  • Outcomes after perioperative SARS-CoV-2 infection in patients with proximal femoral fractures: an international cohort study

    • Studies have demonstrated high rates of mortality in people with proximal femoral fracture and SARS-CoV-2, but there is limited published data on the factors that influence mortality for clinicians to make informed treatment decisions. This study aims to report the 30-day mortality associated with perioperative infection of patients undergoing surgery for proximal femoral fractures and to examine the factors that influence mortality in a multivariate analysis.SettingProspective, international, multicentre, observational cohort study.ParticipantsPatients undergoing any operation for a proximal femoral fracture from 1 February to 30 April 2020 and with perioperative SARS-CoV-2 infection (either 7 days prior or 30-day postoperative).Primary outcome30-day mortality. Multivariate modelling was performed to identify factors associated with 30-day mortality.ResultsThis study reports included 1063 patients from 174 hospitals in 19 countries. Overall 30-day mortality was 29.4% (313/1063). In an adjusted model, 30-day mortality was associated with male gender (OR 2.29, 95% CI 1.68 to 3.13, p<0.001), age >80 years (OR 1.60, 95% CI 1.1 to 2.31, p=0.013), preoperative diagnosis of dementia (OR 1.57, 95% CI 1.15 to 2.16, p=0.005), kidney disease (OR 1.73, 95% CI 1.18 to 2.55, p=0.005) and congestive heart failure (OR 1.62, 95% CI 1.06 to 2.48, p=0.025). Mortality at 30 days was lower in patients with a preoperative diagnosis of SARS-CoV-2 (OR 0.6, 95% CI 0.6 (0.42 to 0.85), p=0.004). There was no difference in mortality in patients with an increase to delay in surgery (p=0.220) or type of anaesthetic given (p=0.787).ConclusionsPatients undergoing surgery for a proximal femoral fracture with a perioperative infection of SARS-CoV-2 have a high rate of mortality. This study would support the need for providing these patients with individualised medical and anaesthetic care, including medical optimisation before theatre. Careful preoperative counselling is needed for those with a proximal femoral fracture and SARS-CoV-2, especially those in the highest risk groups.Trial registration numberNCT04323644

  • A control oriented strategy of disruption prediction to avoid the configuration collapse of tokamak reactors

    • The objective of thermonuclear fusion consists of producing electricity from the coalescence of light nuclei in high temperature plasmas. The most promising route to fusion envisages the confinement of such plasmas with magnetic fields, whose most studied configuration is the tokamak. Disruptions are catastrophic collapses affecting all tokamak devices and one of the main potential showstoppers on the route to a commercial reactor. In this work we report how, deploying innovative analysis methods on thousands of JET experiments covering the isotopic compositions from hydrogen to full tritium and including the major D-T campaign, the nature of the various forms of collapse is investigated in all phases of the discharges. An original approach to proximity detection has been developed, which allows determining both the probability of and the time interval remaining before an incoming disruption, with adaptive, from scratch, real time compatible techniques. The results indicate that physics based prediction and control tools can be developed, to deploy realistic strategies of disruption avoidance and prevention, meeting the requirements of the next generation of devices.

    • The DO experiment enjoyed a very successful data-collection run at the Fermilab Tevatron collider between 1992 and 1996. Since then, the detector has been upgraded to take advantage of improvements to the Tevatron and to enhance its physics capabilities. We describe the new elements of the detector, including the silicon microstrip tracker, central fiber tracker, solenoidal magnet, preshower detectors, forward muon detector, and forward proton detector. The uranium/liquid -argon calorimeters and central muon detector, remaining from Run 1, are discussed briefly. We also present the associated electronics, triggering, and data acquisition systems, along with the design and implementation of software specific to DO.

  • An open-access database and analysis tool for perovskite solar cells based on the FAIR data principles

    • Large datasets are now ubiquitous as technology enables higher-throughput experiments, but rarely can a research field truly benefit from the research data generated due to inconsistent formatting, undocumented storage or improper dissemination. Here we extract all the meaningful device data from peer-reviewed papers on metal-halide perovskite solar cells published so far and make them available in a database. We collect data from over 42,400 photovoltaic devices with up to 100 parameters per device. We then develop open-source and accessible procedures to analyse the data, providing examples of insights that can be gleaned from the analysis of a large dataset. The database, graphics and analysis tools are made available to the community and will continue to evolve as an open-source initiative. This approach of extensively capturing the progress of an entire field, including sorting, interactive exploration and graphical representation of the data, will be applicable to many fields in materials science, engineering and biosciences.

