    • The Industry 4.0 revolution relies heavily on data to generate value, innovation, new services, and optimize current processes [1]. Technologies such as Internet of Things (IoT), machine learning, digital twins, and much more depend directly on data to bring value and innovation to both discrete manufacturing and process industries. The origin of data may vary from sensor data to financial statements and even strictly confidential user or business data. In data-driven ecosystems, collaboration between different actors is often needed to provide services such as analytics, logistics, predictive maintenance, process improvement, and more. Data therefore cannot be considered a corporate internal asset only. Hence, data needs to be shared among organizations in a data-driven ecosystem for it to be used as a strategic resource for creating desired values, innovations, or process improvements [2]. When sharing business critical and sensitive data, the access to the data needs to be accurately controlled to prevent leakage to authorized users and organizations. Access control is a mechanism to control actions of users over objects, e.g., to read, write, and delete files, accessing data, writing over registers, and so on. This thesis studies one of the latest access control mechanisms in Attribute Based Access Control (ABAC) for industrial data sharing. ABAC emerges as an evolution of the commonly industry-wide used Role-based Access Control. ABAC presents the idea of attributes to create access policies, rather than manually assigned roles or ownerships, enabling for expressive fine-granular access control policies. Furthermore, this thesis presents approaches to implement ABAC into industrial IoT data sharing applications, with special focus on the manageability and granularity of the attributes and policies.  The thesis also studies the implications of outsourced data storage on third party cloud servers over access control for data sharing and explores how to integrate cryptographic techniques and paradigms into data access control. In particular, the combination of ABAC and Attribute-Based Encryption (ABE) is investigated to protect privacy over not-fully trusted domains. In this, important research gaps are identified.

  • Investigation of Microservice-Based Workflow Management Solutions for Industrial Automation

    • In an era ruled by data and information, engineers need new tools to cope with the increased complexity of industrial operations. New architectural models for industry enable open communication environments, where workflows can play a major role in providing flexible and dynamic interactions between systems. Workflows help engineers maintain precise control over their factory equipment and Information Technology (IT) services, from the initial design stages to plant operations. The current application of workflows departs from the classic business workflows that focus on office automation systems in favor of a manufacturing-oriented approach that involves direct interaction with cyber-physical systems (CPSs) on the shop floor. This paper identifies relevant industry-related challenges that hinder the adoption of workflow technology, which are classified within the context of a cohesive workflow lifecycle. The classification compares the various workflow management solutions and systems used to monitor and execute workflows. These solutions have been developed alongside the Eclipse Arrowhead framework, which provides a common infrastructure for designing systems according to the microservice architectural principles. This paper investigates and compares various solutions for workflow management and execution in light of the associated industrial requirements. Further, it compares various microservice-based approaches and their implementation. The objective is to support industrial stakeholders in their decision-making with regard to choosing among workflow management solutions.

    • In an era ruled by data and information, workflows are the tools used by engineers to obtain precise control over their factory equipment and Information Technology (IT) services. The study of workflows differentiates two kinds: scientific workflows, focus on the data-flow, and business workflows, task and control-flow oriented. In this work we focus on the latter type, studying and comparing various workflow management approaches, the systems used to monitor and execute workflows. The development inside the Arrowhead framework has lead to different alternatives for workflow management and execution, some closer to the original business process field where the term workflow management originated, while others deal with lower-level systems, the kind of cyber-physical systems that will populate factories in the forthcoming Industry 4.0

    • Morphine and its derivatives play inevitably important role in the m-opioid receptor (MOR) targeted antinociception. A structure-activity relationship study is presented for novel and known orvinol and thevinol derivatives with varying 3-O, 6-O, 17-N and 20-alkyl substitutions starting from agonists, antagonists and partial agonists. In vitro competition binding experiments with [H-3]DAMGO showed low subnanomolar affinity to MOR. Generally, 6-O-demethylation increased the affinity toward MOR and decreased the efficacy changing the pharmacological profile in some cases. In vivo tests in osteoarthritis inflammation model showed significant antiallodynic effects of thevinol derivatives while orvinol derivatives did not. The pharmacological character was modelled by computational docking to both active and inactive state models of MOR. Docking energy difference for the two states separates agonists and antagonists well while partial agonists overlapped with them. An interaction pattern of the ligands, involving the interacting receptor atoms, showed more efficient separation of the pharmacological profiles. In rats, thevinol derivatives showed antiallodynic effect in vivo. The orvinol derivatives, except for 6-O-desmethyl-dihydroetorfin (2c), did not show antiallodynic effect.

    • Opiate alkaloids and their synthetic derivatives are still widely used in pain management, drug addiction, and abuse. To avoid serious side effects, compounds with properly designed pharmacological profiles at the opioid receptor subtypes are long needed. Here a series of 17-N-substituted derivatives of normorphine and noroxymorphone analogues with five- and six-membered ring substituents have been synthesized for structure–activity study. Some compounds showed nanomolar affinity to MOR, DOR and KOR in in vitro competition binding experiments with selective agonists [3H]DAMGO, [3H]Ile5,6-deltorphin II and [3H]HS665, respectively. Pharmacological characterization of the compounds in G-protein signaling was determined by [35S]GTPγS binding assays. The normorphine analogues showed higher affinity to KOR compared to MOR and DOR, while most of the noroxymorphone derivatives did not bind to KOR. The presence of 14-OH substituent resulted in a shift in the pharmacological profiles in the agonist > partial agonist > antagonist direction compared to the parent compounds. A molecular docking-based in silico method was also applied to estimate the pharmacological profile of the compounds. Docking energies and the patterns of the interacting receptor atoms, obtained with experimentally determined active and inactive states of MOR, were used to explain the observed pharmacological features of the compounds.
