  • New Dual Inhibitors of Bacterial Topoisomerases with Broad-Spectrum Antibacterial Activity and In Vivo Efficacy against Vancomycin-Intermediate Staphylococcus aureus
  • 2023
  • Ingår i: Journal of Medicinal Chemistry. - : American Chemical Society (ACS). - 0022-2623 .- 1520-4804. ; 66:6, s. 3968-3994
  • Tidskriftsartikel (refereegranskat)abstract
    • A new series of dual low nanomolar benzothiazole inhibitors of bacterial DNA gyrase and topoisomerase IV were developed. The resulting compounds show excellent broad-spectrum antibacterial activities against Gram-positive Enterococcus faecalis, Enterococcus faecium and multidrug resistant (MDR) Staphylococcus aureus strains [best compound minimal inhibitory concentrations (MICs): range, <0.03125–0.25 μg/mL] and against the Gram-negatives Acinetobacter baumannii and Klebsiella pneumoniae (best compound MICs: range, 1–4 μg/mL). Lead compound 7a was identified with favorable solubility and plasma protein binding, good metabolic stability, selectivity for bacterial topoisomerases, and no toxicity issues. The crystal structure of 7a in complex with Pseudomonas aeruginosa GyrB24 revealed its binding mode at the ATP-binding site. Expanded profiling of 7a and 7h showed potent antibacterial activity against over 100 MDR and non-MDR strains of A. baumannii and several other Gram-positive and Gram-negative strains. Ultimately, in vivo efficacy of 7a in a mouse model of vancomycin-intermediate S. aureus thigh infection was also demonstrated.

  • Lysophophatidylcholine 16:0 mediates chronic joint pain associated to rheumatic diseases through acid-sensing ion channel 3
  • 2022
  • Ingår i: Pain. - : International Association for the Study of Pain. - 0304-3959 .- 1872-6623. ; 163:10, s. 1999-2013
  • Tidskriftsartikel (refereegranskat)abstract
    • Rheumatic diseases are often associated to debilitating chronic pain, which remains difficult to treat and requires new therapeutic strategies. We had previously identified lysophosphatidylcholine (LPC) in the synovial fluids from few patients and shown its effect as a positive modulator of acid-sensing ion channel 3 (ASIC3) able to induce acute cutaneous pain in rodents. However, the possible involvement of LPC in chronic joint pain remained completely unknown. Here, we show, from 2 independent cohorts of patients with painful rheumatic diseases, that the synovial fluid levels of LPC are significantly elevated, especially the LPC16:0 species, compared with postmortem control subjects. Moreover, LPC16:0 levels correlated with pain outcomes in a cohort of osteoarthritis patients. However, LPC16:0 do not appear to be the hallmark of a particular joint disease because similar levels are found in the synovial fluids of a second cohort of patients with various rheumatic diseases. The mechanism of action was next explored by developing a pathology-derived rodent model. Intra-articular injections of LPC16:0 is a triggering factor of chronic joint pain in both male and female mice, ultimately leading to persistent pain and anxiety-like behaviors. All these effects are dependent on ASIC3 channels, which drive sufficient peripheral inputs to generate spinal sensitization processes. This study brings evidences from mouse and human supporting a role for LPC16:0 via ASIC3 channels in chronic pain arising from joints, with potential implications for pain management in osteoarthritis and possibly across other rheumatic diseases.

  • Elevated inflammatory proteins in cerebrospinal fluid from patients with painful knee osteoarthritis are associated with reduced symptom severity
  • 2020
  • Ingår i: Journal of Neuroimmunology. - : ELSEVIER. - 0165-5728 .- 1872-8421. ; 349
  • Tidskriftsartikel (refereegranskat)abstract
    • Neuroinflammation and periphery-to-CNS neuroimmune cross-talk in patients with painful knee osteoarthritis (OA) are poorly understood. We utilized proximity extension assay to measure the level of 91 inflammatory proteins in CSF and serum from OA patients and controls. The patients had elevated levels of 48 proteins in CSF indicating neuroinflammation. Ten proteins were correlated between CSF and serum and potentially involved in periphery-to-CNS neuroimmune cross-talk. Seven CSF proteins, all with previously reported neuroprotective effects, were associated with lower pain intensity and milder knee-related symptoms. Our findings indicate that neuroinflammation in OA could be protective and associated with less severe symptoms.

  • Unraveling the neuroimmune interface in chronic pain-the association between cytokines in the cerebrospinal fluid and pain in patients with lumbar disk herniation or degenerative disk disease
  • 2024
  • Ingår i: Pain. - : Wolters Kluwer. - 0304-3959 .- 1872-6623. ; 165:7, s. e65-e79
  • Tidskriftsartikel (refereegranskat)abstract
    • Supplemental Digital Content is Available in the Text.A cross-sectional study finding elevated cerebrospinal fluid protein levels in patients with degenerative disk disease and lumbar disk herniation, suggesting neuroimmune activity. The interaction between neuroimmunity and pain was complex. Recent evidence highlights the importance of the neuroimmune interface, including periphery-to-central nervous system (CNS) neuroimmune crosstalk, in chronic pain. Although neuroinflammatory processes have been implicated in central sensitization for a long time, their potential neuroprotective and analgesic effects remain relatively elusive. We have explored the relationships between cytokine expression and symptom severity, and candidates for periphery-to-CNS crosstalk. Patients with degenerative disk disease (DDD) (nociceptive pain) or patients with lumbar disk herniation (LDH) with radiculopathy (predominantly neuropathic pain) completed questionnaires regarding pain and functional disability, underwent quantitative sensory testing, and provided blood and cerebrospinal fluid (CSF) samples. Proximity extension assay (PEA) was used to measure the levels of 92 inflammatory proteins in the CSF and serum from a total of 160 patients and controls, and CSF/serum albumin quotients was calculated for patients with DDD and patients with LDH. We found signs of neuroimmune activation, in the absence of systemic inflammation. Regarding periphery-to-CNS neuroimmune crosstalk, there were significant associations between several cytokines and albumin quotient, despite the latter being primarily at subclinical levels. The cytokines CCL11, CD5, IL8, and MMP-10 were elevated in the CSF, had positive correlations between CSF and serum levels, and associated in a nonlinear manner with back, but not leg, pain intensity in the LDH, but not the DDD, group. In conclusion, we found evidence for neuroimmune activation in the CNS of both patient groups in the absence of systemic inflammation and signs of a communication between CSF and serum. Complex and disease-specific associations were found between cytokines in CSF and back pain intensity.

    • (1) Background: Overweight and obesity in children have increased worldwide and tend to persist into adolescence and adulthood. The Child Health Service (CHS) has an important role in providing health-promotive interventions, and such interventions are required to be documented in a child’s health record. The aim of the study was to investigate Child Health Care (CHC) nurses’ documentation of weight-related, health-promotive interventions in the Child Health Care Record (CHCR) regarding lifestyle habits in connection to the four-year visit. (2) Methods: A record review of 485 CHCRs using a review template was accomplished. Of the included CHS units, four used electronic records and two used paper records. Chi-square tests and Spearman’s rank-order correlations were used to analyse data. (3) Results: The results showed that CHC nurses document interventions regarding lifestyle habits to a low extent, although children with overweight/obesity seemed to undergo more interventions. There was also a difference between electronic and paper records. (4) Conclusions: The consequences of not documenting the interventions in the CHCR make it difficult to follow up and demonstrate the quality of the CHC nurse’s work. There is a need for more research to gain a deeper understanding of the reasons that the work of CHC nurses is not visible in children’s health records.

