    • Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale1–3. Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained 4–5 driver mutations when combining coding and non-coding genomic elements; however, in around 5% of cases no drivers were identified, suggesting that cancer driver discovery is not yet complete. Chromothripsis, in which many clustered structural variants arise in a single catastrophic event, is frequently an early event in tumour evolution; in acral melanoma, for example, these events precede most somatic point mutations and affect several cancer-associated genes simultaneously. Cancers with abnormal telomere maintenance often originate from tissues with low replicative activity and show several mechanisms of preventing telomere attrition to critical levels. Common and rare germline variants affect patterns of somatic mutation, including point mutations, structural variants and somatic retrotransposition. A collection of papers from the PCAWG Consortium describes non-coding mutations that drive cancer beyond those in the TERT promoter4; identifies new signatures of mutational processes that cause base substitutions, small insertions and deletions and structural variation5,6; analyses timings and patterns of tumour evolution7; describes the diverse transcriptional consequences of somatic mutation on splicing, expression levels, fusion genes and promoter activity8,9; and evaluates a range of more-specialized features of cancer genomes8,10–18.

  • Significant effects of transport on nanoparticles during new particle formation events in the atmosphere of Beijing

    • The mechanisms of new particle formation (NPF) events that occurred under high aerosol loadings ("polluted" NPF) in the atmosphere have been unclear, which has inhibited the precision of particle pollution control. To deepen the understanding of how the "polluted" NPF events occur, a one-month comprehensive measurement was conducted in the atmosphere of Beijing during the summer of 2016. The "clean" NPF events (frequency = 22%) (condensation sink, CS < 0.015 s-1) were found to be caused by local nucleation and growth. The "polluted" NPF events (frequency = 28%) (CS > 0.015 s-1) were influenced by both local nucleation-growth and regional transport, and the contributions from the two factors to 6-25 nm particle number concentration were 60% and 40%, respectively. This study emphasized the importance of the transport for nanoparticles in relatively polluted atmospheres, and for that the regional joint particle pollution control would be an essential policy.   • Low-frequency Whistler Waves Modulate Electrons and Generate Higher-frequency Whistler Waves in the Solar Wind

    • The role of whistler-mode waves in the solar wind and the relationship between their electromagnetic fields and charged particles is a fundamental question in space physics. Using high-temporal-resolution electromagnetic field and plasma data from the Magnetospheric MultiScale spacecraft, we report observations of low-frequency whistler waves and associated electromagnetic fields and particle behavior in the Earth's foreshock. The frequency of these whistler waves is close to half the lower-hybrid frequency (similar to 2 Hz), with their wavelength close to the ion gyroradius. The electron bulk flows are strongly modulated by these waves, with a modulation amplitude comparable to the solar wind velocity. At such a spatial scale, the electron flows are forcibly separated from the ion flows by the waves, resulting in strong electric currents and anisotropic ion distributions. Furthermore, we find that the low-frequency whistler wave propagates obliquely to the background magnetic field ( B (0)), and results in spatially periodic magnetic gradients in the direction parallel to B (0). Under such conditions, large pitch-angle electrons are trapped in wave magnetic valleys by the magnetic mirror force, and may provide free perpendicular electron energy to excite higher-frequency whistler waves. This study offers important clues and new insights into wave-particle interactions, wave generation, and microscale energy conversion processes in the solar wind.

  • Broadband Multi-wavelength Properties of M87 during the 2017 Event Horizon Telescope Campaign

