  • What can we learn from long-term groundwater data to improve climate change impact studies?

    • Future risks for groundwater resources, due to global change are usually analyzed by driving hydrological models with the outputs of climate models. However, this model chain is subject to considerable uncertainties. Given the high uncertainties it is essential to identify the processes governing the groundwater dynamics, as these processes are likely to affect groundwater resources in the future, too. Information about the dominant mechanisms can be achieved by the analysis of long-term data, which are assumed to provide insight in the reaction of groundwater resources to changing conditions (weather, land use, water demand). Referring to this, a dataset of 30 long-term time series of precipitation dominated groundwater systems in northern Switzerland and southern Germany is collected. In order to receive additional information the analysis of the data is carried out together with hydrological model simulations. High spatio-temporal correlations, even over large distances could be detected and are assumed to be related to large-scale atmospheric circulation patterns. As a result it is suggested to prefer innovative weather-type-based downscaling methods to other stochastic downscaling approaches. In addition, with the help of a qualitative procedure to distinguish between meteorological and anthropogenic causes it was possible to identify processes which dominated the groundwater dynamics in the past. It could be shown that besides the meteorological conditions, land use changes, pumping activity and feedback mechanisms governed the groundwater dynamics. Based on these findings, recommendations to improve climate change impact studies are suggested. © Author(s) 2011.

  • RNA interference in Lepidoptera : An overview of successful and unsuccessful studies and implications for experimental design

    • Gene silencing through RNA interference (RNAi) has revolutionized the study of gene function, particularly in non-model insects. However, in Lepidoptera (moths and butterflies) RNAi has many times proven to be difficult to achieve. Most of the negative results have been anecdotal and the positive experiments have not been collected in such a way that they are possible to analyze. In this review, we have collected detailed data from more than 150 experiments including all to date published and many unpublished experiments. Despite a large variation in the data, trends that are found are that RNAi is particularly successful in the family Saturniidae and in genes involved in immunity. On the contrary, gene expression in epidermal tissues seems to be most difficult to silence. In addition, gene silencing by feeding dsRNA requires high concentrations for success. Possible causes for the variability of success in RNAi experiments in Lepidoptera are discussed. The review also points to a need to further investigate the mechanism of RNAi in lepidopteran insects and its possible connection to the innate immune response. Our general understanding of RNAi in Lepidoptera will be further aided in the future as our public database at http://insectacentral.org/RNAi will continue to gather information on RNAi experiments.

    • Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease with a lifetime risk of one in 350 people and an unmet need for disease-modifying therapies. We conducted a cross-ancestry genome-wide association study (GWAS) including 29,612 patients with ALS and 122,656 controls, which identified 15 risk loci. When combined with 8,953 individuals with whole-genome sequencing (6,538 patients, 2,415 controls) and a large cortex-derived expression quantitative trait locus (eQTL) dataset (MetaBrain), analyses revealed locus-specific genetic architectures in which we prioritized genes either through rare variants, short tandem repeats or regulatory effects. ALS-associated risk loci were shared with multiple traits within the neurodegenerative spectrum but with distinct enrichment patterns across brain regions and cell types. Of the environmental and lifestyle risk factors obtained from the literature, Mendelian randomization analyses indicated a causal role for high cholesterol levels. The combination of all ALS-associated signals reveals a role for perturbations in vesicle-mediated transport and autophagy and provides evidence for cell-autonomous disease initiation in glutamatergic neurons.

  • CLINICAL UTILITY OF β-AMYLOID PET IMAGING IN PEOPLE LIVING WITH HIV WITH COGNITIVE SYMPTOMS.

    • Imaging with β-amyloid (Aβ) PET has the potential to aid the diagnosis of the cause of cognitive impairment affecting people living with HIV (PLWH) when neurodegenerative disorders are considered. We evaluated the clinical utility of [18F]Florbetaben (FBB) in PLWH with cognitive symptoms.Imaging with FBB PET was performed in 20 patients with cognitive concerns about dementia. Neuropsychological testing, plasma neurofilament light protein, plasma Aβ40, Aβ42 and CSF Aβ42, tau, and HIV RNA were obtained. FBB PET images were assessed visually by three readers blinded to the clinical diagnosis, and quantitatively by obtaining a composite cortical to cerebellar cortex standardized uptake value ratio (SUVR). FBB SUVR from 10 age-matched healthy controls were compared to SUVR of PLWH.Most participants were male (90%) of white ethnicity (90%) with a median age (IQR) of 59 (43-79) years. Median CD4 count was 682 (74-1056). All patients were on cART with plasma and CSF HIV RNA< 40 copies/mL. Fourteen patients had objective cognitive impairment including two who met clinical criteria for a diagnosis of dementia. No significant differences in composite SUVRs between PLWH and controls [mean (SD): 1.18 (0.03) vs 1.16 (0.09); p=0.37] were observed. Four patients were FBB+ with the highest SUVR in the posterior cingulate, superior temporal and frontal superior lobe. Amyloid PET results contributed to a change in diagnosis and treatment for 10 patients.[18F]Florbetaben PET has potential as an adjunctive tool in the diagnosis of PLWH with cognitive impairment, increasing diagnostic certainty and optimizing management.

  • Control of the phonon band gap with isotopes in hexagonal boron nitride

    • The isotopic mass of constituent elements of materials has a well-known effect on the energy of vibrational modes. By means of monochromated scanning transmission electron microscopy we have experimentally studied the phonon bandstructure of hexagonal BN, where a phonon band gap appears between in-plane optical phonon modes and the lower energy part of the phonon spectrum. The size of the phonon band gap can be manipulated by the isotopic mass of the boron. While in 11BN the phonon band gap is about 7 meV wide, in 10BN the gap nearly closes, being an order of magnitude smaller (below 0.5 meV). This opens exciting options for manipulating terahertz wave propagation through isotopically structured devices having otherwise no interfaces between chemically distinct components.
