  • Precision electroweak measurements on the Z resonance

    • We report on the final electroweak measurements performed with data taken at the Z resonance by the experiments operating at the electron-positron colliders SLC and LEP. The data consist of 17 million Z decays accumulated by the ALEPH, DELPHI, L3 and OPAL experiments at LEP, and 600 thousand Z decays by the SLID experiment using a polarised beam at SLC. The measurements include cross-sections, forward-backward asymmetries and polarised asymmetries. The mass and width of the Z boson, m(Z) and Gamma(Z), and its couplings to fermions, for example the p parameter and the effective electroweak mixing angle for leptons, are precisely measured: m(Z) = 91.1875 +/- 0.0021 GeV, Gamma(Z) = 2.4952 +/- 0.0023 GeV, rho(l) = 1.0050 +/- 0.0010, sin(2)theta(eff)(lept) = 0.23153 +/- 0.00016. The number of light neutrino species is determined to be 2.9840 +/- 0.0082, in agreement with the three observed generations of fundamental fermions. The results are compared to the predictions of the Standard Model (SM). At the Z-pole, electroweak radiative corrections beyond the running of the QED and QCD coupling constants are observed with a significance of five standard deviations, and in agreement with the Standard Model. Of the many Z-pole measurements, the forward-backward asymmetry in b-quark production shows the largest difference with respect to its SM expectation, at the level of 2.8 standard deviations. Through radiative corrections evaluated in the framework of the Standard Model, the Z-pole data are also used to predict the mass of the top quark, m(t) = 173(+10)(+13) GeV, and the mass of the W boson, m(W) = 80.363 +/- 0.032 GeV. These indirect constraints are compared to the direct measurements, providing a stringent test of the SM. Using in addition the direct measurements of m(t) and m(W), the mass of the as yet unobserved SM Higgs boson is predicted with a relative uncertainty of about 50% and found to be less than 285 GeV at 95% confidence level.   • A direct search for stable magnetic monopoles produced in positron-proton collisions at HERA

    • A direct search has been made for magnetic monopoles produced in e(+)p collisions at a centre of mass energy of 300 GeV at HERA. The beam pipe surrounding the interaction region in 1995-1997 was investigated using a SQUID magnetometer to look for stopped magnetic monopoles. During this time an integrated luminosity of 62 pb(-1) was delivered. No magnetic monopoles were observed and charge and mass dependent upper limits on the e(+)p production cross section are set.

    • Using the deep inelastic e(+) p and e(-) p charged and neutral current scattering cross sections previously published, a combined electroweak and QCD analysis is performed to determine electroweak parameters accounting for their correlation with parton distributions. The data used have been collected by the HI experiment in 1994-2000 and correspond to an integrated luminosity of 117.2 pb(-1). A measurement is obtained of the W propagator mass in charged current ep scattering. The weak mixing angle sin(2)theta w is determined in the on-mass-shell renormalisation scheme. A first measurement at HERA is made of the light quark weak couplings to the Z(0) boson and a possible contribution of right-handed isospin components to the weak couplings is investigated. The charm and beauty fractions are determined using a method based on the impact parameter, in the transverse plane, of tracks to the primary vertex, as measured by the H1 vertex detector. The data are divided into four regions in Q(2) and Bjorken x, and values for the structure functions F-2(c (c) over bar) and F-2(b (b) over bar) are obtained. The results are found to be compatible with the predictions of perturbative quantum chromodynamics.

The fractions of events containing charm and beauty quarks are determined using a method based on the impact parameter, in the transverse plane, of tracks to the primary vertex, as measured by the H1 vertex detector. Values for the structure functions F-2(cc) and F-2(bb) are obtained. This is the first measurement of F-2(bb) in this kinematic range. The results are found to be compatible with the predictions of perturbative quantum chromodynamics and with previous measurements of F-2(cc).

