    • Ett övergripande syfte med boken är att fördjupa kunskapen om och förståelsen för ojämställdhet och jämställdhet mellan könen. I centrum står det obetalda arbetet i hemmet och det betalda arbetet på den reguljära arbetsmarknaden. Innehållet och syftet kan delas i två huvuddelar. Den första delen kretsar kring frågan vilka faktorer som påverkar hur mycket män och kvinnor engagerar sig i förvärvsarbete respektive hushållsarbete, samt hur dessa olika typer av arbeten hänger samman och påverkar varandra. Den andra centrala forskningsfrågan är vad den tid och energi som kvinnor och män ägnar åt förvärvsarbete respektive hushållsarbete betyder för välbefinnande och upplevelse av rollkonflikter. Resultaten grundar sig på empiriska studier av sambanden mellan arbetsliv, familjeliv och kön som genomförts under 1990-talets senare del samt under den inledande delen av 2000-talet. De flesta har publicerats i tidskrifter och böcker som separata artiklar, företrädesvis på engelska. Artiklarna har omvandlats och integrerats så att de tillsammans bildar en helhet som syftar till att öka förståelsen för vad som påverkar kvinnors och mäns engagemang i arbetsliv och familjeliv, samt vad detta betyder för välbefinnandet. Exempel på resultat: - Det är först när mannen är öppet arbetslös och kvinnan arbetar heltid i ett förhållande som hushållsarbetet är någorlunda jämnt fördelat. - Skillnader i könsrollsvärderingar förklarar en del av de länderskillnader som finns rörande hushållsarbetets fördelning och kvinnors arbetsmarknadsdeltagande. - Cirka 40 procent av alla heltidsarbetande mödrar skulle vilja arbeta mindre, vilket kan jämföras med 11 procent bland män i samma situation. - Det är fler kvinnor i Sverige, jämfört med svenska män och kvinnor från andra länder, som upplever att arbetet ofta kommer i konflikt med familjelivet.

  • Effect of caffeine on exercise capacity and function in prostate cancer survivors.

    • PURPOSE: This study aimed to examine the acute effect of caffeine on exercise capacity, exercise-related fatigue, and functional performance in prostate cancer survivors.METHODS: In this randomized, placebo-controlled, double-blind crossover study, 30 prostate cancer survivors (age, 70.3 ± 7.7 yr; body mass, 80.5 ± 13.0 kg; mean ± SD) consumed 6.04 ± 0.16 mg·kg(-1) of anhydrous caffeine or a placebo 1 h before completing a battery of exercise capacity and functional performance tests. Testing sessions were separated by 3-4 wk. Immediate fatigue and perceived exertion were measured directly pre- and postexercise at both testing sessions.RESULTS: Caffeine increased exercise capacity by 7.93 s (+3.0%; P = 0.010); however, postexercise fatigue and perception of exertion were comparable with the placebo session (P = 0.632 and P = 0.902, respectively). Increases in isometric grip strength trended toward significance in both dominant (+2.9%; P = 0.053) and nondominant (+2.1%; P = 0.061) hands in the caffeine trial compared with placebo. Caffeine ingestion did not result in improvements in performance for any of the remaining functional measures, including the timed up-and-go test, repeated chair stands, 6-m fast walk, and 6-m backward tandem walk. Systolic blood pressure and HR were significantly increased (P = 0.006 and P = 0.040, respectively) upon completion of the testing battery when compared with placebo.CONCLUSIONS: Consumption of caffeine 1 h before exercise induced improvements in exercise capacity and muscular strength in prostate cancer survivors. However, there was no change in exercise-related fatigue when compared with placebo despite reduction in timed performance of the 400-m walk. Caffeine seems to enhance exercise tolerance through improved performance with no subsequent increase in fatigue or perception of exertion and may be an appropriate strategy to promote exercise participation in prostate cancer survivors.

