  • Platelet-Related Variants Identified by Exomechip Meta-analysis in 157,293 Individuals

    • Platelet production, maintenance, and clearance are tightly controlled processes indicative of platelets' important roles in hemostasis and thrombosis. Platelets are common targets for primary and secondary prevention of several conditions. They are monitored clinically by complete blood counts, specifically with measurements of platelet count (PLT) and mean platelet volume (MPV). Identifying genetic effects on PLT and MPV can provide mechanistic insights into platelet biology and their role in disease. Therefore, we formed the Blood Cell Consortium (BCX) to perform a large-scale meta-analysis of Exomechip association results for PLT and MPV in 157,293 and 57,617 individuals, respectively. Using the low-frequency/rare coding variant-enriched Exomechip genotyping array, we sought to identify genetic variants associated with PLT and MPV. In addition to confirming 47 known PLT and 20 known MPV associations, we identified 32 PLT and 18 MPV associations not previously observed in the literature across the allele frequency spectrum, including rare large effect (FCER1A), low-frequency (IQGAP2, MAP1A, LY75), and common(ZMIZ2, SMG6, PEAR1, ARFGAP3/PACSIN2) variants. Several variants associated with PLT/MPV(PEAR1, MRVI1, PTGES3) were also associated with platelet reactivity. In concurrent BCX analyses, there was overlap of platelet-associated variants with red (MAP1A, TMPRSS6, ZMIZ2) and white (PEAR1, ZMIZ2, LY75) blood cell traits, suggesting common regulatory pathways with shared genetic architecture among these hematopoietic lineages. Our large-scale Exomechip analyses identified previously undocumented associations with platelet traits and further indicate that several complex quantitative hematological, lipid, and cardiovascular traits share genetic factors.

  • An Open Resource for Non-human Primate Imaging

    • Non-human primate neuroimaging is a rapidly growing area of research that promises to transform and scale translational and cross-species comparative neuroscience. Unfortunately, the technological and methodological advances of the past two decades have outpaced the accrual of data, which is particularly challenging given the relatively few centers that have the necessary facilities and capabilities. The PRIMatE Data Exchange (PRIME-DE) addresses this challenge by aggregating independently acquired non-human primate magnetic resonance imaging (MRI) datasets and openly sharing them via the International Neuroimaging Data-sharing Initiative (INDI). Here, we present the rationale, design, and procedures for the PRIME-DE consortium, as well as the initial release, consisting of 25 independent data collections aggregated across 22 sites (total = 217 non-human primates). We also outline the unique pitfalls and challenges that should be considered in the analysis of non-human primate MRI datasets, including providing automated quality assessment of the contributed datasets.

    • Terahertz pulses are very popular because of their numerous applications, for example in security. Located between microwaves and optical waves in the electromagnetic spectrum, their spectral domain can now be exploited for molecular spectroscopy using terahertz emission from plasmas formed by femtosecond laser pulses ionizing gases such as air. Down-conversion of broadband optical spectra in a plasma produces intense radiation suitable for the detection of suspect materials remotely. The different physical mechanisms involved to create terahertz radiation by laser-matter interaction are reviewed. The new potentialities offered by intense ultrafast lasers allow the acquisition of unique spectral signatures characterizing various materials. Copyright (C) EPLA, 2019

    • Experimental research in low-power wireless networking lacks a reference benchmark. While other communities such as databases or machine learning have standardized bench-marks, our community still uses ad-hoc setups for its exper-iments and struggles to provide a fair comparison between communication protocols. Reasons for this include the di-versity of network scenarios and the stochastic nature of wireless experiments. Leveraging on the excellent testbeds and tools that have been built to support experimental val-idation, we make the case for a reference benchmark to pro-mote a fair comparison and reproducibility of results. This abstract describes early design elements and a benchmark-ing methodology with the goal to gather feedback from the community rather than propose a definite solution.

