    • Differences between sexes contribute to variation in the levels of fasting glucose and insulin. Epidemiological studies established a higher prevalence of impaired fasting glucose in men and impaired glucose tolerance in women, however, the genetic component underlying this phenomenon is not established. We assess sex-dimorphic (73,089/50,404 women and 67,506/47,806 men) and sex-combined (151,188/105,056 individuals) fasting glucose/fasting insulin genetic effects via genome-wide association study meta-analyses in individuals of European descent without diabetes. Here we report sex dimorphism in allelic effects on fasting insulin at IRS1 and ZNF12 loci, the latter showing higher RNA expression in whole blood in women compared to men. We also observe sex-homogeneous effects on fasting glucose at seven novel loci. Fasting insulin in women shows stronger genetic correlations than in men with waist-to-hip ratio and anorexia nervosa. Furthermore, waist-to-hip ratio is causally related to insulin resistance in women, but not in men. These results position dissection of metabolic and glycemic health sex dimorphism as a steppingstone for understanding differences in genetic effects between women and men in related phenotypes.

    • Background: Glycated hemoglobin (HbA1c) is used to diagnose type 2 diabetes (T2D) and assess glycemic control in patients with diabetes. Previous genome-wide association studies (GWAS) have identified 18 HbA1c-associated genetic variants. These variants proved to be classifiable by their likely biological action as erythrocytic (also associated with erythrocyte traits) or glycemic (associated with other glucose-related traits). In this study, we tested the hypotheses that, in a very large scale GWAS, we would identify more genetic variants associated with HbA1c and that HbA1c variants implicated in erythrocytic biology would affect the diagnostic accuracy of HbA1c. We therefore expanded the number of HbA1c-associated loci and tested the effect of genetic risk-scores comprised of erythrocytic or glycemic variants on incident diabetes prediction and on prevalent diabetes screening performance. Throughout this multiancestry study, we kept a focus on interancestry differences in HbA1c genetics performance that might influence race-ancestry differences in health outcomes.Methods & findings: Using genome-wide association meta-analyses in up to 159,940 individuals from 82 cohorts of European, African, East Asian, and South Asian ancestry, we identified 60 common genetic variants associated with HbA1c. We classified variants as implicated in glycemic, erythrocytic, or unclassified biology and tested whether additive genetic scores of erythrocytic variants (GS-E) or glycemic variants (GS-G) were associated with higher T2D incidence in multiethnic longitudinal cohorts (N = 33,241). Nineteen glycemic and 22 erythrocytic variants were associated with HbA1c at genome-wide significance. GS-G was associated with higher T2D risk (incidence OR = 1.05, 95% CI 1.04-1.06, per HbA1c-raising allele, p = 3 x 10-29); whereas GS-E was not (OR = 1.00, 95% CI 0.99-1.01, p = 0.60). In Europeans and Asians, erythrocytic variants in aggregate had only modest effects on the diagnostic accuracy of HbA1c. Yet, in African Americans, the X-linked G6PD G202A variant (T-allele frequency 11%) was associated with an absolute decrease in HbA1c of 0.81%-units (95% CI 0.66-0.96) per allele in hemizygous men, and 0.68%-units (95% CI 0.38-0.97) in homozygous women. The G6PD variant may cause approximately 2% (N = 0.65 million, 95% CI0.55-0.74) of African American adults with T2Dto remain undiagnosed when screened with HbA1c. Limitations include the smaller sample sizes for non-European ancestries and the inability to classify approximately one-third of the variants. Further studies in large multiethnic cohorts with HbA1c, glycemic, and erythrocytic traits are required to better determine the biological action of the unclassified variants.Conclusions: As G6PD deficiency can be clinically silent until illness strikes, we recommend investigation of the possible benefits of screening for the G6PD genotype along with using HbA1c to diagnose T2D in populations of African ancestry or groups where G6PD deficiency is common. Screening with direct glucose measurements, or genetically-informed HbA1c diagnostic thresholds in people with G6PD deficiency, may be required to avoid missed or delayed diagnoses.

  • Origin of the anomalous temperature dependence of coercivity in soft ferromagnets

    • We report on the origin of the anomalous temperature dependence of coercivity observed in some soft ferromagnets by studying the magnetic and electronic properties of FeZr films doped using ion implantation by H, He, B, C, and N. The anomalous increase of the coercivity with temperature was observed only in the C- and B-doped samples. Using x-ray photoelectron spectroscopy, we show that the anomalous behavior of the coercivity coincides with the occurrence of an electron charge transfer for those implanted samples. The origin of the anomaly is discussed in terms of (i) magnetic softness, (ii) nature of the Fe-C and -B covalent bonds, and (iii) large charge transfer.

