  • Synthetic Heparan Sulfate Mimetic Pixatimod (PG545) Potently Inhibits SARS-CoV-2 by Disrupting the Spike-ACE2 Interaction

    • Heparan sulfate (HS) is a cell surface polysaccharide recently identified as a coreceptor with the ACE2 protein for the S1 spike protein on SARS-CoV-2 virus, providing a tractable new therapeutic target. Clinically used heparins demonstrate an inhibitory activity but have an anticoagulant activity and are supply-limited, necessitating alternative solutions. Here, we show that synthetic HS mimetic pixatimod (PG545), a cancer drug candidate, binds and destabilizes the SARS-CoV-2 spike protein receptor binding domain and directly inhibits its binding to ACE2, consistent with molecular modeling identification of multiple molecular contacts and overlapping pixatimod and ACE2 binding sites. Assays with multiple clinical isolates of SARS-CoV-2 virus show that pixatimod potently inhibits the infection of monkey Vero E6 cells and physiologically relevant human bronchial epithelial cells at safe therapeutic concentrations. Pixatimod also retained broad potency against variants of concern (VOC) including B.1.1.7 (Alpha), B.1.351 (Beta), B.1.617.2 (Delta), and B.1.1.529 (Omicron). Furthermore, in a K18-hACE2 mouse model, pixatimod significantly reduced SARS-CoV-2 viral titers in the upper respiratory tract and virus-induced weight loss. This demonstration of potent anti-SARS-CoV-2 activity tolerant to emerging mutations establishes proof-of-concept for targeting the HS-Spike protein-ACE2 axis with synthetic HS mimetics and provides a strong rationale for clinical investigation of pixatimod as a potential multimodal therapeutic for COVID-19.

    • Multiple roles of reactive oxygen species (ROS) and their consequences for health and disease are emerging throughout biological sciences. This development has led researchers unfamiliar with the complexities of ROS and their reactions to employ commercial kits and probes to measure ROS and oxidative damage inappropriately, treating ROS (a generic abbreviation) as if it were a discrete molecular entity. Unfortunately, the application and interpretation of these measurements are fraught with challenges and limitations. This can lead to misleading claims entering the literature and impeding progress, despite a well-established body of knowledge on how best to assess individual ROS, their reactions, role as signalling molecules and the oxidative damage that they can cause. In this consensus statement we illuminate problems that can arise with many commonly used approaches for measurement of ROS and oxidative damage, and propose guidelines for best practice. We hope that these strategies will be useful to those who find their research requiring assessment of ROS, oxidative damage and redox signalling in cells and in vivo. Reactive oxygen species (ROS) have important roles in health and disease, but are chemically complex and difficult to measure accurately. This consensus statement proposes guidelines and best practices on the nomenclature and assessment of ROS, oxidative reactions and oxidative damage in cells, tissues and in vivo.

  • Standardized Map of Iodine Status in Europe

    • Background:Knowledge about the population's iodine status is important, because it allows adjustment of iodine supply and prevention of iodine deficiency. The validity and comparability of iodine-related population studies can be improved by standardization, which was one of the goals of the EUthyroid project. The aim of this study was to establish the first standardized map of iodine status in Europe by using standardized urinary iodine concentration (UIC) data. Materials and Methods:We established a gold-standard laboratory in Helsinki measuring UIC by inductively coupled plasma mass spectrometry. A total of 40 studies from 23 European countries provided 75 urine samples covering the whole range of concentrations. Conversion formulas for UIC derived from the gold-standard values were established by linear regression models and were used to postharmonize the studies by standardizing the UIC data of the individual studies. Results:In comparison with the EUthyroid gold-standard, mean UIC measurements were higher in 11 laboratories and lower in 10 laboratories. The mean differences ranged from -36.6% to 49.5%. Of the 40 postharmonized studies providing data for the standardization, 16 were conducted in schoolchildren, 13 in adults, and 11 in pregnant women. Median standardized UIC was <100 mu g/L in 1 out of 16 (6.3%) studies in schoolchildren, while in adults 7 out of 13 (53.8%) studies had a median standardized UIC <100 mu g/L. Seven out of 11 (63.6%) studies in pregnant women revealed a median UIC Conclusions:We demonstrate that iodine deficiency is still present in Europe, using standardized data from a large number of studies. Adults and pregnant women, particularly, are at risk for iodine deficiency, which calls for action. For instance, a more uniform European legislation on iodine fortification is warranted to ensure that noniodized salt is replaced by iodized salt more often. In addition, further efforts should be put on harmonizing iodine-related studies and iodine measurements to improve the validity and comparability of results.

  • Register-based information on thyroid diseases in Europe: lessons and results from the EUthyroid collaboration

    • ObjectiveRegisters of diagnoses and treatments exist in different forms in the European countries and are potential sources to answer important research questions. Prevalence and incidence of thyroid diseases are highly dependent on iodine intake and, thus, iodine deficiency disease prevention programs. We aimed to collect European register data on thyroid outcomes to compare the rates between countries/regions with different iodine status and prevention programs. DesignRegister-based cross-sectional study. MethodsNational register data on thyroid diagnoses and treatments were requested from 23 European countries/regions. The provided data were critically assessed for suitability for comparison between countries/regions. Sex- and age-standardized rates were calculated. ResultsRegister data on >= 1 thyroid diagnoses or treatments were available from 22 countries/regions. After critical assessment, data on medication, surgery, and cancer were found suitable for comparison between 9, 10, and 13 countries/regions, respectively. Higher rates of antithyroid medication and thyroid surgery for benign disease and lower rates of thyroid hormone therapy were found for countries with iodine insufficiency before approx. 2001, and no relationship was observed with recent iodine intake or prevention programs. ConclusionsThe collation of register data on thyroid outcomes from European countries is impeded by a high degree of heterogeneity in the availability and quality of data between countries. Nevertheless, a relationship between historic iodine intake and rates of treatments for hyper- and hypothyroid disorders is indicated. This study illustrates both the challenges and the potential for the application of register data of thyroid outcomes across Europe.

