  • Identification of novel candidate protein biomarkers for the post-polio syndrome — Implications for diagnosis, neurodegeneration and neuroinflammation

    • Survivors of poliomyelitis often develop increased or new symptoms decades after the acute infection, a condition known as post-polio syndrome (PPS). The condition affects 20-60% of previous polio patients, making it one of the most common causes of neurological deficits worldwide. The underlying pathogenesis is not fully understood and accurate diagnosis is not feasible. Herein we investigated whether it was possible to identify proteomic profile aberrations in the cerebrospinal fluid (CSF) of PPS patients. CSF from 15 patients with well-defined PPS were analyzed for protein expression profiles. The results were compared to data obtained from nine healthy controls and 34 patients with other non-inflammatory diseases which served as negative controls. In addition, 17 samples from persons with secondary progressive multiple sclerosis (SPMS) were added as relevant age-matched references for the PPS samples. The CSF of persons with PPS displayed a disease-specific and highly predictive (p=0.0017) differential expression of five distinct proteins: gelsolin, hemopexin, peptidylglycine alpha-amidating monooxygenase, glutathione synthetase and kallikrein 6, respectively, in comparison with the control groups. An independent ELISA confirmed the increase of kallikrein 6. We suggest that these five proteins should be further evaluated as candidate biomarkers for the diagnosis and development of new therapies for PPS patients.

  • Position Sensitive Detectors : Device Technology and Applications in Spectroscopy

    • This thesis deals with the development, processing and characterization of position sensitive detectors and, in addition, to the development of compact and cost effective spectrometers. Position sensitive detectors are used to measure, with great accuracy and speed, the position of a light spot incident on the surface. Their main use is for triangulation, displacement and vibration measurements. A type of position sensitive detector based on the MOS principle and using optically transparent indium tin oxide as a gate contact has been developed. This type of detector utilizes the MOS principle where an induced channel forms beneath the gate oxide in the surface of the Silicon substrate. One and two dimensional detectors have both been fabricated and characterized. The first measurements showed that the linearity did not fulfil expectations and it was suspected that stress induced by the gate contact could be the reason for the seemingly high nonlinearity. Further investigations into both the p-n junction and the MOS type position sensitive detectors lead to the conclusion that the indium tin oxide gate is responsible for inducing a substantial stress in the surface of the detector, thus giving rise to increased position nonlinearity. The heat treatment step which was conducted was determined to be critical as either a too short or too long heat treatment resulted in stress in the gate and channel leading to position nonlinearity. If a correctly timed heat treatment is performed then the detector’s linearity is in parity with the best commercial position sensitive detectors. In addition, the development of very small, compact and cost effective spectrometers has been performed with the aim of constructing devices for use in the process industry. The development of a wedge shaped array of Fabry-Perot interferometers that can be mounted directly on top of a detector makes it possible to construct a very compact spectrometer using the minimum amount of optics. This wedge interferometer has been evaluated by means of array pixel detectors and position sensitive detectors for both the infrared and the visible wavelength ranges. When used with a position sensitive detector it is necessary to use a slit to record the intensity of the interferogram for many points over the detector, equivalent to pixels on an array detector. Usually the use of moving parts in a spectrometer will impose the use of high precision scanning mechanisms and calibration. By using a position sensitive detector for the interferogram readout both the position and the intensity are known for every measurement point and thus the demands placed on the scanning system are minimized.

    • Type 1 diabetes in children is characterised by autoimmune destruction of pancreatic beta cells and the presence of certain risk genotypes. In adults the same situation is often referred to as latent autoimmune diabetes in adults (LADA). We tested whether genetic markers associated with type 1 or type 2 diabetes could help to discriminate between autoimmune and non-autoimmune diabetes in young (15-34 years) and middle-aged (40-59 years) diabetic patients. In 1,642 young and 1,619 middle-aged patients we determined: (1) HLA-DQB1 genotypes; (2) PTPN22 and INS variable-number tandem repeat (VNTR) polymorphisms; (3) two single nucleotide polymorphisms (rs7903146 and rs10885406) in the TCF7L2 gene; (4) glutamic acid decarboxylase (GAD) and IA-2-protein tyrosine phosphatase-like protein (IA-2) antibodies; and (5) fasting plasma C-peptide. Frequency of risk genotypes HLA-DQB1 (60% vs 25%, p =9.4x10(-34); 45% vs 18%, p= 1.4x10(-16)), PTPN22 CT/TT (34% vs 26%, p=0.0023; 31% vs 23%, p=0.034), INS VNTR class I/I (69% vs 53%, p=1.3x10(-8); 69% vs 51%, p=8.5x10(-5)) and INS VNTR class IIIA/IIIA (75% vs 63%, p=4.3x10(-6); 73% vs 60%, p=0.008) was increased in young and middle-aged GAD antibodies (GADA)-positive compared with GADA-negative patients. The type 2 diabetes-associated genotypes of TCF7L2 CT/TT of rs7903146 were significantly more common in young GADA-negative than in GADA-positive patients (53% vs 43%; p=0.0004). No such difference was seen in middle-aged patients, in whom the frequency of the CT/TT genotypes of TCF7L2 was similarly increased in GADA-negative and GADA-positive groups (55% vs 56%). Common variants in the TCF7L2 gene help to differentiate young but not middle-aged GADA-positive and GADA-negative diabetic patients, suggesting that young GADA-negative patients have type 2 diabetes and that middle-aged GADA-positive patients are different from their young GADA-positive counterparts and share genetic features with type 2 diabetes.