  • Human growth patterns - with focus on pubertal growth and secular changes

    • Introduction: Human growth is a dynamic process, an indicator of health and disease. Previous used growth models have been limited in describing the pubertal growth spurt. Aim: The overall aim of this thesis is to increase knowledge regarding human growth. The specific aims were to; explore pubertal growth in detail with new estimates from the QEPS model (paper I), investigate associations between peak BMI-SDS in childhood and subsequent growth (II), evaluate secular changes in adult height (AH) for Nordic countries (in Sweden including parental heights of study populations) and analyse during which growth phases (fetal/infancy/childhood/ puberty) changes occur ( III), study changes in growth patterns from birth to AH in two Swedish population based cohorts born in 1974 and 1990 (IV). Methods: The main study material was based on longitudinal growth data (height/ weight) from two population based Swedish growth cohorts (~4000) born around 1974 and 1990. The novel QEPS growth model was used for analysing height/growth patterns. By applying four mathematical functions, QEPS describes the individual height gain; Quadratic (ongoing from before birth to AH), Exponential (rapid gain during fetal life/infancy), Puberty (adding the specific pubertal growth), Stop (ending gain in height to AH). The Nordic study analysed height data from present and past growth references (Denmark/Finland/Norway) and Swedish growth studies, comparing height at different ages up to AH (in Sweden including parental height). Results and conclusions: New estimates from QEPS model including markers of quality (CI) and SDS for onset, middle and end of puberty showed: the later onset of puberty, the greater the AH. Pubertal gain due to the specific pubertal P-function was independent of age at onset of puberty; boys had higher total gain during puberty due to P-function growth than to QESfunction, reversed for girls. The novel pubertal growth estimates enable a more detailed analyse of pubertal growth than previously possible (I). Higher childhood BMI-SDS was associated with more growth before onset of puberty, earlier pubertal growth and less pubertal height gain for both sexes, over the entire BMI-spectrum (II). The Nordic countries have similar positive secular changes in AH (females +4-7/males +5-15mm/decade), mainly due to increased height in childhood, the change was more pronounced in parental heights, i.e. earlier decades and for males. Earlier pubertal growth was seen in the most recent growth study in all four countries (III). When studying changes in height between the 1974 and 1990 cohorts, a positive change in AH was found (1990), due to more growth during childhood in both sexes and during puberty in girls. QEPS is effective detecting small changes of growth patterns, in cohorts born only 16 years apart (IV).

    • This study reports the in situ crystal growth of HKUST-1 on electrospun cellulose nanofibers. Two different methods for introducing carboxyl groups on the nanofiber surface were used; HKUST-1 was then synthesized on the cellulose nanofiber surface using a layer-by-layer approach. The distribution of HKUST-1 on the nanofiber surface was highly dependent on the type of anionic pretreatment. The loading of HKUST-1 on the nanofiber surface could be controlled by the layer-by-layer synthesis and the BET surface area could be increased by a factor of 44 to 440 m2 g-1.

  • Fire-Induced Radiological Integrated Assessment : Fire properties of selected materials and products

    • Characterization of emissions from fires in a laboratory-controlled environment are presented in this report. The project is initiated by the CERN HSE Unit and is called FIRIA, Fire-Induced Radiological Integrated Assessment. The objective of FIRIA is to enhance the knowledge of aerosols emitted from fires in order to develop dispersion models of radiologically-activated material in case of fire. In this report, several normally occurring combustible products and materials are tested in a standardized setup for fire tests, the cone calorimeter. In the cone calorimeter, standardized fire tests according to ISO 5660-1:2015 have been performed as well as fire tests at reduced oxygen concentrations in a vitiated air chamber. As an additional setup, aerosol measurement equipment was coupled to the cone calorimeter ventilation duct to characterize the emitted aerosols as in the particle size distribution, mass yield and elemental analysis.The results show peak heat release rates for oil at 1100 kW/m2 at an incident heat flux of 50 kW/m2. Similar results for the plastic materials were 800 kW/m2. For cables and insulating plastic materials peak heat release rates at an incident heat flux of 50 kW/m2 were around 350 kW/m2. Significant for most of the cables was a heat release rate curve with two distinct peaks. This is proposed to be due to the outer combustible sheath burning first followed by the interior plastic insulating material of the cables burning. There could also be heat transfer effects and cracking of the material surface contributing to the two peaks. Nevertheless, for some cables a low incident heat flux led to only one peak indicating that only the sheath ignited. Time to ignition varied between the materials but was increased as the incident heat flux decreased. Reduced oxygen concentration in the vitiated air chamber also prolonged the ignition time as well as the heat release rates. The critical heat flux to ignite the cables was calculated to be just below 10 kW/m2. The oil and two cable types were tested in the vitiated air chamber to perform tests at reduced oxygen concentrations. These tests were performed to retrieve specific fire properties as well as specific emissions from such conditions. The tests were performed with a progressively lower oxygen concentrations until no ignition of the sample occurred. Results showed an ignition limit around 11-13 % oxygen at incident heat fluxes of 20-30 kW/m2. The tests in the vitiated air chamber is described in subreport FIRIA- Fire properties of selected materials and products in reduced oxygen conditions.