    • Rapid changes in ocean circulation and climate have been observed in marine-sediment and ice cores over the last glacial period and deglaciation, highlighting the non-linear character of the climate system and underlining the possibility of rapid climate shifts in response to anthropogenic greenhouse gas forcing. To date, these rapid changes in climate and ocean circulation are still not fully explained. One obstacle hindering progress in our understanding of the interactions between past ocean circulation and climate changes is the difficulty of accurately dating marine cores. Here, we present a set of 92 marine sediment cores from the Atlantic Ocean for which we have established age-depth models that are consistent with the Greenland GICC05 ice core chronology, and computed the associated dating uncertainties, using a new deposition modeling technique. This is the first set of consistently dated marine sediment cores enabling paleoclimate scientists to evaluate leads/lags between circulation and climate changes over vast regions of the Atlantic Ocean. Moreover, this data set is of direct use in paleoclimate modeling studies.

    • After having emerged as primary contenders in the race for highly efficient optoelectronics materials, organolead halide perovskites (OHLP) are now being investigated in the nanoscale regime as promising building blocks with unique properties. For example, unlike their bulk counterpart, quantum dots of OHLP are brightly luminescent, owing to large exciton binding energies that cannot be rationalized solely on the basis of quantum confinement. Here, we establish the direct correlation between the structure and the electronic band-edge properties of CH3NH3PbBr3 nanoparticles. Complementary structural and spectroscopic measurements probing long-range and local order reveal that lattice strain influences the nature of the valence band and modifies the subtle stereochemical activity of the Pb2+ lone-pair. More generally, this work demonstrates that the stereochemical activity of the lone-pair at the metal site is a specific physicochemical parameter coupled to composition, size and strain, which can be employed to engineer novel functionalities in OHLP nanomaterials.

    • OBJECTIVE: To evaluate newly developed equations predicting relative glomerular filtration rate(GFR) in adult Swedish Caucasians and to compare with the Modification of Diet in Renal Disease(MDRD) and Mayo Clinic equations using enzymatic and zero-calibrated plasma creatinine assays. MATERIAL AND METHODS: GFR was measured with iohexol clearance adjusted to 1.73 m(2). One population sample (n=436/Lund) was used to derive an equation based on plasma-creatinine/age/gender, and a second with the addition of lean body mass (LBM). Both equations were validated in a separate sample (n=414/Malmö). The coefficients of the equations were eventually fine-tuned using all 850 patients and yielding Lund-Malmö equations without (LM) and with LBM-term (LM(LBM)).Their performance was compared with the MDRD(CC) (conventional creatinine calibration), MDRD(IDMS) (isotope dilution mass spectroscopy traceable calibration) and Mayo Clinic equations. RESULTS: The Lund equations performed similarly in both samples. In the combined set, the Mayo Clinic/MDRD(CC) resulted in +19.0/+10.2 % median bias, while bias for the other equations was < 10 %. LM(LBM) had the highest accuracy (86 % of estimates within 30 % of measured GFR), significantly (p < 0.001) better than for MDRD(IDMS) (80 %). In men with BMI < 20 kg/m(2), MDRD(IDMS)/LM had +46 %/+19 % median bias. MDRD(IDMS) also overestimated GFR by 22 %/14 % in men/women above 80 years of age. The LM(LBM) equation had < 10 % bias irrespective of BMI, age or GFR except for a 15 % negative bias at GFR > 90 mL/min/1.73 m(2). CONCLUSION: The newly developed Lund-Malmö equations for GFR estimation performed better than the MDRD(IDMS) and Mayo Clinic equations in a Swedish Caucasian sample. Inclusion of an LBM term improved performance markedly in certain subgroups.

  • A new algorithm for constructing optimal experimental designs

    • In any experimental science we are sometimes confronted with new experimental situations, where underlying physical models are unknown. After a first attempt at identifying physical variables that could effect the experimental result, we need to perform what is called a screening experiment. The objective is to conduct a small number of experiments that span as much of the experimental space as possible, to merit further experiments in smaller, more promising test systems. In this report we present an algorithm that, given a set of possible experiments, selects a subset of these. The selection is designed to cover as much variation as possible in the experimental space, and to assure a minimized expected mean squared error in the model parameter estimation. We verified the algorithm with an example taken from organic synthesis.

