  • Guidelines for the use and interpretation of assays for monitoring autophagy
  • 2012
  • Ingår i: Autophagy. - : Informa UK Limited. - 1554-8635 .- 1554-8627. ; 8:4, s. 445-544
  • Forskningsöversikt (refereegranskat)abstract
    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Robust estimation of bacterial cell count from optical density
  • 2020
  • Ingår i: Communications Biology. - : Springer Science and Business Media LLC. - 2399-3642. ; 3:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • 2020
  • Ingår i: Military Medical Research. - : Springer Science and Business Media LLC. - 2054-9369. ; 7:1
  • Tidskriftsartikel (refereegranskat)abstract
    • The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of a rapidly spreading illness, coronavirus disease 2019 (COVID-19), affecting more than seventeen million people around the world. Diagnosis and treatment guidelines for clinicians caring for patients are needed. In the early stage, we have issued "A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus (2019-nCoV) infected pneumonia (standard version)"; now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline. We formed a working group of clinical experts and methodologists. The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas: chemoprophylaxis, diagnosis, treatments, and discharge management. We searched the literature for direct evidence on the management of COVID-19, and assessed its certainty generated recommendations using the Grading of Recommendations, Assessment, Development and Evaluation (GRADE) approach. Recommendations were either strong or weak, or in the form of ungraded consensus-based statement. Finally, we issued 34 statements. Among them, 6 were strong recommendations for, 14 were weak recommendations for, 3 were weak recommendations against and 11 were ungraded consensus-based statement. They covered topics of chemoprophylaxis (including agents and Traditional Chinese Medicine (TCM) agents), diagnosis (including clinical manifestations, reverse transcription-polymerase chain reaction (RT-PCR), respiratory tract specimens, IgM and IgG antibody tests, chest computed tomography, chest x-ray, and CT features of asymptomatic infections), treatments (including lopinavir-ritonavir, umifenovir, favipiravir, interferon, remdesivir, combination of antiviral drugs, hydroxychloroquine/chloroquine, interleukin-6 inhibitors, interleukin-1 inhibitors, glucocorticoid, qingfei paidu decoction, lianhua qingwen granules/capsules, convalescent plasma, lung transplantation, invasive or noninvasive ventilation, and extracorporeal membrane oxygenation (ECMO)), and discharge management (including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge). We also created two figures of these recommendations for the implementation purpose. We hope these recommendations can help support healthcare workers caring for COVID-19 patients.

  • Efficient Synthesis of Angular Selective Surfaces Based on Accurate Equivalent Circuit Analysis
  • 2023
  • Ingår i: IEEE Transactions on Microwave Theory and Techniques. - 0018-9480. ; 71:6, s. 2625-2638
  • Tidskriftsartikel (refereegranskat)abstract
    • A new efficient synthesis method of angular selective surfaces (ASSs) based on the accurate equivalent circuit model is proposed. The angular dispersion of different components under oblique incidence is modeled by virtual lumped elements in the angular domain. The processes of obtaining the equivalent circuit for substrates, electrically small grids and patch arrays are presented in detail. The synthesized circuits are simple and accurate. Full-wave simulations are carried out to obtain the angular dispersion of different structures, and the calculated results are in good agreement with those of the equivalent circuit. A third-order ASS with bandpass response and a second-order ASS with a tunable bandwidth are given as design examples. The implemented second-order ASS shows the 0.33 to 0.52 tunable bandwidth in the angular domain. The transmission loss is better than 1.2 dB when the incidence angle is 65°-77°. The simulation results and calculated results are in good agreement with the measured results.

