    • One of the main challenges of modern chemical industry is the inevitable transition to greener, more sustainable manufacturing processes that utilize raw materials more effectively, minimize waste streams, and avoid the use of toxic and hazardous materials. The development of new iron-based catalytic systems can eliminate the need for the currently widely used, costly and potentially toxic heavy metal catalysts in industrially relevant chemical reactions. Prominent examples of such processes include transition metal catalyzed cross-coupling reactions for formation of carbon-carbon (C -C) bonds. This minireview outlines recent developments in the area of iron-catalyzed cross-coupling chemistry during the last couple of decades, with special emphasis on the use of propargylic substrates and Grignard reagents as coupling partners.

  • Reindeer husbandry in peril?-How extractive industries exert multiple pressures on an Arctic pastoral ecosystem

    • Environmental changes and their consequences on biodiversity are known to have far-reaching effects on the resilience of animal populations and associated livelihoods around the world. To counteract negative demographic and economic effects on pastoralism, knowledge about the historical and current status of the environment is essential. In this study, we show how extractive industries, especially large-scale mining, induced a cascade of land conversions which are affecting animal populations and pastoralists' adaptive responses in northern Sweden. We examine social-ecological vulnerability in Arctic reindeer husbandry by integrating herders' knowledge, population statistics for semi-domesticated reindeer Rangifer t. tarandus, public data on socio-economic variables and geospatial tools. We determine that approximately 34% of Laevas reindeer herding community's grazing grounds are functionally unavailable to reindeer at present due to the accumulation of multiple competing land use pressures. Reindeer numbers currently only remain stable due to increased management efforts. Moreover, we identified current hotspots of high cumulative impact and mineral exploration as the spatially dominating land use factor in this area. Our approach and results provide new insights for scientifically robust cumulative impact assessments of anthropogenic stressors by creating a baseline of current developments via a combination of reindeer herder's knowledge with historical data of trends and extents of human activity over the last century.

    • Structural Maintenance of Chromosomes (SMC) proteins are vital for a wide range of processes including chromosome structure and dynamics, gene regulation and DNA repair. Eukaryotes have three SMC complexes, consisting of heterodimeric pairs of six different SMC proteins along with several specific regulatory subunits. In addition to their other functions, all three SMC complexes play distinct roles in DNA repair. Cohesin (SMC1-SMC3) is involved in DNA double-strand break repair, condensin (SMC2-SMC4) participates in single-strand break (SSB) repair, and the SMC5-SMC6 complex functions in various DNA repair pathways. SMC proteins consist of N- and C-terminal domains that fold back onto each other to create an ATPase ’head’ domain, connected to a central ’hinge’ domain via long coiled-coils. The hinge domain mediates dimerization of SMC proteins and binds DNA, but it is not clear to what purpose this activity serves. We studied the structure and function of the condensin hinge domain from mouse. While the SMC hinge domain structure is largely conserved from prokaryotes to eukaryotes, its function seems to have diversified throughout the course of evolution. The condensin hinge domain preferentially binds single-stranded DNA. We propose that this activity plays a role in the SSB repair function of the condensin complex.

    • Ultrasonic consolidation has been shown to be a viable metal-matrix-based smart composite additive layer manufacturing process. Yet, high quantity fibre integration has presented the requirement for a method of accurate positioning and fibre protection to maintain the fibre layout during ultrasonic consolidation. This study presents a novel approach for fibre integration during ultrasonic consolidation: channels are manufactured by laser processing on an ultrasonically consolidated sample. At the same time, controlled melt ejection is applied to aid accurate fibre placement and simultaneously reducing fibre damage occurrences. Microscopic, scanning electron microscopic and energy dispersive X-ray spectroscopic analyses are used for samples containing up to 10.5% fibres, one of the highest volumes in an ultrasonically consolidated composite so far. Up to 98% of the fibres remain in the channels after consolidation and fibre damage is reduced to less than 2% per sample. This study furthers the knowledge of high volume fibre embedment via ultrasonic consolidation for future smart material manufacturing. © Institution of Mechanical Engineers.

