Expression of Prostate Specific Antigen on the Surface of a Filamentous Phage

- We have constructed two phagemid vectors containing the gene coding for human Prostate Specific Antigen (PSA) translationally fused to a pelB signal sequence and minor coat protein III of phage fd leading to phage particles that carry PSA on their tips. Phages were characterized by Western blotting and by panning, using biotinylated monoclonal anti-PSA antibodies immobilized onto streptavidin-coated microtitration wells. Binding of PSA-phages was 102- to 5×103-fold more effective than that of wildtype phages. Trypsin treatment of the phage abolished the specific binding. Competitive panning with different concentrations of added purified PSA demonstrated that binding was PSA specific. Finally, Western-blot analysis confirmed that full-length and shorter degradation products of the fusion between PSA and protein III were visible as probed with anti-PSA antibody.

- Fifteen patients with isolated old rupture of the anterior cruciate ligament (ACL) were examined to judge their functional disability. The following basic factors were used for evaluation: thigh muscle strength testing, stabilometry in a one-leg stance with straight and flexed knee, gait analysis, performance tests, knee function score, activity grading scale, and clinical joint laxity tests. We found that these rehabilitated patients mainly had impaired performance in those tests which greatly stressed the knee joint's sagittal stability. One-leg hop length was impaired for the injured limb. This group of patients had bilateral impairment of postural control compared with a reference group. This dysfunction was not revealed when comparing limbs. A derotation brace (ECKO) had neither a positive nor a negative effect in the tests. Different aspects of knee function evaluation are emphasized. Some of the altered functions are suggested to be due to central adjustments of motor control.

- Objectives.: To assess the comparability of the Tandem-R and IMx serumprostate-specific antigen (PSA) assays across levels of the ratio of free-to-total serum PSA found in a community-based population of healthy men. Methods.: Banked serum samples from the baseline component of the Olmsted CountyStudy of Urinary Symptoms and Health Status Among Men were thawed and analyzed using the Tandem-R and IMx PSA assays. Serum levels also were determined for the free, noncomplexed form of PSA, PSA complexed to alpha-1 antichymotrypsin, and total PSA with a research-based immunofluorometric assay. Results.: The results of the Tandem-R and IMx assays were strongly correlated at alllevels of the ratio of free-to-total serum PSA. The Spearman correlation coefficients ranged from 0.87 to 0.98 (all p < 0.001). The relationship between the Tandem-R and IMx assays, however, differed at low levels of free-to-total serum PSA compared with high levels. In the lowest 10th percentile of the ratio of free-to-total serum PSA (0.04 to 0.18), the IMx assay read lower than the Tandem-R (slope ± standard error = 0.92±0.04, intercept ± standard error = 0.21 ± 0.14); whereas in the upper 10th percentile of free-to-total ratio (0.46 to 0.65) the IMx assay yielded values higher than the Tandem-R assay (slope = 1.21 ± 0.07, intercept = 0.14 ± 0.05). In the middle 90%, the slope did not statistically differ from 1.0. Conclusions.: For the majority of men, results of the Tandem-R and IMx PSA assays were virtually identical. The small differences found would not be of clinical significance for most men but should be considered when comparing results of different assays in sequential determinations for a specific man.

- Under 1990 beslöt Trafiksäkerhetsverket (TSV) att genomföra försök med haveriundersökningar. Föreliggande rapport redovisar resultaten och en utvärdering av denna försöksverksamhet samt en rekommendation rörande fortsatt verksamhet.Inledningsvis ges en kort historik och en beskrivning av den aktuella situationen vad gäller haveriundersökningar i Sverige och utomlands.Rapporten ger vidare en kort beskrivning av planläggningsarbetet för projektet och av den utbildning som personalen fick före projektets start. Utbildningen avsåg såväl praktiska aspekter - alarmeringsrutiner, datainsamling och rapportering - som bakomliggande teoretiska resonemang och modeller.Den metodik som användes i projektet är en fallstudiemetodik med inriktning på händelseförloppet före kollision. Den teoretiska modell som ligger till grund för såväl datainsamling som analysarbete betonar systemaspekten hos trafiken dvs den ser trafik som ett samspel mellan trafikanter, fordon samt väg- och trafikmiljön. Analysen av olyckorna sker i termer av den informationsprocess som de inblandade trafikanterna svarat för i samband med olycksförloppet. Med detta som utgångspunkt förklaras alltid en olycka som resultatet av att trafikanten gått miste om någon information som varit nödvändig för att kunna undvika olyckan.

- The structures of the anionic Cu, clusters with n I 10 have been investigated by using correlated wave functions includingthe effects of core (3d)-valence correlation through a model operator. The Cu core orbitals (including 3d) are describedby an effective core potential (ECP). The accuracy of both the ECP description and the treatment of corevalence correlationeffects is established by comparing with all-electron calculations for the smaller clusters. Recent experimental data for theelectron detachment energy (EDE) and HOMO-LUMO separations is combined with the computed total energies to establishthe structure of the observed Cui clusters. The EDE's are found to be strongly structure dependent, which is used to determinewhich of the low-lying structures is observed experimentally.
