  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Pan-cancer analysis of whole genomes

    • Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale1–3. Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained 4–5 driver mutations when combining coding and non-coding genomic elements; however, in around 5% of cases no drivers were identified, suggesting that cancer driver discovery is not yet complete. Chromothripsis, in which many clustered structural variants arise in a single catastrophic event, is frequently an early event in tumour evolution; in acral melanoma, for example, these events precede most somatic point mutations and affect several cancer-associated genes simultaneously. Cancers with abnormal telomere maintenance often originate from tissues with low replicative activity and show several mechanisms of preventing telomere attrition to critical levels. Common and rare germline variants affect patterns of somatic mutation, including point mutations, structural variants and somatic retrotransposition. A collection of papers from the PCAWG Consortium describes non-coding mutations that drive cancer beyond those in the TERT promoter4; identifies new signatures of mutational processes that cause base substitutions, small insertions and deletions and structural variation5,6; analyses timings and patterns of tumour evolution7; describes the diverse transcriptional consequences of somatic mutation on splicing, expression levels, fusion genes and promoter activity8,9; and evaluates a range of more-specialized features of cancer genomes8,10–18.

    • To identify new susceptibility loci to lung cancer among diverse populations, we performed cross-ancestry genome-wide association studies in European, East Asian and African populations and discovered five loci that have not been previously reported. We replicated 26 signals and identified 10 new lead associations from previously reported loci. Rare-variant associations tended to be specific to populations, but even common-variant associations influencing smoking behavior, such as those with CHRNA5 and CYP2A6, showed population specificity. Fine-mapping and expression quantitative trait locus colocalization nominated several candidate variants and susceptibility genes such as IRF4 and FUBP1. DNA damage assays of prioritized genes in lung fibroblasts indicated that a subset of these genes, including the pleiotropic gene IRF4, potentially exert effects by promoting endogenous DNA damage.

  • Modulating electron density of vacancy site by single Au atom for effective CO2 photoreduction

    • The surface electron density significantly affects the photocatalytic efficiency, especially the photocatalytic CO2 reduction reaction, which involves multi-electron participation in the conversion process. Herein, we propose a conceptually different mechanism for surface electron density modulation based on the model of Au anchored CdS. We firstly manipulate the direction of electron transfer by regulating the vacancy types of CdS. When electrons accumulate on vacancies instead of single Au atoms, the adsorption types of CO2 change from physical adsorption to chemical adsorption. More importantly, the surface electron density is manipulated by controlling the size of Au nanostructures. When Au nanoclusters downsize to single Au atoms, the strong hybridization of Au 5d and S 2p orbits accelerates the photo-electrons transfer onto the surface, resulting in more electrons available for CO2 reduction. As a result, the product generation rate of AuSA/Cd1−xS manifests a remarkable at least 113-fold enhancement compared with pristine Cd1−xS.

  • Global consortium for the classification of fungi and fungus-like taxa

    • The Global Consortium for the Classification of Fungi and fungus-like taxa is an international initiative of more than 550 mycologists to develop an electronic structure for the classification of these organisms. The members of the Consortium originate from 55 countries/regions worldwide, from a wide range of disciplines, and include senior, mid-career and early-career mycologists and plant pathologists. The Consortium will publish a biannual update of the Outline of Fungi and fungus-like taxa, to act as an international scheme for other scientists. Notes on all newly published taxa at or above the level of species will be prepared and published online on the Outline of Fungi website (https://www.outlineoffungi.org/), and these will be finally published in the biannual edition of the Outline of Fungi and fungus-like taxa. Comments on recent important taxonomic opinions on controversial topics will be included in the biannual outline. For example, 'to promote a more stable taxonomy in Fusarium given the divergences over its generic delimitation', or 'are there too many genera in the Boletales?' and even more importantly, 'what should be done with the tremendously diverse 'dark fungal taxa?' There are undeniable differences in mycologists' perceptions and opinions regarding species classification as well as the establishment of new species. Given the pluralistic nature of fungal taxonomy and its implications for species concepts and the nature of species, this consortium aims to provide a platform to better refine and stabilise fungal classification, taking into consideration views from different parties. In the future, a confidential voting system will be set up to gauge the opinions of all mycologists in the Consortium on important topics. The results of such surveys will be presented to the International Commission on the Taxonomy of Fungi (ICTF) and the Nomenclature Committee for Fungi (NCF) with opinions and percentages of votes for and against. Criticisms based on scientific evidence with regards to nomenclature, classifications, and taxonomic concepts will be welcomed, and any recommendations on specific taxonomic issues will also be encouraged; however, we will encourage professionally and ethically responsible criticisms of others' work. This biannual ongoing project will provide an outlet for advances in various topics of fungal classification, nomenclature, and taxonomic concepts and lead to a community-agreed classification scheme for the fungi and fungus-like taxa. Interested parties should contact the lead author if they would like to be involved in future outlines.

  • Rapid personalized AMR diagnostics using two-dimensional antibiotic resistance profiling strategy employing a thermometric NDM-1 biosensor

    • Antimicrobial resistance (AMR) threatens global public health and modern surgical medicine. Expression of β-lactamase genes is the major mechanism by which pathogens become antibiotic resistant. Pathogens expressing extended spectrum β-lactamases (ESBL) and carbapenemases (CP) are especially difficult to treat and are associated with increased hospitalization and mortality rates. Despite considerable effort, identification of ESBLs and CPs in a clinically relevant timeframe remains challenging. In this study, a two-dimensional AMR profiling assay strategy was developed employing panels of antibiotics (penicillins, cephamycins, oximino-cephalosporins and carbapenems) and β-lactamases inhibitors (avibactam and EDTA). The assay required the development of a novel biosensor that employed New Delhi metallo-β-lactamase-1 (NDM-1) as the sensing element. Functionally probing β-lactamase activity using substrates and inhibitors combinatorically increased the informational content that enabled the development of assays capable of simultaneous, differential identification of multiple β-lactamases expressed in a single bacterial isolate. More specifically, the assay enabled the simultaneous identification of ESBL and CP in mock samples, as well as in an engineered construct which co-expressed these β-lactamases. The NDM-1 biosensor assay was 16 times and 8 times more sensitive than the ESBL Nordmann/Dortet/Poirel (NDP) and Carba Nordmann/Poirel (NP) assays, respectively. In a retrospective study, NDM-1 biosensor assays were able to differentially identify ESBLs, metallo-CPs and serine-CPs β-lactamases in 23 clinical isolates with 100% accuracy. An assay algorithm was developed which accelerated data analytics reducing turnaround to <1 h. The assay strategy integrated with AI-based data analytics has the potential to provide physicians with a comprehensive readout of patient AMR status.

