och kontrollera status. Bådas erfarenhet är att utväxlingen på den insatta tiden är avsevärd om experimentet utformas med en balans mellan företagens och forskarnas behov. Företagen bidrar med datainsamling och erfarenheter från praktisk tillämpning, vilket ger viktiga insikter till den teoretiska utvecklingen. Sammantaget blir nettoeffekten positiv för alla inblandade parter. Metoden innebär både fler och längre iterationer mellan företag och forskare vilket gör att tillgången på kalendertid är central. Avslutningsvis sammanfattas ett antal erfarenheter som kan vara till nytta för företag och forskare som vill använda experiment som forskningsmetod.
