    • Cyclic nucleotide phosphodiesterases (PDEs) are important regulators of signal transduction processes mediated by cAMP and cGMP. One PDE family member, PDE3B, plays an important role in the regulation of a variety of metabolic processes such as lipolysis and insulin secretion. In this study, the cellular localization and the role of PDE3B in the regulation of triglyceride, cholesterol and glucose metabolism in hepatocytes were investigated. PDE3B was identified in caveolae, specific regions in the plasma membrane, and smooth endoplasmic reticulum. In caveolin-1 knock out mice, which lack caveolae, the amount of PDE3B protein and activity were reduced indicating a role of caveolin-1/caveolae in the stabilization of enzyme protein. Hepatocytes from PDE3B knock out mice displayed increased glucose, triglyceride and cholesterol levels, which was associated with increased expression of gluconeogenic and lipogenic genes/enzymes including, phosphoenolpyruvate carboxykinase, peroxisome proliferator-activated receptor gamma, sterol regulatory element-binding protein 1c and hydroxyl-3-methylglutaryl coenzyme A reductase. In conclusion, hepatocyte PDE3B is localized in caveolae and smooth endoplasmic reticulum and plays important roles in the regulation of glucose, triglyceride and cholesterol metabolism. Dysregulation of PDE3B could have a role in the development of fatty liver, a condition highly relevant in the context of type 2 diabetes.

  • Mammographic calcifications undergoing percutaneous biopsy : outcome in women with and without a personal history of breast cancer

    • Purpose: To compare the positive predictive values (PPVs) of BI-RADS categories used to assess pure mammographic calcifications in women with and without a previous history of breast cancer (PHBC). Materials and methods: In this retrospective study, all consecutive pure mammographic calcifications (n = 320) undergoing a stereotactic biopsy between 2016 and 2018 were identified. Mammograms were evaluated in consensus by two radiologists according to BI-RADS and blinded to patient history and pathology results. Final pathologic results were used as the standard of reference. PPV of BI-RADS categories were compared between the two groups. Data were evaluated using standard statistics, Mann–Whitney U tests and Chi-square tests. Results: Two hundred sixty-eight patients (274 lesions, median age 54 years, inter-quartile range, 50–65 years) with a PHBC (n = 46) and without a PHBC (n = 222) were included. Overall PPVs were the following: BI-RADS 2, 0% (0 of 56); BI-RADS 3, 9.1% (1 of 11); BI-RADS 4a, 16.2% (6 of 37); BI-RADS 4b, 37.5% (48 of 128); BI-RADS 4c, 47.3% (18 of 38) and BI-RADS 5, 100% (4 of 4). The PPV of BI-RADS categories was similar in patients with and without a PHBC (P =.715). Calcifications were more often malignant in patients with a PHBC older than 10 years (47.3%, 9 of 19) compared to 1–2 years (25%, 1 of 4), 2–5 years (20%, 2 of 10) and 5–10 years (0%, of 13) from the first breast cancer (P =.005). Conclusion: PPV of mammographic calcifications is similar in women with or without PHBC when BI-RADS classification is strictly applied. A higher risk of malignancy was observed in patients with a PHBC longer than 10 years.

  • Scaling the right answers – Creating and maintaining hope through social entrepreneurship in light of humanitarian crises

    • Triggered by the Russo–Ukrainian war starting early in 2022 and the subsequent movement of refugees toward various European countries, this rapid response paper provides five reflections on the role of social entrepreneurship in light of humanitarian crises. We validate two problems with the help of a problem owner from social entrepreneurial practice and suggest answers to them grounded in existing evidence documented in the academic literature (translational research approach). First, we show how social entrepreneurs can focus on solving the right problems in chaotic and fast-paced crises, and second, we illustrate measures to scale appropriately. Finally, on a meta-level, hope emerges as an additional answer. Even if social entrepreneurs should not address the “right” problems and even if they scale inappropriately, in light of any humanitarian crises, they still contribute value by creating hope for their societies, their stakeholders, and for themselves. © 2022 The Authors

