  • 2019
  • Ingår i: Computer Vision – ECCV 2018 Workshops. - Cham : Springer Publishing Company. - 9783030110086 - 9783030110093 ; , s. 3-53
  • Konferensbidrag (refereegranskat)abstract
    • The Visual Object Tracking challenge VOT2018 is the sixth annual tracker benchmarking activity organized by the VOT initiative. Results of over eighty trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis and a “real-time” experiment simulating a situation where a tracker processes images as if provided by a continuously running sensor. A long-term tracking subchallenge has been introduced to the set of standard VOT sub-challenges. The new subchallenge focuses on long-term tracking properties, namely coping with target disappearance and reappearance. A new dataset has been compiled and a performance evaluation methodology that focuses on long-term tracking capabilities has been adopted. The VOT toolkit has been updated to support both standard short-term and the new long-term tracking subchallenges. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website (http://votchallenge.net).

  • Comparative analysis of optimal operation strategies for district heating and cooling system based on design and actual load
  • 2017
  • Ingår i: Applied Energy. - : Elsevier BV. - 0306-2619 .- 1872-9118. ; 205, s. 577-588
  • Tidskriftsartikel (refereegranskat)abstract
    • Computational load significantly influences energy and cost savings when developing an operation strategy for a district heating and cooling system. In this study, a model was identified to study the effects of the difference between design load and actual load on an optimal operation strategy. The established model is strongly dependent on the economy principle, and the proposed optimal strategy could achieve a dynamic balance between the users' load and the system energy supply. This model was validated at 30% load rate, which demonstrated an obvious cost saving of 63.6% under the actual load and 42.2% under the design load. Based on the current strategy, the optimal strategy at different load rates was analyzed with respect to two characteristics of each subsystem: energy outputs and operation costs. Furthermore, in the optimal strategy, changes in total operation costs and cost savings rates under different load rates are also discussed. The results showed that, when the load rate was changed from 30 to 75%, the savings rates based on the design load were 42.2, 17.9, 2.5, and 12.6%, and the savings rates based on the actual load were 63.6, 49.8, 34.3, and 25.7%, respectively. Based on the actual load, the energy savings advantage of the optimal operation strategy could be maximized, in particular, during the initial stage of project construction. Furthermore, the commercial software MATLAB was used for programming and calculations. The simulation results indicated that the application of the combined cooling, heating, and power system could significantly improve the cost-effectiveness.

  • Analysis of Heritability and Shared Heritability Based on Genome-Wide Association Studies for 13 Cancer Types
  • 2015
  • Ingår i: Journal of the National Cancer Institute. - : Oxford University Press (OUP). - 0027-8874 .- 1460-2105. ; 107:12
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: Studies of related individuals have consistently demonstrated notable familial aggregation of cancer. We aim to estimate the heritability and genetic correlation attributable to the additive effects of common single-nucleotide polymorphisms (SNPs) for cancer at 13 anatomical sites. Methods: Between 2007 and 2014, the US National Cancer Institute has generated data from genome-wide association studies (GWAS) for 49 492 cancer case patients and 34 131 control patients. We apply novel mixed model methodology (GCTA) to this GWAS data to estimate the heritability of individual cancers, as well as the proportion of heritability attributable to cigarette smoking in smoking-related cancers, and the genetic correlation between pairs of cancers. Results: GWAS heritability was statistically significant at nearly all sites, with the estimates of array-based heritability, h(l)(2), on the liability threshold (LT) scale ranging from 0.05 to 0.38. Estimating the combined heritability of multiple smoking characteristics, we calculate that at least 24% (95% confidence interval [CI] = 14% to 37%) and 7% (95% CI = 4% to 11%) of the heritability for lung and bladder cancer, respectively, can be attributed to genetic determinants of smoking. Most pairs of cancers studied did not show evidence of strong genetic correlation. We found only four pairs of cancers with marginally statistically significant correlations, specifically kidney and testes (rho = 0.73, SE = 0.28), diffuse large B-cell lymphoma (DLBCL) and pediatric osteosarcoma (rho = 0.53, SE = 0.21), DLBCL and chronic lymphocytic leukemia (CLL) (rho = 0.51, SE = 0.18), and bladder and lung (rho = 0.35, SE = 0.14). Correlation analysis also indicates that the genetic architecture of lung cancer differs between a smoking population of European ancestry and a nonsmoking Asian population, allowing for the possibility that the genetic etiology for the same disease can vary by population and environmental exposures. Conclusion: Our results provide important insights into the genetic architecture of cancers and suggest new avenues for investigation.