    • In 2017, the Event Horizon Telescope (EHT) Collaboration succeeded in capturing the first direct image of the center of the M87 galaxy. The asymmetric ring morphology and size are consistent with theoretical expectations for a weakly accreting supermassive black hole of mass ∼6.5 × 109 M o˙. The EHTC also partnered with several international facilities in space and on the ground, to arrange an extensive, quasi-simultaneous multi-wavelength campaign. This Letter presents the results and analysis of this campaign, as well as the multi-wavelength data as a legacy data repository. We captured M87 in a historically low state, and the core flux dominates over HST-1 at high energies, making it possible to combine core flux constraints with the more spatially precise very long baseline interferometry data. We present the most complete simultaneous multi-wavelength spectrum of the active nucleus to date, and discuss the complexity and caveats of combining data from different spatial scales into one broadband spectrum. We apply two heuristic, isotropic leptonic single-zone models to provide insight into the basic source properties, but conclude that a structured jet is necessary to explain M87's spectrum. We can exclude that the simultaneous γ-ray emission is produced via inverse Compton emission in the same region producing the EHT mm-band emission, and further conclude that the γ-rays can only be produced in the inner jets (inward of HST-1) if there are strongly particle-dominated regions. Direct synchrotron emission from accelerated protons and secondaries cannot yet be excluded.

  • An optimal charging scheduling model and algorithm for electric buses

    • Electrification poses a promising low-carbon or even zero-carbon transportation solution, serving as a strategic approach to reducing carbon emissions and promoting carbon neutrality in the transportation sector. Along the transportation electrification pathway, the goal of carbon neutrality can be further accelerated with an increasing amount of electricity being generated from renewable energies. The past decade observed the rapid development of battery technologies and deployment of electricity infrastructure worldwide, fostering transportation electrification to expand from railways to light and then heavy vehicles on roadways. In China, a massive number of electric buses have been employed and operated in dozens of metropolises. An important daily operations issue with these urban electric buses is how to coordinate their charging activities in a cost-effective manner, considering various physical, financial, institutional, and managerial constraints. This paper addresses a general charging scheduling problem for an electric bus fleet operated across multiple bus lines and charging depots and terminals, aiming at finding an optimal set of charging location and time decisions given the available charging windows. The charging windows for each bus are predetermined in terms of its layovers at depots and terminals and each of them is discretized into a number of charging slots with the same time duration. A mixed linear integer programming model with binary charging slot choice and continuous state-of-charge (SOC) variables is constructed for minimizing the total charging cost of the bus fleet subject to individual electricity consumption rates, electricity charging rates, time-based charging windows, battery SOC bounds, time-of-use (TOU) charging tariffs, and station-specific electricity load capacities. A Lagrangian relaxation framework is employed to decouple the joint charging schedule of a bus fleet into a number of independent single-bus charging schedules, which can be efficiently addressed by a bi-criterion dynamic programming algorithm. A real-world regional electric bus fleet of 122 buses in Shanghai, China is selected for validating the effectiveness and practicability of the proposed charging scheduling model and algorithm. The optimization results numerically reveal the impacts of TOU tariffs, station load capacities, charging infrastructure configurations, and battery capacities on the bus system performance as well as individual recharging behaviors, and justify the superior solution efficiency of our algorithm against a state-of-the-art commercial solver.

  • Optimizing transformation-induced plasticity in CoCrNi alloys by combined grain refinement and chemical tuning

    • Grain refinement increases yield strength, but usually at the cost of ductility reduction. In the present work, we explored the strategy of grain refinement and composition tuning in CoCrNi alloys to optimize mechanical properties. Grain refinement was found to strongly suppress deformation-induced martensitic transformation in metastable CoCrNi alloys, which was utilized to modulate the transformation-induced plasticity (TRIP) effect in combination with composition tuning guided by theoretical calculations. We demonstrated in a non-equiatomic CoCrNi TRIP medium-entropy alloy (MEA) that our approach resulted in an excellent combination of strength and ductility. The proposed strategy is expected to be useful in exploring superior mechanical properties of MEAs and high-entropy alloys in varying systems.