    • Gene expression patterns help to measure and characterize the effect of environmental perturbations at the cellular and organism-level. Complicating interpretation is the presence of uncharacterized or "hypothetical" gene functions for a large percentage of genomes. This is particularly evident in Daphnia genomes, which contains many regions coding for "hypothetical proteins" and are significantly divergent from many of the available arthropod model species, but might be ecologically important. In the present study, we developed a gene expression database, the Daphnia stressor database (http://www.daphnia-stressordb.uni-hamburg.de/dsdbstart.php), built from 90 published studies on Daphnia gene expression. Using a comparative genomics approach, we used the database to annotate D. galeata transcripts. The extensive body of literature available for Daphnia species allowed to associate stressors with gene expression patterns. We believe that our stressor based annotation strategy allows for better understanding and interpretation of the functional role of the understudied hypothetical or uncharacterized Daphnia genes, thereby increasing our understanding of Daphnia's genetic and phenotypic variability.

    • Over the decades, several natural history studies on patients with primary (PPMS) or secondary progressive multiple sclerosis (SPMS) were reported from international registries. In PPMS, a consistent heterogeneity on long-term disability trajectories was demonstrated. The aim of this study was to identify subgroups of patients with SPMS with similar longitudinal trajectories of disability over time.MethodsAll patients with MS collected within Big MS registries who received an SPMS diagnosis from physicians (cohort 1) or satisfied the Lorscheider criteria (cohort 2) were considered. Longitudinal Expanded Disability Status Scale (EDSS) scores were modelled by a latent class growth analysis (LCGA), using a non-linear function of time from the first EDSS visit in the range 3–4.ResultsA total of 3613 patients with SPMS were included in the cohort 1. LCGA detected three different subgroups of patients with a mild (n=1297; 35.9%), a moderate (n=1936; 53.6%) and a severe (n=380; 10.5%) disability trajectory. Median time to EDSS 6 was 12.1, 5.0 and 1.7 years, for the three groups, respectively; the probability to reach EDSS 6 at 8 years was 14.4%, 78.4% and 98.3%, respectively. Similar results were found among 7613 patients satisfying the Lorscheider criteria.ConclusionsContrary to previous interpretations, patients with SPMS progress at greatly different rates. Our identification of distinct trajectories can guide better patient selection in future phase 3 SPMS clinical trials. Additionally, distinct trajectories could reflect heterogeneous pathological mechanisms of progression.

