  • Flagella-mediated secretion of a novel Vibrio cholerae cytotoxin affecting both vertebrate and invertebrate hosts

    • Using Caenorhabditis elegans as an infection host model for Vibrio cholerae predator interactions, we discovered a bacterial cytotoxin, MakA, whose function as a virulence factor relies on secretion via the flagellum channel in a proton motive force-dependent manner. The MakA protein is expressed from the polycistronic makDCBA (motility-associated killing factor) operon. Bacteria expressing makDCBA induced dramatic changes in intestinal morphology leading to a defecation defect, starvation and death in C. elegans. The Mak proteins also promoted V. cholerae colonization of the zebrafish gut causing lethal infection. A structural model of purified MakA at 1.9 Å resolution indicated similarities to members of a superfamily of bacterial toxins with unknown biological roles. Our findings reveal an unrecognized role for V. cholerae flagella in cytotoxin export that may contribute both to environmental spread of the bacteria by promoting survival and proliferation in encounters with predators, and to pathophysiological effects during infections.

    • The PREDICTS project-Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)-has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

Using structural MRI data from 45,615 individuals aged 3-96 years, we demonstrate distinct patterns of apparent brain aging in several brain disorders and reveal genetic pleiotropy between apparent brain aging in healthy individuals and common brain disorders.

    • The Carnegie Supernova Project-II (CSP-II) was an NSF-funded, four-year program to obtain optical and near-infrared observations of a Cosmology sample of similar to 100 Type. Ia supernovae located in the smooth Hubble flow (0.03 less than or similar to z less than or similar to 0.10). Light curves were also obtained of a Physics sample composed of 90 nearby Type. Ia supernovae at z <= 0.04 selected for near-infrared spectroscopic timeseries observations. The primary emphasis of the CSP-II is to use the combination of optical and near-infrared photometry to achieve a distance precision of better than 5%. In this paper, details of the supernova sample, the observational strategy, and the characteristics of the photometric data are provided. In a companion paper, the near-infrared spectroscopy component of the project is presented.

  • Conjugation factors controlling F-plasmid antibiotic resistance transmission

    • The rapid horizontal transmission of many antibiotic resistance genes between bacterial host cells on conjugative plasmids is a major cause of the accelerating antibiotic resistance crisis. Preventing understanding and targeting conjugation, there currently are no experimental platforms for fast and cost-efficient screening of genetic effects on antibiotic resistance transmission by conjugation. We introduce a novel experimental framework to screen for conjugation based horizontal transmission of antibiotic resistance between >60,000 pairs of cell populations in parallel. Plasmid-carrying donor strains are constructed in high throughput. We then mix the resistance plasmid carrying donors with recipients in a design where only transconjugants can reproduce, measure growth in dense intervals and extract transmission times as the growth lag. As proof-of-principle, we exhaustively explored chromosomal genes controlling F plasmid donation within E. coli populations, by screening the Keio deletion collection at high replication. We recover all six known chromosomal gene mutants affecting conjugation and identify >50 novel factors, all of which diminish antibiotic resistance transmission. We verify 10 of the novel genes' effects in a liquid mating assay. The new framework holds great potential for exhaustive disclosing of candidate targets for helper drugs that delay resistance development in patients and societies and improves the longevity of current and future antibiotics.

  • K2-141 b: A 5-M⊕ super-Earth transiting a K7 V star every 6.7 hours

    • We report on the discovery of K2-141 b (EPIC 246393474 b), an ultra-short-period super-Earth on a 6.7 h orbit transiting an active K7 V star based on data from K2 campaign 12. We confirmed the planet's existence and measured its mass with a series of follow-up observations: seeing-limited Muscat imaging, NESSI high-resolution speckle observations, and FIES and HARPS high-precision radial-velocity monitoring. K2-141 b has a mass of 5.31 ± 0.46 M ⊗ and radius of 1.54 -0.09 +0.10 R ⊗ , yielding a mean density of 8.00 -1.45 +1.83 g cm -3 and suggesting a rocky-iron composition. Models indicate that iron cannot exceed ∼70% of the total mass. With an orbital period of only 6.7 h, K2-141 b is the shortest-period planet known to date with a precisely determined mass.

    • We present a detailed analysis of HARPS-N radial velocity observations of K2-100, a young and active star in the Praesepe cluster, which hosts a transiting planet with a period of 1.7 d. We model the activity-induced radial velocity variations of the host star with a multidimensional Gaussian Process framework and detect a planetary signal of 10.6 ± 3.0 m s−1, which matches the transit ephemeris, and translates to a planet mass of 21.8 ± 6.2 M. We perform a suite of validation tests to confirm that our detected signal is genuine. This is the first mass measurement for a transiting planet in a young open cluster. The relatively low density of the planet, 2.04+−006661 g cm−3, implies that K2-100b retains a significant volatile envelope. We estimate that the planet is losing its atmosphere at a rate of 1011–1012 g s−1 due to the high level of radiation it receives from its host star.

