    • We have investigated the electronic structure of one and two-dimensional C60 polymers with regard to both their low-lying excitations and C 1s excitation spectra by means of electron energy-loss spectroscopy in transmission. We compare the results with those for pristine C60. In general, the spectra for the polymers resemble those for pristine C60 but show a broadening due to both the lowering of the symmetry and the increased intermolecular overlap. This is also reflected by a reduction of the optical gap in comparison with pristine C60.

    • A simulation model, ORWARE (ORganic WAste REsearch), for the handling of organic waste in urban areas has been constructed. The model provides a comprehensive view of the environmental effects, plant nutrient utilisation and energy turnover for this large and complex system. The ORWARE model consists of several sub-models; sewage plant, incineration, landfill, compost, anaerobic digestion, truck transport, transport by sewers, residue transport and spreading of residues on arable land. The model is intended for simulating different scenarios, and the results are: emissions to air and water, energy turnover and the amount of residues returned to arable land. All results are presented, both as the gross figure for the entire system and figures for each process. Throughout the model all physical flows are described by the same variable vector, consisting of 43 substances. This extensive vector facilitates a thorough analysis of the results, but involves some difficulties in acquiring relevant data. In this paper, the model is described. Results from a hypothetical case study are presented in a companion paper.

  • Human antibody reactivity against the lower matrix protein (pp65) produced by cytomegalovirus

    • The lower matrix protein (pp65) is a major product of many laboratory strains of cytomegalovirus (CMV). It is thus an integral part of many CMV serological assays based on native antigen. Recombinant fragments of pp65 have previously been investigated for their usefulness in more-defined assays. The latter antigens have, however, failed to develop a positive response with serum samples derived from a substantial number of infected individuals. Here we show that the human humoral immune response to CMV pp65 is highly diverse and recognizes at least seven distinct but in some cases partly overlapping epitopes. Most of these epitopes could not be mimicked by any of the investigated recombinant or synthetic antigens. Furthermore, when we investigated the ability of human CMV-seropositive serum samples to block the reactivity of pp65 specific antibodies recognizing five different epitopes within pp65, it was evident that several sera did not contain significant levels of antibodies against any of these or overlapping structures. It was thus concluded that the antibody response against CMV pp65 is weak in some CMV-infected individuals, making this antigen unsuitable for use alone in serological screening systems for CMV infection.

    • The structure and lattice properties of a polymerized C60 sample were investigated by X-ray powder diffraction over the range from liquid-nitrogen to room temperature. The aim of these studies was to establish the effect of low-pressure compression (up to 1.1 GPa) on the phase composition of the sample, to determine the structure of the phases involved, to obtain the variation of the lattice parameters with temperature, to observe eventual phase transformation in polymerized C60, and to elucidate the evolution of the orientational order with varying temperature. Analysis of the room-temperature diffraction patterns indicates that the sample contained at least two major phases, namely rhombohedral and and tetragonal with the polymerization array close to two-dimensional. Our data obtained on samples annealed at 300 C in the air allow us to conclude that polymerized C60 converts to a mixture of the fcc structure plus a new phase, presumably C60O.

    • NADPH: protochlorophyllide (POR) is a vital enzyme in the biosynthesis of chlorophyl where it catalyzes the reduction of Pchlide into Chlide in a light-dependent manner. POR is nucleus-encoded and imported into the plastids where it is found at the inner membranes. Together with its substrate and the co-factor NADPH it forms a ternary complex which is needed for catalytical activity. The anomaly of a decreasing POR level during active chlorophyll synthesis was cleared with the discovery of two different POR proteins, POR-A and POR-B, in barley and Arabidopsis thaliana. During greening, POR-A is negatively regulated by light both at transcriptional and proteolytical levels. In addition, the import of POR-A, but not POR-B, has been suggestedto require Pchlide in order to be translocated into the plastid. In this respect, POR-A differs from other known nucleus-encoded plastid proteins, and as it appears, this requirements represents a novel and exclusive import characteristic. In pea, only one POR gene has been found indicating that the situation for the regulation of POR import and accumulation is far from clear. We here present a characterization of the import conditions of the pea POR, including the potentional role of Pchlide inthe translocation step.

