Genome-wide association study identifies variation at 6q25.1 associated with survival in multiple myeloma.

- Survival following a diagnosis of multiple myeloma (MM) varies between patients and some of these differences may be a consequence of inherited genetic variation. In this study, to identify genetic markers associated with MM overall survival (MM-OS), we conduct a meta-analysis of four patient series of European ancestry, totalling 3,256 patients with 1,200 MM-associated deaths. Each series is genotyped for ∼600,000 single nucleotide polymorphisms across the genome; genotypes for six million common variants are imputed using 1000 Genomes Project and UK10K as the reference. The association between genotype and OS is assessed by Cox proportional hazards model adjusting for age, sex, International staging system and treatment. We identify a locus at 6q25.1 marked by rs12374648 associated with MM-OS (hazard ratio=1.34, 95% confidence interval=1.22-1.48, P=4.69 × 10(-9)). Our findings have potential clinical implications since they demonstrate that inherited genotypes can provide prognostic information in addition to conventional tumor acquired prognostic factors.

Effects of source material on epitaxial growth of fluorescent SiC

- The growth of fluorescent SiC using Fast Sublimation Growth Process was demonstrated using different types of SiC source materials. These were prepared by (i) high-temperature hot pressing, (ii) chemical vapor deposition and (iii) physical vapor transport. The optimized growth rates of 50 μm/h, 170 μm/h and 200 μm/h were achieved using the three types of sources, respectively. The best results in respect to growth rates are obtained using higher density sources. Fluorescent SiC layers with mirror-like morphology, very good crystal quality and yellowish or warm white light photoluminescence at room temperature were grown using all three types of the source materials.

A Randomized Controlled Trial on the Effects of a 12-Week High- vs. Low-Intensity Exercise Intervention on Hippocampal Structure and Function in Healthy, Young Adults

- Physical exercise affects hippocampal structure and function, but the underlying neural mechanisms and the effects of exercise intensity remain incompletely understood. Therefore, we undertook a comprehensive, multi-modal 3T and 7T MRI randomized controlled trial (Netherlands Trial Register - NL5847) in which we randomized 52 young, non-athletic volunteers to a 12-week low- or high-intensity exercise program. Using state-of-the-art methods, we investigated changes in hippocampal volume, as well as changes in vasculature, neuro-metabolites, and peripheral growth factors as potential underpinnings. Cardiorespiratory fitness improved over time (p < 0.001), but no interaction with exercise intensity was found (p = 0.48). Accordingly, we did not observe significant interactions between exercise condition and time on MRI measures (all p > 0.06). However, we found a significant decrease in right hippocampal volume (p < 0.01), an increase in left hippocampal glutathione (p < 0.01), and a decrease of left hippocampal cerebral blood volume (p = 0.01) over time, regardless of exercise condition. Additional exploratory analyses showed that changes in brain-derived neurotrophic factor (p = 0.01), insulin-like growth-factor (p = 0.03), and dorsal anterior cingulate cortex N-acetyl-aspartate levels (p = 0.01) were positively associated with cardiorespiratory fitness changes. Furthermore, a trend toward a positive association of fitness and gray-matter cerebral blood flow (p = 0.06) was found. Our results do not provide evidence for differential effects between high-intensity (aerobic) and low-intensity (toning) exercise on hippocampal structure and function in young adults. However, we show small but significant effects of exercise on hippocampal volume, neurometabolism and vasculature across exercise conditions. Moreover, our exploratory results suggest that exercise might not specifically only benefit hippocampal structure and function, but rather has a more widespread effect. These findings suggest that, in agreement with previous MRI studies demonstrating moderate to strong effects in elderly and diseased populations, but none to only mild effects in young healthy cohorts, the benefits of exercise on the studied brain measures may be age-dependent and restorative rather than stimulatory. Our study highlights the importance of a multi-modal, whole-brain approach to assess macroscopic and microscopic changes underlying exercise-induced brain changes, to better understand the role of exercise as a potential non-pharmacological intervention.

Pharmaceutical consumption patterns in four coastal regions of the South Baltic Sea : Germany, Sweden, Poland and Lithuania

- This report of MORPHEUS Project Partners documents the outcome of WP3 – consumption patterns. Herein, the project aims to identify and quantify the emission of selected pharmaceuticals especially discharged via wastewater of WWTP. A comprehensive data research was performed and prioritized pharmaceutical consumption loads have been calculated for the model areas in Lithuania, Poland, Sweden and Germany. The outcome of this report can be summarized in the following key facts: 1. Consumption data is based on either sales data of wholesalers/pharmacies or data from health care institutions. The unit of consumption is number of reimbursed packages andDDD, respectively. 2. The resolution and data coverage (only partly including OTC, hospital etc.) is country- specific: The best time-resolution was found in Poland (monthly), the best spatial resolution in Lithuania (population group <3000). 3. Country-specific consumption was comparable by a developed value intake load per inhabitant per year. 4. For Swedish and German data, a comparison of different distributing sites of pharmaceuticals was possible (over the counter sales, application in hospitals, prescriptions/pharmacies). 5. For Lithuanian data, the high spatial resolution enabled a potential correlation with the local demographic data in regard of consumption of beta blocking pharmaceuticals. 6. For Polish data, a seasonal variation can be clearly shown for antibiotics. 7. For German data, an accumulation within the river catchment shows that a complete picture of the whole system is essential to understand the actual burden of pharmaceuticals in the environment. 8. Comparing the model areas, all results have been in the same order of magnitude more or less depending on consumption and doses. Metformin and diverse analgesics revealed high intake loads. In general, mostlyGerman intake loads exceed the others which may lies in the fact that the selection of pharmaceuticals is mostly based on German literature and statistics due to availability/accessibility. An adjustment to each country-specific list may lead to other results. 9. Some substances remain unclear: Contrast media and hormones are still a matter of burden to the environment but caused some difficulties during data collection andresearch.

Quantum phase dynamics in an LC shunted Josephson junction

- We have studied both theoretically and experimentally how an LC series circuit connected in parallel to a Josephson junction influences the Josephson dynamics. The presence of the shell circuit introduces two energy scales, which in specific cases, can strongly differ from the plasma frequency of the isolated junction. Josephson junctions were manufactured using the Nb/Al-AlOx/Nb fabrication technology with various on-chip LC shunt circuits. Spectroscopic measurements in the quantum limit show excellent agreement with theory taking into account the shunt inductance and capacitance in the resistively and capacitively shunted junction model. The results clearly show that the dynamics of the system are two-dimensional, resulting in two resonant modes of the system. These findings have important implications for the design and operation of Josephson junction based quantum bits. (C) 2011 American Institute of Physics. [doi: 10.1063/1.3581139]
