  • ApoAI-derived peptide increases glucose tolerance and prevents formation of atherosclerosis in mice

    • AIMS/HYPOTHESIS: Finding new treatment alternatives for individuals with diabetes with severe insulin resistance is highly desired. To identify novel mechanisms that improve glucose uptake in skeletal muscle, independently from insulin levels and signalling, we have explored the therapeutic potential of a short peptide sequence, RG54, derived from apolipoprotein A-I (ApoA-I).METHODS: INS-1E rat clonal beta cells, C2C12 rat muscle myotubes and J774 mouse macrophages were used to study the impact of RG54 peptide on glucose-stimulated insulin secretion, glucose uptake and cholesterol efflux, respectively. GTTs were carried out on diet-induced insulin-resistant and Leprdb diabetic mouse models treated with RG54 peptide, and the impact of RG54 peptide on atherosclerosis was evaluated in Apoe-/- mice. Control mice received ApoA-I protein, liraglutide or NaCl.RESULTS: The synthetic RG54 peptide induced glucose uptake in cultured muscle myotubes by a similar amount as insulin, and also primed pancreatic beta cells for improved glucose-stimulated insulin secretion. The findings were verified in diet-induced insulin-resistant and Leprdb diabetic mice, jointly confirming the physiological effect. The RG54 peptide also efficiently catalysed cholesterol efflux from macrophages and prevented the formation of atherosclerotic plaques in Apoe-/- mice.CONCLUSIONS/INTERPRETATION: The RG54 peptide exhibits good prospects for providing glucose control and reducing the risk of cardiovascular disease in individuals with severe insulin resistance.

  • Characterisation of a flavonoid ligand of the fungal protein Alt a 1

    • Spores of pathogenic fungi are virtually ubiquitous and cause human disease and severe losses in crops. The endophytic fungi Alternaria species produce host-selective phytotoxins. Alt a 1 is a strongly allergenic protein found in A. alternata that causes severe asthma. Despite the well-established pathogenicity of Alt a 1, the molecular mechanisms underlying its action and physiological function remain largely unknown. To gain insight into the role played by this protein in the pathogenicity of the fungus, we studied production of Alt a 1 and its activity in spores. We found that Alt a 1 accumulates inside spores and that its release with a ligand is pH-dependent, with optimum production in the 5.0-6.5 interval. The Alt a 1 ligand was identified as a methylated flavonoid that inhibits plant root growth and detoxifies reactive oxygen species. We also found that Alt a 1 changes its oligomerization state depending on the pH of the surrounding medium and that these changes facilitate the release of the ligand. Based on these results, we propose that release of Alt a 1 should be a pathogenic target in approaches used to block plant defenses and consequently to favor fungal entry into the plant.

    • The analysis of the Coulomb excitation experiment conducted on 104Sn required a strict selection of the data in order to reduce the large background present in the gamma-ray energy spectra and identify the gamma-ray peak corresponding to the Coulomb excitation events. As a result the B(E2; 0 + -> 2+) value could be extracted, which established the downward trend towards 100Sn and therefore the robustness of the N=Z=50 core against quadrupole excitations.

    • A measurement of the reduced transition probability for the excitation of the ground state to the first 2(+) state in Sn-104 has been performed using relativistic Coulomb excitation at GSI. Sn-104 is the lightest isotope in the Sn chain for which this quantity has been measured. The result is a key point in the discussion of the evolution of nuclear structure in the proximity of the doubly magic nucleus Sn-100. The value B(E2; 0(+) -> 2(+)) = 0.10(4) e(2)b(2) is significantly lower than earlier results for Sn-106 and heavier isotopes. The result is well reproduced by shell model predictions and therefore indicates a robust N = Z = 50 shell closure.

    • Early depictions of anthropomorphs in rock art provide unique insights into life during the deep past. This includes human engagements with the environment, socio-cultural practices , gender and uses of material culture. In Australia, the Dynamic Figure rock paintings of Arnhem Land are recognized as the earliest style in the region where humans are explicitly depicted. Important questions, such as the nature and signicance of body adornment in rock art and society, can be explored, given the detailed nature of the human gurative art and the sheer number of scenes depicted. In this paper, we make a case for Dynamic Figure rock art having some of the earliest and most extensive depictions of complex an-thropomorph scenes found anywhere in the world.

    • This paper presents findings from a recent study of the Anbangbang Gallery in the Burrungkuy (Nourlangie) site complex of Kakadu National Park, Australia. Using new technologies alongside established methods for rock art documentation, we discuss the complexity and uniqueness of Anbangbang Gallery as an icon of Australian rock art. We have taken a comprehensive approach to our investigations, deliberately linking new technologies and scientific analysis with other archaeological and anthropological research methods. In particular, using evidence from a detailed site recording, oral histories, and pXRF analysis, we explore aspects of the site chronology, the nature of painting activity, and the retouching and repainting of earlier imagery. The findings force us to rethink the existing interpretative narrative for Anbangbang Gallery, the motivations behind previously held beliefs relating to recent rock art, and the implications this has had for ongoing conservation work in the region.
