    • We present the catalog of optical and infrared counterparts of the Chandra COSMOS-Legacy Survey, a 4.6 Ms Chandra program on the 2.2 deg2 of the COSMOS field, combination of 56 new overlapping observations obtained in Cycle 14 with the previous C-COSMOS survey. In this Paper we report the i, K, and 3.6 μm identifications of the 2273 X-ray point sources detected in the new Cycle 14 observations. We use the likelihood ratio technique to derive the association of optical/infrared (IR) counterparts for 97% of the X-ray sources. We also update the information for the 1743 sources detected in C-COSMOS, using new K and 3.6 μm information not available when the C-COSMOS analysis was performed. The final catalog contains 4016 X-ray sources, 97% of which have an optical/IR counterpart and a photometric redshift, while ≃54% of the sources have a spectroscopic redshift. The full catalog, including spectroscopic and photometric redshifts and optical and X-ray properties described here in detail, is available online. We study several X-ray to optical (X/O) properties: with our large statistics we put better constraints on the X/O flux ratio locus, finding a shift toward faint optical magnitudes in both soft and hard X-ray band. We confirm the existence of a correlation between X/O and the the 2-10 keV luminosity for Type 2 sources. We extend to low luminosities the analysis of the correlation between the fraction of obscured AGNs and the hard band luminosity, finding a different behavior between the optically and X-ray classified obscured fraction.

  • Resolving the ISM at the Peak of Cosmic Star Formation with ALMA: The Distribution of CO and Dust Continuum in z ∼2.5 Submillimeter Galaxies

    • We use Atacama Large Millimeter Array (ALMA) observations of four submillimeter galaxies (SMGs) at z ∼2-3 to investigate the spatially resolved properties of the interstellar medium (ISM) at scales of 1-5 kpc (0.″1-0.″6). The velocity fields of our sources, traced by the12CO(J =3-2) emission, are consistent with disk rotation to the first order, implying average dynamical masses of ∼3 1011within two half-light radii. Through a Bayesian approach we investigate the uncertainties inherent to dynamically constraining total gas masses. We explore the covariance between the stellar mass-to-light ratio and CO-to-H2conversion factor, αCO, finding values of for dark matter fractions of 15%. We show that the resolved spatial distribution of the gas and dust continuum can be uncorrelated to the stellar emission, challenging energy balance assumptions in global SED fitting. Through a stacking analysis of the resolved radial profiles of the CO(3-2), stellar, and dust continuum emission in SMG samples, we find that the cool molecular gas emission in these sources (radii ∼5-14 kpc) is clearly more extended than the rest-frame ∼250 μm dust continuum by a factor >2. We propose that assuming a constant dust-to-gas ratio, this apparent difference in sizes can be explained by temperature and optical depth gradients alone. Our results suggest that caution must be exercised when extrapolating morphological properties of dust continuum observations to conclusions about the molecular gas phase of the interstellar medium (ISM).

  • Phenome-wide association analysis of LDL-cholesterol lowering genetic variants in PCSK9

    • Background: We characterised the phenotypic consequence of genetic variation at the PCSK9 locus and compared findings with recent trials of pharmacological inhibitors of PCSK9. Methods: Published and individual participant level data (300,000+ participants) were combined to construct a weighted PCSK9 gene-centric score (GS). Seventeen randomized placebo controlled PCSK9 inhibitor trials were included, providing data on 79,578 participants. Results were scaled to a one mmol/L lower LDL-C concentration. Results: The PCSK9 GS (comprising 4 SNPs) associations with plasma lipid and apolipoprotein levels were consistent in direction with treatment effects. The GS odds ratio (OR) for myocardial infarction (MI) was 0.53 (95% CI 0.42; 0.68), compared to a PCSK9 inhibitor effect of 0.90 (95% CI 0.86; 0.93). For ischemic stroke ORs were 0.84 (95% CI 0.57; 1.22) for the GS, compared to 0.85 (95% CI 0.78; 0.93) in the drug trials. ORs with type 2 diabetes mellitus (T2DM) were 1.29 (95% CI 1.11; 1.50) for the GS, as compared to 1.00 (95% CI 0.96; 1.04) for incident T2DM in PCSK9 inhibitor trials. No genetic associations were observed for cancer, heart failure, atrial fibrillation, chronic obstructive pulmonary disease, or Alzheimer's disease - outcomes for which large-scale trial data were unavailable. Conclusions: Genetic variation at the PCSK9 locus recapitulates the effects of therapeutic inhibition of PCSK9 on major blood lipid fractions and MI. While indicating an increased risk of T2DM, no other possible safety concerns were shown; although precision was moderate.