  • A reporting format for leaf-level gas exchange data and metadata

    • Leaf-level gas exchange data support the mechanistic understanding of plant fluxes of carbon and water. These fluxes inform our understanding of ecosystem function, are an important constraint on parameterization of terrestrial biosphere models, are necessary to understand the response of plants to global environmental change, and are integral to efforts to improve crop production. Collection of these data using gas analyzers can be both technically challenging and time consuming, and individual studies generally focus on a small range of species, restricted time periods, or limited geographic regions. The high value of these data is exemplified by the many publications that reuse and synthesize gas exchange data, however the lack of metadata and data reporting conventions make full and efficient use of these data difficult. Here we propose a reporting format for leaf-level gas exchange data and metadata to provide guidance to data contributors on how to store data in repositories to maximize their discoverability, facilitate their efficient reuse, and add value to individual datasets. For data users, the reporting format will better allow data repositories to optimize data search and extraction, and more readily integrate similar data into harmonized synthesis products. The reporting format specifies data table variable naming and unit conventions, as well as metadata characterizing experimental conditions and protocols. For common data types that were the focus of this initial version of the reporting format, i.e., survey measurements, dark respiration, carbon dioxide and light response curves, and parameters derived from those measurements, we took a further step of defining required additional data and metadata that would maximize the potential reuse of those data types. To aid data contributors and the development of data ingest tools by data repositories we provided a translation table comparing the outputs of common gas exchange instruments. Extensive consultation with data collectors, data users, instrument manufacturers, and data scientists was undertaken in order to ensure that the reporting format met community needs. The reporting format presented here is intended to form a foundation for future development that will incorporate additional data types and variables as gas exchange systems and measurement approaches advance in the future. The reporting format is published in the U.S. Department of Energy?s ESS-DIVE data repository, with documentation and future development efforts being maintained in a version control system.

  • Testing Syndromes of Psychopathology in Parent and Youth Ratings Across Societies

    • As societies become increasingly diverse, mental health professionals need instruments for assessing emotional, behavioral, and social problems in terms of constructs that are supported within and across societies. Building on decades of research findings, multisample alignment confirmatory factor analyses tested an empirically based 8-syndrome model on parent ratings across 30 societies and youth self-ratings across 19 societies. The Child Behavior Checklist for Ages 6–18 and Youth Self-Report for Ages 11–18 were used to measure syndromes descriptively designated as Anxious/ Depressed, Withdrawn/Depressed, Somatic Complaints, Social Problems, Thought Problems, Attention Problems, Rule-Breaking Behavior, and Aggressive Behavior. For both parent ratings (N = 61,703) and self-ratings (N = 29,486), results supported aggregation of problem items into 8 first-order syndromes for all societies (configural invariance), plus the invariance of item loadings (metric invariance) across the majority of societies. Supported across many societies in both parent and self-ratings, the 8 syndromes offer a parsimonious phenotypic taxonomy with clearly operatio- nalized assessment criteria. Mental health professionals in many societies can use the 8 syndromes to assess children and youths for clinical, training, and scientific purposes.

  • Preliminary interpretation of the REMS pressure data from the first 100 sols of the MSL mission

    • We provide a preliminary interpretation of the Rover Environmental Monitoring Station (REMS) pressure data from the first 100 Martian solar days (sols) of the Mars Science Laboratory mission. The pressure sensor is performing well and has revealed the existence of phenomena undetected by previous missions that include possible gravity waves excited by evening downslope flows, relatively dust-free convective vortices analogous in structure to dust devils, and signatures indicative of the circulation induced by Gale Crater and its central mound. Other more familiar phenomena are also present including the thermal tides, generated by daily insolation variations, and the CO2 cycle, driven by the condensation and sublimation of CO2 in the polar regions. The amplitude of the thermal tides is several times larger than those seen by other landers primarily because Curiosity is located where eastward and westward tidal modes constructively interfere and also because the crater circulation amplifies the tides to some extent. During the first 100 sols tidal amplitudes generally decline, which we attribute to the waning influence of the Kelvin wave. Toward the end of the 100 sol period, tidal amplitudes abruptly increased in response to a nearby regional dust storm that did not expand to global scales. Tidal phases changed abruptly during the onset of this storm suggesting a change in the interaction between eastward and westward modes. When compared to Viking Lander 2 data, the REMS daily average pressures show no evidence yet for the 1-20 Pa increase expected from the possible loss of CO 2 from the south polar residual cap.   • Secular Climate Change on Mars : An Update Using MSL Pressure Data