  • Geomorphology of the Huang He ice sheet area: towards a reconstruction of the glacial history of the northeastern Tibetan Plateau

    • Recent terrestrial cosmogenic nuclide (TCN) studies on end moraines of the Tibetan Plateau have yielded a first synthesis of the timing of mountain glacier and ice field maximum extents at discrete times in the past. Although these allow important constraints on the presence of expanded ice on the plateau, they don’t address the question of the presence of areally more extensive ice sheet configurations where mountain ice complexes from discrete mountain blocks coalesce to form larger bodies. Two ice sheets hypothesised to have covered parts of the Tibetan Plateau are the Tibetan ice sheet and the areally much more restricted Huang He ice sheet. In this study we have focussed on the hypothesised Huang He ice sheet area in the headwaters of the Huang He and Yangtze rivers on the north-eastern margin of the Tibetan Plateau.Two mountain blocks from which ice might have emanated to inundate the plateau surface around it and form the ice sheet are the marginally-located Anyemaqen and centrally-located Bayan Har Mountains. Of these the Anyemaqen is located closer to the edge of the plateau, is higher, wetter, and is ornamented with glaciers today, which, according to TCN studies have been more extensive during marine oxygen isotope stages 3, 2, and 1.Using Landsat 7 ETM+ satellite imagery we have mapped the glacial geomorphology of the entire hypothesised Huang He ice sheet area (50,000-70,000 km2) and concentrated our TCN field sampling to its core area, the Bayan Har Mountains. The area displays widespread morphological evidence of glacial erosion and deposition, particularly around the higher mountain blocks. The erosional landforms include large-scale glacial troughs, U-shaped valleys and occasional lake basins, and small-scale lateral meltwater channels. The depositional landforms include primarily lateral and end-moraines, but also hummocky moraines and drumlins. Field inspection has yielded observations of tills and erratic boulders. Taken together, these traces comprise an impressive record of multiple large-scale erosional events as witnessed by cross-cutting relationships of glacial valleys and multiple glacier advances through the Bayan Har Mountain valleys, some of which terminated onto the plateau surface, by the presence of suites of end-moraines and associated meltwater traces.The mapping exercise thus far has established a clear patchiness to the erosional imprint of ice in the uplands comprising the Huang He ice sheet area. Although the integrated imprint of erosion is clear and displays a pattern of topographically-forced selective linear erosion, the rates of glacial landscape change in the absence of TCN measurements remains unknown.We note that except for the arguable presence of tills and the reported, but not confirmed, presence of erratics beyond the mountain fronts, we have not been able to establish firm evidence of ice coverage on the intervening plateau surfaces. Rather, many areas display a distinct non-glacial morphology with welldeveloped fluvial valley systems and basins infilled with alluvial deposits. This casts some doubt on the concept of the Huang He ice sheet, although one may argue that, if of considerable age, few glacial traces may have survived degradational processes. Moreover, we conclude that the break in slope between the youthful steep fluvial landscapes of the Huang He and Yangtze rivers and the relict gentle sloping surface of the Tibetan Plateau almost entirely coincides with the outline of the Huang He ice sheet bordering these rivers. This could be used to further question the reality of the Huang He ice sheet or, if indisputable further evidence can be uncovered in the years to come, the coincidence of borders could indicate that the ice sheet was larger but that evidence for this is now flowing down the rivers.Finally, an ambitious TCN and OSL sampling campaign in the Bayan Har Mountains region with our colleagues from the USA (Caffee, Harbor, Li) and China (Zhou, Liu, Ma) will likely shed light on the timing of glacial advances through the dating of end moraines, erratics and till stratigraphies and establish contemporary landscape catchment erosion rates through the analysis of river bank sediment TCN concentrations.

    • Outcome of 333 children with acute myeloid leukaemia relapsing after a first allogeneic haematopoietic stem cell transplantation was analyzed. Four-year probability of overall survival (4y-pOS) was 14%. 4y-pOS for 122 children receiving a second haematopoietic stem cell transplantation was 31% and 3% for those that did not (P = <0·0001). Achievement of a subsequent remission impacted survival (P = <0·0001). For patients receiving a second transplant survival with or without achieving a subsequent remission was comparable. Graft source (bone marrow vs. peripheral blood stem cells, P = 0·046) and donor choice (matched family vs. matched unrelated donor, P = 0·029) positively impacted survival after relapse. Disease recurrence and non-relapse mortality at four years reached 45% and 22%.

