  • Rapid measurement of heteronuclear transverse relaxation rates using non-uniformly sampled R1ρ accordion experiments

    • Multidimensional, heteronuclear NMR relaxation methods are used extensively to characterize the dynamics of biological macromolecules. Acquisition of relaxation datasets on proteins typically requires significant measurement time, often several days. Accordion spectroscopy offers a powerful means to shorten relaxation rate measurements by encoding the “relaxation dimension” into the indirect evolution period in multidimensional experiments. Time savings can also be achieved by non-uniform sampling (NUS) of multidimensional NMR data, which is used increasingly to improve spectral resolution or increase sensitivity per unit time. However, NUS is not commonly implemented in relaxation experiments, because most reconstruction algorithms are inherently nonlinear, leading to problems when estimating signal intensities, relaxation rate constants and their error bounds. We have previously shown how to avoid these shortcomings by combining accordion spectroscopy with NUS, followed by data reconstruction using sparse exponential mode analysis, thereby achieving a dramatic decrease in the total length of longitudinal relaxation experiments. Here, we present the corresponding transverse relaxation experiment, taking into account the special considerations required for its successful implementation in the framework of the accordion-NUS approach. We attain the highest possible precision in the relaxation rate constants by optimizing the NUS scheme with respect to the Cramér-Rao lower bound of the variance of the estimated parameter, given the total number of sampling points and the spectrum-specific signal characteristics. The resulting accordion-NUS R1ρ relaxation experiment achieves comparable precision in the parameter estimates compared to conventional CPMG (Carr-Purcell-Meiboom-Gill) R2 or spin-lock R1ρ experiments while saving an order of magnitude in experiment time.

    • Insects that hibernate as adults have a life span of almost a whole year. Hence, they must have extraordinary adaptations for adult survival. In this paper, we study winter survival in two butterflies that hibernate as adults and have multimodal anti-predator defences-the peacock, Inachisio, which has intimidating eyespots that are effective against bird predation, and the small tortoiseshell, Aglais urticae, which does not have an effective secondary defence against birds. We assessed predation on wild butterflies hibernating in the attic of an unheated house, as well as survival of individually marked butterflies placed by hand on different sites in the attic. Our objectives were to assess (1) the number of butterflies that were killed during hibernation, (2) whether survival differed between butterfly species, and (3) how predation was related to hibernation site and the identity of the predator. There was a strong pulse of predation during the first 2 weeks of hibernation: 58% of A. urticae and 53% of I. io were killed during this period. Thereafter, predation decreased and butterfly survival equalled 98% during the final 16 weeks of hibernation. There was no difference in survival between the two butterfly species, but predation was site-specific and more pronounced under light conditions in locations accessible to a climbing rodent, such as the common yellow-necked mouse, Apodemus flavicollis. We contend that small rodents are likely important predators on over-wintering butterflies, both because rodents are active throughout winter when butterflies are torpid and because they occur at similar sites

    • Adrenocortical carcinoma (ACC) is a rare tumour disease with sinister prognosis also after attempts to radical surgery; better prognosis is seen for low-stage tumours. Adjuvant treatment with the adrenolytic drug mitotane has been attempted, but not proven to prevent from recurrence. The drug may offer survival advantage in case of recurrence. The aim of this single-centre study (1979-2007) of 43 consecutive patients was to evaluate the long-term survival after active surgical treatment combined with monitored mitotane (to reduce side effects of the drug). The series is unique, since all patients were offered a period of mitotane as adjuvant or palliative treatment; six patients refused mitotane. Despite a high proportion of high-stage tumours (67%), the complete resection rate was high (77%). The disease-specific 5-year survival was high (64.1%); very high for patients with low-stage tumours without evident relation to mitotane levels. Patients with high-stage tumours had a clear survival advantage with mitotane levels above a threshold of 14 mg/l in serum. The hazard ratio for patients with high mitotane levels versus all patients indicates a significant effect of the drug. The results indicate that adjuvant mitotane may be the standard of care for patients with high-stage ACC after complete resection.

    • The MYC genes are the most frequently activated oncogenes in human tumors and are hence attractive therapeutic targets. MYCN amplification leads to poor clinical outcome in childhood neuroblastoma, yet strategies to modulate the function of MYCN do not exist. Here we show that 10058-F4, a characterized c-MYC/Max inhibitor, also targets the MYCN/Max interaction, leading to cell cycle arrest, apoptosis, and neuronal differentiation in MYCN-amplified neuroblastoma cells and to increased survival of MYCN transgenic mice. We also report the discovery that inhibition of MYC is accompanied by accumulation of intracellular lipid droplets in tumor cells as a direct consequence of mitochondrial dysfunction. This study expands on the current knowledge of how MYC proteins control the metabolic reprogramming of cancer cells, especially highlighting lipid metabolism and the respiratory chain as important pathways involved in neuroblastoma pathogenesis. Together our data support direct MYC inhibition as a promising strategy for the treatment of MYC-driven tumors.
