  • The VLA/ALMA Nascent Disk and Multiplicity (VANDAM) Survey of Orion Protostars. II. A Statistical Characterization of Class 0 and Class i Protostellar Disks

    • We have conducted a survey of 328 protostars in the Orion molecular clouds with the Atacama Large Millimeter/submillimeter Array at 0.87 mm at a resolution of ∼0.″1 (40 au), including observations with the Very Large Array at 9 mm toward 148 protostars at a resolution of ∼0.″08 (32 au). This is the largest multiwavelength survey of protostars at this resolution by an order of magnitude. We use the dust continuum emission at 0.87 and 9 mm to measure the dust disk radii and masses toward the Class 0, Class I, and flat-spectrum protostars, characterizing the evolution of these disk properties in the protostellar phase. The mean dust disk radii for the Class 0, Class I, and flat-spectrum protostars are 44.9-3.4+5.8, 37.0-3.0+4.9, and 28.5-2.3+3.7 au, respectively, and the mean protostellar dust disk masses are 25.9-4.0+7.7, 14.9-2.2+3.8, 11.6-1.9+3.5 M⊙, respectively. The decrease in dust disk masses is expected from disk evolution and accretion, but the decrease in disk radii may point to the initial conditions of star formation not leading to the systematic growth of disk radii or that radial drift is keeping the dust disk sizes small. At least 146 protostellar disks (35% of 379 detected 0.87 mm continuum sources plus 42 nondetections) have disk radii greater than 50 au in our sample. These properties are not found to vary significantly between different regions within Orion. The protostellar dust disk mass distributions are systematically larger than those of Class II disks by a factor of >4, providing evidence that the cores of giant planets may need to at least begin their formation during the protostellar phase.

    • Sjögren’s syndrome (SS) is an exocrinopathy characterized by the hypofunction of salivary glands (SGs). Aquaporin-5 (AQP5); a water channel involved in saliva formation; is aberrantly dis-tributed in SS SG acini and contributes to glandular dysfunction. We aimed to investigate the role of ezrin in AQP5 mislocalization in SS SGs. The AQP5–ezrin interaction was assessed by immuno-precipitation and proteome analysis and by proximity ligation assay in immortalized human SG cells. We demonstrated, for the first time, an interaction between ezrin and AQP5. A model of the complex was derived by computer modeling and in silico docking; suggesting that AQP5 interacts with the ezrin FERM-domain via its C-terminus. The interaction was also investigated in human minor salivary gland (hMSG) acini from SS patients (SICCA-SS); showing that AQP5–ezrin complexes were absent or mislocalized to the basolateral side of SG acini rather than the apical region compared to controls (SICCA-NS). Furthermore, in SICCA-SS hMSG acinar cells, ezrin immunore-activity was decreased at the acinar apical region and higher at basal or lateral regions, accounting for altered AQP5–ezrin co-localization. Our data reveal that AQP5–ezrin interactions in human SGs could be involved in the regulation of AQP5 trafficking and may contribute to AQP5-altered localization in SS patients.

    • Saliva secretion requires effective translocation of aquaporin 5 (AQP5) water channel to the salivary glands (SGs) acinar apical membrane. Patients with Sjögren’s syndrome (SS) display abnormal AQP5 localization within acinar cells from SGs that correlate with sicca manifestation and glands hypofunction. Several proteins such as Prolactin-inducible protein (PIP) may regulate AQP5 trafficking as observed in lacrimal glands from mice. However, the role of the AQP5-PIP complex remains poorly understood. In the present study, we show that PIP interacts with AQP5 in vitro and in mice as well as in human SGs and that PIP misexpression correlates with an altered AQP5 distribution at the acinar apical membrane in PIP knockout mice and SS hMSG. Furthermore, our data show that the protein-protein interaction involves the AQP5 C-terminus and the N-terminal of PIP (one molecule of PIP per AQP5 tetramer). In conclusion, our findings highlight for the first time the role of PIP as a protein controlling AQP5 localization in human salivary glands but extend beyond due to the PIP-AQP5 interaction described in lung and breast cancers.