  • Cancer incidence in a petrochemical industry area in Sweden

    • Emissions from petrochemical industries may contain suspected or established carcinogens. As increased incidence of cancer in residential areas close to petrochemical industries has been reported in the literature, we conducted a study of cancer incidence in Stenungsund, Sweden, where petrochemical industries were established in the mid 1960s. A number of cancer cases in the central parts of Stenungsund were collected from the regional cancer registry for each year between 1974 and 2005. In addition to the total number of cases, the numbers of leukemia, lymphoma, liver cancer, lung cancer, and brain cancer were also collected. Expected numbers for each year were calculated based on age- and sex-specific incidence rates in reference areas. Levels of carcinogenic volatile hydrocarbons (VOC) were estimated from measurements and emission data. A dispersion model was used to classify Stenungsund into a "low" and "high" ethylene level area. Standardized Incidence Ratio (SIR) for all cancer for the entire period was 1.02 (95% CI 0.97-1.08). The occurrence of leukemia, lymphoma, and cancer in the central nervous system was slightly lower than expected for the entire period. SIR for lung cancer was 1.37 (95% CI 1.10-1.69), and SIR for liver cancer was 1.50 (0.82-2.53). VOC levels were low. Taking estimated exposure and demographic factors into account, our assessment is that occurrence of cancer was not affected by industrial emissions in any of the studied sites.

    • Ecosystem services have become a mainstream concept for the expression of values assigned by people to various functions of ecosystems. Even though the introduction of the concept has initiated a vast amount of research, progress in using this knowledge for sustainable resource use remains insufficient. We see a need to broaden the scope of research to answer three key questions that we believe will improve incorporation of ecosystem service research into decision-making for the sustainable use of natural resources to improve human well-being: (i) how are ecosystem services co-produced by social–ecological systems, (ii) who benefits from the provision of ecosystem services, and (iii) what are the best practices for the governance of ecosystem services? Here, we present these key questions, the rationale behind them, and their related scientific challenges in a globally coordinated research programme aimed towards improving sustainable ecosystem management. These questions will frame the activities of ecoSERVICES, formerly a DIVERSITAS project and now a project of Future Earth, in its role as a platform to foster global coordination of multidisciplinary sustainability science through the lens of ecosystem services.

    • Ecosystem management that attempts to maximize the production of one ecosystem service often results in substantial declines in the provision of other ecosystem services. For this reason, recent studies have called for increased attention to development of a theoretical understanding behind the relationships among ecosystem services. Here, we review the literature on ecosystem services and propose a typology of relationships between ecosystem services based on the role of drivers and the interactions between services. We use this typology to develop three propositions to help drive ecological science towards a better understanding of the relationships among multiple ecosystem services. Research which aims to understand the relationships among multiple ecosystem services and the mechanisms behind these relationships will improve our ability to sustainably manage landscapes to provide multiple ecosystem services.

  • Effects of hyperglycemia on interdigestive gastrointestinal motility in humans

    • BACKGROUND: Gastrointestinal motility disorders are common in patients with diabetes mellitus. Recent studies indicate that hyperglycemia can affect gastric emptying and gastric motility in healthy subjects and diabetics. METHODS: The effect of acute hyperglycemia on gastrointestinal motility was studied with a manometric technique in healthy subjects. Seven individuals, four men and three women, 23-34 years old, were studied on 2 different days. On 1 of the days a 5-h registration was performed after an overnight fast. On another day and after an initial basal period, acute steady-state hyperglycemia was induced by intravenous glucose infusion for 90 min. Motility variables were evaluated in four segments: in the gastric antrum, the proximal duodenum, the distal duodenum, and the proximal jejunum. RESULTS: Fasting migrating motor complex rhythm including migration of phase III prevailed during hyperglycemia. Compared with euglycemia, the motility index in phase II was lower during hyperglycemia in all segments studied. In the antrum the difference was 62% (p < 0.01); in the proximal duodenum, 37% (p < 0.01); in the distal duodenum, 44% (p < 0.05); and in the jejunum, 58% (p < 0.01). During hyperglycemia the prevalence of propagated contractions in phase II was significantly lower than during euglycemia both in the antrum and the proximal duodenum. In the last part of phase III in proximal duodenum most individual contractions were propagated in orad direction compared with early phase III, and this difference persisted during hyperglycemia. The number of long clusters was significantly increased during hyperglycemia as compared with euglycemia: 2.0 +/- 0.6 per hour versus 0.4 +/- 0.14 (p < 0.01). In late phase II plasma levels of motilin and pancreatic polypeptide were significantly decreased during hyperglycemia. CONCLUSION: Hyperglycemia not only reduces the motility in the stomach but also inhibits motility in both the duodenum and the jejunum. The results show that acute hyperglycemia has an important impact on small-intestinal motility.