  • Subendocardial versus transmural ischaemia in myocardial perfusion SPECT - a Monte Carlo study

    • Myocardial perfusion imaging with single-photon emission computed tomography (SPECT) is useful for the evaluation of patients with known or suspected coronary artery disease. Parameters of interest are the reduction in the blood perfusion (severity) and the lesion volume (extent). The aim of this study was to evaluate these parameters, as calculated by automatic quantification software, for different cases of subendocardial and transmural myocardial lesions. Methods A computer phantom was used to simulate 32 male patients with different defect locations and activity uptakes, which were based on clinical patient studies. The Monte Carlo program SIMIND was used to simulate realistic SPECT projections which were reconstructed to give short-axis images, analysed by the AutoQUANT program using the same procedure as for a real patient. Results The results showed a disparity between the quantification of transmural and subendocardial lesions with the same lesion activity uptake reduction and this could be confirmed by visual interpretation. Neither the parameters given by the quantification program nor visual interpretation could distinguish between the transmural lesions and the subendocardial lesions with activity uptake reduction twice as high as in the corresponding transmural lesions. Conclusion Transmural lesions and the corresponding subendocardial lesions with the same activity uptake reduction could be separated by the quantification software for SPECT imaging and visual analysis. The subendocardial lesions with activity uptake reduction twice as high as in the corresponding transmural lesions could not be differentiated neither by the quantification software nor by visual interpretation. Thus these lesions will get the same scoring when analysed by the AutoQUANT program.

    • Young vertebrates have limited capacity to synthesize antibodies and are dependent on the protection of maternally transmitted antibodies for humoral disease resistance early in life. However, mothers may enhance fitness by priming their offspring's immune systems to elevate disease resistance. Transgenerational induced defences have been documented in plants and invertebrates, but maternal priming of offspring immunity in vertebrates has been essentially neglected. To test the ability of mothers to stimulate the immune systems of offspring, we manipulated maternal and offspring antigen exposure in a wild population of birds, pied flycatchers (Ficedula hypoleuca). We show that immunization of the mother before egg laying apparently stimulates a transgenerational defence against pathogens by elevating endogenous offspring antibody production. If the disease environments encountered by mothers and offspring are similar, this transgenerational immune priming may allow young to better cope with the local pathogen fauna.

  • Frequent Flyers Programmes as Mobility Boosters? Implications for Sustainable Aviation.

    • Aviation is an important driver of individual and global mobility and emissions associated with transports. Recent studies indicate that a relatively small, highly mobile part of society may account for a large share of the total distances travelled. This phenomenon is as yet insufficiently understood. In addressing this situation, the paper reviews the processes leading to growth in mobility, and goes on to focus on frequent flyer programmes (FFPs) as an institutionalized framework for high mobility, detailing how these programmes reward and thus increase interest in mobility. Results are linked to a number of observations regarding the interrelationship of high mobility and social status, and substantiated by a survey of FFP members and their perspectives on benefits provided by such programmes. The paper ends with a discussion of the implications of FFPs for growth in aeromobility and its sustainability in a climate change context.

  • Advances in Separation Science : . Molecular Imprinting: Development of Spherical Beads and Optimization of the Formulation by Chemometrics.

    • An intrinsic mathematical model for simulation of fixed bed chromatography was demonstrated and compared to more simplified models. The former model was shown to describe variations in the physical, kinetic, and operating parameters better than the latter ones. This resulted in a more reliable prediction of the chromatography process as well as a better understanding of the underlying mechanisms responsible for the separation. A procedure based on frontal liquid chromatography and a detailed mathematical model was developed to determine effective diffusion coefficients of proteins in chromatographic gels. The procedure was applied to lysozyme, bovine serum albumin, and immunoglobulin γ in Sepharose™ CL-4B. The effective diffusion coefficients were comparable to those determined by other methods.Molecularly imprinted polymers (MIPs) are traditionally prepared as irregular particles by grinding monoliths. In this thesis, a suspension polymerization providing spherical MIP beads is presented. Droplets of pre-polymerization solution were formed in mineral oil with no need of stabilizers by vigorous stirring. The droplets were transformed into solid spherical beads by free-radical polymerization. The method is fast and the performance of the beads comparable to that of irregular particles. Optimizing a MIP formulation requires a large number of experiments since the possible combinations of the components are huge. To facilitate the optimization, chemometrics was applied. The amounts of monomer, cross-linker, and porogen were chosen as the factors in the model. Multivariate data analysis indicated the influence of the factors on the binding and an optimized MIP composition was identified. The combined use of the suspension polymerization method to produce spherical beads with the application of chemometrics was shown in this thesis to drastically reduce the number of experiments and the time needed to design and optimize a new MIP.