  • Going to extremes - a metagenomic journey into the dark matter of life
  • 2021
  • Ingår i: FEMS Microbiology Letters. - : Oxford University Press (OUP). - 1574-6968. ; 368:12
  • Forskningsöversikt (refereegranskat)abstract
    • The Virus-X-Viral Metagenomics for Innovation Value-project was a scientific expedition to explore and exploit uncharted territory of genetic diversity in extreme natural environments such as geothermal hot springs and deep-sea ocean ecosystems. Specifically, the project was set to analyse and exploit viral metagenomes with the ultimate goal of developing new gene products with high innovation value for applications in biotechnology, pharmaceutical, medical, and the life science sectors. Viral gene pool analysis is also essential to obtain fundamental insight into ecosystem dynamics and to investigate how viruses influence the evolution of microbes and multicellular organisms. The Virus-X Consortium, established in 2016, included experts from eight European countries. The unique approach based on high throughput bioinformatics technologies combined with structural and functional studies resulted in the development of a biodiscovery pipeline of significant capacity and scale. The activities within the Virus-X consortium cover the entire range from bioprospecting and methods development in bioinformatics to protein production and characterisation, with the final goal of translating our results into new products for the bioeconomy. The significant impact the consortium made in all of these areas was possible due to the successful cooperation between expert teams that worked together to solve a complex scientific problem using state-of-the-art technologies as well as developing novel tools to explore the virosphere, widely considered as the last great frontier of life.

  • Crystal structure of DNA polymerase I from Thermus phage G20c
  • 2022
  • Ingår i: Acta crystallographica. Section D, Structural biology. - 2059-7983. ; 78:Pt 11, s. 1384-1398
  • Tidskriftsartikel (refereegranskat)abstract
    • This study describes the structure of DNA polymerase I from Thermus phage G20c, termed PolI_G20c. This is the first structure of a DNA polymerase originating from a group of related thermophilic bacteriophages infecting Thermus thermophilus, including phages G20c, TSP4, P74-26, P23-45 and phiFA and the novel phage Tth15-6. Sequence and structural analysis of PolI_G20c revealed a 3'-5' exonuclease domain and a DNA polymerase domain, and activity screening confirmed that both domains were functional. No functional 5'-3' exonuclease domain was present. Structural analysis also revealed a novel specific structure motif, here termed SβαR, that was not previously identified in any polymerase belonging to the DNA polymerases I (or the DNA polymerase A family). The SβαR motif did not show any homology to the sequences or structures of known DNA polymerases. The exception was the sequence conservation of the residues in this motif in putative DNA polymerases encoded in the genomes of a group of thermophilic phages related to Thermus phage G20c. The structure of PolI_G20c was determined with the aid of another structure that was determined in parallel and was used as a model for molecular replacement. This other structure was of a 3'-5' exonuclease termed ExnV1. The cloned and expressed gene encoding ExnV1 was isolated from a thermophilic virus metagenome that was collected from several hot springs in Iceland. The structure of ExnV1, which contains the novel SβαR motif, was first determined to 2.19 Å resolution. With these data at hand, the structure of PolI_G20c was determined to 2.97 Å resolution. The structures of PolI_G20c and ExnV1 are most similar to those of the Klenow fragment of DNA polymerase I (PDB entry 2kzz) from Escherichia coli, DNA polymerase I from Geobacillus stearothermophilus (PDB entry 1knc) and Taq polymerase (PDB entry 1bgx) from Thermus aquaticus.

  • Increased noise levels cause behavioural and distributional changes in Atlantic cod and saithe in a large public aquarium—A case study
  • 2023
  • Ingår i: Aquaculture, Fish and Fisheries. - : John Wiley & Sons. - 2693-8847.
  • Tidskriftsartikel (refereegranskat)abstract
    • Abstract Investigating the effects of underwater noise on aquatic animals is a research field that is receiving rapidly increasing attention. Despite this, surprisingly few studies have addressed the potential impacts of noise in a marine animal husbandry setting. In this regard, the behaviour of fish in public aquariums can be used as an indicator of well-being, and noise is known to cause behavioural changes. This case study investigates the behaviour of cod (Gadus morhua) and saithe (Pollachius virens) in a large public aquarium when exposed to increased noise levels originating from an aquarium renovation carried out by construction divers. Swimming behaviour, group formation and vertical distribution, along with yawning and scratching frequencies of the fish, were analysed from video recordings made before, during and after the exposure to increased noise levels. The same parameters were also analysed to evaluate potential effects of the presence of divers when not making renovation noise, compared to fish behaviour prior to the renovation. There was a slight change in the depth distribution of both species and a decrease in the number of scratches in cod due to the presence of divers that were not making renovation noise. In the presence of construction noises in the tank, however, both cod and saithe showed a wider array of behavioural changes, including increased swimming speed, changes in depth distribution and increased yawning frequencies. The results from this case study demonstrate that an underwater renovation with increased noise levels impacts fish behaviour and suggests that underwater noise should be considered during the management of aquatic environments, including public aquaria.

  • The FASD Eye Code: a complementary diagnostic tool in fetal alcohol spectrum disorders
  • 2021
  • Ingår i: Bmj Open Ophthalmology. - : BMJ. - 2397-3269. ; 6:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective To create an easy-to-use complementary ophthalmological tool to support a fetal alcohol spectrum disorder (FASD) diagnosis. Methods and Analysis The FASD Eye Code was derived from 37 children with FASD evaluated along with 65 healthy age-matched and sex-matched controls. Four ophthalmological categories, which are abnormalities commonly found in children with FASD, were ranked independently on a 4-point scale, with 1 reflecting normal finding and 4 a strong presence of an abnormality: visual acuity, refraction, strabismus/binocular function and ocular structural abnormalities. The tool was validated on 33 children with attention deficit/hyperactivity disorder (ADHD), 57 children born moderate-to-late premature (MLP) and 16 children with Silver-Russell syndrome (SRS). Among children with ADHD none was born prematurely or small for gestational age (SGA) or diagnosed with FASD. Among children born MLP none was SGA, had a diagnosis of ADHD or FASD, or a history of retinopathy of prematurity. Children with SRS were all born SGA, half were born preterm and none had FASD. Children with FASD were re-examined as young adults. Results An FASD Eye Code cut-off total score of >= 10 showed an area under the curve (AUC) of 0.78 (95% CI 0.69 to 0.87), with 94% specificity and 43% sensitivity, in discriminating between FASD and controls, MLP and ADHD, corresponding to a positive likelihood ratio (LR+) of 7.5. Between FASD and controls, an AUC of 0.87 (CI 0.80 to 0.95), with 100% specificity and 43% sensitivity, was found; between FASD and SRS, an AUC of 0.60 (CI 0.45 to 0.75) was found, with 88% specificity and 43% sensitivity. A cut-off score of >= 9 showed a specificity of 98% and a sensitivity of 57% for FASD versus controls, corresponding to an LR+ of 36.9. Scores in individuals with FASD were stable into young adulthood. Conclusion The FASD Eye Code has the potential to serve as a complementary tool and help to strengthen an FASD diagnosis.