  • Comparing approaches for studying the effects of climate extremes : a case study of hospital admissions in Sweden during an extremely warm summer

    • Background: Health effects induced by climate, weather and climatic change may act directly or indirectly on human physiology. The future total burden of global warming is uncertain, but in some areas and for specific outcomes, mortality and morbidity are likely to increase. One likely effect of global warming is an increasing number of extreme weather events, such as floods, storms and heat waves. The excess numbers of specific health outcomes attributable to climate-induced events can be estimated. This paper compares approaches for estimating excess numbers of outcomes associated with climate extremes, exemplified by a case study of hospital admissions during the extremely warm summer of 2006 in southern Sweden.Materials and methods: Daily hospital admission data were obtained from the Swedish National Board of Health and Welfare for six hospitals in the Skåne region of southern Sweden for the period 1998 to 2006. Daily temperature data for the region were obtained from the meteorological station in the city of Malmö. We used four established approaches for estimating the daily excess numbers associated with extreme heat. Time series of daily event rates were assumed to follow a Poisson distribution. Excess event rates were compared by using several approaches, such as standardised event ratios and generalised additive models to estimate the health risks attributable to the extreme climate event.Results: The four approaches yielded vastly different results. The estimates of excess were considerably biased when not accounting for time trends in previous years’ data. Three of four approaches showed a significant increase in excess hospitalisation rates attributable to the heat episode in Skåne in 2006. However, modelling the effect of temperature failed to describe the risks induced by the extreme heat.Conclusion: Estimates of excess events depend greatly on the approach used. Further research is needed to identify which method yielded the most accurate estimates. However, one of the approaches used generally seem to perform better than the others in estimating the excess rates associated with the heat episode. Further on, estimating relative risks of temperature or other determinants of disease may fail to incorporate the unique characteristics of particular weather events, such as the effect caused by very persistent heat exposure. Unless this can be incorporated into predictive models, such models may be less appropriate to use when predicting the future burden of heat waves on human health.