  • Final deglaciation of the Scandinavian Ice Sheet and implications for the Holocene global sea-level budget

    • The last deglaciation of the Scandinavian Ice Sheet (SIS) from similar to 21,000 to 13,000 yr ago is well constrained by several hundred Be-10 and C-14 ages. The subsequent retreat history, however, is established primarily from minimum-limiting C-14 ages and incomplete Baltic-Sea varve records, leaving a substantial fraction of final SIS retreat history poorly constrained. Here we develop a high-resolution chronology for the final deglaciation of the SIS based on 79 Be-10 cosmogenic exposure dates sampled along three transects spanning southern to northern Sweden and Finland. Combining this new chronology with existing Be-10 ages on deglaciation since the Last Glacial Maximum shows that rates of SIS margin retreat were strongly influenced by deglacial millennial-scale climate variability and its effect on surface mass balance, with regional modulation of retreat associated with dynamical controls. Ice-volume estimates constrained by our new chronology suggest that the SIS contributed 8 m sea-level equivalent to global sea-level rise between similar to 14.5 ka and 10 ka. Final deglaciation was largely complete by similar to 10.5 ka, with highest rates of sea-level rise occurring during the Bolling-Allerod, a 50% decrease during the Younger Dryas, and a rapid increase during the early Holocene. Combining our SIS volume estimates with estimated contributions from other remaining Northern Hemisphere ice sheets suggests that the Antarctic Ice Sheet (AIS) contributed 14.4 +/- 5.9 m to global sea-level rise since 13 ka. This new constraint supports those studies that indicate that an ice volume of 15 m or more of equivalent sea-level rise was lost from the AIS during the last deglaciation.

    • Intense bottom-ice algal blooms, often dominated by diatoms, are an important source of food for grazers, organic matter for export during sea ice melt, and dissolved organic carbon. Sea-ice diatoms have a number of adaptations, including accumulation of compatible solutes, that allows them to inhabit this highly variable environment characterized by extremes in temperature, salinity, and light. In addition to protecting them from extreme conditions, these compounds present a labile, nutrient-rich source of organic matter, and include precursors to climate active compounds (e.g., dimethyl sulfide [DMS]), which are likely regulated with environmental change. Here, intracellular concentrations of 45 metabolites were quantified in three sea-ice diatom species and were compared to two temperate diatom species, with a focus on compatible solutes and free amino acid pools. There was a large diversity of metabolite concentrations between diatoms with no clear pattern identifiable for sea-ice species. Concentrations of some compatible solutes (isethionic acid, homarine) approached 1 M in the sea-ice diatoms, Fragilariopsis cylindrus and Navicula cf. perminuta, but not in the larger sea-ice diatom, Nitzschia lecointei or in the temperate diatom species. The differential use of compatible solutes in sea-ice diatoms suggests different adaptive strategies and highlights which small organic compounds may be important in polar biogeochemical cycles.

  • Cardiovagal Function Measured by the Deep Breathing Test : Relationships With Coronary Atherosclerosis

    • Background The cardiovagal function can be assessed by quantification of respiratory sinus arrhythmia (RSA) during a deep breathing test. However, population studies of RSA and coronary atherosclerosis are lacking. This population-based study examined the relationship between RSA during deep breathing and coronary atherosclerosis, assessed by coronary artery calcium score (CACS). Methods and Results SCAPIS (Swedish Cardiopulmonary Bioimage Study) randomly invited men and women aged 50 to 64 years from the general population. CACS was obtained from computed tomography scanning, and deep breathing tests were performed in 4654 individuals. Expiration-inspiration differences (E-Is) of heart rates were calculated, and reduced RSA was defined as E-I in the lowest decile of the population. The relationship between reduced RSA and CACS (CACS≥100 or CACS≥300) was calculated using multivariable-adjusted logistic regression. The proportion of CACS≥100 was 24% in the lowest decile of E-I and 12% in individuals with E-I above the lowest decile (P<0.001), and the proportion of CACS≥300 was 12% and 4.8%, respectively (P<0.001). The adjusted odds ratio (OR) for CACS≥100 was 1.42 (95% CI, 1.10-1.84) and the adjusted OR for CACS≥300 was 1.62 (95% CI, 1.15-2.28), when comparing the lowest E-I decile with deciles 2 to 10. Adjusted ORs per 1 SD lower E-I were 1.17 (P=0.001) for CACS≥100 and 1.28 (P=0.001) for CACS≥300. Conclusions Low RSA during deep breathing is associated with increased coronary atherosclerosis as assessed by CACS, independently of traditional cardiovascular risk factors. Cardiovagal dysfunction could be a prevalent and modifiable risk factor for coronary atherosclerosis in the general population.