    • To study the physical mechanisms of phenomena such as polar mesospheric summer echoes, the SCRAP (Scattering of Radar waves on Aerosols in Plasmas) experiment aimed to validate theories on density fluctuations in dusty plasmas. The SCRAP team developed two identical free falling units (FFUs) designed to create a cloud of copper particles once they eject from the REXUS17 sounding rocket 124 seconds after launch. By using the EISCAT incoherent scatter radar system to observe the cloud, the SCRAP experiment proposed to relate theoretical predictions to a controlled object. The SCRAP experiment was launched from ESRANGE on March the 17th 2015. The FFUs GPS signal was lost during launch and the units were therefore not found. Moreover, no backscattering from the copper cloud was observed by the radar.

    • Staphylococcus aureus is implicated in disease progression in cutaneous T-cell lymphoma (CTCL). Here, we demonstrate that malignant T cell lines derived from CTCL patients as well as primary malignant CD4+ T cells from Sézary syndrome patients are considerably more resistant to alpha-toxin-induced cell death than their non-malignant counterparts. Thus, in a subset of Sézary syndrome patients the ratio between malignant and non-malignant CD4+ T cells increases significantly following exposure to alpha-toxin. Whereas toxin-induced cell death is ADAM10 dependent in healthy CD4+ T cells, resistance to alpha-toxin in malignant T cells involves both downregulation of ADAM10 as well as other resistance mechanisms. In conclusion, we provide first evidence that Staphylococcus aureus derived alpha-toxin can tilt the balance between malignant and non-malignant CD4+ T cells in CTCL patients. Consequently, alpha-toxin may promote disease progression through positive selection of malignant CD4+ T cells, identifying alpha-toxin as a putative drug target in CTCL.

  • Single-cell heterogeneity in Sézary syndrome

    • Sezary syndrome (SS) is an aggressive leukemic variant of cutaneous T-cell lymphoma (CTCL) with a median life expectancy of less than 4 years. Although initial treatment responses are often good, the vast majority of patients with SS fail to respond to ongoing therapy. We hypothesize that malignant T cells are highly heterogeneous and harbor subpopulations of SS cells that are both sensitive and resistant to treatment. Here, we investigate the presence of single-cell heterogeneity and resistance to histone deacetylase inhibitors (HDACi) within primary malignant T cells from patients with SS. Using single-cell RNA sequencing and flow cytometry, we find that malignant T cells from all investigated patients with SS display a high degree of single-cell heterogeneity at both the mRNA and protein levels. We show that this heterogeneity divides the malignant cells into distinct subpopulations that can be isolated by their expression of different surface antigens. Finally, we show that treatment with HDACi (suberanilohydroxamic acid and romidepsin) selectively eliminates some subpopulations while leaving other subpopulations largely unaffected. In conclusion, we show that patients with SS display a high degree of single-cell heterogeneity within the malignant T-cell population, and that distinct subpopulations of malignant T cells carry HDACi resistance. Our data point to the importance of understanding the heterogeneous nature of malignant SS cells in each individual patient to design combinational and new therapies to counter drug resistance and treatment failure.

    • To evaluate clinical performance of a new automated cell-free (cf)DNA assay in maternal plasma screening for trisomies 21, 18, and 13, and to determine fetal sex.Maternal plasma samples from 1200 singleton pregnancies were analyzed with a new non-sequencing cfDNA method, which is based on imaging and counting specific chromosome targets. Reference outcomes were determined by either cytogenetic testing, of amniotic fluid or chorionic villi, or clinical examination of neonates.The samples examined included 158 fetal aneuploidies. Sensitivity was 100% (112/112) for trisomy 21, 89% (32/36) for trisomy 18, and 100% (10/10) for trisomy 13. The respective specificities were 100%, 99.5%, and 99.9%. There were five first pass failures (0.4%), all in unaffected pregnancies. Sex classification was performed on 979 of the samples and 99.6% (975/979) provided a concordant result.The new automated cfDNA assay has high sensitivity and specificity for trisomies 21, 18, and 13 and accurate classification of fetal sex, while maintaining a low failure rate. The study demonstrated that cfDNA testing can be simplified and automated to reduce cost and thereby enabling wider population-based screening.

    • Deficient expression of SATB1 hampers thymocyte development and results in inept T-cell lineages. Recent data implicate dysregulated SATB1 expression in the pathogenesis of mycosis fungoides, the most frequent variant of cutaneous T-cell lymphoma. Here, we report on a disease stage-associated decrease of SATB1 expression and an inverse expression of STAT5 and SATB1 in situ. STAT5 inhibited SATB1 expression through induction of microRNA-155. Decreased SATB1 expression triggered enhanced expression of IL-5 and IL-9 (but not IL-6 and IL-32), whereas increased SATB1 expression had the opposite effect, indicating that the microRNA-155 target SATB1 is a repressor of IL-5 and IL-9 in malignant T cells. In accordance, inhibition of STAT5 and its upstream activator JAK3 triggered increased SATB1 expression and a concomitant suppression of IL-5 and IL-9 expression in malignant T cells. In conclusion, we provide a mechanistic link between the proto-oncogenic JAK3/STAT5/microRNA-155 pathway, SATB1, and cytokines linked to CTCL severity and progression, indicating that SATB1 dysregulation is involved in cutaneous T-cell lymphoma pathogenesis.