  • Visual and ocular findings in children with fetal alcohol spectrum disorders (FASD): validating the FASD Eye Code in a clinical setting
  • 2023
  • Ingår i: Bmj Open Ophthalmology. - : BMJ. - 2397-3269. ; 8:1
  • Tidskriftsartikel (refereegranskat)abstract
    • ObjectiveFetal alcohol spectrum disorders (FASD) is an umbrella term covering a spectrum of medical conditions caused by prenatal alcohol exposure. The FASD Eye Code is a new complementary ophthalmological diagnostic tool created to corroborate the complex FASD diagnosis. The aim of this work was to validate the FASD Eye Code by testing it on a second group of children diagnosed with FASD in a clinical setting. Methods and analysisA clinical study was carried out in a group of 21 children (13 males, 8 females, mean age 13.3 years) investigated for suspected FASD and a healthy sex-matched and age-matched control group (n=21). The participants underwent a detailed ophthalmological examination including visual perception problems (VPPs) assessment. Clinical examination results were compiled, and total scores were calculated according to the FASD Eye Code protocol (range 4-16).ResultsThe median total score in the FASD group was 8. Eight individuals in the FASD group and none of the controls obtained a total score of >= 9 corresponding to 38% sensitivity and 100% specificity with an area under the curve of 0.90. A cut-off total score of >= 8 showed 52% sensitivity and 95% specificity. One individual in the FASD group versus 12 controls had a total score of 4, representing normal findings. No significant difference between the two groups regarding VPPs was seen.ConclusionThe FASD Eye Code can be used as a complementary diagnostic tool for FASD to assist in diagnosis and to detect ophthalmological abnormalities in individuals with suspected FASD.

  • Self-report tool for identification of individuals with coronary atherosclerosis : the Swedish cardiopulmonary bioimage study
  • 2024
  • Ingår i: Journal of the American Heart Association. - : John Wiley & Sons. - 2047-9980. ; 13:14
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND: Coronary atherosclerosis detected by imaging is a marker of elevated cardiovascular risk. However, imaging involves large resources and exposure to radiation. The aim was, therefore, to test whether nonimaging data, specifically data that can be self-reported, could be used to identify individuals with moderate to severe coronary atherosclerosis.METHODS AND RESULTS: We used data from the population-based SCAPIS (Swedish CardioPulmonary BioImage Study) in individuals with coronary computed tomography angiography (n=25 182) and coronary artery calcification score (n=28 701), aged 50 to 64 years without previous ischemic heart disease. We developed a risk prediction tool using variables that could be assessed from home (self-report tool). For comparison, we also developed a tool using variables from laboratory tests, physical examinations, and self-report (clinical tool) and evaluated both models using receiver operating characteristic curve analysis, external validation, and benchmarked against factors in the pooled cohort equation. The self-report tool (n=14 variables) and the clinical tool (n=23 variables) showed high-to-excellent discriminative ability to identify a segment involvement score ≥4 (area under the curve 0.79 and 0.80, respectively) and significantly better than the pooled cohort equation (area under the curve 0.76, P<0.001). The tools showed a larger net benefit in clinical decision-making at relevant threshold probabilities. The self-report tool identified 65% of all individuals with a segment involvement score ≥4 in the top 30% of the highest-risk individuals. Tools developed for coronary artery calcification score ≥100 performed similarly.CONCLUSIONS: We have developed a self-report tool that effectively identifies individuals with moderate to severe coronary atherosclerosis. The self-report tool may serve as prescreening tool toward a cost-effective computed tomography-based screening program for high-risk individuals.

  • Tidskriftsartikel (refereegranskat)abstract
    • Chest pain and acute dyspnoea are frequent causes of emergency medical services activation. The pre-hospital management of these conditions is heterogeneous across different regions of the world and Europe, as a consequence of the variety of emergency medical services and absence of specific practical guidelines. This position paper focuses on the practical aspects of the pre-hospital treatment on board and transfer of patients taken in charge by emergency medical services for chest pain and dyspnoea of suspected cardiac aetiology after the initial assessment and diagnostic work-up. The objective of the paper is to provide guidance, based on evidence, where available, or on experts’ opinions, for all emergency medical services’ health providers involved in the pre-hospital management of acute cardiovascular care.

  • The Role of Legal Theory in Legal Education: A Reflection on Professional and Scholarly Ideals in Nordic Legal Education
  • 2021
  • Ingår i: Nordic Journal on Law and Society. - : Umea University Library. - 2002-7788. ; 4:3, s. 1-31
  • Tidskriftsartikel (refereegranskat)abstract
    • The article addresses the role of legal theory in legal education. Today, a multitude of perspectives is present within legal theory and, as a subject, it is not as distinct as it is sometimes claimed to be. It is evident when considering syllabuses in the Nordic countries that nearly all LLM programmes have ambitions to teach legal theory as embracing a multitude of theoretical and methodological perspectives. This multitude of perspectives promises adaptable content for the subject of legal theory in various legal contexts, and could facilitate a reflection on how knowledge is acquired and why. The challenge addressed in the article is the presence of explicit or implicit ideas from the subject of legal theory as comprising a coherent “legal method” and a specific list of accepted theories. The persistence of such ideas is scrutinised in this article with help of the concepts of “professional knowledge” and “scholarly knowledge”. In order to navigate the complex field of legal theories, students need meta-reflective skills, which means the ability to reflect upon the underlying complexity and multitude of the subject. This is shown by a case study from the Department of Law at the University of Gothenburg, together with examples from curricula and textbooks from legal theory courses across the Nordic countries.

  • Anthropogenic noise disrupts early-life development in a fish with paternal care
  • 2024
  • Ingår i: Science of the Total Environment. - 0048-9697 .- 1879-1026. ; 935
  • Tidskriftsartikel (refereegranskat)abstract
    • Anthropogenic noise is a global pollutant but its potential impacts on early life-stages in fishes are largely unknown. Here, using controlled laboratory experiments, we tested for impacts of continuous or intermittent exposure to low-frequency broadband noise on early life-stages of the common goby (Pomatoschistus microps), a marine fish with exclusive paternal care. Neither continuous nor intermittent noise exposure had an effect on filial cannibalism, showing that males were capable and willing to care for their broods. However, broods reared in continuous noise covered a smaller area and contained fewer eggs than control broods. Moreover, although developmental rate was the same in all treatments, larvae reared by males in continuous noise had, on average, a smaller yolk sac at hatching than those reared in the intermittent noise and control treatments, while larvae body length did not differ. Thus, it appears that the increased consumption of the yolk sac reserve was not utilised for increased growth. This suggests that exposure to noise in early life-stages affects fitness-related traits of surviving offspring, given the crucial importance of the yolk sac reserve during the early life of pelagic larvae. More broadly, our findings highlight the wide-ranging impacts of anthropogenic noise on aquatic wildlife living in an increasingly noisy world.

