  • Variants of the interleukin-1 receptor antagonist gene are associated with fat mass in men

    • Context: Immune functions seem to have connections to variations in body fat mass. Studies of knockout mice indicate that endogenous interleukin (IL)-1 can suppress mature-onset obesity. Objective: To systematically investigate our hypotheses that single- nucleotide polymorphisms (SNPs) and/or haplotypes variants in the IL-1 gene system are associated with fat mass. Subjects: The Gothenburg osteoporosis and obesity determinants (GOOD) study is a population-based cross-sectional study of 18-20 year-old men (n = 1068), from Gothenburg, Sweden. Major findings were confirmed in elderly men (n = 3014) from the Swedish part of the osteoporotic fractures in men (MrOS) multicenter population-based study. Main Outcome Measure: The genotype distributions and their association with body fat mass in different compartments, measured with dual-energy X-ray absorptiometry (DXA). Results: Out of 15 investigated SNPs in the IL-1 receptor antagonist (IL1RN) gene, a recently identified 30 untranslated region C4T (rs4252041, minor allele frequency 4%) SNP was associated with the primary outcome total fat mass (P = 0.003) and regional fat masses, but not with lean body mass or serum IL-1 receptor 1 (IL1RN) levels. This SNP was also associated with body fat when correcting the earlier reported IL1RN_2018 T4C (rs419598) SNP (in linkage disequilibrium with a well-studied variable number tandem repeat of 86 bp). The association between rs4252041 SNP and body fat was confirmed in the older MrOS population (P = 0.03). The rs4252041 SNP was part of three haplotypes consisting of five adjacent SNPs that were identified by a sliding window approach. These haplotypes had a highly significant global association with total body fat (P < 0.001). None of the other investigated members of the IL-1 gene family displayed any SNPs that have not been described previously to be significantly associated with body fat. Conclusions: The IL1RN gene, shown to enhance obesity by suppressing IL-1 effects in experimental animals, have no previously described gene polymorphisms and haplotypes that are associated with fat, but not lean mass in two populations of men.   • Composite design for a foiling Optimist dinghy

    • In April 2017, a foiling Optimist dingy designed entirely by students, was successfully tested under standard sailing conditions in the waters outside Gothenburg. In order to achieve take of wind speeds as low as 6 m/s, a stiff and lightweight design of the dinghy and its foiling components was necessary. There have been few successful attempts to make an Optimist foil in a stable manner, as such there were no standards or recommendations available for the design. Therefore, a simulation driven structural design methodology for hydrofoils, centreboards, centreboard-to-hull connections, and necessary hull reinforcements using sandwich structures was adopted. The proposed design was then manufactured, allowing for a significantly stiffer hull and a 20% decrease in weight over a conventional Optimist. Excluding the rig and sail, the final weight came to 27 kg.

  • Hur konsumtionen digitaliseras : Texter från forskningsprojektet Digcon - Digitaliseringen av konsumtionskulturen

    • Digital teknik håller radikalt på att förändra vår konsumtionskultur. Appar, QR-koder och ständigt mer avancerade mobiltelefoner, samt de digitala spår användningen av dem lämnar efter sig skapar nya konsumtionsbeteenden, nya metoder för marknadsföring och försäljning av produkter, men också nya metoder för forskare att studera konsumtionsmönster. De omformar även vilka vi är som konsumenter. Att forska om den här omvandlingen innebär att undersöka tekniken i sig, de som tillverkar den, de som marknadsför den liksom de som använder den. I forskningsprojektet Digcon – Digitaliseringen av konsumtionskulturen, har forskare från Sverige och Frankrike undersökt olika aspekter av digitalisering och konsumtion. Vilka betydelser har digitala verktyg och apparater i människors vardagsliv och hur förändrar de konsumtionsmönster? Hur påverkar digitaliseringen vad det innebär att vara konsument och hur en konsument uppfattas vara. Hur omformas förståelser av kön, till exempel genom uppkomsten av modebloggar och modeappar? Vilka möjligheter till etisk konsumtion skapas genom digital teknik och vilken etik är det som skapas? I den här rapporten har vi samlat populärvetenskapliga texter från forskningsprojektet. Digcon bedrivs vid Centrum för konsumtionsvetenskap och Centre for retailing, Handelshögskolan vid Göteborgs universitet, Handelshögskolan i Stockholm och Université Tolouse II, Frankrike.

  • PRISMA Mission Extension : Adapting Mission Operations to New and Changing Mission Objectives

    • The PRISMA in-orbit test-bed was launched in June 2010 to demonstrate strategies and technologies for formation flying and rendezvous. OHB Sweden is the prime contractor for the project which is funded by the Swedish National Space Board (SNSB) with support from DLR, CNES, and DTU. In early September of 2011, 15 months after launch, all primary mission objectives of the PRISMA formation flying satellites had been achieved and mission success was declared. Since a significant amount of delta-V capability still remained an open call for new experiments was issued, inviting both old and new experimenters to use the capabilities of the formation. Several interested parties took the opportunity to perform their own experiments with an existing platform, each coming with new mission objectives not previously planned to be flown on the PRISMA satellites. Some of these experiments were close to what had already been achieved within the nominal mission, but some included new ways of using the formation not envisioned by the spacecraft designers. The new experiments span from data collection in specific relative orbits, with a separation from a few meters to several kilometers, to entirely new modules within the on-board software. Changing from a pre-planned technology demonstration mission to operating a commercial resource required adaptation of the original operational concept, taking into account the different levels of experience of the customers and managing the satellites between experiments. This paper describes how these new mission objectives were integrated in operations and how a sometimes very short turn-around between initial concept and experiment execution was implemented with the aid of well established validation processes, high degrees of on-board autonomy and a flexible operations team.