    • Rhenium(I)-carbonyl-diimine complexes have emerged as promising photocatalysts for carbon dioxide reduction with covalent organic frameworks recognized as perfect sensitizers and scaffold support. Such Re complexes/covalent organic frameworks hybrid catalysts have demonstrated high carbon dioxide reduction activities but with strong excitation energy-dependence. In this paper, we rationalize this behavior by the excitation energy-dependent pathways of internal photo-induced charge transfer studied via transient optical spectroscopies and time-dependent density-functional theory calculation. Under band-edge excitation, the excited electrons are quickly injected from covalent organic frameworks moiety into catalytic RheniumI center within picosecond but followed by fast backward geminate recombination. While under excitation with high-energy photon, the injected electrons are located at high-energy levels in RheniumI centers with longer lifetime. Besides those injected electrons to RheniumI center, there still remain some long-lived electrons in covalent organic frameworks moiety which is transferred back from RheniumI. This facilitates the two-electron reaction of carbon dioxide conversion to carbon monoxide.

  • Perceived professional benefits and their associated factors among Chinese registered nurses caring for women diagnosed with gynecological cancer

    • ProposeRegistered nurses (RNs) are considered to be a major source of professional supportive care for women diagnosed with gynecological cancer (GC). This study described the level of perceived professional benefits and explored association between perceived professional benefits, sense of coherence (SOC), and coping strategies in Chinese RNs caring for women diagnosed with GC.MethodA cross-sectional survey was employed to collect data using the Nurses' Perceived Professional Benefits Questionnaire (NPPBQ), Sense of coherence scale (SOC-13), and Brief Coping Orientation to Problems Experienced (Brief COPE). The questionnaires were administered to 250 RNs in China. The correlations between NPPBQ, SOC-13, and Brief COPE were evaluated with Pearson's correlation coefficient. Multiple regression analysis was performed to assess the relative contribution of each possible factor in explaining variance in the increased NPPBQ.ResultsTotal score for NPPBQ was 142.4 (range 33.0–165.0). SOC, dysfunctional coping strategies, and problem-focused coping strategies were recognized as predictors of RNs' perceived professional benefit, while, emotion-focused coping strategies were not significantly associated with RNs’ perceived professional benefits.ConclusionsThe findings indicate that RNs who have high levels of SOC, dysfunctional coping strategies, and problem-focused coping strategies tend to experience more perceived professional benefit. These findings propose new perspectives for nursing managers to promote RNs' perceived professional benefit by helping RNs to find meaningfulness when caring for women diagnosed with GC, increasing RNs’ communication skills to improve their relationship with patients, and training RNs to use coping strategies effectively.

    • Real-time monitoring of patient’s blood metabolites, such as glucose and lactate, could potentially improve surgery and recovery outcomes for patients in surgical and intensive care units. Our enzyme thermometric biosensor which is based on flow injected calorimetric determination of immobilized enzyme reaction is capable of performing continuous, fast, and quantitative analysis of metabolites using whole blood. A key technical advantage the assay affords is the ability to use unpretreated whole blood. In this article, the enzyme thermometric biosensor was used, for the first time, to determine glucose and lactate concentrations in the blood of ICU patients. The linear detection range for glucose was 0.5–30 mM and 0.25–12 mM for lactate, using a 20 μL sample volume. A maximum sampling rate of 15 measurements per hour was achieved using venous blood samples, which corresponds to a 4-min measurement interval. In order to validate the accuracy of the results, a comparative analysis between the thermometric biosensor and the clinically applied instrument (LifeScan’s OneTouch®) which is based on disposable dry chemical reaction was performed using samples from 33 patients. The results showed a good correlation between the two methods for both glucose (r = 0.843, p < 0.0001) and lactate (r = 0.78, p = 0.0105). The ability to monitor metabolite levels and trends on a clinically relevant timescale of 5 min is critical for intensive monitoring of ICP and operative patients.

    • Flotation separation, which is the most important mineral beneficiation technique, is dependent on gas dispersion (hydrodynamic conditions). Thus, many investigations have focused on the precise determination of hydrodynamic conditions such as Reynolds number of the bubbles, bubble velocity, and bubble diameter. However, few studies have examined their relationships with pressure fluctuations in a column flotation. This study introduced the differential pressure fluctuations as an actual variable that could be considered to determine the collection zone’s hydrodynamic conditions in a cyclonic microbubble flotation column. In general, the outcomes indicated that superficial gas velocity had the most substantial relationship with the differential pressure fluctuations among other flotation factors (such as pump speed, superficial gas velocity, superficial water velocity, and frother dosage). Furthermore, a high coefficient of determination (R2 > 0.77) for the equation generated to assess the relationships demonstrated that differential pressure fluctuations could be used as a promising tool to determine the hydrodynamic parameters’ characteristics in the flotation columns.

    • Joint 2D cardiac segmentation and 3D volume reconstruction are fundamental in building statistical cardiac anatomy models and understanding functional mechanisms from motion patterns. However, due to the low through-plane resolution of cine MR and high inter-subject variance, accurately segmenting cardiac images and reconstructing the 3D volume are challenging. In this study, we propose an end-to-end latent-space-based framework, DeepRecon, that generates multiple clinically essential outcomes, including accurate image segmentation, synthetic high-resolution 3D image, and 3D reconstructed volume. Our method identifies the optimal latent representation of the cine image that contains accurate semantic information for cardiac structures. In particular, our model jointly generates synthetic images with accurate semantic information and segmentation of the cardiac structures using the optimal latent representation. We further explore downstream applications of 3D shape reconstruction and 4D motion pattern adaptation by the different latent-space manipulation strategies. The simultaneously generated high-resolution images present a high interpretable value to assess the cardiac shape and motion. Experimental results demonstrate the effectiveness of our approach on multiple fronts including 2D segmentation, 3D reconstruction, downstream 4D motion pattern adaption performance.

