    • Municipal wastewater contains emergent chemical and biological pollutants that are resistant to conventional wastewater treatments. Therefore, the focus of the current study was to address the challenge of removing emergent chemical and biological pollutants present in municipal wastewater. To achieve this, a photo electro-catalytic (PEC) treatment approach was employed, focusing on the removal of both micro and biological pollutants that are of emergent concern, as well as the reduction of Chemical Oxidation Demand (COD) and Total Organic Carbon (TOC). The treatment involved the use of a modified multi-layer catalytic anode photo-electroactive anode as an effective anode for PEC treatment of municipal wastewater. In the continuous mode of operation, %COD removal was optimized for the treatment of municipal wastewater under Ultra-Violet C (UVc), 280 nm, and Visible (Vis) radiation, 400 nm. Therefore, a comparative study was performed to investigate the effect of Vis radiation on %COD removal, micropollutants removal, and disinfection of municipal wastewater. Micropollutants present in municipal wastewater were effectively oxidized/degraded with the highest reduction rate between 100% and 80% under the influence of UVc and Vis radiation respectively by the PEC treatment process. Disinfection of various microorganisms present in the wastewater with the effect of UVc and Vis assisted PEC treatment was also monitored. Overall, 75–80% of the disinfection of municipal wastewater was contributed by the modified multi-layer catalytic anode. The UVc in the PEC system, contributes approximately 20–25% to the overall disinfection of municipal wastewater.

    • Cytotoxic T lymphocytes (CTLs) recognize peptides presented by HLA class I molecules on the cell surface. The C terminus of these CTL epitopes is considered to be produced by the proteasome. Here we demonstrate that the cytosolic endopeptidases nardilysin and thimet oligopeptidase (TOP) complemented proteasome activity. Nardilysin and TOP were required, either together or alone, for the generation of a tumor-specific CTL epitope from PRAME, an immunodominant CTL epitope from Epstein-Barr virus protein EBNA3C, and a clinically important epitope from the melanoma protein MART-1. TOP functioned as C-terminal trimming peptidase in antigen processing, and nardilysin contributed to both the C-terminal and N-terminal generation of CTL epitopes. By broadening the antigenic peptide repertoire, nardilysin and TOP strengthen the immune defense against intracellular pathogens and cancer.

  • Epidemiological and Pathological Characteristics of Cutaneous Leishmaniasis from Baluchistan Province of Pakistan

    • Cutaneous Leishmaniasis (CL) is considered a neglected tropical disease which in Pakistan can now be considered as growing public health problem. The exact figures on the magnitude of disease are lacking both at national and regional level and only a few health centers are available for diagnosis of CL. The present study was designed to identify the epidemiology of CL infection from August 2018 to December 2019 and to assess clinical aspects of CL in Baluchistan Province of Pakistan. A total of 4072 clinically suspected CL cases were analysed statistically. The highest number of CL cases were reported in May, followed by April, January and then July, February, and June and lowest number of cases were observed in March and November. The highest prevalence rate was found in males where 38% of reported cases were aged 0-9 years. The majority (24.4%) of lesions were found on the hands followed by the face in which cheeks, ears and nose were the effected organs. About 50% of the participants have single lesion while 14% of the participants had two and nearly 3% of the participants have six lesions. The atypical clinical presentations were observed in Baluchistan and common unusual presentations were lupus erythematosus. The study findings suggest that more epidemiological studies and health education campaigns are needed for the population awareness regarding CL in Baluchistan. It is recommended that risk factors should be evaluated to establish the control and management strategies to prevent disease at individual and community level.

  • Fabrication, spark plasma consolidation, and thermoelectric evaluation of nanostructured CoSb3

    • Nanostructured powders of thermoelectric (TE) CoSb3 compounds were synthesized using a chemical alloying method. This method involved co-precipitation of oxalate precursors in aqueous solution with controlled pH, followed by thermochemical treatments including calcination and reduction to produce stoichiometric nanostructured CoSb3. Moreover, CoSb3 nanoparticles were consolidated by spark plasma sintering (SPS) with a very brief processing time. Very high compaction densities (>95%) were achieved and the grain growth was almost negligible during consolidation. An iterative procedure was developed to maintain pre-consolidation particle size and to compensate Sb evaporation during reduction. Significant changes in particle size and morphology were observed, and the post-reduction cooling was found to be an important stage in the process. The spark plasma sintering (SPS) parameters were optimized to minimize the grain growth while achieving sufficient densification. Grain sizes in the range of 500 nm to 1 mu m, with compaction density of 95-98% were obtained. Preliminary measurements of thermal diffusivity and conductivity showed the dependence on grain size as well as on porosity. TE transport properties were measured in the temperature range of 300-650 K. Sample showed p-type behavior with a positive Seebeck coefficient, which increases with increasing temperature. Electrical conductivity measurements indicate metallic behavior and it decreases with increasing temperature. Thermal conductivity also decreases with increasing temperature and major contribution is due to the lattice component. A TE figure of merit of 0.15 was achieved for high purity CoSb3 nanostructured TE material at 650 K and these results are comparable with the values reported for the best unfilled/undoped CoSb3 in the literature.

  • Functional characterization of human Kindlin-2 core promoter identifies a key role of SP1 in Kindlin-2 transcriptional regulation

    • Kindlin-2 is a recently identified FERM and PH domain containing integrin interacting protein. Kindlin-2 is ubiquitously expressed in normal tissues. So far, much effort has been spent exploring the functional aspects of Kindlin-2. However, the transcriptional regulation of Kindlin-2 has not yet been investigated. In this study we identified and functionally characterized the promoter of the human Kindlin-2 gene. We show that the core promoter of Kindlin-2 is a 39 base pair long GC rich fragment located −122/-83 upstream of the Kindlin-2 transcription start site. Functional characterization of this core promoter region by both in silico as well as in vitro/in vivo analysis shows that the transcription factor SP1 plays an important role in regulation of Kindlin-2 expression.
