  • Cadmium Causes Misfolding and Aggregation of Cytosolic Proteins in Yeast

    • Cadmium is a highly poisonous metal and is classified as a human carcinogen. While its toxicity is undisputed, the underlying in vivo molecular mechanisms are not fully understood. Here, we demonstrate that cadmium induces aggregation of cytosolic proteins in living Saccharomyces cerevisiae cells. Cadmium primarily targets proteins in the process of synthesis or folding, probably by interacting with exposed thiol groups in not-yet-folded proteins. On the basis of in vitro and in vivo data, we show that cadmium-aggregated proteins form seeds that increase the misfolding of other proteins. Cells that cannot efficiently protect the proteome from cadmium-induced aggregation or clear the cytosol of protein aggregates are sensitized to cadmium. Thus, protein aggregation may contribute to cadmium toxicity. This is the first report on how cadmium causes misfolding and aggregation of cytosolic proteins in vivo. The proposed mechanism might explain not only the molecular basis of the toxic effects of cadmium but also the suggested role of this poisonous metal in the pathogenesis of certain protein-folding disorders.

  • High-spin spectroscopy of Nd-140

    • The population of the high-spin states in Nd-140 was investigated using the reaction Zr-96(Ca-48,4n). The results from two experiments, one with the EUROBALL array and one with the JUROGAM II + RITU + GREAT setup employing the recoil decay tagging technique, have been combined to develop a very detailed level scheme for Nd-140. Twelve bands of quadrupole transitions and eleven bands of dipole transitions were identified and their connections to low-lying states were established. Calculations using the cranked Nilsson-Strutinsky and the tilted axis cranking models were used to interpret the observed structures. The overall good agreement between the experimental results and the calculations assuming a triaxial shape of the nucleus strongly support the existence of a stable triaxial shape at high spins in this mass region.

  • First identification of rotational band structures in Re-166(75)91

    • Excited states in the odd-odd, highly neutron-deficient nucleus Re-166 have been investigated via the Mo-92(Kr-78, 3p1n)Re-166 reaction. Prompt gamma rays were detected by the JUROGAM II. gamma-ray spectrometer, and the recoiling fusion-evaporation products were separated by the recoil ion transport unit (RITU) gas-filled recoil separator and implanted into the Gamma Recoil Electron Alpha Tagging spectrometer located at the RITU focal plane. The tagging and coincidence techniques were applied to identify the gamma-ray transitions in Re-166, revealing two collective, strongly coupled rotational structures, for the first time. The more strongly populated band structure is assigned to the pi h(11/2)[514]9/2(-) circle times vi(13/2)[660]1/2(+) Nilsson configuration, while the weaker structure is assigned to be built on a two-quasiparticle state of mixed pi h(11/2)[514]9/2(-) circle times v[h(9/2)f(7/2)]3/2(-) character. The configuration assignments are based on the electromagnetic characteristics and rotational properties, in comparison with predictions from total Routhian surface and particle-rotor model calculations.

  • Recoil-decay tagging spectroscopy of W-162(74)88

    • Excited states in the highly neutron-deficient nucleus W-162 have been investigated via the Mo-92(Kr-78, 2 alpha) W-162 reaction. Prompt gamma rays were detected by the JUROGAM II high-purity germanium detector array and the recoiling fusion-evaporation products were separated by the recoil ion transport unit (RITU) gas-filled recoil separator and identified with the gamma recoil electron alpha tagging (GREAT) spectrometer at the focal plane of RITU. gamma rays from W-162 were identified uniquely using mother-daughter and mother-daughter-granddaughter alpha-decay correlations. The observation of a rotational-like ground-state band is interpreted within the framework of total Routhian surface (TRS) calculations, which suggest an axially symmetric ground-state shape with a gamma-soft minimum at beta(2) approximate to 0.15. Quasiparticle alignment effects are discussed based on cranked shell model calculations. New measurements of the W-162 ground-state alpha-decay energy and half-life were also performed. The observed alpha-decay energy agrees with previous measurements. The half-life of W-162 was determined to be t(1/2) = 990(30) ms. This value deviates significantly from the currently adopted value of t(1/2) = 1360(70) ms. In addition, the alpha-decay energy and half-life of Os-166 were measured and found to agree with the adopted values.

  • Hepatocyte-specific loss of GPS2 in mice reduces non-alcoholic steatohepatitis via activation of PPARα

    • Obesity triggers the development of non-alcoholic fatty liver disease (NAFLD), which involves alterations of regulatory transcription networks and epigenomes in hepatocytes. Here we demonstrate that G protein pathway suppressor 2 (GPS2), a subunit of the nuclear receptor corepressor (NCOR) and histone deacetylase 3 (HDAC3) complex, has a central role in these alterations and accelerates the progression of NAFLD towards non-alcoholic steatohepatitis (NASH). Hepatocyte-specific Gps2 knockout in mice alleviates the development of diet-induced steatosis and fibrosis and causes activation of lipid catabolic genes. Integrative cistrome, epigenome and transcriptome analysis identifies the lipid-sensing peroxisome proliferator-activated receptor α (PPARα, NR1C1) as a direct GPS2 target. Liver gene expression data from human patients reveal that Gps2 expression positively correlates with a NASH/fibrosis gene signature. Collectively, our data suggest that the GPS2-PPARα partnership in hepatocytes coordinates the progression of NAFLD in mice and in humans and thus might be of therapeutic interest.