  • Microbial experience through housing in a farmyard-type environment alters intestinal barrier properties in mouse colons

    • To close the gap between ultra-hygienic research mouse models and the much more environmentally exposed conditions of humans, we have established a system where laboratory mice are raised under a full set of environmental factors present in a naturalistic, farmyard-type habitat-a process we have called feralization. In previous studies we have shown that feralized (Fer) mice were protected against colorectal cancer when compared to conventionally reared laboratory mice (Lab). However, the protective mechanisms remain to be elucidated. Disruption of the protective intestinal barrier is an acknowledged player in colorectal carcinogenesis, and in the current study we assessed colonic mucosal barrier properties in healthy, feralized C57BL/6JRj male mice. While we found no effect of feralization on mucus layer properties, higher expression of genes encoding the mucus components Fcgbp and Clca1 still suggested mucus enforcement due to feralization. Genes encoding other proteins known to be involved in bacterial defense (Itln1, Ang1, Retnlb) and inflammatory mechanisms (Zbp1, Gsdmc2) were also higher expressed in feralized mice, further suggesting that the Fer mice have an altered intestinal mucosal barrier. These findings demonstrate that microbial experience conferred by housing in a farmyard-type environment alters the intestinal barrier properties in mice possibly leading to a more robust protection against disease. Future studies to unravel regulatory roles of feralization on intestinal barrier should aim to conduct proteomic analyses and in vivo performance of the feralized mice intestinal barrier.

    • Objective: There are few methods that focus on therapists' experiences of supervision. To facilitate the development of psychologist students, a supervisee-centred supervision, based on deliberate practice, feedback informed treatment and self-compassion, was introduced.Methods: This study examines six supervisees’ experiences of a supervisee-centred supervision. A semi- structured interview was used for the collection of the data, which identified two main themes: Learning and Development and five associated sub-themes: structure and purposesfulness, prerequisites, experience-based learning, therapeutic skills and personal development.Conclusion: The experience- and feedback-based approach was perceived as efficient, structured and goal oriented. This created high-focused activity and participation, a strong group dynamic and a good alliance with the supervisors, providing a good climate for learning and development. Focusing on performance and feedback was perceived as a potential obstacle that could create stress and anxiety.

  • Exploring the relationship between personality and gaming disorder symptoms in a sample of Dota 2 players

    • This study explored the relationship between the big five personality traits and gaming disorder symptoms (GDS) in a sample of Dota 2 players. Recent research has indicated that the relationship between personality traits and GDS may depend on the video game genre investigated. However, the association between GDS and personality has yet to be investigated within a specific game, which may be even more relevant to explore as each game offers unique gameplay mechanics that can influence player behavior differently. Thus, the present study investigated the relationship between the big five personality traits and GDS in a sample of video game players from a specific game, DOTA 2 (n = 321, M = 23.25 years, SD = 4.51). Multiple linear regression was used to analyze GDS formatted as a composite score, and multinomial logistic regressions were further conducted for analyses in which gamers were classified into normal gamers (i.e., less than three criteria endorsed), moderate-risk gamers (i.e., four criteria endorsed), and high-risk gamers (i.e., all criteria endorsed). The analysis of the composite score showed a significant relationship between neuroticism, conscientiousness, agreeableness, and GDS. Analyses of the gaming classifications revealed that neuroticism was the most crucial factor concerning differentiating moderate-risk and high-risk from normal gamers. The findings and their practical implications are further discussed.

  • Mining players’ experience in computer games: Immersion affects flow but not presence

    • Understanding how different levels of immersion influence the experiences of flow and presence can shed light on the intricate interplay between these constructs and provide valuable insights into the factors that contribute to engaging and immersive gameplay. The independent variable, immersion, was manipulated in three conditions (high, moderate, and low) in a between-subject design within the video game Minecraft. Participants were asked to complete 15 min of gameplay and then fill out the questionnaires concerning flow and presence. The experiment was conducted remotely on a video-sharing platform. Bayesian analysis revealed an effect of immersion level on flow, while no evidence of an effect was found for the experience of presence. This study provides evidence in favor of a relation between flow and immersion while supporting a presumed double dissociation of immersion from presence. Future research using a Bayesian approach is encouraged to build further knowledge on this research topic.