    • The automotive industry always strives to achieve light weight components to reduce fuel consumption and to meet environmental requirements. One way to obtain weight reduction is to replace steel components with components made of aluminium or other light weight materials. Aluminium has good corrosion properties and a high strength to weight ratio which makes it favourable in many applications. The increased use of aluminium castings in the automotive industry does also imply that the need for design data for aluminium, increases. Especially for castings, the influence of casting defects are always an issue. For this reason fatigue properties for as-cast sand and permanent mould specimens with different contents of porosity have been studied. Sand cast and permanent mould cast aluminium specimens of two different geometries were fatigue tested in cyclic bending at R= -1. Prior to fatigue test specimens were examined by X-ray and sorted into three quality groups depending on the porosity level. The aim of this work was to investigate the fatigue life for sand cast and permanent mould cast AlSi10Mg with different amounts of porosity. An additional aim was to predict the largest defect contained in a specified volume of a component, by using a statistical analysis of extreme values, and relate it to the fatigue life. The results showed that fatigue strength for a smooth specimen geometry decreases by up to 15% with increased porosity. For specimens with a notched geometry, no influence of porosity on the fatigue strength was found. This is believed to be due to a much smaller volume subject to high stress than for specimens with low stress concentration.

  • Cruise tourism in the Baltic and Bothnian Sea - : A pilot study on maritime tourism : Seagull an EU Interreg III B project

    • Just as globally, tourism is a fast growing phenomenon in the Baltic Sea region. Especially for the new EU member countries tourism appears to be one of the most important instruments for economic development. This has led to increased competition among destinations around the Baltic Sea. The reports shows that cruise tourism is one of the areas in which obvious advantage and synergies could be gained through co-operation. The Baltic Sea countries are visited by about 10 % of the world´s cruise tourists and there is a tendency that even more cruise tourists will choose the Baltic Sea because of safety and security reasons as well as new interesting destinations. The survey and analysis suggest a number of specific opportunities for co-operation, which can contribute to a better utilisation of the potential for cruise tourism and thereby positively influende the economic development in the Baltic Sea region.

  • Fracture toughness properties of duplex stainless steels

    • Good toughness properties in base and weld material enable the use of duplex stainless steels (DSS) in critical applications. DSS offer high strength compared to common austenitic stainless steels. The high strength can be utilized to reduce the wall thickness and accordingly accomplish reduction of cost, welding time and transportation weight, contributing to ecological and energy savings. Although DSS have been used successfully in many applications the last decades, the full utilisation in pressure vessels has been restricted due to conservative design rules. The consequences of failure in a pressure vessel are often very severe and it is accordingly important to verify a high ductility and fracture toughness. In this study fracture toughness data has been generated that has been used to analyse the brittle failure model in the European pressure vessel code EN 13445. The evaluation of the results has been made successfully by the master curve analysis, previously applied to ferritic steels. The master curve analysis includes calculation of a reference temperature, which can be correlated to an impact toughness transition temperature. A correlation between fracture and impact toughness results is necessary for a practically applicable design code. The heat distribution and austenite reformation have been modelled to verify satisfactory toughness properties in the heat affected zone. A similar model was used to evaluate the nucleation and diffusional growth of sigma phase during isothermal heat treatment or continuous cooling. For future stainless steel development, the availability of satisfactory correlations between composition, microstructure and mechanical properties are essential to optimize alloy design. Stainless steel data has been analysed to find approximate relations between mechanical properties and the chemical composition, grain size, ferrite content, product thickness and solution hardening size misfit parameter. The solution hardening effect was successfully predicted by the Labusch-Nabarro relation and multiple regression analyses were used to evaluate hardening equations for stainless steel.

  • A human protein atlas for normal and cancer tissues based on antibody proteomics

    • Antibody-based proteomics provides a powerful approach for the functional study of the human proteome involving the systematic generation of protein-specific affinity reagents. We used this strategy to construct a comprehensive, antibody-based protein atlas for expression and localization profiles in 48 normal human tissues and 20 different cancers. Here we report a new publicly available database containing, in the first version, similar to 400,000 high resolution images corresponding to more than 700 antibodies toward human proteins. Each image has been annotated by a certified pathologist to provide a knowledge base for functional studies and to allow queries about protein profiles in normal and disease tissues. Our results suggest it should be possible to extend this analysis to the majority of all human proteins thus providing a valuable tool for medical and biological research.