    • The South Polar Residual Cap (SPRC) on Mars is an icy reservoir of CO2. If all the CO2 trapped in the SPRC were released to the atmosphere the mean annual global surface pressure would rise by ~20 Pa. Repeated MOC and HiRISE imaging of scarp retreat rates within the SPRC have led to the suggestion that the SPRC is losing mass. Estimates for the loss rate vary between 0. 5 Pa per Mars Decade to 13 Pa per Mars Decade. Assuming 80% of this loss goes directly into the atmosphere, and that the loss is monotonic, the global annual mean surface pressure should have increased between ~1-20 Pa since the Viking mission (19 Mars years ago). Surface pressure measurements by the Phoenix Lander only 2 Mars years ago were found to be consistent with these loss rates. Here we compare surface pressure data from the MSL mission with that from Viking Lander 2 (VL-2) to determine if the trend continues. We use VL-2 because it is at the same elevation as MSL (-4500 m). However, based on the first 100 sols of data there does not appear to be a significant difference between the dynamically adjusted pressures of the two landers. This result implies one of several possibilities: (1) the cap is not losing mass and the difference between the Viking and Phoenix results is due to uncertainties in the measurements; (2) the cap has lost mass between the Viking and Phoenix missions but it has since gone back to the cap or into the regolith; or (3) that our analysis is flawed. The first possibility is real since post-mission analysis of the Phoenix sensor has shown that there is a 3 (±2) Pa offset in the data and there may also be uncertainties in the Viking data. The loss/gain scenario for the cap seems unlikely since scarps continue retreating, and regolith uptake implies something unique about the past several Mars years. That our analysis is flawed is certainly possible owing to the very different environments of the Viking and MSL landers. MSL is at the bottom of a deep crater in the southern tropics (~5°S), whereas VL-2 is at a high latitude (~48°N) in the northern plains. And in spite of the fact that the two landers are at nearly identical elevations, they are in very different thermal environments (e.g., MSL is warm when VL-2 is cold), which can have a significant affect on pressures. For these reasons, our confidence in the comparison will increase as more MSL data become available. We will report the results up through sol 360 at the meeting.

    • REMS-P, the pressure measurement subsystem of the Mars Science Laboratory (MSL) Rover Environmental Measurement Station (REMS), is performing accurate observations of the Martian atmospheric surface pressure. It has demonstrated high data quality and good temporal coverage, carrying out the first in situ pressure observations in the Martian equatorial regions. We describe the REMS-P initial results by MSL mission sol 100 including the instrument performance and data quality and illustrate some initial interpretations of the observed features. The observations show both expected and new phenomena at various spatial and temporal scales, e.g., the gradually increasing pressure due to the advancing Martian season signals from the diurnal tides as well as various local atmospheric phenomena and thermal vortices. Among the unexpected new phenomena discovered in the pressure data are a small regular pressure drop at every sol and pressure oscillations occurring in the early evening. We look forward to continued high-quality observations by REMS-P, extending the data set to reveal characteristics of seasonal variations and improved insights into regional and local phenomena.     • Mars Science Laboratory Curiosity rover observations of the 2018/Mars year 34 global/planet‐encircling dust storm represent the first in situ measurements of a global dust storm with dedicated meteorological sensors since the Viking Landers. The Mars Science Laboratory team planned and executed a science campaign lasting approximately 100 Martian sols to study the storm involving an enhanced cadence of environmental monitoring using the rover's meteorological sensors, cameras, and spectrometers. Mast Camera 880‐nm optical depth reached 8.5, and Rover Environmental Monitoring Station measurements indicated a 97% reduction in incident total ultraviolet solar radiation at the surface, 30K reduction in diurnal range of air temperature, and an increase in the semidiurnal pressure tide amplitude to 40 Pa. No active dust‐lifting sites were detected within Gale Crater, and global and local atmospheric dynamics were drastically altered during the storm. This work presents an overview of the mission's storm observations and initial results.

    • Resulting from impaired collagen turnover, fibrosis is a hallmark of adipose tissue (AT) dysfunction and obesity-associated insulin resistance (IR). Prolidase, also known as peptidase D (PEPD), plays a vital role in collagen turnover by degrading proline-containing dipeptides but its specific functional relevance in AT is unknown. Here we show that in human and mouse obesity, PEPD expression and activity decrease in AT, and PEPD is released into the systemic circulation, which promotes fibrosis and AT IR. Loss of the enzymatic function of PEPD by genetic ablation or pharmacological inhibition causes AT fibrosis in mice. In addition to its intracellular enzymatic role, secreted extracellular PEPD protein enhances macrophage and adipocyte fibro-inflammatory responses via EGFR signalling, thereby promoting AT fibrosis and IR. We further show that decreased prolidase activity is coupled with increased systemic levels of PEPD that act as a pathogenic trigger of AT fibrosis and IR. Thus, PEPD produced by macrophages might serve as a biomarker of AT fibro-inflammation and could represent a therapeutic target for AT fibrosis and obesity-associated IR and type 2 diabetes. Obesity-associated AT fibro-inflammation and metabolic disturbances are linked to PEPD activity and PEPD extracellular levels.