    • STUDY QUESTION: Can human pre- and peri-pubertal testicular cells obtained from childhood cancer patients, previously treated with chemotherapy, form testicular organoids (TOs)?SUMMARY ANSWER: Organoid formation from testicular tissue collected from childhood cancer patients positively correlates with SRY-Box transcription factor 9 (SOX9) expression in Sertoli cells, which in turn negatively correlates with previous exposure to alkylating chemotherapy.WHAT IS KNOWN ALREADY: Pre- and peri-pubertal boys exposed to highly gonadotoxic therapies can only safeguard their fertility potential through testicular tissue cryopreservation. Today, there is no established clinical tool to restore fertility using these testicular samples. Organoids hold promise in providing fundamental early insights in creating such platforms. However, the generation of TOs that closely resemble the innate testis, to enable a thorough monitoring of the necessary steps for germ cell differentiation and somatic functionalities, remains a challenge.STUDY DESIGN, SIZE, DURATION: We used a Matrigel-based three-layer gradient culture system to generate human TOs and to reveal whether chemotherapy exposure affects TO formation capacity and the functionality of pre- and peri-pubertal testicular somatic cells. Testicular cells of 11 boys (aged 7.7 ± 4.1 (mean ± SD) years) were assessed for TO formation in relation to previous chemotherapy exposure and SOX9 expression in histological sections of paraffin-embedded testicular tissue samples collected on the day of biopsy and compared with testicular tissue samples obtained from 28 consecutive patients (aged 6.9 ± 3.8 (mean ± SD) years). All 39 patients were part of the fertility preservation project NORDFERTIL; an additional 10 samples (from boys aged 5.5 ± 3.5 (mean ± SD) years, without an underlying pathology) in an internal biobank collection were used as controls.PARTICIPANTS/MATERIALS, SETTING, METHODS: We obtained 49 testicular tissue samples from boys aged 0.8-13.4 years. Fresh samples (n = 11) were dissociated into single-cell suspensions and applied to a three-layer gradient culture system for organoid formation. Histological sections of another 28 samples obtained as part of the fertility preservation project NORDFERTIL, and 10 samples from a sample collection of a pathology biobank were used to evaluate the effects of prior exposure to alkylating agents on testicular samples. Testicular organoid formation was defined based on morphological features, such as compartmentalized structures showing cord formation, and protein expression of testicular cell-specific markers for germ and somatic cells was evaluated via immunohistochemical staining. Hormone secretion was analysed by specific enzyme-linked immunosorbent assays for testosterone and anti-Müllerian hormone (AMH) production.MAIN RESULTS AND THE ROLE OF CHANCE: Our results revealed that 4 out of 11 prepubertal testicular samples formed TOs that showed compartmentalized cord-like structures surrounded by interstitial-like areas and increasing levels of both testosterone as well as AMH over a 7-day culture period. We observed that SOX9 expression was correlated positively with TO formation. Moreover, exposure to alkylating agents before biopsy was inversely correlated with SOX9 expression (P = 0.006).LARGE SCALE DATA: N/A. LIMITATIONS, REASONS FOR CAUTION: Due to the limited amount of material available, only 11 out of the 39 pre- and peri-pubertal testicular tissue samples could be used for the organoid formation experiments. The testicular tissue samples obtained from a sample collection of the internal biobank of Department of Pathology, Karolinska University Hospital were considered normal and included in the study if no testicular pathology was reported.     • This manuscript reflects on the affordances and limitations of methodological approaches commonly adopted by science education researchers examining learner identities. Our aims are to unpack the relative strengths and weaknesses of such approaches and note their respective prevalence. In so doing, we identify and critique studies which we consider exemplify the different approaches and, in turn, note the direction of fruitful developments and the nature of key challenges. From our review of the field, we suggest that three discrete methodological approaches can be identified: macro-studies within a psychological tradition; macro-studies within a sociological tradition; and micro-studies within an interpretive tradition. Our review comprised a critical analysis of papers included in the Web of Science databases published between 1998 and 2018. A total of 198 papers examining aspects of learner identity relating to science were identified. Of these, the majority (146) were categorised as micro-studies within an interpretive tradition. We discuss the implications of methodological choices for the advancement of understanding and further note ambiguities in the field particularly in relation to the ways in which learner identity research is conceived. We also raise questions for the field relating to the ways in which findings may be scaled, and how the field might develop to allow stronger theoretical and conceptual coherence.

    • The purpose of this manuscript is to explore how students perceive that online practices have enabled their participation in university physics programmes. In order to conceptualise how students bridge their science participation across physical and online spaces, we make use of the learning ecology perspective. This perspective is complemented with the notion of science capital, analysing how students have been able to strengthen different aspects of science capital through online participation. Data has been generated through semi-structured interviews guided by a timeline, constructed in collaboration between the interviewer and the interviewee. Twenty-one students enrolled in higher education physics have been interviewed, with a focus on their trajectories into higher education physics. The findings focus on four students who in various ways all have struggled to access science learning resources and found ways to utilise online spaces as a complement to their physical learning ecologies. In the manuscript, we show how online practices have contributed to the students’ learning ecologies, e.g. in terms of building networks and functioning as learning support, and how resources acquired through online science practices have both use and exchange value in the wider science community. Online science participation is thus both curiosity driven and founded in instrumental reasons (using online tutoring to pass school science). Furthermore, we argue that online spaces have the potential to offer opportunities for participation and network building for students who do not have access to science activities and science people in their everyday surroundings.

  • Replacing eye trackers in ongoing studies : A comparison of eye‐tracking data quality between the Tobii Pro TX300 and the Tobii Pro Spectrum

    • The Tobii Pro TX300 is a popular eye tracker in developmental eye-tracking research, yet it is no longer manufactured. If a TX300 breaks down, it may have to be replaced. The data quality of the replacement eye tracker may differ from that of the TX300, which may affect the experimental outcome measures. This is problematic for longitudinal and multi-site studies, and for researchers replacing eye trackers between studies. We, therefore, ask how the TX300 and its successor, the Tobii Pro Spectrum, compare in terms of eye-tracking data quality. Data quality—operationalized through precision, accuracy, and data loss—was compared between eye trackers for three age groups (around 5-months, 10-months, and 3-years). Precision was better for all gaze position signals obtained with the Spectrum in comparison to the TX300. Accuracy of the Spectrum was higher for the 5-month-old and 10-month-old children. For the three-year-old children, accuracy was similar across both eye trackers. Gaze position signals from the Spectrum exhibited lower proportions of data loss, and the duration of the data loss periods tended to be shorter. In conclusion, the Spectrum produces gaze position signals with higher data quality, especially for the younger infants. Implications for data analysis are discussed.

  • Effect of the isotope mass on pedestal structure, transport and stability in D, D/T and T plasmas at similar β N and gas rate in JET-ILW type I ELMy H-modes

    • The work describes the pedestal structure, transport and stability in an effective mass (A eff) scan from pure deuterium to pure tritium plasmas using a type I ELMy H-mode dataset in which key parameters that affect the pedestal behaviour (normalized pressure, ratio of the separatrix density to the pedestal density, pedestal ion Larmor radius, pedestal collisionality and rotation) are kept as constant as possible. Experimental results show a significant increase of the density at the pedestal top with increasing A eff, a modest reduction in the temperature and an increase in the pressure. The variations in the pedestal heights are mainly due to a change in the pedestal gradients while only small differences are observed in the pedestal width. A clear increase in the pedestal density and pressure gradients are observed from deuterium to tritium. The experimental results suggest a reduction of the pedestal inter-edge localized mode (inter-ELM) transport from deuterium to tritium. The reduction is likely in the pedestal inter-ELM particle transport, as suggested by the clear increase of the pedestal density gradients. The experimental results suggest also a possible reduction of the pedestal inter-ELM heat transport, however, the large experimental uncertainties do not allow conclusive claims on the heat diffusivity. The clear experimental reduction of eta e (the ratio between density and temperature gradient lengths) in the middle/top of the pedestal with increasing A eff suggests that there may be a link between increasing A eff and the reduction of electron scale turbulent transport. From the modelling point of view, an initial characterization of the behaviour of pedestal microinstabilities shows that the tritium plasma is characterized by growth rates lower than the deuterium plasmas. The pedestal stability of peeling-ballooning modes is assessed with both ideal and resistive magnetohydrodynamics (MHD). No significant effect of the isotope mass on the pedestal stability is observed using ideal MHD. Instead, resistive MHD shows a clear increase of the stability with increasing isotope mass. The resistive MHD results are in reasonable agreement with the experimental results of the normalized pedestal pressure gradient. The experimental and modelling results suggest that the main candidates to explain the change in the pedestal are a reduction in the inter-ELM transport and an improvement of the pedestal stability from deuterium to tritium.

