  • Circumpolar distribution and carbon storage of thermokarst landscapes

    • Thermokarst is the process whereby the thawing of ice- rich permafrost ground causes land subsidence, resulting in development of distinctive landforms. Accelerated thermokarst due to climate change will damage infrastructure, but also impact hydrology, ecology and biogeochemistry. Here, we present a circumpolar assessment of the distribution of thermokarst landscapes, defined as landscapes comprised of current thermokarst landforms and areas susceptible to future thermokarst development. At 3.6 x 10(6) km(2), thermokarst landscapes are estimated to cover similar to 20% of the northern permafrost region, with approximately equal contributions from three landscape types where characteristic wetland, lake and hillslope thermokarst landforms occur. We estimate that approximately half of the below-ground organic carbon within the study region is stored in thermokarst landscapes. Our results highlight the importance of explicitly considering thermokarst when assessing impacts of climate change, including future landscape greenhouse gas emissions, and provide a means for assessing such impacts at the circumpolar scale.

    • Unbleached wood fibers and nanofibers are environmentally friendly bio-based candidates for material production, in particular, as reinforcements in polymer matrix biocomposites due to their low density and potential as carbon sink during the materials production phase. However, producing high reinforcement content biocomposites with degradable or chemically recyclable matrices is troublesome. Here, we address this issue with a new concept for facile and scalable in-situ polymerization of polyester matrices based on functionally balanced oligomers in pre-formed lignocellulosic networks. The idea enabled us to create high reinforcement biocomposites with well-dispersed mechanically undamaged fibers or nanocellulose. These degradable biocomposites have much higher mechanical properties than analogs in the literature. Reinforcement geometry (fibers at 30 mu m or fibrils at 10-1000 nm diameter) influenced the polymerization and degradation of the polyester matrix. Overall, this work opens up new pathways toward environmentally benign materials in the context of a circular bioeconomy. Cellulose biocomposites from nanocellulose or plant fibers with polymer matrix are often not degradable and suffer from insufficient mechanical properties to replace established materials. Here, the authors demonstrate the fabrication of hydrolytically degradable polymers through in-situ polymerization of new functionally balanced oligomers within high-content lignocellulose reinforcement networks.

  • Regular perturbation on the group-velocity dispersion parameter for nonlinear fibre-optical communications

    • Communication using the optical fibre channel can be challenging due to nonlinear effects that arise in the optical propagation. These effects represent physical processes that originate from light propagation in optical fibres. To obtain fundamental understandings of these processes, mathematical models are typically used. These models are based on approximations of the nonlinear Schrödinger equation, the differential equation that governs the propagation in an optical fibre. All available models in the literature are restricted to certain regimes of operation. Here, we present an approximate model for the nonlinear optical fibre channel in the weak-dispersion regime, in a noiseless scenario. The approximation is obtained by applying regular perturbation theory on the group-velocity dispersion parameter of the nonlinear Schrödinger equation. The proposed model is compared with three other models using the normalized square deviation metric and shown to be significantly more accurate for links with high nonlinearities and weak dispersion.

  • Groundwater discharge as a driver of methane emissions from Arctic lakes

    • Lateral CH4 inputs to Arctic lakes through groundwater discharge could be substantial and constitute an important pathway that links CH4 production in thawing permafrost to atmospheric emissions via lakes. Yet, groundwater CH4 inputs and associated drivers are hitherto poorly constrained because their dynamics and spatial variability are largely unknown. Here, we unravel the important role and drivers of groundwater discharge for CH4 emissions from Arctic lakes. Spatial patterns across lakes suggest groundwater inflows are primarily related to lake depth and wetland cover. Groundwater CH4 inputs to lakes are higher in summer than in autumn and are influenced by hydrological (groundwater recharge) and biological drivers (CH4 production). This information on the spatial and temporal patterns on groundwater discharge at high northern latitudes is critical for predicting lake CH4 emissions in the warming Arctic, as rising temperatures, increasing precipitation, and permafrost thawing may further exacerbate groundwater CH4 inputs to lakes.

