    • Background: Telomere length is a putative biomarker of ageing, morbidity and mortality. Its application is hampered by lack of widely applicable reference ranges and uncertainty regarding the present limits of measurement reproducibility within and between laboratories. Methods: We instigated an international collaborative study of telomere length assessment: 10 different laboratories, employing 3 different techniques [Southern blotting, single telomere length analysis (STELA) and real-time quantitative PCR (qPCR)] performed two rounds of fully blinded measurements on 10 human DNA samples per round to enable unbiased assessment of intra- and inter-batch variation between laboratories and techniques. Results: Absolute results from different laboratories differed widely and could thus not be compared directly, but rankings of relative telomere lengths were highly correlated (correlation coefficients of 0.63-0.99). Intra-technique correlations were similar for Southern blotting and qPCR and were stronger than inter-technique ones. However, inter-laboratory coefficients of variation (CVs) averaged about 10% for Southern blotting and STELA and more than 20% for qPCR. This difference was compensated for by a higher dynamic range for the qPCR method as shown by equal variance after z-scoring. Technical variation per laboratory, measured as median of intra- and inter-batch CVs, ranged from 1.4% to 9.5%, with differences between laboratories only marginally significant (P = 0.06). Gel-based and PCR-based techniques were not different in accuracy. Conclusions: Intra- and inter-laboratory technical variation severely limits the usefulness of data pooling and excludes sharing of reference ranges between laboratories. We propose to establish a common set of physical telomere length standards to improve comparability of telomere length estimates between laboratories.

  • Determinants of genetic variation across eco-evolutionary scales in pinnipeds

    • The effective size of a population (N-e), which determines its level of neutral variability, is a key evolutionary parameter. N-e can substantially depart from census sizes of present-day breeding populations (N-C) as a result of past demographic changes, variation in life-history traits and selection at linked sites. Using genome-wide data we estimated the long-term coalescent N-e for 17 pinniped species represented by 36 population samples (total n = 458 individuals). N-e estimates ranged from 8,936 to 91,178, were highly consistent within (sub)species and showed a strong positive correlation with N-C (R-adj(2) = 0.59; P = 0.0002). N-e/N-C ratios were low (mean, 0.31; median, 0.13) and co-varied strongly with demographic history and, to a lesser degree, with species' ecological and life-history variables such as breeding habitat. Residual variation in N-e/N-C, after controlling for past demographic fluctuations, contained information about recent population size changes during the Anthropocene. Specifically, species of conservation concern typically had positive residuals indicative of a smaller contemporary N-C than would be expected from their long-term N-e. This study highlights the value of comparative population genomic analyses for gauging the evolutionary processes governing genetic variation in natural populations, and provides a framework for identifying populations deserving closer conservation attention.

  • Heatwave-Like Events During Development Are Sufficient to Impair Bumblebee Worker Responses to Sensory Stimuli

    • Heatwaves are increasingly common globally and are known to have detrimental impacts on animal morphology and behaviour. These impacts can be severe, especially if heatwaves occur during development, even on animals that can regulate the temperature of their developing young. The onset and duration of heatwaves are stochastic and therefore may affect all or only part of development. In the heterothermic bumblebee Bombus terrestris, elevated temperatures over the entirety of development cause morphological changes in adults, despite their capability to regulate brood temperature. However, the effects of heatwaves that occur during a short period of development are unclear. We test the impact of elevated developmental temperature during the latter fraction of development on the behaviour and morphology of adult worker B. terrestris. We show that exposure to elevated temperature over a portion of late development is sufficient to impair the initial behavioural responses of workers to various sensory stimuli. Despite this, exposure to elevated temperatures during a period of development did not have any significant impact on body or organ size. The negative effect of elevated developmental temperatures was independent of the exposure time, which lasted from 11–20 days at the end of the workers’ developmental period. Thus, heat stress in bumblebees can manifest without morphological indicators and impair critical behavioural responses to relevant sensory stimuli, even if only present for a short period of time at the end of development. This has important implications for our understanding of deleterious climactic events and how we measure indicators of stress in pollinators.