  • Continuous and intermittent noise has a negative impact on reproductive success and early life survival in marine fish
  • 2022
  • Konferensbidrag (refereegranskat)abstract
    • Anthropogenic underwater noise is a global pollutant of increasing concern and its effect on marine organisms is largely unknown. Importantly, direct assessments of fitness consequences are lacking especially in fish. The effect of noise pattern with continuous or intermittent noise are poorly understood and the few existing studies investigating the effect highlight contradictory responses in fish. Working in aquaria, we experimentally tested the impact of broadband noise exposure (similar frequency range as anthropogenic boat noise; added either continuously or intermittently) on the behaviour and reproductive success, assessed by the number of obtained eggs, of the common goby (Pomatoschistus microps), a vocal fish with exclusive paternal care.  The continuous noise treatment had the most detrimental effect by reducing spawning probability and females took longer to spawn under continuous noise. Males exposed to continuous noise got significantly fewer egg clutches (4 compared to 11 and 15 in the intermittent noise and silence treatments).  Clutch area did not differ among treatments but clutches in the intermittent and continuous noise treatment had significantly more eggs per cm2. In addition, eggs in the control tanks hatched earlier than in the intermittent and noisy treatments. Larvae reared in continuous noise treatment were larger and had a smaller yolk-sac at hatching than larvae in the intermittent noise treatment and the control. Taken together, we show that noise, particularly a continuous noise exposure, negatively affects reproductive success and early life survival in fish larvae.

  • Male acoustic display in the sand goby – Essential cue in female choice, but unaffected by supplemental feeding
  • 2022
  • Ingår i: Journal of Experimental Marine Biology and Ecology. - : Elsevier. - 0022-0981 .- 1879-1697. ; 556
  • Tidskriftsartikel (refereegranskat)abstract
    • Many teleost fishes use acoustic and visual signalling during courtship. Such displays may convey information about body condition. Here we experimentally altered body condition of sand goby (Pomatoschistus minutus) males to examine effects on acoustic and visual courtship and subsequent spawning decisions. Over two weeks, males fed in excess were fed daily, whereas food-deprived males were fed once a week. Females only spawned with males that produced courtship sound. However, there were no treatment effects on the occurrence of spawning and males fed in excess did not invest more in visual or acoustic courtship than food-deprived males. That said, males fed in excess built more well-covered nests, with more sand piled on top, compared to food-deprived males. Male condition measured as lipid content differed significantly between treatments. However, only males fed in excess differed in lipid content from wild caught males, indicating that in nature, males are of similar condition to males in the low condition treatment group. Apart from the importance of courtship sound, the only male or female behaviour predicting reproductive success was if male displayed in the nest opening. Males often produce courtship sounds together with a visual display in this position. A female dark-eye display did not associate with reproductive success which, together with previous results, suggest a non-ornamental function of this trait. We conclude that male courtship sounds appear to be crucial in female mate choice, but the information content of the courtship sounds and how it relates to male condition remains elusive.

    • Workplace-collected blood spots deposited on filter paper were analysed with multiplexed affinity-based protein assays and found to be suitable for proteomics analysis. The protein extension assay (PEA) was used to characterize 92 proteins using 1.2 mm punches in repeated samples collected from 20 workers. Overall, 97.8% of the samples and 91.3% of the analysed proteins passed quality control. Both within and between spot correlations using six replicates from the same individual were above 0.99, suggesting that comparable levels are obtained from multiple punches from the same spot and from consecutive spots. Protein levels from dried blood and wet serum from the same individuals were compared and the majority of the analysed proteins were found to be significantly correlated. These results open up for simplified sample collection of blood in field conditions for proteomic analysis, but also highlight that not all proteins can be robustly measured from dried whole blood.

  • Is good muscle function a protective factor for early signs of knee osteoarthritis after anterior cruciate ligament reconstruction? The SHIELD cohort study protocol
  • 2020
  • Ingår i: Osteoarthritis and Cartilage Open. - : Elsevier. - 2665-9131. ; 2:4
  • Tidskriftsartikel (refereegranskat)abstract
    • Introduction: Knee injury history and increased joint load, respectively, are major risk factors for the development of knee osteoarthritis (OA). Lower extremity muscle function, such as knee muscle strength, influence joint load and may be important for the onset of knee OA. However, the role of muscle function as a possible modifiable protective mechanism for the development of OA after anterior cruciate ligament reconstruction (ACLR) is not clear. Methods and analysis: In this prospective cohort study, 100 patients (50% women, 18-35 years) with ACLR will be recruited from Skåne University Hospital, Sweden and Oslo University Hospital, Norway. They will be assessed with a comprehensive test battery of muscle function including muscle strength, muscle activation, hop performance, and postural orientation as well as patient-reported outcomes, one year (baseline) and three years (follow-up) after ACLR. Primary predictor will be knee extension strength, primary outcome will be patient-reported knee pain (Knee injury and Osteoarthritis Outcome Score, subscale pain) and secondary outcomes include compositional MRI (T2 mapping) and turnover of cartilage and bone biomarkers. Separate linear regression model will be used to elucidate the influence of each baseline muscle function variable on the outcomes at follow-up, adjusted for baseline values. Twenty non-injured individuals will also be assessed with MRI. This study is approved by The Regional Ethical Review Board in Lund (Sweden) and Oslo (Norway).Discussion: This study may have important clinical implications for using muscle function to screen for risk of early-onset knee OA and for optimizing exercise therapy after knee injury.

  • Biokuma - Biokulturellt arv och alternativ matproduktion : Slutrapport, oktober 2020-september 2022
  • 2023

  • A systematic map of studies testing the relationship between temperature and animal reproduction
  • 2024
  • Ingår i: Ecological Solutions and Evidence. - : John Wiley & Sons. - 2688-8319. ; 5:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Exposure to extreme temperatures can negatively affect animal reproduction, by disrupting the ability of individuals to produce any offspring (fertility), or the number of offspring produced by fertile individuals (fecundity). This has important ecological consequences, because reproduction is the ultimate measure of population fitness: a reduction in reproductive output lowers the population growth rate and increases the extinction risk. Despite this importance, there have been no large-scale summaries of the evidence for effect of temperature on reproduction. We provide a systematic map of studies testing the relationship between temperature and animal reproduction. We systematically searched for published studies that statistically test for a direct link between temperature and animal reproduction, in terms of fertility, fecundity or indirect measures of reproductive potential (gamete and gonad traits).Overall, we collated a large and rich evidence base, with 1654 papers that met our inclusion criteria, encompassing 1191 species. The map revealed several important research gaps. Insects made up almost half of the dataset, but reptiles and amphibians were uncommon, as were non-arthropod invertebrates. Fecundity was the most common reproductive trait examined, and relatively few studies measured fertility. It was uncommon for experimental studies to test exposure of different life stages, exposure to short-term heat or cold shock, exposure to temperature fluctuations, or to independently assess male and female effects. Studies were most often published in journals focusing on entomology and pest control, ecology and evolution, aquaculture and fisheries science, and marine biology. Finally, while individuals were sampled from every continent, there was a strong sampling bias towards mid-latitudes in the Northern Hemisphere, such that the tropics and polar regions are less well sampled. This map reveals a rich literature of studies testing the relationship between temperature and animal reproduction, but also uncovers substantial missing treatment of taxa, traits, and thermal regimes. This database will provide a valuable resource for future quantitative meta-analyses, and direct future studies aiming to fill identified gaps.