    • OBJECT: The dyskinetic subtype of cerebral palsy is difficult to manage, and there is no established gold standard for treatment. External rotation of the shoulder(s) is often managed nonsurgically using injections of botulinum toxin A into the external rotator muscles. This article reports a new surgical technique for managing external rotation when botulinum toxin A treatment is not sufficient or possible.METHODS: Six patients with dyskinetic cerebral palsy underwent denervation of the infraspinatus muscle and release of the posterior part of the deltoid muscle. Postoperative questionnaires were given to the patients/caregivers, and video recordings were made both pre- and postoperatively. Preoperative and postoperative Assisting Hand Assessment was possible in only 1 case.RESULTS: Five patients were very satisfied with their outcome. Four patients' video recordings showed improvement in their condition. One patient developed postoperative complications.CONCLUSIONS: The results indicate that denervation of the infraspinatus muscle and posterior deltoid release can be an option for patients with dyskinetic cerebral palsy to manage external rotation of the shoulder when other treatment alternatives are insufficient.

    • This paper addresses women’s engagement across ethnic boundaries to promote reconciliation and women’s rights in the divided city of Mitrovica, Kosovo. It examines the positive assumptions often made in international peace-building projects about the ‘natural’ effectiveness of women-based reconciliation work. The data are drawn from recent fieldwork on local notions of danger and safety in relation to place and gender in this polarized city. The paper takes its point of departure in local NGO-work and highlights two interrelated dynamics essential to understanding women’s reconciliation work: One is between gender and ethno-political space in the context of the specific community in which moral is crucial for safety; the other, between the local gender order and internationally formulated aims to engage women. These dynamics, the study finds, pose obstacles to women-based reconciliation work in Mitrovica. A conclusion is that, in Mitrovica at least, women’s role and effectiveness are related to the ways in which the local politics of gender and security are played out in an ethnically polarized setting.

    • The most pronounced effects of FSH signaling are potentially displayed in follicle fluid (FF), which acts as a reservoir for FSH induced granulosa cell (GC) secreted hormones. This study investigates the effects of two common polymorphisms of FSHR, FSHR 307 (rs6165) and FSHR 680 (rs6166), by evaluating the hormone and gene expression profiles of human small antral follicles collected under physiological conditions in connection with fertility preservation. In total 69 women at various time during the menstrual cycle was included in this study. The intrafollicular hormone content of 179 FF and the gene expression levels of 85 GC samples were correlated to the genotype of both FSHR polymorphisms. The following parameters were evaluated: follicle diameter, levels of Anti-Müllerian Hormone (AMH), progesterone, oestradiol, testosterone and androstenedione, and gene expression levels of FSHR, luteinizing hormone receptor (LHR), androgen receptor (AR), aromatase cytochrome p450 (CYP19A1,) AMH and AMH receptor II (AMHR2). There was 100 % concordance between the FSHR 307 and the FSHR 680 genotypes: A/A (p.307Thr/Thr and p.680Asn/Asn), A/G (p.307Thr/Ala and p.680Asn/Ser), and G/G (p.307Ala/Ala and p.680Ser/Ser). Considering all follicles, compared to the other genotypes the G/G genotype was associated with significantly elevated gene expression levels for LHR, while AMHR2 gene expression levels were significantly reduced. In follicles 3-6 mm in diameter LHR gene expression was significantly increased, whereas AMH gene expression was significantly reduced for the G/G genotype. In follicles>6 mm, oestradiol and CYP19A1 gene expression levels were significantly higher for the G/G genotype. In conclusion, significant changes were observed between the FSHR 307/680 polymorphisms in human small antral follicles collected under physiological FSH conditions.