  • Genetic variants associated with subjective well-being, depressive symptoms, and neuroticism identified through genome-wide analyses

    • Very few genetic variants have been associated with depression and neuroticism, likely because of limitations on sample size in previous studies. Subjective well-being, a phenotype that is genetically correlated with both of these traits, has not yet been studied with genome-wide data. We conducted genome-wide association studies of three phenotypes: subjective well-being (n = 298,420), depressive symptoms (n = 161,460), and neuroticism (n = 170,911). We identify 3 variants associated with subjective well-being, 2 variants associated with depressive symptoms, and 11 variants associated with neuroticism, including 2 inversion polymorphisms. The two loci associated with depressive symptoms replicate in an independent depression sample. Joint analyses that exploit the high genetic correlations between the phenotypes (vertical bar(p) over cap vertical bar approximate to 0.8) strengthen the overall credibility of the findings and allow us to identify additional variants. Across our phenotypes, loci regulating expression in central nervous system and adrenal or pancreas tissues are strongly enriched for association.

  • Cancer classification using the Immunoscore : a worldwide task force

    • Prediction of clinical outcome in cancer is usually achieved by histopathological evaluation of tissue samples obtained during surgical resection of the primary tumor. Traditional tumor staging (AJCC/UICC-TNM classification) summarizes data on tumor burden (T), presence of cancer cells in draining and regional lymph nodes (N) and evidence for metastases (M). However, it is now recognized that clinical outcome can significantly vary among patients within the same stage. The current classification provides limited prognostic information, and does not predict response to therapy. Recent literature has alluded to the importance of the host immune system in controlling tumor progression. Thus, evidence supports the notion to include immunological biomarkers, implemented as a tool for the prediction of prognosis and response to therapy. Accumulating data, collected from large cohorts of human cancers, has demonstrated the impact of immune-classification, which has a prognostic value that may add to the significance of the AJCC/UICC TNM-classification. It is therefore imperative to begin to incorporate the ` Immunoscore' into traditional classification, thus providing an essential prognostic and potentially predictive tool. Introduction of this parameter as a biomarker to classify cancers, as part of routine diagnostic and prognostic assessment of tumors, will facilitate clinical decision-making including rational stratification of patient treatment. Equally, the inherent complexity of quantitative immunohistochemistry, in conjunction with protocol variation across laboratories, analysis of different immune cell types, inconsistent region selection criteria, and variable ways to quantify immune infiltration, all underline the urgent requirement to reach assay harmonization. In an effort to promote the Immunoscore in routine clinical settings, an international task force was initiated. This review represents a follow-up of the announcement of this initiative, and of the J Transl Med. editorial from January 2012. Immunophenotyping of tumors may provide crucial novel prognostic information. The results of this international validation may result in the implementation of the Immunoscore as a new component for the classification of cancer, designated TNM-I (TNM-Immune).

    • The sustainable supply of raw materials to an expanding global population and rising consumption requires novel governance approaches. Such approaches have recently come to include the topic of ‘minerals safeguarding’. In Austria, the objective of minerals safeguarding has translated into an internationally recognised policy instrument, the Austrian Raw Materials Plan. Our paper investigates how the novel policy approach of safeguarding is translated into provincial and regional land use planning policy in Austria. Following a comparative approach of nine Austrian provinces via document analysis and interviews with policy makers, the analysis indicates diverse forms of implementation: We identify different degrees of uptake and implementation in land use planning policy across goals, instruments and mechanisms. Given the limited amount of research about safeguarding, this paper contributes to a broader understanding of safeguarding practices in public policy (goals, process and instruments), offers insights of’ strong, mixed, and soft’ safeguarding approaches, and provides input for a conceptual model of safeguarding. Our study highlights the importance of political agenda setting for the deployment of land use planning instruments, the need for coordination and communication (between and amongst different levels of government) in the policy process, and a need to assess effectiveness of safeguarding policies.