  • Immune tolerance by interferon-alpha in experimental arthritis

    • Type I Interferons (mainly IFN-α & IFN-β) belong to a family of cytokines that possess strong antiviral and immunomodulatory properties. Pro- and/or anti-inflammatory effects of type I IFN have been observed in infectious diseases and several autoimmune diseases including SLE, MS, RA and experimental models thereof, but what defines either outcome is largely obscure. The main aim of this thesis is to understand how IFN-α may act anti-inflammatory in a model of antigen-induced arthritis (AIA). In this model, mice are sensitised with methylated-BSA (mBSA) emulsified in Freund’s adjuvant at day 1 and 7 followed by intra-articular injection of mBSA in the knee joint at day 21, which induces arthritis within 1 week.Administration of IFN-α at the time of mBSA sensitisations (day 1 and day 7) but not at induction of arthritis (day 21) clearly protected against arthritis in a type I IFN receptor dependent manner. Humoral immunity might not be involved in this protection as the levels of antigen-specific IgG (total, IgG1, IgG2a and IgG2b), IgA, IgE in serum were not altered in IFN-α treated mice. However, IFN-α-protection was accompanied by delayed and decreased antigen-specific proliferative responses in spleen and lymph node cells ex vivo, including impaired proliferative recall responses after intra-articular antigenic challenge.In the course of AIA, IFN-α inhibited the increase of circulatory IL-6, IL-10, IL-12, and TNF in the sensitisation phase (day 0-21) and also the re-call response of IL-1β, IL-10, IL-12, TNF, IFN-γ, and IL-17 induced by intra-articular mBSA challenge in arthritis phase (day 21-28). This IFN-α-inhibition of cytokines was also apparent in mBSA-re-stimulated spleen and lymph node cell cultures ex vivo, including inhibited cytokine production in CD4+ T helper cells and macrophages. In contrast to the inhibition of pro-inflammatory cytokines, the levels of immunomodulatory TGF-β was clearly enhanced in IFN-α-treated mice, both in serum and in re-stimulated leucocytes cultures including both macrophages, especially in the sensitisation phase, and in CD4+ T cells in the arthritis phase. By  inhibiting TGF-β signalling in vivo, the protective effect of IFN-α was  shown to be dependent on TGF-β signalling in the sensitisation phase.The cytokine TGF-β is an activator of the indoleamine 2,3 dioxygnese (IDO1), a potent immuneregulatory component that acts via enzymatic production of kynurenine (Kyn) and signalling activity. The IFN-α-protective effect in AIA was associated with both increased expression and enzymatic activity of IDO1 and the IFN-α-protection was totally ablated in mice lacking IDO1 expression (IDO1 KO mice) and in mice treated with the inhibitor of the enzymatic activity of IDO1 (1-Methyl Tryptophan; 1-MT). Interestingly, administration of the IDO-metabolite Kyn protected mice from AIA in an IFNARindependent manner. These observations show that the IDO1 enzymatic activity is important for the protective effect of IFN-α. Using 1-MT, it was further shown that the enzymatic activity of IDO1 was, like TGF-β, crucial only at the sensitisation but not in the arthritis phase of AIA for IFN-α to protect against arthritis. Instead, IDO1’s non-enzymatic signalling activity, characterized by sustained expression of IDO1 and non-canonical NF-κB activation in pDCs, was observed in the arthritis phase in spleen cells from mice treated with IFN-α.Regulatory T cells (Treg cells) were also found to be important for IFN-α-protection in AIA. Transient depletion of Treg cells by diphtheria toxin in DEREG mice in the arthritis phase, but not during the sensitisation phase abolished IFN-α-protection. Treatment with IFN-α enhanced the numbers of Treg cells in the course of AIA and their function; compared to untreated mice, Treg cells isolated at day 10 and 20 of AIA from IFN-α- treated mice exhibited higher suppressive activity against mBSA-stimulated proliferation of responder T cells. The enhancing effect of IFN-α on Treg cell numbers was observed in blood, spleen, LNs and also in ex-vivo cultures of leucocytes re-stimulated with mBSA and IFN-α. Although IFN-α clearly increased the suppressive activity of Treg cells, adoptive transfer of Treg cells from mBSA immunized mice, regardless of IFN-α treatment, prevented the development of arthritis.ConclusionIn the presence of IFN-α during antigen sensitisation, a state of tolerance is established, which is able to prevent joint inflammation induced by antigenic re-challenge. This immunological tolerance is created in the sensitisation phase of AIA and is characterized by inhibition of pro-inflammatory cytokines, increased TGF-β production and activity of the IDO1 enzyme, the latter two being indispensable for IFN-α-induced protection. Administration of Kyn, the metabolite of the enzymatic activity of IDO1, in the sensitisation phase also protected against AIA downstream of type I IFN signalling. In the arthritis phase regulatory T cells, whose numbers and suppressive capacity was clearly enhanced by IFN-α, mediate the actual prevention of arthritis development in IFN-α-treated animals. We have thus identified molecular and cellular components of the anti-inflammatory program elicited by IFN-α including Kyn that may not have the pro-inflammatory effects associated with IFN.

  • Integrating omics to characterize eco‐physiological adaptations : How moose diet and metabolism differ across biogeographic zones

    • 1. With accelerated land conversion and global heating at northern latitudes, it becomes crucial to understand, how life histories of animals in extreme environments adapt to these changes. Animals may either adapt by adjusting foraging behavior or through physiological responses, including adjusting their energy metabolism or both. Until now, it has been difficult to study such adaptations in free‐ranging animals due to methodological constraints that prevent extensive spatiotemporal coverage of ecological and physiological data.2. Through a novel approach of combining DNA‐metabarcoding and nuclear magnetic resonance (NMR)‐based metabolomics, we aim to elucidate the links between diets and metabolism in Scandinavian moose Alces alces over three biogeographic zones using a unique dataset of 265 marked individuals.3. Based on 17 diet items, we identified four different classes of diet types that match browse species availability in respective ecoregions in northern Sweden. Individuals in the boreal zone consumed predominantly pine and had the least diverse diets, while individuals with highest diet diversity occurred in the coastal areas. Males exhibited lower average diet diversity than females.4. We identified several molecular markers indicating metabolic constraints linked to diet constraints in terms of food availability during winter. While animals consuming pine had higher lipid, phospocholine, and glycerophosphocholine concentrations in their serum than other diet types, birch‐ and willow/aspen‐rich diets exhibit elevated concentrations of several amino acids. The individuals with highest diet diversity had increased levels of ketone bodies, indicating extensive periods of starvation for these individuals.5. Our results show how the adaptive capacity of moose at the eco‐physiological level varies over a large eco‐geographic scale and how it responds to land use pressures. In light of extensive ongoing climate and land use changes, these findings pave the way for future scenario building for animal adaptive capacity.