  • TESS's first planet: A super-Earth transiting the naked-eye star π Mensae

    • We report on the confirmation and mass determination of π Men c, the first transiting planet discovered by NASA's TESS space mission. π Men is a naked-eye (V = 5.65 mag), quiet G0 V star that was previously known to host a sub-stellar companion (π Men b) on a longperiod (Porb = 2091 days), eccentric (e = 0.64) orbit. Using TESS time-series photometry, combined with Gaia data, published UCLES at AAT Doppler measurements, and archival HARPS at ESO-3.6m radial velocities, we found that π Men c is a close-in planet with an orbital period of Porb = 6.27 days, a mass of Mc = 4.52 ± 0.81 Mo, and a radius of Rc = 2.06 ± 0.03 Ro. Based on the planet's orbital period and size, π Men c is a super-Earth located at, or close to, the radius gap, while its mass and bulk density suggest it may have held on to a significant atmosphere. Because of the brightness of the host star, this system is highly suitable for a wide range of further studies to characterize the planetary atmosphere and dynamical properties. We also performed an asteroseismic analysis of the TESS data and detected a hint of power excess consistent with the seismic values expected for this star, although this result depends on the photometric aperture used to extract the light curve. This marginal detection is expected from pre-launch simulations hinting at the asteroseismic potential of the TESS mission for longer, multi-sector observations and/or for more evolved bright stars.

  • The Transiting Multi-planet System HD 3167: A 5.7 M ⊕ Super-Earth and an 8.3 M ⊕ Mini-Neptune

    • HD 3167 is a bright (V = 8.9 mag) K0 V star observed by NASA’s K2 space mission during its Campaign 8. It has recently been found to host two small transiting planets, namely, HD 3167b, an ultra-short-period (0.96 days) super-Earth, and HD 3167c, a mini-Neptune on a relatively long-period orbit (29.85 days). Here we present an intensive radial velocity (RV) follow-up of HD 3167 performed with the FIES@NOT, HARPS@ESO-3.6 m, and HARPS-N@TNG spectrographs. We revise the system parameters and determine radii, masses, and densities of the two transiting planets by combining the K2 photometry with our spectroscopic data. With a mass of 5.69 ± 0.44 M⊕, a radius of 1.574 ± 0.054 R⊕, and a mean density of {8.00}-0.98+1.10 g cm^-3, HD 3167b joins the small group of ultra-short-period planets known to have rocky terrestrial compositions. HD 3167c has a mass of 8.33-1.85+1.79 M⊕ and a radius of 2.74}-0.100+0.106 R⊕, yielding a mean density of 2.21-0.53+0.56 g cm^-3, indicative of a planet with a composition comprising a solid core surrounded by a thick atmospheric envelope. The rather large pressure scale height (∼350 km) and the brightness of the host star make HD 3167c an ideal target for atmospheric characterization via transmission spectroscopy across a broad range of wavelengths. We found evidence of additional signals in the RV measurements but the currently available data set does not allow us to draw any firm conclusions on the origin of the observed variation.

    • We present the first ~7.′5 × 11.′5 velocity-resolved (~0.2 km/s) map of the [C II] 158 μm line toward the Orion molecular cloud1 (OMC1) taken with the Herschel/HIFI instrument. In combination with far-IR (FIR) photometric images and velocity-resolved maps of the H41α hydrogen recombination and CO J = 2–1 lines, this data set provides an unprecedented view of the intricate small-scale kinematics of the ionized/photodissociation region (PDR)/molecular gas interfaces and of the radiative feedback from massive stars. The main contribution to the [C II] luminosity (~85%) is from the extended, FUV-illuminated face of the cloud (G0 > 500, nH > 5 × 10^3 cm^−3) and from dense PDRs (G0>~10^4, nH>~10^5 cm^−3) at the interface between OMC 1 and the H II region surrounding the Trapezium cluster. Around ~15% of the [C II] emission arises from a different gas component without a CO counterpart. The [C II] excitation, PDR gas turbulence, line opacity (from [13C II]), and role of the geometry of the illuminating stars with respect to the cloud are investigated. We construct maps of the L[CII]/LFIR and LFIR/MGas ratios and show that L[CII]/LFIR decreases from the extended cloud component (~10^−2–10^−3) to the more opaque star-forming cores (~10^-3-10−4). The lowest values are reminiscent of the “[C II] deficit” seen in local ultraluminous IR galaxies hosting vigorous star formation. Spatial correlation analysis shows that the decreasing L[C II]/LFIR ratio correlates better with the column density of dust through the molecular cloud than with LFIR/MGas. We conclude that the [C II]-emitting column relative to the total dust column along each line of sight is responsible for the observed L[C II]/LFIR variations through the cloud.