  • Search for leptoquark bosons in ep collisions at HERA

    • A search for scalar and vector leptoquarks coupling to first generation fermions is performed using the e(+) p and e(-) p scattering data collected by the H1 experiment between 1994 and 2000. The data correspond to a total integrated luminosity of 117 pb(-1). No evidence for the direct or indirect production of such particles is found in data samples with a large transverse momentum final state electron or with large missing transverse momentum. Constraints on leptoquark models are established. For leptoquark couplings of electromagnetic strength, leptoquarks with masses up to 275-325 GeV are ruled out at 95% confidence level. These limits improve and supercede earlier H1 limits based on subsamples of the data used here.

Together with the corresponding cross section obtained from the previously published unpolarised data, the polarisation dependence of the charged current cross section is measured for the first time at high Q(2) and found to be in agreement with the Standard Model prediction.     • A measurement of charm and beauty photoproduction at the electron proton collider HERA is presented based on the simultaneous detection of a D*(+/-) meson and a muon. The correlation between the D* meson and the muon serves to separate the charm and beauty contributions and the analysis provides comparable sensitivity to both. The total and differential experimental cross sections are compared to LO and NLO QCD calculations. The measured charm cross section is in good agreement with QCD predictions including higher order effects while the beauty cross section is higher.

    • A measurement is presented of elastic deeply virtual Compton scattering gamma* p -> gamma p made using e(+) p collision data corresponding to a luminosity of 46.5 pb(-1), taken with the H1 detector at HERA. The cross section is measured as a function of the photon virtuality, Q(2), the invariant mass of the gamma* p system, W, and for the first time, differentially in the squared momentum transfer at the proton vertex, t, in the kinematic range 2 < Q(2) < 80 GeV2, 30 < W < 140 GeV and vertical bar t vertical bar < 1 GeV2. QCD based calculations at next-to-leading order using generalized parton distributions can describe the data, as can colour dipole model predictions.

    • Inclusive production cross sections are measured in deep inelastic scattering at HERA for meson states composed of a charm quark and a light antiquark or the charge conjugate. The measurements cover the kinematic region of photon virtuality 2 < Q(2) < 100 GeV2, inelasticity 0.05 < y < 0.7, D meson transverse momenta p(t)( D) greater than or equal to 2.5 GeV and pseudorapidity |eta( D)| less than or equal to 1.5. The identification of the D-meson decays and the reduction of the combinatorial background profit from the reconstruction of displaced secondary vertices by means of the H1 silicon vertex detector. The production of charmed mesons containing the light quarks u, d and s is found to be compatible with a description in which the hard scattering is followed by a factorisable and universal hadronisation process.

  • Measurement of beauty production at HERA using events with muons and jets

    • A measurement of the beauty production cross section in ep collisions at a centre-of-mass energy of 319 GeV is presented. The data were collected with the H1 detector at the HERA collider in the years 1999-2000. Events are selected by requiring the presence of jets and muons in the final state. Both the long lifetime and the large mass of b-flavoured hadrons are exploited to identify events containing beauty quarks. Differential cross sections are measured in photoproduction, with photon virtualities Q(2) < 1 GeV2, and in deep inelastic scattering, where 2 < Q(2) < 100 GeV2. The results are compared with perturbative QCD calculations to leading and next-to-leading order. The predictions are found to be somewhat lower than the data.

  • Measurement of prompt photon cross sections in photoproduction at HERA

    • Results are presented on the photoproduction of isolated prompt photons, inclusively and associated with jets, in the gammap center of mass energy range 142 < W < 266 GeV. The cross sections are measured for the transverse momentum range of the photons 5 < E. T < 10 GeV and for associated jets with E-T(jet) > 4.5 GeV. They are measured differentially as a function of E-T(gamma), E-T(jet), the pseudorapidities eta(gamma) and eta(jet) and estimators of the momentum fractions x(gamma) and x(p) of the incident photon and proton carried by the constituents participating in the hard process. In order to further investigate the underlying dynamics, the angular correlation between the prompt photon and the jet in the transverse plane is studied. Predictions by perturbative QCD calculations in next to leading order are about 30% below the inclusive prompt photon data after corrections for hadronisation and multiple interactions, but are in reasonable agreement with the results for prompt photons associated with jets. Comparisons with the predictions of the event generators PYTHIA and HERWIG are also presented.