    • The paper describes a new engineering Master's program called MMII (manufacturing management and industrial informatics) that is co-located at universities in Sweden, Spain and the United Kingdom. One reason for developing the program was that the changing manufacturing landscape due to globalisation, increasing complexity of manufacturing systems itself and an increased need to integrate manufacturing systems with corporate information systems forces educators to find solutions that provide industry with engineers who have the right skills. Apart from "hard" skills related to the above-mentioned issues, industry increasingly also requires engineers to have well-developed "soft" skills such as an ability to work in an international environment and willingness to work abroad. A program given at only one location would not provide a truly European dimension and besides, it would draw heavily upon the teaching resources; hence the decision to seek international partners with complementing competences and resources; these were found at universities in Skövde, Valencia and Loughborough. In Loughborough, students read capita selecta from CAE (computer aided engineering) during one semester. In Valencia, they spend a project-based semester on international industrial management. In Skövde, they read virtual manufacturing during one semester and carry out their degree project during the final semester.

    • Direct neuronal reprogramming of a somatic cell into therapeutic neurons, without a transient pluripotent state, provides new promise for the large number of individuals afflicted by neurodegenerative diseases or brain injury. This approach could be potentially applied directly in the brain by targeting resident cells as a source of new neurons. Direct neuronal conversion of resident glial cells is advantageous since they are ubiquitously distributed brain cells able to self-renew and replenish their number, making them ideal candidates for endogenous repair.In this thesis, human glia-to-neuron direct conversion and engineered viral vectors are explored using pre-clinical in vitro and ex vivo models. The first part of the thesis (Paper I, II, III) shows the development and improvement of a hESC-based system of for virus-mediated direct reprogramming of human glial progenitor cells into both induced dopaminergic neurons (iDANs) and GABAergic interneurons. An extensive functional and molecular analysis confirms the maturation and properties of these neurons and we show the expression of subtype-specific markers in vitro. We have also increased our understanding on the reprogramming process by employing single-nucleus RNA sequencing for assessment of cell composition before and after conversion into iDANs. The last part of the thesis (Paper IV) is aimed at investigating a better targeting strategy for future in vivo conversion applications and demonstrates that selective infection of human glia transplanted in the rodent brain can be achieved by engineering the AAV capsid surface.Collectively, the work thereby presented contributes to better understanding of the potential of this technique and further advanced the development of therapeutic reprogramming strategies as a future avenue for brain repair.

Investigation of the essential spectrum of the corresponding self-adjoint operator is continued but; now without assuming that the quasi-regularity conditions are satisfied. New conditions that guarantee that the operator is semi-bounded from below are derived. It is proven that the essential spectrum of any self-adjoint operator associated with the matrix differential operator is given by the range range((mp - beta(2))/px(2)) in the case where the quasi-regularity conditions are not satisfied.

    • Denna rapport är en processutvärdering av projektet Trygg Fotboll i Göteborg (TFG) som genomförts med hjälp av analys av dokument samt intervjuer med deltagande aktörer. I TFG samverkar polis, kommun och fotbollsklubb med syfte att minska otryggheten, brottsligheten och ordningsstörningarna med hjälp av förbättrad samverkan samt förebyggande arbete. I projektets planering låg fokus framför allt på att utveckla en bristfällig samverkan samt att ta fram nya, och mindre repressiva, metoder att hantera risksupportrar – vilket kan betecknas social prevention. Utvärderingen visar att projektet verkar ha medfört förbättrade rutiner för samverkan, men att det inte gjorts så mycket som planerat avseende social prevention, och att istället situationell prevention kommit i fokus. En förutsättning för att få arbetet att fungera väl är tydliga strukturer och processtöd. Ett hinder som identifierats i implementeringen är tröghet i organisationerna, vilket gör att saker tar tid och genererar frustration. Dessutom hade delar av verksamheten troligen mått bra av viss extern kommunikation eller marknadsföring, något som samtidigt är vanskligt med en verksamhet som är under uppförande. Sammantaget är utvärderingens slutsats att metoden verkar vara ett rimligt sätt att arbeta kring problem med fotbollsmatcher, och att det fokus mot situationell prevention som verkar ha växt fram under projektets gång kanske också initialt är mer passande för denna typ av samverkan än mer komplicerade insatser som syftar till att förändra risksupportrars brottsbenägenhet.