    • A reduction of the pedestal pressure with increasing separatrix density over pedestal density (n (e) (sep)/n (e) (ped)) has been observed in JET. The physics behind this correlation is investigated. The correlation is due to two distinct mechanisms. The increase of n (e) (sep)/n (e) (ped) till approximate to 0.4 shifts the pedestal pressure radially outwards, decreasing the peeling-balloning stability and reducing the pressure height. The effect of the position saturates above n (e) (sep)/n (e) (ped) approximate to 0.4. For higher values, the reduction of the pedestal pressure is ascribed to increased turbulent transport and, likely, to resistive MHD effects. The increase of n (e) (sep)/n (e) (ped) above approximate to 0.4 reduces backward difference n (e) /n (e), increasing eta (e) and the pedestal turbulent transport. This reduces the pressure gradient and the pedestal temperature, producing an increase in the pedestal resistivity. The work suggests that the increase in resistivity might destabilize resistive balloning modes, further reducing the pedestal stability.

    • For young women, inbound identity trajectories into physics are generally regarded as exceptional. In this study, we investigated the experiences that young women have which may support their sustained interest and achievement in physics, and their ongoing inbound trajectories into post-secondary physics education. To understand these experiences, we look to the role of informal physics learning (IPL) environments as spaces which can offer resources that support women's trajectories into physics. In this paper, we highlight the important role of what we call "university-adjacent" IPL experiences-internships, summer schools, and associations that connect secondary students with the research lives of physicists. Focusing on case studies of six women enrolled in post-secondary physics programs across Sweden, we identify the various forms of resources made available through IPL environments, and how these create possibilities for young women to engage in forms of identity work that contribute to the construction of new possible selves in physics. Findings suggest that young women can access important relational and ideational resources through university-adjacent IPL programs. Relational resources included (a) supportive social networks, (b) enduring relationships, and (c) relatability. Importantly, our research finds that IPL opportunities that emphasize relationship building can create immersive experiences which go beyond representation and rather emphasize opportunities to develop practice-linked identities. Ideational resources emerged as (a) sources of information which possibilized physics for participants, and (b) types of information that provided possibilities to learn about the life of a physicist. Finally, while we claim that IPL experiences provide important possibilities for young women to immerse themselves in the practices of physics, we also discuss that these kinds of experiences remain inaccessible to most students, and thus reproduce a certain elitism in the field.

    • Visual routines theory posits that vision is critical for guiding sequential actions in the world. Most studies on the link between vision and sequential action have considered individual agents, while substantial human behaviour is characterized by multi-party interaction. Here, the actions of each person may affect what the other can subsequently do. We investigated task execution and gaze allocation of 19 dyads completing a Duplo-model copying task together, while wearing the Pupil Invisible eye tracker. We varied whether all blocks were visible to both participants, and whether verbal communication was allowed. For models in which not all blocks were visible, participants seemed to coordinate their gaze: The distance between the participants' gaze positions was smaller and dyads looked longer at the model concurrently than for models in which all blocks were visible. This was most pronounced when verbal communication was allowed. We conclude that the way the collaborative task was executed depended both on whether visual information was available to both persons, and how communication took place. Modelling task structure and gaze allocation for human-human and human-robot collaboration thus requires more than the observable behaviour of either individual. We discuss whether an interactive visual routines theory ought to be pursued.

    • In this paper, we present a review of how the various aspects of any study using an eye tracker (such as the instrument, methodology, environment, participant, etc.) affect the quality of the recorded eye-tracking data and the obtained eye-movement and gaze measures. We take this review to represent the empirical foundation for reporting guidelines of any study involving an eye tracker. We compare this empirical foundation to five existing reporting guidelines and to a database of 207 published eye-tracking studies. We find that reporting guidelines vary substantially and do not match with actual reporting practices. We end by deriving a minimal, flexible reporting guideline based on empirical research (Section "An empirically based minimal reporting guideline").

    • Eye trackers are applied in many research fields (e.g., cognitive science, medicine, marketing research). To give meaning to the eye-tracking data, researchers have a broad choice of classification methods to extract various behaviors (e.g., saccade, blink, fixation) from the gaze signal. There is extensive literature about the different classification algorithms. Surprisingly, not much is known about the effect of fixation and saccade selection rules that are usually (implicitly) applied. We want to answer the following question: What is the impact of the selection-rule parameters (minimal saccade amplitude and minimal fixation duration) on the distribution of fixation durations? To answer this question, we used eye-tracking data with high and low quality and seven different classification algorithms. We conclude that selection rules play an important role in merging and selecting fixation candidates. For eye-tracking data with good-to-moderate precision (RMSD < 0.5∘), the classification algorithm of choice does not matter too much as long as it is sensitive enough and is followed by a rule that selects saccades with amplitudes larger than 1.0∘ and a rule that selects fixations with duration longer than 60 ms. Because of the importance of selection, researchers should always report whether they performed selection and the values of their parameters.

    • How well can modern wearable eye trackers cope with head and body movement? To investigate this question, we asked four participants to stand still, walk, skip, and jump while fixating a static physical target in space. We did this for six different eye trackers. All the eye trackers were capable of recording gaze during the most dynamic episodes (skipping and jumping). The accuracy became worse as movement got wilder. During skipping and jumping, the biggest error was 5.8 ∘. However, most errors were smaller than 3 ∘. We discuss the implications of decreased accuracy in the context of different research scenarios.