  • The JCMT BISTRO-2 Survey: Magnetic Fields of the Massive DR21 Filament

    • We present 850 mu m dust polarization observations of the massive DR21 filament from the B-fields In STar-forming Region Observations (BISTRO) survey, using the POL-2 polarimeter and the SCUBA-2 camera on the James Clerk Maxwell Telescope. We detect ordered magnetic fields perpendicular to the parsec-scale ridge of the DR21 main filament. In the subfilaments, the magnetic fields are mainly parallel to the filamentary structures and smoothly connect to the magnetic fields of the main filament. We compare the POL-2 and Planck dust polarization observations to study the magnetic field structures of the DR21 filament on 0.1-10 pc scales. The magnetic fields revealed in the Planck data are well-aligned with those of the POL-2 data, indicating a smooth variation of magnetic fields from large to small scales. The plane-of-sky magnetic field strengths derived from angular dispersion functions of dust polarization are 0.6-1.0 mG in the DR21 filament and similar to 0.1 mG in the surrounding ambient gas. The mass-to-flux ratios are found to be magnetically supercritical in the filament and slightly subcritical to nearly critical in the ambient gas. The alignment between column density structures and magnetic fields changes from random alignment in the low-density ambient gas probed by Planck to mostly perpendicular in the high-density main filament probed by James Clerk Maxwell Telescope. The magnetic field structures of the DR21 filament are in agreement with MHD simulations of a strongly magnetized medium, suggesting that magnetic fields play an important role in shaping the DR21 main filament and subfilaments.

    • A major subset of solar-terrestrial relations, responsible, in particular, for the driver of space weather phenomena, is the interaction between the Earth's magnetosphere and the solar wind. As one of the most important modes of the solar-wind-magnetosphere interaction, magnetic reconnection regulates the energy transport and energy release in the solar-terrestrial relation. In situ measurements in the near-Earth space are crucial for understanding magnetic reconnection. Past and existing spacecraft constellation missions mainly focus on the measurement of reconnection on plasma kinetic-scales. Resolving the macro-scale and cross-scale aspects of magnetic reconnection is necessary for accurate assessment and predictions of its role in the context of space weather. Here, we propose the AME (self-Adaptive Magnetic reconnection Explorer) mission consisting of a cross-scale constellation of 12+ CubeSats and one mother satellite. Each CubeSat is equipped with instruments to measure magnetic fields and thermal plasma particles. With multiple CubeSats, the AME constellation is intended to make simultaneous measurements at multiple scales, capable of exploring cross-scale plasma processes ranging from kinetic scale to macro scale.

    • Integration of high activity, selectivity, and stability is urgently desired to achieve more ideal photocatalysts. Herein, we reported the rational design of MoS2-MnS@(InxCu1-x)2S3 (M-M@IC) catalysts with dual interface to integrate separated redox sites for boosting photocatalytic hydrogen sulphide (H2S) splitting and the resource utilization of sacrificial reagents (Na2S/Na2SO3). The spatially separated reduction (MnS) and oxidation (In2S3) sites in MnS/In2S3 heterojunction, on which MoS2 and Cu were selectively loaded, can drive electrons and holes near the surface to flow along opposite directions, while the heterojunction between MnS and In2S3 inhibits the bulk charge recombination. Furthermore, the introduction of Cu atoms creates a d-band center, which favours mass diffusion of reactants/products species and greatly facilitates sunlight response. The MoS2 serves to provide abundant sites for proton reduction due to the unsaturated-sulfur-edge-rich (US-rich) nature. As a result, the M−M@IC shows a state-of-the-art visible-light photocatalytic H2 evolution rate (126.5 mmol g-1h−1), inspiring stability of >50 h, and nearly 100% selectivity toward value-added Na2S2O3 production under optimized condition. This work opens up new opportunities for the construction and design of spatially separated catalytic site in photocatalysts.

    • Simultaneous hydrogen (H2) evolution and value−added chemicals production are highly attractive but have not drawn enough attention. Here, we demonstrate a hydrogen sulphide (H2S)−induced product−targeting (HIPT) strategy for the coproduction of H2 and valuable chemical feedstocks from Na2S/Na2SO3 via overall H2S splitting using a MnxCd1−xS/CdyMn1−yS catalyst driven by visible light excitation. With this chemistry, 113 mmol g−1 h−1 of hydrogen evolution rate is achieved, surpassing most of the previously reported state-of-the-art photocatalyst, together with the production of value−added Na2S2O3 with nearly 100% selectivity. This work not only provides a good example for solar energy conversion via overall H2S splitting, but also offers new insights into the resource utilization of sacrificial donor (Na2S/Na2SO3) in various catalytic fields such as H2O splitting and CO2 reduction.

    • Whole-genome sequencing projects are increasingly populating the tree of life and characterizing biodiversity(1-4). Sparse taxon sampling has previously been proposed to confound phylogenetic inference(5), and captures only a fraction of the genomic diversity. Here we report a substantial step towards the dense representation of avian phylogenetic and molecular diversity, by analysing 363 genomes from 92.4% of bird families-including 267 newly sequenced genomes produced for phase II of the Bird 10,000 Genomes (B10K) Project. We use this comparative genome dataset in combination with a pipeline that leverages a reference-free whole-genome alignment to identify orthologous regions in greater numbers than has previously been possible and to recognize genomic novelties in particular bird lineages. The densely sampled alignment provides a single-base-pair map of selection, has more than doubled the fraction of bases that are confidently predicted to be under conservation and reveals extensive patterns of weak selection in predominantly non-coding DNA. Our results demonstrate that increasing the diversity of genomes used in comparative studies can reveal more shared and lineage-specific variation, and improve the investigation of genomic characteristics. We anticipate that this genomic resource will offer new perspectives on evolutionary processes in cross-species comparative analyses and assist in efforts to conserve species. A dataset of the genomes of 363 species from the Bird 10,000 Genomes Project shows increased power to detect shared and lineage-specific variation, demonstrating the importance of phylogenetically diverse taxon sampling in whole-genome sequencing.