    • A search for gravitinos produced in e(+/-)p collisions is performed using the H1 detector at HERA. The data were taken at a centre-of-mass energy of 319 GeV and correspond to an integrated luminosity of 64.3 pb(-1) for e(+)p collisions and 13.5 pb(-1) for e(-)p collisions. If R-parity is not conserved, the t-channel exchange of a selectron can produce a neutralino, which, in models where the gravitino is the lightest supersymmetric particle, subsequently decays into a photon and a light gravitino. The resulting event signature, which involves an isolated photon, a jet and missing transverse energy, is analysed for the first time at HERA. No deviation from the Standard Model is found. Exclusion limits on the cross section and on R-parity-violating Yukawa couplings are derived in a Gauge Mediated Supersymmetry Breaking scenario. The results are independent of the squark sector. Neutralinos and supersymmetric partners of the left-handed electron with masses up to 112 GeV and 164 GeV, respectively, can be ruled out at the 95% confidence level for R-parity-violating couplings lambda' equal to 1, in some parts of the parameter space of the considered model.

    • Measurements are reported of the production of dijet events with a leading neutron in ep interactions at HERA. Differential cross sections for photoproduction and deep inelastic scattering are presented as a function of several kinematic variables. Leading order QCD simulation programs are compared with the measurements. Models in which the real or virtual photon interacts with a parton of an exchanged pion are able to describe the data. Next-to-leading order perturbative QCD calculations based on pion exchange are found to be in good agreement with the measured cross sections. The fraction of leading neutron dijet events with respect to all dijet events is also determined. The dijet events with a leading neutron have a lower fraction of resolved photon processes than do the inclusive dijet data.

    • Background: In this study we aimed to evaluate the role of a SNP in intron 1 of the ERCC4 gene (rs744154), previously reported to be associated with a reduced risk of breast cancer in the general population, as a breast cancer risk modifier in BRCA1 and BRCA2 mutation carriers. Methods: We have genotyped rs744154 in 9408 BRCA1 and 5632 BRCA2 mutation carriers from the Consortium of Investigators of Modifiers of BRCA1/2 (CIMBA) and assessed its association with breast cancer risk using a retrospective weighted cohort approach. Results: We found no evidence of association with breast cancer risk for BRCA1 (per-allele HR: 0.98, 95% CI: 0.93-1.04, P0.5) or BRCA2 (per-allele HR: 0.97, 95% CI: 0.89-1.06, P0.5) mutation carriers. Conclusion: This SNP is not a significant modifier of breast cancer risk for mutation carriers, though weak associations cannot be ruled out.

    • Scientific findings from the last decades have clearly highlighted the need for a more comprehensive approach to atmospheric change processes. In fact, observation of atmospheric composition variables has been an important activity of atmospheric research that has developed instrumental tools (advanced analytical techniques) and platforms (instrumented passenger aircrafts, ground-based in-situ and remote sensing stations, earth observation satellite instruments) providing essential information on the composition of the atmosphere. The variability of the atmospheric system and the extreme complexity of the atmospheric cycles for short-lived gaseous and aerosol species have led to the development of complex models to interpret observations, test our theoretical understanding of atmospheric chemistry and predict future atmospheric composition. The validation of numerical models requires accurate information concerning the variability of atmospheric composition for targeted species via comparison with observations and measurements. In this paper, we provide an overview of recent advances in instrumentation and methodologies for measuring atmospheric composition changes from space, aircraft and the surface as well as recent improvements in laboratory techniques that permitted scientific advance in the field of atmospheric chemistry. Emphasis is given to the most promising and innovative technologies that will become operational in the near future to improve knowledge of atmospheric composition. Our current observation capacity, however, is not satisfactory to understand and predict future atmospheric composition changes, in relation to predicted climate warming. Based on the limitation of the current European observing system, we address the major gaps in a second part of the paper to explain why further developments in current observation strategies are still needed to strengthen and optimise an observing system not only capable of responding to the requirements of atmospheric services but also to newly open scientific questions.

