  • Synthesizing implications of a collaborative project on socially sustainable housing policies for the aging population

    • Objective: Based on findings from four transdisciplinary original research studies on housing issues for the aging population, whereof three had a particular focus on marginalized groups, we report a co-produced synthesis of implications from a collaborative research project on socially sustainable housing policies. Researchers and non-academic partners in the ongoing Thematic Collaboration Initiative Social Rights and Housing for the Aging Population at Lund University collaborated in co-creative activities aiming at policy recommendations. Results: Seven types of implications (i.e., themes) that represent macro and meso level approaches with potential to generate impact on social rights and housing for the aging population were identified. The content of legislation and regulations, financial institution strategies, and housing and neighbourhood development exemplify macro level implications. The three themes education and training, communicating with a diversity of stakeholders, and communicating with the public all relate to an overall need for integrated knowledge translation. The theme involving older adults as a resource delivered insight into underutilized capacities of the diverse target group. As the quest for integrated knowledge translation is growing stronger, this research note contributes to development of co-production approaches to synthesize implications of transdisciplinary collaboration, connecting research, practice and policy on societal challenges that ay population aging.

    • Several technological challenges delay the adoption of electrified powertrains in the heavy-duty transport sector. For fuel-cell hybrid electric trucks, key issues include slow cold start, reduced cooling power during high ambient temperatures, and uncertainties regarding durability. In addition, the engineers must handle the complexity of the system. In this article, a Matlab/Simulink library is introduced, which has been developed to aid engineers in the design and optimization of energy management systems and strategies of this complex system that consider mechanical, electrochemical, and thermal energy flows. The library is introduced through five example vehicle models, and through case studies that highlight the various kinds of analysis that can be performed using the provided models. All library code is open source, open for commercial use, and runs in Matlab/Simulink without any need for external libraries.

  • Protein Kinase R Is Constitutively Expressed in the Human Pancreas

    • Viral infection of the insulin-producing cells in the pancreas has been proposed in the etiology of type 1 diabetes. Protein kinase R (PKR) is a cytoplasmic protein activated through phosphorylation in response to cellular stress and particularly viral infection. As PKR expression in pancreatic beta-cells has been interpreted as a viral footprint, this cross-sectional study aimed at characterizing the PKR expression in non-diabetic human pancreases. PKR expression was evaluated in pancreas tissue from 16 non-diabetic organ donors, using immunohistochemistry, qPCR, and western blot. Immunohistochemistry and western blot showed readily detectable PKR expression in the pancreatic parenchyma. The qPCR detected PKR mRNA in both endocrine and exocrine samples, with a slightly higher expression in the islets. In conclusion, PKR is constitutively expressed in both endocrine and exocrine parts of the pancreas and its expression should not be interpreted as a viral footprint in pancreatic beta cells.

    • In experimental studies, pancreatic islet microvasculature is essential for islet endocrine function and mass, and islet vascular morphology is altered in diabetic subjects. Even so, almost no information is available concerning human islet microvascular endothelial cell (MVEC) physiology and gene expression. In this study, islets and exocrine pancreatic tissue were acquired from organ donors with normoglycemia or impaired glucose metabolism (IGM) immediately after islet isolation. Following single-cell dissociation, primary islet- and exocrine MVECs were obtained through fluorescence-activated cell sorting (FACS) and transcriptional profiles were generated using AmpliSeq. Multiple gene sets involved in general vascular development and extracellular matrix remodeling were enriched in islet MVEC. In exocrine MVEC samples, multiple enriched gene sets that relate to biosynthesis and biomolecule catabolism were found. No statistically significant enrichment was found in gene sets related to autophagy or endoplasmic reticulum (ER) stress. Although ample differences were found between islet- and exocrine tissue endothelial cells, no differences could be observed between normoglycemic donors and donors with IGM at gene or gene set level. Our data is consistent with active angiogenesis and vascular remodeling in human islets and support the notion of ongoing endocrine pancreas tissue repair and regeneration even in the adult human.