    • A microwave sensor to safely measure quality of muscle tissue for diagnosis and screening of diseases and medical conditions characterized by fat infiltration in muscle is presented. Fat infiltration in muscle may be seen by a lower dielectric constant of muscle at microwave frequencies corresponding to the large contrast between fat and muscle tissues. A planar resonator based on a bandstop filter and optimized to noninvasively interrogate muscle in the thigh on tissue quality is proposed. Currently, a study based on clinical trials is carried out, and, here, we present a preliminary correlation between skin and fat thicknesses and rectus femoris cross sectional area (CSA) measured with ultrasound and the proposed sensor's resonance frequency. CST simulations based on the ultrasound information guide the analysis. We see that although there are signs of a potential correlation between CSA and resonance, skin and fat variability is still an issue to overcome.

  • Mineral analysis reveals extreme manganese concentrations in wild harvested and commercially available edible termites

    • Termites are widely used as a food resource, particularly in Africa and Asia. Markets for insects as food are also expanding worldwide. To inform the development of insect-based foods, we analysed selected minerals (Fe-Mn-Zn-Cu-Mg) in wild-harvested and commercially available termites. Mineral values were compared to selected commercially available insects. Alate termites, of the genera Macrotermes and Odontotermes, showed remarkably high manganese (Mn) content (292-515 mg/100 gdw), roughly 50-100 times the concentrations detected in other insects. Other mineral elements occur at moderate concentrations in all insects examined. On further examination, the Mn is located primarily in the abdomens of the Macrotermes subhyalinus; with scanning electron microscopy revealing small spherical structures highly enriched for Mn. We identify the fungus comb, of Macrotermes subhyanus, as a potential biological source of the high Mn concentrations. Consuming even small quantities of termite alates could exceed current upper recommended intakes for Mn in both adults and children. Given the widespread use of termites as food, a better understanding the sources, distribution and bio-availability of these high Mn concentrations in termite alates is needed.

  • QUANTIFYING THE SUNK CARBON COSTS OF CITIES: A case study of 50 years of construction in Odense, Denmark

    • Constructing buildings and infrastructure in cities generates immense emissions of carbon dioxide. Understanding these emissions can aid in low-carbon urban planning and development. Current efforts to estimate these emissions use the Carbon Replacement Value method, which calculates the carbon cost of replacing the urban stock using current technologies. The CRV is useful for forecasting the carbon costs of near-term urban development. However, the perspective of historical emissions from technologies from the time of construction is missing. Quantifying historical emissions would improve our understanding of the embodied carbon spent to build the cities we live in today. This study proposes a methodology to estimate the Sunk Carbon Cost of urban material stocks. This method combines urban material cadastral maps with temporally dynamic life cycle assessment (LCA) of previous construction technologies to quantify and spatialize the historical carbon investment in the urban built form. We demonstrate the method by estimating the historical carbon emissions from concrete in all construction since 1961 (~105,000 individual buildings) in the city of Odense, Denmark. We estimate that 20.2 Mt of concrete was used in these buildings which released 3.0 Mt of carbon dioxide. We find that the sunk carbon cost for all construction since 1961 may be up to ~36% greater than estimates using carbon replacement value, but that these differences decrease for newer buildings. The spatial analysis identifies low-density settlement patterns as spatial hotspots of historical emissions, highlighting the material and carbon efficiencies of high-density neighborhoods. Applying this method to a city’s entire material stock can identify hotspots of embodied emissions across the urban fabric and suggest how to best utilize spent carbon through the adaptive reuse of existing buildings and low-carbon urban design.

  • Missing water in Class i protostellar disks

    • Context. Water is a key volatile that provides insight into the initial stages of planet formation. The low water abundances inferred from water observations toward low-mass protostellar objects may point to a rapid locking of water as ice by large dust grains during star and planet formation. However, little is known about the water vapor abundance in newly formed planet-forming disks. Aims. We aim to determine the water abundance in embedded Keplerian disks through spatially-resolved observations of H218O lines to understand the evolution of water during star and planet formation. Methods. We present H218O line observations with ALMA and NOEMA millimeter interferometers toward five young stellar objects. NOEMA observed the 31,3-22,0 line (Eup? kB = 203.7 K) while ALMA targeted the 41,4-32,1 line (Eup? kB = 322.0 K). Water column densities were derived considering optically thin and thermalized emission. Our observations were sensitive to the emission from the known Keplerian disks around three out of the five Class I objects in the sample. Results. No H218O emission is detected toward any of our five Class I disks. We report upper limits to the integrated line intensities. The inferred water column densities in Class I disks are NH218O < 1015 cm-2 on 100 au scales, which include both the disk and envelope. The upper limits imply a disk-averaged water abundance of ? 10-6 with respect to H2 for Class I objects. After taking the physical structure of the disk into account, the upper limit to the water abundance averaged over the inner warm disk with T > 100 K is between ~10-7 and 10-5. Conclusions. Water vapor is not abundant in warm protostellar envelopes around Class I protostars. Upper limits to the water vapor column densities in Class I disks are at least two orders of magnitude lower than values found in Class 0 disk-like structures.