  • The history of settlement and agrarian land use in a boreal forestin Värmland, Sweden, new evidence from pollen analysis
  • 2021
  • Ingår i: Vegetation History and Archaeobotany. - : Springer. - 0939-6314 .- 1617-6278. ; , s. 1-13
  • Tidskriftsartikel (refereegranskat)abstract
    • Shielings are the historically known form of transhumance in Scandinavia, where livestock were moved from the farmsteadto sites in the outlands for summer grazing. Pollen analysis has provided a valuable insight into the history of shielings. Thispaper presents a vegetation reconstruction and archaeological survey from the shieling Kårebolssätern in northern Värmland, western Sweden, a renovated shieling that is still operating today. The first evidence of human activities in the area nearKårebolssätern are Hordeum- and Cannabis-type pollen grains occurring from ca. 100 bc. Further signs of human impactare charcoal and sporadic occurrences of apophyte pollen from ca. ad 250 and pollen indicating opening of the canopy ca.ad 570, probably a result of modification of the forest for grazing. A decrease in land use is seen between ad 1000 and 1250,possibly in response to a shift in emphasis towards large scale commodity production in the outlands. Emphasis on bloomeryiron production and pitfall hunting may have caused a shift from agrarian shieling activity. The clearest changes in the pollenassemblage indicating grazing and cultivation occur from the mid-thirteenth century, coinciding with wetter climate at thebeginning of the Little Ice Age. The earliest occurrences of anthropochores in the record predate those of other shieling sitesin Sweden. The pollen analysis reveals evidence of land use that predates the results of the archaeological survey. The studyhighlights how pollen analysis can reveal vegetation changes where early archaeological remains are obscure.

  • Promoting Gender Equality in Media Content: A Limitation or Extension of Freedom of Expression?
  • 2023
  • Ingår i: The Handbook on Gender, Communication, and Women's Human Rights, Margaret Gallagher & Aimee Vega Montiel (Eds.). - Hoboken, New Jersey : Wiley. - 9781119800682 ; , s. 195-212
  • Bokkapitel (refereegranskat)abstract
    • Is promoting gender equality in media content a limitation or an extension of freedom of expression? This question addresses the complicated intertwining of two political values essential in a democratic system. Whereas freedom of expression has long been safeguarded in international and national legal instruments, gender equality was established as a human right more recently. Fulfilling the goal of gender equality in media content through regulation is often perceived as a limitation of freedom of expression by the media industry as well as policymakers. Both self-regulation and regulation through legislation are needed to achieve gender equality in media content when it comes to eliminating gender stereotypes and combating sexist hate speech. Measures to safeguard gender equality in the media should be understood as expanding freedom of expression. It demands a system that goes far beyond protecting the individual right to free speech and the right to freedom from discrimination.

  • Influence of horizontal resolution and complexity of aerosol–cloud interactions on marine stratocumulus and stratocumulus-to-cumulus transition in HadGEM3-GC3.1
  • 2023
  • Ingår i: Quarterly Journal of the Royal Meteorological Society. - 0035-9009 .- 1477-870X. ; 755:149, s. 2049-2066
  • Tidskriftsartikel (refereegranskat)abstract
    • Stratocumulus (Sc) clouds and stratocumulus-to-cumulus transitions (SCTs) are challenging to represent in global models and they contribute to a large spread in modeled subtropical cloud feedbacks. We evaluate the impact of increasing the horizontal model resolution (∼135, 60 and 25 km, respectively) and increasing the complexity of the aerosol–cloud interaction parameterization (interactive versus non-interactive at medium resolution) on springtime subtropical marine Sc properties and SCTs in the atmosphere-only version of HadGEM3-GC3.1. No significant impact on the spatial location of the SCT could be found between the different model versions. Increasing horizontal resolution led to small but significant increases in liquid water content and a stronger (more negative) shortwave (SW) cloud radiative effect (CRE), in particular over the southern-hemisphere Sc regions. However, for two out of the four studied regions, the stronger SW CRE also brought the model outside the range of satellite-derived values of the SW CRE. Applying non-interactive aerosols instead of interactive aerosols also led to significantly higher liquid water content and a stronger SW CRE over the southern-hemisphere Sc regions, while over the northern-hemisphere Sc regions, a competition between a substantial increase in the cloud droplet number concentration and small changes in the liquid water content resulted in a weaker SW CRE or non-significant changes. In general, using interactive instead of non-interactive aerosol–cloud interactions brought the model closer to satellite-retrieved mean values of the SW CRE. Our results suggest that increasing the horizontal resolution or the complexity of the aerosol–cloud parameterization has a small but statistically significant effect on the SW CRE of marine Sc, in particular over regions with high liquid water content. For these regions, the effect of introducing non-interactive versus interactive aerosol–cloud interactions is about as large as increasing the horizontal resolution from medium to high.

  • Methane potentials and organic matter characterization of wood fibres from pulp and paper mills : The influence of raw material, pulping process and bleaching technique
  • 2020
  • Ingår i: Biomass and Bioenergy. - : Elsevier. - 0961-9534 .- 1873-2909. ; 143
  • Tidskriftsartikel (refereegranskat)abstract
    • During the process of pulp- and papermaking, large volumes of fibre-rich primary sludge are generated. Anaerobic digestion of primary sludge offers a substantial potential for methane production as an alternative approach to the inefficient energy recoveries by commonly used incineration techniques. However, a systematic study of the importance of upstream process techniques for the methane potential of pulp fibres is lacking. Therefore, biochemical methane potentials were determined at mesophilic conditions for 20 types of fibres processed by a variety of pulping and bleaching techniques and from different raw materials. This included fibres from kraft, sulphite, semi-chemical, chemical thermo-mechanical (CTMP) and thermo-mechanical pulping plants and milled raw wood. The pulping technique was clearly important for the methane potential, with the highest potential achieved for kraft and sulphite fibres (390–400 Nml CH4 g VS−1). For raw wood and CTMP, hardwood fibres gave substantially more methane than the corresponding softwood fibres (240 compared to 50 Nml CH4 g VS−1 and 300 compared to 160 Nml CH4 g VS−1, respectively). Nuclear magnetic resonance characterization of the organic content demonstrated that the relative lignin content of the fibres was an important factor for methane production, and that an observed positive effect of bleaching on the methane potential of softwood CTMP fibres was likely related to a higher degree of deacetylation and improved accessibility of the hemicellulose. In conclusion, fibres from kraft and sulphite pulping are promising substrates for methane production irrespective of raw material or bleaching, as well as fibres from CTMP pulping of hardwood.