  • The optical afterglow of the short gamma-ray burst associated with GW170817

    • The binary neutron star merger GW170817 was the first multi-messenger event observed in both gravitational and electromagnetic waves(1,2). The electromagnetic signal began approximately two seconds post-merger with a weak, short burst of gamma rays(3), which was followed over the next hours and days by the ultraviolet, optical and near-infrared emission from a radioactively powered kilonova(4-11). Later, non-thermal rising X-ray and radio emission was observed(12,13). The low luminosity of the gamma rays and the rising non-thermal flux from the source at late times could indicate that we are outside the opening angle of the beamed relativistic jet. Alternatively, the emission could be arising from a cocoon of material formed from the interaction between a jet and the merger ejecta(13-15). Here we present late-time optical detections and deep near-infrared limits on the emission from GW170817 at 110 days post-merger. Our new observations are at odds with expectations of late-time emission from kilonova models, being too bright and blue(16,17). Instead, the emission arises from the interaction between the relativistic ejecta of GW170817 and the interstellar medium. We show that this emission matches the expectations of a Gaussian-structured relativistic jet, which would have launched a high-luminosity, short gamma-ray burst to an aligned observer. However, other jet structure or cocoon models can also match current data-the future evolution of the afterglow will directly distinguish the origin of the emission.

  • High precision astrometry mission for the detection and characterization of nearby habitable planetary systems with the Nearby Earth Astrometric Telescope (NEAT)

    • A complete census of planetary systems around a volume-limited sample of solar-type stars (FGK dwarfs) in the Solar neighborhood (d a parts per thousand currency signaEuro parts per thousand 15 pc) with uniform sensitivity down to Earth-mass planets within their Habitable Zones out to several AUs would be a major milestone in extrasolar planets astrophysics. This fundamental goal can be achieved with a mission concept such as NEAT-the Nearby Earth Astrometric Telescope. NEAT is designed to carry out space-borne extremely-high-precision astrometric measurements at the 0.05 mu as (1 sigma) accuracy level, sufficient to detect dynamical effects due to orbiting planets of mass even lower than Earth's around the nearest stars. Such a survey mission would provide the actual planetary masses and the full orbital geometry for all the components of the detected planetary systems down to the Earth-mass limit. The NEAT performance limits can be achieved by carrying out differential astrometry between the targets and a set of suitable reference stars in the field. The NEAT instrument design consists of an off-axis parabola single-mirror telescope (D = 1 m), a detector with a large field of view located 40 m away from the telescope and made of 8 small movable CCDs located around a fixed central CCD, and an interferometric calibration system monitoring dynamical Young's fringes originating from metrology fibers located at the primary mirror. The mission profile is driven by the fact that the two main modules of the payload, the telescope and the focal plane, must be located 40 m away leading to the choice of a formation flying option as the reference mission, and of a deployable boom option as an alternative choice. The proposed mission architecture relies on the use of two satellites, of about 700 kg each, operating at L2 for 5 years, flying in formation and offering a capability of more than 20,000 reconfigurations. The two satellites will be launched in a stacked configuration using a Soyuz ST launch vehicle. The NEAT primary science program will encompass an astrometric survey of our 200 closest F-, G- and K-type stellar neighbors, with an average of 50 visits each distributed over the nominal mission duration. The main survey operation will use approximately 70% of the mission lifetime. The remaining 30% of NEAT observing time might be allocated, for example, to improve the characterization of the architecture of selected planetary systems around nearby targets of specific interest (low-mass stars, young stars, etc.) discovered by Gaia, ground-based high-precision radial-velocity surveys, and other programs. With its exquisite, surgical astrometric precision, NEAT holds the promise to provide the first thorough census for Earth-mass planets around stars in the immediate vicinity of our Sun.

    • The number of supernovae known to be connected with long-duration gamma-ray bursts (GRBs) is increasing and the link between these events is no longer exclusively found at low redshift (z less than or similar to 0.3) but is well established also at larger distances. We present a new case of such a liaison at z = 0.33 between GRB171010A and SN 2017htp. It is the second closest GRB with an associated supernova of only three events detected by Fermi-LAT. The supernova is one of the few higher redshift cases where spectroscopic observations were possible and shows spectral similarities with the well-studied SN 1998bw, having produced a similar Ni mass (M-Ni = 0.33 +/- 0.02 M-circle dot) with slightly lower ejected mass (M-ej = 4.1 +/- 0.7 M-circle dot) and kinetic energy (E-K = 8.1 +/- 2.5 x 10(51) erg). The host-galaxy is bigger in size than typical GRB host galaxies, but the analysis of the region hosting the GRB revealed spectral properties typically observed in GRB hosts and showed that the progenitor of this event was located in a very bright H II region of its face-on host galaxy, at a projected distance of similar to 10 kpc from its galactic centre. The star-formation rate (SFRGRB similar to 0.2 M-circle dot yr(-1)) and metallicity (12 + log(O/H) similar to 8.15 +/- 0.10) of the GRB star-forming region are consistent with those of the host galaxies of previously studied GRB-SN systems.