    • BackgroundMany ultraprocessed food (UPF)-derived by-products may play a role in the development of chronic kidney disease (CKD). Although several studies have assessed the association of UPFs with kidney function decline or CKD in various countries, no evidence has been shown in China and the United Kingdom.ObjectivesThis study aims to evaluate the association between UPF consumption and risk of CKD in 2 large cohort studies from China and the United Kingdom.MethodsIn total, 23,775 and 102,332 participants without baseline CKD were enrolled in the Tianjin Chronic Low-Grade Systemic Inflammation and Health (TCLSIH) and UK Biobank cohort studies, respectively. Information on UPF consumption was obtained from a validated food frequency questionnaire in the TCLSIH and 24-h dietary recalls in the UK Biobank cohort. CKD was defined as an estimated glomerular filtration rate of ResultsAfter a median follow-up of 4.0 and 10.1 y, the incidence rates of CKD were around 1.1% and 1.7% in the TCLSIH and UK Biobank cohorts, respectively. The multivariable hazard ratio [95% confidence interval] of CKD across increasing quartiles (quartiles 1–4) of UPF consumption were 1 (reference), 1.24 (0.89, 1.72), 1.30 (0.91, 1.87), and 1.58 (1.07, 2.34) (P for trend = 0.02) in the TCLSIH cohort and 1 (reference), 1.14 (1.00, 1.31), 1.16 (1.01, 1.33), and 1.25 (1.09, 1.43) (P for trend < 0.01) in the UK Biobank cohort, respectively.ConclusionsOur finding indicated that higher UPF consumption is associated with a higher risk of CKD. Moreover, restricting UPF consumption may potentially benefit the prevention of CKD. Further clinical trials are required to clarify the causality.

    • For polar ship operating in ice-covered areas, numerical prediction and relevant research on maneuverability are particularly important. In this paper, a numerical method considering circumferential crack fracture of level ice based on former research is applied to calculate ice load and predict motion of ship turning in level ice. The difference between bending failure and crushing failure are also taken into consideration during the turning simulation, meanwhile crushing failure pattern is validated with measured data. The turning performance of icebreaker AHTS/IB Tor Viking Ⅱ based on present method is analyzed and compared with full scale measurement. The simulated icebreaking capability of the ship is also validated with sea trials and previous work. The results show that good agreement is achieved and the accuracy of the numerical prediction is improved by including crushing failure into previous research.

    • Three nearly degenerate pairs of doublet bands are identified in Ba-131. Two of them, with positive-parity, are interpreted as pseudospin-chiral quartet bands. This is the first time that a complete set of chiral doublet bands built on the pseudospin partners pi(d(5/2), g(7/2)) is observed. The chiral bands with opposite parity built on 3-quasiparticle configurations are directly connected by many E1 transitions, without involving an intermediary non-chiral configuration. The observed band structures in Ba-131 have been investigated by using the reflection-asymmetric particle rotor model. The energies and the electromagnetic transition ratios of the three pairs of doublet bands observed in Ba-131 are reproduced and they are interpreted as chiral doublet bands with three-quasiparticle configurations. It is the first time that multiple chiral bands are observed in the presence of enhanced octupole correlations and pseudospin symmetry.

    • This study focuses on one of the critical design parameters of lean premixed (LPM) combustors-the air split ratio (ASR) between the pilot and main stages. The effect of the air split ratio on thermoacoustic instability is experimentally studied based on a centrally staged model combustor. It is found that the thermoacoustic instability is suppressed with the ASR higher than 20%. The acoustic modes of ASR lower than 20% are dominated by the main stage mode, while the others are controlled by the pilot stage mode. The time-averaged flame shapes and the flame dynamics are processed and discussed. With the increase in ASR, the pilot flame becomes longer, whereas the main flame becomes shorter. Meanwhile, the pilot and main flames become less separated. Moreover, the flame in the outer shear layer (OSL) is extinguished, which is the key to suppressing the thermoacoustic instability. The flame with lower ASR exhibits large-scale periodic axial motion of flame dynamics. The flow fields of ASR = 15% (unstable) and ASR = 30% (stable) are analyzed and discussed to help reveal the differences in thermoacoustic instability between the two cases. It is found that the primary recirculation zone is smaller with ASR = 30%. Higher vorticity and strain rate distributions found in the OSL are believed to quench the flame there. The present study illustrates that a higher air split ratio improves the thermoacoustic stability, which is helpful in designing LPM combustors.

This paper proposes a numerical model for analyzing coursestability of a gravity-based structure during towing operation in managed ice fields, considering coupledmotions of tugs, towed platform, and ice floes. A discrete element method is applied to simulateinteraction between drifting ice floes and structures, and verified by existing model tests. The motions oftug and towed platform are coupled through a catenary towline model. A proportional-differentialcontroller is employed to adjust heading and lateral position of tug. The effects of main factors, such astugs’ number, towline length, ice concentration, rudder area and controller gains on course stability arediscussed through numerical case studies. The presented numerical approach is expected to providepractical guidance for towing design, and ensure safety of towing operation.