  • High-pressure polymerized phases of C60

    • Data from recent experimental studies of C60 under high pressures are collected and analyzed, concentrating on the polymerized states where covalent intermolecular bonds have been formed through treatment of molecular C60 at high pressures and temperatures at or above room temperature. We give an overview of the observed phase transformations and the structures of metastable polymeric phases, both crystalline and disordered, as analyzed under ambient conditions, and we present and discuss a pressure-temperature diagram showing which synthesis conditions result in which final phases in the pressure range up to 20 GPa (200 kbar) and at temperatures up to 2300 K. The physical properties of the various phases are discussed whenever enough data are available. The pressure-temperature stability limits for C60 and the conditions for the transformation of C60 into graphite, diamond and chaoite-type carbon are derived.

    • A poliovirus-binding inhibition test (PoBI test) was established for the quantitative determination of antibodies to polioviruses and was evaluated in comparison with the conventional neutralization test (NT). The first step of the PoBI test is an incubation of serial dilutions of test samples with inactivated poliovirus followed by the detection of free viral epitopes by a double antibody sandwich enzyme-linked immunosorbent assay with type-specific capture polyclonal antisera and type-specific neutralizing monoclonal indicator antibodies. A comparison of the PoBI test with the conventional NT for antibodies to all three types in 100 human serum samples showed excellent correlations (r > 0.95) over a wide range of antibody concentrations. The PoBI test, not necessitating live virus and tissue culture facilities, could be a simple alternative to the NT, and the principle of the assay is potentially applicable to other microbial systems.

    • We have measured the bulk modulus K for C61D2 up to 1 GPa in the temperature range 175–343 K. For face-centered cubic C61D2 above 290 K, we find an anomalously low value for K below about 0.15 GPa, possibly indicating pressure-induced changes in the structure. The (extrapolated) zero-p bulk modulus K(0) decreases with increasing T from 6.7 GPa at 175 K to 5.2 GPa at 343 K. A comparison with hypothetical expanded f.c.c. C60 with the same lattice constant shows that K(0) values are similar, indicating that the main intermolecular interactions are still between molecular bellies, not the sidegroups.

    • The uterine mucosa in pregnancy, the decidua, allows placenta formation and survival of the fetus despite the fact that it is semiallogeneic. Decidua contains large numbers of lymphocytes, of which CD56+ cells dominate, followed by T cells expressing either alpha beta or gamma delta TCR. We have investigated the developmental relationship between the CD56- and TCR gamma delta-expressing cells in early pregnancy decidua using dual labeling immunoelectron microscopy, immunoflow cytometry, and cell fractionation. Lymphocyte subpopulations were, in addition, analyzed for expression of the cytokine receptor for IL-7 and c-kit and for mRNA expression of recombinase-activating genes 1 and 2. Four different cell populations could be distinguished: CD56+bright, CD56+dim/TCR gamma delta+low, CD56+dim/TCR gamma delta+high, and TCR gamma delta+low. Recombinase-activating genes 1 and 2 were expressed in the CD56+bright cells and to a limited degree in CD56+dim/TCR gamma delta+low cells. c-kit was preferentially expressed on the CD56+bright cells, while IL-7R was preferentially expressed on CD56+dim/TCR gamma delta+low and CD56+dim/TCR gamma delta+high cells. The CD56+dim TCR gamma delta+low and CD56+dim/TCR gamma delta+high cells displayed the characteristic morphology of large granular lymphocytes, while single positive TCR gamma delta+low cells were usually smaller and did not contain cytoplasmic granules. The gamma delta 1 gene segment was almost exclusively used in the TCR. Gamma delta T cells in mitosis were seen. We suggest that human early pregnancy decidua is a transient site for extrathymic maturation and that the progenitors of TCR gamma delta+ cells are bone marrow-derived immature cells expressing the CD56 (neural cell adhesion molecule) homing receptor.

    • The membrane-impermeable thiol blocker 5′5-dithiobis 2- nitrobenzoic acid (DTNB) blocked infectivity of human cytomegalovirus (CMV) although the virus still bound to cells. DTNB-treated CMV regained 65% of its infectivity after incubation with the disulfide bond-reducing agent dithiothreitol. These observations suggest that free thiol groups on CMV are required for infectivity and may participate in disulfide bond formation during virus entry.

    • Literature data for the in-plane electrical resistivity ρ of single-crystal La2-xSrxCuO4 with 0.07 ≤x≤0.34 have been corrected to constant crystal volume V using literature data for the pVT properties and for the resistivity vs pressure p. The original data at constant p show the usual anomalies observed for ceramic high-transition-temperature superconductors, such as linearity of ρ to extremely high T at optimum doping and an upwards curvature (ρ∝T1.5) for overdoped material. After correction, the data for ρ at constant volume are linear in T for overdoped material only. We show that phonons cannot be ruled out as the major carrier scattering agent.
