    • Economic studies concerning environmental functions of forests are often partial in the sense that they focus on either benefits or costs. In other words, benefit/cost analyses indicating whether it is economically motivated to change forestry to make it compatible with environmental demands are relatively rare. This benefit/cost analysis deals with the forest as recreation environment, where benefit estimates from a study conducted in the county of Västerbotten, Sweden, are compared with cost estimates from another study in the same county. It is shown that adjustments of forest management to meet recreational demands do largely affect both benefits and costs, and that the results are sensitive to how soon the effects on the recreation environment occur after the adjustments of forest management.

    • The growth of both tumors and nonneoplastic tissues may be influenced by signals from the vascular endothelium. In the present investigation we show that purified proliferating endothelial cells from pancreatic islets can stimulate beta-cell proliferation through secretion of hepatocyte growth factor (HGF). This secretion could be induced by soluble signals from the islets, such as vascular endothelial growth factor-A (VEGF-A) and insulin. During pregnancy, the pancreatic beta-cells display a highly reproducible physiological proliferation. We show that islet endothelial cell proliferation precedes beta-cell proliferation in pregnant animals. Vascular growth was closely associated with endocrine cell proliferation, and prominent expression of HGF was observed in islet endothelium on d 15 of pregnancy, i.e. coinciding with the peak of beta-cell proliferation. In summary, our results suggest the existence of an endothelial-endocrine axis within adult pancreatic islets, which is of importance for adult beta-cell proliferation.

    • OBJECTIVE—Clinical islet transplantations are performed through infusion of islets via the portal vein into the liver. This study aimed at characterizing the influence of the implantation microenvironment on islet graft metabolism and function. RESEARCH DESIGN AND METHODS—Islets were transplanted into their normal environment, i.e., the pancreas, or intraportally into the liver of mice. One month posttransplantation, the transplanted islets were retrieved and investigated for changes in function and gene expression. RESULTS—Insulin content, glucose-stimulated insulin release, (pro)insulin biosynthesis, and glucose oxidation rate were markedly decreased in islets retrieved from the liver, both when compared with islets transplanted into the pancreas and endogenous islets. Islets transplanted into the pancreas showed normal insulin content, (pro)insulin biosynthesis, and glucose oxidation rate but increased basal insulin secretion and impaired glucose stimulation index. Gene expression data for retrieved islets showed downregulation of pancreatic and duodenal homeobox gene-1, GLUT-2, glucokinase, mitochondrial glycerol-phosphate dehydrogenase, and pyruvate carboxylase, preferentially in intraportally transplanted islets. CONCLUSIONS—Islets transplanted into their normal microenvironment, i.e., the pancreas, display gene expression changes when compared with endogenous islets but only moderate changes in metabolic functions. In contrast, site-specific properties of the liver markedly impaired the metabolic functions of intraportally transplanted islets.

  • Oxygen Diffusion and Photon-Induced Decomposition of Acetone on Zr- and Nb-Doped TiO2 Nanoparticles

    • We report on the photoinduced decomposition of acetone on Zr- and Nb-doped anatase TiO2 nanoparticles prepared by the sol−gel method in oxygen-free environment (N2) and synthetic air, respectively. Physical properties of the nanoparticles were determined by TEM, SEM, AFM, XRD, and UV−vis spectroscopy. Photoinduced surface reactions were monitored by in situ Fourier transform infrared (FTIR) spectroscopy. Acetone photo-oxidation occurs in the absence of O2 in the reaction gas and is proposed to be due to reactions with photoactivated surface oxygen. In N2 atmosphere a new parallel reaction pathway is found that stimulates surface carbonate formation. A coupled diffusion-reaction model was developed to quantitatively determine the role of O diffusion. The results yield quantitative support to an oxygen surface diffusion mechanism, which depletes the surface from oxygen and gradually deactivates the particles in the absence of external oxygen supply. The diffusion reaction pathway is significant on the doped TiO2 particles. The contribution of this reaction pathway amounts to up to 65% of the total PID rate on Nb- and Zr-doped TiO2 in synthetic air environment, while it gives only a minor contribution on pure TiO2.

  • Endothelial cells in endogenous and transplanted pancreatic islets : differences in the expression of angiogenic peptides and receptors

    • Background/Aims: An important reason for the large amount of islets required for successful islet transplantation is likely to be inadequate engraftment of the transplanted islets. Thus, the revascularization is of major importance for graft survival. In order to study the expression of angiogenic peptides and receptors on islet endothelial cells (EC), we needed methods giving access to such endothelium. Therefore, we developed methods to isolate EC from islets transplanted intraportally or beneath the kidney capsule. Methods: Pancreatic islets were isolated, cultured and syngeneically transplanted into the liver or beneath the kidney capsule of C57BL/6 mice. One month post-transplantation, the islets were retrieved and EC from these islets were explanted. EC were also collected from freshly isolated and cultured non-transplanted islets. The EC were purified with Dynabeads and identified with immunocytochemistry. Angiogenesis GEArray technology was used to study angiogenic gene expression. Results: Several angiogenic genes were expressed in EC; e. g. endostatin, pigment-epithelial derived factor, vascular endothelial growth factor and angiopoietin-2, and their expression were affected by culture. Conclusion: The expression of angiogenesis-related genes in islet EC from non-transplanted islets is affected by culture. Moreover, we also describe a technique, which makes it possible to obtain EC from transplanted islets.