  • Environmental Dynamics and the Habitability Potential at Gale Crater, Mars

    • The assessment of environmental habitability potential involves measurement of the chemical and physical attributes of the system as well as their dynamic interplay. The environmental dynamics describe the availability of both energy sources and raw materials for meeting the requirements of organisms and for altering the environment. Energetic exchange can also determine the preservation potential for organic materials in the rock record. During its first year at Gale Crater, the Mars Science Laboratory payload has directly measured the chemistry and physical attributes, e.g., temperature, humidity, radiation, pressure, etc. of the martian atmosphere. Curiosity has also acquired chemical and mineralogical data, both from a wind drift deposit of fines and from two examples of a sedimentary rock formation in a region of Gale Crater called Yellowknife Bay, some 445 meters to the east of Bradbury Landing, where Curiosity initially touched down. These data enabled inferences to be made regarding depositional environment and past habitability potential at Gale Crater. The rock chemistry data reveal signs of aqueous interaction i.e., H2O, OH and H2 and sufficient elemental basis (C, H, O, S and possibly N) for plausible nutrient supply, should Mars have ever had autotrophic prokaryotes to exploit it, and a range of redox conditions tolerable to Earth microbes is indicated by the presence of clay minerals. Curiosity’s observations of the chemical, physical and geologic features of Yellowknife Bay point to a formerly habitable environment.

  • Mars Science Laboratory relative humidity observations : Initial results

    • The Mars Science Laboratory (MSL) made a successful landing at Gale crater early August 2012. MSL has an environmental instrument package called the Rover Environmental Monitoring Station (REMS) as a part of its scientific payload. REMS comprises instrumentation for the observation of atmospheric pressure, temperature of the air, ground temperature, wind speed and direction, relative humidity (REMS-H), and UV measurements. We concentrate on describing the REMS-H measurement performance and initial observations during the first 100 MSL sols as well as constraining the REMS-H results by comparing them with earlier observations and modeling results. The REMS-H device is based on polymeric capacitive humidity sensors developed by Vaisala Inc., and it makes use of transducer electronics section placed in the vicinity of the three humidity sensor heads. The humidity device is mounted on the REMS boom providing ventilation with the ambient atmosphere through a filter protecting the device from airborne dust. The final relative humidity results appear to be convincing and are aligned with earlier indirect observations of the total atmospheric precipitable water content. The water mixing ratio in the atmospheric surface layer appears to vary between 30 and 75 ppm. When assuming uniform mixing, the precipitable water content of the atmosphere is ranging from a few to six precipitable micrometers.

    • The photoionization of Cl+ leading to Cl2+ was measured in the photon energy range of 19.5-28.0 eV. A spectrum with a photon energy resolution of 15 meV normalized to absolute cross-section measurements is presented. The measurements were carried out by merging a Cl+ ion beam with a photon beam of highly monochromatic synchrotron radiation at the Advanced Light Source at Lawrence Berkeley National Laboratory. The measured photoionization of Cl+ consists of several autoionization resonances surperimposed on the direct photoionization signal. Most of the prominent resonances are assigned to members of Rydberg series originating from the singlet ground state and from metastable triplet levels within the ground-state configuration of Cl+.     • Background: The first interim analysis of the phase III, randomized, double-blind, placebo-controlled, multinational TITAN study demonstrated improved overall survival (OS) and radiographic progression-free survival (rPFS) with apalutamide added to ongoing androgen deprivation therapy (ADT) in patients with metastatic castration-sensitive prostate cancer. The final analysis confirmed improvement in OS and other long-term outcomes. We evaluated prostate-specific antigen (PSA) kinetics and the association between PSA decline and outcomes in patients with metastatic castration-sensitive prostate cancer from TITAN. Patients and methods: Patients received apalutamide (240 mg/day) or placebo plus ADT (1: 1). This post hoc exploratory analysis evaluated PSA kinetics and decline in relation to rPFS (22.7 months’ follow-up) and OS, time to PSA progression, and time to castration resistance (44.0 months’ follow-up) in patients with or without confirmed PSA decline using a landmark analysis, the Kaplan–Meier method, and Cox proportional hazards model. Results: One thousand and fifty-two patients (apalutamide, 525; placebo, 527) were enrolled. Best confirmed PSA declines (≥50% or ≥90% from baseline or to ≤0.2 ng/ml) were achieved at any time during the study in 90%, 73%, and 68% of apalutamide-treated versus 55%, 29%, and 32% of placebo-treated patients, respectively. By 3 months of apalutamide treatment, best deep PSA decline of ≥90% or to ≤0.2 ng/ml occurred in 59% and 51% of apalutamide- and in 13% and 18% of placebo-treated patients, respectively. Achievement of deep PSA decline at landmark 3 months of apalutamide treatment was associated with longer OS [hazard ratio (HR) 0.35; 95% confidence interval (CI) 0.25-0.48), rPFS (HR 0.44; 95% CI 0.30-0.65), time to PSA progression (HR 0.31; 95% CI 0.22-0.44), and time to castration resistance (HR 0.38; 95% CI 0.27-0.52) compared with no decline (P < 0.0001 for all). Similar results were observed at landmark 6 and 12 months of apalutamide treatment. Conclusions: Apalutamide plus ADT demonstrated a robust (rapid, deep, and durable) PSA decline that was associated with improved clinical outcomes, including long-term survival.