    • The discriminant-analysis method has been applied to optimize the exotic-beam charge recognition in a projectile fragmentation experiment. The experiment was carried out at the GSI using the fragment separator (FRS) to produce and select the relativistic secondary beams, and the ALADIN setup to measure their fragmentation products following collisions with Sn target nuclei. The beams of neutron poor isotopes around La-124 and Sn-107 were selected to study the isospin dependence of the limiting temperature of heavy nuclei by comparing with results for stable Sn-124 projectiles. A dedicated detector to measure the projectile charge upstream of the reaction target was not used, and alternative methods had to be developed. The presented method, based on the multivariate discriminant analysis, allowed to increase the efficacy of charge recognition up to about 90%, which was about 20% more than achieved with the simple scalar methods.

    • Vertical profiles of stratospheric water vapour measured by the Michelson Interferometer for Passive Atmospheric Sounding (MIPAS) with the full resolution mode between September 2002 and March 2004 and retrieved with the IMK/IAA scientific retrieval processor were compared to a number of independent measurements in order to estimate the bias and to validate the existing precision estimates of the MIPAS data. The estimated precision for MIPAS is 5 to 10% in the stratosphere, depending on altitude, latitude, and season. The independent instruments were: the Halogen Occultation Experiment (HALOE), the Atmospheric Chemistry Experiment Fourier Transform Spectrometer (ACE-FTS), the Improved Limb Atmospheric Spectrometer-II (ILAS-II), the Polar Ozone and Aerosol Measurement (POAM III) instrument, the Middle Atmospheric Water Vapour Radiometer (MIAWARA), the Michelson Interferometer for Passive Atmospheric Sounding, balloon-borne version (MIPAS-B), the Airborne Microwave Stratospheric Observing System(AMSOS), the Fluorescent Stratospheric Hygrometer for Balloon (FLASH-B), the NOAA frostpoint hygrometer, and the Fast In Situ Hygrometer (FISH). For the in-situ measurements and the ground based, air- and balloon borne remote sensing instruments, the measurements are restricted to central and northern Europe. The comparisons to satellite-borne instruments are predominantly at mid- to high latitudes on both hemispheres. In the stratosphere there is no clear indicationof a bias in MIPAS data, because the independent measurements in some cases are drier and in some cases are moister than the MIPAS measurements. Compared to the infrared measurements of MIPAS, measurements in the ultraviolet and visible have a tendency to be high, whereas microwave measurements have a tendency to be low. Theresults of chi2-based precision validation are somewhat controversial among the comparison estimates. However, for comparison instruments whose error budget also includes errors due to uncertainties in spectrally interfering species and where good coincidences were found, the chi2 values found are in the expected range or even below. This suggests that there is no evidence of systematically underestimated MIPAS random errors.