    • The pupil size artefact (PSA) is the gaze deviation reported by an eye tracker during pupil size changes if the eye does not rotate. In the present study, we ask three questions: 1) how stable is the PSA over time, 2) does the PSA depend on properties of the eye tracker set up, and 3) does the PSA depend on the participants' viewing direction? We found that the PSA is very stable over time for periods as long as 1 year, but may differ between participants. When comparing the magnitude of the PSA between eye trackers, we found the magnitude of the obtained PSA to be related to the direction of the eye-tracker-camera axis, suggesting that the angle between the participants' viewing direction and the camera axis affects the PSA. We then investigated the PSA as a function of the participants' viewing direction. The PSA was non-zero for viewing direction 0∘ and depended on the viewing direction. These findings corroborate the suggestion by Choe et al. (Vision Research 118(6755):48-59, 2016), that the PSA can be described by an idiosyncratic and a viewing direction-dependent component. Based on a simulation, we cannot claim that the viewing direction-dependent component of the PSA is caused by the optics of the cornea.

    • BACKGROUND: Treatment with glucagon-like peptide-1 receptor agonists (GLP-1 RAs) leads to multiple metabolic changes, reduction in glucose levels and body weight are well established. In people with type 2 diabetes, GLP-1 RAs reduce the risk of cardiovascular (CV) disease and may also potentially represent a treatment for fatty liver disease. The mechanisms behind these effects are still not fully elucidated. The aim of the study was to investigate whether treatment with liraglutide is associated with favourable metabolic changes in cases of both CV disease and fatty liver disease.METHODS: In a prespecified post-hoc analysis of a double-blind, placebo-controlled trial in 62 individuals with type 2 diabetes (GLP-1 RA liraglutide or glimepiride, both in combination with metformin), we evaluated the changes in plasma molecular lipids and polar metabolites after 18 weeks of treatment. The lipids and polar metabolites were measured by using ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UHPLC-QTOFMS).RESULTS: In total, 340 lipids and other metabolites were identified, covering 14 lipid classes, bile acids, free fatty acids, amino acids and other polar metabolites. We observed more significant changes in the metabolome following liraglutide treatment compared to with glimepiride, particularly as regards decreased levels of cholesterol esters hexocyl-ceramides, lysophosphatidylcholines, sphingolipids and phosphatidylcholines with alkyl ether structure. In the liraglutide-treated group, lipids were reduced by approximately 15% from baseline, compared to a 10% decrease in the glimepiride group. At the pathway level, the liraglutide treatment was associated with lipid, bile acid as well as glucose metabolism, while glimepiride treatment was associated with tryptophan metabolism, carbohydrate metabolism, and glycerophospholipid metabolism.CONCLUSIONS: Compared with glimepiride, liraglutide treatment led to greater changes in the circulating metabolome, particularly regarding lipid metabolism involving sphingolipids, including ceramides. Our findings are hypothesis-generating and shed light on the underlying biological mechanisms of the CV benefits observed with GLP-1 RAs in outcome studies. Further studies investigating the role of GLP-1 RAs on ceramides and CV disease including fatty liver disease are warranted.TRIAL REGISTRATION: NCT01425580.

    • Cerebrospinal fluid (CSF) biomarkers play an important role in diagnosing Alzheimer's disease (AD) which is characterized by amyloid-beta (A beta) amyloidosis. Here, we used two App knock-in mouse models, App(NL-F/NL-F) and App(NL-G-F/NL-G-F), exhibiting AD-like A beta pathology to analyze how the brain pathologies translate to CSF proteomes by label-free mass spectrometry (MS). This identified several extracellular matrix (ECM) proteins as significantly altered in App knock-in mice. Next, we compared mouse CSF proteomes with previously reported human CSF MS results acquired from patients across the AD spectrum. Intriguingly, the ECM protein decorin was similarly and significantly increased in both App(NL-F/NL-F) and App(NL-G-F/NL-G-F) mice, strikingly already at three months of age in the App(NL-F/NL-F) mice and preclinical AD subjects having abnormal CSF-A beta 42 but normal cognition. Notably, in this group of subjects, CSF-decorin levels positively correlated with CSF-A beta 42 levels indicating that the change in CSF-decorin is associated with early A beta amyloidosis. Importantly, receiver operating characteristic analysis revealed that CSF-decorin can predict a specific AD subtype having innate immune activation and potential choroid plexus dysfunction in the brain. Consistently, in App(NL-F/NL-F) mice, increased CSF-decorin correlated with both AP plaque load and with decorin levels in choroid plexus. In addition, a low concentration of human A beta 42 induces decorin secretion from mouse primary neurons. Interestingly, we finally identify decorin to activate neuronal autophagy through enhancing lysosomal function. Altogether, the increased CSF-decorin levels occurring at an early stage of A beta amyloidosis in the brain may reflect pathological changes in choroid plexus, present in a subtype of AD subjects.

    • This systematic review aims to assess the effectiveness of virtual reality (VR) and gamification interventions in addressing anxiety and depression. The review also seeks to identify gaps in the current VR treatment landscape and provide guidelines for future research and development. A systematic literature search was conducted using Scopus, Web of Science, and PubMed databases, focusing on studies that utilized VR and gamification technology to address anxiety and depression disorders. A total of 2,664 studies were initially identified, 15 of those studies fulfilled the inclusion criteria for this systematic review. The efficacy of VR in addressing anxiety and depression was evident across all included studies. However, the diversity among VR interventions highlights the need for further investigation. It is advised to incorporate more diverse participant samples and larger cohorts and explore a broader spectrum of therapeutic approaches within VR interventions for addressing anxiety and depression to enhance the credibility of future research. Additionally, conducting studies in varying socioeconomic contexts would contribute to a more comprehensive understanding of their real-world applicability.