    • Mini-abstract The objective of this study was to review and describe the current state of Swedish osteoporosis care and to highlight ongoing challenges. This report encompasses quantitative health outcomes based on Swedish registry data as well as organizational and management aspects. Executive summary Swedish osteoporosis care is characterized by a significant burden of disease, difficulties in identifying high-risk patients, and fragmented pathways for patients in need of secondary fracture prevention. This report aimed to describe the current state, gaps, and challenges in Swedish osteoporosis care, using Swedish national databases, questionnaires, and interviews with healthcare representatives. A secondary aim was to develop quality and process measures to compare differences between counties and to use those measures to describe the interaction between quantitative health outcomes and aspects of care organization and management. In conjunction with fractures, a considerably smaller proportion of men are treated than women, and a smaller proportion of older women are treated compared to younger groups. Between 3 and 16 % of patients receive treatment after a fracture, and the treatment rate in this patient group can likely increase. In addition to an unsatisfactory treatment rate, a limited number of those treated continue treatment throughout the recommended treatment durations, leading to increased risk of fracture. With a substantial variation between counties, there is a clear difficulty to identify non-persistent patients and switch to an alternative treatment. Collaboration around the patient across specialties has been lacking, and systems for secondary prevention have been concentrated to a few counties. However, when this study was conducted, there was a general trend towards implementing regional care programs. This report suggests possible strategies for improving quality of care and, hopefully, it can provide a basis for future evaluations and regional improvement of osteoporosis care in Sweden and other countries.

  • Experimental and Numerical Study of Laminar-Turbulent Transition on a Low-Pressure Turbine Outlet Guide Vane

    • In modern commercial aviation engines, the low-pressure turbine (LPT) has a high outlet swirl to maximize turbine power to weight ratio. Downstream of the last LPT rotor is the turbine rear structure (TRS) that with relatively few low-aspect-ratio outlet guide vanes (OGV) de-swirls the flow to maximize the thrust. The performance of the TRS is strongly connected to secondary flow structures, which in turn are strongly influenced by the laminar-turbulent transition. Transition can be challenging to predict in turbomachinery due to the highly complex flow present. At the design point the TRS can have both by-pass transition and laminar separation with transition and a following turbulent reattachment. In addition, a TRS needs to perform well in a large off-design envelope, with large variations of the inlet swirl angle. Accurately predicting transition, both at the design point and in important off-design points, is critical for the development of future TRS modules. In modern geared and ultra-high by-pass engines the TRS swirl angle off-design requirements are also increasing. There are several available transition models in RANS simulations and most of them need parameter tuning when introduced to new conditions. Evaluation of these models for different turbomachinery components is relatively well covered in the literature even though the model specifics often is a classified property of engine manufacturers. However, there are no cases in the literature of transition prediction with experimental verification in the TRS at engine-realistic conditions. This work presents the first experimental verification of laminar-turbulent transition in a TRS module tested in the LPT-OGV experimental facility at Chalmers Laboratory of Thermal and Fluid Science. The facility is a semi-closed rig using a rotating 1.5 stage shrouded low-pressure turbine stage to create engine representative inlet conditions for the TRS downstream of the LPT stage. Transition was measured using differential IR-thermography (DIT) which is a non-intrusive two-dimensional measurement technique. The technique was specially developed at Chalmers for this particular purpose and validated by boundary layer hot-wire measurements. The numerical analysis was done using commercially available transition models in Fluent and Ansys CFX. Gamma-theta transition model was used with the k-omega SST turbulence model. Experiments and numerical simulations were performed at a chord Reynold number of 235000 and with LPT outlet swirl angles covering both the design point (ADP) and relevant off-design points. Numerical and experimental results show that agreement between transition models and experiments can be achieved at these conditions. Boundary layers on the pressure side and suction side undergo laminar-turbulent transition for the selected test range. At decreased OGV aerodynamic load, the boundary layer on the pressure side near the leading edge is laminar along most of the span. At higher OGV loads the secondary flow is influencing the region near the shroud on the pressure side as well as near the hub on the suction side. The transition on the suction side midspan is significantly influenced by the vane load. The numerical analysis was used to better understand the involved flow mechanisms.