    • Quantifying the relationship between pollen and vegetation is an essential step in the pollen-based quantitative reconstruction of past vegetation cover. In this study, we use the Extended R-Value (ERV) model and a modern dataset of pollen (collected from moss polsters) and related vegetation from 50 sites in the Daba Mountains (subtropical China) to (i) estimate the relevant source area of pollen (RSAP) of the moss samples and the relative pollen productivities (RPPs) of nine major plant taxa-characteristic of the region, and (ii) evaluate the obtained RPPs. The RSAP estimates of moss polsters vary between 225 and 610 m depending on the ERV submodels and models of pollen dispersal and deposition used. The RPP estimates are different from values published in previous studies from temperate and subtropical China. This may be explained by differences in methodology, climate and vegetation (species composition and spatial distribution), of which vegetation is probably the most important factor. The ranking of the RPP estimates for the nine taxa is Pinus > Juglandaceae > D - Quercus (deciduous Quercus) > Poaceae > Rosaceae > Cyperaceae > Anacardiaceae > Castanea > Fabaceae. We use a 'leave-one-out' cross-validation strategy and the Landscape Reconstruction Algorithm (LRA) for pollen-based reconstruction of regional and local plant cover to evaluate the ERV model-based RPP estimates. Both the REVEALS (Regional Estimates of VEgetation Abundance from Large Sites)-based and the LOVE (LOcal Vegetation Estimates)-based plant cover using the RPP estimates are closer to the modern vegetation composition than pollen percentages, thus confirming the applicability of the ERV model and the LRA approach in subtropical China.

    • We present observations of polarized dust emission at 850 mu m from the L43 molecular cloud, which sits in the Ophiuchus cloud complex. The data were taken using SCUBA-2/POL-2 on the James Clerk Maxwell Telescope as a part of the BISTRO large program. L43 is a dense (N-H2 similar to 10(22) - 10(23) cm(-2)) complex molecular cloud with a submillimeter-bright starless core and two protostellar sources. There appears to be an evolutionary gradient along the isolated filament that L43 is embedded within, with the most evolved source closest to the Sco OB2 association. One of the protostars drives a CO outflow that has created a cavity to the southeast. We see a magnetic field that appears to be aligned with the cavity walls of the outflow, suggesting interaction with the outflow. We also find a magnetic field strength of up to similar to 160 +/- 30 mu G in the main starless core and up to similar to 90 +/- 40 mu G in the more diffuse, extended region. These field strengths give magnetically super- and subcritical values, respectively, and both are found to be roughly trans-Alfvenic. We also present a new method of data reduction for these denser but fainter objects like starless cores.

    • Truck platooning has gained increasing attention due to the benefits in energy and operation efficiency in freight transportation. One significant challenge for deploying truck platoons is the safe and efficient interaction with surrounding traffic, especially at freeway discontinuities where mandatory lane changes usually lead to the decoupling of truck platoons. This study proposes a cooperative truck platoon lane-changing model (CTPLC) to prevent the decoupling of truck platoons in a mixed traffic environment. Specifically, a two-step control strategy is presented, where vehicles in the target lane firstly cooperatively adjust speeds to create an appropriate gap for a truck platoon, and then trucks within the truck platoon conduct lane change sequentially. The cooperative speed profiles are generated by solving an optimization problem considering the lane-changing influence and energy consumption. Based on that, a two-dimensional nonlinear model predictive control (MPC) algorithm is employed to generate vehicular acceleration and steering angle for each truck. A series of numerical simulation experiments were conducted to validate the proposed strategy. As shown by the results, our proposed method truck platoon could conduct a lane change in a traffic-efficient and safe manner, and meanwhile, our method was more energy-efficient than a benchmark strategy.

    • Sighthounds, a distinctive group of hounds comprising numerous breeds, have their origins rooted in ancient artificial selection of dogs. In this study, we performed genome sequencing for 123 sighthounds, including one breed from Africa, six breeds from Europe, two breeds from Russia, and four breeds and 12 village dogs from the Middle East. We gathered public genome data of five sighthounds and 98 other dogs as well as 31 gray wolves to pinpoint the origin and genes influencing the morphology of the sighthound genome. Population genomic analysis suggested that sighthounds originated from native dogs independently and were comprehensively admixed among breeds, supporting the multiple origins hypothesis of sighthounds. An additional 67 published ancient wolf genomes were added for gene flow detection. Results showed dramatic admixture of ancient wolves in African sighthounds, even more than with modern wolves. Whole-genome scan analysis identified 17 positively selected genes (PSGs) in the African population, 27 PSGs in the European population, and 54 PSGs in the Middle Eastern population. None of the PSGs overlapped in the three populations. Pooled PSGs of the three populations were significantly enriched in "regulation of release of sequestered calcium ion into cytosol"(gene ontology: 0051279), which is related to blood circulation and heart contraction. In addition, ESR1, JAK2, ADRB1, PRKCE, and CAMK2D were under positive selection in all three selected groups. This suggests that different PSGs in the same pathway contributed to the similar phenotype of sighthounds. We identified an ESR1 mutation (chr1: g.42,177,149-T > C) in the transcription factor (TF) binding site of Stat5a and a JAK2 mutation (chr1: g.93,277,007-T > A) in the TF binding site of Sox5. Functional experiments confirmed that the ESR1 and JAK2 mutation reduced their expression. Our results provide new insights into the domestication history and genomic basis of sighthounds.

    • Partial discharge (PD) test has been widely used in condition assessment for power cables. However, discharge extinguish phenomenon occurred in PD detection hinders the effective detection of potential defects in cable accessories. For this reason, a novel PD detection method based on thermal excitation is proposed in this paper to eliminate the extinguish phenomenon and to detect the potential defects in earlier stage. First, the physical processes in PD activity and the reasons for discharge extinguishing are introduced. Then, the principle and mechanism of the thermal excitation detection method are described and analyzed respectively, that is excitation by external heating to break the dynamic equilibrium of charge movement within the defect and further stimulate new discharges. And a finite element simulation model is constructed to determine the thermal excitation parameters. Finally, the effectiveness is validated by performing thermal excitation-based PD test on some defective cables. The experimental results show that this method can significantly increase the number of discharges in off-line test, while also stimulating PD of higher amplitudes in on-line monitoring. This method can improve the sensitivity of PD detection for cable accessories and detect potential defects in early stage.