  • Analysis of Heritability and Shared Heritability Based on Genome-Wide Association Studies for 13 Cancer Types
  • 2015
  • Ingår i: Journal of the National Cancer Institute. - : Oxford University Press (OUP). - 0027-8874 .- 1460-2105. ; 107:12
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: Studies of related individuals have consistently demonstrated notable familial aggregation of cancer. We aim to estimate the heritability and genetic correlation attributable to the additive effects of common single-nucleotide polymorphisms (SNPs) for cancer at 13 anatomical sites. Methods: Between 2007 and 2014, the US National Cancer Institute has generated data from genome-wide association studies (GWAS) for 49 492 cancer case patients and 34 131 control patients. We apply novel mixed model methodology (GCTA) to this GWAS data to estimate the heritability of individual cancers, as well as the proportion of heritability attributable to cigarette smoking in smoking-related cancers, and the genetic correlation between pairs of cancers. Results: GWAS heritability was statistically significant at nearly all sites, with the estimates of array-based heritability, h(l)(2), on the liability threshold (LT) scale ranging from 0.05 to 0.38. Estimating the combined heritability of multiple smoking characteristics, we calculate that at least 24% (95% confidence interval [CI] = 14% to 37%) and 7% (95% CI = 4% to 11%) of the heritability for lung and bladder cancer, respectively, can be attributed to genetic determinants of smoking. Most pairs of cancers studied did not show evidence of strong genetic correlation. We found only four pairs of cancers with marginally statistically significant correlations, specifically kidney and testes (rho = 0.73, SE = 0.28), diffuse large B-cell lymphoma (DLBCL) and pediatric osteosarcoma (rho = 0.53, SE = 0.21), DLBCL and chronic lymphocytic leukemia (CLL) (rho = 0.51, SE = 0.18), and bladder and lung (rho = 0.35, SE = 0.14). Correlation analysis also indicates that the genetic architecture of lung cancer differs between a smoking population of European ancestry and a nonsmoking Asian population, allowing for the possibility that the genetic etiology for the same disease can vary by population and environmental exposures. Conclusion: Our results provide important insights into the genetic architecture of cancers and suggest new avenues for investigation.

  • Characterizing prostate cancer risk through multi-ancestry genome-wide discovery of 187 novel risk variants
  • 2023
  • Ingår i: Nature Genetics. - : Springer Nature. - 1061-4036 .- 1546-1718. ; 55:12, s. 2065-2074
  • Tidskriftsartikel (refereegranskat)abstract
    • The transferability and clinical value of genetic risk scores (GRSs) across populations remain limited due to an imbalance in genetic studies across ancestrally diverse populations. Here we conducted a multi-ancestry genome-wide association study of 156,319 prostate cancer cases and 788,443 controls of European, African, Asian and Hispanic men, reflecting a 57% increase in the number of non-European cases over previous prostate cancer genome-wide association studies. We identified 187 novel risk variants for prostate cancer, increasing the total number of risk variants to 451. An externally replicated multi-ancestry GRS was associated with risk that ranged from 1.8 (per standard deviation) in African ancestry men to 2.2 in European ancestry men. The GRS was associated with a greater risk of aggressive versus non-aggressive disease in men of African ancestry (P = 0.03). Our study presents novel prostate cancer susceptibility loci and a GRS with effective risk stratification across ancestry groups.

  • Imputation and subset-based association analysis across different cancer types identifies multiple independent risk loci in the TERT-CLPTM1L region on chromosome 5p15.33
  • 2014
  • Ingår i: Human Molecular Genetics. - : Oxford University Press (OUP). - 0964-6906 .- 1460-2083. ; 23:24, s. 6616-6633
  • Tidskriftsartikel (refereegranskat)abstract
    • Genome-wide association studies (GWAS) have mapped risk alleles for at least 10 distinct cancers to a small region of 63 000 bp on chromosome 5p15.33. This region harbors the TERT and CLPTM1L genes; the former encodes the catalytic subunit of telomerase reverse transcriptase and the latter may play a role in apoptosis. To investigate further the genetic architecture of common susceptibility alleles in this region, we conducted an agnostic subset-based meta-analysis (association analysis based on subsets) across six distinct cancers in 34 248 cases and 45 036 controls. Based on sequential conditional analysis, we identified as many as six independent risk loci marked by common single-nucleotide polymorphisms: five in the TERT gene (Region 1: rs7726159, P = 2.10 × 10(-39); Region 3: rs2853677, P = 3.30 × 10(-36) and PConditional = 2.36 × 10(-8); Region 4: rs2736098, P = 3.87 × 10(-12) and PConditional = 5.19 × 10(-6), Region 5: rs13172201, P = 0.041 and PConditional = 2.04 × 10(-6); and Region 6: rs10069690, P = 7.49 × 10(-15) and PConditional = 5.35 × 10(-7)) and one in the neighboring CLPTM1L gene (Region 2: rs451360; P = 1.90 × 10(-18) and PConditional = 7.06 × 10(-16)). Between three and five cancers mapped to each independent locus with both risk-enhancing and protective effects. Allele-specific effects on DNA methylation were seen for a subset of risk loci, indicating that methylation and subsequent effects on gene expression may contribute to the biology of risk variants on 5p15.33. Our results provide strong support for extensive pleiotropy across this region of 5p15.33, to an extent not previously observed in other cancer susceptibility loci.