    • Isotopic effects in projectile fragmentation at relativistic energies have been studied with the ALADIN forward spectrometer at SIS. Stable and radioactive Sn and La beams with an incident energy of 600 MeV per nucleon have been used in order to explore a wide range of isotopic compositions. Chemical freeze-out temperatures are found to be nearly invariant with respect to the A/Z ratio of the produced spectator sources, consistent with predictions for expanded systems. Consequences for the proposed interpretation of chemical breakup temperatures as representing the limiting temperatures predicted by microscopic models are discussed. (C) 2009 Elsevier B.V.     • Nutrigenomics is the study of how constituents of the diet interact with genes, and their products, to alter phenotype and, conversely, how genes and their products metabolise these constituents into nutrients, antinutrients, and bioactive compounds. Results from molecular and genetic epidemiological studies indicate that dietary unbalance can alter gene–nutrient interactions in ways that increase the risk of developing chronic disease. The interplay of human genetic variation and environmental factors will make identifying causative genes and nutrients a formidable, but not intractable, challenge. We provide specific recommendations for how to best meet this challenge and discuss the need for new methodologies and the use of comprehensive analyses of nutrient–genotype interactions involving large and diverse populations. The objective of the present paper is to stimulate discourse and collaboration among nutrigenomic researchers and stakeholders, a process that will lead to an increase in global health and wellness by reducing health disparities in developed and developing countries.

    • An experiment was performed using the FRS-LAND setup at GSI to study the dipole strength distributions above neutron separation threshold for neutron-rich Ni isotopes. Measurements, using the same experimental setup, were also carried out to extract single particle occupancies via knockout reactions to investigate the structure and magicity of the neutron-rich Ni isotopes.     • Dust production and accumulation present potential safety and operational issues for the ITER. Dust diagnostics can be divided into two groups: diagnostics of dust on surfaces and diagnostics of dust in plasma. Diagnostics from both groups are employed in contemporary tokamaks; new diagnostics suitable for ITER are also being developed and tested. Dust accumulation in ITER is likely to occur in hidden areas, e.g., between tiles and under divertor baffles. A novel electrostatic dust detector for monitoring dust in these regions has been developed and tested at PPPL. In the DIII-D tokamak dust diagnostics include Mie scattering from Nd:YAG lasers, visible imaging, and spectroscopy. Laser scattering is able to resolve particles between 0.16 and 1.6 mu m in diameter; using these data the total dust content in the edge plasmas and trends in the dust production rates within this size range have been established. Individual dust particles are observed by visible imaging using fast framing cameras, detecting dust particles of a few microns in diameter and larger. Dust velocities and trajectories can be determined in two-dimension with a single camera or three-dimension using multiple cameras, but determination of particle size is challenging. In order to calibrate diagnostics and benchmark dust dynamics modeling, precharacterized carbon dust has been injected into the lower divertor of DIII-D. Injected dust is seen by cameras, and spectroscopic diagnostics observe an increase in carbon line (CI, CII, C(2) dimer) and thermal continuum emissions from the injected dust. The latter observation can be used in the design of novel dust survey diagnostics.

    • We provide new evidence for the development of a stable estuarinecirculation characterized by stagnating water bodies, nutrientrecycling and increased primary productivity in the South ChinaSea (SCS) during glacial intervals. This circulation was caused bythe closure of the shallow and narrow straits connecting the SCSin the south and west. Our main evidence is derived from newlymeasured Mn concentrations and Mn/Al ratios in two sedimentcores from the northern and southeastern SCS covering the last500 ky. Concentrations and Mn/Al ratios of the redox sensitive elementMnshowclearglacial–interglacial cycles with maxima duringinterglacial periods and minima during glacial periods. These cyclesindicate ventilation cycles of the bottom water, and are connectedto the glacial–interglacial changes in sea level. In contrast, totalorganic carbon (TOC) concentrations display an opposite patternwith pronounced maxima during glacial times, especially in thesouthern part of the basin. The variations in TOC can be ascribed totwo factors. Firstly, variations in primary productivity are controlledby variations in the intensity of the winter monsoon. Secondlyto the degree of preservation of TOC controlled by variations inventilation, which in turn is ultimately controlled by sea level. Consequently, variations in TOC represent a superimposition ofprimarily sea level influenced preservation control and wintermonsoon driven variations in primary productivity intensity. Thedecrease in Mn correspond to times when sea level dropped40–60m below the present level. The larger amplitude of the variationsin TOC and Mn in the southern part of the basin compared tothe northern site suggest that oxygen depletion and nutrient recyclingwas stronger in the parts of the basin situated the furthest away from the only remaining opening to the open Pacific, the LuzonStrait.

