  • Worldwide prevalence of baseline resistance-associated polymorphisms and resistance mutations in HCV against current direct-acting antivirals

    • Background: HCV infections can now be completely cured, thanks to the currently marketed direct-acting antivirals (DAAs). It is known that HCV patients carry viral populations with baseline polymorphisms and/or mutations that make them resistant against some of these DAAs, which can negatively impact the patient's treatment outcome. Using complete HCV coding sequences isolated from 1,306 treatment-naive patients of genotypes (GTs) 1, 2, 3, 4 and 6 from around the globe, we studied the prevalence of baseline resistance-associated polymorphisms (RAPs) and resistance mutations (RMs) against DAAs that are currently on the market or in clinical trials.Methods: The HCV genome sequences used in this study were retrieved from the NCBI database. RAPs and RMs, with reference to HCV GT1a, were identified using the HCV Geno2pheno web server.Results: Nearly 50% of the total amino acid positions (including NS3 protease, NS5A and NS5B) studied are baseline polymorphisms that differentiated one GT from the rest. A proportion of these baseline polymorphisms and baseline non-polymorphic RMs could confer a significant increase in resistance against DAAs.Conclusions: In this study, we show the presence and prevalence of RAPs and RMs in DAA treatment-naive patients against currently used DAAs or DAAs in clinical trials. Our study suggests that RAPs and RMs profiling of HCV patients should be performed before the start of the therapy. Our results should be relevant especially in low- and middle-income countries, where the patients have a large variation of GTs and subtypes, and where the generic HCV treatment is now increasingly available.

  • Enhanced anti-wetted PVDF membrane for pulping RO brine treatment by vacuum membrane distillation

    • Large amounts of lignin, hemicellulose, organic matter and inorganic salts are present in pulping and papermaking wastewater. Therefore, the treatment and discharge of pulping wastewater are important issues. We have designed and developed the world's first zero liquid discharge (ZLD) system for it based on pretreatment-membrane-evaporative crystallization integrated technologies. However, for the treatment of high salt reverse osmosis (RO) brine, there are still challenges that need to be addressed. In this work, the feasibility of using a membrane distillation crystallization (MDC) process to treat high saline RO brine was investigated. Two different coating materials, Hyflon AD 40H and perfluoropolyether (PFPE), were applied to improve the membrane surface hydrophobicity for stable and efficient performance. After modification, the water contact angle was up to 138.4o and 157.7o for Hyflon/polyvinylidene fluoride (PVDF) and PFPE/PVDF composite membranes, respectively. Moreover, the PFPE/PVDF membrane showed a higher repellency toward RO brine, which is better for the following MDC treatment. During the MDC progress using composite PFPE/PVDF membranes, the salt crystals precipitated and gradually accumulated. More importantly, the final NaCl purity can be up to 94%. Therefore, it is hopeful to improve of the existing ZLD system by MDC operation.

  • Bark Beetle-Associated Blue-Stain Fungi Increase Antioxidant Enzyme Activities and Monoterpene Concentrations in Pinus yunnanensis

    • Yunnan pine is the most important tree species in SW China in both economical and ecological terms, but it is often killed by pine shoot beetles (Tomicus spp.). Tomicus beetles are secondary pests in temperate regions and the aggressiveness of the beetles in SW China is considered to be due to the warm subtropical climates as well as the beetles' virulent fungal associates. Here, we assessed the virulence of three blue-stain fungi (Leptographium wushanense, L. sinense and Ophiostoma canum) associated with pine shoot beetles to Yunnan pine (Pinus yunnanensis) in SW China. Following fungal inoculation, we measured necrotic lesion lengths, antioxidant enzyme activities and monoterpene concentrations in the stem phloem of Yunnan pine. Leptographium wushanense induced twice as long lesions as L. sinense and O. canum, and all three fungi induced significantly longer lesions than sterile agar control inoculations. The activity of three tested antioxidant enzymes (peroxidase, polyphenol oxidase, and superoxide dismutase) increased after both fungal inoculation and control inoculation. However, L. wushanense and L. sinense generally caused a greater increase in enzyme activities than O. canum and the control treatment. Fungal inoculation induced stronger increases in six major monoterpenes than the control treatment, but the difference was significant only for some fungus-monoterpene combinations. Overall, our results show that L. wushanense and L. sinense elicit stronger defense responses and thus are more virulent to Yunnan pine than O. canum. The two Leptographium species may thus contribute to the aggressiveness of their beetle vectors and could damage Yunnan pine across SW China if they spread from the restricted geographical area they have been found in so far.