  • A global survey on the use of the international classification of diseases codes for metabolic dysfunction-associated fatty liver disease.
  • 2024
  • Ingår i: Hepatology international. - 1936-0541.
  • Tidskriftsartikel (refereegranskat)abstract
    • With the implementation of the 11th edition of the International Classification of Diseases (ICD-11) and the publication of the metabolic dysfunction-associated fatty liver disease (MAFLD) nomenclature in 2020, it is important to establish consensus for the coding of MAFLD in ICD-11. This will inform subsequent revisions of ICD-11.Using the Qualtrics XM and WJX platforms, questionnaires were sent online to MAFLD-ICD-11 coding collaborators, authors of papers, and relevant association members.A total of 890 international experts in various fields from 61 countries responded to the survey. We also achieved full coverage of provincial-level administrative regions in China. 77.1% of respondents agreed that MAFLD should be represented in ICD-11 by updating NAFLD, with no significant regional differences (77.3% in Asia and 76.6% in non-Asia, p=0.819). Over 80% of respondents agreed or somewhat agreed with the need to assign specific codes for progressive stages of MAFLD (i.e. steatohepatitis) (92.2%), MAFLD combined with comorbidities (84.1%), or MAFLD subtypes (i.e., lean, overweight/obese, and diabetic) (86.1%).This global survey by a collaborative panel of clinical, coding, health management and policy experts, indicates agreement that MAFLD should be coded in ICD-11. The data serves as a foundation for corresponding adjustments in the ICD-11 revision.

  • Clinical Characteristics of COVID-19 Patients With Gastrointestinal Symptoms : An Analysis of Seven Patients in China
  • 2020
  • Ingår i: Frontiers in Medicine. - : Frontiers Media SA. - 2296-858X. ; 7
  • Tidskriftsartikel (refereegranskat)abstract
    • Objectives: Patients with novel coronavirus disease 2019 (COVID-19) can present with gastrointestinal symptoms as their initial symptoms or as the main manifestations during disease progression, but the clinical characteristics of these patients are still unknown. Methods: We identified COVID-19 patients who admitted to Xiangyang No. 1 People's Hospital and presented with gastrointestinal symptoms as their initial or main symptoms. Their medical records were reviewed by two independent clinical scientists. The epidemiological and clinical characteristics as well as the clinical outcomes were analyzed. Results: Among 142 confirmed COVID-19 cases, 7 (4.9%) of them presented with gastrointestinal symptoms. Three patients had gastrointestinal symptoms as the initial symptoms and chief complaints, and 4 patients as the main symptoms during disease progression. Six patients had symptoms of diarrhea (3–16 days), 7 with anorexia (7–22 days), 6 with upper abdominal discomfort (1–7 days), and 4 with nausea (1–7 days), 1 with heartburn lasting 2 days, and 2 with vomiting symptoms (1 day). The chest CT scan showed typical COVID-19 imaging features, and associated with the progression of the disease. During treatment, 2 patients died due to organ failure. Discussion: COVID-19 patients with gastrointestinal symptoms are relatively rare and might be misdiagnosed. The clinical features include watery stools, anorexia, and upper abdominal discomfort. These patients may have severe disease and be associated with a poor prognosis. The underlying mechanisms of SARS-CoV-2 related gastrointestinal symptoms need to clarify in future studies.

  • Identification and analysis of functional elements in 1% of the human genome by the ENCODE pilot project
  • 2007
  • Ingår i: Nature. - : Springer Science and Business Media LLC. - 0028-0836 .- 1476-4687. ; 447:7146, s. 799-816
  • Tidskriftsartikel (refereegranskat)abstract
    • We report the generation and analysis of functional data from multiple, diverse experiments performed on a targeted 1% of the human genome as part of the pilot phase of the ENCODE Project. These data have been further integrated and augmented by a number of evolutionary and computational analyses. Together, our results advance the collective knowledge about human genome function in several major areas. First, our studies provide convincing evidence that the genome is pervasively transcribed, such that the majority of its bases can be found in primary transcripts, including non-protein-coding transcripts, and those that extensively overlap one another. Second, systematic examination of transcriptional regulation has yielded new understanding about transcription start sites, including their relationship to specific regulatory sequences and features of chromatin accessibility and histone modification. Third, a more sophisticated view of chromatin structure has emerged, including its inter-relationship with DNA replication and transcriptional regulation. Finally, integration of these new sources of information, in particular with respect to mammalian evolution based on inter- and intra-species sequence comparisons, has yielded new mechanistic and evolutionary insights concerning the functional landscape of the human genome. Together, these studies are defining a path for pursuit of a more comprehensive characterization of human genome function.

