  • The Type Icn SN 2021csp : Implications for the Origins of the Fastest Supernovae and the Fates of Wolf-Rayet Stars

    • We present observations of SN 2021csp, the second example of a newly identified type of supernova (SN) hallmarked by strong, narrow, P Cygni carbon features at early times (Type Icn). The SN appears as a fast and luminous blue transient at early times, reaching a peak absolute magnitude of −20 within 3 days due to strong interaction between fast SN ejecta (v ≈ 30,000 km s−1) and a massive, dense, fast-moving C/O wind shed by the WC-like progenitor months before explosion. The narrow-line features disappear from the spectrum 10–20 days after explosion and are replaced by a blue continuum dominated by broad Fe features, reminiscent of Type Ibn and IIn supernovae and indicative of weaker interaction with more extended H/He-poor material. The transient then abruptly fades ∼60 days post-explosion when interaction ceases. Deep limits at later phases suggest minimal heavy-element nucleosynthesis, a low ejecta mass, or both, and imply an origin distinct from that of classical Type Ic SNe. We place SN 2021csp in context with other fast-evolving interacting transients, and discuss various progenitor scenarios: an ultrastripped progenitor star, a pulsational pair-instability eruption, or a jet-driven fallback SN from a Wolf–Rayet (W-R) star. The fallback scenario would naturally explain the similarity between these events and radio-loud fast transients, and suggests a picture in which most stars massive enough to undergo a W-R phase collapse directly to black holes at the end of their lives.

  • The Challenges Ahead for Multimessenger Analyses of Gravitational Waves and Kilonova : A Case Study on GW190425

    • In recent years, there have been significant advances in multimessenger astronomy due to the discovery of the first, and so far only confirmed, gravitational wave event with a simultaneous electromagnetic (EM) counterpart, as well as improvements in numerical simulations, gravitational wave (GW) detectors, and transient astronomy. This has led to the exciting possibility of performing joint analyses of the GW and EM data, providing additional constraints on fundamental properties of the binary progenitor and merger remnant. Here, we present a new Bayesian framework that allows inference of these properties, while taking into account the systematic modeling uncertainties that arise when mapping from GW binary progenitor properties to photometric light curves. We extend the relative binning method presented in Zackay et al. to include extrinsic GW parameters for fast analysis of the GW signal. The focus of our EM framework is on light curves arising from r-process nucleosynthesis in the ejected material during and after merger, the so-called kilonova, and particularly on black hole-neutron star systems. As a case study, we examine the recent detection of GW190425, where the primary object is consistent with being either a black hole or a neutron star. We show quantitatively how improved mapping between binary progenitor and outflow properties, and/or an increase in EM data quantity and quality are required in order to break degeneracies in the fundamental source parameters.

  • A multi-stage genome-wide association study of bladder cancer identifies multiple susceptibility loci

    • We conducted a multi-stage, genome-wide association study of bladder cancer with a primary scan of 591,637 SNPs in 3,532 affected individuals (cases) and 5,120 controls of European descent from five studies followed by a replication strategy, which included 8,382 cases and 48,275 controls from 16 studies. In a combined analysis, we identified three new regions associated with bladder cancer on chromosomes 22q13.1, 19q12 and 2q37.1: rs1014971, (P = 8 × 10⁻¹²) maps to a non-genic region of chromosome 22q13.1, rs8102137 (P = 2 × 10⁻¹¹) on 19q12 maps to CCNE1 and rs11892031 (P = 1 × 10⁻⁷) maps to the UGT1A cluster on 2q37.1. We confirmed four previously identified genome-wide associations on chromosomes 3q28, 4p16.3, 8q24.21 and 8q24.3, validated previous candidate associations for the GSTM1 deletion (P = 4 × 10⁻¹¹) and a tag SNP for NAT2 acetylation status (P = 4 × 10⁻¹¹), and found interactions with smoking in both regions. Our findings on common variants associated with bladder cancer risk should provide new insights into the mechanisms of carcinogenesis.