    • Previous studies report high and increasing iron (Fe) concentrations in boreal river mouths. This Fe has shown relatively high stability to salinity-induced aggregation in estuaries. The aim of this study was to understand how the speciation of Fe affects stability over salinity gradients. For Fe to remain in suspension interactions with organic matter (OM) are fundamental and these interactions can be divided in two dominant phases: organically complexed Fe, and colloidal Fe (oxy)hydroxides, stabilized by surface interactions with OM. The stability of these two Fe phases was tested using mixing experiments with river water and artificial seawater. Fe speciation of river waters and salinity-induced aggregates was determined by synchrotron-based extended X-ray absorption fine structure (EXAFS) spectroscopy. The relative contribution of the two Fe phases varied widely across the sampled rivers. Moreover, we found selective removal of Fe (oxy)hydroxides by aggregation at increasing salinity, while organically complexed Fe was less affected. However, Fe-OM complexes were also found in the aggregates, illustrating that the control of Fe stability is not explained by the prevalence of the respective Fe phases alone. Factors such as colloid size and the chemical composition of the OM may also impact the behavior of Fe species.

  • Exoplanets around Low-mass Stars Unveiled by K2

    • We present the detection and follow-up observations of planetary candidates around low-mass stars observed by the K2 mission. Based on light-curve analysis, adaptive-optics imaging, and optical spectroscopy at low and high resolution (including radial velocity measurements), we validate 16 planets around 12 low-mass stars observed during K2 campaigns 5–10. Among the 16 planets, 12 are newly validated, with orbital periods ranging from 0.96 to 33 days. For one of the planets (K2-151b), we present ground-based transit photometry, allowing us to refine the ephemerides. Combining our K2 M-dwarf planets together with the validated or confirmed planets found previously, we investigate the dependence of planet radius R p on stellar insolation and metallicity [Fe/H]. We confirm that for periods P ≲ 2 days, planets with a radius Rp≳ 2 R⊕ are less common than planets with a radius between 1–2 R⊕. We also see a hint of the “radius valley” between 1.5 and 2 R⊕, which has been seen for close-in planets around FGK stars. These features in the radius/period distribution could be attributed to photoevaporation of planetary envelopes by high-energy photons from the host star, as they have for FGK stars. For the M dwarfs, though, the features are not as well defined, and we cannot rule out other explanations such as atmospheric loss from internal planetary heat sources or truncation of the protoplanetary disk. There also appears to be a relation between planet size and metallicity: the few planets larger than about 3 R⊕ are found around the most metal-rich M dwarfs.

    • We report on the discovery of three transiting super-Earths around K2-155 (EPIC 210897587), a relatively bright early M dwarf (V = 12.81 mag) observed during Campaign 13 of the NASA K2 mission. To characterize the system and validate the planet candidates, we conducted speckle imaging and high-dispersion optical spectroscopy, including radial velocity measurements. Based on the K2 light curve and the spectroscopic characterization of the host star, the planet sizes and orbital periods are 1.55 -0.17 +0.20 R ⊕ and 6.34365 ±0.00028 days for the inner planet; 1.95 -0.22 +0.27 R ⊕ and 13.85402 ±0.00088 days for the middle planet; and 1.64 -0.17 +0.18 R ⊕ and 40.6835 ±0.0031 days for the outer planet. The outer planet (K2-155d) is near the habitable zone, with an insolation 1.67 ±0.38 times that of the Earth. The planet's radius falls within the range between that of smaller rocky planets and larger gas-rich planets. To assess the habitability of this planet, we present a series of three-dimensional global climate simulations, assuming that K2-155d is tidally locked and has an Earth-like composition and atmosphere. We find that the planet can maintain a moderate surface temperature if the insolation proves to be smaller than ∼1.5 times that of the Earth. Doppler mass measurements, transit spectroscopy, and other follow-up observations should be rewarding, as K2-155 is one of the optically brightest M dwarfs known to harbor transiting planets.

    • We report the discovery of two transiting planets orbiting K2-290 (EPIC 249624646), a bright (V = 11.11) late F-type star residing in a triple-star system. It was observed during Campaign 15 of the K2 mission, and in order to confirm and characterize the system, follow-up spectroscopy and AO imaging were carried out using the FIES, HARPS, HARPS-N, and IRCS instruments. From AO imaging and Gaia data we identify two M-dwarf companions at a separation of 113 ± 2 and 2467+−177155 au. From radial velocities, K2 photometry, and stellar characterization of the host star, we find the inner planet to be a mini-Neptune with a radius of 3.06 ± 0.16 R and an orbital period of P = 9.2 d. The radius of the mini-Neptune suggests that the planet is located above the radius valley, and with an incident flux of F ∼ 400 F, it lies safely outside the super-Earth desert. The outer warm Jupiter has a mass of 0.774 ± 0.047 MJ and a radius of 1.006 ± 0.050 RJ, and orbits the host star every 48.4 d on an orbit with an eccentricity e < 0.241. Its mild eccentricity and mini-Neptune sibling suggest that the warm Jupiter originates from in situ formation or disc migration.

