    • Steady-state absorption and femtosecond time-resolved spectroscopic studies have been carried out on astaxanthin dissolved in CS2, methanol, and acetonitrile, and in purified alpha-crustacyanin. The spectra of the S-0 -> S-2 and S-1 -> S-n transitions were found to be similarly dependent on solvent environment. The dynamics of the excited-state decay processes were analyzed with both single wavelength and global fitting procedures. In solution, the S-1 lifetime of astaxanthin was found to be similar to 5 ps and independent of solvent. In alpha-crustacyanin, the lifetime was noticeably shorter at similar to 1.8 ps. Both fitting procedures led to the conclusion that the lifetime of the S-2 state was either comparable to or shorter than the instrument response time. The data support the idea that dimerization of astaxanthin in alpha-crustacyanin is the primary molecular basis for the bathochromic shift of the S-0 -> S-2 and S-1 - S-n transitions. Planarization of the astaxanthin molecule, which leads to a longer effective pi-electron conjugated chain and a lower S-1 energy, accounts for the shorter tau(1) in the protein.

  • A BALB/c IGHV Reference Set, Defined by Haplotype Analysis of Long-Read VDJ-C Sequences From F1 (BALB/c x C57BL/6) Mice

    • The immunoglobulin genes of inbred mouse strains that are commonly used in models of antibody-mediated human diseases are poorly characterized. This compromises data analysis. To infer the immunoglobulin genes of BALB/c mice, we used long-read SMRT sequencing to amplify VDJ-C sequences from F1 (BALB/c x C57BL/6) hybrid animals. Strain variations were identified in the Ighm and Ighg2b genes, and analysis of VDJ rearrangements led to the inference of 278 germline IGHV alleles. 169 alleles are not present in the C57BL/6 genome reference sequence. To establish a set of expressed BALB/c IGHV germline gene sequences, we computationally retrieved IGHV haplotypes from the IgM dataset. Haplotyping led to the confirmation of 162 BALB/c IGHV gene sequences. A musIGHV398 pseudogene variant also appears to be present in the BALB/cByJ substrain, while a functional musIGHV398 gene is highly expressed in the BALB/cJ substrain. Only four of the BALB/c alleles were also observed in the C57BL/6 haplotype. The full set of inferred BALB/c sequences has been used to establish a BALB/c IGHV reference set, hosted at https://ogrdb.airr-community.org. We assessed whether assemblies from the Mouse Genome Project (MGP) are suitable for the determination of the genes of the IGH loci. Only 37 (43.5%) of the 85 confirmed IMGT-named BALB/c IGHV and 33 (42.9%) of the 77 confirmed non-IMGT IGHV were found in a search of the MGP BALB/cJ genome assembly. This suggests that current MGP assemblies are unsuitable for the comprehensive documentation of germline IGHVs and more efforts will be needed to establish strain-specific reference sets.

  • Clonal Hematopoiesis and risk of atherosclerotic cardiovascular disease

    • BACKGROUND Clonal hematopoiesis of indeterminate potential (CHIP), which is defined as the presence of an expanded somatic blood-cell clone in persons without other hematologic abnormalities, is common among older persons and is associated with an increased risk of hematologic cancer. We previously found preliminary evidence for an association between CHIP and atherosclerotic cardiovascular disease, but the nature of this association was unclear. METHODS We used whole-exome sequencing to detect the presence of CHIP in peripheral-blood cells and associated such presence with coronary heart disease using samples from four case-control studies that together enrolled 4726 participants with coronary heart disease and 3529 controls. To assess causality, we perturbed the function of Tet2, the second most commonly mutated gene linked to clonal hematopoiesis, in the hematopoietic cells of atherosclerosis-prone mice. RESULTS In nested case-control analyses from two prospective cohorts, carriers of CHIP had a risk of coronary heart disease that was 1.9 times as great as in noncarriers (95% confidence interval [CI], 1.4 to 2.7). In two retrospective case-control cohorts for the evaluation of early-onset myocardial infarction, participants with CHIP had a risk of myocardial infarction that was 4.0 times as great as in noncarriers (95% CI, 2.4 to 6.7). Mutations in DNMT3A, TET2, ASXL1, and JAK2 were each individually associated with coronary heart disease. CHIP carriers with these mutations also had increased coronary-artery calcification, a marker of coronary atherosclerosis burden. Hypercholesterolemia-prone mice that were engrafted with bone marrow obtained from homozygous or heterozygous Tet2 knockout mice had larger atherosclerotic lesions in the aortic root and aorta than did mice that had received control bone marrow. Analyses of macrophages from Tet2 knockout mice showed elevated expression of several chemokine and cytokine genes that contribute to atherosclerosis. CONCLUSIONS The presence of CHIP in peripheral-blood cells was associated with nearly a doubling in the risk of coronary heart disease in humans and with accelerated atherosclerosis in mice.   • Energy production and water savings from floating solar photovoltaics on global reservoirs