    • Hexagonal boron nitride (hBN) has recently been demonstrated to contain optically polarized and detected electron spins that can be utilized for implementing qubits and quantum sensors in nanolayered-devices. Understanding the coherent dynamics ofmicrowave driven spins in hBN is of crucial importance for advancing these emerging new technologies. Here, we demonstrate and study the Rabi oscillation and related phenomena of a negatively charged boron vacancy (V-B(-)) spin ensemble in hBN. We report on different dynamics of the V-B(-) spins at weak and strong magnetic fields. In the former case the defect behaves like a single electron spin system, while in the latter case it behaves like a multi-spin system exhibiting multiple-frequency dynamical oscillation as beat in the Ramsey fringes. We also carry out theoretical simulations for the spin dynamics of V-B(-) and reveal that the nuclear spins can be driven via the strong electron nuclear coupling existing in V-B(-) center, which can be modulated by the magnetic field and microwave field.

Stronger error correction code (ECC) and adaptive reading strategies are proposed to improve the reliability and performance taking a threshold voltage (Vth) distribution model as the backbone. However, the existing modeling methods are challenged to develop such a Vth distribution model for 3D NAND flash memory. To facilitate it, in this paper, we propose a machine learning-based modeling method. It employs a neural network taking advantage of the existing modeling methods and fully considers multiple interferences and variations in 3D NAND flash memory. Compared with state-of-the-art models, evaluations demonstrate it is more accurate and efficient for predicting Vth distribution.

    • In this work, we explored the photoinduced charge carriers dynamics rationalizing the photocatalytic oxidation of NO over N-doped Bi2O2CO3/graphene quantum dots composites(N-BOC/GQDs) via time-resolved photoluminescence (TRPL). Under visible light illumination, only GQDs can be photoexcited and inject electrons to N-BOC within 0.5 ns. Under UV light irradiation, the interfacial Z-scheme heterojunction recombination between the electrons in N-BOC and holes in GQDs dominate the depopulation of excited states within 0.36 ns. Such efficient Z-scheme recombination regardless of the large energy difference (1.66 eV) is mediated by the interfacial oxygen vacany defect states characterized by both density functional theory calculations (DFT) and electron paramagnetic resonance (EPR) measurement. This finding provide a novel strategic view to improve the photocatalytic performance of the nanocomposite by interfacial engineering

    • Photoinduced carrier dynamical processes dominate the optical excitation properties of photocatalysts and further determine the photocatalytic performance. In addition, as the electrons generally possess a faster transfer rate than holes, hole transfer and accumulation are critical, and they play the key efficiency-limiting step during the photocatalytic process. Therefore, a comprehensive understanding of the dynamics of photogenerated holes and their determining factors in the photocatalytic system is highly essential to rationalize the full catalytic mechanism and develop highly efficient photocatalysts, which have not yet been revealed. In this work, the photoinduced charge carrier dynamics in InP/ZnS quantum dots (QDs) capped with long-chain L-typed ligands (oleylamine) and inorganic ligands (sulfide ion (S2−)) were explored. Time-resolved photoluminescence and femtosecond transient-absorption spectroscopy unambiguously confirmed the ultrafast hole transfer from the InP core to S2− ligands. Moreover, by probing the bleach of vibrational stretching of the ligands with transient midinfrared absorption spectroscopy, the hole transfer time was determined to be 4.2 ps. The injected holes are long-lived at the S2− ligands (>4.5 ns), and they can remove electrostatically attached surfactants to compensate for the spatial charge redistribution. Finally, compared with other inorganic ligands such as Cl− and PO43−, S2− balances the ionic radii and net charge to ensure the optimal condition for charge transfer. Such observation rationalizes the excellent photocatalytic H2 evolution (213.6 µmol mg−1 within 10 h) in InP/ZnS QDs capped with S2− compared with those capped with other ligands and elucidates the role of surface ligands in the photocatalytic activity of colloidal QDs.

    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

    • In recent years, lactate has been recognized as an important circulating energy substrate rather than only a dead-end metabolic waste product generated during glucose oxidation at low levels of oxygen. The term "aerobic glycolysis" has been coined to denote increased glucose uptake and lactate pro-duction despite normal oxygen levels and functional mito-chondria. Hence, in "aerobic glycolysis," lactate production is a metabolic choice, whereas in "anaerobic glycolysis," it is a metabolic necessity based on inadequate levels of oxygen. Interestingly, lactate can be taken up by cells and oxidized to pyruvate and thus constitutes a source of pyruvate that is in-dependent of insulin. Here, we show that the transcription factor Foxp1 regulates glucose uptake and lactate production in adipocytes and myocytes. Overexpression of Foxp1 leads to increased glucose uptake and lactate production. In addition, protein levels of several enzymes in the glycolytic pathway are upregulated, such as hexokinase 2, phosphofructokinase, aldolase, and lactate dehydrogenase. Using chromatin immu-noprecipitation and real-time quantitative PCR assays, we demonstrate that Foxp1 directly interacts with promoter consensus cis-elements that regulate expression of several of these target genes. Conversely, knockdown of Foxp1 suppresses these enzyme levels and lowers glucose uptake and lactate production. Moreover, mice with a targeted deletion of Foxp1 in muscle display systemic glucose intolerance with decreased muscle glucose uptake. In primary human adipocytes with induced expression of Foxp1, we find increased glycolysis and glycolytic capacity. Our results indicate Foxp1 may play an important role as a regulator of aerobic glycolysis in adipose tissue and muscle.

The crash mechanism of a consecutive crash series is distinctive, as it is different from common primary and secondary crashes mainly caused by queuing effects and chain-reaction crashes that involve multiple collisions in one crash. It commonly affects a large area of road space and possibly causes congestions and significant delays in evacuation and clearance. This study identified the influential factors determining the severity of primary and secondary crashes in a consecutive crash series. Basic, random-effects, random-parameters, and two-level binary logistic regression models were established based on crash data collected on the freeway network of Guizhou Province, China in 2018, of which 349 were identified as consecutive crashes. According to the model performance metrics, the two-level logistic model outperformed the other three models. On the crash level, double-vehicle primary crash had a negative association with the severity of secondary consecutive crashes, and the involvement of trucks in the secondary consecutive crash had a positive contribution to its crash severity. On a road segment level, speed limit, traffic volume, tunnel, and extreme weather conditions such as rainy and cloudy days had positive effects on consecutive crash severity, while the number of lanes was negatively associated with consecutive crash severity. Policy suggestions are made to alleviate the severity of consecutive crashes by reminding the drivers with real-time potential hazards of severe consecutive crashes and providing educative programs to specific groups of drivers.