  • Muscle-like Artificial Molecular Actuators for Nanoparticles
  • 2018
  • Ingår i: Chem. - : Cell Press. - 2451-9308 .- 2451-9294. ; 4:11, s. 2670-2684
  • Tidskriftsartikel (refereegranskat)abstract
    • Muscle tissue performs crucial contraction/extension motions that generate mechanical force and work by consuming chemical energy. Inspired by this naturally created biomolecular machine, artificial molecular muscles are designed and synthesized to undertake linear actuation functions. However, most of these muscle-like actuators are performed at large ensembles, while to realize the nanoscale actuation at the single-to few-molecule level remains challenging. Herein, we developed an artificial muscle-like molecular actuator that can reversibly control the proximity of the attached nano-objects, gold nanoparticles, within the single-molecule length level by its stimuli-responsive muscle-like linear contraction/extension motion. The molecular actuation motion is accompanied by an optical signal output resulting from the plasmonic resonance properties of gold nanoparticles. Meanwhile, the thermal noise of the muscle-like molecular actuator can be overcome by integrating the optical signal over a sufficiently long period.

  • Mutations in TUBB8 and Human Oocyte Meiotic Arrest.
  • 2016
  • Ingår i: The New England journal of medicine. - 1533-4406. ; 374:3, s. 223-232
  • Tidskriftsartikel (refereegranskat)abstract
    • Background Human reproduction depends on the fusion of a mature oocyte with a sperm cell to form a fertilized egg. The genetic events that lead to the arrest of human oocyte maturation are unknown. Methods We sequenced the exomes of five members of a four-generation family, three of whom had infertility due to oocyte meiosis I arrest. We performed Sanger sequencing of a candidate gene, TUBB8, in DNA samples from these members, additional family members, and members of 23 other affected families. The expression of TUBB8 and all other β-tubulin isotypes was assessed in human oocytes, early embryos, sperm cells, and several somatic tissues by means of a quantitative reverse-transcriptase-polymerase-chain-reaction assay. We evaluated the effect of the TUBB8 mutations on the assembly of the heterodimer consisting of one α-tubulin polypeptide and one β-tubulin polypeptide (α/β-tubulin heterodimer) in vitro, on microtubule architecture in HeLa cells, on microtubule dynamics in yeast cells, and on spindle assembly in mouse and human oocytes. Results We identified seven mutations in the primate-specific gene TUBB8 that were responsible for oocyte meiosis I arrest in 7 of the 24 families. TUBB8 expression is unique to oocytes and the early embryo, in which this gene accounts for almost all the expressed β-tubulin. The mutations affect chaperone-dependent folding and assembly of the α/β-tubulin heterodimer, disrupt microtubule behavior on expression in cultured cells, alter microtubule dynamics in vivo, and cause catastrophic spindle-assembly defects and maturation arrest on expression in mouse and human oocytes. Conclusions TUBB8 mutations have dominant-negative effects that disrupt microtubule behavior and oocyte meiotic spindle assembly and maturation, causing female infertility. • Aerodynamic effect of wind barriers and running safety of trains on high-speed railway bridges under cross winds
  • 2015
  • Ingår i: Wind and Structures. - : Techno-Press. - 1226-6116 .- 1598-6225. ; 20:2, s. 213-236
  • Tidskriftsartikel (refereegranskat)abstract
    • For high-speed railways (HSR) in wind prone regions, wind barriers are often installed on bridges to ensure the running safety of trains. This paper analyzes the effect of wind barriers on the running safety of a high-speed train to cross winds when it passes on a bridge. Two simply-supported (S-S) PC bridges in China, one with 32 m box beams and the other with 16 m trough beams, are selected to perform the dynamic analyses. The bridges are modeled by 3-D finite elements and each vehicle in a train by a multi-rigid-body system connected with suspension springs and dashpots. The wind excitations on the train vehicles and the bridges are numerically simulated, using the static tri-component coefficients obtained from a wind tunnel test, taking into account the effects of wind barriers, train speed and the spatial correlation with wind forces on the deck. The whole histories of a train passing over the two bridges under strong cross winds are simulated and compared, considering variations of wind velocities, train speeds and without or with wind barriers. The threshold curves of wind velocity for train running safety on the two bridges are compared, from which the windbreak effect of the wind barrier are evaluated, based on which a beam structure with better performance is recommended.