    • As the standardization of full-dimension MIMO systems in the Third Generation Partnership Project progresses, the research community has started to explore the potential of very large arrays as an enabler technology for meeting the requirements of fifth generation systems. Indeed, in its final deliverable, the European 5G project METIS identifies massive MIMO as a key 5G enabler and proposes specific technology components that will allow the cost-efficient deployment of cellular systems taking advantage of hundreds of antennas at cellular base stations. These technology components include handling the inherent pilot-data resource allocation trade-off in a near optimal fashion, a novel random access scheme supporting a large number of users, coded channel state information for sparse channels in frequency-division duplexing systems, managing user grouping and multi-user beamforming, and a decentralized coordinated transceiver design. The aggregate effect of these components enables massive MIMO to contribute to the METIS objectives of delivering very high data rates and managing dense populations.

  • Culprit Vessel-Only Versus Multivessel Percutaneous Coronary Intervention in Patients With Cardiogenic Shock Complicating ST-Segment-Elevation Myocardial Infarction : A Collaborative Meta-Analysis

    • Background The optimal revascularization strategy in patients with multivessel disease presenting with cardiogenic shock complicating ST-segment-elevation myocardial infarction remains unknown. Methods and Results Databases were searched from 1999 to October 2016. Studies comparing immediate/single-stage multivessel percutaneous coronary intervention (MV-PCI) versus culprit vessel-only PCI (CO-PCI) in patients with multivessel disease, ST-segment-elevation myocardial infarction, and cardiogenic shock were included. Primary end point was short-term (in-hospital or 30 days) mortality. Secondary end points included long-term mortality, cardiovascular death, reinfarction, and repeat revascularization. Safety end points were in-hospital stroke, renal failure, and major bleeding. The meta-analysis included 11 nonrandomized studies and 5850 patients (1157 MV-PCI and 4693 CO-PCI). There was no significant difference in short-term mortality with MV-PCI versus CO-PCI (odds ratio [OR], 1.08; 95% confidence interval [CI], 0.81-1.43; P=0.61). Similarly, there were no significant differences in long-term mortality (OR, 0.84; 95% CI, 0.54-1.30; P=0.43), cardiovascular death (OR, 0.72; 95% CI, 0.42-1.23; P=0.23), reinfarction (OR, 1.65; 95% CI, 0.84-3.26; P=0.15), or repeat revascularization (OR, 1.13; 95% CI, 0.76-1.69; P=0.54) between the 2 groups. There was a nonsignificant trend toward higher in-hospital stroke (OR, 1.64; 95% CI, 0.98-2.72; P=0.06) and renal failure (OR, 1.30; 95% CI, 0.98-1.72; P=0.06), with no difference in major bleeding (OR, 1.47; 95% CI, 0.39-5.63; P=0.57) with MV-PCI when compared with CO-PCI. Conclusions This meta-analysis of nonrandomized studies suggests that in patients with cardiogenic shock complicating ST-segment-elevation myocardial infarction, there may be no significant benefit with single-stage MV-PCI compared with CO-PCI. Given the limitations of observational data, randomized trials are needed to determine the role of MV-PCI in this setting.

  • Don’t believe the hype : a grounded exploratory six country wine purchasing study

    • The purpose of this exploratory study was to understand the extent that consumers report purchasing wine on mobile devices and to empirically examine potential drivers of m-wine purchasing across six countries to guide theoretical research enquiry moving forward. Purposive sampling was employed. An online survey involving 2853 respondents from France, Germany, Greece, Canada, US and South Africa forms the basis for the current study. The results of the study indicate that though mobile phone usage, wine consumption and purchasing rates are high, mobile-wine purchasing prevalence is low within all six countries. While technology hype has us believe an online presence is essential for business revenue growth and performance; the current study indicates wineries should carefully consider consumer readiness towards mobile-wine purchasing. Limitations and recommendations for future research are identified.

  • Terahertz emission from laser-driven gas plasmas: a plasmonic point of view

    • We disclose an unanticipated link between plasmonics and nonlinear frequency down-conversion in laser-induced gasplasmas. For two-color femtosecond pump pulses, a plasmonic resonance is shown to broaden the terahertz emission spectra significantly. We identify the resonance as a leaky mode, which contributes to the emission spectra whenever electrons are excited along a direction where the plasma size is smaller than the plasma wavelength. As a direct consequence, such resonances can be controlled by changing the polarization properties of elliptically shaped driving laser pulses. Both experimental results and 3D Maxwell consistent simulations confirm that a significant terahertz pulse shortening and spectral broadening can be achieved by exploiting the transverse driving laser beam shape as an additional degree of freedom.
