  • The Genome Sequence of Taurine Cattle : A Window to Ruminant Biology and Evolution

    • To understand the biology and evolution of ruminants, the cattle genome was sequenced to about sevenfold coverage. The cattle genome contains a minimum of 22,000 genes, with a core set of 14,345 orthologs shared among seven mammalian species of which 1217 are absent or undetected in noneutherian (marsupial or monotreme) genomes. Cattle-specific evolutionary breakpoint regions in chromosomes have a higher density of segmental duplications, enrichment of repetitive elements, and species-specific variations in genes associated with lactation and immune responsiveness. Genes involved in metabolism are generally highly conserved, although five metabolic genes are deleted or extensively diverged from their human orthologs. The cattle genome sequence thus provides a resource for understanding mammalian evolution and accelerating livestock genetic improvement for milk and meat production.

  • Heterogeneity of breast cancer associations with five susceptibility loci by clinical and pathological characteristics

    • A three-stage genome-wide association study recently identified single nucleotide polymorphisms (SNPs) in five loci (fibroblast growth receptor 2 (FGFR2), trinucleotide repeat containing 9 (TNRC9), mitogen-activated protein kinase 3 K1 (MAP3K1), 8q24, and lymphocyte-specific protein 1 (LSP1)) associated with breast cancer risk. We investigated whether the associations between these SNPs and breast cancer risk varied by clinically important tumor characteristics in up to 23,039 invasive breast cancer cases and 26,273 controls from 20 studies. We also evaluated their influence on overall survival in 13,527 cases from 13 studies. All participants were of European or Asian origin. rs2981582 in FGFR2 was more strongly related to ER-positive (per-allele OR (95%CI) = 1.31 (1.27-1.36)) than ER-negative (1.08 (1.03-1.14)) disease (P for heterogeneity = 10(-13)). This SNP was also more strongly related to PR-positive, low grade and node positive tumors (P = 10(-5), 10(-8), 0.013, respectively). The association for rs13281615 in 8q24 was stronger for ER-positive, PR-positive, and low grade tumors (P = 0.001, 0.011 and 10(-4), respectively). The differences in the associations between SNPs in FGFR2 and 8q24 and risk by ER and grade remained significant after permutation adjustment for multiple comparisons and after adjustment for other tumor characteristics. Three SNPs (rs2981582, rs3803662, and rs889312) showed weak but significant associations with ER-negative disease, the strongest association being for rs3803662 in TNRC9 (1.14 (1.09-1.21)). rs13281615 in 8q24 was associated with an improvement in survival after diagnosis (per-allele HR = 0.90 (0.83-0.97). The association was attenuated and non-significant after adjusting for known prognostic factors. Our findings show that common genetic variants influence the pathological subtype of breast cancer and provide further support for the hypothesis that ER-positive and ER-negative disease are biologically distinct. Understanding the etiologic heterogeneity of breast cancer may ultimately result in improvements in prevention, early detection, and treatment.

  • A common coding variant in CASP8 is associated with breast cancer risk

    • The Breast Cancer Association Consortium (BCAC) has been established to conduct combined case-control analyses with augmented statistical power to try to confirm putative genetic associations with breast cancer. We genotyped nine SNPs for which there was some prior evidence of an association with breast cancer: CASP8 D302H (rs1045485), IGFBP3 -202 C --> A (rs2854744), SOD2 V16A (rs1799725), TGFB1 L10P (rs1982073), ATM S49C (rs1800054), ADH1B 3' UTR A --> G (rs1042026), CDKN1A S31R (rs1801270), ICAM5 V301I (rs1056538) and NUMA1 A794G (rs3750913). We included data from 9-15 studies, comprising 11,391-18,290 cases and 14,753-22,670 controls. We found evidence of an association with breast cancer for CASP8 D302H (with odds ratios (OR) of 0.89 (95% confidence interval (c.i.): 0.85-0.94) and 0.74 (95% c.i.: 0.62-0.87) for heterozygotes and rare homozygotes, respectively, compared with common homozygotes; P(trend) = 1.1 x 10(-7)) and weaker evidence for TGFB1 L10P (OR = 1.07 (95% c.i.: 1.02-1.13) and 1.16 (95% c.i.: 1.08-1.25), respectively; P(trend) = 2.8 x 10(-5)). These results demonstrate that common breast cancer susceptibility alleles with small effects on risk can be identified, given sufficiently powerful studies.

  • Short-range order and Fe clustering in Mg1-xFexO under high pressure

    • By combining high-pressure and high-temperature Moumlssbauer spectroscopic studies of (Mg,Fe)O with results of ab initio simulations, several important properties of this material were established. Under high pressure (Mg,Fe)O shows changes in the short-range order with the tendency to form iron clusters. These changes were found to be irreversible, implying sluggish kinetics of these processes at ambient conditions. The pressure-induced spin crossover is interpreted here as a gradual noncooperative transition. The onset and width of spin crossover depends, therefore, not only on pressure, temperature, and composition, but also on short-range order in the FeO-MgO solid solution.