    • We present the discovery and confirmation of two new transiting giant planets from the Kepler extended mission K2. K2-260 b is a hot Jupiter transiting a V = 12.7 F6V star in K2 Field 13, with a mass and radius of M = 1.39-0.06+0.05M⊙and R = 1.69 ± 0.03 R. The planet has an orbital period of P = 2.627 d, and a mass and radius of MP= 1.42-0.32+0.31MJand RP= 1.552-0.057+0.048RJ. This is the first K2 hot Jupiter with a detected secondary eclipse in the Kepler bandpass, with a depth of 71 ± 15 ppm, which we use to estimate a geometric albedo of Ag~ 0.2. We also detected a candidate stellar companion at 0.6 arcsec from K2-260; we find that it is very likely physically associated with the system, in which case it would be an M5-6V star at a projected separation of ~400 au. K2-261 b is a warm Saturn transiting a bright (V = 10.5) G7IV/V star in K2 Field 14. The host star is a metal rich ([Fe/H] = 0.36 ± 0.06), mildly evolved 1.10-0.02+0.01M⊙star with R = 1.65 ± 0.04 R. Thanks to its location near the main-sequence turn-off, we can measure a relatively precise age of 8.8-0.3+0.4Gyr. The planet has P = 11.633 d, MP= 0.223 ± 0.031 MJ, and RP= 0.850-0.022+0.026RJ, and its orbit is eccentric (e = 0.39 ± 0.15). Its brightness and relatively large transit depth make this one of the best-known warm Saturns for follow-up observations to further characterize the planetary system.

    • Background: We aimed to evaluate differences in outcome between patients admitted to intensive care unit (ICU) after elective versus acute surgery in a multinational cohort of very old patients (80 years; VIP). Predictors of mortality, with special emphasis on frailty, were assessed.Methods: In total, 5063 VIPs were induded in this analysis, 922 were admitted after elective surgery or intervention, 4141 acutely, with 402 after acute surgery. Differences were calculated using Mann-Whitney-U test and Wilcoxon test. Univariate and multivariable logistic regression were used to assess associations with mortality.Results: Compared patients admitted after acute surgery, patients admitted after elective surgery suffered less often from frailty as defined as CFS (28% vs 46%; p < 0.001), evidenced lower SOFA scores (4 +/- 5 vs 7 +/- 7; p < 0.001). Presence of frailty (CFS >4) was associated with significantly increased mortality both in elective surgery patients (7% vs 12%; p = 0.01), in acute surgery (7% vs 12%; p = 0.02).Conclusions: VIPs admitted to ICU after elective surgery evidenced favorable outcome over patients after acute surgery even after correction for relevant confounders. Frailty might be used to guide clinicians in risk stratification in both patients admitted after elective and acute surgery.

    • In cutaneous T cell lymphomas (CTCL), miR-21 is aberrantly expressed in skin and peripheral blood and displays anti-apoptotic properties in malignant T cells. It is, however, unclear exactly which cells express miR-21 and what mechanisms regulate miR-21. Here, we demonstrate miR-21 expression in situ in both malignant and reactive lymphocytes as well as stromal cells. qRT-PCR analysis of 47 patients with mycosis fungoides (MF) and Sezary Syndrome (SS) confirmed an increased miR- 21 expression that correlated with progressive disease. In cultured malignant T cells miR-21 expression was inhibited by Tofacitinib (CP-690550), a clinical-grade JAK3 inhibitor. Chromatin immunoprecipitation (ChIP) analysis showed direct binding of STAT5 to the miR-21 promoter. Cytokine starvation ex vivo triggered a decrease in miR-21 expression, whereas IL-2 induced an increased miR-21 expression in primary SS T cells and cultured cytokine-dependent SS cells (SeAx). siRNA-mediated depletion of STAT5 inhibited constitutive- and IL-2- induced miR-21 expression in cytokine- independent and dependent T cell lines, respectively. IL-15 and IL-2 were more potent than IL-21 in inducing miR-21 expression in the cytokine-dependent T cells. In conclusion, we provide first evidence that miR-21 is expressed in situ in CTCL skin lesions, induced by IL-2 and IL-15 cytokines, and is regulated by STAT5 in malignant T cells. Thus, our data provide novel evidence for a pathological role of IL-2Rg cytokines in promoting expression of the oncogenic miR-21 in CTCL.