    • The spallation of Fe-56 in collisions with hydrogen at 1A GeV has been studied in inverse kinematics with the large-aperture setup SPALADIN at GSI. Coincidences of residues with low-center-of-mass kinetic energy light particles and fragments have been measured allowing the decomposition of the total reaction cross section into the different possible deexcitation channels. Detailed information on the evolution of these deexcitation channels with excitation energy has also been obtained. The comparison of the data with predictions of several deexcitation models coupled to the INCL4 intranuclear cascade model shows that only GEMINI can reasonably account for the bulk of collected results, indicating that in a light system with no compression and little angular momentum, multifragmentation might not be necessary to explain the data.

    • The dielectronic recombination rate coefficients of Nv and Ne viii ions have been measured at a heavy-ion storage ring. The investigated energy ranges covered all dielectronic recombination resonances attached to 2s -> 2p (delta_n = 0) core excitations. The rate coefficients in a plasma are derived and parameterized by using a convenient fit formula. The experimentally derived rate coefficients are compared with theoretical data by Colgan et al. (2004, A&A, 417, 1183) and Nahar & Pradhan (1997, ApJ, 111, 339) as well as with the recommended rate coefficients by Mazzotta et al. (1998, A&A, 133, 403). The data of Colgan et al. and Nahar & Pradhan reproduce the experiment very well over the temperature ranges where Nv and Ne viii are expected to exist in photoionized as well as in collisionally ionized plasmas. In contrast the recommendation of Mazzotta et al. agrees with the experimental rate coefficient only in the collisionally ionized temperature range. At lower temperatures it deviates from the measured rate coefficient by orders of magnitude. In addition the influence of external electric fields with field strengths up to 1300 V/cm on the dielectronic recombination rate coefficient has been investigated.

    • OBJECTIVE: To explore the involvement of M-type potassium channels KCNQ2, Q3, and Q5 in the pathogenesis of common idiopathic epilepsies. METHODS: Sequence analysis of the KCNQ2, Q3, and Q5 coding regions was performed in a screening sample consisting of 58 nuclear families with rolandic epilepsy. Subsequently, an association study was conducted for all discovered variants in a case-control sample comprising 459 German patients with idiopathic generalized epilepsy (IGE) and 462 population controls. RESULTS: An in-frame deletion of codon 116 in KCNQ2 (p.Lys116del) and a missense mutation in KCNQ3 (p.Glu299Lys) were detected in two index cases exhibiting rolandic epilepsy and benign neonatal convulsions. Both mutations resulted in reduced potassium current amplitude in Xenopus oocytes. Mutation analysis of families with rolandic epilepsy without neonatal seizures discovered three novel missense variations (KCNQ2 p.Ile592Met, KCNQ3 p.Ala381Val, KCNQ3 p.Pro574Ser). The KCNQ2 p.Ile592Met variant displayed a significant reduction of potassium current amplitude in Xenopus oocytes and was present only once in 552 controls. Both missense variants identified in KCNQ3 (p.Ala381Val and p.Pro574Ser) were present in all affected family members and did not occur in controls, but did not show obvious functional abnormalities. The KCNQ3 missense variant p.Pro574Ser was also detected in 8 of 455 IGE patients but not in 454 controls (p = 0.008). In KCNQ2, a silent single nucleotide polymorphism (rs1801545) was found overrepresented in both epilepsy samples (IGE, p = 0.004). CONCLUSION: Sequence variations of the KCNQ2 and KCNQ3 genes may contribute to the etiology of common idiopathic epilepsy syndromes.