    • Over the past 3.5 million years, there have been several intervals when climate conditions were warmer than during the pre-industrial Holocene. Although past intervals of warming were forced differently than future anthropogenic change, such periods can provide insights into potential future climate impacts and ecosystem feedbacks, especially over centennial-to-millennial timescales that are often not covered by climate model simulations. Our observation-based synthesis of the understanding of past intervals with temperatures within the range of projected future warming suggests that there is a low risk of runaway greenhouse gas feedbacks for global warming of no more than 2 °C. However, substantial regional environmental impacts can occur. A global average warming of 1–2 °C with strong polar amplification has, in the past, been accompanied by significant shifts in climate zones and the spatial distribution of land and ocean ecosystems. Sustained warming at this level has also led to substantial reductions of the Greenland and Antarctic ice sheets, with sea-level increases of at least several metres on millennial timescales. Comparison of palaeo observations with climate model results suggests that, due to the lack of certain feedback processes, model-based climate projections may underestimate long-term warming in response to future radiative forcing by as much as a factor of two, and thus may also underestimate centennial-to-millennial-scale sea-level rise.

    • Objective. To evaluate the Cockcroft - Gault (CG) equation, using various body weight expressions, and the Sawyer equation in predicting glomerular filtration rate (GFR) using an enzymatic and zero-calibrated Jaffe plasma-creatinine assay, and to derive a new robust equation in adults. Material and methods. The CG weight measures included total, ideal and adjusted body weight (ABW; lowest of total and ideal) and two lean body mass (LBM) expressions, while the Sawyer equation is based primarily on LBM. Iohexol clearance was used to measure GFR. One derivation set (n = 436; enzymatic assay) was used to evaluate and bias-adjust existing equations when indicated, and to derive a new equation based on plasma-creatinine, age, gender and the body weight measure yielding the best adjusted R-2. All equations were then validated in a separate set (n = 414; Jaffe assay). Results. The existing equations all performed similarly in both sets. Prediction errors of equations based on LBM showed no correlation with BMI. The CG(ABW) and Sawyer equations performed best. The new equation with LBM yielded the highest adjusted R-2. In the combined set (n = 850), its accuracy (86%/98% of estimates within 30%/50% of measured GFR) was significantly better than for the CGABW (79%/95%) and Sawyer equations (79%/93%) (p < 0.001) for each 30 mL/min GFR subgroup within +/- 30% and +/- 50%, except within +/- 30% > 120 mL/min. Prediction error did not correlate with BMI, age or gender. Conclusion. A new creatinine-based equation derived in a mainly Caucasian patient sample is a better predictor of GFR than CG-type equations irrespective of the body weight measure used or, if bias-adjusted, when using zero-calibrated creatinine assays.

    • Two interglacial epochs are included in the suite of Paleoclimate Modeling Intercomparison Project (PMIP4) simulations in the Coupled Model Intercomparison Project (CMIP6). The experimental protocols for simulations of the mid-Holocene (midHolocene, 6000 years before present) and the Last Interglacial (lig127k, 127 000 years before present) are described here. These equilibrium simulations are designed to examine the impact of changes in orbital forcing at times when atmospheric greenhouse gas levels were similar to those of the preindustrial period and the continental configurations were almost identical to modern ones. These simulations test our understanding of the interplay between radiative forcing and atmospheric circulation, and the connections among large-scale and regional climate changes giving rise to phenomena such as land-sea contrast and high-latitude amplification in temperature changes, and responses of the monsoons, as compared to today. They also provide an opportunity, through carefully designed additional sensitivity experiments, to quantify the strength of atmosphere, ocean, cryosphere, and land-surface feedbacks. Sensitivity experiments are proposed to investigate the role of freshwater forcing in triggering abrupt climate changes within interglacial epochs. These feedback experiments naturally lead to a focus on climate evolution during interglacial periods, which will be examined through transient experiments. Analyses of the sensitivity simulations will also focus on interactions between extratropical and tropical circulation, and the relationship between changes in mean climate state and climate variability on annual to multi-decadal timescales. The comparative abundance of paleoenvironmental data and of quantitative climate reconstructions for the Holocene and Last Interglacial make these two epochs ideal candidates for systematic evaluation of model performance, and such comparisons will shed new light on the importance of external feedbacks (e.g., vegetation, dust) and the ability of state-of-the-art models to simulate climate changes realistically.