    • Solutions that can support multiple sustainability goals related to clean energy, and resource use efficiency, will be crucial in the near future. The study estimates the potential of floating solar panels on reservoirs globally to generate renewable energy, reduce water losses and conserve land. Growing global energy use and the adoption of sustainability goals to limit carbon emissions from fossil fuel burning are increasing the demand for clean energy, including solar. Floating photovoltaic (FPV) systems on reservoirs are advantageous over traditional ground-mounted solar systems in terms of land conservation, efficiency improvement and water loss reduction. Here, based on multiple reservoir databases and a realistic climate-driven photovoltaic system simulation, we estimate the practical potential electricity generation for FPV systems with a 30% coverage on 114,555 global reservoirs is 9,434 +/- 29 TWh yr(-1). Considering the proximity of most reservoirs to population centres and the potential to develop dedicated local power systems, we find that 6,256 communities and/or cities in 124 countries, including 154 metropolises, could be self-sufficient with local FPV plants. Also beneficial to FPV worldwide is that the reduced annual evaporation could conserve 106 +/- 1 km(3) of water. Our analysis points to the huge potential of FPV systems on reservoirs, but additional studies are needed to assess the potential long-term consequences of large systems.

  • Climbing the cosmic ladder with stellar twins in RAVE with Gaia

    • We apply the twin method to determine parallaxes to 232 545 stars of the RAVE survey using the parallaxes of Gaia DR1 as a reference. To search for twins in this large data set, we apply the t-student stochastic neighbour embedding projection that distributes the data according to their spectral morphology on a two-dimensional map. From this map, we choose the twin candidates for which we calculate a χ2 to select the best sets of twins. Our results show a competitive performance when compared to other model-dependent methods relying on stellar parameters and isochrones. The power of the method is shown by finding that the accuracy of our results is not significantly affected if the stars are normal or peculiar since the method is model free. We find twins for 60 per cent of the RAVE sample that are not contained in Tycho-Gaia Astrometric Solution (TGAS) or that have TGAS uncertainties that are larger than 20 per cent. We could determine parallaxes with typical errors of 28 per cent. We provide a complementary data set for the RAVE stars not covered by TGAS, or that have TGAS uncertainties which are larger than 20 per cent, with model-free parallaxes scaled to the Gaia measurements.

    • One of the major goals of pharmacogenetics is to elucidate mechanisms and identify patients at increased risk of adverse events (AEs). To date, however, there have been only a few successful examples of this type of approach. In this paper, we describe a retrospective case–control pharmacogenetic study of an AE of unknown mechanism, characterized by elevated levels of serum alanine aminotransferase (ALAT) during long-term treatment with the oral direct thrombin inhibitor ximelagatran. The study was based on 74 cases and 130 treated controls and included both a genome-wide tag single nucleotide polymorphism and large-scale candidate gene analysis. A strong genetic association between elevated ALAT and the MHC alleles DRB1*07 and DQA1*02 was discovered and replicated, suggesting a possible immune pathogenesis. Consistent with this hypothesis, immunological studies suggest that ximelagatran may have the ability to act as a contact sensitizer, and hence be able to stimulate an adaptive immune response.

    • We report on the synthesis, electrochemical, photophysical, and photovoltaic properties of a series of three organic dyads comprising a perylene monoimide (PMI) dye connected to a naphthalene diimide (NDI) or a fullerene (C-60) for application in dye-sensitized solar cells (DSCs) with nanocrystalline NiO electrodes. It was found that the secondary electron acceptor (NDI or C-60) in all the three dyads extends the charge separated state lifetime by about five orders of magnitude compared to the respective parent PMI dye. Nanosecond pump-probe experiments of the NiO/dyads in the presence of the electrolyte show that the reduction of triiodide by the secondary electron acceptor is slow in all the dyads, which we ascribe to a weak driving force for this reaction. This reaction is significantly faster with the cobalt electrolyte (tris(4,4'-di-tert-butyl-2,2'-bipyridine)cobalt(II/III)), whose driving force is larger; however, its reaction with the reduced dyads is still rather slow. We demonstrate that the larger photovoltage observed with the cobalt electrolyte (V-OC = 285 mV) relative to the iodide electrolyte (V-OC = 120 mV) is due to a decrease in the dark current for the former owing to slower interfacial electron transfer of the reduced mediator with the injected holes into the NiO electrode. In terms of photovoltaic performances, the most efficient dyad is the system in which the NDI is directly connected to the PMI (eta = 0.14% under AM 1.5 with the cobalt electrolyte), but the dyad containing the fullerene acceptor exhibits the highest IPCE and the highest short circuit current density (IPCE = 57%, J(SC) = 1.88 mA cm(-2)) with the iodide electrolyte. The latter performances are attributed to the slightly stronger reducing power of C-60 relative to NDI, which favours the reduction of the mediator in the electrolyte.