    • Virtual reality is an emerging field in mental health and has gained widespread acceptance due to its potential to treat various disorders, such as anxiety and depression. This paper presents a bibliometric analysis of virtual reality (VR) use in addressing depression and anxiety from 1995 to 2022. The study analysed 1872 documents using the Scopus database, identifying the field’s most relevant journals and authors. The results indicate that using VR for addressing anxiety and depression is a multidisciplinary field with a wide variety of research topics, leading to significant collaborative research in this area. The Annual Review of Cybertherapy and Telemedicine was identified as the most relevant journal, while Behavior Research and Therapy was found to be the most cited journal. The analysis of keywords suggests that there is more research on using VR for anxiety and related disorders than for depression. Riva G. was identified as the top author in producing research outputs on VR-AD, and the University of Washington emerged as the leading institution in scientific outputs on VR-AD. Thematic and intellectual analyses helped identify the main themes within the research domain, providing valuable insight into the current and future directions of the field.

    • Utbildningar inom fysik är i både svensk och internationell kontext bland de minst jämställda och med lägst mångfald både när det gäller naturvetenskapliga och ingenjörsprogram [1]. I det pågående VR-finansierade projektet “De oväntade naturvetarna” ställer vi frågan vad vi kan lära oss av studenter som gått mot strömmen och valt att läsa fysik. Projektet bygger på livsberättelseintervjuer med 21 fysikstudenter som på något sätt uppfattar sin egen bana till fysiken som icke-konventionell. Vi har undersökt vad som möjliggjort för dessa studenter att ta sig till universitetet, och specifikt en fysikutbildning. I presentationen sammanfattas projektet. Resultatet visar hur aktiviteter utanför skolan kan ha betydelse; informella men universitetsrelaterade aktiviteter som sommarskolor och forskarbesök, men också deltagande i naturvetenskapliga sammanhang online, kan spela stor roll för relations- och identitetsbyggande som möjliggör ett deltagande i fysiken och en väg till en universitetsutbildning [2], [3]. Pågående delstudier vänder på perspektivet och ställer frågorna: Vad möjliggörs för individen genom ett deltagande i fysik, och hur blir vissa val av studier och karriär möjliga och legitima medan andra omöjliggörs i relation till samhälleliga normer, kön, klass och etnicitet. För några av våra informanter har fysik och naturvetenskap varit central för att möjliggöra en starkare känsla av sammanhang, med världen och andra människor. Detta har dock inte erbjudits av skolundervisningen i fysik, utan handlat om andra upplevelser, och först med stöd av vuxenutbildning på folkhögskola och basår har fysiken kunnat ta en central plats i deras liv, vilket har inneburit ett brott från annars alienerande liv och karriärer.   • Performing legitimate choice narratives in physics: possibilities for under-represented physics students

    • Higher education physics has long been a field with a disproportionately skewed representation in terms of gender, class, and ethnicity. Responding to this challenge, this study explores the trajectories of "unexpected" (i.e., demographically under-represented) students into higher education physics. Based on timeline-guided life-history interviews with 21 students enrolled in university physics programs across Sweden, the students' accounts of their trajectories into physics are analyzed as choice narratives. The analysis explores what ingredients are used to tell a legitimate story of physics participation, in relation to dominant discourses in physics culture, and wider social and political discourses. Results indicate that students narrate their choice as based on motivations of physics being a prestigious and challenging subject, of a deep interest in and a natural ability for physics, as well as a wish to use physics for contributing to the world. While most of these affiliations to physics has been documented in earlier research, the study shows how they are negotiated in relation to social locations such as gender, class and migration history, and used to perform an authentic and legitimate choice narrative in the interview situation. Furthermore, the study reports and discusses the possibility of conceiving the role of physics in students' lives as something beyond a "pure", intellectually challenging, and "prestigious" subject. In contrast, and with implications for widening participation, the stories of "unexpected" physics students indicate that physics can be reconceived as socially and altruistically oriented.

    • Background and purpose: Yoga has shown promise as a treatment for depression and anxiety. The present pilot study investigated the feasibility of an eight-week grouptreatment integrating emotion-focused psychoeducation, compassion-focused therapy, and Virya yoga for depression and anxiety in primary healthcare.Materials and methods: Patients seeking treatment for depression and anxiety in a primary healthcare centre completed either an integrative group-treatment (N = 14) or treatment as usual (TAU, N = 17). Outcome measures were analysed pre- and posttreatment. Correlations in the intervention group were investigated between treatment outcomes and amount of yoga practice between sessions.Results: Large within-group effect sizes on all outcome measures were found at posttreatment. Symptom reduction did not differ between groups (p = 0.155). Improvement in alexithymia correlated significantly (p < 0.05) with amount of yoga practice between sessions.Conclusion: Integrating yoga with a psychological group-treatment is a somewhat feasible approach to treatment for depression and anxiety in primary healthcare.

    • Fertility preservation for male childhood cancer survivors not yet capable of producing mature spermatozoa, relies on experimental approaches such as testicular explant culture. Although the first steps in somatic maturation can be observed in human testicular explant cultures, germ cell depletion is a common obstacle. Hence, understanding the spermatogonial stem cell (SSC) niche environment and in particular, specific components such as the seminiferous basement membrane (BM) will allow progression of testicular explant cultures. Here, we revealed that the seminiferous BM is established from 6 weeks post conception with the expression of laminin alpha 1 (LAMA 1) and type IV collagen, which persist as key components throughout development. With prepubertal testicular explant culture we found that seminiferous LAMA 1 expression is disrupted and depleted with culture time correlating with germ cell loss. These findings highlight the importance of LAMA 1 for the human SSC niche and its sensitivity to culture conditions.