  • Colour-tunable fluorescence of single molecules based on the vibration induced emission of phenazine
  • 2015
  • Ingår i: Chemical Communications. - : Royal Society of Chemistry (RSC). - 1359-7345 .- 1364-548X. ; 51:21, s. 4462-4464

    • Due to the vibration of the phenazine unit, compound S1 exhibits dual fluorescence in solution but one peak in the solid state. Based on this novel phenomenon and combined with the intramolecular energy transfer (IET) effect, a colour-tunable luminescence, even near white emission from a single molecule could be achieved in two different ways: controlling the polarity of the solvent and the aggregation index.

  • Enantiospecific photoresponse of sterically hindered diarylethenes for chiroptical switches and photomemories
  • 2015
  • Ingår i: Scientific Reports. - : Springer Science and Business Media LLC. - 2045-2322. ; 5
  • Tidskriftsartikel (refereegranskat)abstract
    • Light-driven transcription, replication and enzyme catalysis are critically dependent upon a delicate transfer between molecular and supramolecular chirality. Chemists have well realized the impressive stereospecificity over many thermally accessible cycloaddition with chiral catalysts, but making light work in the enantiomer control of diarylethene photocyclization has proved to be more challenging. Here, we report a unique sterically hindered diarylethene (BBTE) system with absolute enantiospecific photocyclization and cycloreversion. Moreover, we have fully separated all the five thermally stable isomers, consisting of one achiral parallel conformer, one pair of anti-parallel ring-open enantiomers, and another pair of ring-closed enantiomers, whose absolute chiral configurations are entirely elucidated by single X-ray crystallographic analyses. The photo-responsive feature exhibits a reversible, complete enantio-control transformation without racemism, offering an unrivaled unimolecular enantiospecific platform for potential applications as bistable chiroptical switches and all-photonic photomemories with optical rotation as non-destructive readout.

  • Sterically hindered diarylethenes with a benzobis(thiadiazole) bridge : photochemical and kinetic studies
  • 2015
  • Ingår i: Journal of Materials Chemistry C. - : Royal Society of Chemistry (RSC). - 2050-7526 .- 2050-7534. ; 3:33, s. 8665-8674
  • Tidskriftsartikel (refereegranskat)abstract
    • Four rationally designed diarylethenes (DAEs) 1-4 with a benzobis(thiadiazole) bridge are specifically designed for gaining insights into steric effects on photochromic performances. It is shown that, upon increasing steric hindrance, the exchanging rate between two main conformers in the ring-open form gradually slows down, offering the opportunity for isolating photoactive anti-parallel conformers. Impressively, the separated anti-parallel conformer shows high cyclization quantum yields over the unresolved common DAEs. The typical donor-pi-acceptor (D-pi-A) feature in ring-open DAEs 1-4 endows their prominent fluorescence, which can be conveniently modulated by photocyclization. In the ring-closed form, the excess steric hindrance is found to seriously disrupt the thermal bistability, and particularly 3c fades quickly with a half-life of several hours at ambient temperature. In contrast, both 1c and 2c exhibit excellent stability, which originates from the stabilization effects of intramolecular hydrogen bonds. This work demonstrates the steric effects on the photochemical and kinetic behaviors of DAEs, providing a unique approach to develop photochromic DAEs with high photosensitivity.