    • We present the discovery of three small planets transiting K2-136 (LP 358 348, EPIC 247589423), a late K dwarf in the Hyades. The planets have orbital periods of 7.9757 ± 0.0011, 17.30681-0.00036+0.00034, and 25.5715-0.0040+0.0038 days, and radii of 1.05 ± 0.16, 3.14 ± 0.36, and 1.55-0.21+0.24 Rearth , respectively. With an age of 600–800 Myr, these planets are some of the smallest and youngest transiting planets known. Due to the relatively bright (J = 9.1) host star, the planets are compelling targets for future characterization via radial velocity mass measurements and transmission spectroscopy. As the first known star with multiple transiting planets in a cluster, the system should be helpful for testing theories of planet formation and migration.

    • Low-cost sensors continuously measuring blood glucose levels in intervals of a few minutes and mobile platforms combined with machine-learning (ML) solutions enable personalized precision health and disease management. ML solutions must be adapted to different sensor technologies, analysis tasks and individuals. This raises the issue of scale for creating such adapted ML solutions. We present an approach for predicting blood glucose levels for diabetics up to one hour into the future. The approach is based on recurrent neural networks trained in an end-to-end fashion, requiring nothing but the glucose level history for the patient. The model outputs the prediction along with an estimate of its certainty, helping users to interpret the predicted levels. The approach needs no feature engineering or data pre-processing, and is computationally inexpensive.

The Kepler extended mission, also known as K2, has provided the community with a wealth of planetary candidates that orbit stars typically much brighter than the targets of the original mission. These planet candidates are suitable for further spectroscopic follow-up and precise mass determinations, leading ultimately to the construction of empirical mass-radius diagrams. Particularly interesting is to constrain the properties of planets that are between Earth and Neptune in size, the most abundant type of planet orbiting Sun-like stars with periods of less than a few years. Aims. Among many other K2 candidates, we discovered a multi-planetary system around EPIC 246471491, referred to henceforth as K2-285, which contains four planets, ranging in size from twice the size of Earth to nearly the size of Neptune. We aim here at confirming their planetary nature and characterizing the properties of this system. Methods. We measure the mass of the planets of the K2-285 system by means of precise radial-velocity measurements using the CARMENES spectrograph and the HARPS-N spectrograph. Results. With our data we are able to determine the mass of the two inner planets of the system with a precision better than 15%, and place upper limits on the masses of the two outer planets. Conclusions. We find that K2-285b has a mass of M b = 9.68 -1.37+1.21 M · and a radius of R b = 2.59 -0.06+0.06 R · , yielding a mean density of ρ b = 3.07 -0.45+0.45 g cm -3 , while K2-285c has a mass of M c = 15.68 -2.13+2.28 M · , radius of R c = 3.53 -0.08+0.08 R · , and a mean density of ρ c = 1.95 -0.28+0.32 g cm -3 . For K2-285d (R d = 2.48 -0.06+0.06 R · ) and K2-285e (R e = 1.95 -0.05+0.05 R · ), the upper limits for the masses are 6.5 M · and 10.7 M · , respectively. The system is thus composed of an (almost) Neptune-twin planet (in mass and radius), two sub-Neptunes with very different densities and presumably bulk composition, and a fourth planet in the outermost orbit that resides right in the middle of the super-Earth/sub-Neptune radius gap. Future comparative planetology studies of this system would provide useful insights into planetary formation, and also a good test of atmospheric escape and evolution theories.

  • Greening of the brown-dwarf desert EPIC 212036875b: a 51 M-J object in a 5-day orbit around an F7V star

    • Context. Although more than 2000 brown dwarfs have been detected to date, mainly from direct imaging, their characterisation is difficult due to their faintness and model-dependent results. In the case of transiting brown dwarfs, however, it is possible to make direct high-precision observations. Aims. Our aim is to investigate the nature and formation of brown dwarfs by adding a new well-characterised object, in terms of its mass, radius and bulk density, to the currently small sample of less than 20 transiting brown dwarfs. Methods. One brown dwarf candidate was found by the KESPRINT consortium when searching for exoplanets in the K2 space mission Campaign 16 field. We combined the K2 photometric data with a series of multicolour photometric observations, imaging, and radial velocity measurements to rule out false positive scenarios and to determine the fundamental properties of the system. Results. We report the discovery and characterisation of a transiting brown dwarf in a 5.17-day eccentric orbit around the slightly evolved F7V star EPIC 212036875. We find a stellar mass of 1.15 +/- 0.08 M-circle dot, a stellar radius of 1.41 +/- 0.05 R-circle dot, and an age of 5.1 +/- 0.9 Gyr. The mass and radius of the companion brown dwarf are 51 +/- 2 M-J and 0.83 +/- 0.03 R-J, respectively, corresponding to a mean density of 108(-13)(+15) g cm(-3). Conclusions. EPIC 212036875 b is a rare object that resides in the brown-dwarf desert. In the mass-density diagram for planets, brown dwarfs, and stars, we find that all giant planets and brown dwarfs follow the same trend from similar to 0.3 M-J to the turn-over to hydrogen burning stars at similar to 73 M-J. EPIC 212036875 b falls close to the theoretical model for mature H/He dominated objects in this diagram as determined by interior structure models. We argue that EPIC 212036875 b formed via gravitational disc instabilities in the outer part of the disc, followed by a quick migration. Orbital tidal circularisation may have started early in its history for a brief period when the brown dwarf's radius was larger. The lack of spin-orbit synchronisation points to a weak stellar dissipation parameter (Q(star)' greater than or similar to 10(8)), which implies a circularisation timescale of greater than or similar to 23 Gyr, or suggests an interaction between the magnetic and tidal forces of the star and the brown dwarf.