Diabetic nephropathy (DN) is the most common cause of end-stage renal disease, affecting similar to 30% of the rapidly growing diabetic population, and strongly associated with cardiovascular risk. Despite this, the molecular mechanisms of disease remain unknown. Methods. RNA sequencing (RNAseq) was performed on paired, micro-dissected glomerular and tubulointerstitial tissue from patients diagnosed with DN [n = 19, 15 males, median (range) age: 61 (30-85) years, chronic kidney disease stages 1-4] and living kidney donors [n = 20, 12 males, median (range) age: 56 (30-70) years]. Results. Principal component analysis showed a clear separation between glomeruli and tubulointerstitium transcriptomes. Differential expression analysis identified 1550 and 4530 differentially expressed genes, respectively (adjusted P < 0.01). Gene ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses highlighted activation of inflammation and extracellular matrix (ECM) organization pathways in glomeruli, and immune and apoptosis pathways in tubulointerstitium of DN patients. Specific gene modules were associated with renal function in weighted gene co-expression network analysis. Increased messengerRNA (mRNA) expression of renal damage markers lipocalin 2 (LCN) and hepatitis A virus cellular receptor1 (HAVCR1) in the tubulointerstitial fraction was observed alongside higher urinary concentrations of the corresponding proteins neutrophil gelatinase-associated lipocalin (NGAL) and kidney injury molecule-1 (KIM-1) in DN patients. Conclusions. Here we present the first RNAseq experiment performed on paired glomerular and tubulointerstitial samples from DN patients. We show that prominent disease-specific changes occur in both compartments, including relevant cellular processes such as reorganization of ECM and inflammation (glomeruli) as well as apoptosis (tubulointerstitium). The results emphasize the potential of utilizing high-throughput transcriptomics to decipher disease pathways and treatment targets in this high-risk patient population.

FCM-MRD performed in 18 laboratories across seven countries was used for risk stratification of 1487 patients with BCP-ALL enrolled in the NOPHO ALL2008 protocol. When no informative FCM-marker was available, risk stratification was based on real-time quantitative PCR. An informative FCM-marker was found in 96.2% and only two patients (0.14%) had non-informative FCM and non-informative PCR-markers. The overall 5-year event-free survival was 86.1% with a cumulative incidence of relapse (CIR5y) of 9.5%. FCM-MRD levels on days 15 (HzR 4.0, p<0.0001), 29 (HzR 2.7, p<0.0001), and 79 (HzR 3.5, p<0.0001) associated with hazard of relapse adjusted for age, cytogenetics, and WBC. The early (day 15) response associated with CIR5y adjusted for day 29 FCM-MRD, with higher levels in adults (median 2.4×10-2 versus 5.2×10-3, p<0.0001). Undetectable FCM- and/or PCR-MRD on day 29 identified patients with a very good outcome (CIR5y=3.2%). For patients who did not undergo transplantation, day 79 FCM-MRD>10-4 associated with a CIR5y=22.1%. In conclusion, FCM-MRD performed in a multicenter setting is a clinically useful method for MRD-based treatment stratification in BCP-ALL.

    • The notion of secure base explains how a child can grow and become independent through access to a significant other (i.e., parent) who is available, encouraging, and noninterfering. The purpose of the current study was to develop an understanding of parental secure base support within the context of youth sport in Sweden, with a specific focus on: (a) what parental behaviors constitute a secure base, and (b) how these behaviors differ across contexts (at home before and after sport, at practice and during competitions). An interpretive descriptive methodology (Thorne, 2016) was used. Interviews were conducted with 13 family triads (children aged 12-15 years) and 1 dyad living in Sweden. Analysis was conducted to illuminate associations, patterns, and relationships within the sample. Analysis led to the development of nine categories of parental behaviors that were perceived to underpin a secure base. Availability was seen to comprise physical presence and support provision, being responsive, and developing positive mental representations. Encouragement encompassed demonstrating that sport participation is valued, motivating to explore sporting endeavors, and reinforcing and rewarding persistence in sports. Interference was described as unrequested interference, requested interference, and intentionally constrained involvement. Additionally, influencing factors such as communication, family structure and culture, were identified. The findings provide an empirical illustration for several behaviors that have been perceived as positive in previous literature, as well as highlighting numerous further complexities, particularly as it relates to interference.

    • Mobile head-worn eye trackers allow researchers to record eye-movement data as participants freely move around and interact with their surroundings. However, participant behavior may cause the eye tracker to slip on the participant’s head, potentially strongly affecting data quality. To investigate how this eye-tracker slippage affects data quality, we designed experiments in which participants mimic behaviors that can cause a mobile eye tracker to move. Specifically, we investigated data quality when participants speak, make facial expressions, and move the eye tracker. Four head-worn eye-tracking setups were used: (i) Tobii Pro Glasses 2 in 50 Hz mode, (ii) SMI Eye Tracking Glasses 2.0 60 Hz, (iii) Pupil-Labs’ Pupil in 3D mode, and (iv) Pupil-Labs’ Pupil with the Grip gaze estimation algorithm as implemented in the EyeRecToo software. Our results show that whereas gaze estimates of the Tobii and Grip remained stable when the eye tracker moved, the other systems exhibited significant errors (0.8–3.1∘ increase in gaze deviation over baseline) even for the small amounts of glasses movement that occurred during the speech and facial expressions tasks. We conclude that some of the tested eye-tracking setups may not be suitable for investigating gaze behavior when high accuracy is required, such as during face-to-face interaction scenarios. We recommend that users of mobile head-worn eye trackers perform similar tests with their setups to become aware of its characteristics. This will enable researchers to design experiments that are robust to the limitations of their particular eye-tracking setup.

  • Effect of resistivity on the pedestal MHD stability in JET

    • The ELM triggering mechanism in tokamaks is not yet fully understood. For example, in the JET tokamak with ITER-like wall (commonly called JET-ILW), the ELMs are sometimes triggered before the ideal peeling-ballooning (PB) boundary is reached. This typically occurs for shots with high input power and high gas rate. The discrepancy between model and experiment has in previous works been clearly correlated with the relative shift between the electron temperature and density pedestals. The discrepancy has also been correlated with the resistivity in the middle-bottom of the pedestal. The present work shows that resistive MHD can have a significant impact on the PB stability of JET pedestals. The inclusion of resistivity removes the correlation between the discrepancy from the PB stability and the relative shift (the difference between the position of the electron temperature and density pedestals) and significantly improves the agreement between PB model and experimental results. The work also shows that the key parameter is the resistivity at the pedestal bottom, near the separatrix, while the resistivity near the middle/top of the pedestal has a negligible effect on the PB stability of JET plasmas.