  • Enhanced Photocurrent Density by Spin-Coated NiO Photocathodes for N-Annulated Perylene-Based p-Type Dye-Sensitized Solar Cells
  • 2016
  • Ingår i: ACS Applied Materials and Interfaces. - : American Chemical Society (ACS). - 1944-8244 .- 1944-8252. ; 8:30, s. 19393-19401
  • Tidskriftsartikel (refereegranskat)abstract
    • The low photocurrent density of p-type dye sensitized solar cells (p-DSSCs) has limited the development of high-efficiency tandem cells due to the inadequate light harvesting ability of sensitizers and the low hole mobility of semiconductors. Hereby, two new "push-pull" type organic dyes (PQ-1 and PQ-2) containing N-annulated perylene as electron donor have been synthesized, where the PQ-2-based p-DSSCs show higher photoelectric conversion efficiency (PCE) of 0.316% owing to the higher molar extinction compared to of that PQ-1. Additionally, the photocurrent densities were remarkably increased from 2.20 to 5.85 mA cm(-2) for PQ-1 and 2.45 to 6.69 mA cm(-2) for PQ-2 by spin coated NiO photocathode based-p-DSSCs, respectively. This results are ascribed to the enhancement of hole transport rate, dye loading amounts and transparency of NiO films in comparison to that prepared by screen-printing method. Electrochemical impedance spectroscopy and theoretical calculations studies indicate that the molecular dipole moment approaching closer to the NiO surface shifts the quasi-Fermi level to more positive levels, improving,open-circuit voltage (V-oc). Intensity-modulated photocurrent spectroscopy illustrates that the hole transit time in NiO films prepared in spin-coating is shorter than that prepared by screen-printing method.

  • Molecular engineering of D-A-pi-A sensitizers for highly efficient solid-state dye-sensitized solar cells
  • 2017
  • Ingår i: Journal of Materials Chemistry A. - : RSC Publishing. - 2050-7488 .- 2050-7496. ; 5:7, s. 3157-3166
  • Tidskriftsartikel (refereegranskat)abstract
    • Two newquinoxaline-based D-A-pi-A organic sensitizers AQ309 and AQ310 have been designed and synthesized employing 3,4-ethylenedioxythiophene (EDOT) and cyclopentadithiophene (CPDT) as plinker units, respectively. The new AQ309 and AQ310 dyes have been applied in all-solid-state dye-sensitized solar cells (ssDSSCs). An impressive record photoelectric conversion efficiency (PCE) of 8.0% for AQ310-based ssDSSCs using Spiro-OMeTAD as the hole transport material (HTM) was obtained under standard AM 1.5 (100 mW cm (2)) solar intensity. This clearly outperforms the PCE of the state-of-theart organic D-pi-A dye LEG4-based devices showing a PCE of 7.3% under the same conditions. Moreover, an excellent high PCE of 8.6% was also recorded for AQ310-based devices under 50% solar intensity. Meanwhile, the AQ310-based ssDSSCs showed a much longer electron lifetime according to the transient photovoltage decay measurement, demonstrating lower charge recombination losses in the devices. Photo-induced absorption spectroscopy (PIA) indicated that AQ310 could be more efficiently regenerated by Spiro-OMeTAD. These results show that molecular engineering is a promising strategy to develop D-A-pi-A organic sensitizers for highly efficient ssDSSCs.

  • Theoretical insight into the enhanced hindrance, thermal stability and optical properties of diarylethene with a benzobis(thiadiazole) bridge and benzothiophene rings
  • 2016
  • Ingår i: Dyes and pigments. - : ELSEVIER SCI LTD. - 0143-7208 .- 1873-3743. ; 125, s. 348-355
  • Tidskriftsartikel (refereegranskat)abstract
    • To rationalize the effects of intramolecular steric hindrance on the performance of diarylethene photoswitches, we here present a density functional theory study on the thermal bistability and optical properties of a photochromic diarylethene BBTE consisting of a benzobis(thiadiazole) bridge and benzothiophene rings, in which prominent steric hindrance exists owing to the extended structures of is-conjugated groups. Our calculations not only provide rational explanations for the isolation of enantiopure anti-parallel conformers of the open-ring isomer, but also elucidate the detailed pathway of a two-step ground-state ring-opening process, where the thermal stability of the closed-ring isomer is guaranteed by an overall free energy barrier of around 113 kJ mol(-1) (27 kcal mol(-1)). In addition, the tunable intramolecular charge transfer of the donor acceptor structure formed by the benzobis(thiadiazole) bridge and the bulky benzothiophene rings is also addressed within time-dependent density functional theory. Charge transfer excitations in the open- and the closed-ring isomers are characterized as long-range and medium-range, respectively. We show that the diarylethene derivative under investigation can serve as a promising platform for future development of optoelectronic materials.