    • Streptococcus pyogenes of the M1 serotype is commonly associated with invasive streptococcal infections and development of streptococcal toxic shock syndrome. The M1 protein is a powerful inducer of inflammatory responses for several human cell types, but the reason why M1 protein-related strains is over-represented in invasive streptococcal diseases is still not understood. This study was undertaken to investigate if soluble M1 protein can aggravate the severity of streptococcal skin infections in respect to inflammation, leucocyte recruitment, and tissue remodelling as seen in patients with cellulitis and necrotizing fasciitis. We found that HaCaT cells are able to recruit activated leucocytes when encountering M1 protein. Neither the bacterial protein nor activated leucocytes caused cell damage on HaCaT cells, instead HaCaT cells responded to the bacterial virulence factor by releasing several proteins protective against bacterial infection and leucocyte responses. However, although not cytotoxic, M1 protein completely abolished wound healing abilities of HaCaT cells. Taken together, our results demonstrate that M1 protein is a critical virulence factor that can augment streptococcal skin infection suggesting that the protein is an interesting target for drug development.

    • Context. Multiplanet systems are excellent laboratories to test planet formation models as all planets are formed under the same initial conditions. In this context, systems transiting bright stars can play a key role, since planetary masses, radii, and bulk densities can be measured. Aims. GJ 9827 (K2-135) has recently been found to host a tightly packed system consisting of three transiting small planets whose orbital periods of 1.2, 3.6, and 6.2 days are near the 1:3:5 ratio. GJ 9827 hosts the nearest planetary system (~30 pc) detected by NASA's Kepler or K2 space mission. Its brightness (V = 10.35 mag) makes the star an ideal target for detailed studies of the properties of its planets. Methods. Combining the K2 photometry with high-precision radial-velocity measurements gathered with the FIES, HARPS, and HARPS-N spectrographs we revised the system parameters and derive the masses of the three planets. Results. We find that GJ 9827 b has a mass of Mb = 3.69-0.46+0.48 M and a radius of Rb = 1.58-0.13+0.14 R, yielding a mean density of ρb = 5.11-1.27+1.74 g cm-3. GJ 9827 c has a mass of Mc = 1.45-0.57+0.58 M, radius of Rc = 1.24-0.11+0.11 R, and a mean density of ρc = 4.13-1.77+2.31 g cm-3. For GJ 9827 d, we derive Md = 1.45-0.57+0.58 M, Rd = 1.24-0.11+0.11 R, and ρd = 1.51-0.53+0.71 g cm-3. Conclusions. GJ 9827 is one of the few known transiting planetary systems for which the masses of all planets have been determined with a precision better than 30%. This system is particularly interesting because all three planets are close to the limit between super-Earths and sub-Neptunes. The planetary bulk compositions are compatible with a scenario where all three planets formed with similar core and atmosphere compositions, and we speculate that while GJ 9827 b and GJ 9827 c lost their atmospheric envelopes, GJ 9827 d maintained its primordial atmosphere, owing to the much lower stellarirradiation. This makes GJ 9827 one of the very few systems where the dynamical evolution and the atmosphericescape can be studied in detail for all planets, helping us to understand how compact systems form and evolve.

    • The mechanisms underlying longitudinal displacements of the arterial wall, that is, displacements of the wall layers along the artery, and the resulting intramural shear strain remain largely unknown. We have already found that these displacements undergo profound changes in response to catecholamines. Wall shear stress, closely related to wall shear rate, represents the viscous drag exerted on the vessel wall by flowing blood. The aim of the work described here was to study possible relations between the wall shear rate and the longitudinal displacements. We investigated the carotid arteries of five anesthetized pigs in different hemodynamic situations using in-house developed non-invasive ultrasound techniques. The study protocol included administration of epinephrine, norepinephrine and β-blockade (metoprolol). No significant correlation between longitudinal displacement of the intima-media complex and wall shear rate was found. This result suggests that one or multiple pulsatile forces other than wall shear stress are also working along arteries, strongly influencing arterial wall behavior.