  • Sentinel node biopsy in malignant melanoma : Swedish experiences 1997-2005

    • The sentinel node biopsy (SNB) procedure is a multidisciplinary technique, invented to gain prognostic information in different malignant tumors. The aim of the present study was to study the cohort of patients with malignant melanoma, operated with SNB, from the introduction of the technique in Sweden, concerning the prognostic information retrieved and the outcome of the procedures. In Sweden all patients with malignant melanoma are registered at regional Oncological Centers. From these databases ten centers were identified, treating malignant melanoma and performing sentinel node biopsy. Consecutive data concerning tumor characteristics, outcome of the procedure and disease related events during the follow-up time were collected from these ten centers. All cases from the very first in each centre were included. The SNB procedure was performed in 422 patients with a sentinel node (SN) detection rate of 97%, the mean Breslow thickness of the primary tumors was 3.2 mm (median 2.4 mm) and the proportion of ulcerated melanomas 38%. Metastasis in the SN was found in 19% of the patients but there was a wide range in the proportion of SN metastases between the different centers (5-52%). After a follow-up of median 12 months of 361 patients, SN negative patients had better disease-free survival than SN positive (p<0.0001). A false negative rate of 14% was found during the follow-up time. In this study the surgical technique seemed acceptable, but the non-centralized pathology work-up sub-optimal. However, SNB was still found to be a significant prognostic indicator, concerning disease free survival.

  • Ventilation filter efficiency for particles and grass pollen allergens

    • In an experimental study, commonly used ventilation fine filters were tested regarding their efficiency in collecting airborne particles and grass pollen allergens from outdoor air. Grass pollen allergen and particle number concentrations were measured before and after the filters, enabling collection efficiency calculations. Simultaneously, the size distribution of grass pollen allergens was measured in outdoor air using a cascade impactor. The study confirms previous indications that pollen allergens occur in the outdoor air as particles much smaller than pollen grains, and can penetrate ventilation filters to a larger extent than might be expected. The initially high collection efficiency of synthetic, electrostatically charged filters declined significantly with time of use (dust load), whereas glass fiber filters showed steady performance. A slight tendency for pollen allergenic matter to penetrate ventilation filters more than other airborne particles was noted, but no difference in the response to electrostatic charge of filters could be seen.

    • The aim of the study was to investigate the possibility to increase the therapeutic gain of the cytotoxic agent, cisplatin, by incorporation of radioactive platinum. In this study, we investigated how organs at risk (i.e., kidneys, bone marrow, and liver) are affected by treatment with 191Pt-cisplatin, compared to treatment with conventional cisplatin. Rats (total, n = 69) were divided into three groups and given 5 mg/kg 191Pt-cisplatin and 5 mg/kg nonradioactive cisplatin or saline. The weight of the animals and blood samples, including analysis of creatinine, bilirubin, alanine and aspartate aminotransferases and platelet count, was followed for 6 weeks after treatment. Histopathology examinations of kidney and liver tissues were performed. An initial decrease in weight gain was seen from 3 days after treatment with cisplatin and 191Pt-cisplatin and for 1 week onward; thereafter, the weight gain continued, following the same pattern as for the control group. Concentration of plasma creatinine was increased for both cisplatin groups but with no significant difference between treatment groups. No other significant differences in effect parameters were found. There was no increase in toxicity for radioactive cisplatin on liver, kidneys, and bone marrow, compared to conventional cisplatin. Further experimental and clinical studies on preparations of this type are thus warranted.

  • Distribution of intraportally implanted microspheres and fluorescent islets in mice

    • The aim of the study was to evaluate the distribution of intraportally transplanted islets in mice. We initially administered 2000 polystyrene microspheres with a diameter of 50 microm intraportally into normoglycemic C57BL/6 mice. In separate experiments other mice were injected similarly with 300 microspheres each with a diameter of 100 or 200 microm. One week later the animals were killed, and the lungs and livers were removed and divided into lobes. The number of microspheres in each individual liver lobe and in the lungs was counted using a stereomicroscope. In other experiments, athymic C57BL/6 mice were similarly implanted with 250 islets isolated from transgenic mice expressing the enhanced yellow fluorescent protein in the islet cells. The distribution of microspheres and islets was independent of size, and fairly homogenous within the liver, with the exception of the caudate lobe, which contained fewer microspheres and islets, respectively. Approximately one third of all microspheres and islets were present as aggregates. Eighty-five to 90% of the implanted microspheres were identified in the liver sections, whereas 60-65% of the implanted islets were recovered. Aggregates or single fluorescent cells were observed in the liver of islet-implanted mice. We conclude that islets and microspheres implanted into the liver distribute fairly homogenously and quite a few of them exist as aggregates or, with respect to islets, as fragments.

  • Cost-effectiveness of conservation strategies implemented in boreal forests: The area selection process

    • To protect land from commercial exploitation is a common conservation practice. However, this requires large financial resources and it is therefore important to evaluate the cost-effectiveness of different strategies used in the selection of these conservation areas. In this study we compare four strategies and relate the differences in cost-effectiveness to differences in the selection process. We measure conservation benefits both as the amount of three tree structures and as the number of species in three species groups. We also estimate both the information cost associated with selecting conservation areas and the opportunity cost. we found the key habitat strategy to be the over-all most cost-effective. in this strategy, the areas have a flexible size and are selected by the authorities in a national field survey. The least cost-effective strategy was one where the selection was based only on forest classes in a satellite map. Intermediate were the retention group strategy, where small areas are left by the forest owner at harvesting, and the nature reserve strategy, where large areas are selected by the authorities. We emphasize that the differences we found are associated with the selection process and that other aspects, such as long-term survival of species, may rank the strategies differently. We conclude that the cost-effectiveness of a selection strategy depends on the size of the planning area for selection of conservation areas, the size of the conservation areas, the objective of the agent making the selection, and the amount and type of information on which the selection is based.