  • Optimus Pråm - Semi-submersible wind farm installation vessel for Blekinge Offshore

    • In Sweden the government is investing lots of resources in order to meet the energy need with clean and renewable alternatives. Since wind is an unlimited source of energy the exploitations of wind farms is of great interest. This report describes a conceptual design of an innovative offshore wind turbine installation vessel for inland sea conditions, with highest possible energy efficiency and environmental friendly performance in every detail. The customer, Blekinge Offshore, main requirement is to receive a concept with an as low installation cost as possible.The final concept Optimus Pråm includes one installation vessel, which is a semi-submersible barge, and one support vessel that supply the installation vessel with power and propulsion. The power is distributed from the support vessel to the installation vessel through a power cable. The installation vessel will be self-propelled during the installation phase using electrical motors and thrusters.The installation vessel shall be able to handle gravity foundations and fully assembled wind turbines, transported vertically to the installation site. Foundations and windmills will be fully assembled in Karlshamn port before transit to site. During transit the installation and support vessel are connected to each other in all motions except pitch. This almost total fixed connection makes the two vessels acts like one, which gives the joint vessel excellent manoeuvrability. When installing foundations, the installation vessel and the support vessel disconnect. The installation vessel places the foundation at the planned location by ballasting until the foundation has reached the seabed. After de-ballasting the installation vessel and support vessel connects again and transit back to the harbour. When installing wind turbines the two vessels are kept connected and makes highly accurate positioning for the installation possible

    • All techniques needed for proteomic analyses of plant plasma membranes are described in detail, from isolation of plasma membranes to protein identification by mass spectrometry (MS). Plasma membranes are isolated by aqueous two-phase partitioning yielding vesicles with a cytoplasmic side-in orientation and a purity of about 95%. These vesicles are turned inside-out by treatment with Brij 58, which removes soluble contaminating proteins enclosed in the vesicles as well as loosely attached proteins. The final plasma membrane preparation thus retains all integral proteins and many peripheral proteins. Proteins are separated by one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE), and protein bands are excised and digested with trypsin. Peptides in tryptic digests are separated by nanoflow liquid chromatography and either fed directly into an ESI-MS or spotted onto matrix-assisted laser desorption ionization (MALDI) plates for analysis with MALDI-MS. Finally, data processing and database searching are used for protein identification to define a plasma membrane proteome.

    • Objective: To monitor changes in serum concentrations of cartilage oligomeric matrix protein (COMP) during a 24-h period to determine any diurnal variation, and to estimate the half life of COMP in the circulation in patients with symptomatic knee osteoarthritis and in those with rheumatoid arthritis. Methods: Serum samples were drawn every 4 h (7 samples/patient over 24 h) in 10 patients with knee osteoarthritis and 14 patients with rheumatoid arthritis. Osteoarthritis was defined radiographically and clinically (American College of Rheumatology (ACR) criteria) and rheumatoid arthritis according to the 1987 ACR criteria. Serum COMP was measured by sandwich ELISA. A statistical model for the diurnal variation in the COMP levels was developed using the computer program NONMEM. Results: No considerable changes in COMP levels were observed during the day between 08:00 and 21:00 in either group. A significant decrease in serum COMP was apparent during bed rest at night, reaching the lowest levels between 04:00 and 05:00 (p < 0.03 or better v all other time points) in patients with osteoarthritis and in those with rheumatoid arthritis. From the rate of decreasing serum COMP levels, a putative half life of COMP in the circulation was estimated to be 7.4 h. Conclusion: During normal daytime activities, serum COMP levels are constant. The decrease during the night indicates a rapid elimination of COMP once it has reached the circulation. The stable COMP levels during the day suggest that it is not necessary to further standardise the time of serum sampling in clinical practice.

    • CONTEXT: Regulation of fat mass appears to be associated with immune functions. Studies of knockout mice show that endogenous interleukin (IL)-6 can suppress mature-onset obesity. OBJECTIVE: To systematically investigate associations of single nucleotide polymorphisms (SNPs) near the IL-6 (IL6) and IL-6 receptor (IL6R) genes with body fat mass, in support for our hypothesis that variants of these genes can be associated with obesity. DESIGN AND STUDY SUBJECTS: The Gothenburg Osteoporosis and Obesity Determinants (GOOD) study is a population-based cross-sectional study of 18- to 20-year-old men (n=1049), from the Gothenburg area (Sweden). Major findings were confirmed in two additional cohorts consisting of elderly men from the Osteoporotic Fractures in Men (MrOS) Sweden (n=2851) and MrOS US (n=5611) multicenter population-based studies. MAIN OUTCOME: The genotype distributions and their association with fat mass in different compartments, measured with dual-energy X-ray absorptiometry. RESULTS: Out of 18 evaluated tag SNPs near the IL6 and IL6R genes, a recently identified SNP rs10242595 G/A (minor allele frequency=29%) 3' of the IL6 gene was negatively associated with the primary outcome total body fat mass (effect size -0.11 standard deviation (s.d.) units per A allele, P=0.02). This negative association with fat mass was also confirmed in the combined MrOS Sweden and MrOS US cohorts (effect size -0.05 s.d. units per A allele, P=0.002). When all three cohorts were combined (n=8927, Caucasian subjects), rs10242595(*)A showed a negative association with total body fat mass (effect size -0.05 s.d. units per A allele, P<0.0002). Furthermore, the rs10242595(*)A was associated with low body mass index (effect size -0.03, P<0.001) and smaller regional fat masses. None of the other SNPs investigated in the GOOD study were reproducibly associated with body fat. CONCLUSIONS: The IL6 gene polymorphism rs10242595(*)A is associated with decreased fat mass in three combined cohorts of 8927 Caucasian men.