  • Ingår i: Nature. - : Springer Science and Business Media LLC. - 0028-0836 .- 1476-4687. ; 578:7793, s. 129-136
  • Tidskriftsartikel (refereegranskat)abstract
    • Transcript alterations often result from somatic changes in cancer genomes1. Various forms of RNA alterations have been described in cancer, including overexpression2, altered splicing3 and gene fusions4; however, it is difficult to attribute these to underlying genomic changes owing to heterogeneity among patients and tumour types, and the relatively small cohorts of patients for whom samples have been analysed by both transcriptome and whole-genome sequencing. Here we present, to our knowledge, the most comprehensive catalogue of cancer-associated gene alterations to date, obtained by characterizing tumour transcriptomes from 1,188 donors of the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA)5. Using matched whole-genome sequencing data, we associated several categories of RNA alterations with germline and somatic DNA alterations, and identified probable genetic mechanisms. Somatic copy-number alterations were the major drivers of variations in total gene and allele-specific expression. We identified 649 associations of somatic single-nucleotide variants with gene expression in cis, of which 68.4% involved associations with flanking non-coding regions of the gene. We found 1,900 splicing alterations associated with somatic mutations, including the formation of exons within introns in proximity to Alu elements. In addition, 82% of gene fusions were associated with structural variants, including 75 of a new class, termed 'bridged' fusions, in which a third genomic location bridges two genes. We observed transcriptomic alteration signatures that differ between cancer types and have associations with variations in DNA mutational signatures. This compendium of RNA alterations in the genomic context provides a rich resource for identifying genes and mechanisms that are functionally implicated in cancer.

  • Efficient Semitransparent Organic Solar Cells with Tunable Color enabled by an Ultralow-Bandgap Nonfullerene Acceptor
  • 2017
  • Ingår i: Advanced Materials. - : WILEY-V C H VERLAG GMBH. - 0935-9648 .- 1521-4095. ; 29:43
  • Tidskriftsartikel (refereegranskat)abstract
    • Semitransparent organic solar cells (OSCs) show attractive potential in power-generating windows. However, the development of semitransparent OSCs is lagging behind opaque OSCs. Here, an ultralow-bandgap non-fullerene acceptor, "IEICO-4Cl", is designed and synthesized, whose absorption spectrum is mainly located in the near-infrared region. When IEICO-4Cl is blended with different polymer donors (J52, PBDB-T, and PTB7-Th), the colors of the blend films can be tuned from purple to blue to cyan, respectively. Traditional OSCs with a nontransparent Al electrode fabricated by J52: IEICO-4Cl, PBDB-T: IEICO-4Cl, and PTB7-Th: IEICO-4Cl yield power conversion efficiencies (PCE) of 9.65 +/- 0.33%, 9.43 +/- 0.13%, and 10.0 +/- 0.2%, respectively. By using 15 nm Au as the electrode, semitransparent OSCs based on these three blends also show PCEs of 6.37%, 6.24%, and 6.97% with high average visible transmittance (AVT) of 35.1%, 35.7%, and 33.5%, respectively. Furthermore, via changing the thickness of Au in the OSCs, the relationship between the transmittance and efficiency is studied in detail, and an impressive PCE of 8.38% with an AVT of 25.7% is obtained, which is an outstanding value in the semitransparent OSCs.

  • Over 16% efficiency organic photovoltaic cells enabled by a chlorinated acceptor with increased open-circuit voltages
  • 2019
  • Ingår i: Nature Communications. - : NATURE PUBLISHING GROUP. - 2041-1723. ; 10
  • Tidskriftsartikel (refereegranskat)abstract
    • Broadening the optical absorption of organic photovoltaic (OPV) materials by enhancing the intramolecular push-pull effect is a general and effective method to improve the power conversion efficiencies of OPV cells. However, in terms of the electron acceptors, the most common molecular design strategy of halogenation usually results in down-shifted molecular energy levels, thereby leading to decreased open-circuit voltages in the devices. Herein, we report a chlorinated non-fullerene acceptor, which exhibits an extended optical absorption and meanwhile displays a higher voltage than its fluorinated counterpart in the devices. This unexpected phenomenon can be ascribed to the reduced non-radiative energy loss (0.206 eV). Due to the simultaneously improved short-circuit current density and open-circuit voltage, a high efficiency of 16.5% is achieved. This study demonstrates that finely tuning the OPV materials to reduce the bandgap-voltage offset has great potential for boosting the efficiency.