  • Viscosity dynamics and the production of extracellular polymeric substances and soluble microbial products during anaerobic digestion of pulp and paper mill wastewater sludges
  • 2020
  • Ingår i: Bioprocess and biosystems engineering (Print). - : SPRINGER. - 1615-7591 .- 1615-7605. ; 43:2, s. 283-291
  • Tidskriftsartikel (refereegranskat)abstract
    • The production processes of the pulp and paper industry often run in campaigns, leading to large variations in the composition of wastewaters and waste sludges. During anaerobic digestion (AD) of these wastes, the viscosity or the production of extracellular polymeric substances (EPS) and soluble microbial products (SMP) may be affected, with the risk of foam formation, inefficient digester mixing or poor sludge dewaterability. The aim of this study was to investigate how viscosity and production of EPS and SMP during long-term AD of pulp and paper mill sludge is affected by changes in organic loading rate (OLR) and hydraulic retention time (HRT). Two mesophilic lab-scale continuous stirred tank reactors (CSTRs) were operated for 800 days (R1 and R2), initially digesting only fibre sludge, then co-digesting fibre sludge and activated sludge. The HRT was lowered, followed by an increase in the OLR. Reactor fluids were sampled once a month for rheological characterization and analysis of EPS and SMP. The production of the protein fraction of SMP was positively correlated to the OLR, implicating reduced effluent qualities at high OLR. EPS formation correlated with the magnesium content, and during sulphate deficiency, the production of EPS and SMP increased. At high levels of EPS and SMP, there was an increase in viscosity of the anaerobic sludges, and dewatering efficiency was reduced. In addition, increased viscosity and/or the production of EPS and SMP were important factors in sludge bulking and foam formation in the CSTRs. Sludge bulking was avoided by more frequent stirring.

  • Anti-satellite glia cell IgG antibodies in fibromyalgia patients are related to symptom severity and to metabolite concentrations in thalamus and rostral anterior cingulate cortex.
  • 2023
  • Ingår i: Brain, behavior, and immunity. - : Elsevier. - 0889-1591 .- 1090-2139. ; 114, s. 371-382
  • Tidskriftsartikel (refereegranskat)abstract
    • Recent translational work has shown that fibromyalgia might be an autoimmune condition with pathogenic mechanisms mediated by a peripheral, pain-inducing action of immunoglobulin G (IgG) antibodies binding to satellite glia cells (SGC) in the dorsal root ganglia. A first clinical assessment of the postulated autoimmunity showed that fibromyalgia subjects (FMS) had elevated levels of antibodies against SGC (termed anti-SGC IgG) compared to healthy controls and that anti-SGC IgG were associated with a more severe disease status. The overarching aim of the current study was to determine whether the role of anti-SGC IgG in driving pain is exclusively through peripheral mechanisms, as indirectly shown so far, or could be attributed also to central mechanisms. To this end, we wanted to first confirm, in a larger cohort of FMS, the relation between anti-SGC IgG and pain-related clinical measures. Secondly, we explored the associations of these autoantibodies with brain metabolite concentrations (assessed via magnetic resonance spectroscopy, MRS) and pressure-evoked cerebral pain processing (assessed via functional magnetic resonance imaging, fMRI) in FMS. Proton MRS was performed in the thalamus and rostral anterior cingulate cortex (rACC) of FMS and concentrations of a wide spectrum of metabolites were assessed. During fMRI, FMS received individually calibrated painful pressure stimuli corresponding to low and high pain intensities. Our results confirmed a positive correlation between anti-SGC IgG and clinical measures assessing condition severity. Additionally, FMS with high anti-SGC IgG levels had higher pain intensity and a worse disease status than FMS with low anti-SGC IgG levels. Further, anti-SGC IgG levels negatively correlated with metabolites such as scyllo-inositol in thalamus and rACC as well as with total choline and macromolecule 12 in thalamus, thus linking anti-SGC IgG levels to the concentration of metabolites in the brain of FMS. However, anti-SGC IgG levels in FMS were not associated with the sensitivity to pressure pain or the cerebral processing of evoked pressure pain. Taken together, our results suggest that anti-SGC IgG might be clinically relevant for spontaneous, non-evoked pain. Our current and previous translational and clinical findings could provide a rationale to try new antibody-related treatments in FMS.

  • Effects of a palaeolithic diet on obstructive sleep apnoea occurring in females who are overweight after menopause-a randomised controlled trial
  • 2022
  • Ingår i: International Journal of Obesity. - : Springer Science and Business Media LLC. - 0307-0565 .- 1476-5497. ; 46, s. 1833-1839
  • Tidskriftsartikel (refereegranskat)abstract
    • Background/Objectives Obesity is the main risk factor for obstructive sleep apnoea, commonly occurring in females who are overweight after menopause. We aimed to study the effect of a palaeolithic diet on sleep apnoea in females with overweight after menopause from the population. Methods Seventy healthy, non-smoking females with a mean age of 60 years and a mean BMI of 33 kg/m(2) were randomised to a palaeolithic diet or to a control low-fat diet according to Nordic Nutritional Recommendations, for 2 years. The apnoea-hypopnoea index was measured and daytime sleepiness was estimated during the intervention. Results The mean apnoea-hypopnoea index at baseline was 11.6 (95% CI 8.6-14.5). The mean weight loss was 7.2 kg (95% CI 5.3-9.2 kg) in the palaeolithic diet group and 3.9 kg in the control group (95% CI 1.9-5.9 kg); p < 0.021 for the group difference. The reduction in weight corresponded to a reduction in the apnoea-hypopnoea index in the palaeolithic diet group (r = 0.38, p = 0.034) but not in the control group (r = 0.08, p = 0.69). The apnoea-hypopnoea index was reduced in the palaeolithic diet group when the weight was reduced by more than 8 kg. Daytime sleepiness according to the Epworth Sleepiness Scale score and the Karolinska Sleepiness Scale score was unaffected by dietary group allocation. Conclusions A substantial decrease in body weight of 8 kg was needed to achieve a reduction in sleep apnoea in this small trial of women who are overweight after menopause. The palaeolithic diet was more effective for weight reduction than a control low-fat diet and the reduction in sleep apnoea was related to the degree of weight decrement within this diet group.

  • Ingår i: Poverty in Contemporary Economic Thought. - London : Routledge. - 9780367354268 - 9780429331312 ; , s. 188-198
  • Bokkapitel (refereegranskat)abstract
    • This chapter describes the research by Abhijit Banerjee, Esther Duflo, and Michael Kremer, who pioneered the experimental approach to understanding poverty alleviation, for which they were awarded the 2019 Sveriges Riksbank Prize in Economic Sciences in Memory of Alfred Nobel. Economic well-being doubled in the poorest countries between 1995 and 2018. Child mortality has been cut in half relative to 1995, and the proportion of children attending school has gone up from 56 to 80 percent. Any post-intervention differences in the outcome of interest can thus be solely attributed to the treatment. Studies using field experiments are therefore characterized by high internal validity. A key issue is whether experimental results can be generalized to other contexts, whether or not they are externally valid. In the mid-1990s, Michael Kremer and colleagues conducted the first field experiments within development economics in rural Western Kenya.