    • Photoionization of Cl III ions into Cl IV was studied theoretically using the ab initio relativistic Breit-Pauli R-matrix (BPRM) method and experimentally at the Advanced Light Source (ALS) synchrotron at the Lawrence Berkeley National Laboratory. A relative-ion-yield spectrum of Cl IV was measured with a photon energy resolution of 10 meV. The theoretical study was carried out using a large wave-function expansion of 45 levels of configurations 3s(2)3p(2), 3s3p(3), 3s(2)3p3d, 3s(2)3p4s, 3s3p(2)3d, and 3p(4). The resulting spectra are complex. We have compared the observed spectrum with photoionization cross sections (sigma PI) of the ground state 3s(2)3p(3)(S-4(3/2)o) and the seven lowest excited levels 3s(2)3p(3)(D-2(5/2)o), 3s(2)3p(3)(D-2(3/2)o), 3s(2)3p(3)(P-2(3/2)o), 3s(2)3p3(P-2(1/2)o), 3s(3)p(4)((4)P5/2), 3s(3)p(4)(P-4(3/2)) and 3s3p(4)(P-4(1/2)) of Cl III, as these can generate resonances within the energy range of the experiment. We were able to identify most of the resonances as belonging to various specific initial levels within the primary Cl III ion beam. Compared to the first five levels, resonant structures in the sigma PI of excited levels of 3s3p(4) appear to have a weaker presence. We have also produced combined theoretical spectra of the levels by convolving the cross sections with a Gaussian profile of experimental width and summing them using statistical weight factors. The theoretical and experimental features show good agreement with the first five levels of Cl III. These features are also expected to elucidate the recent observed spectra of Cl III by Sloan Digital Scan Survey project.

    • An experimental and theoretical study of the single photoionization cross section of the P+ cation of phosphorus is presented. Photoionization (PI) cross sections are instrumental for the determination of abundances in the interstellar medium. The experiment was performed by merging an ion beam with a photon beam. The photon beam was nearly monochromatic and had an energy resolution of 24 meV. Calculations were carried out using the Breit–Pauli R-matrix method. The combined study was developed in the photon energy interval from 18 eV (68.9 nm) to 50 eV (24.8 nm). Comparison between the measured and the calculated cross section shows good agreement in general and identifies features of the process and existence of states in the experimental beam. The present results should provide for more accurate modeling of P+. © 2016 Elsevier Ltd