    • Optimizing the molecular structures of organic photovoltaic (OPV) materials is one of the most effective methods to boost power conversion efficiencies (PCEs). For an excellent molecular system with a certain conjugated skeleton, fine tuning the alky chains is of considerable significance to fully explore its photovoltaic potential. In this work, the optimization of alkyl chains is performed on a chlorinated nonfullerene acceptor (NFA) named BTP-4Cl-BO (a Y6 derivative) and very impressive photovoltaic parameters in OPV cells are obtained. To get more ordered intermolecular packing, the n-undecyl is shortened at the edge of BTP-eC11 to n-nonyl and n-heptyl. As a result, the NFAs of BTP-eC9 and BTP-eC7 are synthesized. The BTP-eC7 shows relatively poor solubility and thus limits its application in device fabrication. Fortunately, the BTP-eC9 possesses good solubility and, at the same time, enhanced electron transport property than BTP-eC11. Significantly, due to the simultaneously enhanced short-circuit current density and fill factor, the BTP-eC9-based single-junction OPV cells record a maximum PCE of 17.8% and get a certified value of 17.3%. These results demonstrate that minimizing the alkyl chains to get suitable solubility and enhanced intermolecular packing has a great potential in further improving its photovoltaic performance.

  • 2019
  • Ingår i: NATURE ENERGY. - : NATURE PUBLISHING GROUP. - 2058-7546. ; 4:9, s. 768-775
  • Tidskriftsartikel (refereegranskat)abstract
    • Organic photovoltaic cells are potential candidates to drive low power consumption off-grid electronics for indoor applications. However, their power conversion efficiency is still limited by relatively large losses in the open-circuit voltage and a non-optimal absorption spectrum for indoor illumination. Here, we carefully designed a non-fullerene acceptor named IO-4CI and blend it with a polymer donor named PBDB-TF to obtain a photoactive layer whose absorption spectrum matches that of indoor light sources. The photovoltaic characterizations reveal a low energy loss below 0.60 eV. As a result, the organic photovoltaic cell (1 cm(2)) shows a power conversion efficiency of 26.1% with an open-circuit voltage of 1.10 V under a light-emitting diode illumination of 1,000 lux (2,700 K). We also fabricated a large-area cell (4 cm(2)) through the blade-coating method. Our cell shows an excellent stability, maintaining its initial photovoltaic performance under continuous illumination of the indoor light source for 1,000 hours.

  • Konferensbidrag (refereegranskat)abstract
    • IPTV has been hailed as a “killer app'' of the Internet for a long time, but it has yet to really make a major breakthrough, due to various technical and political reasons. The recent influx of community-driven video distribution has re-actualized the research into efficient distribution methods for streaming video. In this paper, we suggest a number of modifications and extensions to the BitTorrent distribution and replication system to make it suitable for use in providing a streaming video delivery service, and implement parts of these in a simulator. Also, we report on a simulation study as well as a large-scale real-world study of the extensions on PlanetLab. Results show that BitTorrent can be used as a bandwidth-efficient alternative to traditional IPTV solutions.

It builds upon the QoE Provisioning-Delivery Hysteresis (PDH) and denotes the maximal throughput at which QoE deteriorations can be kept below a quantifiable level, which in turn allows to compare the EE of different video streaming solutions on QoE-fair grounds. In this work, we particularly focus on delivery problems stemming from outage-prone links, as they are typical for mobile systems. Well-adapted to the nature of the video-associated data streams and disturbances, a stochastic fluid flow model is used that allows for straightforward calculation of sustainable throughput values. We also discuss the application of the sustainable throughput for comparisons among different streaming solutions and their offered QoE and EE, respectively.