    • Elevated ground-level ozone (O-3) concentrations decrease photosynthetic biochemistry more than stomatal conductance (g(s)), leading to an overall reduction in leaf-scale water use efficiency (WUE). Global warming is expected to lead to more severe and frequent droughts resulting in stomatal closure, increased WUE, and potentially in reduced plant O-3 uptake and damage. It is currently unclear how the physiological responses to O-3 and water limitation interact to affect overall leaf WUE and how these WUE responses might affect ecosystem productivity. In this study, we used open top chambers to expose O-3-sensitive poplar saplings to elevated O-3 (E-O-3) and limited water availability to explore the individual and interactive effects of these stressors on WUE. We found that leaf-scale intrinsic water-use efficiency based on gas exchange measurements (iWUE(ge)) decreased under E-O-3 due to significantly reduced photosynthetic capacity, mesophyll conductance and apparent quantum yield, while g(s) was not affected by the treatment. Leaf-scale iWUE(ge) and intrinsic WUE based on isotope measurements (iWUE(iso)) increased in the plants receiving less water due to higher photosynthetic capacities and lower transpiration rates indicated by delta O-18 measurements. The overall plant growth (total number of leaves, height, stem diameter and projected area of individual leaf) was significantly reduced under low water supply. Elevated O-3 resulted in significant leaf senescence, but had no other significant main effect on morphological variables. Reduced water availability prevented O-3 -induced decreases in leaf mass per area and increases in leaf loss. No other significant O-3-water availability interactions were detected in the measured physiological or morphological variables. Our results thus suggest that drought conditions will not prevent O-3 damage to photosynthetic biochemistry in poplar and that high O-3 concentrations will decrease leaf-scale iWUE(ge) regardless of future changes in plant water availability.

  • Gamma-ray observations of MAXI J1820+070 during the 2018 outburst

    • MAXIJ1820+070 is a low-mass X-ray binary with a black hole (BH) as a compact object. This binary underwent an exceptionally bright X-ray outburst from 2018 March to October, showing evidence of a non-thermal particle population through its radio emission during this whole period. The combined results of 59.5 h of observations of the MAXI J1820+070 outburst with the H.E.S.S., MAGIC and VERITAS experiments at energies above 200 GeV are presented, together with Fermi-LAT data between 0.1 and 500 GeV, and multiwavelength observations from radio to X-rays. Gamma-ray emission is not detected from MAXI J1820+070, but the obtained upper limits and the multiwavelength data allow us to put meaningful constraints on the source properties under reasonable assumptions regarding the non-thermal particle population and the jet synchrotron spectrum. In particular, it is possible to show that, if a high-energy (HE) gamma-ray emitting region is present during the hard state of the source, its predicted flux should be at most a factor of 20 below the obtained Fermi-LAT upper limits, and closer to them for magnetic fields significantly below equipartition. During the state transitions, under the plausible assumption that electrons are accelerated up to similar to 500 GeV, the multiwavelength data and the gamma-ray upper limits lead consistently to the conclusion that a potential HE and very-HE gamma-ray emitting region should be located at a distance from the BH ranging between 10(11) and 10(13) cm. Similar outbursts from low-mass X-ray binaries might be detectable in the near future with upcoming instruments such as CTA.

  • Observation of the Gamma-Ray Binary HESS J0632+057 with the HESS, MAGIC, and VERITAS Telescopes

    • The results of gamma-ray observations of the binary system HESS J0632 + 057 collected during 450 hr over 15 yr, between 2004 and 2019, are presented. Data taken with the atmospheric Cherenkov telescopes H.E.S.S., MAGIC, and VERITAS at energies above 350 GeV were used together with observations at X-ray energies obtained with Swift-XRT, Chandra, XMM-Newton, NuSTAR, and Suzaku. Some of these observations were accompanied by measurements of the H alpha emission line. A significant detection of the modulation of the very high-energy gamma-ray fluxes with a period of 316.7 +/- 4.4 days is reported, consistent with the period of 317.3 +/- 0.7 days obtained with a refined analysis of X-ray data. The analysis of data from four orbital cycles with dense observational coverage reveals short-timescale variability, with flux-decay timescales of less than 20 days at very high energies. Flux variations observed over a timescale of several years indicate orbit-to-orbit variability. The analysis confirms the previously reported correlation of X-ray and gamma-ray emission from the system at very high significance, but cannot find any correlation of optical H alpha parameters with fluxes at X-ray or gamma-ray energies in simultaneous observations. The key finding is that the emission of HESS J0632 + 057 in the X-ray and gamma-ray energy bands is highly variable on different timescales. The ratio of gamma-ray to X-ray flux shows the equality or even dominance of the gamma-ray energy range. This wealth of new data is interpreted taking into account the insufficient knowledge of the ephemeris of the system, and discussed in the context of results reported on other gamma-ray binary systems.