    • Keratinocytes and fibroblasts have been grown onto a series of triblock copolymers based on 1,5-dioxepan-2-one (DXO) and L-lactide (LLA). The molar ratio of DXO and LLA were varied in the copolymers. This resulted in different degrees of hydrophilicity, which in turn influenced the cell growth. On these surfaces, the morphological appearance of the cells with their cell movements and growth were investigated by means of scanning electron microscopy, time-lapse videomicroscopy and immunohistochemistry. All results clearly showed that the keratinocytes and fibroblasts adhered best to the most hydrophilic copolymers. A majority of the keratinocytes seeded on the most hydrophilic copolymer also presented a polarized morphology indicating a migration tendency. The cell growth onto these materials are interesting since a possible application for these unique materials is as polymeric membranes for guided cutaneous and/or periodontal tissue generation.

    • The purpose of the European Master’s Programme in Sport and Exercise Psychology (EMPSEP) is to pool expertise of 12 European universities within one Master’s programme (see http://www.fepsac.com/). The 60 ECTS European programme provides students with advanced knowledge and skills. The EMPSEP comprises a joint intensive course, a study module similar in all the participating universities, lectures and seminars, a Master’s thesis, and a mobility period of 4-5 months at an EMPSEP host university. Ten years after the graduation of the first students the EMPSEP consortium conducted an online survey. Seventy of the invited 174 former students participated in the study (mean age 31.5 years, SD= 4.7). The aim of the survey was to discover the participants’ employment status and how their participation in the master’s program was related to this. Results indicate that 86% of the participants have started working since they completed their degree. Forty percent of those participants who have started working in their first job have managed to receive a permanent position, 27% a fixed term or temporary job, 25% a part time job, 6% are self employed and 1 person (2%) was employed by subsidies in his/ her first job after graduation. On a scale from extremely dissatisfied (1) to extremely satisfied (6), the participants rated their satisfaction with the program in relation to their career as 4.72 (SD=1.13) on average. Sixty nine of the participants felt that they had benefited from the international network provided by the students and teachers within the programme, and 94% would recommend the European Master’s program to other students in their field.

  • Monosomy 7 and deletion 7q in children and adolescents with acute myeloid leukemia : an international retrospective study.

    • Monosomy 7 (-7) and deletion 7q \del(7q)] are rare in childhood acute myeloid leukemia (AML). We retrospectively collected data on 258 children with AML or refractory anemia with excess blasts in transformation (RAEB-T) and -7 or del(7q) with or without other cytogenetic aberrations \+/- other]. Karyotypes included -7 (n = 90), -7 other (n = 82), del(7q) (n = 21), and del(7q) other (n = 65). Complete remission (CR) was achieved in fewer patients with -7 +/- other compared with del(7q) +/- other (61% versus 89%, P < .001). Overall, the 5-year survival rate was 39% (SE, 3%). Survival was superior in del(7q) +/- other compared with -7 +/- other (51% versus 30%, P < .01). Cytogenetic aberrations considered favorable in AML \t(8;21)(q22;q22), inv(16)(p13q22), t(15;17)(q22;q21), t(9;11)(p22;q23)] (n = 24) were strongly associated with del(7q) and a higher 5-year survival rate compared with del(7q) without favorable cytogenetics (75% versus 46%, P = .03). Patients with -7 and inv(3),-5/del(5q), or + 21 had a 5-year survival rate of 5%. Stem cell transplantation analyzed as a time-dependent variable had no impact on overall survival. However, patients not achieving CR had a 31% survival rate after stem cell transplantation. Childhood AML with chromosome 7 aberrations represents a heterogeneous group of disorders with additional cytogenetic aberrations having a major prognostic impact which should be reflected in future risk-group stratification.

    • Dorsoventral axis formation in the legs of the fly Drosophila melanogaster requires the T-box genes optomotor-blind (omb) and H15. Evolutionary conservation of the patterning functions of these genes is unclear, because data on H15 expression in the spider Cupiennius salei did not support a general role of H15 in ventral fate specification. However, H15 has a paralogous gene, midline (mid) in Drosophila and H15 duplicates are also present in Cupiennius and the millipede Glomeris marginata. H15 therefore seems to have been subject to gene duplication opening the possibility that the previous account on Cupiennius has overlooked one or several paralogs. We have studied omb- and H15-related genes in two additional spider species, Tegenaria atrica and Achearanea tepidariorum and show that in both species one of the H15 genes belongs to a third group of spider H15 genes that has an expression pattern very similar to the H15 pattern in Drosophila. The expression patterns of all omb-related genes are also very similar to the omb expression pattern in Drosophila. These data suggest that the dorsoventral patterning functions of omb and H15 are conserved in the arthropods and that the previous conclusions were based on an incomplete data set in Cupiennius. Our results emphasize the importance of a broad taxon sampling in comparative studies.

    • Mates and females are in conflict over parental care, as it would be favourable for one parent to shift labour to the other. Yolk hormones may offer a mechanism through which female birds could influence offspring traits in ways that increase the relative investment by the male. We studied the role of yolk androgens in mediating sexual conflict over parental care in the collared flycatcher (Ficedula albicollis). In a cross-fostering experiment, the male's proportion of total feeding visits increased with increasing androgen levels in the foster eggs. This could suggest that sexual conflict over parental care may be influenced by the female's differential allocation of yolk androgens or a maternal effect associated with yolk androgens. However, when we experimentally elevated yolk androgen levels, male feeding rates did not differ between control and androgen-manipulated nests. This suggests that other egg components correlated with yolk androgen levels, rather than yolk androgen levels per se, may influence male parental effort. In conclusion, yolk androgens per se do not appear to mediate sexual conflict over parental investment in the collared flycatcher. (C) 2009 Elsevier Inc. All rights reserved.