  • Numerical simulation of slipstreams and wake flows of trains with different nose lengths passing through a tunnel

    • This study examined the slipstreams induced by high-speed trains (HSTs) passing through a tunnel using the improved delayed detached eddy simulation (IDDES) method. First, the flow fields in the open air and in a tunnel were compared. Furthermore, the flow in a tunnel was analyzed in detail, considering the development of both instantaneous flow structures and slipstream profiles at various measurement points. Finally, by considering four different nose lengths (4 m, 7 m, 9 m, and 12 m), the differences in the slipstream profiles and the wake flow induced by HSTs passing through a tunnel were determined. The results show that the piston effect had a significant influence on the slipstream profiles, causing a larger positive peak when a train passed through a tunnel. The peaks of the slipstream profiles decrease as the distances from the center of the track (COT) and the top of the rail (TOR) increases. The results show that a long nose length can reduce the scale and strength of the instantaneous x-vorticity and y-vorticity in the wake propagation region, thereby lowering the maximum slipstream peaks. The 12-m nose length train induced 56.7% lower velocity than the 4-m nose length train at y = 2 m beside the COT and z = 0.2 m above the TOR. In particular, the standard deviations of the positive peaks of the seven cross-sections decrease by 38.4% with the increase in the nose length from 4 m to 12 m, which means that a longer nose length can reduce the turbulence level in the wake propagation region. Consequently, from the perspectives of the safety and comfort of trackside people, a long nose length train is strongly recommended.

  • Expressions of miR-302a, miR-105, and miR-888 Play Critical Roles in Pathogenesis, Radiotherapy, and Prognosis on Rectal Cancer Patients : A Study From Rectal Cancer Patients in a Swedish Rectal Cancer Trial of Preoperative Radiotherapy to Big Database Analyses

    • Differential expressions and functions of various micoRNAs (miRNAs) have been intensively studied in both colon and rectal cancers. However, the importance of miRNAs on radiotherapy (RT) response and clinical outcome in rectal cancer patients remains unclear. In this study, we used real-time polymerase chain reaction to examine the expressions of miR-302a, miR-105, and miR-888 in normal mucosa and cancer tissue from rectal cancer patients with and without preoperative RT. The biological function of miR-302a, miR-105, and miR-888 expression was further analyzed and identified through the public databases: TCGA (The Cancer Genome Atlas) and GEPIA (Gene Expression Profiling Interactive Analysis). The results showed that the expression of miR-105 in rectal cancer was higher than that in normal mucosa in RT (P = 0.042) and non-RT patients (P = 0.003) and was associated with mucinous histological type (P = 0.004), COX-2 (P = 0.042), and p73 expression (P = 0.030). The expression of miR-302a was shown more frequently in cancers with necrosis (P = 0.033) and with WRAP53 expression (P = 0.015), whereas miR-888 expression occurred more frequently in tumors with protein the expression of survivin (P = 0.015), AEG-1 (astrocyte elevated gene-1) (P = 0.003), and SATB1 (special AT-rich sequence binding protein 1) (P = 0.036). Moreover, TargetScan also predicted AEG-1 and SATB1 as putative targets for miR-888. The miRNA-gene network analysis showed that ABI2 was associated with all the three miRNAs, with lower expression and good diagnostic value in rectal cancers. The TCGA database demonstrated the association of miR-105 expression with high carcinoembryonic antigen level (P = 0.048). RT reduced the expressions of miR-302a, miR-105, and miR-888. Prognostic analysis showed that miR-888 expression was independently associated with worse survival of patients without RT [overall survival, P = 0.001; disease-free survival, P = 0.009]. Analysis of biological function revealed that the protein serine/threonine kinase activity and PI3K-AKT signaling pathway were the most significantly enriched functions and pathways, respectively. Our findings suggest that miR-105 is involved in rectal cancer pathogenesis and miR-888 is associated with prognosis. MiR-302a, miR-105, and miR-888 have potential influence on the pathogenesis, RT, and prognosis of rectal cancer.

    • In the present work, irregular Cu nanoparticle-decorated boron-carbon-nitrogen (Cu-BCN) nanosheets were successfully synthesized. A Cu-BCN dispersion was deposited on a bare glassy carbon electrode (GCE) to prepare an electrochemical sensor (Cu-BCN/GCE) for the detection of chloramphenicol (CAP) in the environment. Cu-BCN was characterized using high-resolution scanning transmission electron microscopy (HRSTEM), scanning electron microscopy (SEM), X-ray diffraction (XRD) analysis, and X-ray photoelectron spectroscopy (XPS). The performance of the Cu-BCN/GCE was studied using electrochemical impedance spectroscopy (EIS), and its advantages were proven by electrode comparison. Differential pulse voltammetry (DPV) was used to optimize the experimental conditions, including the amount of Cu-BCN deposited, enrichment potential, deposition time, and pH of the electrolyte. A linear relationship between the CAP concentration and current response was obtained under the optimized experimental conditions, with a wide linear range and a limit of detection (LOD) of 2.41 nmol/L. Cu-BCN/GCE exhibited high stability, reproducibility, and repeatability. In the presence of various organic and inorganic species, the influence of the Cu-BCN-based sensor on the current response of CAP was less than 5%. Notably, the prepared sensor exhibited excellent performance in real-water samples, with satisfactory recovery.