    • Immunoglobulin A (IgA) mediates mucosal responses to food antigens and the intestinal microbiome and is involved in susceptibility to mucosal pathogens, celiac disease, inflammatory bowel disease, and IgA nephropathy. We performed a genome-wide association study of serum IgA levels in 41,263 individuals of diverse ancestries and identified 20 genome-wide significant loci, including 9 known and 11 novel loci. Co-localization analyses with expression QTLs prioritized candidate genes for 14 of 20 significant loci. Most loci encoded genes that produced immune defects and IgA abnormalities when genetically manipulated in mice. We also observed positive genetic correlations of serum IgA levels with IgA nephropathy, type 2 diabetes, and body mass index, and negative correlations with celiac disease, inflammatory bowel disease, and several infections. Mendelian randomization supported elevated serum IgA as a causal factor in IgA nephropathy. African ancestry was consistently associated with higher serum IgA levels and greater frequency of IgA-increasing alleles compared to other ancestries. Our findings provide novel insights into the genetic regulation of IgA levels and its potential role in human disease.

    • Hydrides based on magnesium and intermetallic compounds provide a viable solution to the challenge of energy storage from renewable sources, thanks to their ability to absorb and desorb hydrogen in a reversible way with a proper tuning of pressure and temperature conditions. Therefore, they are expected to play an important role in the clean energy transition and in the deployment of hydrogen as an efficient energy vector. This review, by experts of Task 40 'Energy Storage and Conversion based on Hydrogen' of the Hydrogen Technology Collaboration Programme of the International Energy Agency, reports on the latest activities of the working group 'Magnesium- and Intermetallic alloys-based Hydrides for Energy Storage'. The following topics are covered by the review: multiscale modelling of hydrides and hydrogen sorption mechanisms; synthesis and processing techniques; catalysts for hydrogen sorption in Mg; Mg-based nanostructures and new compounds; hydrides based on intermetallic TiFe alloys, high entropy alloys, Laves phases, and Pd-containing alloys. Finally, an outlook is presented on current worldwide investments and future research directions for hydrogen-based energy storage.

    • Nickel (Ni)-based superalloys for high-temperature applications are often designed to form a continuous and slow-growing oxide scale by adding Al and Cr and other beneficial elements. In the present work, the critical Al concentration in Ni–Al alloys needed to establish an α-Al2O3 scale in contrast to internal oxide formation is predicted as a function of temperature by means of the CALPHAD approach coupled with models in the literature, which account for the thermodynamics and kinetics of oxidation. The present thermodynamic remodeling of the Ni–O system results in a better agreement with experimental data of oxygen solubility in Ni at high temperatures. The oxygen solubility is combined with kinetic parameters to determine oxygen permeability in Ni, and the critical Al concentration needed to establish an α-Al2O3 scale at a given exposure temperature. Good agreement is found with available experimental data for both oxygen permeability and critical Al concentration, indicating the capacity of the CALPHAD approach to tailor oxidation resistance for materials of interest using thermodynamic and kinetic knowledge.