  • Gender Equality and Media Regulation Study : Sweden
  • 2022
  • Samlingsverk (redaktörskap) (populärvet., debatt m.m.)abstract
    • Freedom of expression and gender equality are core human rights. But women’s voices and perspectives are still missing in most of the news media all over the world, The 6th Global Media Monitoring project indicate that women make up only 25 percent of the news subjects and sources. Could regulation and/or self- regulation of the media be part of the problem - or part of the solution - to increase gender balance in the media? This mapping project, initiated by Fojo Media Institute in collaboration with Gothenburg University’s Department of Journalism, Media & Communication (JMG) and other partners1 is one in a set of country case studies conducted to understand how gender equality and women’s freedom of expression are integrated in media regulation, self-regulation and within media inhouse policies. The foundation for this work is The Beijing Declaration and Platform for Action for the Advancement of women (BPfA) adopted at the Fourth United Nations World Conference on Women (1995) that called on governments and other actors to “increase the participation and access of women to expression and decision making in and through the media and new technologies of communication”, (Strategic objective J.1.) and “promote a balanced and non- stereotyped portrayal of women in the media” (Strategic objective J.2.).Similar to provisions in other policy instruments prior to and following the BPfA’s adoption, area J intimated at possible tensions between guaranteeing (women’s) freedom of expression on the one hand and media independence on the other hand. Freedom of expression and media freedom, however, are not mutually exclusive. Both are necessary for democracy and protected in international standards. Only in a society where there is a plurality of voices that participate in an accessible and dynamic public sphere is there room for the development and improvement of democracy. The case studies were implemented in some of the countries where Fojo is active, namely Armenia, Bangladesh, Rwanda, Somalia, Sweden and Zimbabwe. The case studies accompany a broader global study of gender in media law and policy at international, regional and country levels spanning more than 100 nations. The national studies are based on a pre-formulated framework of questions and tables that then was adapted for the individual country. The inclusion of Sweden as a case was due to both the fact that Fojo Media Institute is based in Sweden and that Sweden has a high standard of living that includes a strong infrastructure both regarding gender equality and freedom of expression. (Due to time restrictions, the Swedish report is quite brief).Ultimately, the global study and the individual country studies presented within the framework of Fojo Media Institute and its partners, seek to guide media development bodies and other stakeholders in their work to promote gender equality in and through the media with full respect for other human rights involved, particularly the right to freedom of expression and freedom of information.Kersti ForsbergDirector, Fojo Media Institute

  • 2021
  • Ingår i: Journal of Clinical Investigation. - : American Society For Clinical Investigation. - 0021-9738 .- 1558-8238. ; 131:13
  • Tidskriftsartikel (refereegranskat)abstract
    • Fibromyalgia syndrome (FMS) is characterized by widespread pain and tenderness, and patients typically experience fatigue and emotional distress. The etiology and pathophysiology of fibromyalgia are not fully explained and there are no effective drug treatments. Here we show that IgG from FMS patients produced sensory hypersensitivity by sensitizing nociceptive neurons. Mice treated with IgG from FMS patients displayed increased sensitivity to noxious mechanical and cold stimulation, and nociceptive fibers in skin-nerve preparations from mice treated with FMS IgG displayed an increased responsiveness to cold and mechanical stimulation. These mice also displayed reduced locomotor activity, reduced paw grip strength, and a loss of intraepidermal innervation. In contrast, transfer of IgG-depleted serum from FMS patients or IgG from healthy control subjects had no effect. Patient IgG did not activate naive sensory neurons directly. IgG from FMS patients labeled satellite glial cells and neurons in vivo and in vitro, as well as myelinated fiber tracts and a small number of macrophages and endothelial cells in mouse dorsal root ganglia (DRG), but no cells in the spinal cord. Furthermore, FMS IgG bound to human DRG. Our results demonstrate that IgG from FMS patients produces painful sensory hypersensitivities by sensitizing peripheral nociceptive afferents and suggest that therapies reducing patient IgG titers may be effective for fibromyalgia.

• Bokkapitel (refereegranskat)abstract
    • The high scoring of the Nordic countries indicates that the Nordic way of striving for gender equality has been successful in a global perspective. Gender equality is an essential element of the Nordic welfare state model, interwoven with and dependent on general welfare measures that shape everyday life for individuals and the organization of society. Anti-discrimination legislation, primarily directed towards the protection of individuals, forms just part of the comprehensive and progressive measures that have been adopted in Sweden in order to achieve gender equality. Despite the high ratings given to gender equality in Sweden in international comparisons and its ambitious gender equality policy and extensive legislation aimed at promoting gender equality and eliminating discrimination, many challenges remain. Together with the other Nordic countries, Sweden is internationally considered a role model and a forerunner for gender equality.

  • Bokkapitel (refereegranskat)abstract
    • A shift from politics to law, inefficient enforcement tools, and dislodgment from action to information as well as criticism of gender equality from various angles have all had an impact on the law’s ability to contribute to the achievement of gender equality in Sweden. It is not primarily through anti-discrimination legislation and litigation that gender equality has been achieved in Sweden, and such an approach will probably not be the main strategy in the future. Still, we have to continue to improve the protection provided to victims of individual discrimination and harassment. The Swedish case shows how the individual’s right to access to justice can be undermined in a context where state bodies – such as the Ombudsman – are given wide discretionary powers to promote equality as a collective public interest.

  • Ophthalmologic Findings in Fetal Alcohol Spectrum Disorders : A Cohort Study From Childhood to Adulthood
  • 2020
  • Ingår i: American Journal of Ophthalmology. - : Elsevier. - 0002-9394 .- 1879-1891. ; 214, s. 14-20
  • Tidskriftsartikel (refereegranskat)abstract
    • PURPOSE: To investigate whether ophthalmologic findings in children with fetal alcohol spectrum disorders (FASD) persist into young adulthood.DESIGN: Prospective cohort study.METHODS: Thirty children (13 female) adopted from eastern Europe to Sweden in the 1990s and diagnosed with FASD by a multidisciplinary team at the median age of 7.9 years were followed up by the same team 13-18 years later. Visual acuity (VA), refraction, stereoacuity, strabismus, ocular media, and fundus were investigated.RESULTS: Median VA in right/left eye (OD/OS) was 20/32/20/32 (0.2/0.2 logMAR) in childhood and 20/22/20/20 (0.05/0.0 logMAR) in adulthood. Median (range) refraction OD/OS was +0.88/+1.25 (-8.75 to +4.75/-9.38 to +5.25) spherical equivalent diopter (D) in childhood and -0.25/-0.25 (-12 to +2.75/-13.25 to +2.63) in adulthood. Astigmatism (≥1 D) was the most common refractive error, in 13 (40%) and 14 (47%) subjects, respectively. Defective stereoacuity (>60 arc second) was noted in 20 subjects (67%) in childhood and 22 (73%) in adulthood. Heterotropia occurred in 12 subjects (40%) in childhood and 13 (43%) in adulthood. Increased tortuosity of the retinal vessels was found in 8 (27%) subjects in childhood vs 11 (37%) in adulthood. Optic nerve hypoplasia was recorded in 3 children and in 4 young adults.CONCLUSIONS: Ophthalmologic findings such as refractive errors, strabismus, and fundus abnormalities are frequent in children with FASD and persist into early adulthood. The facial features characteristic of FAS diminish with age, making a dysmorphology evaluation in adulthood less reliable. An ophthalmologic examination is an important part of the evaluation of FASD in childhood as well as in young adulthood.