  • Substantial variability in morphological scaling among bumblebee colonies

    • Differences in organ scaling among individuals may play an important role in determining behavioural variation. In social insects, there are well-documented intraspecific differences in colony behaviour, but the extent that organ scaling differs within and between colonies remains unclear. Using 12 different colonies of the bumblebee Bombus terrestris, we aim to address this knowledge gap by measuring the scaling relationships between three different organs (compound eyes, wings and antennae) and body size in workers. Though colonies were exposed to different rearing temperatures, this environmental variability did not explain the differences of the scaling relationships. Two colonies had differences in wing versus antenna slopes, three colonies showed differences in wing versus eye slopes and a single colony has differences between eye versus antenna slopes. There are also differences in antennae scaling slopes between three different colonies, and we present evidence for putative trade-offs in morphological investment. We discuss the utility of having variable scaling among colonies and the implication for understanding variability in colony fitness and behaviour.

  • A multi-scale hydroclimatic analysis of runoff generation in the Athabasca River, western Canada

    • A multi-scale hydroclimatic study of runoff generation in the Athabasca River watershed located in western Canada is presented. Mann-Kendall trend detection tests performed on hydrometric data for the lower Athabasca River (LAR) revealed predominantly significant (p<0.05) declines in annual and open-water season median/mean runoff indices over 1958-2009, with the ice-influenced season experiencing significant declines in the median and not the mean. The presence or absence of significant declines in the 25th and 75th runoff percentiles helped explain these results. The only noteworthy result from the mid-point of the watershed was a probable (p<0.10) decline in median open-water runoff, which was not seen over the 1913-2009 period. Divergent seasonal runoff trends from the headwater zone were seen since 1958 and back to 1913, increasing ice-influenced and declining open-water season runoff trends. Although precipitation was observed to decline over 1958-2009, only the LAR watershed scale annual index emerged as a probable decline. Multiple non-linear regression analysis indicated that variation in precipitation explained >67% of the annual median/mean LAR runoff variability since 1958. A first-order precipitation driven hindcasting model suggested that LAR watershed scale runoff may have increased since 1913, warranting further study. A correlation analysis of climatic teleconnections with median/mean runoff indices revealed that the winter North Pacific American index showed a strong, positive association with open-water runoff. The results from our study demonstrated that potentially inconsistent and/or divergent trend results can be obtained when using different time periods and/or regions of the watershed, emphasizing that extreme caution should be exercised when extrapolating sub-watershed results to the watershed scale, or to adjacent watersheds. Our multi-scale study approach also identified the drainage area between Athabasca and Fort McMurray as a zone that influenced runoff declines observed at the LAR watershed scale since 1958, which warrants further investigation with competent hydrological models.     • Two signatures of quantum effects on radiation reaction in the collision of a similar to GeV electron beam with a high intensity (>3 x 10(20) W cm(-2)) laser pulse have been considered. We show that the decrease in the average energy of the electron beam may be used to measure the Gaunt factor g for synchrotron emission. We derive an equation for the evolution of the variance in the energy of the electron beam in the quantum regime, i.e. quantum efficiency parameter eta (sic) 1. We show that the evolution of the variance may be used as a direct measure of the quantum stochasticity of the radiation reaction and determine the parameter regime where this is observable. For example, stochastic emission results in a 25 % increase in the standard deviation of the energy spectrum of a GeV electron beam, 1 fs after it collides with a laser pulse of intensity 10(21) W cm(-2). This effect should therefore be measurable using current high-intensity laser systems.

    • Imaging the visual systems of bumblebees and other pollinating insects may increase understanding of their dependence on specific habitats and how they will be affected by climate change. Current high-resolution imaging methods are either limited to two dimensions (light- and electron microscopy) or have limited access (synchrotron radiation x-ray tomography). For x-ray imaging, heavy metal stains are often used to increase contrast. Here, we present micron-resolution imaging of compound eyes of buff-tailed bumblebees (Bombus terrestris) using a table-top x-ray nanotomography (nano-CT) system. By propagation-based phase-contrast imaging, the use of stains was avoided and the microanatomy could more accurately be reconstructed than in samples stained with phosphotungstic acid or osmium tetroxide. The findings in the nano-CT images of the compound eye were confirmed by comparisons with light- and transmission electron microscopy of the same sample and finally, comparisons to synchrotron radiation tomography as well as to a commercial micro-CT system were done.