    • Ongoing changes in disturbance regimes are predicted to cause acute changes in ecosystem structure and function in the coming decades, but many aspects of these predictions are uncertain. A key challenge is to improve the predictability of postdisturbance biogeochemical trajectories at the ecosystem level. Ecosystem ecologists and paleoecologists have generated complementary data sets about disturbance (type, severity, frequency) and ecosystem response (net primary productivity, nutrient cycling) spanning decadal to millennial timescales. Here, we take the first steps toward a full integration of these data sets by reviewing how disturbances are reconstructed using dendrochronological and sedimentary archives and by summarizing the conceptual frameworks for carbon, nitrogen, and hydrologic responses to disturbances. Key research priorities include further development of paleoecological techniques that reconstruct both disturbances and terrestrial ecosystem dynamics. In addition, mechanistic detail from disturbance experiments, long-term observations, and chronosequences can help increase the understanding of ecosystem resilience.

    • This paper gives an overview of an ongoing project using real-time 3D visualization to display nautical charts in a way used by 3D computer games. By displaying the map in an egocentric perspective the need to make cognitively demanding mental rotations are suggested to be removed, leading to faster decision-making and less errors. Experimental results support this hypothesis. Practical tests with limited success have been performed this year.

    • Background: Mast cells are tissue-resident inflammatory cells defined by their high granularity and surface expression of the high-affinity IgE receptor, Fc + RI, and CD117/KIT, the receptor for stem cell factor (SCF). There is a considerable heterogeneity among mast cells, both phenotypically and functionally. Human mast cells are generally divided into two main subtypes based on their protease content; the mucosa-associated MCT (tryptase positive and chymase negative mast cell) and the connective tissue associated-residing MCTC (tryptase and chymase positive mast cell). Human lung mast cells exhibit heterogeneity in terms of cellular size, expression of cell surface receptors, and secreted mediators. However, knowledge about human lung mast cell heterogeneity is restricted to studies using immunohistochemistry or purified mast cells. Whereas the former is limited by the number of cellular markers that can be analyzed simultaneously, the latter suffers from issues related to cell yield.Aim: To develop a protocol that enables isolation of human lung mast cells at high yields for analysis of functional properties and detailed analysis using single-cell based analyses of protein (flow cytometry) or RNA (RNA-sequencing) expression.Methods: Mast cells were isolated from human lung tissue by a sequential combination of washing, enzymatic digestion, mechanical disruption, and density centrifugation using Percoll (WEMP). As a comparison, we also isolated mast cells using a conventional enzyme-based protocol. The isolated cells were analyzed by flow cytometry.Results: We observed a significant increase in the yield of total human lung CD45(+) immune cells and an even more pronounced increase in the yield of CD117(+) mast cells with the WEMP protocol in comparison to the conventional protocols. In contrast, the frequency of the rare lymphocyte subset innate lymphoid cells group 2 (ILC2) did not differ between the two methods.Conclusion: The described WEMP protocol results in a significant increase in the yield of human lung mast cells compared to a conventional protocol. Additionally, the WEMP protocol enables simultaneous isolation of different immune cell populations such as lymphocytes, monocytes, and granulocytes while retaining their surface marker expression that can be used for advanced single-cell analyses including multi-color flow cytometry and RNA-sequencing.