  • Breastfeeding shows a protective trend toward adolescents with higher abdominal adiposity

    • <b><i>Objective: </i></b>The question of whether breastfeeding has a protective effect against the development of overweight or obesity later in life remains controversial, especially during adolescence. The objective was to assess the relationship between breastfeeding and adolescents' body composition. <b><i>Methods: </i></b>The HELENA study is a cross-sectional study involving 3,528 adolescents from 10 European cities. The outcome measures were body weight and height, subscapular skinfolds as well as waist circumferences. Breastfeeding, smoking status, and parental socioeconomic status were assessed by self-administered questionnaires. Dietary intake was recorded using two 24-hour recall surveys. Two adjustment approaches were used: i) covariance analysis adjusted for confounding factors (propensity score adjustment) and ii) multivariate quantile regression. <b><i>Results: </i></b>After adjustment, no significant associations were observed between breastfeeding and body composition parameters (BMI Z-score; sum of skinfolds; waist-to-height ratio). An adjusted quantile regression analysis showed a non-significant trend for a protective effect of breastfeeding toward the highest percentiles of adiposity in boys but not in girls. This is of particular interest with respect to the superiority of the waist-to-height ratio over waist circumference and BMI for detecting cardiometabolic risk factors. <b><i>Conclusion: </i></b>This first European study, including a large set of factors influencing adolescents' body composition, showed a non-significant trend toward a protective effect of breastfeeding on highest percentiles of adolescent's abdominal adiposity.

    • This is the second iteration of the European Society of Cardiology (ESC) and European Association for the Study of Diabetes (EASD) joining forces to write guidelines on the management of diabetes mellitus (DM), pre-diabetes, and cardiovascular disease (CVD), designed to assist clinicians and other healthcare workers to make evidence-based management decisions. The growing awareness of the strong biological relationship between DM and CVD rightly prompted these two large organizations to collaborate to generate guidelines relevant to their joint interests, the first of which were published in 2007. Some assert that too many guidelines are being produced but, in this burgeoning field, five years in the development of both basic and clinical science is a long time and major trials have reported in this period, making it necessary to update the previous Guidelines.

    • The fourth workshop of the Multidisciplinary European Low Dose Initiative (MELODI) was organised by STUK-Radiation and Nuclear Safety Authority of Finland. It took place from 12 to 14 September 2012 in Helsinki, Finland. The meeting was attended by 179 scientists and professionals engaged in radiation research and radiation protection. We summarise the major scientific findings of the workshop and the recommendations for updating the MELODI Strategic Research Agenda and Road Map for future low dose research activities.

    • The gut epithelium serves to maximize the surface for nutrient and fluid uptake, but at the same time must provide a tight barrier to pathogens and remove damaged intestinal epithelial cells (IECs) without jeopardizing barrier integrity. How the epithelium coordinates these tasks remains a question of significant interest. We used imaging and an optical flow analysis pipeline to study the dynamicity of untransformed murine and human intestinal epithelia, cultured atop flexible hydrogel supports. Infection with the pathogen Salmonella Typhimurium (S.Tm) within minutes elicited focal contractions with inward movements of up to similar to 1,000 IECs. Genetics approaches and chimeric epithelial monolayers revealed contractions to be triggered by the NAIP/NLRC4 inflammasome, which sensed type-III secretion system and flagellar ligands upon bacterial invasion, converting the local tissue into a contraction epicenter. Execution of the response required swift sublytic Gasdermin D pore formation, ion fluxes, and the propagation of a myosin contraction pulse across the tissue. Importantly, focal contractions preceded, and could be uncoupled from, the death and expulsion of infected IECs. In both two-dimensional monolayers and three-dimensional enteroids, multiple infection-elicited contractions coalesced to produce shrinkage of the epithelium as a whole. Monolayers deficient for Caspase-1(-11) or Gasdermin D failed to elicit focal contractions but were still capable of infected IEC death and expulsion. Strikingly, these monolayers lost their integrity to a markedly higher extent than wild-type counterparts. We propose that prompt NAIP/NLRC4/Caspase-1/Gasdermin D/myosin-dependent contractions allow the epithelium to densify its cell packing in infected regions, thereby preventing tissue disintegration due to the subsequent IEC death and expulsion process.