  • Local vibrational modes of Si vacancy spin qubits in SiC

    • Silicon carbide is a very promising platform for quantum applications because of the extraordinary spin and optical properties of point defects in this technologically friendly material. These properties are strongly influenced by crystal vibrations, but the exact relationship between them and the behavior of spin qubits is not fully investigated. We uncover the local vibrational modes of the Si vacancy spin qubits in as-grown 4H-SiC. We apply microwave-assisted spectroscopy to isolate the contribution from one particular type of defects, the so-called V2 center, and observe the zero-phonon line together with seven equally separated phonon replicas. Furthermore, we present first-principles calculations of the photoluminescence line shape, which are in excellent agreement with our experimental data. To boost up the calculation accuracy and decrease the computation time, we extract the force constants using machine-learning algorithms. This allows us to identify the dominant modes in the lattice vibrations coupled to an excited electron during optical emission in the Si vacancy. A resonance phonon energy of 36 meV and a Debye-Waller factor of about 6% are obtained. We establish experimentally that the activation energy of the optically induced spin polarization is given by the local vibrational energy. Our findings give insight into the coupling of electronic states to vibrational modes in SiC spin qubits, which is essential to predict their spin, optical, mechanical, and thermal properties. The approach described can be applied to a large variety of spin defects with spectrally overlapped contributions in SiC as well as in other three-and two-dimensional materials.

  • Effect of elevated ozone, nitrogen availability and mesophyll conductance on the temperature responses of leaf photosynthetic parameters in poplar

    • Although ozone (O3) concentration and nitrogen (N) availability are well known to affect plant physiology, their impacts on the photosynthetic temperature response are poorly understood. We addressed this knowledge gap by exposing seedlings of hybrid poplar clone '107' (Populous euramericana cv. '74/76') to elevated O3 (E-O3) and N availability variation in a factorial experiment. E-O3 decreased light-saturated net photosynthesis (Asat), mesophyll conductance (gm) and apparent maximum rate of carboxylation (Vcmax, based on intercellular CO2 concentration) but not actual Vcmax (based on chloroplast CO2 concentration) and increased respiration in light (Rd) at 25 °C. Nitrogen fertilization increased Asat, gm, Vcmax and the maximum rate of electron transport (Jmax) and reduced Rd at 25 °C and the activation energy of actual Vcmax. No E-O3 or E-O3 x N interaction effects on the temperature response parameters were detected, simplifying the inclusion of O3 impacts on photosynthesis in vegetation models. gm peaked at 30 °C, apparent Vcmax and Jmax at 32-33 °C, while the optimum temperatures of actual Vcmax and Jmax exceeded the measured temperature range (15-35 °C). Ignoring gm would, thus, have resulted in mistakenly attributing the decrease in Asat at high temperatures to reduced biochemical capacity rather than to greater diffusion limitation. © The Author(s) 2020.   • Stomatal response drives between-species difference in predicted leaf water-use efficiency under elevated ozone

    • Ozone-induced changes in the relationship between photosynthesis (An) and stomatal conductance (gs) vary among species, leading to inconsistent water use efficiency (WUE) responses to elevated ozone (O3). Thus, few vegetation models can accurately simulate the effects of O3 on WUE. Here, we conducted an experiment exposing two differently O3-sensitive species (Cotinus coggygria and Magnolia denudata) to five O3 concentrations and investigated the impact of O3 exposure on predicted WUE using a coupled An-gs model. We found that increases in stomatal O3 uptake caused linear reductions in the maximum rates of Rubisco carboxylation (Vcmax) and electron transport (Jmax) in both species. In addition, a negative linear correlation between O3-induced changes in the minimal gs of the stomatal model (g0) derived from the theory of optimal stomatal behavior and light-saturated photosynthesis was found in the O3-sensitive M. denudata. When the O3 dose-based responses of Vcmax and Jmax were included in a coupled An-gs model, simulated An under elevated O3 were in good agreement with observations in both species. For M. denudata, incorporating the O3 response of g0 into the coupled model further improved the accuracy of the simulated gs and WUE. In conclusion, the modified Vcmax, Jmax and g0 method presented here provides a foundation for improving the prediction for O3-induced changes in An, gs and WUE. © 2020 Elsevier Ltd Stomatal response to elevated O3 determined predicted leaf WUE. © 2020 Elsevier Ltd