  • Quantitative proteomic analysis of gastric cancer tissue reveals novel proteins in platelet-derived growth factor B signaling pathway
  • 2017
  • Ingår i: Oncotarget. - : IMPACT JOURNALS LLC. - 1949-2553. ; 8:13, s. 22059-22075
  • Tidskriftsartikel (refereegranskat)abstract
    • Gastric cancer is one of the most common cancers in Asian countries. Searching for reliable biomarkers involving the development of gastric cancer is important for clinical practice. Quantitative proteomics has become an important method contributed to the discovery of novel diagnostic or therapeutic targets for the management of cancer. Here, we identified differently expressed proteins in gastric cancer and normal gastric tissues by using the high resolution mass spectrometer. Among the total of 2280 identified proteins, 87 were differentially expressed between gastric cancer and normal gastric tissues. Notably, several significant proteins are in the PDGF-B signaling pathway, including peroxiredoxin5 (PRDX5), S100A6, calreticulin (CALR) and cathepsin D (CTSD), which were validated by western blot. Furthermore, upstream regulators including PDGF-B, PDGFR-beta, Akt, eIF4E and p70s6K were found significantly increased in the gastric cancer tissues. In addition, silencing of PRDX5 and PDGF-B suppressed the proliferation of gastric cancer cells in vitro. The administration of exogenous PDGF-BB recovered the reduced expression of PDGF-B signaling pathway in PDGF-B knockdown cells. Taken together, our findings suggested that PDGF-B signaling pathway plays an important role in the regulation of gastric cancer proliferation and the inhibition of this pathway may be a potential approach for treatment of gastric cancer.

  • Toward more realistic projections of soil carbon dynamics by Earth system models
  • 2016
  • Ingår i: Global Biogeochemical Cycles. - 0886-6236. ; 30:1, s. 40-56
  • Tidskriftsartikel (refereegranskat)abstract
    • Soil carbon (C) is a critical component of Earth system models (ESMs), and its diverse representations are a major source of the large spread across models in the terrestrial C sink from the third to fifth assessment reports of the Intergovernmental Panel on Climate Change (IPCC). Improving soil C projections is of a high priority for Earth system modeling in the future IPCC and other assessments. To achieve this goal, we suggest that (1) model structures should reflect real-world processes, (2) parameters should be calibrated to match model outputs with observations, and (3) external forcing variables should accurately prescribe the environmental conditions that soils experience. First, most soil C cycle models simulate C input from litter production and C release through decomposition. The latter process has traditionally been represented by first-order decay functions, regulated primarily by temperature, moisture, litter quality, and soil texture. While this formulation well captures macroscopic soil organic C (SOC) dynamics, better understanding is needed of their underlying mechanisms as related to microbial processes, depth-dependent environmental controls, and other processes that strongly affect soil C dynamics. Second, incomplete use of observations in model parameterization is a major cause of bias in soil C projections from ESMs. Optimal parameter calibration with both pool- and flux-based data sets through data assimilation is among the highest priorities for near-term research to reduce biases among ESMs. Third, external variables are represented inconsistently among ESMs, leading to differences in modeled soil C dynamics. We recommend the implementation of traceability analyses to identify how external variables and model parameterizations influence SOC dynamics in different ESMs. Overall, projections of the terrestrial C sink can be substantially improved when reliable data sets are available to select the most representative model structure, constrain parameters, and prescribe forcing fields.