  • MAS : Standalone Microwave Resonator to Assess Muscle Quality

    • Microwave-based sensing for tissue analysis is recently gaining interest due to advantages such as non-ionizing radiation and non-invasiveness. We have developed a set of transmission sensors for microwave-based real-time sensing to quantify muscle mass and quality. In connection, we verified the sensors by 3D simulations, tested them in a laboratory on a homogeneous three-layer tissue model, and collected pilot clinical data in 20 patients and 25 healthy volunteers. This report focuses on initial sensor designs for the Muscle Analyzer System (MAS), their simulation, laboratory trials and clinical trials followed by developing three new sensors and their performance comparison. In the clinical studies, correlation studies were done to compare MAS performance with other clinical standards, specifically the skeletal muscle index, for muscle mass quantification. The results showed limited signal penetration depth for the Split Ring Resonator (SRR) sensor. New sensors were designed incorporating Substrate Integrated Waveguides (SIW) and a bandstop filter to overcome this problem. The sensors were validated through 3D simulations in which they showed increased penetration depth through tissue when compared to the SRR. The second-generation sensors offer higher penetration depth which will improve clinical data collection and validation. The bandstop filter is fabricated and studied in a group of volunteers, showing more reliable data that warrants further continuation of this development.

  • Effects of aquatic exercise on appetitive responses in adolescents with obesity : An exploratory study

    • Aquatic exercise has been suggested as a beneficial modality to improve weight loss, cardiorespiratory fitness and quality of life in adolescents with obesity; however, its impact on appetite control in youth remains unknown. The aim of this preliminary study was to examine the effect of an acute aquatic exercise session on energy intake (EI), appetite feelings and food reward in adolescents with obesity. Twelve adolescents with obesity (12-16 years, Tanner stage 3-5, 9 males) randomly completed two conditions: i) control (CON); ii) aquatic exercise session (AQUA). One hour before lunch, the adolescents stayed at rest outside the water in a quiet room for 45 min on CON while they performed a 45-min aquatic exercise session on AQUA. Ad libitum EI and macronutrients were assessed at lunch and dinner, subjective appetite feelings taken at regular intervals, and food reward measured before and after lunch. Paired T-test showed that EI was not different between CON and AQUA at lunch (1333 +/- 484 kcal vs 1409 +/- 593 kcal; p = 0.162) and dinner (528 +/- 218 kcal vs 513 +/- 204 kcal; p = 0.206). Total daily ad libitum EI was significantly higher on AQUA (1922 +/- 649 kcal) compared with CON (1861 +/- 685 kcal; p = 0.044) but accounting for the exercise-induced energy expenditure, relative energy intake did not differ (2263 +/- 732 kcal vs 2117 +/- 744 kcal, p = 0.304). None of the appetite feelings (hunger, fullness, prospective food consumption and desire to eat) and food reward dimensions were significantly different between conditions. These preliminary and exploratory results suggest that an acute aquatic-exercise session might not induce energy compensatory responses in adolescents with obesity.

  • Robust Assembly of Cross-Linked Protein Nanofibrils into Hierarchically Structured Microfibers

    • Natural, high-performance fibers generally have hierarchically organized nanosized building blocks. Inspired by this, whey protein nanofibrils (PNFs) are assembled into microfibers, using flow-focusing. By adding genipin as a nontoxic cross-linker to the PNF suspension before spinning, significantly improved mechanical properties of the final fiber are obtained. For curved PNFs, with a low content of cross-linker (2%) the fiber is almost 3 times stronger and 4 times stiffer than the fiber without a cross-linker. At higher content of genipin (10%), the elongation at break increases by a factor of 2 and the energy at break increases by a factor of 5. The cross-linking also enables the spinning of microfibers from long straight PNFs, which has not been achieved before. These microfibers have higher stiffness and strength but lower ductility and toughness than those made from curved PNFs. The fibers spun from the two classes of nanofibrils show clear morphological differences. The study demonstrates the production of protein-based microfibers with mechanical properties similar to natural protein-based fibers and provides insights about the role of the nanostructure in the assembly process.