    • We report the discovery and characterization of HD 89345b (K2-234b; EPIC 248777106b), a Saturn-sized planet orbiting a slightly evolved star. HD 89345 is a bright star (V = 9.3 mag) observed by the K2 mission with 1 min time sampling. It exhibits solar-like oscillations. We conducted asteroseismology to determine the parameters of the star, finding themass and radius to be 1.12-0.01+0.04M⊙and 1.657-0.004+0.020R⊙, respectively. The star appears to have recently left the main sequence, based on the inferred age, 9.4-1.3+0.4Gyr, and the non-detection of mixed modes. The star hosts a 'warm Saturn' (P = 11.8 d, Rp= 6.86 ± 0.14 R⊕). Radial-velocity follow-up observations performed with the FIbre-fed Echelle Spectrograph, HARPS, and HARPS-N spectrographs show that the planet has a mass of 35.7 ± 3.3 M⊕. The data also show that the planet's orbit is eccentric (e≈0.2). An investigation of the rotational splitting of the oscillation frequencies of the star yields no conclusive evidence on the stellar inclination angle. We further obtained Rossiter-McLaughlin observations, which result in a broad posterior of the stellar obliquity. The planet seems to confirm to the same patterns that have been observed for other sub-Saturns regarding planet mass and multiplicity, orbital eccentricity, and stellar metallicity.

    • Background: Many translational MR biomarkers derive from measurements of the water proton longitudinal relaxation rate R 1 , but evidence for between-site reproducibility of R 1 in small-animal MRI is lacking. Objective: To assess R 1 repeatability and multi-site reproducibility in phantoms for preclinical MRI. Methods: R 1 was measured by saturation recovery in 2% agarose phantoms with five nickel chloride concentrations in 12 magnets at 5 field strengths in 11 centres on two different occasions within 1–13 days. R 1 was analysed in three different regions of interest, giving 360 measurements in total. Root-mean-square repeatability and reproducibility coefficients of variation (CoV) were calculated. Propagation of reproducibility errors into 21 translational MR measurements and biomarkers was estimated. Relaxivities were calculated. Dynamic signal stability was also measured. Results: CoV for day-to-day repeatability (N = 180 regions of interest) was 2.34% and for between-centre reproducibility (N = 9 centres) was 1.43%. Mostly, these do not propagate to biologically significant between-centre error, although a few R 1 -based MR biomarkers were found to be quite sensitive even to such small errors in R 1 , notably in myocardial fibrosis, in white matter, and in oxygen-enhanced MRI. The relaxivity of aqueous Ni 2+ in 2% agarose varied between 0.66 s −1 mM −1 at 3 T and 0.94 s −1 mM −1 at 11.7T. Interpretation: While several factors affect the reproducibility of R 1 -based MR biomarkers measured preclinically, between-centre propagation of errors arising from intrinsic equipment irreproducibility should in most cases be small. However, in a few specific cases exceptional efforts might be required to ensure R 1 -reproducibility.

    • Detta hypotestestförprojekt har haft som mål att validera potentialen för ett nyttmultifunktionellt kaskadbioraffinaderi som producerar fyra viktigaplattformsprodukter från ett Kraftverk: cellulosa, lignobränsle, biovätgas ochbiopolymer. Eftersom cellulosa- och ligninproduktionslinjer är väl karakteriseradefokuserar Bioplastogen på att utvärdera och identifiera möjligheter ochutmaningar vid vätgas- och bioplastproduktion från hemicellulosarestprodukter.Utförandet har varit indelat i tre faser dvs 1) identifierandet av hemicellulosarikaströmmar för biovätgasprocessen, 2) fermenteringexperiment för produktion avvätgas och organisk syra, 3) omvandling av organisk syra till PHA genomaktiverat slam. Valet av hemicellulosafraktion baserades på ett litteraturstudieoch genom diskussioner med personal på olika bruk och på LignoBoostpilotanläggning. Eftersom Lignoboost restprodukt har alternativaanvändningsområdet och hög halt inhibitorer togs ett beslut att användahemicellulosa som extraheras innan den blir en del av svartlut.Hemicellulosafraktionen bestod av 19g/L organiskt material varav sockerarternafrämst var pentoser. Fraktionens effekt på vätgasproduktionen utvärderas iserumflaskor och bioreaktorer med osmotoleranta stammar avCaldicellulosiruptor sacchrolyticus. Bioprocessen utvecklades även förindustriella applikation. Försök i serumflaskor visade potential förbiovätgasproduktion som inte kunde replikeras i reaktorsförsöken. Förslag på nystrategi för att förbättra biovätgasproduktionen också i reaktorförsken kan vara attantinger anpassa mikroorganismerna till de nya substraten och/eller att användablandkulturer som kan vara mer robusta mot denna typ av substrat.Den organiska biprodukten från biovätgasprocessen användes förbiopolymerproduktionsprocessen. Två olika slam från Sjölundavattenreningsanläggning användes och visade en bra potential genom attackumulera mer än 25% av gPHA/gVSS. Detta resultat stärkte projekthypotesen.Nästa steg blir att använda ett acklimatiserat slam från biologisk vattenrening frånett pappersmassabruk.Genom Bioplastogen har vi stärkt våra hypoteser men också identifieradestrategier för fortsatt förbättring som förhoppningsvis kan bli implementerade iansökt fortsättningsprojekt – HyPer