  • 2018
  • Ingår i: Applied Microbiology and Biotechnology. - : Springer Science and Business Media LLC. - 1432-0614 .- 0175-7598. ; 102:22, s. 9541-9548
  • Forskningsöversikt (refereegranskat)abstract
    • © 2018, The Author(s). The oleaginous yeast Yarrowia lipolytica is widely used for the production of both bulk and fine chemicals, including organic acids, fatty acid-derived biofuels and chemicals, polyunsaturated fatty acids, single-cell proteins, terpenoids, and other valuable products. Consequently, it is becoming increasingly popular for metabolic engineering applications. Multiple gene manipulation tools including URA blast, Cre/LoxP, and transcription activator-like effector nucleases (TALENs) have been developed for metabolic engineering in Y. lipolytica. However, the low efficiency and time-consuming procedures involved in these methods hamper further research. The emergence of the CRISPR/Cas system offers a potential solution for these problems due to its high efficiency, ease of operation, and time savings, which can significantly accelerate the genomic engineering of Y. lipolytica. In this review, we summarize the research progress on the development of CRISPR/Cas systems for Y. lipolytica, including Cas9 proteins and sgRNA expression strategies, as well as gene knock-out/knock-in and repression/activation applications. Finally, the most promising and tantalizing future prospects in this area are highlighted.

  • 2016
  • Ingår i: Nature Photonics. - : NATURE PUBLISHING GROUP. - 1749-4885 .- 1749-4893. ; 10:11, s. 699-
  • Tidskriftsartikel (refereegranskat)abstract
    • Organometal halide perovskites can be processed from solutions at low temperatures to form crystalline direct-bandgap semiconductors with promising optoelectronic properties(1-5). However, the efficiency of their electroluminescence is limited by non-radiative recombination, which is associated with defects and leakage current due to incomplete surface coverage(6-9). Here we demonstrate a solution-processed perovskite light-emitting diode (LED) based on self-organized multiple quantum wells (MQWs) with excellent film morphologies. The MQW-based LED exhibits a very high external quantum efficiency of up to 11.7%, good stability and exceptional highpower performance with an energy conversion efficiency of 5.5% at a current density of 100 mA cm(-2). This outstanding performance arises because the lower bandgap regions that generate electroluminescence are effectively confined by perovskite MQWs with higher energy gaps, resulting in very efficient radiative decay. Surprisingly, there is no evidence that the large interfacial areas between different bandgap regions cause luminescence quenching.

    • Logic is the key to computing. Traditionally, logic devices have been fabricated by the top-down approach, whose dimensions are drastically limited. The ultimate goal is to use molecular tailorability to design logics using the "bottom-up'' approach. Here we report an unprecedented photochromic molecule that undergoes unimolecular logic switching when excited anywhere in the entire UV-visible spectrum, thus a bottom-up, all-photonic, molecular logic gate. Specifically, these molecular photonic logics embedded in the polymer thin films function as the "AND'' or "OR'' gate at different temporal responses. To achieve high information-processing density, moreover, a ternary flip-flap-flop gate is realized in the molecular logic because the fact that this photochromic molecule can be photoswitched anywhere in its UV-vis spectrum enabled three different lasers (532, 473, and 561 nm) as the inputs to deliver the complex logic optical outputs.

    • Three new metal-free organic sensitizers with a D-A-p-A configuration based on a triarylamine donor, 2,1,3-benzothiadiazole (BTD) and different pi units have been synthesized. Because a long alkyl side chain has been attached to the thiophene adjacent to BDT, HECA achieves the best performance. Under standard global AM 1.5 solar light conditions, the HECA based-device gives the best conversion efficiency of 9.13% with a J(sc) of 16.14 mA cm(-2), a V-oc of 0.72 V and a FF of 0.77. Although EDCA displays superior optical properties, it does not give the desired results. The device based on EDCA achieves efficiency of 7.37% with a J(sc) of 15.56 mA cm(-2), a V-oc of 0.67 V and a FF of 0.70. EIS analysis confirms the existence of fast recombination in the EDCA system. Further computational analysis indicates that the coplanarity and the very strong affinity to iodine molecules of EDCA may be the origin of the low performance.