  • Outcome for sinonasal malignancies : a population-based survey
  • 2022
  • Ingår i: European Archives of Oto-Rhino-Laryngology. - : Springer Science and Business Media LLC. - 0937-4477 .- 1434-4726. ; 279:5, s. 2611-2622
  • Tidskriftsartikel (refereegranskat)abstract
    • Purpose: Sinonasal malignancies (SNM) represent a rare and complex group of cancers that includes a wide range of histopathological subtypes. Data from population-based cohorts are scarce but warranted as a basis for randomized controlled treatment trials (RCTs). Our aim was to assess overall and histology subset-specific outcomes for SNM patients treated at a tertiary referral centre. Methods: A retrospective, population-based, consecutive cohort of patients with SNMs diagnosed from 2001 through 2019 was examined. Outcome was analysed in relation to age, gender, site, stage, histopathology, and treatment. Results: Two-hundred and twenty-six patients were identified, whereof 61% presented with stage IV disease. 80% completed treatment with curative intent, which comprised surgery with neoadjuvant (29%) or adjuvant (37%) radiotherapy, monotherapy with surgery (22%), definitive chemoradiotherapy (7%), or radiotherapy (5%). Median follow-up was 106 months. The 5- and 10-year overall survival rates were 57% and 35%, respectively. Median overall survival was 76 months (esthesioneuroblastoma: 147 months; adenocarcinoma: 117; salivary carcinoma: 88; mucosal melanoma: 69; squamous cell carcinoma: 51, undifferentiated carcinoma: 42; neuroendocrine carcinoma: 9; and NUT-carcinoma 5). The 5- and 10-year disease-free survival rates were 63% and 54%, respectively, and disease-specific survival 83% and 66%. Increasing age, stage IVB, melanoma histopathology, and treatment with definitive chemoradiotherapy emerged as significant independent prognostic risk factors for disease-specific mortality (p ≤ 0.001). Conclusion: The results indicate a seemingly good outcome in comparison to previous reports, particularly for mucosal melanoma, adenocarcinoma, and undifferentiated carcinoma. The study provides additional background for future RCTs focusing on histology subset-specific treatment for SNM.

  • What's leadership got to do with it? Children's sustainable futures
  • 2024
  • Ingår i: Abstract book. ; , s. 60-60
  • Konferensbidrag (refereegranskat)abstract
    • The aim of our Practice-and-Research project HållUt2023-2026 is to establish what constitutes sustainable leadership in preschool in relation to social, ecological, and economic sustainability according to the Swedish National Curriculum for Preschool (Lpfo2018). While we have been awarded a Sustainable Municipality prize, our knowledge and professional competencies require more effort regarding SDG17 to be able to support our youngest members of society. Acutely aware of the climate crisis and its catastrophic effects on societies, the development of robust pedagogies and sustainable leadership may help strengthen democracy, e.g. by inspiring resilience in children. Theories of professions, lifelong learning and sustainable leadership form part of our participatory action research framework. The Reflection-Action-Reflection-Action model (RARA) also draws on theories and science-based facts in a process-oriented manner. The RARA-model includes dialogue, diaries, videos and ongoing interview studies relating to our evolving understanding of social, ecological and economic development in preschools. Ethics in Participatory Action Research habitually exceed those stipulated by ethics committees and we have put safeguarding mechanisms into place to prevent any ethical shortcomings. We are participants and researching parties and participate as equal members. We strictly observe provisions in law and recommendations by social research ethics bodies, by regularly discussing any issues. The participating academic researcher is tasked with meticulously monitoring that all ethical considerations be taken seriously and swiftly dealt with.By the end of 2026 we expect to: present a model for sustainable development and sustainable leadership, contribute towards practice-and-research based methods, offer knowledge-based professional development from inside preschools, vocalise clearly the importance of preschool teachers’ professional development relating to sustainable development, better protect children by adequately preparing them for the future.

  • Invisible and Ignored : The Archaeology of Nineteenth-Century Subalterns in Sweden
  • 2020
  • Ingår i: International Journal of Historical Archaeology. - : Springer Science and Business Media LLC. - 1092-7697 .- 1573-7748. ; 24, s. 1-21
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper aims to discuss subalterns in different social environments in Sweden. The potential of archaeological studies of landless subalterns in rural and urban areas are shown though a number of case studies. It is argued that archaeology can show the multivocality of the lives of the subalterns, in the same way as it shows how the subalterns organized their daily life. This is done through the use of the concepts of matterscape, powerscape, and mindscape. The subalterns used the physical landscape (matterscape) according to prevailing norms and power structures (powerscape), thus creating a perceptive understanding of their daily landscape (mindscape).

  • Clinical features and treatment response to differentiate idiopathic peritonitis from non-strangulating intestinal infarction of the pelvic flexure associated with Strongylus vulgaris infection in the horse
  • 2022
  • Ingår i: BMC Veterinary Research. - : Springer Science and Business Media LLC. - 1746-6148. ; 18
  • Tidskriftsartikel (refereegranskat)abstract
    • Background Peritonitis in horses secondary to non-strangulating infarction (NSII) has a guarded prognosis, even after intestinal resection. In contrast, horses with idiopathic peritonitis respond well to medical treatment. Affected horses in both cases often show signs of both colic and systemic inflammation, but early diagnosis is crucial for optimal treatment and an accurate prognosis. One cause of NSII is thrombus formation secondary to Strongylus vulgaris larval migration. There has been a documented increase in S. vulgaris prevalence in Sweden since the implementation of selective anthelmintic treatment in 2007, which subsequently could result in a rise in NSII cases. In a retrospective clinical study, medical records from cases diagnosed with NSII of the pelvic flexure or idiopathic peritonitis from three equine referral hospitals in Sweden during 2017-2020 were reviewed. Information including demographic data, relevant medical history, and clinical- and laboratory parameters were obtained from patient records. To facilitate the differentiation between cases of idiopathic peritonitis and cases with confirmed NSII of the pelvic flexure, the aim of the study was to compare clinical and laboratory parameters, clinical progression and initial response to antimicrobial treatment. A secondary aim was to compare survival-rates. Results Horses with NSII (n = 20) were significantly more likely to present during the winter months with a poorer response to medical treatment within 48 h. Cases of idiopathic peritonitis (n = 107) had a 100% survival rate with medical treatment, although one case required surgical correction of a colon displacement. In comparison, all confirmed NSII cases were non-responsive to antimicrobial treatment, with a survival rate to discharge of 50% after colon resection. Specific rectal findings and peripheral blood neutropenia were strongly associated with NSII. Conclusions In Sweden, idiopathic peritonitis cases still predominate over S. vulgaris associated NSII cases and have an excellent survival rate with antimicrobial treatment. However, horses presenting with septic peritonitis during the winter months with a palpable rectal mass and displaying fever and colic signs beyond 48 h of medical treatment are likely to suffer from NSII of the pelvic flexure and should be considered for abdominal surgery.