    • In this work an ultrasound-based measurement method is used formonitoring suspension velocity and build-up of magnetic material inside awet Low-Intensity Magnetic Separator, a process used e.g. inbeneficiation of magnetite ores. Today the only available option is tomonitor material transport between unit operations; i.e. flow rate,solids concentration, and particle size distribution of suspension flowin pipes are measured online using standard equipment.An Acoustic Backscatter System is fitted to the tank of a separator, andused to monitor the internal flow. A method called Ultrasonic VelocityProfiling is used to capture internal velocity profiles. Simultaneously,the backscatter signal intensity is used to get indications about localsolids concentration of the flow, and build-up of magnetic material. Themethods are evaluated in realistic conditions, where the effect ofvarying factors relevant to machine performance is investigated. Theincluded factors are; the slurry feed rate, the slurry solidsconcentration, the magnet assembly angle, and the drum rotational speed.The presented method gives useful information about the internal materialflow inside the separator. The velocity measurements capture the,sometimes complex, internal flow patterns, for example the presence andvelocity of a recirculating flow in the dewatering zone. Additionally,keeping a balanced material loading in the concentrate dewatering zone isimportant to separator performance. Using the signal backscatterintensity it is possible to qualitatively monitor this material loading.Generally these direct measurements can aid in improvements to machinedesign, process optimization, and process control.

    • The internal material transport and selection processes of the wet low-intensity magnetic separators (LIMS) are poorly understood; this calls for improved measurement techniques. In this work an ultrasonic velocity profiling (UVP) technique for measuring how material flow velocity varies with penetration depth is presented. A measurement depth of just a couple of centimetres would greatly improve the understanding of the separation process in a LIMS.When applied to flows of mineral suspensions with high volumetric solids concentration, similar to those in the separators, UVP is unique in combining:•Non-intrusive measurements.•Operates using just one sensor element (transducer).•Relatively good spatial resolution.•Penetrates opaque suspensions.•Fast sampling rate.Here, flows are studied in a rectangular duct (50 × 75 mm). Using magnetite suspensions, measurement through the whole depth of 50 mm is made with good accuracy. Velocity profiles are presented for solids concentrations of 5% and 9% solids by volume (20% and 36% by weight). Even at 9 vol% solids it is possible to reach a penetration depth of more than 25 mm.

    • In the mining industry magnetite particles are transported in aqueous suspension through different stages of the process. In some stages it is of interest to monitor both the concentration and particle velocity profiles over a cross-section of the flow. In this paper an ultrasonic flow meter method based on cross-correlation of backscattered sound is presented. High solid particle content (e.g. 20-40 wt%) makes this challenging and therefore the emphasis lies on developing robust signal processing techniques for particle velocity estimation. The developed method is evaluated in laboratory experiments on flows in a rectangular duct. Transducers with centre frequency 1, 2, 4 and 5 MHz are evaluated in flows with a solid content of 20% by weight (4.7 vol% solids). Using the 2 MHz transducer a penetration depth of 50 mm is reached.

    • In the mining industry, magnetite particles are transported in suspensions with water through different stages of the process. In some of these stages, it is of interest to monitor both the concentration and particle velocity over a cross-section of the flow. High particle concentration makes development of flow measurement techniques challenging. An additional challenge is that the flow is often accessible from one side only, which further limits the selection of applicable techniques. Previous work by the authors focused on using pulse-echo ultrasound for flow velocity profile estimation. In this paper the same setup is used to simultaneously study local variations in solids concentration. Ultrasound pulses are transmitted into the suspension, and the resulting backscatter is recorded. The statistics of the backscatter depend on solids concentration, particle size distribution, particle density, etc. We demonstrate how a short-time (windowed) Power Spectral Density (PSD) estimate can be used to obtain qualitative information about local solids concentration variations. For demonstration, a magnetite suspension carrying up to 7.5 vol% particles (29 wt%, mean particle size 34 μm) is pumped through a closed rectangular channel. When the pump is stopped, pulse-echo ultrasound (with a center frequency of 2.25 or 3.5 MHz) is used to monitor the sedimentation process. Nine snapshots of the process are included. These show a time lapse of the sedimentation, with 5 s between each image. It is clear that the short-time PSD is a good indicator of local mass fraction variations.

    • We use quantum Monte Carlo simulations to study effects of disorder on the quantum phase transition occurring versus the ratio g = J/J' in square-lattice dimerized S = 1/2 Heisenberg antiferromagnets with intradimer and interdimer couplings J and J'. The dimers are either randomly distributed (as in the classical dimer model), or come in parallel pairs with horizontal or vertical orientation. In both cases the transition violates the Harris criterion, according to which the correlation-length exponent should satisfy v >= 1. We do not detect any deviations from the three-dimensional O(3) universality class obtaining in the absence of disorder (where v approximate to 0.71). We discuss special circumstances which allow v < 1 for the type of disorder considered here.
