  • An anergic immune signature in the tumor microenvironment of classical Hodgkin lymphoma is associated with inferior outcome

    • Objective: The classical Hodgkin lymphoma (cHL) tumor microenvironment shows anongoing inflammatory response consisting of varying degrees of infiltrating eosinophils,mast cells, macrophages, regulatory T lymphocytes (Tregs), and activated lymphocytes surrounding the malignant cells. Herein, different immune signatures are characterized and correlated with treatment outcome.Methods: Tumor-infiltrating leukocytes were phenotyped in biopsies from 459 patients with cHL. Time to progression (TTP) (primary progression, relapse, or death from cHL) and overall survival were analyzed using Cox proportional hazards regression.Results: The leukocyte infiltration in the microenvironment was highly diverse between patients and was categorized in 4 immune signatures (active, anergic, innate, or mixed). A high proportion of Tregs (anergic) resulted in shorter TTP (median 12.9-year follow-up) in age-adjusted analyses (hazard ratio = 1.82; 95% confidence interval 1.05-3-15). Epstein-Barrvirus (EBV)-positive cases had higher proportions of macrophages and activated lymphocytes than EBV negative, but neither of those leukocytes predicted prognosis.Conclusions: Abundant Tregs (anergic signature) indicate a shorter TTP, particularly in younger patients. This is probably due to a reduced ability of the immune system to attack the tumor cells. Our data warrant further investigation if these suggested immune signatures could predict outcome of immunotherapy such as immune checkpoint inhibitors.

  • Effects of acute spinalization on neurons of postural networks

    • Postural limb reflexes (PLRs) represent a substantial component of postural corrections. Spinalization results in loss of postural functions, including disappearance of PLRs. The aim of the present study was to characterize the effects of acute spinalization on two populations of spinal neurons (F and E) mediating PLRs, which we characterized previously. For this purpose, in decerebrate rabbits spinalized at T12, responses of interneurons from L5 to stimulation causing PLRs before spinalization, were recorded. The results were compared to control data obtained in our previous study. We found that spinalization affected the distribution of F- and E-neurons across the spinal grey matter, caused a significant decrease in their activity, as well as disturbances in processing of posture-related sensory inputs. A two-fold decrease in the proportion of F-neurons in the intermediate grey matter was observed. Location of populations of F- and E-neurons exhibiting significant decrease in their activity was determined. A dramatic decrease of the efficacy of sensory input from the ipsilateral limb to F-neurons and from the contralateral limb to E-neurons was found. These changes in operation of postural networks underlie the loss of postural control after spinalization and represent a starting point for the development of spasticity.

  • The Prognostic Role of LRIG Proteins in Endometrial Cancer

    • Endometrial cancer (EC) is the most common gynecologic malignancy in Sweden and it has various prognostic factors. The LRIG family is a group of three integral surface proteins with a similar domain organization. The study aimed to explore LRIG family as prognostic factor proteins in EC. The initial study cohort included 100 women with EC who were treated at the Department of Women’s and Children’s Health, Karolinska University Hospital Solna, between 2007 and 2012. We assessed the associations between LRIG protein expression and type, grade, and stage of EC, as well as progression-free and overall survival. Immunohistochemistry results revealed that most women in the analytical sample had >50% LRIG1-, LRIG2- and LRIG3-positive cells. A statistically significant association was observed between having a high number of LRIG3-positive cells and superior overall survival (incidence rate ratio = 0.977; 95% confidence interval: 0.958–0.996, p = 0.019). Moreover, positive LRIG3 staining of the cell membrane was associated with reducing in the risk of death (hazard ratio = 0.23; 95% confidence interval: 0.09–0.57). Our results show that LRIG3 expression might be a prognostic factor in EC. The role of LRIG1 and LRIG2 expression remains to be further investigated.

  • Externa kostnader i transportscenarier med utökad användning av sjöfart

    • Studien i denna rapport ger faktamaterial som belyser hur godstrafikens externa kostnader skulle påverkas om dagens transportlösningar ersätts av lösningar med utökad användning av sjöfart. Två fall har studerats:Containerfallet avser en transport från Ostkusten/Stockholm till Hamburg. I Alternativ 1 antas transporten gå på järnväg till Göteborgs hamn och därifrån med ett containerfartyg till Hamburg och i Alternativ 2 med ett containerfartyg direkt till Hamburg.Trailerfallet avser en transport från Ostkusten/Stockholm till Travemünde. I Alternativ 1 antas transporten gå på väg till Trelleborgs hamn och därifrån med ropax-färjan till Travemünde och i Alternativ 2 med ett roro-fartyg direkt till Travemünde.Både i containerfallet och i trailerfallet beräknas de externa kostnaderna vara ungefär lika stora eller lägre för de direkta sjötransportalternativen än för de kombinerade land/sjö-alternativen. I containerfallet med ASEK-värderingar är jämförelsen ofullständig eftersom ASEK inte värderar knapphet på järnvägen. Det är möjligt att de externa kostnaderna för land/sjö-alternativet ökar signifikant om knappheten på järnväg värderas. Sjöfartens relativt låga internaliserande avgifter leder till högre externa kostnader för de direkta sjötransportalternativen än de kombinerade land/sjö-alternativen när skatter och avgifter räknas bort. Resultaten är robusta i trailerfallet (oberoende av värdering) och containerfallet (med EU:s värdering Ricardo dock inte med den svenska värderingen ASEK). I containerfallet med ASEK-värdering saknas de externa kostnaderna kopplade till knappheten på järnväg.

  • A Method to Test the Information Quality of Technical Documentation on Websites

    • In software engineering, testing is one of the corner-stones of quality assurance. The idea of software testing can be applied to information quality as well. Technical documentation has a set of intended uses that correspond to use cases in a software system. Documentation is, in many cases, presented via software systems, e.g., web servers and browsers, and contains software, e.g., Javascript for user interaction, animation, and customization, etc. This makes it difficult to find a clear-cut distinction between a software system and technical documentation. However, we can assume that each use case of a technical documentation involves retrieval of some sort of information that helps a user answer a specific questions. To assess information testing as a method, we implemented QAnalytics, a tool to assess the information quality of documentation that is provided by a website. The tool is web-based and allows test managers and site owners to define test cases and success criteria, disseminate the test cases to testers, and to analyze the test results. This way, information testing is easily manageable even for non-technical stakeholders. We applied our testing method and tool in a case study. According to common perception, the website of Linnaeus University needs to be re-engineered. Our method and tool helped the stakeholders identify what information is presented well and which parts of the website that need to change. The test results allowed the design and development effort to prioritize actual quality issues and potentially save time and resources.
