    • The role of gamma delta T cells in antitumor immunity has been under investigation for the past two decades, but little is known about their contribution to clinical outcomes in patients. Here, we set out to define the clonotypic, phenotypic, and functional features of gamma delta T cells in peripheral blood, ascites, and metastatic tumor tissue from patients with advanced epithelial ovarian cancer. T cell receptor (TCR) sequencing of the gamma chain revealed that tumor-infiltrating gamma delta T cells have a unique and skewed repertoire with high TCR diversity and low clonality. In contrast, ascites-derived gamma delta T cells presented a lower TCR diversity and higher clonality, suggesting a TCR-dependent clonal focusing at this site. Further investigation showed that tumor samples had abundant gamma delta T cells with a tissue-resident, activation-associated phenotype, less usage of V gamma 9 and an impaired response to adaptive-associated stimuli, implying an innate-like activation pathway, rather than an adaptive TCR-engaging pathway, at these tumor sites. Furthermore, high gamma delta T cell cytokine responsiveness upon stimulation was associated with a favorable outcome for patients in terms of both overall survival and reduced residual tumor burden after primary surgery. Last, the functionality of gamma delta T cells and patient survival were negatively affected by the proportions of CD39-expressing T cells, highlighting the potential of CD39 as a target to improve gamma delta T cell responses and unleash their antitumor capabilities.

    • Abstract LTE and LTE-Advanced mobile technologies have integrated discontinuous reception (DRX) power saving method to optimize the power consumption at the user equipment (UE). The DRX method was proposed by the 3rd Generation Partnership Project (3GPP), and since then, the traffic behavior has been analyzed in several studies with a standard 3-state DRX model to describe the trade-off between power saving and delay. In this paper, we presented a novel 4-state and 5-state 3GPP LTE DRX mechanisms. The proposed mechanisms were developed by augmenting (an) active state(s) to deep and/or light sleep cycle of standard 3-state DRX for handling a small burst of packets, thereby bypassing the process of returning to the timer-dependent active mode. We have generated analytical models using a semi-Markov process for bursty packet data traffic and evaluated these augmented DRX mechanisms against a standard 3-state DRX method. Overall, the analytical results from varying timing parameters showed that our augmented DRX (both 4-state and 5-state) improved power saving factor (ranging between 1% and 8%) and reduced delay (ranging between 20% and 60%) compared to the standard 3-state DRX. Furthermore, the magnitude of improvement for both delay and power-saving was somewhat greater in 5-state than 4-state.

  • Defective peripheral B cell selection in common variable immune deficiency patients with autoimmune manifestations

    • Common variable immune deficiency (CVID) is a heterogeneous disorder characterized by recurrent infections, low levels of serum immunoglobulins, and impaired vaccine responses. Autoimmune manifestations are common, but B cell central and peripheral selection mechanisms in CVID are incompletely understood. Here, we find that receptor editing, a measure of central tolerance, is increased in transitional B cells from CVID patients and that these cells have a higher immunoglobulin κ:λ ratio in CVID patients with autoimmune manifestations than in those with infection only. Contrariwise, the selection pressure in the germinal center on CD27bright memory B cells is decreased in CVID patients with autoimmune manifestations. Finally, functionally, T cell-dependent activation showed that naive B cells in CVID patients are badly equipped for activation and induction of mismatch repair genes. We conclude that central tolerance is functional whereas peripheral selection is defective in CVID patients with autoimmune manifestations, which could underpin the development of autoimmunity.