    • We report a new dye (INPA) adopting pyrimidine-2-carboxylic acid as an electron-accepting and anchoring group to be used in dye-sensitized solar cells. IR spectral analysis indicates that the anchoring group may form two coordination bonds with TiO2 and so facilitate the interaction between the anchoring group and TiO2. The INPA-based cell exhibits an overall conversion efficiency of 5.45%, which is considerably higher than that obtained with cyanoacrylic acid commonly used as the electron acceptor.

  • Overexpression of Rac GTPase Activating Protein 1 Contributes to Proliferation of Cancer Cells by Reducing Hippo Signaling to Promote Cytokinesis
  • 2018
  • Ingår i: Gastroenterology. - : Elsevier BV. - 0016-5085. ; 155:4
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND & AIMS: Agents designed to block or alter cytokinesis can kill or stop proliferation of cancer cells. We aimed to identify cytokinesis-related proteins that are overexpressed in hepatocellular carcinoma (HCC) cells and might be targeted to slow liver tumor growth. METHODS: Using the Oncomine database, we compared the gene expression patterns in 16 cancer microarray datasets and assessed gene enrichment sets using gene ontology. We performed immunohistochemical analysis of an HCC tissue microarray and identified changes in protein levels that are associated with patient survival times. Candidate genes were overexpressed or knocked down with small hairpin RNAs in SMMC7721, MHCC97H, or HCCLM3 cell lines; we analyzed their proliferation, viability, and clone-formation ability and their growth as subcutaneous or orthotopic xenograft tumors in mice. We performed microarray analyses to identify alterations in signaling pathways and immunoblot and immunofluorescence assays to detect and localize proteins in tissues. Yeast 2-hybrid screens and mass spectrometry combined with co-immunoprecipitation experiments were used to identify binding proteins. Protein interactions were validated with co-immunoprecipitation and proximity ligation assays. Chromatin immunoprecipitation, promoter luciferase activity, and quantitative real-time polymerase chain reaction analyses were used to identify factors that regulate transcription of specific genes. RESULTS: The genes that were most frequently overexpressed in different types of cancer cells were involved in cell division processes. We identified 3 cytokinesis-regulatory proteins among the 10 genes most frequently overexpressed by all cancer cell types. Rac GTPase activating protein 1 (RACGAP1) was the cytokinesis-regulatory protein that was most highly overexpressed in multiple cancers. Increased expression of RACGAP1 in tumor tissues was associated with shorter survival times of patients with cancer. Knockdown of RACGAP1 in HCC cells induced cytokinesis failure and cell apoptosis. In microarray analyses, we found knockdown of RACGAP1 in SMMC7721 cells to reduce expression of genes regulated by yes-associated protein (YAP) and WW domain containing transcription regulator 1 (WWTR1 or TAZ). RACGAP1 reduced activation of the Hippo pathway in HCC cells by increasing activity of RhoA and polymerization of filamentous actin. Knockdown of YAP reduced phosphorylation of RACGAP1 and redistribution at the anaphase central spindle. We found transcription of the translocated promoter region, nuclear basket protein (TPR) to be regulated by YAP and coordinately expressed with RACGAP1 to promote proliferation of HCC cells. TPR redistributed upon nuclear envelope breakdown and formed complexes with RACGAP1 during mitosis. Knockdown of TPR in HCC cells reduced phosphorylation of RACGAP1 by aurora kinase B and impaired their redistribution at the central spindle during cytokinesis. STAT3 activated transcription of RACGAP in HCC cells. CONCLUSIONS: In an analysis of gene expression patterns of multiple tumor types, we found RACGAP1 to be frequently overexpressed, which is associated with shorter survival times of patients. RACGAP1 promotes proliferation of HCC cells by reducing activation of the Hippo and YAP pathways and promoting cytokinesis in coordination with TPR.