  • Ground-atmosphere interactions at Gale

    • We analyze variations in environmental parameters and regolith properties along Curiosity’s track to determine the possible causes of an abrupt change in the thermal properties of the ground and the atmosphere observed around Sol 120, as the rover transitioned from an area of sandy soil (Rocknest) to an area of fractured bedrock terrain (Yellowknife). Curiosity is instrumented with the Rover Environmental Monitoring Station (REMS) and the Dynamic Albedo of Neutrons (DAN) sensors to measure the air temperature, the ground temperature, and the hydrogen content of the shallow subsurface along Curiosity’s track. Analysis of the REMS data is used to estimate the regolith’s heat budget. This analysis suggests that the abrupt decrease in the ground and atmosphere temperature and the difference between ground and air temperatures observed around Sol 120 is likely caused by an increase in the soil thermal inertia. The changes in thermal inertia have been known for some time so confirming this by the REMS package provides ground truthing. A new unexpected finding is that the regolith water content, as indicated by DAN’s detection of hydrogen content, is higher in the Yellowknife soil. Another interesting finding at this site are the holes and other signs of recent geological activity in the area of fractured terrain that may reflect large volumetric variations and facilitate gas exchange between the ground and atmosphere. Near-surface volumetric changes in soil and bedrock could reflect changes in the volume of subsurface H2O, or in the partitioning of H2O among its three phases. Volume increases could also result from salt crystal growth in rock pores and soil pores associated with the adsorption of water vapor. Crystallization in pores is a significant weathering process on Earth; it could well be active on Mars. Salts also inhibits the exchange of moisture between the ground and the atmosphere, and cements the soils of arid places such as in the McMurdo Dry Valleys in Antarctica. Indeed, salts might be responsible for the ubiquitous martian duricrust. More importantly, salt crusts have the potential to create pockets of wet regolith in the shallow martian subsurface that could be habitable. A better understanding of ground-atmosphere interactions has the potential to shed new light into aqueous processes in the shallow martian subsurface.

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • Background: As part of I-O Optimise, a multinational research platform providing real-world insights into the management of lung cancers, the SCAN-LEAF study aims to describe the epidemiology, clinical care and outcomes for pts with NSCLC in Scandinavia. We report initial Tx and OS for pts with NSCLC prior to the availability of immunotherapies in Sweden. Methods: The analysis includes all adult pts diagnosed with NSCLC at Uppsala and Karolinska (Stockholm) University Hospitals from 2012 to 2015 (follow-up to Dec 2016). Electronic medical record data were extracted using Pygargus CXP software and linked with national registries. Bespoke rule-based algorithms were applied to describe Tx patterns; Kaplan–Meier methods were used to estimate OS. Results: 2779 pts were diagnosed with incident NSCLC (median age, 70 yrs [range: 22–96; 14.2% ≥80]; male, 48.5%; histology: non-squamous (NSQ), 70.9%, squamous (SQ), 17.7%, other, 11.4%; stage distribution: I, 19.3%; II, 7.7%; IIIA, 12.3%; IIIB, 7.2%; IV, 51.2%). Initial Tx (≤6 months from diagnosis) by stage and yr of diagnosis is shown in the table. Median OS (months) for NSQ and SQ pts: not reached and 52.8 in stage I, 43.2 and 23.6 in stage II, 26.7 and 20.4 in stage IIIA, 12.5 and 12.9 in stage IIIB, and 7.6 and 6.1 in stage IV, respectively. Among stage IIIB–IV pts, 60.7% (NSQ) and 53.5% (SQ) had ≥1 line of systemic anti-cancer therapy (SACT); median OS was 12.2 (NSQ) and 10.4 (SQ) months in pts on SACT, and 3.1 (NSQ) and 3.7 (SQ) months in pts not on SACT. Ongoing analyses will assess factors associated with SACT receipt in stage IIIB–IV pts. Conclusions: Swedish pts with NSCLC had a high burden of disease, with most diagnosed at stage IV and a median OS of 1 yr in late-stage pts receiving SACT. There is also scope for improved prognosis in pts diagnosed at early stages, particularly in SQ pts. Future analyses will assess the potential impact of recent improvements in diagnostics and therapeutics on Tx patterns and OS in Swedish NSCLC pts.

Methods: The SCAN-LEAF Danish and Swedish cohorts were established by linking respective national registries and include all adult pts diagnosed with incident NSCLC from Jan 2005 to Dec 2015 (follow-up to Dec 2016). The Kaplan–Meier method was used to estimate OS at 1, 3, and 5 yrs by histology (non-squamous cell [NSQ] or squamous cell [SQ]), TNM stage, and yr of diagnosis; changes in OS over time were assessed using the Cochrane–Armitage test. Results: 31,939 pts in Denmark and 30,067 pts in Sweden were diagnosed with NSCLC from 2005 to 2015. Most were diagnosed at stage IV (51.6% and 48.4%, respectively) and had NSQ histology (54.4% and 60.4%). Statistically significant trends (P < 0.05) for improved OS accompanied by an absolute OS rate increase of > 5% over the analysis period were seen for NSQ pts at 1 yr for all stages in both countries (Table); at 3 yrs for stages I–IIIB in Denmark (P ≤ 0.027), and stages I–II (P ≤ 0.0013) in Sweden; and at 5 yrs for stages I–II (P ≤ 0.026) in both countries. For SQ pts, this was seen only at 1 yr for stage IIIA in Denmark and stage I in Sweden (Table), and at 5 yrs for stage IIIA in Denmark (p = 0.02). Conclusions: Despite some improvements between 2005 and 2015, mainly in the short-term survival of pts with early-stage NSCLC, long-term OS rates for pts with late-stage disease did not change significantly and remained low. Even in pts with early-stage disease, OS outcomes were suboptimal, with a particular unmet need in the SQ population. Future analyses including data after 2015 will evaluate the potential impact on OS of increased use of new TKIs and immune checkpoint inhibitors.