  • A new Software Framework for Heterogeneous Knowledge Sharing in Healthcare system
  • 2016
  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • Today’s demand for healthcare is dramatically increasing as the factor of the aging population and expectations growing during the past few years. This leads to the need of substantial healthcare services with innovative technologies developed from both industry and academia. Designing an efficient healthcare system is however a sophisticated process due to different research issues with the requirement for the provision of high-quality healthcare services. Connected to this requirement, the focus of many studies done so far is widely laid on a well-known problem called knowledge sharing. In recent years, knowledge sharing raises as one of the most demanding applications with references to the dynamic inter-activity among different healthcare actors and the complex data structures involved in this application. Suitable solution approach to knowledge sharing can enhance the efficiency of healthcare delivery, and thus improving the quality of healthcare services. The corresponding development tasks can be accomplished by using different methodologies such as analytical approaches, simulation experiments and practical measurements on the real healthcare system. In our work, the problem of heterogeneous knowledge sharing in the healthcare system is considered. Here, the heterogeneous aspect is expressed in terms ofdifferent healthcare actors and the associated characterizations. To do this, we suggest a new software framework, which mainly consists of three components. The first component is about the ontology based activity theory, which is used to scientifically represent the healthcare actors together with their relationships and interactions. The second component refers to an overlay decision maker, which is responsible for dealing with the decision-making activities such as appointment scheduling. Its advantage is to jointly consider various healthcare parameters and different algorithms for decision-making purposes. Based on these two components, the third component provides the theoretical models to conduct the numerical analysis and performance evaluation on the particular healthcare service.

  • eHASS : A Smart Appointment Scheduling System for eHealth
  • 2016
  • Konferensbidrag (refereegranskat)abstract
    • In the eHealth system, the appointment scheduling is an important task for the delivery of healthcare service among different healthcare actors. The key procedure is to do the decision making on the selection of suitable appointments between the care providers and the care receivers. The appointment decisionis a sophisticated problem in terms of how to efficiently deal with various parameters of involved healthcare actors. To solve this problem, we suggest a smart system called eHealth Appointment Scheduling System (eHASS). eHASS takes into account both heterogeneous aspects and interoperability requirements of eHealth system. As such, eHASS is capable of jointly considering various appointment characterizations and decision making algorithms for conducting appointment scheduling. The paper reports the eHASS architecture as well as the related work-in-progress.

    • Today, knowledge sharing raises as an important issue that challenges for the eHealth management system. It becomes one of the most demanding applications with references to the dynamic inter-activities among different health actors and the complex data structures involved in this application. In this paper, we suggest an activity theory based ontology model to scientifically represent various health actors in the eHealth system. The goal of the suggest model is to enhance the inter activities among these health actors for the efficient knowledge sharing purposes. We also develop a prototype software system based on the suggested ontology model. The survey results collected from the system users show the feasibility of the developed software system.

  • On Performance of Prioritized Appointment Scheduling for Healthcare
  • 2019
  • Ingår i: Journal of Service Science and Management. - : Scientific Research Publishing. - 1940-9893 .- 1940-9907. ; 12, s. 589-604
  • Tidskriftsartikel (refereegranskat)abstract
    • Designing the appointment scheduling is a challenging task for the development of healthcare system. The efficient solution approach can provide high-quality healthcare service between care providers (CP)s and care receivers (CR)s. In this paper, we consider the healthcare system with the heterogeneous CRs in terms of urgent and routine CRs. Our suggested model assumes that the system gives the service priority to the urgent CRs by allowing them to interrupt the ongoing routine appointments. An appointment handoff scheme is suggested for the interrupted routine appointments, and thus the routine CRs can attempt to re-establish the appointment scheduling with other available CPs. With these considerations, we study the scheduling performance of the system by using the Markov chains based modeling approach. The numerical analysis is reported and the simulation experiment is conducted to validate the numerical results.

  • 2021
  • Ingår i: Current Opinion in Chemical Engineering. - : IOP Publishing. - 2211-3398. ; 8:3
  • Tidskriftsartikel (refereegranskat)abstract
    • We report direct measurements via angle-resolved photoemission spectroscopy (ARPES) of the electronic dispersion of single-layer (SL) CoO2. The Fermi contour consists of a large hole pocket centered at the (Gamma) over bar point. To interpret the ARPES results, we use density functional theory (DFT) in combination with the multi-orbital Gutzwiller Approximation (DFT+GA), basing our calculations on crystalline structure parameters derived from x-ray photoelectron diffraction and low-energy electron diffraction. Our calculations are in good agreement with the measured dispersion. We conclude that the material is a moderately correlated metal. We also discuss substrate effects, and the influence of hydroxylation on the CoO2 SL electronic structure.