    • Purified B subunit of cholera toxin retains membrane-binding capacity and protective immunogenicity and yet has no toxic activity as tested in animals. These properties suggest that B subunit might be a promising immunogen, particularly as an oral vaccine, for stimulating protective antitoxic immunity against cholera in man. A method has been elaborated which allows preparation of +/- 10 grams of pure B subunit per fermentor culture cycle. As tested in both Swedish and Bangladeshi volunteers purified B subunit alone or in combination with conventional whole cell vaccine gives no side-effects at all when given orally and only very mild local reactions after parenteral administration. A single peroral or intramuscular immunization with B subunit has given significant intestinal IgA antitoxin antibody formation in 75-85% of Bangladeshi women tested; however, the duration of the response was longer after the oral route. The preliminary results of a recent study (Svennerholm, A.M., Jertborn, M., Gothefors, L., Karim, A., Sack, D. and Holmgren, J., to be published) have further shown that two peroral immunizations of Bangladeshi volunteers with a combined B subunit--whole cell cholera vaccine give rise to mucosal IgA antitoxin as well as anti-lipopolysaccharide antibody formation which closely resemble these antibody responses in cholera convalescents. The combined vaccine also evoked a local immunologic memory comparable to that induced by clinical disease.

    • Mucosal and systemic immune responses to a new oral cholera vaccine, consisting of the B subunit plus killed vibrios, were studied in Bangladeshi volunteers and compared with those to clinical cholera. A single peroral dose of vaccine induced a local IgA antitoxin response in intestinal-lavage fluid of seven of eight vaccinees; the response closely mimicked that of patients convalescing from cholera, and evidence of the induction of local immunologic memory was found as well. Two peroral doses were needed for stimulation of an intestinal IgA immune response to the lipopolysaccharide of Vibrio cholerae that was comparable to the response obtained after clinical cholera. This response to peroral immunization was considerably stronger than that to parenteral vaccination, although the intramuscular route gave rise to the strongest IgG antitoxin and antilipolysaccharide responses in serum. The results suggest that B subunit-whole cell vaccine, when given in at least two oral doses, may be a good candidate for use in cholera prophylaxis.

  • Electrochemical and structural characterization of BaCe 0.7 Zr 0.15 Y 0.1 Zn 0.05 O 3-δ as an electrolyte for SOFC-H

    • As a potential electrolyte for proton-conducting solid oxide fuel cells (SOFC-Hs) and to get better protonic conductivity and stability, zinc doped BCZY material has been found to be promising. In this study, we report a new composition of proton conductors BaCe0.7Zr0.15Y01Zn0.05O3-s (BCZYZn10) which was investigated using XRD, SEM and conductivity measurements. Rietveld refinement of the XRD data revel a cubic perovskite structure with Pm-3m space group. Rietveld analysis of BaCe07Zr0.15Y01Zn0.05O3-5 shows the unit cell parameter is a = 4.3582(7) A. Scanning electron microscopy images shows that the grain sizes are large and compact which gives the sample high density and good protonic conductivity. The total conductivity in wet atmosphere is significantly higher than that of dry condition and the conductivity was found to be 0.004032 Scm-1 and 0.00164 Scm-1 at 600 °C in wet and dry Ar, respectively. This study indicated that perovskite electrolyte BCZYZn10 is a promising material for the next generation intermediate temperature solid oxide fuel cells (IT-SOFCs).

    • Monogalactosyldiacylglycerol (MGDG) is the major lipid constituent of chloroplast membranes and has been proposed to act directly in several important plastidic processes, particularly during photosynthesis. In this study, the effect of MGDG deficiency, as observed in the monogalactosyldiacylglycerol synthase1-1 (mgd1-1) mutant, on chloroplast protein targeting, phototransformation of pigments, and photosynthetic light reactions was analyzed. The targeting of plastid proteins into or across the envelope, or into the thylakoid membrane, was not different from wild-type in the mgd1 mutant, suggesting that the residual amount of MGDG in mgd1 was sufficient to maintain functional targeting mechanisms. In dark-grown plants, the ratio of bound protochlorophyllide (Pchlide, F656) to free Pchlide (F631) was increased in mgd1 compared to the wild type. Increased levels of the photoconvertible pigment-protein complex (F656), which is photoprotective and suppresses photooxidative damage caused by an excess of free Pchlide, may be an adaptive response to the mgd1 mutation. Leaves of mgd1 suffered from a massively impaired capacity for thermal dissipation of excess light due to an inefficient operation of the xanthophyll cycle; the mutant contained less zeaxanthin and more violaxanthin than wild type after 60 min of high-light exposure and suffered from increased photosystem II photoinhibition. This is attributable to an increased conductivity of the thylakoid membrane at high light intensities, so that the proton motive force is reduced and the thylakoid lumen is less acidic than in wild type. Thus, the pH-dependent activation of the violaxanthin de-epoxidase and of the PsbS protein is impaired.