    • Walker Lake, a terminal sodium bicarbonate lake in Western Nevada (Great Basin, USA) contains numerous carbonate structures, including stromatolites. The lake is a remnant of the larger Pleistocene Lake Lahontan system that has been isolated for the last ~12 ka. Stromatolites of unique macroscale morphology were collected at the ancient Lahontan shoreline during the 2014 International Geobiology Course.Initial observations of a stromatolite bed revealed a bowl-shaped carbonate framework composed of stacked, weakly laminated, vertical and horizontal petal-like structures with copious pore space. One laterally-oriented petal was taken off of the main structure and studied. Petrographical observations exhibit two types of alternating microfabrics and three transitions in microfabric. Both sparry crystal fans of calcite, and convex layers of fine micrite with occasional trapped crystals and fossils, were observed.Calibrated 14C ages (IntCal13) for the proximal and the distal end of the stromatolite are 35,540 YBP and 33,580 YBP, respectively. Clumped isotope (D47)-based estimates of temperature steadily increase throughout most of this interval, from the beginning of accretion, to the middle of the structure. By the distal end, values are at their peak, and at the tip temperatures decrease again. D47-temperatures correspond to microfabric, with textural changes associated with evidence for climatic fluctuations.We suggest the stromatolite formation may have been initiated during warmer intervals, induced by the chemical precipitation of calcite fans which served as a substrate for a biofilm growth. Microbial activity trapped the fine sediment and formed micrite. Colder conditions propagated fan precipitation. Microfabric alternation throughout the stromatolite records environmental change in the span of ca. 2000 years of Lake Lahontan history, likely in response to lake level fluctuations.

    • Experimenting is fundamental to the training process of all scientists and engineers. While experiments have been traditionally done inside laboratories, the emergence of Information and Communication Technologies added two alternatives accessible anytime, anywhere. These two alternatives are known as virtual and remote laboratories and are sometimes indistinguishably referred as online laboratories. Similarly to other instructional technologies, virtual and remote laboratories require some effort from teachers in integrating them into curricula, taking into consideration several factors that affect their adoption (i.e., cost) and their educational effectiveness (i.e., benefit). This chapter analyzes these two dimensions and sustains the case where only through international cooperation it is possible to serve the large number of teachers and students involved in engineering education. It presents an example in the area of electrical and electronics engineering, based on a remote laboratory named Virtual Instruments System in Reality, and it then describes how a number of European and Latin American institutions have been cooperating under the scope of an Erasmus+ project, for spreading its use in Brazil and Argentina.

  • Detection of a new molecular cloud in the LHAASO J2108+5157 region supporting a hadronic PeVatron scenario

    • PeVatrons are the most powerful naturally occurring particle accelerators in the Universe. The identification of counterparts associated to astrophysical objects such as dying massive stars, molecular gas, star-forming regions, and star clusters is essential to clarify the underlying nature of the PeV emission, i.e., hadronic or leptonic. We present 12,13CO (J = 2→1) observations made with the 1.85 m radio-Telescope of the Osaka Prefecture University toward the Cygnus OB7 molecular cloud, which contains the PeVatron candidate LHAASO J2108+5157. We investigate the nature of the sub-PeV (gamma-ray) emission by studying the nucleon density determined from the content of H i and H2, derived from the CO observations. In addition to MML[2017]4607, detected via the observations of the optically thick 12CO (J = 1→0) emission, we infer the presence of an optically thin molecular cloud, named [FKT-MC]2022, whose angular size is 1-1 ± 0.}^\circ}}2. We propose this cloud as a new candidate to produce the sub-PeV emission observed in LHAASO J2108+5157. Considering a distance of 1.7 kpc, we estimate a nucleon (H i + H2) density of 37 ± 14 cm-3, and a total nucleon mass(H i + H2) of 1.5 ± 0.6 × 104 M·. On the other hand, we confirm that Kronberger 82 is a molecular clump with an angular size of 0°1, a nucleon density ∼103 cm-3, and a mass ∼103 M·. Although Kronberger 82 hosts the physical conditions to produce the observed emission of LHAASO J2108+5157, [FKT-MC]2022 is located closer to it, suggesting that the latter could be the one associated to the sub-PeV emission. Under this scenario, our results favour a hadronic origin for the emission.

