  • Functional influence of masticatory muscles on the fibre characteristics and capillary distribution in growing ferrets (Mustela putonusfuro)--a histochemical analysis.

    • OBJECTIVE: Changes in the masticatory function of ferrets have been shown to cause alterations in their craniofacial morphology. The aim of the present study was to investigate how the functional changes influence the histochemical characteristics of fibres of the masticatory muscles of the animal. DESIGN: Forty five-week old male ferrets were divided into two groups: one group was fed a diet of hard pellets (HD group), and one group was fed the same diet but softened with water (SD group). After 6 months, specimens of temporalis, masseter and digastricus were taken from each ferret and prepared for enzyme histochemical analysis RESULTS: Type I muscle fibres constituted the majority of the temporalis and the masseter, while the digastricus mainly comprised type II muscle fibres. The fibre-type composition did not differ significantly between the two groups. The mean cross-sectional area of type I and type II fibres in the temporalis and the masseter, and type II fibres in the digastricus was significantly greater in the HD group than the SD group. Furthermore, the HD group had a significantly greater number of capillaries per fibre, but not per surface unit (mm2). CONCLUSIONS: The results suggest that the muscle fibre size and the number of capillaries per fibre of the masticatory muscles are readily adaptesd to the altered masticatory muscle functions.

  • Differential investment in the Banggai cardinalfish : can females adjust egg size close to egg maturation to match the attractiveness of a new partner?

    • To test whether females can change their egg investment according to the different attractiveness ( i.e. size as measured by standard length, Ls) of a new mate after eggs have already matured in response to an earlier mate, female Banggai cardinalfish Pterapogon kauderni were first allowed to produce eggs for small (unattractive) or large (attractive) males. Then, when spawning was initiated, but prior to actual spawning, their partner was switched to either a significantly larger or a significantly smaller partner, respectively. A strong positive correlation between egg size and days until spawning with the second male was found for the females initially paired to a small and then a large male. Within a few days, these females apparently increased their egg size to match the attractiveness of their new male. No correlation between days until spawning and egg size in females initially paired to a large and then a small male, however was found, so apparently females were unable to adjust egg size in response to a decrease in mate attractiveness. Consequently, it is suggested that females can increase their egg size investment even after the onset of egg maturation and that this change can be quite rapid.

    • Deleted in malignant brain tumors 1 (DMBT1) has been proposed as a candidate tumor suppressor for brain and epithelial cancer. Initial studies suggested loss of expression rather than mutation as the predominant mode of DMBT1 inactivation. However, in situ studies in lung cancer demonstrated highly sophisticated changes of DMBT1 expression and localization, pointing to a chronological order of events. Here we report on the investigation of DMBT1 in breast cancer in order to test whether these principles might also be attributable to other tumor types. Comprehensive mutational analyses did not uncover unambiguous inactivating DMBT1 mutations in breast cancer. Expression analyses in the human and mouse mammary glands pointed to the necessity of DMBT1 induction. While age-dependent and hormonal effects could be ruled out, 9 of 10 mice showed induction of Dmbt1 expression after administration of the carcinogen 7,12-dimethybenz(alpha)anthracene prior to the onset of tumorigenesis or other histopathological changes. DMBT1 displayed significant up-regulation in human tumor-flanking tissues compared to in normal breast tissues (P < 0.05). However, the breast tumor cells displayed a switch from lumenal secretion to secretion to the extracellular matrix and a significant down-regulation compared to that in matched normal flanking tissues (P < 0.01). We concluded that loss of expression also is the predominant mode of DMBT1 inactivation in breast cancer. The dynamic behavior of DMBT1 in lung carcinoma is fully reflected in breast cancer, which suggests that this behavior might be common to tumor types arising from monolayered epithelia.