    • The only way to cure leukemia is by cooperative research. To optimize research, the European Leukemia Net integrates 105 national leukemia trial groups and networks, 105 interdisciplinary partner groups and about 1,000 leukemia specialists from 175 institutions. They care for tens of thousands of leukemia patients in 33 countries across Europe. Their ultimate goal is to cure leukemia. Since its inception in 2002, the European Leukemia Net has steadily expanded and has unified leukemia research across Europe. The European Leukemia Net grew from two major roots: 1) the German Competence Network on Acute and Chronic Leukemias; and 2) the collaboration of European Investigators on Chronic Myeloid Leukemia. The European Leukemia Net has improved leukemia research and management across Europe. Its concept has led to funding by the European Commission as a network of excellence. Other sources (European Science Foundation; European Leukemia Net-Foundation) will take over when the support of the European Commission ends.

    • This subtask is divided in two actions: Action 2A: Materials Assessment & Ageing Procedures (Experiments & Simulation) Action 2B: Components & Systems Assessment (Experiments & Simulation) As their structures and microstructures are completely different, Super-Insulating Materials (SIMs) cannot be compared directly to traditional insulating materials. Worldwide acceptance of these materials will be improved if the hygro-thermal and mechanical properties of SIM can be clearly articulated and reproduced. In particular, nano-structured materials used to manufacture a SIM are characterized by a high specific area (m²/g) and narrow pores (smaller than 1 μm) which make them very sensitive to gas adsorption and condensation, especially in contact with water molecules. Therefore, methods of characterization must be adapted, or new methods developed to measure the microstructural, hygro-thermal and mechanical properties of these materials and their barrier films. In parallel, modelling methods to describe heat, moisture and air transfer through nano-structured materials and films will have to be developed (adsorption and desorption models, diffusion models, freeze-thawing …). Of course, a few methods will be common to all SIMs, but due to their structural differences some specific modelling methods have to be developed. SIMs can offer considerable advantages (low thickness, low Uvalue) ; however potential drawback effects should be considered in the planning process in order to optimise the development of these extraordinary properties (very low thermal conductivity) and to prevent negative publicity which could be detrimental to this sector of emerging products. This is why ageing tests will be set according to realistic conditions (temperature, moisture, pressure, load …) as set out in SubTask 3A. One objective of artificial ageing is to understand potential degradation processes that could occur. The durability of hydrophobic treatment is one of these processes and will also be subject to discussion and investigation. At the component scale, additional characterizations are needed as panels or rolls are sold by manufacturers. In particular, thermal bridges will be carefully investigated, as the extraordinary thermal performance of SIMs are sensitive to the influence of thermal bridges.

  • Mineralogy, Morphology, and Emplacement History of the Maaz Formation on the Jezero Crater Floor From Orbital and Rover Observations

    • The first samples collected by the Perseverance rover on the Mars 2020 mission were from the Maaz formation, a lava plain that covers most of the floor of Jezero crater. Laboratory analysis of these samples back on Earth would provide important constraints on the petrologic history, aqueous processes, and timing of key events in Jezero crater. However, interpreting these samples requires a detailed understanding of the emplacement and modification history of the Maaz formation. Here we synthesize rover and orbital remote sensing data to link outcrop-scale interpretations to the broader history of the crater, including Mastcam-Z mosaics and multispectral images, SuperCam chemistry and reflectance point spectra, Radar Imager for Mars' subsurface eXperiment ground penetrating radar, and orbital hyperspectral reflectance and high-resolution images. We show that the Maaz formation is composed of a series of distinct members corresponding to basaltic to basaltic-andesite lava flows. The members exhibit variable spectral signatures dominated by high-Ca pyroxene, Fe-bearing feldspar, and hematite, which can be tied directly to igneous grains and altered matrix in abrasion patches. Spectral variations correlate with morphological variations, from recessive layers that produce a regolith lag in lower Maaz, to weathered polygonally fractured paleosurfaces and crater-retaining massive blocky hummocks in upper Maaz. The Maaz members were likely separated by one or more extended periods of time, and were subjected to variable erosion, burial, exhumation, weathering, and tectonic modification. The two unique samples from the Maaz formation are representative of this diversity, and together will provide an important geochronological framework for the history of Jezero crater.