  • Evidence for a gamma-ray molecular target in the enigmatic PeVatron candidate LHAASO J2108+5157

    • Context . Peta-eV (PeV) astronomy emerged in 2021 with the discovery of ultra-high-energy gamma-ray sources associated with powerful natural particle accelerators known as PeVatrons. In order to determine the nature of their emission, namely whether it has a hadronic or leptonic origin, it is essential to characterise the physical parameters of the environment where it originates. Aims . We unambiguously confirm the association of molecular gas with the PeVatron candidate LHAASO J2108+5157 using unprecedented high angular-resolution (17′) 12,13CO(J = 1 → 0) observations carried out with the Nobeyama 45m radio telescope. Methods . We characterised a molecular cloud in the vicinity of the PeVatron candidate LHAASO J2108+5157 by determining its physical parameters from our 12,13CO(J = 1 → 0) line observations. We used an updated estimation of the distance to the cloud, which provided a more reliable result. The molecular emission was compared with excess gamma-ray images obtained with Fermi-LAT at energies above 2 GeV to search for spatial correlations and test a possible hadronic (π0 decay) origin for the gamma-ray emission. Results . We find that the morphology of the spatial distribution of the CO emission is strikingly similar to that of the Fermi-LAT excess gamma ray. By combining our observations with archival 21 cm HI line data, the nucleons (HI + H2) number density of the target molecular cloud is found to be 133.0 ± 45.0 cm-3, for the measured angular size of 0.55 ± 0.02 at a distance of 1.6 ± 0.1 kpc. The resulting total mass of the cloud is M(HI + H2) = 7.5±2.9×103M⊙. Under a hadronic scenario, we obtain a total energy of protons of Wp = 4.3 ± 1.5 × 1046 erg with a cutoff of 700±300 TeV, which reproduces the sub-PeV gamma-ray emission. Conclusions . We identified a molecular cloud in the vicinity of LHAASO J2107+5157 as the main target where cosmic rays from an unknown PeVatron produce the observed gamma-ray emission via π0 decay.

    • In the first 100 Martian solar days (sols) of the Mars Science Laboratory mission, the Rover Environmental Monitoring Station (REMS) measured the seasonally evolving diurnal cycles of ultraviolet radiation, atmospheric pressure, air temperature, ground temperature, relative humidity, and wind within Gale Crater on Mars. As an introduction to several REMS-based articles in this issue, we provide an overview of the design and performance of the REMS sensors and discuss our approach to mitigating some of the difficulties we encountered following landing, including the loss of one of the two wind sensors. We discuss the REMS data set in the context of other Mars Science Laboratory instruments and observations and describe how an enhanced observing strategy greatly increased the amount of REMS data returned in the first 100 sols, providing complete coverage of the diurnal cycle every 4 to 6 sols. Finally, we provide a brief overview of key science results from the first 100 sols. We found Gale to be very dry, never reaching saturation relative humidities, subject to larger diurnal surface pressure variations than seen by any previous lander on Mars, air temperatures consistent with model predictions and abundant short timescale variability, and surface temperatures responsive to changes in surface properties and suggestive of subsurface layering.     • Alterations in brain cholesterol homeostasis in midlife are correlated with a higher risk of developing Alzheimer’s disease (AD). However, global cholesterol-lowering therapies have yielded mixed results when it comes to slowing down or preventing cognitive decline in AD. We used the transgenic mouse model Cyp27Tg, with systemically high levels of 27-hydroxycholesterol (27-OH) to examine long-term potentiation (LTP) in the hippocampal CA1 region, combined with dendritic spine reconstruction of CA1 pyramidal neurons to detect morphological and functional synaptic alterations induced by 27-OH high levels. Our results show that elevated 27-OH levels lead to enhanced LTP in the Schaffer collateral-CA1 synapses. This increase is correlated with abnormally large dendritic spines in the stratum radiatum. Using immunohistochemistry for synaptopodin (actin-binding protein involved in the recruitment of the spine apparatus), we found a significantly higher density of synaptopodin-positive puncta in CA1 in Cyp27Tg mice. We hypothesize that high 27-OH levels alter synaptic potentiation and could lead to dysfunction of fine-tuned processing of information in hippocampal circuits resulting in cognitive impairment. We suggest that these alterations could be detrimental for synaptic function and cognition later in life, representing a potential mechanism by which hypercholesterolemia could lead to alterations in memory function in neurodegenerative diseases.