  • Tourism water footprint : an empirical analysis of Mount Huangshan
  • 2017
  • Ingår i: Asia Pacific Journal of Tourism Research. - : Routledge. - 1094-1665 .- 1741-6507. ; 22:10, s. 1083-1098
  • Tidskriftsartikel (refereegranskat)abstract
    • Water is an important factor for the sustainable development of tourism. We constructed a comprehensive model of the tourism water footprint (TWF), including tourism sewage and water for management, and used the world heritage Mount Huangshan as an example. The results showed that the total TWF, which included green, blue and grey water of Mount Huangshan in 2012, was about 10.19 million m(3)/year, approximately per tourist 3.39m(3)/day or 3387L/day. Tourism sewage and food were the main factors of water consumption. The spatial transfer of TWF led the spillover of tourism environmental impact, not only affecting Mount Huangshan but the Huangshan City and even nationwide. Tourist flow and temperature had a highly significant positive correlation with the TWF. Quantifying the TWF can reflect the pressure of tourists on water resources, and provide an effective decision-making basis for rational use of water resources.

  • Multicolor Photoluminescence Including White-Light Emission by a Single Host-Guest Complex
  • 2016
  • Ingår i: Journal of the American Chemical Society. - : American Chemical Society (ACS). - 0002-7863 .- 1520-5126. ; 138:41, s. 13541-13550
  • Tidskriftsartikel (refereegranskat)abstract
    • Achieving multicolor photoluminescence, 'especially white-light emission, under mild conditions based on a single "fluorescent compound-is a great challenge. Herein, we report a novel colorful-emission host guest complex BPCY, which is composed of a two-arm fluorescent guest molecule (BPC) and gamma-cyclodextrin (gamma-CD) as the host-molecule. BPC bears a unique asymmetrical donor acceptor donor (D-1-A(+)similar to D-2)-type structure, where D-1, A(+), and D-2 stand for the binaphthol electron donor, pyridinium electron acceptor, and coumarin electron donor, respectively. The luminescence property of BPC shows dual-sensitivity, i.e., toward the excitation wavelength and the cyclodextrin host molecule. Under certain conditions, the complex shows three different emission wavelengths, allowing the realization of multicolor photoluminescence, including red (R), green (G), and blue (B) as well as various intermediate colors by orthogonally modulating these two stimuli. In this way, nearly pure white-light emission with CIE coordinates (0.33, 0.34) could be generated. A combination of structural, spectroscopic, and computational simulation studies revealed the occurrence of synergetic mechanistic processes for the stimuli-responsive multicolor luminescence of the BPCY complex, namely, host-enhanced intramolecular charge-transfer (ICT) and host-induced restriction of intramolecular rotation (RIR). This new supramolecular complex with superior multicolor emission abilities may find wide applications in the fields of information processing and display media. Furthermore, the molecular design rationale presented here may provide a new design strategy for the development of high performance optical materials using a single supramolecular platform.

  • Virtual water trade of agricultural products : A new perspective to explore the Belt and Road
  • 2018
  • Ingår i: Science of the Total Environment. - : Elsevier. - 0048-9697 .- 1879-1026. ; 622, s. 988-996
  • Tidskriftsartikel (refereegranskat)abstract
    • The Belt and Road is an initiative of cooperation and development that was proposed by China. Moreover, most of the spanning countries faced water shortages and agriculture consumed a lot of water. Virtual water links water, food and trade and is an effective tool to ease water shortages. Therefore, this paper aims to understand the Belt and Road from the new perspective of virtual water trade of agricultural products. We considered agricultural products trade from 2001 to 2015. On the whole, the results indicated that China was in virtual water trade surplus with the countries along the Belt and Road. However, in terms of each country, >40 spanning countries were in virtual water trade surplus with China and eased water shortages. Russia had the largest net imported virtual water from China. Furthermore, the proportion of the grey water footprint that China exported to the spanning countries was much higher than that imported, no matter from the whole or different geographical regions. Moreover, more than half of the countries' virtual water trade with China conformed to the virtual water strategy, which helped to ease water crises. Furthermore, the products that they exported to China were mainly advantageous products that each spanning countries have. Virtual water trade is a new perspective to explore the Belt and Road. Agricultural products trade with China definitely benefits both the countries along the Belt and Road and China from the perspective of virtual water. The findings are beneficial for the water management of the countries along the Belt and Road and China, alleviating water shortages, encouraging the rational allocation of water resources in the various departments. They can provide references for optimizing trade structures as well. (c) 2017 Elsevier B.V. All rights reserved.