  • Power Consumption in Remote Gaming : an Empirical Evaluation
  • 2016
  • Ingår i: 2016 IEEE Conference on Computer Communications Workshops (INFOCOM WKSHPS). - : IEEE. - 9781467399555
  • Konferensbidrag (refereegranskat)abstract
    • The thin-client approach for gaming is becoming more popular. For instance, Nvidia Shield, Valve Steam and Shinra technologies have offerings based on the concept. In remote cloud gaming, the game is being executed and processed in the cloud while the user receives a video and audio stream of the game, in a very similar way as with remote desktop clients. At the same time, clouds are moving towards the end-users as "edge clouds" with different standardization bodies, such Open Mobile Edge Cloud and Open Fog Consortium, giving momentum for the efforts. Were remote gaming approaches to utilize edge clouds, the games could be played without installing any infrastructure at the homes of end-users while keeping network delays to the latency-sensitive games low. While waiting for such edge-cloud deployments to substantiate, even regional clouds could be utilized for the purpose. In such environments, remote cloud gaming can already now be utilized by game companies as an alternative to traditional download-and-install games in order to support, e.g., anti-piracy protection. While the incentives for the game companies are relatively clear, the end-user experience has been investigated mainly from the viewpoint of latency. In this paper, we fill a research gap related to energy efficiency by showing that mobile phone users can save between 12 and 32 % power by utilizing remote gaming instead of playing with a native app. Our prototype is based on GamingAnywhere open-source software for which we have also integrated a gamepad for easier controls. We show power measurements both with a 2D and 3D games, and also additional measurements with a smart TV-stick.

  • Inhibition of K-Ras4B-plasma membrane association with a membrane microdomain-targeting peptide
  • 2020
  • Ingår i: Chemical Science. - : Royal Society of Chemistry. - 2041-6520 .- 2041-6539. ; 11:3, s. 826-832
  • Tidskriftsartikel (refereegranskat)abstract
    • The association of K-Ras4B protein with plasma membrane (PM) is required for its signaling activity. Thus, direct inhibition of K-Ras4B-PM interaction could be a potential anti-Ras therapeutic strategy. However, it remains challenging to modulate such protein-PM interaction. Based on Ras isoform-specific PM microdomain localization patterns, we have developed a potent and isoform-selective peptide inhibitor, Memrasin, for detachment of K-Ras4B from the PM. Memrasin is one of the first direct inhibitors of K-Ras4B-PM interaction, and consists of a membrane l(d) region-binding sequence derived from the C-terminal region of K-Ras4B and an endosome-escape enhancing motif that can aggregate on membrane. It forms peptide-enriched domains in the l(d) region, abrogates the tethering of K-Ras4B to the PM and accordingly impairs Ras signaling activity, thereby efficiently decreasing the viability of several human lung cancer cells in a dose-responsive and K-Ras dependent manner. Memrasin provides a useful tool for exploring the biological function of K-Ras4B on or off the PM and a potential starting point for further development into anti-Ras therapeutics.

  • Stable and efficient hybrid Ag-In-S/ZnS@SiO2-carbon quantum dots nanocomposites for white light-emitting diodes
  • 2020
  • Ingår i: Chemical Engineering Journal. - : Elsevier. - 1385-8947 .- 1873-3212. ; 393
  • Tidskriftsartikel (refereegranskat)abstract
    • As a promising energy-saving technique, the eco-friendly and low-cost solid-state white light-emitting diodes (WLEDs) based on quantum dots (QDs) have been widely studied. Herein, a WLED device prepared by core-shell structure nanocomposites based on Ag-In-S/ZnS@SiO2 quantum dots (AIS@SiO2) and carbon quantum dots (CDs) was successfully constructed. CDs were combined onto the surface of AIS@SiO2 QDs to synthesize Ag-In-S/ZnS@SiO2-Carbon quantum dots (AIS-CDs) nanocomposites with a white-light emission, which successfully overcome the quenching effect of CDs induced by conventional aggregation. The as-prepared AIS-CDs nanocomposites presented high stability and a photoluminescence quantum yield (PLQY) of 35%. Moreover, the corresponding AIS-CDs nanocomposites-based WLEDs demonstrated the color coordinate of (0.32, 0.33), which is comparable to the pure white light (0.33, 0.33); furthermore, the luminous efficiency of the as-prepared WLEDs showed 15.1 lm W-1. These results reported herein may open up a new avenue for the development of high-performance, low-cost, and environmentally-friendly WLEDs.