    • Ophiostomatalean fungi may facilitate bark beetle colonization and reproduction. In the present study of the fungal community associated with bark beetle spe-cies belonging to Tomicus in Yunnan, China, six ophios-tomatalean fungi (Ophiostoma canum, O. ips, O. tingens, Leptographium yunnanense, Leptographium sp. 1 and Lep-tographium sp. 2) were isolated from the beetles or their galleries; O. canum was the most common fungal species. The distribution of O. canum was associated with stands heavily damaged by Tomicus species and a higher percentage of valid galleries of Tomicus yunnanensis and T. minor in Yunnan pine (Pinus yunnanensis). After inoculation of Yun-nan pine with the fungus, a phloem reaction zone formed and monoterpenes accumulated in the phloem. These results suggested that O. canum was pathogenic to Yunnan pine and that the wide distribution of the fungus might be beneficial to reproduction of pine shoot beetles in Yunnan pine. How-ever, because the reaction zone and monoterpene accumu-lation were mild, fungal damage of Yunnan pine might be limited. A more integrated study considering all the fungal species should be done to better understand the interactions among bark beetles, blue-stain fungi, and the tree hosts in the region.

  • The Dominant Role of Brewer-Dobson Circulation on 17O-Excess Variations in Snow Pits at Dome A, Antarctica

    • Recent studies have suggested that water isotopologues in snow pits from remote East Antarctica can be influenced by the input of stratospheric water, which has anomalously high 17O-excess values. However, it remains unclear whether the 17O-excess records preserved in snow and ice from this region can be used to reconstruct stratosphere-troposphere exchange (STE). In this study, we present high-resolution 17O-excess records from two snow pits at Dome A, the highest point of the Antarctic ice sheet. The 17O-excess records show a significant positive correlation with the strength of the Brewer-Dobson circulation (BDC), the hemispheric-scale troposphere-stratosphere overturn circulation. Stronger BDC leads to more stratospheric water input over Antarctica and higher 17O-excess, and vice versa. In addition, the 17O-excess records also have a significant positive correlation with the Southern Annular Mode (SAM) index, because SAM modulates Antarctic precipitation, which has a dilution effect on the stratospheric water input. The 17O-excess records do not show significant correlations with local temperature and relative humidity in the moisture source region. These results suggest the dominant effect of BDC on 17O-excess and indicate the potential for using 17O-excess records in ice cores from remote sites in East Antarctica for reconstructing long-term variations of STE, and understanding their mechanisms and climate effects.

  • Evolution of spin excitations from bulk to monolayer FeSe

    • In ultrathin films of FeSe grown on SrTiO (FeSe/STO), the superconducting transition temperature T is increased by almost an order of magnitude, raising questions on the pairing mechanism. As in other superconductors, antiferromagnetic spin fluctuations have been proposed to mediate SC making it essential to study the evolution of the spin dynamics of FeSe from the bulk to the ultrathin limit. Here, we investigate the spin excitations in bulk and monolayer FeSe/STO using resonant inelastic x-ray scattering (RIXS) and quantum Monte Carlo (QMC) calculations. Despite the absence of long-range magnetic order, bulk FeSe displays dispersive magnetic excitations reminiscent of other Fe-pnictides. Conversely, the spin excitations in FeSe/STO are gapped, dispersionless, and significantly hardened relative to its bulk counterpart. By comparing our RIXS results with simulations of a bilayer Hubbard model, we connect the evolution of the spin excitations to the Fermiology of the two systems revealing a remarkable reconfiguration of spin excitations in FeSe/STO, essential to understand the role of spin fluctuations in the pairing mechanism.

  • Targeting Bcl-2 stability to sensitize cells harboring oncogenic ras

    • The pro-survival factor Bcl-2 and its family members are critical determinants of the threshold of the susceptibility of cells to apoptosis. Studies are shown that cells harboring an oncogenic ras were extremely sensitive to the inhibition of protein kinase C (PKC) and Bcl-2 could antagonize this apoptotic process. However, it remains unrevealed how Bcl-2 is being regulated in this apoptotic process. In this study, we investigate the role of Bcl-2 stability in sensitizing the cells harboring oncogenic K-ras to apoptosis triggered by PKC inhibitor GO6976. We demonstrated that Bcl-2 in Swiss3T3 cells ectopically expressing or murine lung cancer LKR cells harboring K-ras rapidly underwent ubiquitin-dependent proteasome pathway after the treatment of GO6976, accompanied with induction of apoptosis. In this process, Bcl-2 formed the complex with Keap-1 and Cul3. The mutation of serine-17 and deletion of BH-2 or 4 was required for Bcl-2 ubiquitination and degradation, which elevate the signal threshold for the induction of apoptosis in the cells following PKC inhibition. Thus, Bcl-2 appears an attractive target for the induction of apoptosis by PKC inhibition in cancer cells expressing oncogenic K-ras.