    • Thioredoxin reductase (TrxR), a NADPH-dependent disulfide oxidoreductase, is vital in numerous cellular processes including defence against reactive oxygen species, cell proliferation and signal transduction. TrxRs occur in 2 forms, a high M-r enzyme characterized by those of mammals, the malaria parasite Plasmodium falciparum and some worms, and a low M-r form is present in bacteria, fungi, plants and some protozoan parasites. Our hypothesis is that the differences between the forms can be exploited in the development of selective inhibitors. In this study, cyclodextrin- and sulfonic acid-derived organotelluriums known to inhibit mammalian TrxR were investigated for their relative efficacy against P. falciparum TrxR (PfTrxR), a high M-r enzyme, and Trichomonas vaginalis TrxR (TvTrxR), a low M-r form of TrxR. The results suggest that selective inhibition of low M-r TrxRs is a feasible goal.

    • In congenital myasthenic syndrome with DOK7 mutations ephedrine was reported to be beneficial in single patients. We carried out a small, open and prospective cohort study in eight European patients manifesting from birth to 12 years. Five patients showed limb-girdle and facial weakness, three a floppy infant syndrome with bulbar symptoms and/or respiratory distress. Ephedrine was started with 25 mg/day and slowly increased to 75-100 mg/day. Within weeks after starting therapy an improvement was observed in all patients and clinical follow-up disclosed positive effects more pronounced on proximal muscle weakness and strength using MRC scale. Effects on facial weakness were less pronounced. Vital capacity measurements and repetitive stimulation tests did not improve in the same way as clinical symptoms did. These investigations are appropriate to confirm the diagnosis in case of pathological results, but they might not be appropriate means to monitor patients under ephedrine therapy.

    • Effector mechanisms in anaphylaxis were reviewed. Current approaches to confirmation of the clinical diagnosis were discussed. Improved methods for distinguishing between allergen sensitization (which is common in the general population) and clinical risk of anaphylaxis (which is uncommon) were deliberated. Innovative techniques that will improve risk assessment in anaphylaxis in the future were described.

    • Risk assessment of individuals with anaphylaxis is currently hampered by lack of (1) an optimal and readily available laboratory test to confirm the clinical diagnosis of an anaphylaxis episode and (2) an optimal method of distinguishing allergen-sensitized individuals who are clinically tolerant from those at risk for anaphylaxis episodes after exposure to the relevant allergen. Our objectives were to review the effector mechanisms involved in the pathophysiology of anaphylaxis; to explore the possibility of developing an optimal laboratory test to confirm the diagnosis of an anaphylaxis episode, and the possibility of improving methods to distinguish allergen sensitization from clinical reactivity; and to develop a research agenda for risk assessment in anaphylaxis. Researchers from the American Academy of Allergy, Asthma & Immunology and the European Academy of Allergology and Clinical Immunology held a PRACTALL (Practical Allergy) meeting to discuss these objectives. New approaches being investigated to support the clinical diagnosis of anaphylaxis include serial measurements of total tryptase in serum during an anaphylaxis episode, and measurement of baseline total tryptase levels after the episode. Greater availability of the test for mature beta-tryptase, a more specific mast cell activation marker for anaphylaxis than total tryptase, is needed. Measurement of chymase, mast cell carboxypeptidase A3, platelet-activating factor, and other mast cell products may prove to be useful. Consideration should be given to measuring a panel of mediators from mast cells and basophils. New approaches being investigated to help distinguish sensitized individuals at minimum or no risk from those at increased risk of developing anaphylaxis include measurement of the ratio of allergen-specific IgE to total IgE, determination of IgE directed at specific allergenic epitopes, measurement of basophil activation markers by using flow cytometry, and assessment of allergen-specific cytokine responses. Algorithms have been developed for risk assessment of individuals with anaphylaxis, along with a research agenda for studies that could lead to an improved ability to confirm the clinical diagnosis of anaphylaxis and to identify allergen-sensitized individuals who are at increased risk of anaphylaxis.