  • Predation and spatial connectivity interact to shape ecosystem resilience to an ongoing regime shift

    • Ecosystem regime shifts can have severe ecological and economic consequences, making it a top priority to understand how to make systems more resilient. Theory predicts that spatial connectivity and the local environment interact to shape resilience, but empirical studies are scarce. Here, we use >7000 fish samplings from the Baltic Sea coast to test this prediction in an ongoing, spatially propagating shift in dominance from predatory fish to an opportunistic mesopredator, with cascading effects throughout the food web. After controlling for the influence of other drivers (including increasing mesopredator densities), we find that predatory fish habitat connectivity increases resilience to the shift, but only when densities of fish-eating top predators (seals, cormorants) are low. Resilience also increases with temperature, likely through boosted predatory fish growth and recruitment. These findings confirm theoretical predictions that spatial connectivity and the local environment can together shape resilience to regime shifts.

    • Myoglobin, encoded by MB, is a small cytoplasmic globular hemoprotein highly expressed in cardiac myocytes and oxidative skeletal myofibers. Myoglobin binds O2, facilitates its intracellular transport and serves as a controller of nitric oxide and reactive oxygen species. Here, we identify a recurrent c.292C>T (p.His98Tyr) substitution in MB in fourteen members of six European families suffering from an autosomal dominant progressive myopathy with highly characteristic sarcoplasmic inclusions in skeletal and cardiac muscle. Myoglobinopathy manifests in adulthood with proximal and axial weakness that progresses to involve distal muscles and causes respiratory and cardiac failure. Biochemical characterization reveals that the mutant myoglobin has altered O2 binding, exhibits a faster heme dissociation rate and has a lower reduction potential compared to wild-type myoglobin. Preliminary studies show that mutant myoglobin may result in elevated superoxide levels at the cellular level. These data define a recognizable muscle disease associated with MB mutation.

  • Tig1 regulates proximo-distal identity during salamander limb regeneration

    • Salamander limb regeneration is an accurate process which gives rise exclusively to the missing structures, irrespective of the amputation level. This suggests that cells in the stump have an awareness of their spatial location, a property termed positional identity. Little is known about how positional identity is encoded, in salamanders or other biological systems. Through single-cell RNAseq analysis, we identified Tig1/Rarres1 as a potential determinant of proximal identity. Tig1 encodes a conserved cell surface molecule, is regulated by retinoic acid and exhibits a graded expression along the proximo-distal axis of the limb. Its overexpression leads to regeneration defects in the distal elements and elicits proximal displacement of blastema cells, while its neutralisation blocks proximo-distal cell surface interactions. Critically, Tig1 reprogrammes distal cells to a proximal identity, upregulating Prod1 and inhibiting Hoxa13 and distal transcriptional networks. Thus, Tig1 is a central cell surface determinant of proximal identity in the salamander limb.

  • An optochemical tool for light-induced dissociation of adherens junctions to control mechanical coupling between cells

    • The cadherin-catenin complex at adherens junctions (AJs) is essential for the formation of cell-cell adhesion and epithelium integrity; however, studying the dynamic regulation of AJs at high spatio-temporal resolution remains challenging. Here we present an optochemical tool which allows reconstitution of AJs by chemical dimerization of the force bearing structures and their precise light-induced dissociation. For the dimerization, we reconstitute acto-myosin connection of a tailless E-cadherin by two ways: direct recruitment of α-catenin, and linking its cytosolic tail to the transmembrane domain. Our approach enables a specific ON-OFF switch for mechanical coupling between cells that can be controlled spatially on subcellular or tissue scale via photocleavage. The combination with cell migration analysis and traction force microscopy shows a wide-range of applicability and confirms the mechanical contribution of the reconstituted AJs. Remarkably, in vivo our tool is able to control structural and functional integrity of the epidermal layer in developing Xenopus embryos.