  • Multiple chiral bands in 137 Nd

    • Two new bands have been identified in 137Nd from a high-statistics JUROGAM II gamma-ray spectroscopy experiment. Constrained density functional theory and particle rotor model calculations are used to assign configurations and investigate the band properties, which are well described and understood. It is demonstrated that these two new bands can be interpreted as chiral partners of previously known three-quasiparticle positive- and negative-parity bands. The newly observed chiral doublet bands in 137Nd represent an important support to the existence of multiple chiral bands in nuclei. The present results constitute the missing stone in the series of Nd nuclei showing multiple chiral bands, which becomes the most extended sequence of odd–even and even-even nuclei presenting multiple chiral bands in the Segré chart.

    • This article focuses on design of an Artificial Neural Network (ANN) model to estimate ship resistance in ice-covered water by using suitable ship and ice parameters. In order to develop a reliable model, as much ice resistance test data as from the ship sea trials and model test measurements are collected to train the neural network. Different features (ship design parameters and ice mechanic properties) are explored to find a suitable combination of input features. Several algorithms are tested and compared to select a good model for resistance prediction. It turns out that seven features and the Radial Basis Function - Particle Swarm Optimization algorithm (RBF-PSO) can provide a reasonable generalization model. This study shows that the ice resistance predicted by the ANN correlates well with the measured data. The model developed herein can be used as an ice resistance prediction tool with high accuracy compared to the conventional semi-empirical formulae used in polar ship design.

    • Metal halide perovskites are an emerging materials platform for optoelectronic, spintronic, and thermoelectric applications. The field of perovskite materials and devices has progressed rapidly over the past decade. For halide perovskite materials, a range of physical and chemical properties such as crystal structure, bandgap, charge carrier density, and stability that govern the device functionalities are critically determined by the oxidation states of the B-site metal ions. However, such an important mechanistic connection unique to halide perovskites is not well established, limiting the pace of development in this area. In this review, we identify the roles of metal oxidation states in perovskite semiconductors. The redox reactions leading to these states, and their effects on the materials properties, are clarified. Finally, we suggest routes to improving device efficiency and stability from the perspective of oxidation state control.

  • Tailoring C─N Containing Compounds into Carbon Nanomaterials with Tunable Morphologies for Electrocatalytic Applications

    • Carbon materials with unique sp2-hybridization are extensively researched for catalytic applications due to their excellent conductivity and tunable physicochemical properties. However, the development of economic approaches to tailoring carbon materials into desired morphologies remains a challenge. Herein, a convenient “bottom-up” strategy by pyrolysis of graphitic carbon nitride (g-C3N4) (or other carbon/nitrogen (C, N)-enriched compounds) together with selected metal salts and molecules is reported for the construction of different carbon-based catalysts with tunable morphologies, including carbon nano-balls, carbon nanotubes, nitrogen/sulfur (S, N) doped-carbon nanosheets, and single-atom catalysts, supported by carbon layers. The catalysts are systematically investigated through various microscopic, spectroscopic, and diffraction methods and they demonstrate promising and broad applications in electrocatalysis such as in the oxygen reduction reaction and water splitting. Mechanistic monitoring of the synthesis process through online thermogravimetric-gas chromatography-mass spectrometry measurements indicates that the release of C─N-related moieties, such as dicyan, plays a key role in the growth of carbon products. This enables to successfully predict other widely available precursor compounds beyond g-C3N4 such as caffeine, melamine, and urea. This work develops a novel and economic strategy to generate morphologically diverse carbon-based catalysts and provides new, essential insights into the growth mechanism of carbon nanomaterials syntheses.

    • Glucagon-like peptide 1 receptor agonist exenatide (exendin-4) has potential protective capabilities against diabetic kidney disease (DKD). However, the underlying mechanism has not been fully elucidated. The expression of thioredoxin-interacting protein (Txnip) is upregulated during DKD progression by histone acetylation. Sirtuin 1 (SIRT1) is a deacetylase and is decreased in DKD, which indicates that it may regulate Txnip in this disease. Here, we used whole-body heterozygous Sirt1 knockout (Sirt1+/-) and kidney-specific Sirt1 knockout (KSK) mice to investigate whether SIRT1 regulates Txnip via histone deacetylation in DKD and exenatide-alleviated DKD. Exenatide substantially improved renal pathological damage, decreased the albumin-to-creatinine ratio (ACR), upregulated SIRT1 expression, and downregulated Txnip expression in kidneys of high-fat diet-treated C57BL/6J mice. However, these effects diminished in Sirt1+/- and KSK mice under exenatide treatment. The downregulation of Txnip expression by exendin-4 in high-glucose-treated SV40 MES13 cells was hampered during Sirt1 knockdown. These results demonstrate that kidney SIRT1 is indispensable in exenatide-improved DKD and downregulation of Txnip expression. Exendin-4 mechanistically downregulated Txnip histone 3 lysine 9 acetylation (H3K9ac) in a SIRT1-dependent manner and decreased spliced X-box binding protein 1 (XBP1s) recruitment to the Txnip promoter. These findings provide epigenetic evidence elucidating the specific mechanism for exenatide-mediated DKD alleviation and highlight the importance of Txnip as a promising therapeutic target for DKD.

    • Human amniotic membranes (hAMs) are widely used as wound management biomaterials, especially as grafts for corneal reconstruction due to the structure of the extracellular matrix and excellent biological properties. However, their fragile nature and rapid degradation rate hinder widespread clinical use. In this work, we engineered a novel self-powered electronic dress (E-dress), combining the beneficial properties of an amniotic membrane and a flexible electrical electrode to enhance wound healing. The E-dress displayed a sustained discharge capacity, leading to increased epidermal growth factor (EGF) release from amniotic mesenchymal interstitial stem cells. Live/dead staining, CCK-8, and scratch-wound-closure assays were performed in vitro. Compared with amniotic membrane treatment alone, the E-dress promoted cell proliferation and migration of mouse fibroblast cells and lower cytotoxicity. In a mouse full-skin defect model, the E-dress achieved significantly accelerated wound closure. Histological analysis revealed that E-dress treatment promoted epithelialization and neovascularization in mouse skin. The E-dress exhibited a desirable flexibility that aligned with tissue organization and displayed maximum bioactivity within a short period to overcome rapid degradation, implying great potential for clinical applications.