    • Estimating the gaze direction with a digital video-based pupil and corneal reflection (P-CR) eye tracker is challenging partly since a video camera is limited in terms of spatial and temporal resolution, and because the captured eye images contain noise. Through computer simulation, we evaluated the localization accuracy of pupil-, and CR centers in the eye image for small eye rotations (≪ 1 deg). Results highlight how inaccuracies in center localization are related to 1) how many pixels the pupil and CR span in the eye camera image, 2) the method to compute the center of the pupil and CRs, and 3) the level of image noise. Our results provide a possible explanation to why the amplitude of small saccades may not be accurately estimated by many currently used video-based eye trackers. We conclude that eye movements with arbitrarily small amplitudes can be accurately estimated using the P-CR eye-tracking principle given that the level of image noise is low and the pupil and CR span enough pixels in the eye camera, or if localization of the CR is based on the intensity values in the eye image instead of a binary representation.

  • Evaluation of Effects of Continuous Glucose Monitoring on Physical Activity Habits and Blood Lipid Levels in Persons With Type 1 Diabetes Managed With Multiple Daily Insulin Injections: An Analysis Based on the GOLD Randomized Trial (GOLD 8)

    • People with type 1 diabetes generally view it easier to exercise when having continuous information of the glucose levels. We evaluated whether patients with type 1 diabetes managed with multiple daily insulin injections (MDI) exercised more after initiating continuous glucose monitoring (CGM) and whether the improved glycemic control and well-being associated with CGM translates into improved blood lipids and markers of inflammation. Method: The GOLD trial was a randomized cross-over trial over 16 months where patients used either CGM or capillary self-monitoring of blood glucose (SMBG) over six months, with a four-month wash-out period between the two treatment periods. We compared grade of physical activity, blood lipids, apolipoproteins, and high-sensitivity C-reactive protein (hsCRP) levels during CGM and SMBG. Results: There were 116 patients with information of physical activity estimated by the International Physical Activity Questionnaire (IPAQ) during both CGM and SMBG. No changes were found during CGM or SMBG, IPAQ scores 3305 versus 3878 ( P = .16). In 136 participants with information of blood lipid levels with no change in lipid-lowering medication during the two treatment periods, HbA1c differed by 4.2 mmol/mol (NGSP 0.39%) between SMBG and CGM treatment ( P < .001). No significant changes existed in low-density lipoprotein, high-density lipoprotein, triglycerides, total cholesterol, apolipoprotein A1, apolipoprotein B1, or hsCRP, during CGM and SMBG. Conclusion: Although many patients experience it easier to perform physical activity when monitoring glucose levels with CGM, it does not influence the amount of physical activity in persons with type 1 diabetes. Blood lipids, apolipoprotein, and hsCRP levels were similar during CGM and SMBG.

    • Recent reports on children and adolescents’ physical activity indicate that approximately 80% do not meet the World Health Organisation’s recommended levels of physical activity. Childhood is a critical period of development; as such, it is important to prioritize health promotion in this phase of one’s life. Use of gamification in mobile apps has been shown to positively influence physical activity levels in children and adolescents. Inclusion of behaviour change techniques (BCTs) has been noted to increase the effect of health promoting interventions. Previous research highlights that children and adolescents find the BCTs of goal setting/planning, feedback, reward, self-monitoring, social comparison, and social support to be of interest. The aim of this study was to explore Swedish adolescents’ experience with physical activity and apps with a focus on the influence of BCTs. Three focus group interviews were undertaken with a total of 18 participants (11-15 years of age). The data were explored using reflective thematic analysis. In addition to the BCTs identified in previous research, participants in the present study indicated they were positive about mobile apps providing instruction on how to perform a behaviour, demonstration of the behaviour, and the influence of removal of reward. Other attractive features of physical activities apps included developing a streak, competition, and provision of an adequate level of challenge. The present study contributes to a deeper understanding of how BCTs and other features can be implemented in physical activity apps, where individualization and renewal appear to be more important than the actual content features. These findings are important for future work to create interventions that increase the level of physical activity among adolescents both in Sweden and other countries.

  • Isotope physics of heat and particle transport with tritium in JET-ILW type-I ELMy H-mode plasmas

    • As part the DTE2 campaign in the JET tokamak, we conducted a parameter scan in T and D-T complementing existing pulses in H and D. For the different main ion masses, type-I ELMy H-modes at fixed plasma current and magnetic field can have the pedestal pressure varying by a factor of 4 and the total pressure changing from beta(N )= 1.0 to 3.0. We investigated the pedestal and core isotope mass dependencies using this extensive data set. The pedestal shows a strong mass dependence on the density, which influences the core due to the strong coupling between both plasma regions. To better understand the causes for the observed isotope mass dependence in the pedestal, we analysed the interplay between heat and particle transport and the edge localised mode (ELM) stability. For this purpose, we developed a dynamic ELM cycle model with basic transport assumptions and a realistic neutral penetration. The temporal evolution and resulting ELM frequency introduce an additional experimental constraint that conventional quasi-stationary transport analysis cannot provide. Our model shows that a mass dependence in the ELM stability or in the transport alone cannot explain the observations. One requires a mass dependence in the ELM stability as well as one in the particle sources. The core confinement time increases with pedestal pressure for all isotope masses due to profile stiffness and electromagnetic turbulence stabilisation. Interestingly, T and D-T plasmas show an improved core confinement time compared to H and D plasmas even for matched pedestal pressures. For T, this improvement is largely due to the unique pedestal composition of higher densities and lower temperatures than H and D. With a reduced gyroBohm factor at lower temperatures, more turbulent drive in the form of steeper gradients is required to transport the same amount of heat. This picture is supported by quasilinear flux-driven modelling using TGLF-SAT2 within Astra. With the experimental boundary condition TGLF-SAT2 predicts the core profiles well for gyroBohm heat fluxes >15, however, overestimates the heat and particle transport closer to the turbulent threshold.
